Selected AbstractsThrough-Water Close Range Digital Photogrammetry in Flume and Field EnvironmentsTHE PHOTOGRAMMETRIC RECORD, Issue 99 2002Justin Butler Measurement of the structure of gravel-bed river surfaces is crucial for understanding both bed roughness and the sediment entrainment process. This paper describes the use of close range digital photogrammetry to measure and monitor change occurring in submerged river gravel-beds in both flume and field environments. High-resolution digital elevation models (DEMs) were obtained and two-media (through air and water) techniques were used to correct for the e.ects of refraction at the air/water interface. Although suitable refractive models have been developed, the use of proprietary software to generate DEMs automatically introduces the problem of how to re-establish collinearity. A simple refraction correction algorithm based upon analytical geometry was developed and is described. This algorithm was designed for use after initial DEM acquisition and allows any photogrammetric software package to be used for data acquisition. Application of this algorithm led to improvements in DEM accuracy by reducing the systematic, depth-dependent bias caused by refraction Research carried out in a flume environment allowed the algorithm to be tested by measuring a flooded and drained bed surface. Results suggest that close range digital photogrammetry can be used to extract high quality DEMs of submerged topography in both flume and field fluvial environments, which represents a particularly exciting development for fluvial geomorphologists [source] Near-Term Travel Speed Prediction Utilizing Hilbert,Huang TransformCOMPUTER-AIDED CIVIL AND INFRASTRUCTURE ENGINEERING, Issue 8 2009Khaled Hamad In this study, we propose an innovative methodology for such prediction. Because of the inherently direct derivation of travel time from speed data, the study was limited to the use of speed only as a single predictor. The proposed method is a hybrid one that combines the use of the empirical mode decomposition (EMD) and a multilayer feedforward neural network with backpropagation. The EMD is the key part of the Hilbert,Huang transform, which is a newly developed method at NASA for the analysis of nonstationary, nonlinear time series. The rationale for using the EMD is that because of the highly nonlinear and nonstationary nature of link speed series, by decomposing the time series into its basic components, more accurate forecasts would be obtained. We demonstrated the effectiveness of the proposed method by applying it to real-life loop detector data obtained from I-66 in Fairfax, Virginia. The prediction performance of the proposed method was found to be superior to previous forecasting techniques. Rigorous testing of the distribution of prediction errors revealed that the model produced unbiased predictions of speeds. The superiority of the proposed model was also verified during peak periods, midday, and night. In general, the method was accurate, computationally efficient, easy to implement in a field environment, and applicable to forecasting other traffic parameters. [source] Behavioural responses of dairy cattle to the stable fly, Stomoxys calcitrans, in an open field environmentMEDICAL AND VETERINARY ENTOMOLOGY, Issue 1 2006B. A. Mullens Abstract., Individual cows (25 in each of four herds) were monitored 8,10 times weekly for 12 weeks (stable fly season) on a southern California dairy, with 100 observations per cow. The numbers of biting stable flies, Stomoxys calcitrans (L.) (Diptera: Muscidae) on the front legs and the frequencies of four fly-repelling behaviours per 2-min observation period [head throws, front leg stamps, skin twitches (panniculus reflex) and tail flicks] were recorded. Fly numbers varied, peaking at 3.0,3.5 flies per leg in week 9 (late May). Weekly herd mean frequencies of fly-repelling behaviours were highly dependent on fly numbers, with a linear regression r2 > 0.8. Head throws and stamps were less frequent than skin twitches and tail flicks. Individual cows differed in numbers of stable flies and behaviours. Behaviours were correlated with flies for individual cows, but at a lower level than were herd means (r = 0.3,0.7). Cows that stamped more within a herd tended to have lower fly counts; other fly-repelling behaviours were less effective. Cows maintained ranks within a herd with regard to fly numbers (r = 0.47), head throws (0.48), leg stamps (0.64), skin twitches (0.69) and tail flicks (0.64). Older cows tended to harbour higher fly numbers and to stamp less relative to younger adult cows. Ratios of leg stamps and head throws to fly numbers dropped significantly through time, suggesting habituation to pain associated with fly biting. Tail flicks were not effective for repelling Stomoxys, but were easiest to quantify and may help in monitoring pest intensity. At this low,moderate fly pressure, no consistent impacts on milk yield were detected, but methods incorporating cow behaviour are recommended for future studies of economic impact. [source] Biosuppression of Botrytis cinerea in grapesPLANT PATHOLOGY, Issue 2 2006P. A. G. Elmer There is increasing interest in the use of biological control agents (BCAs) and plant resistance stimulants to suppress botrytis bunch rot in grapes, caused by Botrytis cinerea. Numerous different filamentous fungi, bacteria and yeasts have been selected as potential BCAs for control of grey mould based upon demonstrated antagonism towards B. cinerea. Biological suppression of the pathogen arises via competition for nutrients and space, the production of inhibitory metabolites and/or parasitism. Preformed and inducible grapevine defence mechanisms also contribute to disease suppression by preventing or delaying pathogenic infection. Furthermore, various biotic and abiotic agents can stimulate grapevine defence mechanisms and so elevate resistance to B. cinerea infection. Biosuppression of B. cinerea in vineyards, using BCAs and resistance stimulants, has been inconsistent when compared with that observed in controlled glasshouse or laboratory conditions. This may be attributable, in part, to the innate variability of the field environment. Research to improve field efficacy has focused on formulation improvement, the use of BCA mixtures and combinational approaches involving BCAs and plant resistance stimulants with complementary modes of action. [source] Short periods of prenatal stress affect growth, behaviour and hypothalamo,pituitary,adrenal axis activity in male guinea pig offspringTHE JOURNAL OF PHYSIOLOGY, Issue 3 2005Amita Kapoor Prenatal stress can have profound long-term influences on physiological function throughout the course of life. We hypothesized that focused periods of moderate prenatal stress at discrete time points in late gestation have differential effects on hypothalamo,pituitary,adrenal (HPA) axis function in adult guinea pig offspring, and that changes in HPA axis function will be associated with modification of anxiety-related behaviour. Pregnant guinea pigs were exposed to a strobe light for 2 h on gestational days (GD) 50, 51, 52 (PS50) or 60, 61, 62 (PS60) (gestation length ,70 days). A control group was left undisturbed throughout pregnancy. Behaviour was assessed in male offspring on postnatal day (PND)25 and PND70 by measurement of ambulatory activity and thigmotaxis (wall-seeking behaviour) in a novel open field environment. Subsequent to behavioural testing, male offspring were cannulated (PND75) to evaluate basal and activated HPA axis function. Body weight was significantly decreased in adult PS50 and PS60 offspring and this effect was apparent soon after weaning. The brain-to-body-weight ratio was significantly increased in adult PS50 males. Basal plasma cortisol levels were elevated in PS50 male offspring throughout the 24 h sampling period compared with controls. In response to an ACTH challenge and to exposure to an acute stressor, PS60 male offspring exhibited elevated plasma cortisol responses. Plasma testosterone concentrations were strikingly decreased in PS50 offspring. Thigmotaxis in the novel environment was increased in PS50 male offspring at PND25 and PND70, suggesting increased anxiety in these animals. In conclusion, prenatal stress during critical windows of neuroendocrine development programs growth, HPA axis function, and stress-related behaviour in adult male guinea pig offspring. Further, the nature of the effect is dependant on the timing of the maternal stress during pregnancy. [source] Turbulent flow over a dune: Green River, ColoradoEARTH SURFACE PROCESSES AND LANDFORMS, Issue 3 2005Jeremy G. Venditti Abstract Detailed echo-sounder and acoustic Doppler velocimeter measurements are used to assess the temporal and spatial structure of turbulent flow over a mobile dune in a wide, low-gradient, alluvial reach of the Green River. Based on the geometric position of the sensor over the bedforms, measurements were taken in the wake, in transitional flow at the bedform crest, and in the internal boundary layer. Spatial distributions of Reynolds shear stress, turbulent kinetic energy, turbulence intensity, and correlation coefficient are qualitatively consistent with those over fixed, two-dimensional bedforms in laboratory flows. Spectral and cospectral analysis demonstrates that energy levels in the lee of the crest (i.e. wake) are two to four times greater than over the crest itself, with minima over the stoss slope (within the developing internal boundary layer). The frequency structure in the wake is sharply defined with single, dominant peaks. Peak and total spectral and cross-spectral energies vary over the bedform in a manner consistent with wave-like perturbations that ,break' or ,roll up' into vortices that amalgamate, grow in size, and eventually diffuse as they are advected downstream. Fluid oscillations in the lee of the dune demonstrate Strouhal similarity between laboratory and field environments, and correspondence between the peak frequencies of these oscillations and the periodicity of surface boils was observed in the field. [source] Phylogenetic diversity of T4-like bacteriophages in Lake Baikal, East SiberiaFEMS MICROBIOLOGY LETTERS, Issue 2 2010Tatyana Vladimirovna Butina Abstract Among the tailed phages, the myoviruses, those with contractile tails, are widespread and diverse. An important component of the Myoviridae family is the genus ,T4-like viruses'. The present study was aimed at elucidating the molecular diversity of T4-type bacteriophages in Lake Baikal by partial sequencing of g23 genes of T4-type bacteriophages. Our study revealed that the g23 gene sequences investigated were highly diverse and different from those of T4-like bacteriophages and from g23 clones obtained from different environments. Phylogenetic analysis showed that all g23 fragments from Lake Baikal, except for the one sequence, were more closely related to marine T4 cyanophages and to previously described subgroups of uncultured T4 phages from marine and rice field environments. [source] Chronic effects of polychlorinated dibenzofurans on mink in laboratory and field environmentsINTEGRATED ENVIRONMENTAL ASSESSMENT AND MANAGEMENT, Issue 2 2009Matthew J Zwiernik Abstract Mink are often used as a sentinel species in ecological risk assessments of chemicals such as polychlorinated biphenyls (PCBs), dibenzo- p -dioxins (PCDDs), and dibenzofurans (PCDFs) that cause toxicity mediated through the aromatic hydrocarbon receptor. Considerable toxicological information is available on the effects of PCBs and PCDDs on mink, but limited toxicological information is available for PCDFs. Thus, exposure concentrations at which adverse effects occur could not be determined reliably for complex mixtures in which PCDFs dominate the total calculated concentration of 2,3,7,8-tetrachlorodibenzo- p -dioxin equivalent (TEQ). Two studies were conducted to evaluate the potential toxicity of PCDFs to mink. The first was a chronic exposure, conducted under controlled laboratory conditions, in which mink were exposed to 2,3,7,8-tetrachlorodibenzofuran (2,3,7,8-TCDF) concentrations as great as 2.4 × 103 ng 2,3,7,8-TCDF/kg wet-weight (ww) diet or 2.4 × 102 ng TEQ2006-WHO-mammal/kg ww diet. In that study, transient decreases in body masses of kits relative to the controls was the only statistically significant effect observed. The second study was a 3-y field study during which indicators of individual health, including hematological and morphological parameters, were determined for mink exposed chronically to a mixture of PCDDs and PCDFs under field conditions. In the field study, there were no statistically significant differences in any of the measured parameters between mink exposed to a median estimated dietary dose of 31 ng TEQ2006-WHO-mammal/kg ww and mink from an upstream reference area where they had a median dietary exposure of 0.68 ng TEQ2006-WHO-mammal/kg ww. In both studies, concentrations of TEQ2006-WHO-mammal to which the mink were exposed exceeded those at which adverse effects, based on studies with PCDD and PCB congeners, would have been expected. Yet in both instances where PCDF congeners were the sole or predominant source of the TEQ2006-WHO-mammal, predicted adverse effects were not observed. Taken together, the results of these studies suggest that the values of the mammalian-specific toxicity equivalency factors suggested by the World Health Organization overestimate the toxic potency of PCDFs to mink. Therefore, hazard cannot be accurately predicted by making comparisons to toxicity reference values derived from exposure studies conducted with PCBs or PCDDs in situations where mink are exposed to TEQ mixtures dominated by PCDFs. Glenn Abstract Aims:, To clone and characterize genes from the mycotoxigenic fungus, Fusarium verticillioides, which are associated with its ability to biotransform allelopathic benzoxazolinones produced by maize, wheat, and rye. Methods and Results:, Suppression subtractive hybridization identified F. verticillioides genes up-regulated in response to 2-benzoxazolinone (BOA), including a cluster of genes along chromosome 3. One of these genes, putatively encoding an arylamine N -acetyltransferase (NAT), was highly represented in the subtracted library and was of particular interest since previous analyses identified the FDB2 locus as possibly encoding transferase activity. The gene was subcloned and complemented a natural fdb2 mutant. Conversely, disruption of the gene eliminated the ability of F. verticillioides to metabolize BOA. Other genes in the cluster also were assessed using a complementation assay. Metabolic profiles of fdb2 mutants suggest that minor acylation activity occurred independently of the NAT activity encoded by FDB2. Conclusions:, The previously defined FDB2 locus was functionally associated with the gene encoding putative NAT activity, and the FDB2 gene was essential for biotransformation of BOA. The flanking gene FDB3 encodes a putative Zn(II)2Cys6 transcription factor and contributes to efficient BOA biotransformation but was not essential. Significance and Impact of the Study:, Biotransformation of benzoxazolinones by F. verticillioides may enhance its ecological fitness in maize field environments and our results provide greater understanding of the genes that modulate the biotransformation process. Additionally, this is the first homologue of the NAT gene family to be characterized in a filamentous fungus. [source] Heritable variation and genetic correlation of quantitative traits within and between ecotypes of Avena barbataJOURNAL OF EVOLUTIONARY BIOLOGY, Issue 3 2008K. M. GARDNER Abstract We examined heritable variation for quantitative traits within and between naturally occurring mesic and xeric ecotypes of the slender wild oat (Avena barbata), and in 188 recombinant inbred lines derived from a cross between the ecotypes. We measured a suite of seedling and adult traits in the greenhouse, as well as performance-related traits in field sites native to the two ecotypes. Although the ecotypes were genetically diverged for most traits, few traits showed significant heritable variation within either ecotype. In contrast, considerable heritable variation was released in the recombinant progeny of the cross, and transgressive segregation was apparent in all traits. Heritabilities were substantially greater in the greenhouse than in the field, and this was associated with an increase in environmental variance in the field, rather than a decrease in genetic variance. Strong genetic correlations were evident among the recombinants, such that 22 measured traits could be well represented by only seven underlying factors, which accounted for 80% of the total variation. The primary axis of variation in the greenhouse described a trade-off between vegetative and reproductive allocation, mediated by the date of first flowering, and fitness was strongly correlated with this trade-off. Other factors in the greenhouse described variation in size and in seedling traits. Lack of correlation among these factors represents the release of multivariate trait variation through recombination. In the field, a separate axis of variation in overall performance was found for each year/site combination. Performance was significantly correlated across field environments, but not significantly correlated between greenhouse and field. [source] Erysiphe trifolii, a newly recognized powdery mildew pathogen of peaPLANT PATHOLOGY, Issue 4 2010R. N. Attanayake Diversity of powdery mildew pathogens infecting pea (Pisum sativum) in the US Pacific Northwest was investigated using both molecular and morphological techniques. Phylogenetic analyses based on rDNA ITS sequences, in combination with assessment of morphological characters, defined two groups of powdery mildews infecting pea. Group I (five field samples and three glasshouse samples) had ITS sequences 99% similar to those of Erysiphe pisi in GenBank and exhibited simple, mycelioid type of chasmothecial appendages typical of E. pisi. Erysiphe pisi is normally considered as the powdery mildew pathogen of pea. Group II (four glasshouse samples and two field samples) had ITS sequences 99% similar to those of E. trifolii and produced chasmothecia with dichotomously branched appendages similar to those of E. trifolii. There are fourteen nucleotide differences in the ITS region between the two groups. The correlation of rDNA ITS sequences with teleomorphic features for each of the two groups confirms their identity. Repeated samplings and artificial inoculations indicate that both E. pisi and E. trifolii infect pea in the US Pacific Northwest. Erysiphe trifolii is not previously known as a pathogen of pea. The existence of two distinct powdery mildew species infecting pea in both glasshouse and field environments may interfere with the powdery mildew-resistance breeding programmes, and possibly explains putative instances of breakdown of resistance in previously resistant pea breeding lines.
