Selected AbstractsSwarmer cell differentiation in Proteus mirabilisENVIRONMENTAL MICROBIOLOGY, Issue 8 2005Philip N. Rather Summary Under the appropriate environmental conditions, the Gram-negative bacterium Proteus mirabilis undergoes a remarkable differentiation to form a distinct cell type called a swarmer cell. The swarmer cell is characterized by a 20- to 40-fold increase in both cell length and the number of flagella per cell. Environmental conditions required for swarmer cell differentiation include: surface contact, inhibition of flagellar rotation, a sufficient cell density and cell-to-cell signalling. The differentiated swarmer cell is then able to carry out a highly ordered population migration termed swarming. Genetic analysis of the swarming process has revealed that a large variety of distinct loci are required for this differentiation including: genes involved in regulation, lipopolysaccharide and peptidoglycan synthesis, cell division, ATP production, putrescine biosynthesis, proteolysis and cell shape determination. The process of swarming is important medically because the expression of virulence genes and the ability to invade cells are coupled to the differentiated swarmer cell. In this review, the genetic and environmental requirements for swarmer cell differentiation will be outlined. In addition, the role, of, the, differentiated, swarmer, cell, in, virulence and its possible role in biofilm formation will be discussed. [source] New in silico insight into the synteny between rice (Oryza sativa L.) and maize (Zea mays L.) highlights reshuffling and identifies new duplications in the rice genomeTHE PLANT JOURNAL, Issue 3 2004Jérôme Salse Summary A unigene set of 1411 contigs was constructed from 2629 redundant maize expressed sequence tags (ESTs) mapped on the maizeDB genetic map. Rice orthologous sequences were identified by blast alignment against the rice genomic sequence. A total of 1046 (74%) maize contigs were associated with their corresponding homologues in the rice genome and 656 (47%) defined as potential orthologous relationships. One hundred and seventeen (8%) maize EST contigs mapped to two distinct loci on the maize genetic map, reflecting the tetraploid nature of the maize genome. Among 492 mono-locus contigs, 344 (484 redundant ESTs) identify collinear blocks between maize chromosomes 2 and 4 and a single rice chromosome, defining six new collinear regions. Fine-scale analysis of collinearity between rice chromosomes 1 and 5 with maize chromosomes 3, 6 and 8 shows the presence of internal rearrangements within collinear regions. Mapping of maize contigs to two distinct loci on the rice sequence identifies five new duplication events in rice. Detailed analysis of a duplication between rice chromosomes 1 and 5 shows that 11% of the annotated genes from the chromosome 1 locus are found duplicated on the chromosome 5 paralogous counterpart, indicating a high degree of re-organisations. The implications of these findings for map-based cloning in collinear regions are discussed. [source] Fusion of PDGFRB to two distinct loci at 3p21 and a third at 12q13 in imatinib-responsive myeloproliferative neoplasmsBRITISH JOURNAL OF HAEMATOLOGY, Issue 2 2010Claire Hidalgo-Curtis Summary We identified four patients who presented with BCR-ABL1 negative myeloproliferative neoplasms and cytogenetically visible abnormalities of chromosome band 5q31-35. Fluorescence in situ hybridization indicated that the platelet-derived growth factor receptor , gene (PDGFRB) was disrupted in all four cases and 5, rapid amplification of cDNA ends identified in-frame mRNA fusions between PDGFRB and WDR48 (3p21), GOLGA4 (3p21) and BIN2 (12q13). Strikingly, all three genes encode proteins involving intracellular trafficking. Imatinib, a known inhibitor of PDGFR,, selectively blocked the growth of t(3;5) myeloid colonies and produced clinically significant responses in all patients. We conclude that PDGFRB fuses to diverse partner genes in atypical myeloproliferative neoplasms (MPNs). Although very rare, identification of these fusions is critical for proper management of affected individuals. [source] The fabp4 gene of zebrafish (Danio rerio) , genomic homology with the mammalian FABP4 and divergence from the zebrafish fabp3 in developmental expressionFEBS JOURNAL, Issue 6 2007Rong-Zong Liu Teleost fishes differ from mammals in their fat deposition and distribution. The gene for adipocyte-type fatty acid-binding protein (A-FABP or FABP4) has not been identified thus far in fishes. We have determined the cDNA sequence and defined the structure of a fatty acid-binding protein gene (designated fabp4) from the zebrafish genome. The polypeptide sequence encoded by zebrafish fabp4 showed highest identity to the Had -FABP or H6-FABP from Antarctic fishes and the putative orthologs from other teleost fishes (83,88%). Phylogenetic analysis clustered the zebrafish FABP4 with all Antarctic fish H6-FABPs and putative FABP4s from other fishes in a single clade, and then with the mammalian FABP4s in an extended clade. Zebrafish fabp4 was assigned to linkage group 19 at a distinct locus from fabp3. A number of closely linked syntenic genes surrounding the zebrafish fabp4 locus were found to be conserved with human FABP4. The zebrafish fabp4 transcripts showed sequential distribution in the developing eye, diencephalon and brain vascular system, from the middle somitogenesis stage to 48 h postfertilization, whereas fabp3 mRNA was located widely in the embryonic and/or larval central nervous system, retina, myotomes, pancreas and liver from middle somitogenesis to 5 days postfertilization. Differentiation in developmental regulation of zebrafish fabp4 and fabp3 gene transcription suggests distinct functions for these two paralogous genes in vertebrate development. [source] Ultraviolet color-magnitude relation of early-type dwarf galaxies in the Virgo clusterASTRONOMISCHE NACHRICHTEN, Issue 9-10 2009S. Kim Abstract We present ultraviolet (UV) color-magnitude relations (CMRs) for dwarf ellipticals (dEs) and dwarf lenticulars (dS0s) in the Virgo cluster. We used the Galaxy Evolution Explorer (GALEX) GR3 photometric data in far-ultraviolet (FUV) and near-ultraviolet (NUV) passbands combined with SDSS DR5 data. We found that dS0s as well as peculiar dEs (dEs with blue center and disk) found by Lisker et al. (2006a, b) show distinct locus from that of ordinary dEs in UV CMRs. The dS0s and peculiar dEs have bluer UV colors than ordinary dEs at a fixed optical luminosity. Furthermore, UV colors of dEs in outer region of the cluster are systematically bluer than those in inner region. Since UV flux is sensitive to the presence of young stellar population from recent star formation activity, our results suggest that there are different star formation histories among different subsample of early-type dwarf galaxies in Virgo cluster concerning their morphology and environmental effect (© 2009 WILEY-VCH Verlag GmbH & Co.
