Knowledge-Based Approach towards Hydrolytic Degradation of Polymer-Based Biomaterials

Abstract The concept of hydrolytically degradable biomaterials was developed to enable the design of temporary implants that substitute or fulfill a certain function as long as required to support (wound) healing processes or to control the release of drugs. Examples are surgical implants, e.g., sutures, or implantable drug depots for treatment of cancer. In both cases degradability can help to avoid a second surgical procedure for explanation. Although degradable surgical sutures are established in the clinical practice for more than 30 years, still more than 40% of surgical sutures applied in clinics today are nondegradable.1 A major limitation of the established degradable suture materials is the fact that their degradation behavior cannot reliably be predicted by applying existing experimental methodologies. Similar concerns also apply to other degradable implants. Therefore, a knowledge-based approach is clearly needed to overcome the described problems and to enable the tailored design of biodegradable polymer materials. In this Progress Report we describe two methods (as examples for tools for this fundamental approach): molecular modeling combining atomistic bulk interface models with quantum chemical studies and experimental investigations of macromolecule degradation in monolayers on Langmuir,Blodgett (LB) troughs. Finally, an outlook on related future research strategies is provided. [source]


Antifungal susceptibility testing by flow cytometry: is it the future?

Summary The current increase in the number and significance of fungal infections, the expanding armamentarium of antifungal agents, and the emergence of the problem of antifungal drug resistance have been intensifying the importance of antifungal susceptibility testing (AST). The Clinical and Laboratory Standards Institute (CLSI, formerly NCCLS) in the United States and the Antifungal Susceptibility Testing Subcommittee of the European Committee on Antimicrobial Susceptibility Testing (AFST-EUCAST) published standard methodologies in order to achieve higher reproducibility and allow direct inter-laboratory comparison of the susceptibility results. Nevertheless, several problems remain unresolved and the methods depend on long incubation periods of a minimum of 24 h (EUCAST) or even 48 h (CLSI). Over the last 15 years, successful applications of flow cytometric techniques to AST of both yeast and moulds have been reported. These techniques are based on the analysis of a great number of fungal cells individually and frequently rely on short incubation times of no more than a few hours. Considering these attributes, flow cytometry (FC) seems to have the potential to achieve clinical usefulness in the near future. The collection of data on the reproducibility of the results and on the correlation with clinical outcomes has barely started, however. Practical validation of the experimental methodologies is not granted before a significant amount of data addressing those questions is available. [source]


Photosynthetic Eukaryotes of Freshwater Wetland Biofilms: Adaptations and Structural Characteristics of the Extracellular Matrix in the Green Alga, Cosmarium reniforme (Zygnematophyceae, Streptophyta)

ABSTRACT. Cosmarium reniforme (Zygnematophyceae, Streptophyta) is a green alga that is commonly found in biofilms of wetlands of the Adirondack region, NY (USA). Two distinctive characteristics that are critical to this alga's survival in a benthic biofilm are its elaborate cell morphology and extracellular matrix (ECM). In this study, ultrastructural, immunocytochemical, and experimental methodologies were employed in order to elucidate the cellular characteristics that are critical for survival in a biofilm. The ECM consists of a thick, outwardly lobed cell wall (CW), which contains a patterned network of structurally complex pores. Each pore consists of a narrow channel, terminating internally at a bulb that invaginates localized regions of the plasma membrane. The outer region of the pore contains arabinogalactan protein-like and extensin epitopes that are likely involved in adhesion mechanisms of the cell. External to the CW is the extracellular polymeric substance that is employed in ensheathment of the cell to the substrate and in gliding motility. The architectural design/biochemical make-up of the CW and a secretory system that encompasses the coordinated activities of the endomembrane and cytomotile/cytoskeletal systems provide the organism with effective mechanisms to support life within the biofilm complex. Current directions at the juncture of clinical and cognitive science: A commentary on the special issue

The 12 experimental papers presented in this special issue brilliantly illustrate the creative and exciting ways in which the exchange of theoretical concepts and experimental methodologies, between cognitive and clinical science, is serving both to deepen our understanding of psychopathology, and to identify new ways of potentially remediating such dysfunction. These papers are not only individually excellent, but collectively they communicate some of the major themes that are presently driving new developments within this rapidly evolving field. In this brief commentary, I reflect upon the contributions made by the current studies to these important themes. I also share some suggestions concerning how future work might profitably build upon this present research, to further enhance theoretical progress and enrich therapeutic innovation. The use of muscle protein for egg production in the Zebra Finch Taeniopygia guttata

Pectoral muscle can be an important source of protein for birds. During egg formation Zebra Finches Taeniopygia guttata are able to compensate for nutritional inadequacies in their diet by utilization of the protein in their flight muscles. This analysis of flight muscle sarcoplasm supported earlier observations of protein depletion during egg production. However, SDS gel electrophoresis of the sarcoplasm produced no evidence to support a previous suggestion of the existence of a high molecular weight storage protein, and it is thought that the original observation may have arisen as an artefact of experimental methodology. During laying, protein removal from the sarcoplasm occurred over a range of different proteins and was not confined to any one specific protein band. Additionally, the protein band most reduced over the course of laying did not contain elevated levels of the amino acids most limiting to egg production. These results indicate that during laying, flight muscle sarcoplasm contributes towards the nutrient requirements of egg production from general protein reserves, rather than from a specific storage protein containing elevated levels of limiting amino acids. Dynamic spatial cognition: Components, functions, and modifiability of body schema1

Abstract:, There has been substantial progress towards the understanding of the classical notion of "body schema," with recent advances in experimental methodology and techniques. Mental rotation of the hands can be used as a tool to investigate body schema. Research has shown that implicit motor imagery (i.e., mental simulated movements) can be generated based on the body schema, by combining both stored and incoming sensory information. Multimodal stimulation of peripersonal space has also served as an experimental paradigm for the study of body schema. Perception of peripersonal space is based on body-part-centered space coding that is considered as a manifestation of the body schema, its function being to integrate visual, tactile, and proprioceptive information, and perhaps motor plans as well. By combining such experimental paradigms with neuroimaging and neurophysiological techniques, research has converged to show that the parietal association cortex and premotor cortex are important for the body schema. Multimodal perception of body parts and peripersonal space have been also studied in relation to prism adaptation and tool use effects, indicating a clear modifiability of the body schema. Following prolonged adaptation to reversed vision, a reversed hand representation can be added to the body schema like a tool. The stored component of the body schema may not be established well in young children. But once established it may not be deleted even after an arm is amputated, although it may be weakened. All of these findings help to specify properties of the body schema, its components, functions, and modifiabilities. [source]


Selection of glycoprotein IIb/IIIa inhibitors for upstream use in patients with diabetes experiencing unstable angina or non-ST segment elevation myocardial infarction.

What have we learned in the last 10 years?
Summary Coronary disease accounts for the majority of deaths among patients with diabetes and the thrombotic milieu accelerated by diabetes results in unstable angina (UA), non-ST segment elevation myocardial infarction (NSTEMI) or ST-segment elevation myocardial infarction (STEMI) or death. Upstream use of a glycoprotein IIb/IIIa (GP IIb/IIIa) inhibitor with percutaneous coronary intervention (PCI) as part of an early invasive approach is preferred. However substantial numbers of patients present to rural or non-teaching hospitals without immediate access to a catheterization laboratory. Enhanced GP IIb/IIIa receptor mobilization, TXA2 production and platelet activation together present an extensive thrombotic challenge that may not be overcome with current doses of GP IIb/IIIa inhibitors when used without PCI. Heterogeneity of platelet aggregometric analysis may have identified GP IIb/IIIa doses used in clinical trials that may not fully overcome the thrombotic challenge in patients with diabetes. GUSTO-IV ACS failed to demonstrate a difference in mortality when used without PCI. The PURSUIT trial provided evidence that eptifibatide decreases death or non-fatal myocardial infarction (MI) in the main group and in the diabetic subgroup. Reductions in this primary endpoint were driven by the reduction in non-fatal MI. The PRISM and PRISM-PLUS trials demonstrated a reduction in death, MI or refractory ischaemia at 48 h or 7 days in the main cohort but not specifically in patients with diabetes. Data supporting use of GP IIb/IIIa inhibitors are inconsistent, raising the question of whether these agents should be used at all without PCI. Variability in experimental methodology of platelet aggregometry and selection of anticoagulant used during dose finding studies may have generated doses that are insufficient to overcome the thrombotic burden. A new marker of active inflammation, sCD40L is found to be upregulated at subtherapeutic doses of GP IIb/IIIa inhibitors, suggesting that rebound inflammatory processes may partially account for absence of clear evidence of benefit with some GP IIb/IIIa inhibitors in patients with diabetes experiencing UA/NSTEMI. Patterns of Ethanol Intake in Preadolescent, Adolescent, and Adult Wistar Rats Under Acquisition, Maintenance, and Relapse-Like Conditions

Background:, Animal behavioral models of voluntary ethanol consumption represent a valuable tool to investigate the relationship between age and propensity to consume alcohol using an experimental methodology. Although adolescence has been considered as a critical age, few are the studies that consider the preadolescence age. This study examines the ethanol consumption/preference and the propensity to show an alcohol deprivation effect (ADE) after a short voluntary ethanol exposure from a developmental perspective. Methods:, Three groups of heterogeneous Wistar rats of both sexes with ad libitum food and water were exposed for 10 days to 3 ethanol solutions at 3 different ontogenetic periods: preadolescence (PN19), adolescence (PN28), and adulthood (PN90). Ethanol intake (including circadian rhythm), ethanol preference, water and food consumption, and ADE were measured. Results:, During the exposure, the 3 groups differed in their ethanol intake; the greatest amount of alcohol (g/kg) was consumed by the preadolescent rats while the adolescents showed a progressive decrease in alcohol consumption as they approached the lowest adult levels by the end of the assessed period. The pattern of ethanol consumption was not fully explained in terms of hyperphagia and/or hyperdipsia at early ages, and showed a wholly circadian rhythm in adolescent rats. After an abstinence period of 7 days, adult rats showed an ADE measured both as an increment in ethanol consumption and preference, whereas adolescent rats only showed an increment in ethanol preference. Preadolescent rats decreased their consumption and their preference remained unchanged. Conclusions:, In summary, using a short period of ethanol exposure and a brief deprivation period the results revealed a direct relationship between chronological age and propensity to consume alcohol, being the adolescence a transition period from the infant to the adult pattern of alcohol consumption. Preadolescent animals showed the highest ethanol consumption level. The ADE was only found in adult animals for both alcohol consumption and preference, whereas adolescents showed an ADE only for preference. No effect of sex was detected in any phase of the experiment. [source]


Manganese-enhanced magnetic resonance imaging (MEMRI): methodological and practical considerations

Abstract Manganese-enhanced MRI (MEMRI) is being increasingly used for MRI in animals due to the unique T1 contrast that is sensitive to a number of biological processes. Three specific uses of MEMRI have been demonstrated: to visualize activity in the brain and the heart; to trace neuronal specific connections in the brain; and to enhance the brain cytoarchitecture after a systemic dose. Based on an ever-growing number of applications, MEMRI is proving useful as a new molecular imaging method to visualize functional neural circuits and anatomy as well as function in the brain in vivo. Paramount to the successful application of MEMRI is the ability to deliver Mn2+ to the site of interest at an appropriate dose and in a time-efficient manner. A major drawback to the use of Mn2+ as a contrast agent is its cellular toxicity. Therefore, it is critical to use as low a dose as possible. In the present work the different approaches to MEMRI are reviewed from a practical standpoint. Treatment of sensory defensiveness in adults

Abstract This pilot study explored the relationship between sensory defensiveness and anxiety, as well as the impact of a sensory integration treatment protocol on normal adults. Fifteen adult subjects identified as having sensory defensiveness completed the Adult Sensory Questionnaire (ASQ), Adult Sensory Interview (ADULT-SI), and Beck Anxiety Inventory (BAI) at pre-test and post-test intervals to measure sensory defensiveness and anxiety. A treatment protocol was implemented which included providing insight into sensory defensiveness, regular and daily sensory input, and engagement in activities of choice providing primarily proprioceptive, vestibular and tactile sensory input. Subjects engaged in an individualized self-treatment protocol for one month. Results indicated a significant correlation between anxiety and sensory defensiveness (r = 0.62,p = 0.027). The differences in pre-test and post-test mean scores of the Adult Sensory Interview (p = 0.048) and the Beck Anxiety Inventory (p = 0.0453) supported the use of a sensory treatment protocol to decrease sensory defensiveness and secondary anxiety. There is a need for further research using experimental methodology with a control group to explore the effectiveness of a sensory treatment protocol on sensory defensiveness and anxiety. Bifurcations on a spring-pendulum oscillator

In this paper we study experimentally the dynamics of a non linear system: a spring pendulum coupled to an oscillator. The system was tested using a hybrid technique called Real Time Dynamic Substructuring (RTDS)[1, 2]. RTDS is a testing technique that involves splitting the system under study into two subsystems: one will be physically tested (physical substructure) and the other will be simulated in the computer (numerical model). These substructures interact in real time through a set of transfer systems. RTDS is a very powerful experimental methodology that not only allows full scale and real time testing but also real-time bifurcation tracking in complex engineering systems [3, 4]. An Update on Data Standards for Gel Electrophoresis

The use of gel electrophoresis to separate and, in some instances, to quantify the abundance of large numbers of proteins from complex mixtures, has been well established for several decades. The quantity of publicly available data is still relatively modest due to a lack of community accepted data standards, tools to facilitate the data sharing process and controlled vocabularies to ensure that consistent terminology is used to describe the experimental methodology. It is becoming widely recognised that there are significant benefits in data sharing for proteomics, allowing results to be verified and new findings to be generated by re-analysis of published studies. We report on standards development by the Gel Analysis Workgroup of the Proteomics Standards Initiative. The workgroup develops reporting requirements, data formats and controlled vocabularies for experimental gel electrophoresis, and informatics performed on gel images. We present a tutorial on how such resources can be used and how the community should get involved with the on-going projects. Finally, we present a roadmap for future developments in this area.
