Wilkes Abstract Dyneins are large protein complexes that produce directed movement on microtubules. In situ, dyneins comprise combinations of heavy, intermediate, light-intermediate, and light chains. The light chains regulate the locations and activities of dyneins but their functions are not completely understood. We have searched the recently sequenced Tetrahymena thermophila macronuclear genome to describe the entire family of dynein light chains expressed in this organism. We identified fourteen genes encoding putative dynein light chains and seven genes encoding light chain-like proteins. RNA-directed PCR revealed that all 21 genes were expressed. Quantitative real time reverse transcription PCR showed that many of these genes were upregulated after deciliation, indicating that these proteins are present in cilia. Using the nomenclature developed in Chlamydomonas, Tetrahymena expresses two isoforms each of LC2, LC4, LC7, and Tctex1, three isoforms of p28, and six LC8/LC8-like isoforms. Tetrahymena also expresses two LC3-like genes. No Tetrahymena orthologue was found for Chlamydomonas LC5 or LC6. This study provides a complete description of the different genes and isoforms of the dynein light chains that are expressed in Tetrahymena, a model organism in which the targeted manipulation of genes is straightforward. [source] Total Family Unit Helicobacter pylori Eradication and Pediatric Re-Infection RatesHELICOBACTER, Issue 4 2004Stephen Farrell ABSTRACT Background., Re-infection with Helicobacter pylori is more common in children than adults, and it is generally accepted that the family unit plays a significant role in primary childhood infection. We investigated whether the family unit plays a significant role in pediatric re-infection and if eradication of H. pylori from the entire family reduces the risk of childhood re-infection. Methods., Fifty families, each with an H. pylori -infected pediatric index case (mean age 9.48 years), were recruited. A 13carbon urea breath test was performed on all family members in the same house as the index case. Each family unit was randomized into a ,family unit treatment' group (all infected family members treated) or an ,index case treatment' group (index case only treated). Results., At long-term follow-up (mean 62.2 months), there were three re-infected children in the ,index case treatment' group compared with one in the ,family unit treatment' group. The re-infection rate was 2.4% per patient per year in the ,index case treatment' group and 0.7% per patient per year in the ,family unit treatment' group (p = .31). Conclusions., This study is the first to evaluate the effect of total family unit H. pylori eradication on pediatric re-infection rates and reports the longest period of re-infection follow-up in children. In childhood, re-infection with H. pylori is not significantly reduced by family unit H. pylori eradication. Tchougréeff Abstract We study numerically the crystal and electronic structure of the room temperature organometallic ferromagnet of general composition V(TCNE)x × y solvent with x , 2, starting from both the experimental structure of its iron analog which results from the EXAFS experiment as well as the theoretical model structure compatible with magnetic measurements on this type of compounds. The results of the numerical study performed at the density functional level of theory show that the experimentally determined structure complies with the magnetic measurements and thus can serve as a prototype structure for the entire family of the M(TCNE)2 organometallic magnets. Both the results of the numerical study and the magnetic experiments are interpreted using a proposed model Hamiltonian. © 2008 Wiley Periodicals, Inc. TAMMENTIE mnsc rn Research has shown that postnatal depression (PND) affects 10,15% of mothers in Western societies. PND is not easily identified and therefore it often remains undetected. Untreated depression has a detrimental effect on the mother and child and the entire family. The purpose of this study was to ascertain the state of family dynamics after delivery and whether the mother's PND was associated with family dynamics. The study used a survey covering the catchment area of one Finnish university hospital. Both primi- and multiparas took part and data were collected using the Edinburgh Postnatal Depression Scale (EPDS) for mothers and the Family Dynamics Measure II (FDM II) for both mothers and fathers. The data were analysed using SPSS statistical programme and frequency and percentage distributions, means and standard deviations were examined. Correlations were analysed using Spearman's correlation coefficients. The significance of any differences between mothers' and fathers' scores was determined with a paired t -test. Of the families participating in the study (373 mothers and 314 partners), 13% of the mothers suffered from PND symptoms (EPDS score of 13 or more). As a whole, family dynamics in the families participating in the study were reported to be rather good. However, mothers having depressive symptoms reported more negative family dynamics compared with other families. With the exception of individuation, mothers having depressive symptoms reported more negative family dynamics than their partners. With the exception of role reciprocity, non-depressed mothers reported more positive family dynamics than their partners. Knowledge of the association of mothers' PND with family dynamics could help to develop nursing care at maternity and child welfare clinics and maternity hospitals. Depressed mothers and their families need support to be able to make family dynamics as good as possible. [source] Molecular phylogeny of Enchytraeidae (Oligochaeta) indicates separate invasions of the terrestrial environmentJOURNAL OF ZOOLOGICAL SYSTEMATICS AND EVOLUTIONARY RESEARCH, Issue 3 2010Bent Christensen Abstract Enchytraeidae is a family of soil inhabiting small- to medium-sized oligochaete worms using degradable plant material as a food source and primarily adapted to terrestrial or semi-terrestrial environments. The molecular phylogeny based upon both mitochondrial and nuclear genes indicates early segregations of the two genera Enchytraeus and Lumbricillus leaving the remaining genera included in this study as a later segregated major monophyletic branch. Extant members of the two former genera dominate in decaying seaweed in the littoral zone along the sea although members of in particular the genus Enchytraeus have also invaded other habitats. Historically the littoral zone of the sea is undoubtedly the first terrestrial or semi-terrestrial habitat where dead plant material accumulates to any greater extent and Enchytraeus and Lumbricillus may represent early successful attempts to exploit this resource. Inland soils probably had to await the emergence of land plants in order to provide a similar food resource and here the major branch of enchytraeid genera diversified into a high number of species in the numerous decomposer networks of this varied environment. A subdivision into the genera Enchytraeus and Lumbricillus on the one hand and a branch of mainly inland genera on the other is supported by differences in two somewhat neglected morphological features. Firstly, in Enchytraeus and Lumbricillus the testes are enclosed in a testis sac within which the male cells mature, by one possible exception a unique feature among Oligochaeta, The other enchytraeid genera studied and Oligochaeta in general lack this sac and the male cells mature directly in the cavity of the testicular segment. Secondly, species of Enchytraeus and Lumbricillus generally have a higher reproductive output than species of the inland terrestrial branch and this may represent an adaptation to the unpredictable littoral zone compared to the more stable nature of inland habitats. In the older literature the genus Mesenchytraeus is considered to have a basic position within the entire family but our molecular data do not support this expectation. In Enchytraeidae the nephridia are elaborate organs of a characteristic and constant shape covering species from different genera in a pattern following the molecular phylogeny. Other much used morphological features such as shape of setae, anteclitellar origin of the dorsal vessel and various modifications of the intestine have arisen more than once. Zusammenfassung Enchytraeidae sind eine Familie kleiner bis mittelgroßer edaphischer Oligochaeten, welche abbaubares Pflanzenmaterial als Nahrungsquelle nutzen und primär an terrestrische oder semiterrestrische Lebensräume angepaßt sind. Die hier vorgestellte, auf mitochondrialen und nukleären Genen beruhende molekulare Phylogenie indiziert eine frühe Abtrennung der beiden Gattungen Enchytraeus und Lumbricillus und beläßt die übrigen der in dieser Studie untersuchten Gattungen als eine später abgezweigte monophyletische Großgruppe. Rezente Glieder der beiden erwähnten Gattungen dominieren in verrottendem Strandanwurf des Meereslitorals, obwohl Arten insbesondere der Gattung Enchytraeus auch andere Lebensräume erobert haben. Historisch gesehen ist das Meereslitoral zweifelsohne das erste terrestrische oder semiterrestrische Habitat, in dem totes Pflanzenmaterial in größerem Ausmaß akkumulierte, und Enchytraeus und Lumbricillus könnten frühe erfolgreiche Versuche der Nutzung dieser Ressource darstellen. Festlandsböden dagegen bedurften wahrscheinlich der Entwicklung von Landpflanzen, um eine vergleichbare Nahrungsquelle zur Verfügung zu stellen, und hier diversifizierte sich der Hauptzweig der Enchytraeidengattungen in viele Arten in den diversen Zersetzernetzen dieses vielgestaltigen Lebensraums. Eine Unterteilung in die Gattungen Enchytraeus und Lumbricillus auf der einen Seite und einen Zweig mit vorwiegend terrestrischen Gattungen auf der anderen wird durch zwei bislang eher vernachlässigte morphologische Merkmale gestützt. Erstens, bei Enchytraeus und Lumbricillus wird der Hoden von einem Testis-Sack, in dem die männlichen Zellen reifen, umschlossen. Dies ist mit einer möglichen Ausnahme einzigartig bei Oligochaeten. Bei den übrigen untersuchten Enchytraeidengattungen und bei Oligochaeten generell fehlt dieser Sack, und die männlichen Zellen reifen unmittelbar in der Körperhöhle des Hodensegmentes. Zweitens, die Reproduktionsrate von Enchytraeus und Lumbricillus ist generell höher als die der Arten des Festlandszweigs. Dies könnte eine Anpassung an die im Vergleich zu Festlandsböden instabileren Bedingungen des Meereslitorals bedeuten. In der älteren Literatur wird der Gattung Mesenchytraeus eine basale Position innerhalb der gesamten Familie zugewiesen, aber unsere molekularen Daten stützen diese Annahme nicht. Die Nephridien der Enchytraeiden sind komplexe Organe mit einer charakteristischen und artübergreifend konstanten Form, deren gattungsmäßige Abwandlungen der molekularen Phylogenie entsprechen. [source] Observation of vanadyl porphyrins and sulfur-containing vanadyl porphyrins in a petroleum asphaltene by atmospheric pressure photonionization Fourier transform ion cyclotron resonance mass spectrometryRAPID COMMUNICATIONS IN MASS SPECTROMETRY, Issue 14 2008Kuangnan Qian Vanadyl (VO) porphyrins and sulfur-containing vanadyl (VOS) porphyrins of a wide carbon number range (C26 to C52) and Z-number range (,28 to ,54) were detected and identified in a petroleum asphaltene by atmospheric pressure photonionization (APPI) and Fourier transform ion cyclotron resonance mass spectrometry (FTICR-MS). APPI provides soft ionization of asphaltene molecules (including VO and VOS porphyrins), generating primarily molecular ions (M+.). The ultra-high mass resolving power (m/,mFWHM ,500,K) of FTICR-MS enabled resolution and positive identification of elemental formulae for the entire family of VO and VOS porphyrins in a complicated asphaltene matrix. Deocophylerythro-etioporphyrin (DPEP) is found to be the most prevalent structure, followed by etioporphyrins (etio)- and rhodo (benzo)-DPEP. The characteristic Z-distribution of VO porphyrins suggests benzene and naphthene increment in the growth of porphyrin ring structures. Bimodal carbon number distributions of VO porphyrins suggest possible different origins of low and high molecular weight species. To our knowledge, the observation of VOS porphyrins in a petroleum product has not previously been reported. The work is also the first direct identification of the entire vanadyl porphyrin family by ultra-high resolution mass spectrometry without chromatographic separation or demetallation. [source] Decreasing household television time: a pilot study of a combined behavioral and environmental interventionBEHAVIORAL INTERVENTIONS, Issue 4 2006Amy Gorin Excessive TV viewing is associated with weight gain across the lifespan; thus developing strategies to decrease household viewing time may be an effective obesity prevention approach. This pilot study (n,=,6 families) examined the feasibility and short-term impact of a 2-pronged environmental plus behavioral approach designed to reduce TV time in the entire family. The environmental manipulation involved placing TV Allowances (programmed to turn off power after family members had watched 75% of their baseline hours) on all TVs in the home. A kit with behavioral strategies for reducing TV time was also sent to the home each week and family members self-monitored viewing time. Viewing was objectively assessed with the TV Allowances at baseline and at 8,weeks. A significant decrease in objectively measured TV viewing hours was observed, t(5),=,3.1, p,=,0.03, 29.8,±,10.3 versus 14.9,±,6.0,h (equivalent of decreasing from 7.5 to 3.7,h per day). Fifty percent of families reduced their viewing time by ,50%. The acceptability of the intervention was high, with 100% of families reporting they would recommend the TV Allowances to others. [source] Findings on the impact of treatment for childhood acute lymphoblastic leukaemia on family relationshipsCHILD & FAMILY SOCIAL WORK, Issue 3 2001Pam McGrath ABSTRACT Paediatric acute lymphoblastic leukaemia (ALL) has provided a landmark in cancer therapy as the first disseminated and otherwise lethal malignancy to be curable in the majority of patients. Although the success rate is high, this benefit is achieved through a long, invasive and very arduous treatment process. The experience of undergoing such intensive treatments affects not only the child, but the entire family. To date, however, psychosocial research is only beginning to describe the depth and breadth of the impact on families of this experience. This paper presents findings from a recent qualitative study of families coping with the initial stage (induction remission) of treatment for childhood ALL. The findings indicate that the experience is so profoundly disturbing and disruptive that it challenges the families' sense of normalcy and stability. These findings provide evidence of significant hardship and stress, with the consequent indication of the need for social work support. Above all, such families need to hear the comforting message from social workers that what they are experiencing is normal under the circumstances. Along with practical assistance, they need reassurance about the intensity of feelings they will experience and an affirmation of the difficulty of the challenges they face. [source] |
