Effect of cream treatment on phospholipids and protein recovery in butter-making process

Summary A simple approach is proposed to recover native protein and phospholipid fractions from butter-making process using equipments available in dairy-processing plant. A washing treatment was used to remove protein from the cream and increase the phospholipids purity in buttermilk. Cream from a first separation was diluted with milk ultrafitration permeate and separated a second time. A 10X dilution factor reduced protein concentration in the cream from 1.6 ± 0.2 to 0.52 ± 0.03%. As a result, the phospholipids to protein ratio in buttermilk increased from 53 ± 10 to 172 ± 7 mg g,1. In butter-making process, an important portion of total phospholipids (,26%) is retained in butter. Butter serum made from washed cream could then be used to produce phospholipid concentrates with phospholipids to protein ratio of 473 ± 3 mg g,1. Interestingly, the extracts from butter serum are characterised by a higher proportion of sphingomyelin compared with those from buttermilk. [source]


Timing of Parental Separation and Attachment to Parents in Adolescence: Results of a Prospective Study from Birth to Age 16

Using prospective longitudinal data from the Christchurch Health and Development Study (CHDS), this paper examines the effects of parental separation on the quality of adolescents' attachment to parents and their perceptions of parental care and overprotection during childhood. Exposure to parental separation was significantly associated with lower attachment to parents in adolescence and more negative perceptions of maternal and paternal care and protection during childhood. When examined in relation to the developmental timing of first separation, a linear relationship between the age at first separation and later parental attachment and perceived parent,child relations was found. The younger the age of the child at the time of separation, the lower their subsequent parental attachment and the more likely they were to perceive both their mother and father as less caring and more overprotective. No gender differences were found in children's responses to parental separation. These findings persisted after control for the confounding effects of family social background, marital conflict, parenting, child behavior, and remarriage. Results supported the importance of the early childhood years for the development of a secure and enduring attachment relationship between children and their parents. [source]


Tailoring orthogonal proteomic routines to understand protein separation during ion exchange chromatography

Abstract Surface charge, molecular weight, and folding state are known to influence protein chromatographic behaviour onto ion exchangers. Experimentally, information related to such factors can be gathered via 2-DE methods. The application of 2-D PAGE under denaturing/reducing conditions was already shown to reveal separation trends within a large protein population from cell extracts. However, ion-exchange chromatography normally runs under native conditions. A tailored protocol consisting in a first separation based on IEF on ImmobilineTM strips under native conditions followed by a second dimension SDS-PAGE run was adopted. The chromatographic versus electrophoretic separation behaviours of two model proteins, thaumatin (TAU) and BSA, were compared to better understand which proteomic routine would be better suited to anticipate IEX chromatographic separations. It was observed that the information contained in the pI value obtained with the adapted 2-DE protocol showed better correlation with the IEX chromatographic behaviour. On the other hand, chromatographic separations performed in the presence of urea as a denaturant have demonstrated the potential influence of hydrodynamic radius/conformation on protein separation. Moreover, the information provided by such 2-D system correlated well with the chromatographic behaviour of an additional set of pure proteins. An initial prediction of protein ion-exchange chromatographic behaviour could be possible utilizing an experimental approach based on 2-DE running under milder chemical conditions. This technique provides information that more closely resembles the separation behaviour observed with a complex biotechnological feedstock. [source]


Spectral color reproduction minimizing spectral and perceptual color differences

Abstract In this article, we are combining minimization criteria in the colorant separation process for spectral color reproduction. The colorant separation is performed by inverting a spectral printer model: the spectral Yule-Nielsen modified Neugebauer model. The inversion of the spectral printer model is an optimization operation in which a criterion is minimized at each iteration. The approach we proposed minimizes a criterion defined by the weighted sum of a spectral difference and a perceptual color difference. The weights can be tuned with a parameter , , [0, 1]. Our goal is to decrease the spectral difference between the original data and its reproduction and also to consider perceptual color difference under different illuminant conditions. In order to find the best , value, we initially compare a pure colorimetric criterion and a pure spectral criterion for the reproduction, then we combine them. We perform four colorant separations: the first separation will minimize the 1976 CIELAB color difference where four illuminants are tested, the second separation will minimize an equally weighted summation of 1976 CIELAB color difference with the four illuminants tested independently, the third colorant separation will minimize a spectral difference, and the fourth colorant separation will combine a weighted sum of a spectral difference and one of the two first colorimetric differences previously introduced. This last colorant separation can be tuned with a parameter in order to emphasize on spectral or colorimetric difference. We use a six colorants printer with artificial inks for our experiments. The prints are simulated by the spectral Yule-Nielsen modified Neugebauer model. Two groups of data are used for our experiments. The first group describes the data printed by our printing system, which is represented by a regular grid in colorant space of the printer and the second group describes the data which is not originally produced by our printing system but mapped to the spectral printer gamut. The Esser test chart and the Macbeth Color Checker test chart have been selected for the second group. Spectral gamut mapping of this data is carried out before performing colorant separation. Our results show improvement for the colorant separations combining a sum of 1976 CIELAB color difference for a set of illuminants and for the colorant separation combining a sum of 1976 CIELAB color difference and spectral difference, especially in the case of spectral data originally produced by the printer. © 2008 Wiley Periodicals, Inc.
