FASTER FERTILIZATION RATE IN CONSPECIFIC VERSUS HETEROSPECIFIC MATINGS IN HOUSE MICE

Barriers to gene flow can arise at any stage in the reproductive sequence. Most studies of reproductive isolation focus on premating or postzygotic phenotypes, leaving the importance of differences in fertilization rate overlooked. Two closely related species of house mice, Mus domesticus and M. musculus, form a narrow hybrid zone in Europe, suggesting that one or more isolating factors operate in the face of ongoing gene flow. Here, we test for differences in fertilization rate using laboratory matings as well as in vitro sperm competition assays. In noncompetitive matings, we show that fertilization occurs significantly faster in conspecific versus heterospecific matings and that this difference arises after mating and before zygotes form. To further explore the mechanisms underlying this conspecific advantage, we used competitive in vitro assays to isolate gamete interactions. Surprisingly, we discovered that M. musculus sperm consistently outcompeted M. domesticus sperm regardless of which species donated ova. These results suggest that in vivo fertilization rate is mediated by interactions between sperm, the internal female environment, and/or contributions from male seminal fluid. We discuss the implications of faster conspecific fertilization in terms of reproductive isolation among these two naturally hybridizing species. [source]


Effect of N Fertilization Rate on Sugar Yield and Non-Sugar Impurities of Sugar Beets (Beta vulgaris) Grown Under Mediterranean Conditions

Abstract For three successive growing seasons (1999,2001), a completely randomized block design experiment was established at the surrounding area of each of four sugar beet processing plants of Hellenic Sugar Industry SA, Greece (a total of 12 experiments). Nitrogen was applied at five rates (0, 60, 120, 180 and 240 kg N ha,1) and six replications per rate. Nitrogen fertilization had site-specific effects on quantitative (fresh root and sugar yields) and qualitative (sucrose content, K, Na, , -amino N) traits. When data were combined over years and sites, fresh root and sugar yields were maximized at high N rates (330.75 and 295 kg N ha,1 respectively), as derived from quadratic functions fitted to data. In three trials, increased N rates had negative effects on root and sugar yield. These sites were characterized by high yield in control plots, light soil texture (sand > 50 %) and low CEC values. When data were converted into relative values (the ratio of the trait values to the control mean of each experiment), root and sugar yield was found to be maximized at higher N rates (350 and 316 kg N ha,1, respectively). Sucrose content was strongly and linearly reduced by the increased N rates when data were combined but a significant reduction with increasing N rates was found in only two sites. Non-sugar impurities (K, Na, , -amino N) were positively related to the increased N rates when data were combined. Sodium and , -amino N showed to be most affected by N fertilization as positive relationships were found in six and eight of 12 locations, respectively. Increased N supply resulted in higher soil NO3 -N concentrations (0,90 cm depth) at harvest which were related with amino N contents in sugar beet roots (in 1999 and 2001). [source]


Optimization of fertilization success in Sander vitreus is influenced by the sperm : egg ratio and ova storage

Walleye Sander vitreum eggs could be stored at 7° C for up to 30 h with 60% fertilization rates. Fertilization rates at 1 and 12° C fell to 17 and 5%, respectively. At spermatozoa : egg ratios <25 000 : 1, survival to the eyed stage declined precipitously. Beyond a ratio of 25 000 : 1, survival to the eyed stage did not increase >c. , 70%. Therefore, a standard spermatozoa : egg ratio of 25 000 : 1 was established and is recommended for studies concerning fertilizing success. [source]


Reversibility of estrogenic sex changes in zebrafish (Danio Rerio),

Abstract Development of male zebrafish (Danio rerio) courtship behavior was studied following estrogenic disruption of sexual differentiation. Sixty zebrafish were exposed at 28C to 5 ng/L (nominal concentration) of 17,-ethinylestradiol (EE2) from the egg stage until adulthood at four months of age, resulting in a female-biased sex ratio. Twenty-five EE2 -exposed phenotypic female zebrafish were subsequently held in clean water for eight months. During this period, eight phenotypic males developed. These phenotypic males demonstrated significant behavioral aberrations and a low fertilization rate compared to control males. [source]


Reproductive effects of the endocrine disruptor fenarimol on a Baltic amphipod Monoporeia affinis

Abstract An endocrine disruptor, the fungicide fenarimol, was investigated regarding its effects on reproduction and hormone (ecdysteroid) levels in the deposit-feeding amphipod Monoporeia affinis. In addition, the influence of food shortage, both by itself and in combination with fenarimol, on reproduction was examined. Field-collected amphipods were exposed in flow-through microcosms during the period of sexual maturation and mating in four treatment series: Control with low food, fenarimol with low food, control with high food, and fenarimol with high food. Fenarimol was added at a concentration of 0.3 mg/L in two pulses/week. Results show that fenarimol has a negative effect on fertilization rate and male mating ability. Results were supported by a tendency toward delayed male sexual development. Food shortage decreased weight in both sexes and retarded female oocyte development. Higher ecdysteroid levels were recorded in males than in females, and food shortage increased male ecdysteroid levels. No effect of fenarimol exposure on ecdysteroid levels was observed. No synergistic effects of fenarimol and food shortage could be distinguished in any variable examined. Thus, M. affinis was vulnerable to reproductive impairment by fenarimol, with effects on the next generation (i.e., a disturbed sexual development and fertilization ability). Food shortage has negative effects on M. affinis, but it does not enhance the effects of fenarimol. [source]

Projecting future N2O emissions from agricultural soils in Belgium

Abstract This study analyses the spatial and temporal variability of N2O emissions from the agricultural soils of Belgium. Annual N2O emission rates are estimated with two statistical models, MCROPS and MGRASS, which take account of the impact of changes in land use, climate, and nitrogen-fertilization rate. The models are used to simulate the temporal trend of N2O emissions between 1990 and 2050 for a 10, latitude and longitude grid. The results are also aggregated to the regional and national scale to facilitate comparison with other studies and national inventories. Changes in climate and land use are derived from the quantitative scenarios developed by the ATEAM project based on the Intergovernmental Panel on Climate Change-Special Report on Emissions Scenarios (IPCC-SRES) storylines. The average N2O flux for Belgium was estimated to be 8.6 × 106 kg N2O-N yr,1 (STD = 2.1 × 106 kg N2O-N yr,1) for the period 1990,2000. Fluxes estimated for a single year (1996) give a reasonable agreement with published results at the national and regional scales for the same year. The scenario-based simulations of future N2O emissions show the strong influence of land-use change. The scenarios A1FI, B1 and B2 produce similar results between 2001 and 2050 with a national emission rate in 2050 of 11.9 × 106 kg N2O-N yr,1. The A2 scenario, however, is very sensitive to the reduction in agricultural land areas (,14% compared with the 1990 baseline), which results in a reduced emission rate in 2050 of 8.3 × 106 kg N2O-N yr,1. Neither the climatic change scenarios nor the reduction in nitrogen fertilization rate could explain these results leading to the conclusion that N2O emissions from Belgian agricultural soils will be more markedly affected by changes in agricultural land areas. Comparison of the outcome of intracytoplasmic sperm injection in obstructive and non-obstructive azoospermia in the first cycle: a report of case series and meta-analysis

Summary To investigate the outcome of intracytoplasmic sperm injection with fresh and cryopreserved-thawed testicular spermatozoa in the first cycle in patients with obstructive azoospermia (OA) and non-obstructive azoospermia (NOA), a total of 90 cases, 48 OA and 42 NOA were studied. All patients underwent sperm retrieval by testicular sperm extraction (TESE) while their wives received conventional ovarian hyperstimulation. The hormone levels, testicular histology, the rates of sperm retrieval, fertilization, implantation and pregnancy were analysed and evaluated. This study and other four similar studies were subjected to meta-analysis. Sperm retrieval was successful in 100% OA and 61% NOA. Fresh spermatozoa were used in 87.5% and 92.4% of OA and NOA cases respectively; while cryopreserved-thawed spermatozoa were used in 12.5% and 7.6% of OA and NOA, respectively. The fertilization, implantation and clinical pregnancy rates were 65.5%, 15% and 25% respectively in OA group, and 54.2%, 5% and 23.1% respectively in NOA group. Sperm status (fresh or thawed), male partner's age, female age and male serum follicle-stimulating hormone had no significant effect upon fertilization rate, implantation rate, or pregnancy rate per embryo transfer. The results of meta-analysis indicate that there is no statistically significant difference in clinical pregnancy rates between the two groups. There was a significantly higher fertilization rate among OA patients in all analysed studies (95% CI = 14.29,15.71, d.f. 832, T = 1.96). In conclusion, although the fertilization rate was significantly higher in the OA group in our study and from the given meta-analysis, there were some differences as regards pregnancy rates. Although the overall effect was more or less similar pregnancy rates in both subtypes of azoospermia, this may not be true if non-male infertility variables were controlled for in all studies. Early life history of the Mediterranean gorgonian Paramuricea clavata: implications for population dynamics

Abstract. Knowledge about early stages of marine sessile invertebrates dispersing by means of free-swimming propagules is fundamental toward understanding their population biology. In this study, we describe and quantify survivorship of early stages of the gorgonian Paramuricea clavata to contribute to the understanding of their implications on population dynamics of this emblematic species of the Mediterranean hard-bottom communities. Spawning was recorded in June for the 3 years studied (2001,2003). High levels of fertilization rate were observed during the main spawning in 2001 and 2002 (62,69%). This appears to be related to the surface brooding mode of development, synchronous spawning, and high fecundity of the species. The timing of development of the blastulae was ,24 h and the planulae appeared after 48,72 h. In the laboratory, metamorphosis into polyp started between 8 and 25 d, but this timing may have been delayed by lack of unknown appropriate cues for settlement. The behavior of first developed planulae exhibited a marked negative phototaxis that may be a strategy to avoid competition with fast-growing algae on photophilous habitats. Despite the high fertilization rate in P. clavata, laboratory and field experiences indicate that survivorship through the planulae and polyp stages was very low, given that none of the settled polyps survived for >7 months in both experimental years. The low survival of the early stages may help explain the low recruitment rates observed in the field in addition to indicating major limitations for colonization of new areas and for persistence of the local population under disturbances. Field Pea Seeding Management for Semi-arid Mediterranean Conditions

Abstract The effects of seeding rate (30, 60 and 90 seeds m,2), seeding date (14 January, 28 January and 12 February), seed weight (0.18 and 0.25 g seed,1), seeding depth (3 and 6 cm), and phosphorus fertilization rate (17.5, 35.0 and 52.5 kg P ha,1) and placement method (banded or broadcasted) on field pea (Pisum sativum L.) development and seed yields were investigated in irrigated field experiments conducted in northern Jordan in 2000 and 2001. Results and treatment responses were consistent in both years. Seeding rate, seeding date, seed weight and rate and method of phosphorus fertilization had significant effects on most traits measured; planting depth however did not affect any of the traits. Generally a positive correlation was observed between each factor and seed yield and yield components, with the exception of a negative correlation between seeding rate and yield components, and seeding date and yield and yield components. Increase in seeding rate from 30 to 90 seeds m,2, and increase in P fertilization from 17.5 to 52.5 kg ha,1 alone increased seed yields by 50 and 41 %, respectively. Each delay of 2 weeks for seeding from mid-January resulted in reductions of 12 % in seed yields. Overall, the results revealed that a combination of early seeding (14 January), of large seeds at an high seeding rate (90 seeds m,2), with P fertilizer banding (52.5 kg P ha,1) maximize field pea yields in irrigated fields in semi-arid Mediterranean environments. With such management pea seed yields can be as high as 2800 kg ha,1. [source]


Developmental biology of medaka fish (Oryzias latipes) exposed to alkalinity stress

Summary Alkalinity stress is common in cultured aquatic animals and considered to be one of the major stress factors for fishes when they are transferred to saline-alkali waters. To evaluate potential effects of alkalinity on the developmental biology of Oryzias latipes, fertilized eggs, larvae and breeding fish were exposed to different carbonate alkalinity concentrations of 1.5,64.5 meq l,1, for 9, 120, and 60 days, respectively. The mortality of embryos significantly increased when exposed to the high concentrations (16.5,64.5 meq l,1). Although more than 50% of survived embryos hatched in 16.5 and 31.4 meq l,1 concentrations of carbonate alkalinity, most were not able to swim up after hatching. Morphological abnormalities such as coagulated embryos, halted embryo development, and hatching failure were observed at stages 15, 29,33 and 38 in high concentrations (31.4, 64.5 meq l,1). Almost all larvae in 16.5 and 31.4 meq l,1 treatments died 70 d post-hatch. Growth of juveniles exposed to carbonate alkalinity of 5.3 and 8.8 meq l,1 was not significantly different at 70 d and 120 d post-hatch. The number of eggs released by breeders, the fertilization rate and the hatching rate of eggs were significantly lower in the 31.4 meq l,1 treatment than in other treatments. Although medaka are capable of surviving in high alkalinities (31.4, 64.5 meq l,1) for an extended period of time, these conditions are stressful to the fish, especially at the embryonic and reproductive stages. Effect of prepared diet and vitamins A, E and C supplementation on the reproductive performance of cage-reared bighead carp Aristichthys nobilis (Richardson)

Twenty-month-old bighead carp, Aristichthys nobilis (Richardson), were fed prepared dry diets for 20 months in cages in Laguna de Bay, Philippines, to determine the effect on reproductive performance. The experimental diets were similar in composition except for the combinations of vitamins being tested. Diet 1 was supplemented with vitamins A, E and C; diets 2, 3 and 4 each lacked one of the supplementary vitamins; and diet 5 did not include any vitamin supplementation. Bighead carp that relied solely on natural food without a prepared diet served as a control. The total of six treatments each had two replicates. Results showed that the onset of gonad maturation was 2,3 months earlier in the fish that were fed the prepared diets regardless of vitamin supplementation, when compared with the fish that were fed natural food (control). Moreover, the prepared diets enhanced egg hatchability which was significantly higher in fish that were fed diet 1 (+ vitamins A, E and C, 80.5 ± 18.1%) and diet 3 (, vitamin E, 78.5 ± 1.1%) than in those fish that were fed natural food (control) (36.5 ± 31.3%). Mean number of 3-day-old larvae was highest in fish fed on diet 1 (34 525 ± 1732), followed closely by fish that were fed diet 3 (32 420 ± 3909). A low number of 3-day-old larvae was obtained from fish fed the natural diet (14 490 ± 4331) as well as in fish that were fed diet 2 (, vitamin A, 14 347 ± 4863), diet 4 (, vitamin C, 21 407 ± 5840) and diet 5 (, vitamin A, E and C, 12 191 ± 1439). Other criteria for reproduction such as relative fecundity, fertilization rate, and hatching rate did not differ significantly (P >,0.05) among treatments. The addition of vitamins also had no significant effects on weight gain of adult fish. [source]


Sustained, Natural Spawning of Southern Flounder Paralichthys lethostigma Under an Extended Photothermal Regime

Hormone-induced spawning of southern flounder Paralichthys lethostigma has produced substantial numbers of viable eggs, but wide variations in fertilization and hatch rates have been reported. Recently, sustained natural spawning of southern flounder broodstock, without hormone induction, has been achieved in our laboratory. Adults (average weight = 1.12 kg; N= 25), including 6 captured as juveniles in 1993 and 19 captured as adults during September 1998, were stocked in two 4.8-m3 controlled-environment tanks in October 1998 and held under natural photothermal conditions until January 1999, when an artificial winter photo-period of 10 L:14 D was initiated and then maintained through April 1999. Sex ratio was approximately 13 females:8 males:7 unknown. Natural spawning was observed in early December 1998 and increased in frequency to a peak in March 1999, before declining in late April. Water temperature ranged from 13.9 to 24.5 C during the spawning period. Natural spawnings over 142 d produced a total of 18.3 × 106 eggs, with a mean fertilization rate of 28.0% (range = 0,100%), yielding 4.94 × 106 fertilized eggs. The mean percentage of eggs that remained buoyant in full-strength seawater (34 ppt) was 41.3% (0,98%), while hatching rate of buoyant eggs was 37.3% (0,99%) and survival of yolksac larvae to the first-feeding stage was 30.2% (0,100%). Gonadal biopsies in late April identified six females from both tanks as probable spawners. A preliminary comparison suggests that natural spawning produced much larger numbers of viable eggs per female, with higher egg quality (i.e., fertilization and hatching success) than hormone-induced spawning. In contrast to natural spawning, hormone-induced strip-spawning enabled timing of spawnings to be more precisely controlled. These results suggest that a combination of both natural and hormone-induced spawning of photothermally conditioned fish will help produce the large numbers of eggs required to support commercial production. Production, Quality, and Low Temperature Incubation of Eggs of Atlantic Cod Gadus morhua and Haddock Melanogrammus aeglefinus in Captivity

Atlantic cod Gadus morhua and haddock Melanogrammus aeglefinus broodstock maintained under altered regimens of temperature and photoperiod spawned up to 8 mo per year. The cod broodstock produced viable embryos from October through June. The haddock broodstock produced viable embryos from December through May. Egg diameters were largest during the middle of the spawning season when water temperature was at a minimum, resulting in an inverse relationship between egg diameter and water temperature in both species. Egg quality was high, as evaluated by buoyancy, fertilization rate, regularity of early cleavage, and percent viable hatch. Low temperature incubation of cod and haddock eggs extended the embryonic period. Cod embryos tolerated a wider range of temperatures than haddock. High mortality (1 90%) was observed before hatching in haddock embryos incubated at 1 C. Atlantic cod embryos hatched at temperatures as low as ,1 C, extending the embryonic period to 59 d. At 8 C Atlantic cod and haddock embryos hatched in 11,12 d. To determine if extending the embryo incubation time by using low temperatures had a detrimental effect, embryos were incubated through hatch at either 1 C or 6 C, and the larvae from both groups reared at 6 C. Growth and early survival of larvae were comparable in both treatments. [source]


Assessing in vivo Fertilizing Capacity of Liquid-Preserved Boar Semen According to the ,Hanover Gilt Model'

Contents The goal of this study was to determine the ability of the Hanover gilt model to assess in vivo fertilizing capacity of preserved sperm and to consider whether any modifications to this model were needed. This model evaluates the fertilizing capacity of semen based on the fertilization rate, the rate of normal embryos and the accessory sperm count of 3,5-day embryos. Its distinguishing characteristics are the use of one-time insemination of sperm in reduced numbers, of spontaneously ovulating gilts and of ovulation detection through ultrasound examination of ovaries. Reduced sperm numbers allow for an accurate evaluation of the fertilizing potential of different semen treatments, thereby avoiding the compensatory effect of doses calibrated to maximize fertility. The model's usefulness was assessed in a trial run designed to compare the fertilizing capacity of liquid boar semen diluted into two different extenders. The diluent, the boar and the backflow, had no significant effect on any of the parameters studied. Gilts inseminated less than 24 h before ovulation had a significantly higher (p < 0.01) fertilization rate and accessory sperm cell count (p < 0.05) than those inseminated more than 24 h before ovulation. Very good/good embryos from homogeneous litters (only very good/good embryos were present) had a significantly higher (p < 0.01) accessory sperm count than those from heterogeneous litters (at least one embryo was of a different quality and/or oocytes were present). Both very good/good and degenerated/retarded embryos from heterogeneous litters had low accessory sperm numbers. This suggests that accessory sperm count is significantly related to the quality of the litter, but not to the quality of the embryo within gilts. It can be concluded that the Hanover gilt model is sensitive enough to show fertility differences (in this study, those associated with in vivo ageing of semen), while using relatively few gilts and little time. Problems, Study on feasibility of pCXN2-mIzumo as a potential immunocontraceptive antigen. Method of study, Two groups of mice received 100 ,g/mouse plasmids of pCXN2-mIzumo and pCXN2 respectively. RT-PCR Immunofluorescence assay and ELISA were performed to observe pCXN2-mIzumo expression and antibody response in the inoculated mice. Sperm penetration assay and animal mating were employed to detect differences of in vitro fertilization (IVF) rate and mean litter size between the experimental and control groups. Results, Izumo cDNA positive bands were detected in sample from mice immunized with pCXN2-mIzumo. IgG response started to rise at 2 weeks after first boost and reached the highest antibody titers at 2 weeks after third boost of immunization with pCXN2-mIzumo in the experimental mice. In vitro fertilization rate in the experimental group (11.57%) was significantly lower than that in control (36.60%). Significant difference of mean litter size between female experimental and control groups was observed, and there was significant negative correlation between individual anti-serum titers and litter size (r = ,0.308, P < 0.05). Conclusion, pCXN2-mIzumo plasmid possesses appreciable anti-fertility potential. [source]


Fibroblast Growth Factor (FGF), Intracellular Adhesion Molecule (sICAM-1) Level in Serum and Follicular Fluid of Infertile Women with Polycystic Ovarian Syndrome, Endometriosis and Tubal Damage, and their Effect on ICSI Outcome

PROBLEM: The objective of this study was to determine the concentration of fibroblast growth factor (FGF) and soluble intracellular adhesions molecule (sICAM-1) in serum and follicular fluid (FF) of polycystic ovary (PCO), endometriosis and tubal factor infertility and male factor infertility patients, and to investigate the relationship between these parameters and the outcome of intracytoplasmic sperm injection (ICSI). METHOD OF STUDY: The concentration of FGF and sICAM-1 in serum and FF were determined in patients undergoing controlled ovarian hyperstimulation (COH) for ICSI therapy for various etiology of infertility and the results of cytokines concentration and ICSI outcome were compared between the groups. Twenty patients with PCO (G.I), 17 with endometriosis (G.II), 19 with tubal damage (G.III) and 19 with male factor infertility (G.IV) were enrolled in this study. Quantitative determination of levels of FGF and sICAM-1 was performed using enzyme-linked immunosorbent assays (ELISAs). RESULTS: The FGF level in serum of PCO patients (G.I) were 4.8 ± 2.3 and in FF were 104.0 ± 39.0 pg/mL. The corresponding values in the endometriosis patients group (G.II) were 5.9 ± 3.1 and 125.4 ± 74.9 pg/mL. The concentration of FGF in tubal factor infertility group (G.III) in serum was significantly higher (P = 0.009) than those observed in the PCO group (G.I) 7.4 ± 4.5 pg/mL, whereas the concentration in FF was at the same level like the other groups investigated, 128.7 ± 75.9 pg/mL. Besides, the sICAM-1 (pg/ml) concentration in FF showed a significant difference between the groups investigated (G.I, 175.3 ± 52.8; G.II 194.4 ± 32.2; G.III 233.1 ± 54.3; and G.IV 215.1 ± 54.4 ng/mL; P = 0.003). The sICAM-1 levels in serum were not significantly different between the groups (217.0 ± 42.9; 216.3 ± 73.6; 254.8 ± 79.6; 237.56 ± 78.4 ng/ml; P = 0.267). The fertilization rate was significantly higher in G.III (66.0 ± 23.89%) in comparison to G.II (38.8 ± 33.9%; P = 0.014) or G.IV (38.7 ± 22.7%; P = 0.012). The pregnancy rates were similar in all groups (30, 35.3 and 35.0, 38.6%, respectively). CONCLUSION: Both, FGF and sICAM-1 are present in serum and FF of patients undergoing controlled ovarian hyperstimulation for ICSI therapy. The FGF concentration in serum differs significantly between the groups investigated, whereas, no significant difference could be observed in the FF concentration of FGF. On the other hand, the sICAM in serum showed no significant difference between the groups, whereas, sICAM in FF demonstrated a significant difference between the patient groups investigated. On the whole, the ICSI outcome was not related to serum or FF concentrations of FGF or sICAM-1. Therefore, the mean concentration of FGF and sICAM-1 in serum and in FF could not be used to predict the fertilization rate in an ICSI program. [source]


Effects of paternal cigarette smoking on testicular function, sperm fertilizing capacity, embryonic development, and blastocyst capacity for implantation in rats

Summary. We evaluated the effects of paternal smoking on testicular function, sperm fertilizing capacity, embryonic development, and blastocyst capacity for implantation. Rats of group A were exposed to cigarette smoke for 10 weeks. Rats of group B were exposed to the smoke of incense sticks for 10 weeks. Rats of group C served as a control group. Rats of group D were exposed to cigarette smoke for 7 weeks only. Experimental period was 10 weeks in all groups. At the end of the experimental period serum testosterone responses to human chorionic gonadotropin stimulation, andro-gen-binding protein activity in testicular cytosols, epididymal sperm motility, and oocyte fertilization rate, oocyte cleavage rate, and blastocyst development rate after in vitro fertilization (IVF) trials were significantly smaller in group A compared with groups B and C. In contrast, fertilization rate, cleavage rate, and blastocyst development rate after intracytoplasmic sperm injection (ICSI) procedures were not significantly different among groups A, B, C, and D. Both after IVF trials and ICSI techniques, the proportion of the alive offspring to the number of transferred oocytes was significantly smaller in group A than in groups B and C. Cigarette smoke-exposure results in a secretory deficiency of Leydig and Sertoli cells leading to an impaired epididymal sperm maturation process and diminished capacity of spermatozoa to penetrate oocytes. In addition paternal cigarette smoke exposure affects the embryonic ability for implantation. [source]


Analysis of segregation distortion and association of the bovine FGF2 with fertilization rate and early embryonic survival

Summary Fibroblast growth factor 2 (FGF2) plays an important role in fertility and early embryo development. The objectives of this study were to test the association of FGF2 polymorphisms with fertilization success in cattle using an in vitro fertilization experimental system and to investigate the mechanisms leading to the presence of rare alleles of FGF2 in the Holstein population. A total of 7502 fertilizations were performed and a total of 5049 embryos were produced to collect fertilization and embryo survival records. A total of 444 ovaries, from which oocytes were aspirated and fertilized, were genotyped for two single nucleotide polymorphisms (SNPs) previously identified in FGF2 (g.23G>T and g.11646A>G). Frequency of the TT genotype of the g.23G>T SNP was low in the ovary population (5.4%) and in a different Holstein cattle population (6.6%) examined in this study. Single SNP analysis showed that both SNPs were associated with early embryonic survival rate. Two-way interaction analysis revealed significant association of epistatic interaction between the SNPs with fertilization rate. To test whether or not low frequency of allele T for the g.23G>T SNP in the population is a result of a fertilization failure of T oocytes, semen from six GG bulls was used to fertilize a total of 458 oocytes obtained from 19 GT ovaries. A significant segregation distortion was observed for 169 embryos genotyped for the g.23G>T SNP. We conclude that oocytes carrying the T allele show a reduced fertilization rate and that segregation distortion leads to rarity of the TT genotype in the population. [source]


Effect of storage time and cryoprotectant concentrations on the fertilization rate and hatching rate of cryopreserved sperm in red seabream (Pagrus major Temminck & Schlegel, 1843)

Abstract This study examined the effects of storage time and cryoprotectant concentrations on the post-thaw sperm of red seabream, Pagrus major. Sperm treated with 12%, 15%, 18% and 21% DMSO were cryopreserved for 10, 30, 60 and 360 days, and fertilization and hatching rates were analysed. For all groups, there were no differences in the fertilization rates and hatching rates between sperm cryopreserved for <60 days and fresh sperm (98.8±0.8%, 96.4±1.3%). However, for sperm cryopreserved for 360 days, both fertilization rates (88.6±3.0% to 7.0±1.9%) and hatching rates (79.4±7.2% to 3.3±0.8%) decreased drastically. Furthermore, the cryoprotectant concentrations affected sperm quality significantly (P<0.05). When cryopreserved for 360 days, sperm treated with 15% DMSO obtained the best results compared with other concentrations. We suggest that 15% DMSO may be an effective cryoprotectant for long-term sperm cryopreservation of red seabream. [source]


Vitamin contents of eggs that produce larvae showing a high survival rate in the Japanese eel Anguilla japonica

Abstract This paper describes the relationship between the egg vitamin concentrations and the egg quality in the Japanese eel Anguilla japonica. No notable relation was found between any vitamin and the fertilization rate. Hatching and survival rates of larvae, however, significantly increased with an elevated level of egg vitamin C (VC). In contrast to VC, the relation between vitamins E (VE) and A (VA) concentrations and survival rate showed a clear peak, with a reduced survival rate at both higher and lower vitamin concentrations. The ratio of VE to lipid or highly unsaturated fatty acid (HUFA) in eggs positively correlated with hatching and survival rates of larvae. High-quality (HQ) eggs were determined as eggs that produced larvae having a survival rate higher than 80% at 8 days post hatch, and low-quality (LQ) eggs were determined as eggs that did not hatch. The level of VC of HQ was significantly higher than LQ. The results of this study suggest that HQ eggs, which produce larvae having a high survival rate, must have high levels of VC and VE/HUFA ratio and contain optimum levels of VA and VE in Japanese eel. Influence of water temperature on spawning induction and larval development of the sea squirt Halocynthia ritteri (Oka, 1906)

Abstract In order to establish a large-scale hatchery technique for the sea squirt Halocynthia ritteri, the influences of water temperature on spawning induction, larval development and survival of H. ritteri were studied under laboratory conditions. The larvae of H. ritteri exhibited high sensitivity to changes in temperature. As temperature increased from 10 to 21 °C, the duration of larval stage was inversely related to temperature and the time to reach attached larvae stage decreased from 42.0 to 59.4 h. The biological minimum temperature for the early development of H. ritteri was estimated to be 0.22 °C. In relation to temperature, quadratic equations showed significant fits to the data of spawning rate, fecundity, fertilization, larval survival and attachment. Optimal temperatures for spawning rate, fecundity, fertilization rate, larval survival rate and attachment rate were estimated to be 14.9, 14.7, 13.2, 14.1 and 14.7 °C respectively. It is concluded that the range of 13,15 °C is optimal for spawning and larval rearing of sea squirts. [source]


Fertilization efficiency of cryopreserved sperm from striped catfish, Pangasius hypophthalmus (Sauvage)

Abstract The fertilization efficiency of cryopreserved sperm was compared with fresh sperm from striped catfish, Pangasius hypophthalmus. Of the two sets of experiments carried out, the first compared four sperm doses using fresh sperm and fresh eggs. The second experiment compared six concentrations of cryopreserved sperm ranging from 6.94 × 107 to 6.94 × 1010 to fertilize 100 eggs per batch. Fertilization, hatch and survival rates were compared between cryopreserved and fresh sperm. The highest fertilization rate (53.75±1.62%) was achieved with a sperm dose of 6.94 × 108. Increasing the sperm dose to 3.47 × 109 did not increase the fertilization rate, indicating that the optimum sperm:egg ratio lies between 6.94 × 106 and 3.47 × 107 sperm per egg. Both highest (6.94 × 1010) and the lowest (6.94 × 107) sperm doses resulted in lower fertilization rates (2.04% and 16.90% respectively). No significant differences were found among four fresh sperm doses compared. Mean hatch and survival rates resulting from fresh and cryopreserved sperm were similar. The experiment shows that while only 1.89 × 106 fresh spermatozoa was required to fertilize a fresh egg, 6.94 × 106 (or 3.67 times more) cryopreserved sperm was required to achieve the same level of fertilization. This provides important information for making decision to cryopreserve sperm for commercial and/or conservation purposes. [source]


Early development of the silver catfish Rhamdia quelen (Quoy & Gaimard, 1824) (Pisces:Heptapteridae) from the São Francisco River Basin, Brazil

Abstract The silver catfish, Rhamdia quelen, is endemic to North, Central and South America with high aquaculture potential and wide acceptance in the market. Breeder fish were subjected to induced reproduction through hypophysation using a crude common carp pituitary extract. Egg characteristics, oocyte surface ultrastructure and histology of larval ontogenesis until whole yolk resorption were described for the first time for this species. Oocytes and semen were obtained by manual extrusion, and fertilization was conducted using the dry method. After fertilization, eggs were kept in incubators at 24 °C. The embryonic development was monitored using a stereomicroscope every 10 min until hatching. To analyse the larval development, larvae samples were collected from incubators daily until the fifth day, fixed in Bouin's fluid and subjected to routine histological techniques. The oocyte extrusion occurred 8 h after the second hormone dose at 26 °C. The oocytes were spherical, non-adhesive and yellow, with a diameter of 1471.75±47.63 ,m. Scanning electron microscopy revealed a thin jelly coat covering the zona radiata in the animal pole around the micropyle. The blastopore closure occurred within 8 h after fertilization, and the fertilization rate was 79.9±5.2% at 24 °C. Embryonic development was completed within 25 h 30 min after fertilization. The complete resorption of the yolk and the formation of the digestive system organs and the mouth opening occurred on the fifth day, indicating a need for exogenous feeding. The results of this study provide information important for improvement in R. quelen culture and management. [source]


Increase in milt production by hormonal treatment in the pejerrey fish Odontesthes bonariensis (Valenciennes 1835)

Abstract In spite of interest in the cultivation of the pejerrey fish Odontesthes bonariensis (Cuvier & Valenciennes 1835), there are few studies on subjects required to advance this activity. One of the problems is the synchronization of female and male maturation to provide eggs and sperm for larval production. The low volume of expressible milt, either in wild or culture fish, is a major problem. The aim of this work was to study the effectiveness of the administration of different hormones on sperm production in pejerrey. Milt production was enhanced by the injection of human chorionic gonadotropin (hCG) (16.7-fold increase, 625 IU kg,1), carp pituitary extracts (13.5-fold increase, 30 mg kg,1), salmon pituitary extracts (12.8-fold increase, 30 mg kg,1), salmon-type gonadotropin-releasing hormone analogue (GnRH) (16.7-fold increase, 10 ,g kg,1) and mammalian-type GnRH analogue (10.8-fold increase, 20 ,g kg,1). Sperm concentration, motility and the fertilization rate were not statistically different compared with control groups. It was also demonstrated that sperm could be obtained off-season. Taken together, hCG is recommended to stimulate pejerrey spermiation because it is effective in low doses is inexpensive and is widely available. [source]


Sperm motility and fertilizing ability in the Persian sturgeon Acipenser persicus

Abstract The motility and fertilizing ability of the Persian sturgeon, Acipenser persicus, spermatozoa were investigated. Optimum ionic content (Na+, K+, Ca2+ and Mg2+) and pH of activation solution as well as the optimum dilution rate were determined. The results show optimum motility characteristics of spermatozoa in buffered solutions containing 25, 0.2, 3 and 10 mM L,1 Na+, K+, Ca2+ and Mg2+, respectively, at dilution rate 1:50 and pH 8.0. To test the fertilizing ability of sperm, two buffered saline solutions were used as activation solution of sperm motility. The present study indicated (1) spermatozoa motility is one of key factors that influence on fertilizing ability of sperm, (2) a high fertilizing ability of sperm is obtained after dilution in saline solutions rather than in freshwater and (3) a maximum fertilization rate occurs in buffered saline solution containing 0.2 mM L,1 K+. There is also a good correlation between biochemical characteristics of seminal plasma and fertilizing ability of sperm. [source]


Effect of cryoprotectants, extenders and freezing rates on the fertilization rate of frozen striped catfish, Pangasius hypophthalmus (Sauvage), sperm

Abstract The effects of four cryoprotectants (methanol, MeOH; dimethyl sulphoxide, DMSO; dimethyl acetamide, DMA; and ethylene glycol, EG), three extenders (calcium-free Hanks' balanced salt solution, C-F HBSS, Hanks' balanced salt solution, HBSS and sodium chloride, NaCl) and two different freezing procedures (one- and two-step) on the cryopreservation of striped catfish (Pangasius hypophthalmus (Sauvage)) sperm were investigated. Sperm were frozen using a controlled-rate freezer in 250 ,L straws and stored for 2 weeks in a liquid nitrogen (LN2) container. They were then airthawed at room temperature, and fertilization, motility and viability were assessed. The highest fertilization rate of 41% (81% of control) was achieved with the combination of 12% DMSO and 0.9% NaCl using a one-step freezing procedure (10°C min,1). Also, DMA resulted in a higher fertilization rate (30% or 51% of the control) than MeOH (18% or 38% of the control) or EG (8% or 12% of the control). In addition, the three extenders used did not affect fertilization rates after cryopreservation with each cryoprotectant. There were no significant differences among the three cryoprotectant concentrations and between the one- and two-step freezing procedures. However, fertilization rates of cryopreserved sperm were significantly lower than the controls (P<0.05). The results of this study indicate that high fertilization rates of striped catfish eggs can be achieved using cryopreserved sperm when frozen at 10°C min,1 in DMSO or DMA with either 0.9% NaCl or C-F HBSS. Fertilization, hatching, survival and growth rates in reciprocal crosses of two strains of an African catfish Heterobranchus longifilis Valenciennes 1840 under controlled hatchery conditions

A diallelic cross between two strains [Layo strain (LS) and Noun strain (NS)] of the catfish Heterobranchus longifilis Valenciennes was carried out under controlled hatchery conditions to estimate their reproductive performance and aquaculture potential in terms of fertilizability, hatchability, survival, growth and heterosis. The average fertilization rate of all mating groups was as high as 90.4%, the fertilization rate of the purebred NS (95.2%) being significantly higher than that of the purebred LS or the reciprocal crosses (P <,0.05). The average hatching rate of all genetic groups was as high as 84.7%, the hatching rate of NS (89.4%) being significantly higher than that of the purebred LS or the reciprocal crosses (P <,0.05). The mean survival rate of all crosses from hatching up to the onset of exogenous feeding stood at 94.2%, without showing a significant difference (P >,0.05) between the crosses. During the larval rearing period, which extended from the onset of exogenous feeding up to 15 days of age, NS displayed a significantly lower growth rate (P <,0.001) than that of the purebred LS and their reciprocal hybrids. There was no significant difference (P >,0.05) in survival rate (mean 84.7%) between the four crosses at the end of the larval rearing period. During the juvenile rearing phase, the mean growth performance of all crossbreds was similar to that of the purebred LS and significantly different from that of the purebred NS (P <,0.001). The final individual weights attained by LS, NS, LS × NS and NS × LS were 3.31, 0.71, 3.97 and 3.66 g respectively. The increase in weight attained by the fast-growing crossbred LS × NS was 562.2%, 20.4% and 8.7% more than that of NS, LS and NS × LS respectively. The survival rate of NS (57.1%) was significantly lower (P <,0.001) than that of all the rest of the crosses. The crossbreds displayed about 15.1% heterosis in mean body weight relative to the fast-growing purebred LS. It was concluded that cross-breeding of H. longifilis strains could be advantageous because of the hybrid vigour in the progeny. [source]


Reproductive isolating barriers between colour-differentiated populations of an African annual killifish, Nothobranchius korthausae (Cyprinodontiformes)

Allopatric populations separated by vicariance events are expected to evolve reproductive isolating mechanisms as a result of disparate selection pressures and genetic drift. The appearance of reproductive isolating mechanisms may vary across taxa with differences in the opportunity for mate choice, and may be asymmetrical. In addition, premating barriers may be affected by individual mating experience. We used choice and no-choice experiments to investigate reproductive isolation between two allopatric (island and mainland) and colour-differentiated populations of an African annual fish, Nothobranchius korthausae. Assortative mating under experimental conditions was limited and asymmetrical. Preference for sympatric males was only expressed in nonvirgin females from one population. Virgin fish from both populations mated indiscriminately. No difference in the number of eggs laid, fertilization rate and hatching success was detected in no-choice experiments. All mating combinations produced viable offspring and no postmating barriers were detected in terms of the performance and fertility of F1 hybrids. Optimization of fertilization success in Sander vitreus is influenced by the sperm : egg ratio and ova storage

Walleye Sander vitreum eggs could be stored at 7° C for up to 30 h with 60% fertilization rates. Fertilization rates at 1 and 12° C fell to 17 and 5%, respectively. At spermatozoa : egg ratios <25 000 : 1, survival to the eyed stage declined precipitously. Beyond a ratio of 25 000 : 1, survival to the eyed stage did not increase >c. , 70%. Therefore, a standard spermatozoa : egg ratio of 25 000 : 1 was established and is recommended for studies concerning fertilizing success. [source]
