Cytological diagnosis of basal cell carcinoma and actinic keratosis, using Papanicolaou and May,Grünwald,Giemsa stained cutaneous tissue smear

Objective:, Cytology may become the diagnostic method of choice with the advent of new non-invasive treatments for non-melanoma skin cancer, as the sampling technique for cytology entails little tissue disfiguration. The aim of this study was to compare and evaluate the diagnostic performance of scrape cytology using two different cytological staining techniques, and to evaluate additional touch imprint cytology, with that of histopathology of basal cell carcinoma (BCC) and actinic keratosis (AK). Methods:, We investigated 50 BCC and 28 AK histologically verified lesions, from 41 and 25 patients, respectively. Two separate skin scrape samples and one touch imprint sample were taken from each lesion. The smears were stained with Papanicolaou (Pap) or May,Grünwald,Giemsa (MGG) stains. All cytological specimens were examined in random order by pathologists without knowledge of the histology. Cytodiagnostic results were compared with the histopathological report. Results:, Scrape cytodiagnosis agreed with histopathology in 48 (Pap) and 47 (MGG) of the 50 BCC cases, and in 26 of 28 (Pap) and 21 of 26 (MGG) AK cases, yielding sensitivities of 96%, 94%, 93% and 81%, respectively. No significant difference in sensitivity between the two staining methods was found but a trend towards higher Pap sensitivity for AK was noted (P = 0.10). Touch imprint cytology confirmed histopathology in 38 of the 77 cases of BCC and AK. Conclusion:, Cytological diagnosis with either Pap or MGG stain for BCC and AK is reliable, and differentiates well between BCC and AK. Imprint cytology proved to be non-diagnostic in half of the examined cases. [source]


Earthquake-induced interaction between adjacent reinforced concrete structures with non-equal heights

Abstract The influence of the structural pounding on the ductility requirements and the seismic behaviour of reinforced concrete structures designed to EC2 and EC8 with non-equal heights is investigated. Special purpose elements of distributed plasticity are employed for the study of the columns. Two distinct types of the problem are identified: Type A, where collisions may occur only between storey masses; and Type B, where the slabs of the first structure hit the columns of the other (72 Type A and 36 Type B pounding cases are examined). Type A cases yielded critical ductility requirements for the columns in the pounding area mainly for the cases where the structures were in contact from the beginning of the excitation. In both pounding types the ductility requirements of the columns of the taller building are substantially increased for the floors above the highest contact storey level probably due to a whiplash behaviour. The most important issue in the pounding type B is the local response of the column of the tall structure that suffers the hit of the upper floor slab of the adjacent shorter structure. In all the examined cases this column was in a critical condition due to shear action and in the cases where the structures were in contact from the beginning of the excitation, this column was also critical due to high ductility demands. It can be summarized that in situations of potential pounding, neglecting its possible effects leads to non-conservative building design or evaluation that may become critical in some cases. DNA mismatch repair protein expression and microsatellite instability in primary mucosal melanomas of the head and neck

Aims:, To examine the expression of DNA mismatch repair (MMR) proteins and the presence of microsatellite instability (MSI) in seven primary mucosal melanomas of the head and neck (MMHN). Methods and results:, Haematoxylin and eosin staining and immunohistochemical analysis for routine diagnostic markers and for MMR proteins were performed. Six cases were examined for MSI. Four cases were monomorphous and three cases were pleomorphic type MMHN. Melanocytic markers were positive in all cases. Immunoreactivity for MMR proteins was weak in normal epithelium. The neoplastic tissue in six cases showed positivity for all MMR proteins with different percentages. One case showed weak positivity for hMSH2 and hMSH6 and no immunoreactivity for hMLH1 or hPMS2. Staining intensity was higher in tumour cells than in matched normal mucosa in three cases for hMSH2 and hMLH1 and in two cases for hPMS2. None of the examined cases showed MSI. Conclusions:, Expression of hMSH2 and hMLH1 proteins was up-regulated in three cases, whereas in two cases that of hPMS2 was increased. hMSH6 expression was comparable to that of normal cells in all cases. The percentage of positive neoplastic cells and the intensity of staining seemed to be greater in pleomorphic melanomas. Six cases were MMR-proficient and microsatellite stable. [source]


Hybrid ,/,3 -peptides with proteinogenic side chains. monosubstituted analogues of the chemotactic tripeptide For-Met-Leu-Phe-OMe

Abstract The ,/,3 -mixed tripeptides R-CO-,3 -HMet-Leu-Phe-OMe (1a,b), R-CO-Met-,3 -HLeu-Phe-OMe (2a,b) and R-CO-Met-Leu-,3 -HPhe-OMe (3a,b) (a, R = tert -butyloxy-; b, R = H,), analogues of the potent chemoattractant For-Met-Leu-Phe-OMe, have been synthesized by classical solution methods and fully characterized. The activities of the new analogues as chemoattractants, superoxide anion producers and lysozyme releasers have been determined on human neutrophils. Whereas all of the three N -formyl derivatives are significantly less active than the parent tripeptide as chemoattractants, compound 1b has been found to be highly active as a superoxide anion producer and 3b as a lysozyme releaser. The results show that the replacement of the native Leu residue at the central position is, in each of the examined cases, the least favourable modification. The three N -Boc derivatives are, as expected, devoid of activity as agonists, but they are all good inhibitors of chemotaxis. Information on the solution conformation has been obtained by examining the involvement of the NH groups in intramolecular H-bonds using 1H NMR. The conformation of the N -Boc analogue 1a has also been determined in the crystal state by x-ray diffraction analysis. The molecule is extended at the ,3 -HMet residue (,1 = ,87°;,1 = 172°;,1 = 126° ) and no intramolecular H-bond is present. Copyright © 2004 European Peptide Society and John Wiley & Sons, Ltd. Detection of Aflatoxin in Aspergillus Species Isolated from Pistachio in Iran

Abstract To estimate the incidence contamination of fresh pistachio nuts by aflatoxigenic fungi in Iran, nut samples were collected from pistachio orchards in Kerman, Rafsanjan and Isfahan regions. Out of the 200 Aspergillus isolates obtained, 11 species were identified as A. alliaceous, A. candidus, A. flavus, A. niger, A. niveus, A. ochraceus, A. parasiticus, A. tamari, A. terreus, A. unguis and A. wentii. For detection of aflatoxin production ability of the isolates, three target genes, namely aflR, aflJ, and omtB, used in PCR amplification. In all the examined cases, the degenerate primer designed for amplification of omtB gene, named omtBII, was able to amplify an expected 611 bp fragment in aflatoxigenic isolates in this study and yielded the same result as those obtained from TLC analysis and fluorescence ability by application of methylated ,-cyclodextrin in culture media. Using this procedure the significant incidence of aflatoxin-producing aspergilli was confirmed in pistachio nuts produced in different regions of Iran. The results indicated that PCR method described here, in combination with fluorescence assay, is a reliable and simple confirmatory test for monitoring pistachio nuts contaminated with aflatoxinogenic aspergilli. The role of productive efficiency on entry and post-entry performance under different strategic orientation: the case of the Greek plastics and rubber industry

The present study formulates and empirically tests the hypothesis that the post-entry performance and growth of new firms is affected by the way in which crucial resources are combined during the decision-making process to enter the industry or not. Further, the study empirically tests the hypothesis that multifaceted productive efficiency influences both the entry decision and the entrants' post-entry performance. The proposed analytical framework allows for testing these hypotheses under different strategic orientations assumed to be followed by entrants. Investigation of bacterial communities associated with asymptomatic and symptomatic endodontic infections by denaturing gradient gel electrophoresis fingerprinting approach

The purpose of the present study was to investigate the bacterial communities associated with asymptomatic and symptomatic endodontic infections and to compare denaturing gradient gel electrophoresis (DGGE) fingerprinting patterns of these two clinical conditions. The root canal microbiota of teeth associated with asymptomatic or symptomatic periradicular lesions was profiled by the PCR-DGGE method and then compared, taking into consideration the banding patterns. Bacteria were present in all examined cases. Comparative analysis of the two clinical conditions revealed bands that were common to both symptomatic and asymptomatic cases, but most DGGE bands appeared to be unique for each clinical condition. No single band occurred in all profiles. The mean number of bands detected in the 16S rDNA community profiles were 12.1 ± 9.4 (range 2,29) for symptomatic samples and 6.7 ± 2.7 (range 2,11) for asymptomatic ones. Clustering methods and principal component analysis of DGGE banding pattern placed the samples according to the presence or absence of symptoms. Four intense bands that were excised from the gel and sequenced showed similarities to species of the Campylobacter genus (found in 5/12 asymptomatic and in 3/11 symptomatic cases), Fusobacterium genus (4/11 symptomatic cases), Acinetobacter genus (5/12 asymptomatic cases), and Enterobacteriaceae family (11/12 asymptomatic and 2/11 symptomatic cases). The profiles of the predominant bacterial community appeared to be unique for each individual. These findings confirm that endodontic infections are polymicrobial and showed that there are significant differences in the predominant bacterial composition between asymptomatic and symptomatic cases. Immunohistochemical studies of the PrPCJD deposition in Creutzfeldt-Jakob disease

The PrPCJD deposition in eight brains of sporadic Creutzfeldt-Jakob disease (CJD) was examined immunohistochemically using both hydrolytic autoclaving and formic acid pretreatment in order to understand the pathogenesis of CJD. Synaptic-type PrP immunoreactivity was revealed in the gray matter in all cases and had a tendency to be weaker in devastated areas in cases with a longer duration of illness. However, in one particular case with numerous prion plaques, the degeneration was relatively mild while PrPCJD immunoreactivity was intense despite the longest duration of illness among the examined cases. Deep layer accentuation of PrPCJD immunoreactivity was observed in the cerebral cortices in most cases. This staining pattern, however, disappeared in a burnt-out lesion exhibiting status spongiosus. The granular layer was damaged mostly in the cerebellum of the advanced cases. PrPCJD and synaptophysin immunoreactivities decreased as the tissue degeneration progressed. Interestingly, the Purkinje cells had no positivity for PrPCJD in all cases, although the neurons in relatively preserved cerebellum showed apparent positivity for synaptophysin. In the Ammon's horn and subiculum the neurons were well preserved despite the marked immunoreactivity for PrPCJD in all cases, although some cases demonstrated severe spongiform change. Approximately half of the cases showed intracytoplasmic inclusion body-like immunoreactivity for PrPCJD in neurons of the dentate nucleus. These findings suggest that PrPCJD deposition may be an event that precedes neuronal degeneration evolving from deeper layers of the cerebral cortex. Although the Ammon's horn and subiculum showed striking PrPCJD deposition and spongiform change, neuronal loss did not take place, suggesting that deposited PrPCJD itself seems not to be directly harmful to the neurons. Some investigators have assumed that microglia activated by PrPCJD plays an important role in neuronal degeneration. Considering this, we speculate that microglia in the Ammon's horn and subiculum may have a unique characteristic of not responding to PrPCJD. [source]
