Selected AbstractsCell-surface phytase on Pichia pastoris cell wall offers great potential as a feed supplementFEMS MICROBIOLOGY LETTERS, Issue 1 2010Piyanun Harnpicharnchai Abstract Cell-surface expression of phytase allows the enzyme to be expressed and anchored on the cell surface of Pichia pastoris. This avoids tedious downstream processes such as purification and separation involved with extracellular expression. In addition, yeast cells with anchored proteins can be used as a whole-cell biocatalyst with high value added. In this work, the phytase was expressed on the cell surface of P. pastoris with a glycosylphosphatidylinositol anchoring system. The recombinant phytase was shown to be located at the cell surface. The cell-surface phytase exhibited high activity with an optimal temperature at 50,55 °C and two optimal pH peaks of 3 and 5.5. The surface-displayed phytase also exhibited similar pH stability and pepsin resistance to the native and secreted phytase. In vitro digestibility test showed that P. pastoris containing cell-surface phytase released phosphorus from feedstuff at a level similar to secreted phytase. Yeast cells expressing phytase also provide additional nutrients, especially biotin and niacin. Thus, P. pastoris with phytase displayed on its surface has a great potential as a whole-cell supplement to animal feed. O. Atteh Summary A perennial schrub, stevia, and its extracts are used as a natural sweetener and have been shown to possess antimicrobial properties. Stevia contains high levels of sweetening glycosides including stevioside which is thought to possess antimicrobial and antifungal properties. Little is known about the nutritional value of the schrub in livestock. This study determined the potential use of the shrub as a prebiotic animal feed supplement in light of the recent ban on the use of antibiotics in animal feed and the role of its constituent stevioside in the effects of the shrub. Male Cobb broiler chicks were fed a basal broiler diet without antibiotic but with performance enhancing enzyme mix (positive control), a basal diet without antibiotic and enzymes (negative control), or diets in which 2% of the negative control diet was replaced with either dried ground stevia leaves or 130 ppm pure stevioside during 2 week starter and 2 week grower periods. Body weight gains, feed conversion, abdominal fat deposition, plasma hormone and metabolites and caecal short chain fatty acids (SCFA) were measured in the broilers at 2 and 4 weeks of age. There was no significant effect of the treatments on feed intake during the starter period but birds fed diet supplemented with stevia leaves and stevioside consumed more feed (p < 0.05) than those fed the positive control diet during the grower period. Weight gain by birds fed the positive control and stevioside diets was higher (p < 0.05) than those fed other diets only during the starter period. Feed/gain ratio of birds fed the positive control and stevioside diets was superior (p < 0.05) to others. There was no effect of the treatments on nutrient retention and water content of the excreta. Dietary stevia leave and stevioside decreased total concentration of SCFA and changed their profile in the ceca. There was no effect of the treatments on pancreas weight. Dietary stevia reduced blood levels of glucose, triglycerides and triiodothyronine (T3) but had no effect on non-esterified fatty acids. In contrast, stevioside only decreased T3. Both the stevia leaves and stevioside diets significantly increased abdominal fat content. It is concluded that dietary enzyme growth promoters are beneficial to the broilers only during the starter stage and that inclusion of stevia leaves or stevioside has no beneficial effect on the performance of broilers. Rao Abstract Aims:, To isolate, clone and express a novel phytase gene (phy) from Bacillus sp. in Escherichia coli; to recover the active enzyme from inclusion bodies; and to characterize the recombinant phytase. Methods and Results:, The molecular weight of phytase was estimated as 40 kDa on SDS-polyacrylamide gel electrophoresis. A requirement of Ca2+ ions was found essential both for refolding and activity of the enzyme. Bacillus phytase exhibited a specific activity of 16 U mg,1 protein; it also revealed broad pH and temperature ranges of 5·0 to 8·0 and 25 to 70°C, respectively. The Km value of phytase for hydrolysis of sodium phytate has been determined as 0·392 mmol l,1. The activity of enzyme has been inhibited by EDTA. The enzyme exhibited ample thermostability upon exposure to high temperatures from 75 to 95°C. After 9 h of cultivation of transformed E. coli in the bioreactor, the cell biomass reached 26·81 g wet weight (ww) per l accounting for 4289 U enzyme activity compared with 1·978 g ww per l producing 256 U activity in shake-flask cultures. In silico analysis revealed a ,-propeller structure of phytase. Conclusions:, This is the first report of its kind on the purification and successful in vitro refolding of Bacillus phytase from the inclusion bodies formed in the transformed E. coli. Significance and Impact of the Study:, Efficient and reproducible protocols for cloning, expression, purification and in vitro refolding of Bacillus phytase enzyme from the transformed E. coli have been developed. The novel phytase, with broad pH and temperature range, renaturation ability and substrate specificity, appears promising as an ideal feed supplement. Identification of site between 179th amino acid leucine and 180th amino acid asparagine offers scope for insertion of small peptides/domains for production of chimeric genes without altering enzyme activity. [source] Effects of urea treatment of potato pulp and inclusion levels of potato pulp silage in supplements on digestibility and ruminal fermentation in beef steersANIMAL SCIENCE JOURNAL, Issue 6 2007Masahito SUGIMOTO ABSTRACT Six Wagyu (Japanese Black) steers fitted with a ruminal cannula were used in a split-plot design experiment comprising a 3 × 3 Latin square design (whole plot) and a randomized blocks design (subplot) to determine the effect of the treatment of potato pulp (PP) with urea and the effect of inclusion levels of PP silage in feed supplement on digestibility, ruminal in situ degradation and ruminal fermentation. The whole plot consisted of 20%, 50% and 80% PP silage (dry matter (DM) basis), with PP silage replacing formula feed. The subplot included untreated or 0.5% (on an as-fed basis) urea-treated PP. The treatment of PP with urea showed no effect on DM intake and digestibility. The percentage of the rapidly degradable DM fraction of the urea-treated PP silage was higher (P < 0.01) and the percentage of its slowly degradable DM fraction was lower (P < 0.01) than for the untreated PP silage. Ruminal ammonia concentration was greater (P < 0.01) for steers fed urea-treated PP silage than that for steers fed the untreated PP silage. The treatment of PP with urea caused a decrease in the molar proportion of acetate and an increase in the proportion of propionate in ruminal fluid after feeding. The rate of DM degradations in hay (linear, P < 0.01) and in PP silage decreased (linear, P < 0.01) as the inclusion level of PP silage increased. Increasing the inclusion level of PP silage in supplement decreased the effective degradability of DM in hay (linear, P < 0.05) and in PP silage (linear, P < 0.05). An increase in the amount of PP silage increased the molar proportion of acetate (linear, P < 0.01) and decreased the butyrate proportion (linear, P < 0.05) in ruminal fluid. The results suggest that urea treatment of PP facilitates microbial access to starch of PP silage in the rumen and that surplus level of PP silage in supplement have adverse effect on ruminal digestion. [source] Evaluation of indigenous marine periphytic Amphora, Navicula and Cymbella grown on substrate as feed supplement in Penaeus monodon postlarval hatchery systemAQUACULTURE NUTRITION, Issue 2 2009H. KHATOON Abstract Three isolated marine diatoms (Amphora, Navicula and Cymbella) grown on substrate were evaluated as feed supplement for Penaeus monodon postlarvae (PL) in hatchery system for a period of 19 days without changing water. Specific growth rate (day,1) (0.27 ± 0.0) and survival (%) (56.3 ± 1.8) of PLs were significantly higher (P < 0.05) in treatment tanks when compared with the control (0.20 ± 0.0; 36.0 ± 1.5, respectively). Shrimp PLs reared in substrate-based tanks had significantly higher (P < 0.05) levels of protein, lipid (521.0 ± 7.0; 304.0 ± 2 g kg,1 dry weight, respectively), ecosapentaenoic acid (EPA) and docosahexaenoic acid (DHA) (189.0 ± 2.0; 176.0 ± 2 g kg,1 of total fatty acid, respectively) than the control (435.0 ± 22.0; 258.0 ± 22 g kg,1 dry weight; 172.0 ± 5.0; 152 ± 2 g kg,1 total fatty acid, respectively). The periphytic diatoms contained protein and lipid (430,490; 230,260 g kg,1 dry weight, respectively), EPA (30,150 g kg,1 of total fatty acids), DHA (20,30 g kg,1 of total fatty acids) and nine essential amino acids. The results showed that isolated marine periphytic diatoms grown on substrate could be used as feed supplement in enhancing the growth and survival of P. monodon postlarvae. [source] Effects of salinity on the growth and proximate composition of selected tropical marine periphytic diatoms and cyanobacteriaAQUACULTURE RESEARCH, Issue 9 2010Helena Khatoon Abstract Marine periphytic cyanobacteria and diatoms have been examined as a potential source of feed supplement for rearing aquatic larvae in the aquaculture industry. Culture of the periphytic diatom Amphora sp., Navicula sp., Cymbella sp. and the cyanobacteria Oscillatoria sp. at different salinities showed significant changes in biomass and specific growth rates. Diatoms growth was significantly higher at 35 g L,1, while for cyanobacteria growth was better at 25 g L,1. Significantly higher levels of protein and lipid were found in diatoms at low salinities (15,25 g L,1) and an increase in carbohydrate at high salinities (30,35 g L,1). Conversely, cyanobacteria showed a significantly higher lipid content at 30,35 g L,1 compared with other salinity levels but no significant changes were observed in the protein and carbohydrate contents at different salinity levels. The present findings can be taken into consideration when culturing marine periphytic Amphora sp., Navicula sp., Cymbella sp. and Oscillatoria sp. to provide appropriate levels of protein, lipid and carbohydrate as feed supplement as well as for bioremediation in aquaculture. [source] Effects of Grazing on Bituminaria bituminosa (L) Stirton: A Potential Forage Crop in Mediterranean GrasslandsJOURNAL OF AGRONOMY AND CROP SCIENCE, Issue 6 2006M. Sternberg Abstract Plant traits of Bituminaria bituminosa, as affected by different intensities of cattle grazing, were studied in a Mediterranean grassland in Israel. B. bituminosa is a widespread Mediterranean perennial legume species that may potentially serve as a fodder crop in Mediterranean grasslands. The aims of the present study were: (i) to evaluate the responses of B. bituminosa to different cattle grazing intensities; (ii) to study functional traits associated with grazing tolerance; and (iii) to evaluate its potential as an alternative forage crop in the region. A total of 100 B. bituminosa plants were monitored in field conditions. During the growing season each individual was sampled five times and the following plant traits were monitored each time: (i) aboveground biomass production, (ii) plant height, (iii) specific leaf area (SLA), (iv) number of flowers, (v) seed mass and size, (vi) tannin concentration in leaves, (vii) total nitrogen in leaves, (viii) fibre concentration in leaves (Neutral Detergent Fiber), and (ix) in vitro dry matter digestibility. The results showed that grazing intensity and history of grazing affected B. bituminosa performance. Plant biomass, height, and flower and seed production were all reduced when plants were exposed to cattle grazing. However, under moderate grazing intensities, its plant cover remained relatively stable indicating a potential tolerance under this stocking rate. The nutritious characteristics of B. bituminosa leaves were good, and the condensed tannins concentration found indicated favourable conditions for digestion. Moreover, the in vitro digestibility studies indicated relatively high values (46,51 %) of digestion. B. bituminosa may be considered as a potential crop for cattle feeding in Mediterranean grasslands. Growing this plant in dense stands in rotational paddocks may provide alternative sources of natural fodder protein, reducing the potential costs of artificial feed supplements. [source] Utilization of prebiotic carbohydrates by yeasts of therapeutic relevanceLETTERS IN APPLIED MICROBIOLOGY, Issue 4 2001G. Mitterdorfer Aims: To investigate 17 strains of therapeutically relevant strains of Saccharomyces cerevisiae (including 10 strains of so-called S. boulardii) isolated from various pharmaceutical products, feed supplements and brewer's yeast for their capability of utilizing selected carbohydrates of prebiotic importance. Methods and Results: Automated turbidimetric measurements and conventional test combinations were used to examine the basic sugar assimilation profiles of the test strains. It was shown that none of the so-called S. boulardii strains utilized galactose and palatinose. Among the prebiotic substrates, the yeasts indicated a pronounced preference for metabolizing the fructo-oligosaccharides. Conclusions: Yeast strains of therapeutic relevance can be successfully combined with certain prebiotics in synbiotic formulations. Significance and Impact of the Study: The results of this study may serve as a basis for the development of new pharmaceutical preparations for medical therapy and a better understanding of intestinal micro-ecology. [source] Assessing the use of a dietary probiotic/prebiotic as an enhancer of spinefoot rabbitfish Siganus rivulatus survival and growthAQUACULTURE NUTRITION, Issue 6 2007A.Y. EL-DAKAR Abstract The use of prebiotics and probiotics as feed supplements that improve efficiency of intestinal bacteria is becoming de rigueur in animal husbandry in many regions worldwide. We tested the effects of a commercial probiotic (Biogen®) containing allicin, high unit hydrolytic enzyme, Bacillus subtilis spores and ginseng extracts on survival, growth, carcass composition and feed cost/benefit in rabbitfish Siganus rivulatus. Fifteen net cages (100 × 100 × 40 cm; L × W × H) were stocked with 10 juvenile rabbitfish (10.3 g per fish) each and placed in a large rectangular tank and offered feed at 4% body weight daily. Cages were offered one of five isonitrogenous and isocaloric diets containing 0, 1, 2, 3 and 4 g kg,1 probiotic at three replicates per treatment for 98 days. Fish in all cages were weighed at 2-week intervals and feed regimen was adjusted accordingly. Rabbitfish offered the control diet exhibited lower growth and feed utilization than all experimental treatments. There was no effect of probiotic inclusion level on survival but growth was better at all inclusion levels than in the control. No significant differences (P > 0.05) in growth were observed among fish groups fed various levels of the probiotic. Carcass composition was not affected by dietary probiotic inclusion. Ultimately, when all variables are considered, Biogen® inclusion to diets appears to reduce feed cost per unit growth of rabbitfish. [source] Effect of dietary probiotic Biogen® supplementation as a growth promoter on growth performance and feed utilization of Nile tilapia Oreochromis niloticus (L.)AQUACULTURE RESEARCH, Issue 14 2006E R EL-Haroun Abstract Probiotic microbial feed supplements are gaining wide acceptance in livestock production, and may be applicable to aquaculture production systems. The present study was conducted to examine probiotic treatment in the fingerling diet of Nile tilapia Oreochromis niloticus (L.). A total of 240 of Nile tilapia fingerlings (weight ranged from 22.96 to 26.40 g) were divided into five experimental groups. The experiment was conducted for 120 days. Experimental diets were identical in all, except for the variation in probiotic levels. A probiotic (Biogen®) was used at 0% (diet 1), 0.5% (diet 2), 1.5% (diet 3), 2.0% (diet 4) and 2.5% (diet 5) inclusion rates in the experimental diets. The growth performance and nutrient utilization of Nile tilapia including weight gain, specific growth rate, protein efficiency ratio, protein productive value and energy retention were significantly (P,0.01) higher in the treatment receiving probiotic (Biogen®) than the control diet. No differences were observed for moisture, ash and protein content (P,0.01) among the experimental diets. The lowest gross energy and lipid contents were recorded for fish fed the diet containing 0.5% Biogen® (P,0.01). The production performance and subsequent cost,benefit analyses clearly indicated that the diets containing probiotic biogen recorded the highest net return and the lowest total cost compared with the control diet.
