Selected AbstractsEfficacy of laser Doppler flowmetry for the diagnosis of revascularization of reimplanted immature dog teethDENTAL TRAUMATOLOGY, Issue 2 2001Kallaya Yanpiset Abstract , This study was performed to assess if laser Doppler flowmetry (LDF) is an improved method for the detection of revascularization of replanted teeth. Teeth were extracted and reimplanted under different experimental conditions. LDF readings were taken before extraction and weekly for 3 months. In control teeth, LDF baseline readings were taken and then repeated after the apical blood vessels were cut surgically. At the end of 3 months it was determined radiographically and histologically whether revascularization had occurred, i.e. vitality had returned. Results: LDF readings correctly predicted the pulp status (vital vs. non-vital) in 83.7% of the readings. 73.9% (17 of 23) were correct for the vital teeth and 95% (19 of 20) were correct for the non-vital teeth. Fisher's exact test (2-tail) indicated that there was no significant association between the efficacy of LDF and tooth type (P=0.166), although P2 was the least accurate tooth tested. Wilcoxon's matched-pair signed rank test demonstrated that in the revascularized (vital) teeth, the flux value between the baseline and week 2 dropped significantly (P=0.0001), increased significantly from week 2 to week 4 (P=0.0001) and then decreased steadily until week 12. However, at week 12 the flux was still significantly higher than at week 2 (P=0.010). In the teeth that failed to revascularize, the flux value dropped significantly by weeks 1 and 2 (P=0.004 and P=0.0001, respectively). Flux values did not increase from week 2. A Fast Fourier Transform (FFT) analysis confirmed a pulse of dominant frequency of 2 Hz in the teeth that returned to vitality and the lack thereof in those that stayed non-vital. One tooth in which the flux value evaluation indicated a non-vital tooth but the radiographic/histologic findings showed vital (false negative) possessed a pulse of dominant frequency and proved by this method to have successfully revascularized. Van Der Bijl DSc Continued interest in the various routes by which sumatriptan may be administered prompted us to investigate its passage through buccal mucosa. Because human buccal mucosa is scarce, we proposed using the relatively abundant vaginal mucosa, which has been shown to have comparable diffusion rates for a number of widely varying molecules, as a model of buccal mucosa. In addition, by comparing these two tissues with respect to their permeability to sumatriptan, the human vaginal/buccal mucosa model could be further evaluated. Clinically healthy human vaginal and buccal mucosa specimens were used in the permeability studies. Permeability to sumatriptan was determined using a continuous flow-through diffusion system in the presence and absence of permeation enhancers. No statistically significant differences in permeability could be demonstrated for both mucosae toward sumatriptan. Flux values obtained in the absence and presence of glycodeoxycholate and lauric acid (1:1 molar ratio) to sumatriptan of buccal and vaginal mucosa, respectively, were not significantly different. The results obtained further support the hypothesis of the vaginal/buccal mucosal in vitro permeability model and suggest that this model may be used in conjunction with various absorption enhancers. Further studies on the buccal route of absorption of sumatriptan are thus warranted. Mimouni Abstract This paper shows that it is possible to extend the principle of field-oriented control (FOC) approach to a double-star induction motor (DSIM). In the first stage, a robust variable structure current controller based on space phasor voltages PWM scheme is established. In this current controller design, only the stator currents and rotor speed sensors are used. In the second stage, the FOC method developed for DSIM is motivated by the minimization of the copper losses. The developed approach uses a loss model controller (LMC) and an adaptive rotor flux observer to compute the adequate rotor flux value minimizing the copper losses. The control variables are the stator currents or the machine input power. Compared to the constant rotor flux approach, it is proved that higher performances are achieved. However, the sensitivity of the FOC to parameter error of the machine still remains a problem. To guarantee the performance of the vector control, the stator and rotor resistances are adapted on-line, based on the Lyapunov theory. An appropriate choice of the reference model allows building a Lyapunov function by means of which the updating law can be found. Legoabe Abstract Objectives The aim of this study was to synthesise and determine the transdermal penetration of cytarabine alkylamide derivatives and assess the correlation of flux with physicochemical properties. Methods The alkylamide derivatives of cytarabine were synthesised by acylation at the N4-amino group by the mixed anhydride method. The in-vitro permeation studies were performed using the Franz diffusion cell methodology. Furthermore, partition coefficients (n -octanol,water) and aqueous solubility of the N4-alkylamide derivatives of cytarabine were determined in order to obtain information about their lipophilicity and hydrophilicity. Key findings The N4-alkylamides of cytarabine (acetyl, butanoyl, hexanoyl, octanoyl, and decanoyl derivatives) showed decreased hydrophilicity and increased lipophilicity. The log D values of the alkylamides were higher than that of the parent compound and increased linearly as the alkyl chain lengthened. N4-hexanoyl-4-amino-1-[(2R,3S,4R,5R)-3,4-dihydroxy-5-(hydroxymethyl)oxolan-2-yl] pyrimidin-2-one) showed the highest median steady-state flux (Jss) of 89.0 nmol/cm2 per h in the series, which shows a high statistical difference with the parent compound flux value (3.70 nmol/cm2 per h). Conclusions The prodrug approach appears to be a promising strategy for the enhancement of transdermal penetration of cytarabine. [source] Factors affecting fluid flow in strike,slip fault systems: coupled deformation and fluid flow modelling with application to the western Mount Isa Inlier, AustraliaGEOFLUIDS (ELECTRONIC), Issue 1 2009A. FORD Abstract Deformation and focused fluid flow within a mineralized system are critical in the genesis of hydrothermal ore deposits. Dilation and integrated fluid flux due to coupled deformation and fluid flow in simple strike,slip fault geometries were examined using finite difference analysis in three dimensions. A series of generic fault bend and fault jog geometries consistent with those seen in the western Mount Isa Inlier were modelled in order to understand how fault geometry parameters influence the dilation and integrated fluid flux. Fault dip, fault width, bend/jog angle, and length were varied, and a cross-cutting fault and contrasting rock types were included. The results demonstrate that low fault dips, the presence of contrasts in rock type, and wide faults produce highest dilation and integrated fluid flux values. Increasing fault bend lengths and angles increases dilation and integrated fluid flux, but increasing fault jog length or angle has the opposite effect. There is minimal difference between the outputs from the releasing and restraining fault bend and jog geometries. Model characteristics producing greater fluid flows and/or gradients can be used in a predictive capacity in order to focus exploration on regions with more favorable fault geometries, provided that the mineralized rocks had Mohr,Coulomb rheologies similar to the ones used in the models. [source] The surface radiation budget over North America: gridded data assessment and evaluation of regional climate modelsINTERNATIONAL JOURNAL OF CLIMATOLOGY, Issue 15 2009Marko Markovic Abstract While surface station observations of downwelling radiation offer accuracy at high temporal resolution, they do not easily allow an evaluation of model surface radiation budgets (SRB) over a wide geographical area. We evaluate three gridded SRB data sets against detailed observations from six surface radiation sites from the US surface radiation (SURFRAD) network. We subsequently use the most accurate surrogate observational data set for evaluation of model-simulated SRB. The data sets assessed are: ERA40,reanalysis of European Centre for Medium-Range Weather Forecasts (ECMWF), North American Regional Reanalysis (NARR),regional reanalysis of National Centres for Environmental Prediction (NCEP) and the surface radiative budget (SRB) from the International Satellite Cloud Climatology Project (ISCCP). Due to varying constraints with respect to temporal coverage of each data set, the evaluation period used in this study is 1996,2001, inclusive. The ERA40 downwelling longwave radiation (DLR) appears the most accurate surrogate observation, while both ERA40 and ISCCP show accurate results when the incoming shortwave radiation (ISR) is considered across the annual cycle. Winter DLR is less accurate in ISCCP with a positive bias and lack of very low (<200 Wm,2) flux values. The NARR SRB shows a large positive bias in the ISR throughout the annual cycle, linked to a significant underestimate of cloud cover. The ERA40 data are subsequently used to evaluate the simulated SRB in three regional climate models across North America. With respect to solar radiation, cloud cover biases are seen to be crucial, while for longwave fluxes both cloud fraction and in-cloud water content are important to simulate correctly. Inclusion of trace gases beyond H2O, CO2 and O3 appears necessary for an accurate calculation of clear-sky longwave radiation. Error compensation frequently occurs between the various components contributing to a model total-sky SRB. This is important to consider when trying to identify the underlying causes of errors in the simulated total SRB. Van der Bijl Abstract: Because alkaloids from areca nut, arecoline and arecaidine, have been implicated in the development of oral submucous fibrosis, we determined their diffusion kinetics through human buccal and vaginal mucosa. Four clinically healthy vaginal mucosa specimens (mean patient age± standard deviation: 47±15 years; age range: 31,60 years) and 4 buccal mucosa specimens from 2 male patients and 2 female patients (mean patient age±standard deviation: 31±9 years; age range: 17,53 years) were obtained during surgery. In vitro flux rates of reduced arecoline and arecaidine (r-arecoline and r-arecaidine) were determined by use of a flow-through diffusion apparatus. Analysis of variance, a Duncan multiple range test, and an unpaired t -test were used to determine steady state kinetics and flux differences over time intervals. Although statistically significant differences were observed between flux values for both alkaloids and tissues at certain time points, these were not considered to be of biological (clinical) significance. However, the flux rates across both mucosa of r-arecoline were significantly higher statistically than those of r-arecaidine. The findings demonstrated the differences in the diffusion kinetics between r-arecoline and r-arecaidine across human buccal and vaginal mucosa, an observation that could be explained in terms of their ionisation characteristics. Additionally, the results obtained further support the hypothesis that human vaginal mucosa can be used as a model for buccal mucosa in studies of permeability to various chemical compounds. Balaguer-Fernández Abstract We have successfully obtained sumatriptan transdermal systems with different polymer compositions: methyl cellulose (MC), polyvinyl pyrrolidone (PVP) and a polyvinyl pyrrolidone (PVP)-polyvinyl alcohol (PVA) mixture. The systems contained 1,2-propilenglycol (MC) or sorbitol as a plasticizer (PVP and PVP-PVA), methacrylate copolymer as an adhesive agent, and an occlusive liner. Azone® (5%, w/w) was incorporated into all the systems as a percutaneous enhancer. Transdermal systems are thin, transparent and non-adhesive when in a dry state. The permeation of sumatriptan succinate across pig ear skin was studied using the systems prepared. The formulation with MC polymer produced a statistically significant increment with respect to the PVP and PVP-PVA formulations (p,<,0.05). Azone® incorporation into the systems produced an increment in the sumatriptan flux values of all three transdermal systems with respect to those of the controls (p,<,0.05). Sloan Abstract It has been shown for homologous series of prodrugs that those members who were the more water soluble ones gave the greatest enhancement in topical delivery of the parent drug and not the more lipophilic ones. However, until recently models for topical delivery and equations to predict topical delivery focused only on lipid solubility (SLIPID) or partition coefficient (KOCT:AQ) and molecular volume (or molecular weight, MW) as parameters. Now several equations (transformed Potts,Guy or Series/Parallel) have been developed which include aqueous solubility (SAQ) as a parameter for predicting flux through skin. Experimental fluxes, solubilities, and MW from seven series of prodrugs have been fit to the transformed Potts,Guy equation to give coefficients for log solubility in isopropyl myristate (log SIPM) and log solubility in water (log SAQ) (0.53 and 0.47, respectively) which show, for parent drugs delivered by prodrugs from IPM in vitro through hairless mouse skin, that water solubility is almost as important as lipid solubility. When the transformed Potts,Guy equation was fit to data for the delivery of NSAID from mineral oil (MO) in vivo through human skin, the coefficients were 0.72 log SMO and 0.28 log SAQ. When the transformed Potts,Guy equation was fit to data for the delivery of their parent drugs by three series of prodrugs from water in vitro through hairless mouse skin the coefficients were 0.66 log SIPM and 0.34 log SAQ. [source] In silico genome-scale reconstruction and validation of the Corynebacterium glutamicum metabolic networkBIOTECHNOLOGY & BIOENGINEERING, Issue 2 2009Kjeld Raunkjćr Kjeldsen Abstract A genome-scale metabolic model of the Gram-positive bacteria Corynebacterium glutamicum ATCC 13032 was constructed comprising 446 reactions and 411 metabolites, based on the annotated genome and available biochemical information. The network was analyzed using constraint based methods. The model was extensively validated against published flux data, and flux distribution values were found to correlate well between simulations and experiments. The split pathway of the lysine synthesis pathway of C. glutamicum was investigated, and it was found that the direct dehydrogenase variant gave a higher lysine yield than the alternative succinyl pathway at high lysine production rates. The NADPH demand of the network was not found to be critical for lysine production until lysine yields exceeded 55% (mmol,lysine,(mmol,glucose),1). The model was validated during growth on the organic acids acetate and lactate. Comparable flux values between in silico model and experimental values were seen, although some differences in the phenotypic behavior between the model and the experimental data were observed.
