Relevance of the genes for bone mass variation to susceptibility to osteoporotic fractures and its implications to gene search for complex human diseases

Abstract We investigate the relevance of the genetic determination of bone mineral density (BMD) variation to that of differential risk to osteoporotic fractures (OF). The high heritability (h2) of BMD and the significant phenotypic correlations between high BMD and low risk to OF are well known. Little is reported on h2 for OF. Extensive molecular genetic studies aimed at uncovering genes for differential risks to OF have focussed on BMD as a surrogate phenotype. However, the relevance of the genetic determination of BMD to that of OF is unknown. This relevance can be characterized by genetic correlation between BMD and OF. For 50 Caucasian pedigrees, we estimated that h2 at the hip is 0.65 (P < 0.0001) for BMD and 0.53 (P < 0.05) for OF; however, the genetic correlation between BMD and OF is nonsignificant (P > 0.45) and less than 1% of additive genetic variance is shared between them. Hence, most genes found important for BMD may not be relevant to OF at the hip. The phenotypic correlation between high BMD and low risk to OF at the hip (approximately ,0.30) is largely due to an environmental correlation (,E = ,0.73, P < 0.0001). The search for genes for OF should start with a significant h2 for OF and should include risk factors (besides BMD) that are genetically correlated with OF. All genes found important for various risk factors must be tested for their relevance to OF. Ideally, employing OF per se as a direct phenotype for gene hunting and testing can ensure the importance and direct relevance of the genes found for the risk of OF. This study may have significant implications for the common practice of gene search for complex diseases through underlying risk factors (usually quantitative traits). Genet. GEOGRAPHY COMPASS (ELECTRONIC), Issue 3 2008

EDITORIAL It is a great privilege to serve as Editor for the Development section of Geography Compass. The journal is an exciting new venture in electronic publishing that aims to publish state-of-the-art peer-reviewed surveys of key contemporary issues in geographical scholarship. As the first Editor of this section, it is my responsibility to establish the key aims and innovations for this section of the journal. These include: publishing reviews of scholarship on topics of contemporary relevance that are accessible and useful to researchers, teachers, students and practitioners; developing the range of topics covered across the spectrum of development geography; helping to set agendas in development geography by identifying gaps in existing empirical and conceptual research; commissioning articles from both established and graduate/early career researchers who are working at the frontiers of development geography; and communicating the distinctiveness of Geography Compass. Part of this distinctiveness is in publishing articles that are both of scholarly excellence and accessible to a wide audience. The first volume of Geography Compass was published in 2007, covering a wide range of topics (e.g. migration, children, technology, grassroots women's organizations, civil society, biodiversity, tourism, inequality, agrarian change, participatory development, disability, spirituality) in a number of specific geographical areas (e.g. Africa/southern Africa, Caribbean, China, Peru). Forthcoming in 2008/2009 are articles on the Gambia, Latin America, the Philippines, Southeast Asia, Bangladesh and South Africa, focusing on topics such as food security, comparative post-socialism, foreign aid and fair trade. Building on these diverse and excellent articles, I plan to communicate the distinctiveness of Development in a number of ways. First, I encourage an ecumenical approach to the notion of ,development geography' and welcome contributions from scholars across a range of social science disciplines whose work would be useful to a geography audience. This is important, not least because both development and geography, in disciplinary terms, are largely European inventions. Many scholars in Latin America, Africa and Asia, for example, do not refer to themselves as either development specialists or geographers but are producing important research in areas of direct relevance to students and researchers of ,development geography'. As the first editions illustrate, I also seek to publish articles that reflect ,development' in its broadest sense, encompassing economic, (geo)political, social, cultural and environmental issues. 2008 will be an interesting year for development, with a number of important issues and events shaping discourse and policy. These include: the Beijing Olympics and increasing focus on China's role in international development; political change in a number of African countries (Kenya, Zimbabwe, South Africa); the US presidential elections and potential shifts in policy on climate change, trade and security; the impacts of the Bali roadmap on climate change in the current economic context; the increasing number of impoverished people in Asia (notably China and India), sub-Saharan Africa and Latin America (notably Brazil) that even the World Bank has acknowledged; the implications of the increasing role of philanthropic foundations (e.g. the Gates Foundation and those emerging in India and Russia) in international development. I hope to see some of these issues covered in this journal. Second, I am keen to break down the association between ,development' and parts of the world variously categorized as ,Third World', ,Global South' or ,Developing World' by publishing articles that cut across North and South, East and West. The intellectual and disciplinary practices within (Western) geography that separate those researching issues in the South and post-socialist contexts from those researching similar issues in advanced capitalist economies are, it seems, no longer sustainable or sensible. Moreover, while studies of transnational and ethical trade, neoliberalism, household economies and ,commodity chains', for example, incorporate a multitude of case studies from across the world, these tend to be understood through conceptual lenses that almost always have their theoretical antecedents in Western theorization. The notion of ,learning from' debates, policy and practice in other parts of the world is still relatively alien within the discipline. There are thus issues in how we research and teach ethically and responsibly in and about different parts of the world, and in which this journal might make a contribution. Third, and related, part of my responsibility is to ensure that Compass reflects the breadth of debate about ,development' by publishing articles written by a truly international range of scholars. This has proved to be a challenge to date, in part reflecting the newness of the journal and the difficulties posed by English language publication. However, an immediate aim is to publish the work and ideas of scholars based outside of Anglophone contexts, in the Global South and in post-socialist contexts, and to use international referees who are able to provide valuable commentaries on the articles. A longer-term aim is to also further internationalize the Editorial Board. Currently, one-third of the Editorial Board is non-UK and I plan to increase this to at least 50% in future. Fourth, I plan to ensure that the Development section takes full advantage of electronic publication and the opportunities this offers. Thus, while I am keen to retain a word limit in the interest of publishing accessible articles, the lack of constraint regarding page space enables authors to include a wide range of illustrative and other material that is impossible in print journals. I plan to encourage authors to make greater use of visual materials (maps, photographs/photo-essays, video, sound recordings, model simulations and datasets) alongside text as well as more innovative forms of presentation where this might be appropriate. Finally, in the coming year, I intend to work more closely with other Compass section Editors to realize the potential for fostering debate that cuts across subdisciplinary and even disciplinary boundaries. The journal publishes across the full spectrum of the discipline and there is thus scope for publishing articles and/or special issues on development-related topics that might best be approached through dialogue between the natural and social sciences. Such topics might include resources (e.g. water, oil, bio-fuels), hazard and risk (from environmental issues to human and state security), and sustainability and quality of life (planned for 2008). Part of the distinctiveness of Compass is that electronic-only publication ensures that articles are published in relatively quick time , in some cases less than 3 months from initial submission to publication. It thus provides an important outlet for researchers working in fast-changing contexts and for those, such as graduate and early-career researchers, who might require swift publication for career purposes. Of course, as Editor I am reliant on referees both engaging with Manuscript Central and providing reports on articles in a relatively short space of time to fully expedite the process. My experience so far has been generally very positive and I would like to thank the referees for working within the spirit of the journal. Editing a journal is, of course, a collaborative and shared endeavour. The Development Editorial Board has been central to the successful launch of Development by working so generously to highlight topics and potential authors and to review articles; I would like to take this opportunity to thank Tony Bebbington, Reg Cline-Cole, Sara Kindon, Claire Mercer, Giles Mohan, Warwick Murray, Richa Nagar, Rob Potter, Saraswati Raju, Jonathan Rigg, Jenny Robinson and Alison Stenning. The Editors-in-Chief , Mike Bradshaw and Basil Gomez , have provided invaluable advice while adding humour (and colour) to the editorial process. Political and social drivers for access to the countryside: the need for research on birds and recreational disturbance

The introduction of a statutory right of access to open country and registered common land in England and Wales in 2005 was a major milestone in a campaign traceable to the 19th century, with views strongly polarized between social classes and political parties, and between land owners and campaigners. More recently, access has also been recognized as a factor contributing to quality-of-life, public health, social diversity and rural economic issues. The mapping of access land revealed that 55% of it is also designated Site of Special Scientific Interest, where wildlife is legally conserved. This has generated a need to assess the implications of access in each case, and take measures to ensure nationally and internationally important features are protected, drawing on sound scientific principles. Early research, although competently undertaken, often failed to address population-scale effects significant at the designated-site level, enabling disputes and polarized ,beliefs' to be articulated. Hence, in addition to drawing up formal and transparent procedures for evaluating impacts and resolving difficulties, funds were released and a major programme initiated, commissioning applied research of direct relevance to the implementation of the legislation. This has pushed forward the boundaries of knowledge in a field which is both difficult and expensive to study. By gradually replacing ,belief' with evidence, this represents a case study in resolving environmental disputes. [source]


Large-scale ecology and hydrology: an introductory perspective from the editors of the Journal of Applied Ecology

1. Five key features characterize large-scale factors in ecology: (a) they incorporate some of the most major of all ecological phenomena , the ranges of organisms, patterns of diversity, variations in ecosystem character and environmental processes such as climate, biogeochemical cycles, dispersal and migration; (b) they involve interactions across scales through both top-down and bottom-up processes; (c) they are multifaceted, and hence require an interdisciplinary perspective; (d) they reflect the cumulative effects of anthropogenic change across all scales, and so have direct relevance to environmental management; (e) they invariably exceed the range of classical ecological experiments, and so require alternative approaches to hypothesis testing. 2. Against this background, a recent research initiative on large-scale ecology and hydrology was funded jointly by the Natural Environment Research Council (NERC) and the Scottish Executive Rural Affairs Department (SERAD). Outputs from this programme are reported in this special issue of the Journal of Applied Ecology, and they illustrate some of the ecological research that is currently in progress in the UK at large spatio-temporal scales. 3. The spatial scales investigated in the papers range from hectares to whole continents, and much of the work reported here involves modelling. Although the model outputs are intrinsically valuable, several authors express the need for improved validation and testing. We suggest that this is an area requiring much development, and will need considerable innovation due to the difficulties at the scales involved (see 1d). Possible methods include: model applications to new circumstances; large-scale environmental manipulations; large-scale surveys that mimic experimental protocols; support from process studies at smaller scales. These alternatives are not mutually exclusive, and all can allow robust hypothesis testing. 4. Much of the work reported here is interdisciplinary linking, for example, geographical, mathematical, hydrological, hydrochemical and ecological concepts (see 1c). We suggest that even stronger links between environmental disciplines will further aid large-scale ecological research. 5. Most important in the context of the Journal of Applied Ecology, the work reported in this issue reveals that large-scale ecology already has applied value. Sectors benefiting include the conservation of biodiversity, the control of invasive species, and the management of land and water resources. 6. Large-scale issues continue to affect many applied ecologists, with roughly 30,40% of papers published in the Journal of Applied Ecology typically confronting such problems. This special issue adds to the growing body of seminal contributions that will add impetus to further large-scale work. Moreover, occurring in a period when other areas of biology are increasingly reductionist, this collection illustrates that, at least with respect to large-scale environmental problems, ecology still holds centre ground. Large-scale processes in ecology and hydrology

1. Several papers published in the 1980s stressed the importance of scaling issues, the inter-relatedness of patterns and processes at different scales of time and space, to our understanding of ecological systems. Scaling issues are of major theoretical interest and increasingly are of considerable applied importance. 2. In recognition of this, the Natural Environment Research Council, in partnership with the Scottish Executive Rural Affairs Department, funded a Thematic Programme entitled ,Large-scale Processes in Ecology and Hydrology'. The principal aim of this Programme was to integrate recent major developments in information resources and technologies with current theory in order to improve understanding of large-scale patterns and processes and their relationship to patterns and processes at smaller scales. 3. The Thematic Programme, which ran from 1995 until 1999, funded six research projects that have generated a large body of published papers. This volume, dedicated to the findings of the Programme, brings together outputs from all six projects with the aim of ensuring a rapid and widespread dissemination of the Programme's findings. A brief résumé of each of the papers is presented. 4. The papers in this volume cover a wide variety of subjects ranging from ions to the flora and fauna of the United Kingdom. Nonetheless, each study has sought in various ways to quantify observed spatio-temporal patterns at a range of scales, to determine whether those patterns are consistent across scales and to identify the interactions between small-scale patterns and processes and those at larger scales. The importance of the spatial and temporal scales at which studies are conducted, the key role played by dispersal in spatial population dynamics, and the diversity of ways in which large-scale patterns and processes relate to those at smaller scales are highlighted in many of the papers. 4. All of the papers presented here have direct relevance to applied issues. These issues are diverse and include the control of invasive alien species, the conservation of declining, threatened or endangered species, the development of survey techniques, strategies for farmland, woodland and forestry management, and the assessment of pollution sensitivity. Thus, the Thematic Programme has addressed issues of considerable theoretical interest and has at the same time generated results and predictive models that are of considerable practical and policy relevance. The Impact of Performance Level Misclassification on the Accuracy and Precision of Percent at Performance Level Measures

No Child Left Behind (NCLB) performance mandates, embedded within state accountability systems, focus school AYP (adequate yearly progress) compliance squarely on the percentage of students at or above proficient. The singular importance of this quantity for decision-making purposes has initiated extensive research into percent proficient as a measure of school quality. In particular, technical discussions have scrutinized the impact of sampling, measurement, and other sources of error on percent proficient statistics. In this article, we challenge the received orthodoxy that measurement error associated with individual students' scores is inconsequential for aggregate percent proficient statistics. Synthesizing current classification accuracy research with techniques from randomized response designs, we establish results which specify the extent to which measurement error,manifest as performance level misclassifications,produces bias and increases error variability for percent at performance level statistics. The results have direct relevance for the design of coherent and fair accountability systems based upon assessment outcomes. [source]


Rice versus fish revisited: On the integrated management of floodplain resources in Bangladesh

Abstract Disproportionately little attention has been paid to the dry season trade-off between rice and (inland capture) fish production on the floodplains of Bangladesh, compared to the same trade-off during the flood season. As the rural economy grows increasingly dominated by dry-season irrigated rice production, and floodplain land and water come under ever-increasing pressure during the dry winter months, there is an urgent need to focus attention on these dry months that are so critical to the survival and propagation of the floodplain resident fish, and to the poor people that depend on these fish for their livelihood. This article examines three important dry-season natural resource constraints to floodplain livelihoods in Bangladesh, and finds a common factor at the heart of all three: rice cultivation on lands at low and very low elevations. The article articulates the system interlinkages that bind these constraints and the long-run trend towards irrigated rice cropping on lower-lying lands, and suggests a management approach based on locally tailored strategies to arrest this trend. Apart from its direct relevance to the floodplains of Bangladesh, which support more than 100 million people, these lessons have relevance for river floodplain systems elsewhere in the developing world, notably the Mekong Delta. Searle, Rationality, and Social Reality

ABSTRACT . This paper makes a critical assessment of some contributions of John Searle of direct relevance to social science,his theory of rationality and his theory of institutions. The former is criticized for being able to account for how people can act for "external reasons," as opposed to their desires, only in cases where an obligation derives from a promise; a rival account is much more general. The latter is criticized on three grounds: (1) his theory of institutions can only account for a narrow range of social reality, that deriving from some kind of "enactment"; (2) his notion of "collective intentionality" is unnecessary to explain institutions and is introduced only so that Searle can stay within an individualistic, Cartesian theory of the mind, something of no interest to social scientists; (3) his account implies that if an institution exists, "we collectively accept it," whereas it may in fact not be accepted, but merely tolerated or submitted to. Monitoring chloramines and bromamines in a humid environment using selected ion flow tube mass spectrometry

The selectivity and sensitivity of selected ion flow tube mass spectrometry (SIFT-MS) for individual breath analysis of haloamines has been improved by heating the flow tube in a commercial instrument to around 106°C. Data is presented showing the marked reduction in the number density of water clusters of product ions of common breath metabolites that are isobaric with the product ions from monochloramine and monobromamine that are used to monitor the haloamine concentrations. These results have direct relevance to the real-time monitoring of chloramines in drinking water, swimming pools and food processing plants. However, once the isobaric overlaps from water cluster ions are reduced at the higher temperatures, there is no conclusive evidence showing the presence of haloamines on single breath exhalations in the mid parts per trillion range from examination of the breaths of volunteers. Reconstruction and Morphometric Analysis of the Nasal Airway of the Dog (Canis familiaris) and Implications Regarding Olfactory Airflow

Abstract The canine nasal airway is an impressively complex anatomical structure, having many functional roles. The complicated branching and intricate scrollwork of the nasal conchae provide large surface area for heat, moisture, and odorant transfer. Of the previous anatomical studies of the canine nasal airway, none have included a detailed rendering of the maxilloturbinate and ethmoidal regions of the nose. Here, we present a high-resolution view of the nasal airway of a large dog, using magnetic resonance imaging scans. Representative airway sections are shown, and a three-dimensional surface model of the airway is reconstructed from the image data. The resulting anatomic structure and detailed morphometric data of the airway provide insight into the functional nature of canine olfaction. A complex airway network is revealed, wherein the branched maxilloturbinate and ethmoturbinate scrolls appear structurally distinct. This is quantitatively confirmed by considering the fractal dimension of each airway, which shows that the maxilloturbinate airways are more highly contorted than the ethmoidal airways. Furthermore, surface areas of the maxilloturbinate and ethmoidal airways are shown to be much different, despite having analogous physiological functions. Functionally, the dorsal meatus of the canine nasal airway is shown to be a bypass for odorant-bearing inspired air around the complicated maxilloturbinate during sniffing for olfaction. Finally, nondimensional analysis is used to show that the airflow within both the maxilloturbinate and ethmoturbinate regions must be laminar. Current and emerging concepts in tumour metastasis,

Abstract Disseminated cancer accounts for most deaths due to malignancy. Despite this, research has focused predominantly on tumour development and progression at the primary site. Recently, attention has shifted towards the field of tumour metastasis. Several new and exciting concepts that have emerged in the past few years may shed light on this complex area. The established canonical theory of tumour metastasis, as a process emerging from a stepwise accumulation of genetic events fuelled by clonal evolution, has been challenged. New evidence suggests that malignant cells can disseminate at a much earlier stage than previously recognized in tumourigenesis. These findings have direct relevance to clinical practice and shed new light on tumour biology. Gene-profiling studies support this theory, suggesting that metastatic ability may be an innate property shared by the bulk of cells present early in a developing tumour mass. There is a growing recognition of the importance of host factors outside the primary site in the development of metastatic disease. The role of the ,pre-metastatic niche' is being defined and with this comes a new understanding of the function of bone marrow-derived progenitor cells in directing the dissemination of malignant cells to distant sites. Current research has highlighted the crucial roles played by non-neoplastic host cells within the tumour microenvironment in regulating metastasis. These new concepts have wide-ranging implications for our overall understanding of tumour metastasis and for the development of cancer therapeutics. Smoking and anaesthesia: the pharmacological implications

Summary Anaesthetists are generally familiar with the peri-operative implications of cigarette smoking. Although there are a number of publications dealing with the wider pharmacological implications of cigarette smoking, the specific interactions which are of direct relevance to anaesthetists are less well known. This review gives an overview of those interactions which are of clinical relevance to anaesthetists and provides, where possible, an explanation of the mechanism. Recent improvements in the understanding of biotransformation of drugs, including streamlining of the classification of hepatic enzymes has allowed better understanding of drug interactions and enables the mechanistic prediction of those involving new drugs. Vine vigour effects on leaf gas exchange and resource utilisation

Abstract Background and Aims:, Variability in vine vigour is a commonly observed feature in vineyard blocks. Although this aspect is well recognised among viticulturalists, impacts of vigour variability on vine function and resource use efficiency are seldom considered in management decision-making. This study examined influences of vine vigour variability on vine physiological performance and resource use in a commercially managed vineyard block. Methods and Results:, The vineyard block was divided into different vigour zones using plant cell density data derived from aerial multi-spectral imaging of the study block. Gas exchange and under-vine soil moisture status were measured in some of the vigour zones. Low vigour vines had consistently lower photosynthesis and stomatal conductance rates than high vigour vines. The differences were evident both during soil-drying and rewetting. Similarly, irrigation water utilisation was markedly lower in low than in high vigour vines. Vine fruit yield was linearly and positively related to vines' capacity to utilise applied water. Conclusions:, Vigour has considerable influence on vine physiological performance and on resource utilisation capacity. Uniform irrigation application to a block with variable vigour (which is the norm currently) leads to spatially inefficient resource use, and consequently to a marked within-block variation in irrigation water productivity. Significance of the Study:, The demonstration that variation in vigour has a corresponding effect on resource utilisation has direct relevance for managing vineyard inputs such as irrigation or fertilisers. A phytase enzyme-based biochemistry practical particularly suited to students undertaking courses in biotechnology and environmental science

Abstract Courses in introductory biochemistry invariably encompass basic principles of enzymology, with reinforcement of lecture-based material in appropriate laboratory practicals. Students undertaking practical classes are more enthusiastic, and generally display improved performance, when the specific experiments undertaken show direct relevance to their core degree area. Herein is described an enzyme-based practical of particular relevance to students undertaking primary degrees in biotechnology, environmental science, nutrition, as well as animal and veterinary science. The practical entails assay of phytase, an enzyme that is now routinely included in monogastric animal feed in order to ameliorate the negative nutritional and environmental consequences of its substrate, dietary phytic acid. The practical is technically straightforward, requiring relatively basic laboratory equipment and reagents that are both easily obtained and relatively inexpensive. To be an effective digestive supplement, the enzyme must display a combination of physicochemical properties, which provide sufficient scope for an interesting and multifaceted post-laboratory discussion. Determining the skills for child protection practice: emerging from the quagmire!

Abstract This paper, the second in a series of two, presents and analyses the results of an extended literature review undertaken for a Master's dissertation, the purpose of which was to establish the evidence base for the learning and teaching of skills for child protection practice. The review, discussed in the previous paper, was carried out in two stages. An initial search yielded a very small number of studies of direct relevance but they provided the trigger and the key words for a second search. Many potentially useful areas had to be omitted from the second stage because of limited resources, but the two stages in the search generated large amounts of material, much of which was indirectly relevant to child protection practice. The review did not achieve the original aim of determining skills that are identified through research and other evidence as being essential for child protection practice, but it was possible to draw some conclusions. For example, evidence was found of the importance of a range of communication skills, whether these be child focused, carer focused or inter-professional. Skills in managing conflict and challenge were found to be crucial, with the importance of role clarification being acknowledged. There was some evidence of the use of decision-making skills and problem-solving skills, but little research that explored procedural skills. Policy interpretation network on children's health and environment

Abstract Aim: The main objective of PINCHE is to provide policy recommendations aiming at protecting children's health and environment based on completed scientific research. The project focused on four themes: indoor and outdoor air pollutants, carcinogens, neurotoxicants, and noise. The data were evaluated in workpackages on exposure assessment, epidemiology, toxicology, and risk and health impact assessment. The data were analysed according to a framework of questions. The workpackage on socioeconomic factors studied the influence of socioeconomic status on exposures and on health effects. In the workpackage on science-policy interface, recommendations on how to improve children's environmental health were formulated. Results: The policy recommendations resulting from the analysis were grouped according to relevant policy levels: European Commission or the European Parliament, member states and other stakeholders at regional or local level. These recommendations are general guidelines for taking action. Regional differences and variation must be reflected when policy is actually implemented. In addition, recommendations related to education and personal behaviour are presented in the reports. Conclusions: The policy recommendations are important input for policy advisers, policy makers and public health authorities at all policy levels. The recommendations are also of direct relevance to interest groups, such as environmental NGOs including child health and advocacy groups. The policy recommendations for each policy level were prioritized. High priorities were given to reduce exposure to environmental tobacco smoke, transport related air pollution, indoor air and mercury. [source]
