Selected AbstractsEffect of mercury and Gpi-2 genotype on standard metabolic rate of eastern mosquitofish (Gambusia holbrooki),ENVIRONMENTAL TOXICOLOGY & CHEMISTRY, Issue 4 2001Christopher Paul Tatara Abstract Previous studies demonstrated differential mortality among mosquitofish of different Gpi-2 genotypes during acute mercury and arsenate exposures. Mercury-exposed mosquitofish also had Gpi-2 genotype-specific differences in glycolytic and Krebs cycle metabolite pools. The mortality and metabolite data suggested that mosquitofish bearing specific Gpi-2 genotypes might differ in metabolic efficiency, with less efficient Gpi-2 genotypes having higher standard metabolic rates (SMRs) and shorter times to death during acute mercury exposure. Effect of Gpi-2 genotype on SMR was assessed with a factorial arrangement of six Gpi-2 genotypes and two exposure sequences (Control , Control; Control , 100 ,g/L Hg). The SMRs were estimated by measuring oxygen consumption using an indirect, closed-circuit, computer-controlled respirometer. A 48-h exposure to 100 ,g/L of mercury resulted in a 16.7% elevation of SMR above control levels (p = 0.001). The Gpi-2 genotype and the number of heterozygous loci per individual had no significant effect on SMR in mercury-exposed mosquitofish. The experimental results do not support the hypothesis that Gpi-2 genotype-specific differences in glycolytic and Krebs cycle metabolite pools and mortality in mosquitofish exposed to mercury are associated with differences in SMR. [source] In vitro gas production profile and the formation of end products from non- washable, insoluble washable and soluble washable fractions in some concentrate ingredientsJOURNAL OF THE SCIENCE OF FOOD AND AGRICULTURE, Issue 7 2007Arash Azarfar Abstract A procedure that mimics washing in the in situ incubation technique, combined with an in vitro gas and volatile fatty acids (VFAs) production technique, was used to verify the assumption that rumen degradation behaviour of material washed out of nylon bags is instantaneous and complete. In a 6 × 4 factorial arrangement of treatments with three replicates, fractions of maize, barley, milo, yellow peas, lupins (a mixture of white and spotted lupins) and round-seeded brown faba beans were subjected to an in vitro incubation technique. Fractions were whole (WHO), non-washable (NWF), insoluble washable (ISWF) and soluble washable (SWF). In a manually operated in vitro fermentation system, another 24 samples of the same substrates were fermented for VFA and ammonia analysis. Except in lupins, ISWF in the concentrate ingredients was very rich in starch. SWF was relatively rich in ash, crude protein, soluble sugars, and a residual unknown fraction but contained only a negligible quantity of starch. Thus, the fermentation characteristics of ISWF were more like WHO and NWF than SWF. Total gas production of SWF was considerably lower than the other fractions. A very rapidly degradable fraction was seen in the first phase of degradation of SWF. The pattern of fermentation end-product formation for SWF differed from that of the other fractions. Copyright © 2007 Society of Chemical Industry [source] The effects of dietary organic or inorganic selenium in rainbow trout (Oncorhynchus mykiss) under crowding conditionsAQUACULTURE NUTRITION, Issue 6 2009F.Z. KÜÇÜKBAY Abstract In the present study, the effects of different sources of selenium (Se; sodium selenite or selenomethionine) supplementation on the growth and serum concentrations of oxidative stress markers [malondialdehyde (MDA), 8-isoprostane, glutathione peroxidase (GSH-Px) activity] and muscle Se, MDA and heat shock protein 70 (Hsp70) levels in rainbow trouts were evaluated. The fish (n = 360; 0 + years old) with initial average weight of 20 ± 0.8 g were randomly assigned to 12 treatment groups consisting of 3 replicates of 10 fish each in a 2 × 2 × 3 factorial arrangement of treatments (stocking densities, Se sources, Se levels). The fish were kept at low (25 kg m,3) or high (100 kg m,3) stocking densities and fed a basal (control) diet or the basal diet supplemented with either 0.15 or 0.30 mg of Se kg,1 of diet from two different forms: sodium selenite or selenomethionine. High stocking density decreased weight gain, feed intake and feed conversion ratio (FCR) when basal diet was fed (P = 0.001). A linear increase in feed intake and weight gain and improvement in FCR were found in sodium selenite (P = 0.01)- or selenomethionine (P = 0.001)-supplemented fish reared under crowding conditions. Serum and muscle Se levels and serum GSH-Px activity increased (P = 0.001) linearly, whereas serum and muscle MDA concentrations and serum 8-isoprostane decreased linearly as dietary sodium selenite (P = 0.01) or selenomethionine (P = 0.001) supplementation increased. Selenomethionine and sodium selenite supplementation decreased Hsp70 in the muscle of fish reared under crowding conditions (P < 0.05). Supplementation with Se improved growth and antioxidant status of fish and the effects of selenomethionine were relatively greater than sodium selenite in the crowded groups. Results suggest that crowding conditions cause significant detrimental effects in rainbow trout indicated by increased oxidative stress, reduced feed intake and body weight gain. ,t also indicates that dietary Se supplementation offers a feasible way of reducing the losses in performance of rainbow trout reared under crowding conditions. Selenomethionine seems to be more effective than sodium selenite and the higer dose in the present study also seems to be more effective than the lower dose. BORBA Abstract Ten isonitrogenous casein,gelatin-based diets were formulated to contain five estimated metabolizable energy concentrations (10.92, 12.29, 13.63, 14.82 and 16.16 kJ g,1) at two carbohydrate-to-lipid ratios (CHO : L, 5.3 and 12.8, g : g) in a 5 × 2 factorial arrangement. Each diet was assigned to triplicate groups of 11 piracanjuba fingerlings (5.25 ± 0.14 g) and fed to apparent satiation twice a day for 90 days. Higher daily weight gain was obtained by fish fed the 13.63 kJ g,1 diets for both CHO : L ratios. There was a significant reduction of feed consumption when dietary energy concentration increased above 13.63 kJ g,1. Feed conversion ratio and apparent net energy retention improved as dietary energy increased. Apparent net protein retention tended to be lower in the highest and lowest dietary energy concentrations. The results suggest that dietary lipid energy was more efficiently utilized by piracanjuba fingerlings than carbohydrate energy. Body composition and hepatosomatic index (HSI) were not influenced by dietary CHO : L ratio. However, an increase in dietary energy concentration beyond 13.63 kJ g,1 resulted in a significant increment in lipid deposition, while body moisture and HSI decreased. Our findings indicate that at 300 g kg,1 dietary crude protein, a CHO : L ratio of 5.3 is recommended for piracanjuba, and the required energy is either 13.63 kJ g,1 if raised for aquaculture or 14.82 kJ g,1 if destined to stock enhancement. [source] Threadfin shad impacts phytoplankton and zooplankton community structures in channel catfish pondsAQUACULTURE RESEARCH, Issue 10 2010Bartholomew W Green Abstract Plankton community structure and chlorophyll a concentration were compared in twelve 0.1 ha earthen ponds co-stocked with channel catfish (Ictalurus punctatus Rafinesque, 1818) in a multiple-batch culture (initial biomass=5458 kg ha,1) and a planktivore, threadfin shad (Dorosoma petenense Güther, 1867; initial biomass=449 kg ha,1), during the April,November growing season. We used a completely randomized design in a 2 × 2 factorial arrangement to test the planktivore level (presence or absence of threadfin shad) and channel catfish feeding frequency (daily or every third day). Channel catfish were fed a 32% protein feed to apparent satiation on days fed. The presence of threadfin shad affected phytoplankton and zooplankton community structure more than did feeding frequency, and the impact in ponds was more pronounced after 1 July. The numbers of all major groups of zooplankton were lower in ponds with threadfin shad, but were unaffected by the feeding frequency. Chlorophyll a concentration before 1 July was higher in ponds with threadfin shad and unaffected by the feeding frequency, whereas after 1 July it was higher in ponds without threadfin shad and that were fed daily. Phytoplankton community structure after 1 July was dominated by nuisance algal bloom genera of cyanobacteria in ponds without threadfin shad and by Bacillariophyceae in ponds with threadfin shad. [source] Manganese alters mitochodrial integrity in the hearts of swine marginally deficient in magnesium,BIOFACTORS, Issue 2 2004Kevin B. Miller Abstract It was previously reported that pigs marginally deficient in magnesium (Mg) and fed diets high in manganese (Mn) died suddenly with signs of sudden cardiac death. Manganese, which has properties similar to Mg, may exacerbate Mg-deficiency and be accumulated by mitochondria resulting in ultrastructural damage. The objective of this study was to determine whether deaths of the type previously observed were mediated by adverse interactions of Mn and Mg resulting in ultrastructural damage to the myocardium, alterations in electrocardiographic recordings and tissue retention of Mn, Mg and calcium (Ca). Forty-eight pigs were fed one of six diets in a 2 × 3 factorial arrangement of Mg (100 or 1000 mg Mg/kg) and Mn (5, 50 or 500 mg Mn/kg) for 8 weeks. Left ventricle muscle samples were collected for examination by transmission electron microscopy. No differences in heart muscle ultrastructure were observed between pigs fed low and adequate dietary Mg. However, marked myocardial necrosis and mitochondrial swelling were observed in pigs fed high dietary Mn when combined with low Mg. Feeding low dietary Mg elevated minimum (P < 0.01), maximum (P < 0.05) and average (P < 0.001) heart rates. Low dietary Mg resulted in a 55% probability of a ventricular beat being recorded (P = 0.05) and lower Mg (P < 0.02) and Ca (P < 0.04) contents in heart atria and ventricles. These results suggest that high Mn, when fed in combination with low Mg, disrupts mitochondrial ultrastructure and is associated with the sudden deaths previously reported. [source] |
