    THERMAL AND HIGH-PRESSURE STABILITY OF PURIFIED PECTIN METHYLESTERASE FROM PLUMS (PRUNUS DOMESTICA)

    ABSTRACT Pectin methylesterase (PME) from greengage plums (Prunus domestica) has been extracted and purified using affinity chromatography. Only one band on sodium dodecyl sulfate,polyacrylamide gel electrophoresis was obtained, with an estimated molecular weight of 31 kDa. On isoelectric focusing electrophoresis, two bands with neutral isoelectric points (6.8 and 7.0) were detected. The optimal pH and temperature for plum PME activity were 7.5 and 65C, respectively. A study of purified plum PME thermostability was performed at pH 7.5 and 4.0, indicating a higher thermostability at pH 7.5 than at pH 4.0. A biphasic inactivation behavior was observed for thermal treatments (54,70C), whereas its pressure inactivation could be described by a first-order kinetic model in a pressure range of 650,800 MPa at 25C. Purified plum PME was found to be relatively stable to thermal and pressure (,600 MPa) treatments, compared to PME from other fruits. [source]


    POLYPHENOLOXIDASE ACTIVITY OF MINIMALLY PROCESSED ,JONAGORED' APPLES (MALUS DOMESTICA)

    ABSTRACT The influence of three chemical dips using ascorbic acid (AA), citric acid (CA) and calcium chloride (CC) on the polyphenoloxidase (PPO) activity and on the total phenolic content of minimally processed (MP) apple (Malus domestica, cv. Jonagored) during cold storage was evaluated and a potential relationship with enzymatic browning was investigated. An ascorbic acid dip (42.6 mM) of 5 min duration was the most efficient chemical treatment in reducing the PPO activity of apple cubes. A 92% inhibition was achieved after 7 days of storage at 4C. All treatments were advantageous in comparison to the control in reducing color changes. Color changes, determined by absorbance at 420 nm (soluble pigments) and lightness (L) (insoluble pigments) of apple cubes treated with ascorbic acid were correlated with total phenolic content. No correlation was observed between PPO activity and tristimulus color parameters, browning index or total phenolic content of AA-treated apple cubes. [source]


    Emx3 is required for the differentiation of dorsal telencephalic neurons

    Abstract emx3 is first expressed in prospective telencephalic cells at the anterior border of the zebrafish neural plate. Knockdown of Emx3 function by morpholino reduces the expression of markers specific to dorsal telencephalon, and impairs axon tract formation. Rescue of both early and late markers requires low-level expression of emx3 at the one- or two-somite stage. Higher emx3 expression levels cause dorsal telencephalic markers to expand ventrally, which points to a possible role of emx3 in specifying dorsal telencephalon and a potential new function for Wnt/beta-catenin pathway activation. In contrast to mice, where Emx2 plays a major role in dorsal telencephalic development, knockdown of zebrafish Emx2 apparently does not affect telencephalic development. Similarly, Emx1 knockdown has little effect. Previously, emx3 was thought to be fish-specific. However, we found all three emx orthologs in Xenopus tropicalis and opossum (Monodelphis domestica) genomes, indicating that emx3 was present in an ancestral tetrapod genome.     Effects of locomotor stimulation and protein synthesis inhibition on circadian rhythms in size changes of L1 and L2 interneurons in the fly's visual system

    Abstract Axons of monopolar cell interneurons L1 and L2 in the first optic lobe (lamina) of the fly Musca domestica undergo cyclical changes in diameter. These axons swell during the day and shrink during the night. In addition, the axons' size depends on light conditions since they are largest in continuous light (LL), somewhat smaller under day/night (LD) conditions, and smallest under constant darkness (DD). In this study we found that sizes of both cells can further increase in free flying flies under LD conditions, while the visual stimulation alone does not have significant effect on the cross-sectional area of L1 and L2 axons. The stimulation of free flying had no effect on L1 and L2 sizes if it was performed at the beginning of subjective day in LL or DD. Our results indicate that a maximal increase in size of L1 and L2 is observed when stimulation of free flying is synchronized with a fly' daily peak of activity.     Functional response and size-dependent foraging on aquatic and terrestrial prey by brown trout (Salmo trutta L.)

Despite this, size-specific use of and efficiency in procuring this resource have not been studied to any great extent. Therefore, we measured the functional responses of three size classes of wild brown trout Salmo trutta (0+, 1+ and ,2+) when fed either benthic- (Gammarus sp.) or surface-drifting prey (Musca domestica) in laboratory experiments. To test for size-specific prey preferences, both benthic and surface prey were presented simultaneously by presenting the fish with a constant density of benthic prey and a variable density of surface prey. The results showed that the functional response of 0+ trout differed significantly from the larger size classes, with 0+ fish having the lowest capture rates. Capture rates did not differ significantly between prey types. In experiments when both prey items were presented simultaneously, capture rate differed significantly between size classes, with larger trout having higher capture rates than smaller trout. However, capture rates within each size class did not change with prey density or prey composition. The two-prey experiments also showed that 1+ trout ate significantly more surface-drifting prey than 0+ trout. In contrast, there was no difference between 0+ and ,2+ trout. Analyses of the vertical position of the fish in the water column corroborated size-specific foraging results: larger trout remained in the upper part of the water column between attacks on surface prey more often than smaller trout, which tended to seek refuge at the bottom between attacks. These size-specific differences in foraging and vertical position suggest that larger trout may be able to use surface-drifting prey to a greater extent than smaller conspecifics. [source]


    Factors influencing the effectiveness of an attracticide formulation against the Oriental fruit moth, Grapholita molesta

    Abstract An attracticide formulation, LastCallÔOFM, was tested against the Oriental fruit moth Grapholita molesta (Busck) (Lepidoptera: Tortricidae) in replicated small plot field trials in apple, Malus domestica (Borkhausen), orchards in South-eastern Pennsylvania, USA. Attracticide treatments were applied using a calibrated hand pump, and treated plots were compared to similar untreated plots. Male moth activity was monitored using virgin female-baited traps, and the potential for reduction in mating activity was assessed using sentinel virgin females. A comparison of application rates showed that 1500 droplets per ha of the attracticide formulation was as effective as 3000 droplets per ha, and both application rates reduced captures in synthetic pheromone-baited traps for prolonged periods. Droplets placed either at high or low positions within the canopy significantly reduced trap capture and mating with sentinel females. In addition, the only sentinel females that mated in the treated plots were located in the untreated portion of the tree canopy. Mate finding behaviour was equally disrupted by formulations with and without insecticide. Therefore, under the test conditions, the mechanism by which the attracticide formulation worked was by disruption of male orientation, and not by the removal of males due to insecticide poisoning. Two field cage experiments tested the impact of population density on the competitiveness of the attracticide formulation compared to virgin females. A significant proportion of males were captured in female-baited traps at the highest female-to-droplet ratio tested. Equal proportions of males were captured in attracticide-baited traps at male moth densities of 10, 20, 40, and 80 males per cage. These results clarify some of the factors influencing the effectiveness and possible mechanisms of an attracticide management tactic against the Oriental fruit moth. [source]


    Quantitative aspects of the regulation of ovarian development in selected anautogenous Diptera: integration of endocrinology and nutrition

    Abstract Aspects of the influence of nutrition on the degree of ovarian development in selected anautogenous Diptera are reviewed. The Diptera considered are several mosquito species, with emphasis on Aedes aegypti, the house fly, Musca domestica, the Australian bush fly, M. vetustissima and the blowflies, Lucilia cuprina and Phormia regina. All the selected species display discrete ovarian cycles in which all oocytes destined to reach maturity in a particular ovarian cycle develop synchronously. In these species, the proportion of females maturing oocytes and, where such data exist, the number of oocytes they mature are positively correlated with the amount of any particular nitrogen-containing material ingested or given by enema or by infusion. In addition, the degree of ovarian development may be affected by the chemical composition of nitrogen-containing food. Possible physiological bases for the observed relationships are discussed. Available evidence suggests that whether or not a female matures any oocytes is hormonally regulated and that the number of oocytes matured is probably regulated by the availability of nutrients. Some approaches that might further elucidate the physiological regulatory mechanisms involved in ovarian development are outlined. [source]


    Relationship between diet composition and the fecundity of Musca domestica

    Characterization of alanyl aminopeptidase from insecticide resistant and susceptible strains of Musca domestica L.

    Abstract To investigate the high activity of intracellular proteases in insecticide resistant strains of Musca domestica L., purification by anion-exchange chromatography and gel filtration of one of the enzymes, alanyl aminopeptidase (Ala AP), in three strains of Musca domestica was carried out. The fractions collected by gel filtration of soluble homogenates of the three strains (571ab, 17bb and Cooper) showed a single peak of Ala AP activity. Partially purified Ala AP of the three strains showed high activity at pH 7.5. The presence or absence of Ca2+ in the assay medium did not produce any difference in activity of Ala AP in the 571ab and Cooper strains, but there was a significant difference in the 17bb strain. The activity of Ala AP in all three strains was essentially unaltered in the presence of inhibitors of serine (PMSF), cysteine (E-64) proteases and carboxypeptidases (pepstatin). Ala AP hydrolyzed alanine amino methylcoumarin (Ala-AMC) maximally, followed by phenyl alanine amino methylcoumarin (Phe-AMC), leucyl amino methylcoumarin (Leu-AMC) and ornithine amino methylcoumarin (Orn-AMC). Ala AP from the three strains showed differential activity towards various substrates. The comparison of alanyl aminopeptidase's activity from different sources is discussed. [source]


    Increased toxicity to invertebrates associated with a mixture of atrazine and organophosphate insecticides

    Abstract This study examined the joint toxicity of atrazine and three organophosphate (OP) insecticides (chlorpyrifos, methyl parathion, and diazinon) exposed to Hyalella azteca and Musca domestica. A factorial design was used to evaluate the toxicity of binary mixtures in which the lethal concentration/lethal dose (LC1/LD1, LC5/LD5, LC15/LD15, and LC50/LD50) of each OP was combined with atrazine concentrations of 0, 10, 40, 80, and 200 ,g/L for H. azteca and 0, 200, and 2,000 ng/mg for M. domestica. Atrazine concentrations (>40 ,g/L) in combination with each OP caused a significant increase in toxicity to H. azteca compared with the OPs dosed individually. Acetylcholinesterase (AChE) activity also was examined for the individual OPs with and without atrazine treatment. Atrazine in combination with each of the OPs resulted in a significant decrease in AChE activity compared with the OPs dosed individually. In addition, H. azteca that were pretreated with atrazine (>40 ,g/L) were much more sensitive to the OP insecticides compared with H. azteca that were not pretreated with atrazine before being tested. Topical exposure to atrazine concentrations did not significantly increase OP toxicity to M. domestica. The results of this study indicate the potential for increased toxicity in organisms exposed to environmental mixtures. [source]

    The Effects of Experimentally Induced Polyandry on Female Reproduction in a Monandrous Mating System

    Females of most insect species maximize their fitness by mating more than once. Yet, some taxa are monandrous and there are two distinct scenarios for the maintenance of monandry. While males should always benefit from inducing permanent non-receptivity to further mating in their mate, this is not necessarily true for females. Since females benefit from remating in many species, cases of monandry may reflect successful male manipulation of female remating (i.e. sexual conflict). Alternatively, monandry may favor both mates, if females maximize their fitness by mating only once in their life. These two hypotheses for the maintenance of monandry make contrasting predictions with regards to the effects of remating on female fitness. Here, we present an experimental test of the above hypotheses, using the monandrous housefly (Musca domestica) as a model system. Our results showed that accessory seminal fluid substances that males transfer to females during copulation have a dual effect: they trigger female non-receptivity but also seem to have a nutritional effect that could potentially enhance female fitness. These results suggest that monandry is maintained in house flies despite potential benefits that females would gain by mating multiply. [source]


    Nursing Synchronization and Milk Ejection Failure as Maternal Strategies to Reduce Allosuckling in Pair-Housed Sows (Sus scrofa domestica)

    Non-offspring nursing (allosuckling) is costly for lactating females. It is reasonable to assume, therefore, that mechanisms have evolved to control the frequency of allosuckling. It is suggested that the synchronization of nursing between females could be such a mechanism. The aim of this study was to investigate whether nursing synchronization is related to the timing of nursing behaviour of paired sows and the behaviour of alien piglets in relation to allosuckling. Specifically, we predicted that: (i) nursing synchronization will increase if there is a real risk of allosuckling compared with a situation where there is none, (ii) nursing synchronization indeed reduces allosuckling and, therefore, those nursings with allosuckling attempts will be less closely synchronized than those without allosuckling attempts and (iii) in non-synchronized nursings, the sows will more often employ another strategy to reduce allosuckling, namely, they will make the nursings non-nutritive by not releasing milk. Ten pairs of sows and their litters were used in this study. For each pair, the two sows were housed individually until their litters were weaned; however, their litters had access to both sows from day 11 post-partum. The behaviour of the 10 pairs of sows with their litters was videotaped for 6 h on days 10, 11, 17 and 24 post-partum. The frequency of allosuckling attempts, non-nutritive nursings and allosuckling during milk ejection was analysed for every pair for every day of observation. The observed frequency of nursing synchronization was high. The median interval between the start of a nursing within one pair of sows was 32 s. The first prediction was supported as synchronization became significantly tighter when piglets were given the possibility to allosuckle on day 11 [Cochran,Mantel,Haenszel (CMH) statistics, p < 0.0001]. The second prediction was also supported; the nursings were less closely synchronized when there were allosuckling attempts (CMH statistics, p < 0.05). Of all nursings, 36% had an allosuckling attempt; however, only 17% resulted in real allosuckling. Evidence in favour of the third prediction was also found as non-nutritive nursings were more frequent in non-synchronized nursings (CMH statistics, p < 0.05). Finally, real allosuckling during milk ejection was unrelated to the degree of nursing synchronization, as in less synchronized nursings, the sows responded to allosuckling attempts by not releasing milk. Our results suggest that sows are not able to completely suppress allosuckling; however, they can reduce its frequency through nursing synchronization and non-nutritive nursings. [source]


    Evolutionary and functional analysis of the tailless enhancer in Musca domestica and Drosophila melanogaster

    SUMMARY To further understand the evolutionary dynamics of the regulatory interactions underlying development, we expand on our previous analysis of hunchback and compare the structure and function of the tailless enhancer between Musca domestica and Drosophila melanogaster. Our analysis shows that although the expression patterns and functional protein domains of tll are conserved between Musca and Drosophila, the enhancer sequences are unalignable. Upon closer investigation, we find that these highly diverged enhancer sequences encode the same regulatory information necessary for Bicoid, Dorsal, and the terminal system to drive tll expression. The binding sites for these transcription factors differ in the sequence, number, spacing, and position between the Drosophila and Musca tll enhancers, and we were unable to establish homology between binding sites from each species. This implies that the Musca and Drosophila Bcd-binding sites have evolved de novo in the 100 million years since these species diverged. However, in transgenic Drosophila embryos the Musca tll enhancer is able to drive the same expression pattern as endogenous Drosophila tll. Therefore, during the rapid evolution of enhancer sequences individual binding sites are continually lost and gained, but the transcriptional output is maintained by compensatory mutations in cis and in trans.     Analysis of nubbin expression patterns in insects

    Summary Previous studies have shown that the gene nubbin (nub) exhibits large differences in expression patterns between major groups of arthropods. This led us to hypothesize that nub may have evolved roles that are unique to particular arthropod lineages. However, in insects, nub has been studied only in Drosophila. To further explore its role in insects in general, we analyzed nub expression patterns in three hemimetabolous insect groups: zygentomans (Thermobia domestica, firebrat), dyctiopterans (Periplaneta americana, cockroach), and hemipterans (Oncopeltus fasciatus, milkweed bug). We discovered three major findings. First, observed nub patterns in the ventral central nervous system ectoderm represent a synapomorphy (shared derived feature) that is not present in other arthropods. Furthermore, each of the analyzed insects exhibits a species-specific nub expression in the central nervous system. Second, recruitment of nub for a role in leg segmentation occurred early during insect evolution. Subsequently, in some insect lineages (cockroaches and flies), this original role was expanded to include joints between all the leg segments. Third, the nub expression in the head region shows a coordinated change in association with particular mouthpart morphology. This suggests that nub has also gained an important role in the morphological diversification of insect mouthparts. Overall, the obtained data reveal an extraordinary dynamic and diverse pattern of nub evolution that has not been observed previously for other developmental genes. [source]


    Early differentiation and migration of cranial neural crest in the opossum, Monodelphis domestica

    SUMMARY Marsupial mammals are born at a highly altricial state. Nonetheless, the neonate must be capable of considerable functional independence. Comparative studies have shown that in marsupials the morphogenesis of many structures critical to independent function are advanced relative to overall development. Many skeletal and muscular elements in the facial region show particular heterochrony. Because neural crest cells are crucial to forming and patterning much of the face, this study investigates whether the timing of cranial neural crest differentiation is also advanced. Histology and scanning electron microscopy of Monodelphis domestica embryos show that many aspects of cranial neural crest differentiation and migration are conserved in marsupials. For example, as in other vertebrates, cranial neural crest differentiates at the neural ectoderm/epidermal boundary and migrates as three major streams. However, when compared with other vertebrates, a number of timing differences exist. The onset of cranial neural crest migration is early relative to both neural tube development and somite formation in Monodelphis. First arch neural crest cell migration is particularly advanced and begins before any somites appear or regional differentiation exists in the neural tube. Our study provides the first published description of cranial neural crest differentiation and migration in marsupials and offers insight into how shifts in early developmental processes can lead to morphological change.     Use of quantitative real-time polymerase chain reaction to estimate the size of the house-fly Musca domestica genome

    Abstract House-flies, Musca domestica, are carriers of more than 100 devastating diseases that have severe consequences for human and animal health. A key bottleneck to progress in controlling the devastating human diseases transmitted by house-flies is lack of knowledge of the basic molecular biology of this species. However, before sequencing of the house-fly genome can be seriously considered it is important to know the size of the genome. In this paper, we used quantitative real-time polymerase chain reaction to calculate genome size of the house-fly in side-by-side experiments with Drosophila melanogaster (known genome size of 180 Mb). Our results indicate the size of the house-fly genome is 295 ± 10 Mb and that of D. melanogaster is 184 Mb. Thus, the house-fly genome is only about 1.6-fold larger than the genome of D. melanogaster. This indicates that the size of the house-fly genome makes it an excellent candidate for whole genome sequencing and that quantitative real-time polymerase chain reaction is an accurate method for the estimation of the size of insect genomes.     Identification of mariner elements from house flies (Musca domestica) and German cockroaches (Blattella germanica)

    Abstract Full-length mariner elements were isolated and sequenced from house flies (Musca domestica) and German cockroaches (Blattella germanica). The amino acid sequence of the house fly mariner element (accession number: AF373028) showed 99.5% identity with Mos1 and peach elements, whereas the German cockroach mariner element (accession number: AF355143) showed 98.8% and 99.8% identity, respectively. Sequence analysis revealed that the mariner elements in house flies and German cockroaches differed from the active Mos1 mariner element by seven and 15 nucleotides, respectively. Four essential nucleotide substitutions at positions 64, 154, 305, and 1203, which have been proposed to contribute to the loss of activity of the inactive elements, were detected in the German cockroach mariner element. In contrast, although the mariner element in house flies contained substitutions at positions 64, 154, and 305, it retained T at position 1203, identical to active mariner elements. Mariner is present in approximately eight copies in the German cockroach genome. [source]


    Isolation and molecular characterization of Musca domestica delta-9 desaturase sequences

    Abstract We have isolated fatty acyl-CoA desaturase cDNA (Mdomd9) and genomic sequences from the housefly, Musca domestica. Two ,1.66 kb cDNAs were recovered. They had identical coding regions and 3, untranslated regions (UTRs), but differed in their 5, UTRs. The open reading frame encodes a 380 amino acid (aa) protein with 82% identity to Drosophila melanogaster desat1, and significant (> 50%) identity with other insect delta-9 desaturases. Functional analyses in a yeast expression system confirmed the cDNA encodes a ,9 desaturase. Northern analysis indicated two transcripts of 1.7 and 2.9 kb that hybridized specifically to the open reading frame. PCR amplification of genomic templates revealed three intron sites that are conserved among other insect species. Southern analysis of genomic DNA indicated at least two desaturase gene copies per haploid genome. There is a high degree of polymorphism, most of which appears to be due to variable intron sequences; curiously, individual flies had varying morphs of intron II and intron III. Together, the data suggest that there are more ,9 desaturase alleles within the population studied than there are loci within the genome, and support other studies suggesting that insect fatty acyl-CoA desaturases are a dynamically evolving gene family.     Structure of Hermes integrations in the germline of the yellow fever mosquito, Aedes aegypti

    Abstract The Hermes transposable element is derived from the house fly, Musca domestica, and can incorporate into the germline of the yellow fever mosquito, Aedes aegypti. Preliminary Southern analyses indicated that Hermes integrated along with the marker gene into the mosquito genomic DNA. Here we show that Hermes integrations are accompanied by the integration of the donor plasmid as well. In addition, breaks in the donor plasmid DNAs do not occur precisely, or at the end of the terminal inverted repeats, and are accompanied by small deletions in the plasmids. Furthermore, integrations do not cause the typical 8-bp duplications of the target site DNA. No integrations are observed in the absence of a source of Hermes transposase. The Hermes transposase clearly did not catalyse precise cut-and-paste transposition in these transformed lines. It may have integrated the transposon through general recombination or through a partial replicative transposition mechanism. The imprecision of Hermes integration may result from interactions of the transposase with an endogenous hAT -like element in the mosquito genome. [source]


    Changes in the oviducal epithelium during the estrous cycle in the marsupial Monodelphis domestica

    Abstract The Monodelphis oviduct can be divided into four anatomical segments: preampulla (comprising fimbriae and infundibulum), ampulla, isthmus with crypts and uterotubal junction. Ovaries are enclosed in a periovarial sac, the bursa, and in some specimens tubules of an epoophoron could be identified. In both structures non-ciliated cells develop small translucent vesicles, which accumulate in the cell apices and presumably produce fluid as often seen in the bursa and in the tubules of the epooophoron. These vesicles do not stain with Alcian blue or PAS. The same applies also to the non-ciliated cells of the fimbriae. The oviducal epithelium of ampulla and the surface epithelium of the isthmus consisting of ciliated and non-ciliated, secretory cells undergo considerable changes during the estrous cycle. Proestrus shows low numbers of ciliated cells, some are in the process of neo-ciliogenesis, non-ciliated cells carry solitary cilia and few remnant secretory granules from the previous cycle may be found. At estrus the amount of ciliated cells in ampulla and isthmus has increased, most non-cililated cells lost the solitary cilia, developed longer microvilli and formed numerous secretory granules in their cell apices. At postestrus secretory products, often surrounded by membranes, are extruded into the oviducal lumen and contribute towards egg coat formation. First signs of deciliation processes are apparent. Solitary cilia reappear. At metestrus only few secretory cells are left with some secretory material. The lumen is often filled with shed cilia and cell apices. Proliferation of basal bodies within non-secretory cells indicate the formation of new ciliated cells. The non-ciliated epithelial cells of the isthmic crypts form no secretory granules but accumulate a great number of translucent vesicles, which in contrast to the secretory granules do not stain with Alcian blue or PAS.     Ultrastructure of the placenta of the tammar wallaby, Macropus eugenii: comparison with the grey short-tailed opossum, Monodelphis domestica

    Abstract The ultrastructure of the tammar placenta was studied throughout pregnancy. The uterine epithelium grows from a columnar to an enlarged, undulating epithelium between early gestation and mid-gestation when the shell coat that surrounds the marsupial conceptus ruptures. Trophectoderm and uterine epithelium do not form syncytia, nor does invasion of the endometrium occur at any stage of pregnancy. Uterine secretion is provided to both the bilaminar and the trilaminar side of the yolk sac placenta up to birth. Fenestrations, abundant vesicles and lumenal processes of maternal capillaries, as well as deep basal folds of the uterine epithelium, suggest that there is transfer of hemotrophes adjacent to both parts of the yolk sac. In contrast, in the grey short-tailed opossum, these structures are lacking. The yolk sacs of adjacent embryos fuse to form a common yolk sac cavity, thus losing most of the bilaminar yolk sac. The bilaminar and trilaminar components of the yolk sac placenta of the tammar are less different in structure and function than those of the grey short-tailed opossum, but both types are fully functional placentas. The extended secretory phase of the tammar uterus and the maternal recognition of early pregnancy appear to be derived characters of macropodid marsupials. [source]


    THERMAL AND HIGH-PRESSURE STABILITY OF PURIFIED PECTIN METHYLESTERASE FROM PLUMS (PRUNUS DOMESTICA)

    ABSTRACT Pectin methylesterase (PME) from greengage plums (Prunus domestica) has been extracted and purified using affinity chromatography. Only one band on sodium dodecyl sulfate,polyacrylamide gel electrophoresis was obtained, with an estimated molecular weight of 31 kDa. On isoelectric focusing electrophoresis, two bands with neutral isoelectric points (6.8 and 7.0) were detected. The optimal pH and temperature for plum PME activity were 7.5 and 65C, respectively. A study of purified plum PME thermostability was performed at pH 7.5 and 4.0, indicating a higher thermostability at pH 7.5 than at pH 4.0. A biphasic inactivation behavior was observed for thermal treatments (54,70C), whereas its pressure inactivation could be described by a first-order kinetic model in a pressure range of 650,800 MPa at 25C. Purified plum PME was found to be relatively stable to thermal and pressure (,600 MPa) treatments, compared to PME from other fruits. [source]


    RADIO FREQUENCY-HOT WATER DIPS FOR POSTHARVEST CODLING MOTH CONTROL IN APPLES,

    ABSTRACT A combination radio frequency-hot water dip method was examined as a potential quarantine treatment against fifth instars of the codling moth, Cydia pomonella L. (Lepidoptera: Tortricidae), in apples, Malus sylvestris (L.) var. domestica (Borkh.) Mansf., which were intended for export to Japan. The apples were initially exposed to 27.12-MHz radio frequency energy at 12 kW for 2.75 min and were then submerged in a range of hot water dips (48,50C) for different durations. Efficacious tests were at 48C for >2 h, at 49C for >50 min and at 50C for >40 min. Fruit quality tests indicated that the best hot water parameters were at 50C for 40 min. Fruit quality after 2 weeks was cultivar dependent where "Fuji" apples tolerated heat treatment better than "Delicious" and "Gala" apples. None of the treated fruits were acceptable after 60 days. Regardless of cultivar, heat treatment resulted in loss of both peel and fresh colors, coupled with reduced firmness and increased external and internal damage. [source]


    POLYPHENOLOXIDASE ACTIVITY OF MINIMALLY PROCESSED ,JONAGORED' APPLES (MALUS DOMESTICA)

    ABSTRACT The influence of three chemical dips using ascorbic acid (AA), citric acid (CA) and calcium chloride (CC) on the polyphenoloxidase (PPO) activity and on the total phenolic content of minimally processed (MP) apple (Malus domestica, cv. Jonagored) during cold storage was evaluated and a potential relationship with enzymatic browning was investigated. An ascorbic acid dip (42.6 mM) of 5 min duration was the most efficient chemical treatment in reducing the PPO activity of apple cubes. A 92% inhibition was achieved after 7 days of storage at 4C. All treatments were advantageous in comparison to the control in reducing color changes. Color changes, determined by absorbance at 420 nm (soluble pigments) and lightness (L) (insoluble pigments) of apple cubes treated with ascorbic acid were correlated with total phenolic content. No correlation was observed between PPO activity and tristimulus color parameters, browning index or total phenolic content of AA-treated apple cubes. [source]


    Insecticidal activity of garlic juice in two dipteran pests

    Abstract 1Botanical products excluding pyrethroids constitute a small, but growing portion of the U.K. pesticides market. With increasing legislative pressure upon chemical pesticides such as organophosphates, interest in this sector is increasing steadily. 2Garlic Allium sativum L. juices and extracts form the basis of several commercially available pest control products, but the performance of these products is variable, possibly due to lack of quality control upon batches of materials in the manufacturing process. 3Some garlic products designed for use in the food industry are subjected to rigorous batch-control to ensure organoleptic consistency. We studied the insecticidal efficacy of a commercially produced food grade garlic juice using two target dipteran pests, Delia radicum (L.) and Musca domestica L. 4Exposure of the two species to different concentrations of garlic juice revealed variability in insecticidal effect across life stages. LC50 values recorded for D. radicum were: eggs (7-day exposure) 0.8%; larvae (24-h exposure) 26.4%; larvae (48-h exposure) 6.8%; and adults (24-h exposure) 0.4%. LC50 values recorded for M. domestica were: eggs (7-day exposure) 1.6%; larvae (24-h exposure) 10.1%; larvae (24-h exposure) 4.5%; and adults (24-h exposure) 2.2%. 5Mortality rates caused by the garlic juice were comparable with those obtained with the organophosphate pesticide Birlane®, indicating parity of effect at various concentrations depending on life stage. 6We conclude that this product may provide an effective, naturally-derived insecticide for use in agricultural systems against dipteran pests.     Influences of Cross Pollination on Pollen Tube Growth and Fruit Set in Zuili Plums (Prunus salicina)

    Abstract Zuili plum (Prunus salicina L.) trees usually set fruit poorly, although they produce high quality fruit. To elucidate the causes of the poor fruit set, pollen tube growth into pistils and fruit set percentage were investigated after cross-, self- and open-pollination. Ovule development in Zuili pistils was also investigated. Pollen tube penetration into the ovules via the obturator and micropyle was best when Zuili pistils were pollinated by cv. Black Amber (P. domestica) pollen grains, although cross-pollinations with Hongxinli and Miili (P. salicina) pollen were more effective than self- and open-pollination. The fruit set percentage was also highest in pistils pollinated with Black Amber pollen grains. Morphological observation of Zuili pistils revealed that the trees produce "double pistils", developing two ovaries from a basal pistil, at a rate as high as 28%. In such abnormal pistils, most ovules were lacking an embryo sac or were entirely degenerated. The percentage of normally developed ovules was 24.3% and 8.9% in normal and double pistils, respectively. From these results, we conclude that the main causes of poor fruit set of Zuili plums are a lack of effective cross-pollination and the production of high percentages of double pistils in which normally developed ovules are scarcely formed.     cDNA cloning of the housefly pigment-dispersing factor (PDF) precursor protein and its peptide comparison among the insect circadian neuropeptides

    Abstract Pigment-dispersing factor (PDF), an 18-amino acid neuropeptide, is a principal circadian neurotransmitter for the circadian rhythms of the locomotor activity in flies. Recently, two completely different types of PDF precursor were clarified; that of the cricket Gryllus bimaculatus and that of the last-summer cicada Meimuna opalifera. The G. bimaculatus PDF precursor is extraordinarily short and comprises a nuclear localization signal (NLS), while the M. opalifera PDF precursor is of ordinary length, comparable to that seen for the precursors of crustacean ,-PDH homologues. Although their PDF peptide regions were exactly the same, the regions containing a signal peptide combined with a PDF-associated peptide (PAP) were remarkably different from each other. Such a grouping suggested a fundamental role for the PAP peptide in the circadian clock, perhaps associated with PDF function. In the present study, the cDNA cloning of PDF from the adult brains of the housefly Musca domestica was carried out and it was found that an isolated clone (527 bp) encodes a PDF precursor protein of ordinary length. The PDF peptide shows a high sequence identity (78%,94%) and similarity (89%,100%) to insect PDFs and also to the crustacean ,-PDH peptides. In particular, there is only a single amino acid difference between the PDFs of Musca and Drosophila; at position 14 Ser for Musca PDF and Asn for Drosophila PDF. A characteristic Ser10 in Drosophila was retained in Musca, indicating the presence of a structural profile unique to these PDFs. The results of sequence analyses suggest that Musca and Drosophila PDFs are to be considered members of a single group that has evolved structurally. When the primary structure of the PAP regions was compared, the Musca PDF precursor also belonged to the same group as that to which the Drosophila PDF precursor belongs.     Early ontogeny and placentation of the grey short-tailed opossum, Monodelphis domestica (Didelphidae: Marsupialia): contribution to the reconstruction of the marsupial morphotype

    This study provides new findings on the placenta of Monodelphis domestica and a reconstruction of the marsupial morphotype. To achieve this, early ontogeny and placentation of the grey short-tailed opossum, M. domestica, from 3 h after copulation until birth (day 15), were studied and compared with other mammals. Both the ultrastructure and histochemistry of egg membranes, foetal membranes, oviduct and uterus were examined. The results of this study provide the first detailed ultrastructural description of a trophoblastic syncytium in a marsupial. In addition, this is the first original documentation of an invasive trophectoderm and an inflammatory reaction at parturition in M. domestica. These findings were compared with literature data and included into the reconstruction of the marsupial morphotype. Based on marsupial phylogeny as proposed by Luckett (J. Mammal. Evol. 2, 255,283, 1994), characters that are consistent at least within didelphids and dasyurids were determined to be characters of the marsupial morphotype. These characters are a central yolk separated from the peripheral yolk-poor cytoplasm in the unfertilized oocyte, the presence of a zona pellucida, a mucoid coat and a shell coat, the absence of a corona radiata, oviductal mucoid secretion, no shell secretion distal to the isthmus of the oviduct, uterine shell secretion, a short tubal passage (1 day at maximum), the apposition of blastomeres to the zona pellucida prior to intercellular association, the absence of a morula stage, the polarity of the zygotic yolk, the localized segmentation of deutoplasm (yolk) during the first cleavage and subsequent extrusion of yolk vesicles during the first two cleavage stages. With regard to the marsupial morphotype, the non-polarized yolk distribution in the zygote [Hartman (J. Morphol. 27, 1,84, 1916); McCrady (Am. Anat. Mem. 16, 1,233, 1938)] is a derived character of Didelphis virginiana. Didelphis virginiana [Hartman (J. Morphol. 27, 1,84, 1916); Hartman (J. Morphol. 32, 1,139, 1919); McCrady (Am. Anat. Mem. 16, 1,233, 1938)] and Didelphis marsupialis (Hill, Q. J. Micr. Sci. 63, 91,139, 1918) share the synapomorphous reduction of deutoplasmolysis to a generalized extrusion of vesicles. The absence of separated yolk and consequently a cleavage without yolk extrusion (Renfree and Lewis, Reprod. Fert. Dev. 8, 725,742, 1996) are apomorphies of macropodids. This is possibly correlated with the association of blastomeres in early cleavage stages (Renfree and Lewis, Reprod. Fert. Dev. 8, 725,742, 1996). A yolk sac placenta and a vascularized allantochorion can be assumed for part of the ontogeny in the marsupial morphotype, irrespective of the formation of an allantoic placenta at near term stages. The character polarization of the mode of placentation and parturition needs further investigation. Es wird die Ultrastruktur und Histochemie der Eihäute, der Fetalmembranen, des Oviductes und des Uterus beschrieben. Erstmalig wird die Ultrastruktur eines trophoblastischen Syncytiums bei einem Beuteltier beschrieben. Weiterhin wird ein invasives Trophektoderm und eine Entzündungsreaktion zum Zeitpunkt der Geburt bei M. domestica festgestellt. Die Befunde dieser Studie und Literaturdaten werden verglichen und in eine Grundplanrekonstruktion integriert. Merkmale, die mindestens zwischen Vertretern der Didelphidae und Dasyuridae übereinstimmen, werden basierend auf dem phylogenetischen System der Marsupialia nach Luckett, J. Mammal. In Bezug auf den Grundplan der Marsupialia ist die unpolare Dotterverteilung in der Zygote ein abgeleitetes Merkmal von Didelphis virginiana. Didelphis virginiana und Didelphis marsupialis teilen als Synapomorphie die Reduktion der Deutoplasmolyse auf eine generelle Vesikelextrusion. Das Fehlen separierten Dotters in der Oocyte und die resultierende Furchung ohne Dotterextrusion [Renfree and Lewis, Reprod. Fert. Dev. 8, 725,742, 1996] ist eine Apomorphie der Macropodidae. Hiermit hängt möglicherweise die frühe Zusammenlagerung der Blastomeren zusammen [Renfree and Lewis, Reprod. Fert. Dev. 8, 725,742, 1996]. Ein vaskularisiertes Allantochorion und eine Dottersackplazenta können für einen Teil der Ontogenese im Grundplan der Marsupialia angenommen werden. Ob das Allantochorion neben der Respiration auch dem Stoffaustausch diente ist unklar. Die Lesrichtung für den Modus der Plazentation und der Geburt bedarf weiterer Untersuchungen. [source]


    Identification of the intermediate hosts of Habronema microstoma and Habronema muscae under field conditions

    Abstract A polymerase chain reaction (PCR)-based assay was used for the specific detection of Habronema microstoma and Habronema muscae (Nematoda, Spirurida) in order to identify the intermediate hosts of both nematode species under field conditions. A total of 1087 netted and 165 laboratory-bred flies were tested. Flies were identified as Musca domestica Linnaeus 1758, Musca autumnalis De Geer 1776, Haematobia irritans (Linnaeus 1758), Haematobia titillans (De Geer 1907) and Stomoxys calcitrans (Linnaeus 1758) (Muscidae). Genomic DNA was extracted from pools of fly heads, thoraces and abdomens, and 703 samples were subjected to a duplex two-step semi-nested PCR assay to specifically detect diagnostic regions within the ribosomal ITS2 sequence of both H. microstoma and H. muscae. Stomoxys calcitrans specimens were positive for H. microstoma DNA and M. domestica specimens were positive for H. muscae DNA. In particular, PCR-positive samples derived from both farm-netted and laboratory-bred flies. The present study represents the first evidence of the vectorial competence of different fly species as intermediate hosts of Habronema stomachworms under field conditions. We discuss the roles of S. calcitrans and M. domestica in transmitting H. microstoma and H. muscae. [source]
