Exposure of three generations of the estuarine sheepshead minnow (Cyprinodon variegatus) to the androgen, 17,-trenbolone: Effects on survival, development, and reproduction

Abstract Estimating long-term effects of endocrine-disrupting chemicals on a species is important to assessing the overall risk to the populations. The present study reports the results of a 42-week exposure of estuarine sheepshead minnows (Cyprinodon variegatus) to the androgen, 17,-trenbolone (Tb) conducted to determine if partial-(F0) or single-generation (F1) fish exposures identify multigenerational (F0,F3) effects of androgens on fish. Adult F0 fish were exposed to 0.007, 0.027, 0.13, 0.87,and 4.1,µg Tb/L, the F1 generation to ,0.87,µg Tb/L, the F2 fish to ,0.13,µg Tb/L, and the F3 fish to ,0.027,µg Tb/L. The highest concentrations with reproducing populations at the end of the F0, F1, and F2 generations were 4.1, 0.87, and 0.027,µg Tb/L, respectively. Reproduction in the F0, F1, and F2 generations was significantly reduced at 0.87, 0.027, and 0.027,µg Tb/L, respectively. Fish were significantly masculinized in the F1 generation exposed to 0.13 µg Tb/L or greater. Female plasma vitellogenin was significantly reduced in F0 fish exposed to ,0.87,µg Tb/L. Gonadosomatic indices of the F0 and F1 generations were significantly increased at 0.87 and 0.13 µg Tb/L in the F0 and F1 generation, respectively, and were accompanied by ovarian histological changes. Reproduction was the most consistently sensitive measure of androgen effects and, after a life-cycle exposure, the daily reproductive rate predicted concentrations affecting successive generations. The present study provides evidence that a multiple generation exposure of fish to some endocrine-disrupting chemicals can result in developmental and reproductive changes that have a much greater impact on the success of a species than was indicated from shorter term exposures. A molecular approach to detect hybridisation between crucian carp (Carassius carassius) and non-indigenous carp species (Carassius spp. and Cyprinus carpio)

Summary 1. Releases of non-native fish into the wild is an increasing problem posing considerable ecological and genetic threats through direct competition and hybridisation. 2. We employed six microsatellite markers to identify first generation hybrids and backcrosses between native crucian carp (Carassius carassius) and introduced goldfish (C. auratus) and common carp (Cyprinus carpio) in the U.K. We also investigated the genetic characteristics of the taxonomically controversial gibel carp (Carassius spp.) from sites across Europe. 3. Natural hybridisation between goldfish and crucian carp occurs frequently, although hybrids between all other species pairs were observed. Only 62% of British crucian carp populations (n = 21) consisted exclusively of pure crucian carp. In some populations hybrids were so frequent, that no pure crucian carp were caught, indicating a high competitive ability of hybrids. 4. Most hybrids belonged to the F1 generation but backcrossing was evident at a low frequency in goldfish × crucian carp hybrids and goldfish × common carp hybrids. Furthermore, some local populations had high frequencies of backcrosses, raising the opportunity for introgression. 5. Gibel carp from Germany and Italy belonged to two triploid clonal lineages that were genetically closely related to goldfish, whereas all individuals identified from British populations proved to be crucian carp × goldfish hybrids. 6. Our study suggests that the release of closely related exotic cyprinids not only poses a threat to the genetic integrity and associated local adaptations of native species, but may also contribute to shifts in community structure through competitive interactions. Sexual reproduction of scleractinian corals in public aquariums: current status and future perspectives

A multiple-choice questionnaire was distributed, mainly via the list servers of the EUAC (European Union of Aquarium Curators) Coral ASP (Animal Sustainability Program) and AquaticInfo, to evaluate the potential of today's aquariums for the captive breeding of scleractinian corals. Sixteen (including the temperate coral Astroides calycularis) of, in total, 24 species (nine families) were recorded as showing reproductive behaviour that could establish an F1 generation. Broadcast spawners (13 species) reproduced mainly in open systems under natural light conditions (in all cases natural moonlight exposure), whereas brooders (11 species) showed less sensitivity towards certain environmental factors known to trigger reproduction in field populations (here moonlight and temperature fluctuations). Except for a few recruits of Galaxea fascicularis and Echinopora lamellosa maintained in a 750 000 litre system, recruits of broadcast spawners could be exclusively obtained by manipulating fertilization and settlement. Brooding corals established generally less than 100 recruits if settlement was not enhanced experimentally. When reproduction was manipulated, it enhanced reproductive success, in most cases to above 100 recruits. We assume that more species, especially brooders, might reproduce in public aquariums without being noticed by the staff owing to the lack of recruitment and of experimental design (larval collection). This study illustrates the great potential for public aquariums to reproduce corals sexually. However, more investigation is necessary to optimize reproductive success and possibly to broaden the spectrum of species reproduced in public aquariums. [source]


How two different host species influence the performance of a gregarious parasitoid: host size is not equal to host quality

Summary 1Hyssopus pallidus Askew (Hymenoptera, Eulophidae) is a gregarious ectoparasitoid of the two tortricid moths species Cydia molesta Busck and C. pomonella L. (Lepidoptera, Tortricidae). It paralyses and parasitizes different larval instars of both species inside the apple fruit, which leads to the death of the caterpillar. 2We assessed the influence of host species characteristics and host food on the performance of the parasitoid female in terms of clutch size decisions and fitness of the F1 generation. 3A comparison of clutch size revealed that female parasitoids deposited similar numbers of eggs on the comparatively smaller C. molesta hosts as on the larger C. pomonella hosts. The number of parasitoid offspring produced per weight unit of host larva was significantly higher in C. molesta than in C. pomonella, which is contrary to the general prediction that smaller hosts yield less parasitoid offspring. However, the sex ratio was not influenced by host species that differed considerably in size. 4Despite the fact that less host resources were available per parasitoid larva feeding on C. molesta caterpillars, the mean weight of emerging female wasps was higher in the parasitoids reared on C. molesta. Furthermore, longevity of these female wasps was neither influenced by host species nor by the food their host had consumed. In addition we did not find a positive relationship between adult female weight and longevity. 5Parasitoid females proved to be able to assess accurately the nutritional quality of an encountered host and adjust clutch size accordingly. These findings indicate that host size is not equal to host quality. Thus host size is not the only parameter to explain the nutritional quality of a given host and to predict fitness gain in the subsequent generation. [source]


Geographic variation in diapause induction and mode of diapause inheritance in Tetranychus pueraricola

Abstract:, Diapause induction and photoperiodic response curves were determined for 33 strains of Tetranychus pueraricola derived from kudzu vine at three constant temperatures (15, 18 and 20°C) under a short-day condition (10 : 14 h; light : dark). Females of all but one of the strains entered diapause at all three temperatures with little variation in diapause percentages among the strains. The exception was the southernmost strain, which was found to be a non-diapause (ND) strain. The critical photoperiod gradually decreased towards the south at a rate of about 1 h for each 5 degrees of latitude. The diapause strains (D1 and D2) exhibited 100% diapause, whereas the ND strain exhibited 0% diapause. By crossing these strains, we determined that ,non-diapause' was a dominant character over ,diapause' and the character was controlled by simple Mendelian inheritance. To clarify why the female progeny from the crosses between the D1 and ND strains did not segregate into the diapause and non-diapause phenotypes in a 1:1 ratio in the B1 generation, round-robin crosses were carried out among the three strains. The results showed that the F1 generation was reproductively compatible and showed high egg hatchability with a female-biased sex ratio. In the B1 generation, the crosses between the D1 and ND strains and between the D1 and D2 strains exhibited extremely low egg hatchability and produced mostly female progeny, whereas offspring from the crosses between the D2 and ND strains showed more than 50% hatchability for B1 eggs and a female-biased sex ratio. Thus, the absence of segregation observed in the crosses between the D1 and ND strains appears to be due to the severe hybrid breakdown that occurred in the B1 generation. [source]


Permethrin resistance ratios compared by two methods of testing nymphs of the German cockroach, Blattella germanica

Summary For the German cockroach, Blattella germanica L. (Dictyoptera: Blattellidae), the permethrin resistance ratio (RR) was assessed by topical application and by tarsal contact tests, using first-instar nymphs of five strains from Tehran, Iran. Each test was replicated three or four times with 10 nymphs aged 2,3 days; mortality was scored 24 h post-treatment. The reference susceptible strain showed LD50 permethrin 0.0175 ,l/nymph from topical application, KT50 of 8.41 min and LT50 of 12.82 following tarsal contact with permethrin 15 mg/m2. In four wild strains (F1 generation) the RR varied from 4.14 to 4.7 for mortality after topical application, from 4.2 to 6.45 for mortality and 17,27 for knockdown following tarsal contact tests. Hence, overall knockdown results gave much higher RRs than for mortality data. Resistance ratios based on both methods of treatment were very similar: one strain showed a slightly higher value by topical application (RR 4.6 vs. 4.2, i.e. 1.1-fold difference) whereas the other three strains gave slightly greater RR (1.2,1.4 fold) by tarsal contact. Resistance was abolished by cotreatment with the synergist piperonyl butoxide plus permethrin (ratio 3 : 1 required for full efficacy), indicating that mixed-function oxidases were inhibited as a major metabolic pathway in all four resistant strains. Genetic and hormonal control of melanization in reddish,brown and albino mutants in the desert locust Schistocerca gregaria

The genetic and hormonal control of body colouration is investigated using two recessive genetic mutant strains, the reddish,brown (RB) mutant and an albino mutant, as well as a normal (pigmented) strain of the desert locust Schistocerca gregaria. The colour patterns of the RB nymphs are similar to those of a normal strain, although the intensity of the melanization is weaker in the former. Reciprocal crosses between the RB and albino mutants produce only normal phenotypes in the F1 generation. In the F2 generation, the normal, RB and albino phenotypes appear in a ratio of 9 : 3 : 4, indicating that two Mendelian units might determine the appearance of dark body colour and the intensity of melanization, respectively. In other words, at least two steps of regulation might be involved in the expression of body colour. Injections of [His7]-corazonin, a neuropeptide inducing dark colour in this locust, fail to induce dark colour in albino nymphs but show a dose-dependent darkening in RB nymphs in the range, 10 pmol to 1 nmol. Some RB nymphs become indistinguishable from normal individuals after injection of the peptide. Implantation of corpora cardiaca (CC) taken from RB mutants into other RB individuals induces darkening in the latter and CC from RB, albino and normal strains have similar dark colour-inducing activity when implanted into albino Locusta migratoria. These results suggest the possibility that the RB mutant gene regulates the intensity of melanization, possibly through controlling the pathway of pigment biosynthesis associated with [His7]-corazonin. Local DNA features affect RNA-directed transcriptional gene silencing and DNA methylation

Summary Transcription of a nopaline synthase promoter (pNOS) inverted repeat provides an RNA signal that can trigger transcriptional gene silencing and methylation of pNOS promoters in trans. The degree of silencing is influenced by the local DNA features close to the target promoter integration sites. Among 26 transgenic Arabidopsis thaliana lines harbouring single copies of a T-DNA including a pNOS- NPTII reporter gene at different chromosomal loci, NPTII RNA levels showed limited variation. When challenged by the silencer transgene providing the pNOS RNA signal, reduction of the NPTII RNA levels in the F1 generation varied by more than 100-fold, ranging from no reduction to reduction to <1% of the non-silenced level. Silencing was generally correlated with proportional DNA methylation in the pNOS region, except for one target transgene showing substantial DNA methylation without adequate silencing. Silencing was progressive through generations. Differences in the degree of silencing among the target transgenes were transmitted at least to the F3 generation, and seemed to be influenced by transgene-flanking sequences. Apparently, close-by repeats promoted, whereas close-by functional genes diminished, the response to the silencing signal. [source]


Grey plumage colouration in the duck is genetically determined by the alleles on two different, interacting loci

Summary Based on the observation of a grey phenotype in the F1 generation from a cross between two white plumage duck varieties, the white Kaiya and the white Liancheng, we hypothesized a possible interaction between two autosomal loci that determine grey plumage. Using the parental and F1 individuals, seven testing combinations including five different F1 intercrosses (F2) and two different backcrosses (BC1 and BC2) were designed to test our hypothesis. It was demonstrated by chi-squared analysis that six test matings produced offspring in the expected ratios between the grey and white, with P- values ranging from 0.50 to 0.99. Another mating, where all white offspring were expected, produced 33 white individuals. These results verified that the interaction between two loci produced the grey phenotype. The C locus, which carries the recessive allele (c), was previously thought to be the only gene responsible for white plumage in the duck. This is the first report that an allele (t), carried by the white Liancheng at a different autosomal locus, also determines white plumage in ducks. Furthermore, the dominant alleles at both loci can interact with each other to produce the grey phenotype, and a new dark phenotype, observed in some F2 individuals, can be attributed to the dosage effect of the T allele. Natural hybridization between Vanilla claviculata (W.Wright) Sw. and V. barbellata Rchb.f. (Orchidaceae): genetic, morphological, and pollination experimental data

Vanilla claviculata (W.Wright) Sw. and K barbellata Rchb.f. grow sympatrically in the southwestern part of herto Rico. At three localities in the range of the two species, individuals with flowers that appeared Merent from previously known species were found. To test whether these were hybrids between V. claviculata and V. barbellata, 11 morphological floral characters were recorded at seven populations and allozyme profiles of the three taxa compared. Principal component analysis on the morphological characters gave three distinct groups with minor overlaps. The first component axis reflects variation in overall size of the flowers, while the second component reflects variation in the size of the distal aperture of the labellum. Allozyme data revealed that the putative hybrids have a significant surplus of heterozygotes (FIS= - 0.65) as expected in a F1 generation. Both data sets support the hypothesis that the individuals are hybrids, which is further augmented by interspecific pollination experiments between the putative parental species. This is the first case of natural hybridization reported in the genus Vanilla. [source]


Evidence that direct DNA uptake through cut shoots leads to genetic transformation of Solanum aviculare Forst

Abstract The reporter genes GUS, NPTII and BAR, either separately or in combination, have been exploited to determine if DNA which can directly enter plants, circulate within the plant and enter nuclei, can also integrate into the genome in a manner which will permit gene expression. Feeding of either seed-derived or adventitious cut shoots of Solanum aviculare with the GUS gene followed by rooting of the shoots and growing on, resulted in all tissues of the plant showing GUS activity as detected cytochemically. Southern blot analysis of plants derived from the adventitious shoots confirmed the presence of the reporter gene in roots. Reporter gene expression was observed also in the F1 generation. If GUS and NPTII or GUS, NPTII and BAR were fed together, then in each case it was possible to have both expression and Southern blot confirmation of each of the genes. Characterizing laboratory colonies of western corn rootworm (Coleoptera: Chrysomelidae) selected for survival on maize containing event DAS-59122-7

Abstract Event DAS-59122-7 is a novel transgenic trait designed to protect the roots and yield potential of maize from the insect pest corn rootworm Diabrotica spp. (Col.: Chrysomelidae). The increased pest status of corn rootworm, exceptional efficacy of this trait, and anticipated increases in farm efficiency and grower and environmental safety will drive adoption of this trait. Strong grower acceptance of this trait highlights the importance of science-based and practical resistance management strategies. A non-diapause trait was introgressed into two laboratory colonies of Diabrotica virgifera virgifera collected from geographically distinct locations: Rochelle, IL and York, NE. Both colonies were divided and each reared on maize containing event DAS-59122-7 or its near isoline. Selected and unselected colonies were evaluated for phenotypic change in larval development, injury potential and survival to adulthood during 10 and 11 generations. The F1 generation of both selected colonies displayed increased larval development, survivorship and measurable, but economically insignificant increases in injury potential on DAS-59122-7 maize. Survival rates of 0.4 and 1.3% in F1 generations of both selected colonies corroborate field estimates of survival on DAS-59122-7 maize. Over later generations, total phenotypic variation declined gradually and irregularly. Despite the absence of random mating, the tolerance trait could not be fixed in either population after 10 or 11 generations of selection. An allele conferring major resistance to DAS-59122-7 was not identified in either selected colony. The assessment also concluded that major resistance gene(s) are rare in populations of D. v. virgifera in the United States, and that a minor trait(s) conferring a low level of survival on DAS-59122-7 maize was present. The tolerance trait identified in this study was considered minor with respect to its impact on DAS-59122-7 maize efficacy, and the role this trait may play in total effective refuge for major resistance genes with recessive inheritance is the basis of future work.
