A Facile and Efficient One-Pot Synthesis of Substituted Quinolines from ,-Arylamino Ketones Under Vilsmeier Conditions

Abstract An efficient one-pot synthesis of substituted quinolines from ,-arylamino ketones in the presence of PBr3 in DMF has been developed. This general protocol provides a novel and facile access to substituted quinolines by sequential Vilsmeier,Haack reaction, intramolecular cyclization and aromatization reactions of ,-arylamino ketones. PBr3 plays a dual role in the quinoline synthesis: as a key component of the Vilsmeier reagent (PBr3/DMF) and as a reducing reagent. (© Wiley-VCH Verlag GmbH & Co. Abstract This communication describes a catalyst-free synthesis of 2-substituted quinazolines and 4H -benzo[d][1,3]oxazines using commericially available sodium hypochlorite as oxidant. Operational simplicity, mild reaction conditions and the ability to construct structurally diverse 2-quinazolines and 2-substituted 4H -benzo[d][1,3]oxazines by this method render it to be a practical alternative for the synthesis of these heterocycles. Abstract A highly efficient one-pot synthesis of carbazoles via palladium-catalyzed double N -arylation of primary amines with 2,2,-dihalobiphenyls is described using a catalyst system comprised of tris(dibenzylideneacetone)dipalladium(0) (Pd2dba3) and the proazaphosphatrane P(i -BuNCH2CH2)3N (8) or its derivative (t- Bu)2PNP(i- BuNCH2CH2)3N (9a) as the ligand. The process is effective for double N -arylation of 2,2,-biphenyl dibromide, diiodide, and even dichloride with a variety of primary amines including neutral, electron-rich, electron-deficient, and sterically hindered anilines as well as aliphatic amines.
