Content of polyunsaturated fatty acids (PUFAs) in serum and liver of rats fed restricted diets supplemented with vitamins B2, B6 and folic acid

Abstract The aim of study was to investigate an influence of nutritional deficiency and dietary addition of vit. B2, B6 and folic acid on PUFAs content in rats' serum and liver. Limitation of consumption full value diet to 50% of its previously determined daily consumption, enriched with m/a vitamins, significant decreased of linoleic (LA) and ,-linolenic (ALA) acids as well as distinctly increased arachidonic (AA) and docosahexaenoic (DHA) acids content in serum in 30th day. In 60th day lower content of AA and DHA fatty acids was found. Nutrition with such diet, lasting 90 days caused decrease of LA content and increase of AA. Diet limitation to its 30% of daily consumption decreased of eicosapentaenoic acid (EPA) and DHA in the 30th day, while AA and DHA content was increased in the 60th day. Distinct decrease of AA content and increase of EPA content were found in the 90th day of experiment. Use of diets, with limited consumption to 50% caused increase of LA and ALA acids content while AA and DHA acids content were significantly decreased in the liver, in 90th day. Limited consumption supplemented diet to 30% caused in liver significant decrease of LA and increase of EPA acids content. [source]


Lipid compounds of human Wharton's jelly and their alterations in preeclampsia

Summary Wharton's jelly is a myxomatous substance which surrounds the umbilical cord vessels protecting them against extension, bending, twisting and compression. Very low number of cells in this tissue produce high amounts of extracellular matrix; collagen, hyaluronate and proteoglycans which bind large quantities of peptide growth factors (PGFs). Preeclampsia (the most common pregnancy-associated syndrome) is accompanied by a significant reduction in hyaluronate and a concomitant increase in sulphated glycosaminoglycans/proteoglycans content in Wharton's jelly. Such a phenomenon corresponds to an ,early ageing' of this tissue. We have evaluated the lipid composition of Wharton's jelly and its alteration in preeclampsia. Thin layer chromatography and high-performance liquid chromatography were employed. It was found that Wharton's jelly contains free fatty acids (FFA), mono-, di- and triacylglycerols, free cholesterol and its esters. The characteristic feature is the presence of relatively high amounts of unsaturated fatty acids, including those (C18:2 and C18:3) which are nutritionally essential. Preeclampsia is associated with a slight increase in the total fatty acid content in Wharton's jelly and with marked changes in the proportional relationships between various lipids. A distinct decrease in the amounts of FFA was observed with a concomitant increase in monoacylglycerols and cholesterol esters. At least in some cases the effects exerted by PGFs are mediated by the lipid second messengers. Thus it is possible that alterations in lipid compounds of Wharton's jelly may participate in the deregulation of various cell functions, including overproduction of sulphated glycosaminoglycans or down-regulation of enzymes which participate in their degradation. Lifetime prediction of CAD/CAM dental ceramics,

Abstract The dynamic fatigue method was used to obtain subcritical crack growth parameters n and A for a commercial feldspathic dental porcelain and for a lanthanum-glass-infiltrated alumina glass ceramic. Five stress rates d,/dt ranging from 50 to 0.01 MPa s,1 were applied. The inert strength values were calculated with the use of Weibull statistics and maximum-likelihood approaches for the Weibull parameter m. Strength,probability,time (SPT) diagrams were derived for both materials. The alumina glass composite showed a high fracture strength ,0 (442 MPa) at a failure probability of PF = 63.2% and a high resistance against subcritical crack growth (n = 36.5). The development of strength under fatigue conditions was calculated for exemplary 1 year. The strength of the alumina glass material dropped to 228 MPa within this period. This fact is due to a low content of infiltrated lanthanum glass phase in the composite material (25 wt%). In contrast, for the high-silica-glass-containing porcelain a distinct decrease of strength ,0 from initial 133 to 47 MPa after 1 year was predicted. This, mainly because of a low crack growth resistance (n = 16.8) of the feldspathic porcelain. Much lower strength values were calculated, assuming a failure probability of PF = 5%. 915 MHz microwaves and 50 Hz magnetic field affect chromatin conformation and 53BP1 foci in human lymphocytes from hypersensitive and healthy persons

Abstract We used exposure to microwaves from a global system for mobile communication (GSM) mobile phone (915 MHz, specific absorption rate (SAR) 37 mW/kg) and power frequency magnetic field (50 Hz, 15 ,T peak value) to investigate the response of lymphocytes from healthy subjects and from persons reporting hypersensitivity to electromagnetic field (EMF). The hypersensitive and healthy donors were matched by gender and age and the data were analyzed blind to treatment condition. The changes in chromatin conformation were measured with the method of anomalous viscosity time dependencies (AVTD). 53BP1 protein, which has been shown to colocalize in foci with DNA double strand breaks (DSBs), was analyzed by immunostaining in situ. Exposure at room temperature to either 915 MHz or 50 Hz resulted in significant condensation of chromatin, shown as AVTD changes, which was similar to the effect of heat shock at 41 °C. No significant differences in responses between normal and hypersensitive subjects were detected. Neither 915 MHz nor 50 Hz exposure induced 53BP1 foci. On the contrary, a distinct decrease in background level of 53BP1 signaling was observed upon these exposures as well as after heat shock treatments. This decrease correlated with the AVTD data and may indicate decrease in accessibility of 53BP1 to antibodies because of stress-induced chromatin condensation. Apoptosis was determined by morphological changes and by apoptotic fragmentation of DNA as analyzed by pulsed-field gel electrophoresis (PFGE). No apoptosis was induced by exposure to 50 Hz and 915 MHz microwaves. In conclusion, 50 Hz magnetic field and 915 MHz microwaves under specified conditions of exposure induced comparable responses in lymphocytes from healthy and hypersensitive donors that were similar but not identical to stress response induced by heat shock.
