Distinct contributions of different CD40 TRAF binding sites to CD154-induced dendritic cell maturation and IL-12 secretion

Abstract The mechanisms by which CD40 controls the maturation and antigen presentation functions of dendritic cells (DC) remains largely undefined in this critical cell type. To examine this question, we have employed retroviral transduction of primary bone marrow-derived mouse DC. Mutation of the distinct binding sites for TNF receptor-associated factor 6 (TRAF6) and for TRAF 2, 3, and 5 in the CD40 cytoplasmic domain revealed their independent contributions to DC maturation and activation of NF-,B. In contrast, disruption of the TRAF6 but not the TRAF 2,3,5 binding site markedly decreased IL-12 p40 secretion along with p38 and JNK activation in response to CD154 stimulation. These data document a clear bifurcation of the CD40 signaling cascade in primary DC at the level of thereceptor's two distinct and autonomous TRAF binding sites, and reveal the predominant role of the TRAF6 binding site in CD40-induced pro-inflammatory cytokine production by these cells. Distinct contributions of the amygdala and hippocampus to fear expression

Abstract The present study attempted to distinguish the independent contributions of the amygdala and hippocampus to fear expression. Rhesus monkeys (Macaca mulatta) with bilateral excitotoxic amygdala lesions (n = 4), bilateral excitotoxic hippocampal lesions (n = 8) and unoperated controls (n = 9) were allowed to reach over a neutral junk object or fear-provoking stimulus (i.e., a rubber snake or a jumping rubber spider) to retrieve a food reward. Monkeys were exposed to each stimulus for 30 s. On each trial we recorded the monkey's latency to retrieve the food reward and scored their whole-body reactions to the object. Confirming previous work we found that, relative to controls, both operated groups showed shorter food-retrieval latencies and exhibited fewer defensive and more approach behaviors when exposed to the fear-provoking stimuli. However, only monkeys with amygdala lesions showed an abnormal, excessive visual interest in the snake and spider. By contrast, monkeys with hippocampal lesions displayed behaviors that were unrelated to the presence of the fear stimuli, thereby indicating a lack of interest in, and emotional reactivity towards, the snake and spider. These data show that the hippocampus and amygdala contribute independently to the overall expression of defensive responses. [source]


The network of global corporations and elite policy groups: a structure for transnational capitalist class formation?

This study situates five top transnational policy,planning groups within the larger structure of corporate power that is constituted through interlocking directorates among the world's largest companies. Each group makes a distinct contribution towards transnational capitalist hegemony both by building consensus within the global corporate elite and by educating publics and states on the virtues of one or another variant of the neo,liberal paradigm. Analysis of corporate,policy interlocks reveals that a few dozen cosmopolitans , primarily men based in Europe and North America and actively engaged in corporate management , knit the network together via participation in transnational interlocking and/or multiple policy groups. As a structure underwriting transnational business activism, the network is highly centralized, yet from its core it extends unevenly to corporations and individuals positioned on its fringes. The policy groups pull the directorates of the world's major corporations together, and collaterally integrate the lifeworld of the global corporate elite, but they do so selectively, reproducing regional differences in participation. These findings support the claim that a well,integrated global corporate elite has formed, and that global policy groups have contributed to its formation. Whether this elite confirms the arrival of a transnational capitalist class is a matter partly of semantics and partly of substance. [source]


Jim Bulpitt's Territory and Power in the United Kingdom and Interpreting Political Development: Bringing the State and Temporal Analysis Back In

This article addresses the relative neglect of Territory and Power in informing the study of general state political development, both as a theoretical approach and in its application to the UK. It locates Territory and Power as a distinct contribution to two major schools of comparative research. The first section argues that Territory and Power provided an approach that was part of the intellectual turn during the 1980s to bring the state back into the analysis of politics. The second part argues that Territory and Power should be seen also as a contribution to the intellectual turn since the 1980s towards temporal analysis of political development. On these bases future researchers may find Territory and Power more accessible as a work that they can incorporate in their own research. [source]


Do delay aversion and executive function deficits make distinct contributions to the functional impact of ADHD symptoms?

A study of early academic skill deficits
Background:, The present study examined the distinct properties of executive functioning in relation to ADHD symptoms, as well as functional outcomes associated with ADHD. In line with the dual-pathway model of ADHD, executive functioning and delay aversion were expected to show independent effects on ADHD symptoms. Furthermore, relations to early academic skills were examined, and it was hypothesized that the two processes of the dual-pathway model can be differentiated in terms of their effect on academic skill deficits, such that EF deficits, but not delay aversion, mediate the link between ADHD and academic functioning. Results:, As hypothesized, both EF deficits and delay aversion were independently related to ADHD symptoms. However, when conducting separate analyses for the two ADHD symptom domains, only the effect of EF deficits was independently related to symptoms of inattention, whereas only the effect of delay aversion was independently related to symptoms of hyperactivity/impulsivity. The mediation analysis showed that EF deficits, but not delay aversion, act as a mediator in the relation between symptoms of inattention and both mathematics and language skills. In addition, there was also a significant direct effect of inattention on early academic skills. Conclusions:, The findings of the present study are of importance for current models of heterogeneity in ADHD as they 1) provide further support for the notion that EF deficits and delay aversion are two possible pathways to ADHD, 2) add new interesting knowledge by showing that EF deficits and delay aversion can be differentiated in terms of their relations to the two ADHD symptom domains, and 3) indicate that the two processes of the dual-pathway model can also be differentiated in terms of their effect on functional impairments associated with ADHD. Retrieving meaning after temporal lobe infarction: The role of the basal language area

During speech comprehension the auditory association cortex in the superior temporal cortex is involved in perceptual analysis of the speech signal, whereas the basal language area in the inferior temporal cortex mediates access to word meaning. Disruption of the interaction between the superior and inferior temporal cortices is one factor that may determine recovery from aphasic stroke. We used positron emission tomography to investigate semantic processing within inferior temporal cortex in control subjects and after infarction involving the superior temporal cortex. In the control group, semantic decision making on clear speech activated both anterior fusiform gyri. Chronic aphasic patients were impaired at the task and demonstrated reduced activation within the left anterior fusiform gyrus. A similar pattern of impaired performance and reduced left anterior fusiform gyrus activation was observed when control subjects heard perceptually degraded speech. Performance in both groups predicted activity in the right anterior fusiform gyrus and the temporal poles, where accuracy linearly correlated with activity. These results demonstrate that the function of the basal language area is sensitive to changes in the quality of perceptual input. In addition, different profiles of response observed in each hemisphere suggest distinct contributions of both left and right inferior temporal cortices to the semantic processing of speech.
