Selected AbstractsSensitivity Analysis for Principal Stratum Direct Effects, with an Application to a Study of Physical Activity and Coronary Heart DiseaseBIOMETRICS, Issue 2 2009Arvid Sjölander Summary In many studies, the aim is to learn about the direct exposure effect, that is, the effect not mediated through an intermediate variable. For example, in circulation disease studies it may be of interest to assess whether a suitable level of physical activity can prevent disease, even if it fails to prevent obesity. It is well known that stratification on the intermediate may introduce a so-called posttreatment selection bias. To handle this problem, we use the framework of principal stratification (Frangakis and Rubin, 2002, Biometrics58, 21,29) to define a causally relevant estimand,the principal stratum direct effect (PSDE). The PSDE is not identified in our setting. We propose a method of sensitivity analysis that yields a range of plausible values for the causal estimand. We compare our work to similar methods proposed in the literature for handling the related problem of "truncation by death." [source] Senior Management Support in the New Product Development ProcessCREATIVITY AND INNOVATION MANAGEMENT, Issue 4 2001Jorge Gomes This paper studies the relationship between senior management support to new product development activities by means of a quantitative and qualitative analysis of questionnaire and interview data collected in the United Kingdom and the Netherlands. The quantitative analysis showed that there is a small to medium association between senior management support to new product development and project performance in the dimensions of time, cost, and end product quality. The qualitative analysis suggests that these weak links could be explained by separating the influence of senior management support on new product development activities into direct and indirect effects. Direct effects include issues such as the use of multifunctional senior teams and process champions, whereas indirect effects include issues such as organization mission and goals, and learning and knowledge management systems. Cruickshank ABSTRACT Aims To examine the literature regarding clinical pharmacokinetics, direct effects and adverse clinical outcomes associated with methamphetamine use. Methods Relevant literature was identified through a PubMed search. Additional literature was obtained from relevant books and monographs. Findings and conclusions The mean elimination half-life for methamphetamine is approximately 10 hours, with considerable inter-individual variability in pharmacokinetics. Direct effects at low-to-moderate methamphetamine doses (5,30 mg) include arousal, positive mood, cardiac stimulation and acute improvement in cognitive domains such as attention and psychomotor coordination. At higher doses used typically by illicit users (,50 mg), methamphetamine can produce psychosis. Its hypertensive effect can produce a number of acute and chronic cardiovascular complications. Repeated use may induce neurotoxicity, associated with prolonged psychiatric symptoms, cognitive impairment and an increased risk of developing Parkinson's disease. Abrupt cessation of repeated methamphetamine use leads to a withdrawal syndrome consisting of depressed mood, anxiety and sleep disturbance. Acute withdrawal lasts typically for 7,10 days, and residual symptoms associated with neurotoxicity may persist for several months. [source] Copper-induced modifications of the trophic relations in riverine algal-bacterial biofilmsENVIRONMENTAL TOXICOLOGY & CHEMISTRY, Issue 6 2003Christiane Barranguet Abstract The effects of copper (Cu) on photosynthetic riverine biofilms were studied in artificial stream channels. Direct effects on the composition and functioning of the biofilms were investigated using plant pigments, community-level physiological profiles (CLPP), and pulse-amplitude-modulated (PAM) fluorescence. Copper caused a significant reduction of microalgal biomass and induced a shift in the population from diatoms to cyanobacteria. However, a decrease in biomass indicated that the replacement of species was not totally effective to counteract the toxic effects of Cu. A direct effect of Cu could also be shown in the bacterial community, and, furthermore, changes in the CLPP could be related to the Cu treatment. Copper-exposed biofilms lost the capacity to use between 11 and 15% of the substrates, but many of the remaining capacities became more robust, indicating an increased Cu tolerance due to the exposure. The change in the biofilm microbial composition points to the indirect effects of Cu on biofilms due to the close interdependence between biofilm autotrophic and heterotrophic compartments. Grazing by snails, which appeared to be an important factor structuring biofilms without any Cu addition, had a very minor effect on Cu-exposed biofilms. Although grazing changed the bacterial composition, its effects were not detected either on the algal community or on the biofilm community tolerance to Cu. [source] Cyclosporine Induces Epileptiform Activity in an In Vitro Seizure ModelEPILEPSIA, Issue 3 2000Michael Wong Summary: Purpose: Cyclosporine (CSA) toxicity represents a common cause of seizures in transplant patients, but the specific mechanisms by which CSA induces seizures are unknown. Although CSA may promote seizure activity by various metabolic, toxic, vascular, or structural mechanisms, CSA also has been hypothesized to modulate neuronal excitability directly. The objective of this study was to determine if CSA exerts direct epileptogenic actions on neurons in an in vitro seizure model. Methods: Combined hippocampal-entorhinal cortex slices from juvenile rats were exposed directly to artificial cerebro-spinal fluid (ACSF) containing either (a) 1.0 mM magnesium sulfate (control), (b) 1.0 mM sodium sulfate (low-magnesium), or (c) 1.0 mM magnesium sulfate + CSA (1,000,10,000 ng/ml). Spontaneous and evoked extracellular field potentials were recorded simultaneously from the dentate gyrus (DG) and CA3 hippocampal regions. Evoked synaptic responses were elicited by stimulation of the entorhinal cortex/perforant pathway. Results: CSA elicited spontaneous or stimulation-induced epileptiform activity in the DG or CA3 region of ,40% of slices, consisting of brief repetitive "interictal" discharges or prolonged stereotypical "ictal" discharges. Mean latency to epileptiform activity was ,100 min after onset of CSA application. The interictal discharges were inhibited by the non-NMDA antagonist, NBQX. Similar epileptiform activity was observed in low-magnesium ACSF without CSA. In control ACSF alone, epileptiform activity was not seen, except for rare spontaneous potentials in the DG. Conclusions: Direct effects of CSA on neuronal excitability and synaptic transmission may contribute to seizures seen in clinical CSA neurotoxicity. Hailer Abstract Acute CNS lesions lead to neuronal injury and a parallel glial activation that is accompanied by the release of neurotoxic substances. The extent of the original neuronal damage can therefore be potentiated in a process called secondary damage. As astrocytes are known to secrete immunomodulatory and neuroprotective substances, we investigated whether astrocytic factors can attenuate the amount of neuronal injury as well as the degree of microglial activation in a model of excitotoxic neurodegeneration. Treatment of organotypic hippocampal slice cultures with N-methyl- d -aspartate (NMDA) resulted in a reproducible loss of viable granule cells, partial destruction of the regular hippocampal cytoarchitecture and a concomitant accumulation of amoeboid microglial cells at sites of neuronal damage. Astrocyte-conditioned media reduced the amount of NMDA-induced neuronal injury by 45.3%, diminished the degree of microglial activation and resulted in an improved preservation of the hippocampal cytoarchitecture. Transforming growth factor (TGF)-, failed to act as a neuroprotectant and even enhanced the amount of neuronal injury by 52.5%. Direct effects of astrocytic factors on isolated microglial cells consisted of increased microglial ramification and down-regulated expression of intercellular adhesion molecule-1, whereas incubation with TGF-, had no such effects. In summary, our findings show that hitherto unidentified astrocyte-derived factors that are probably not identical with TGF-, can substantially enhance neuronal survival, either by eliciting direct neuroprotective effects or by modulating the microglial response to neuronal injury. [source] Actions of Arachidonic Acid on Contractions and Associated Electrical Activity in Guinea-Pig Isolated Ventricular MyocytesEXPERIMENTAL PHYSIOLOGY, Issue 4 2001M. A. Mamas The actions of arachidonic acid (AA) were investigated in guinea-pig isolated ventricular myocytes. Exposure of myocytes to 10 ,M AA reduced the amplitude of contractions and calcium transients accompanying action potentials at a frequency of 1 Hz. AA (10 ,M) also reduced the amplitude of calcium currents recorded under voltage-clamp conditions. The suppression of contraction by AA was not prevented by either 10 ,M trihydroindomethicin (to inhibit cyclo-oxygenase) or 10 ,M ETYA (5,8,11,14-eicosatetraynoic acid, to inhibit AA metabolising enzymes), showing that the actions of AA appeared not to be mediated by these metabolites. The reduction of contraction by 10 ,M AA was also not prevented by the protein kinase C inhibitor, Ro31-8220 (1 ,M), showing that this pathway appeared not to be required for the observed effect. Direct effects of AA may be involved. A further action of 10 ,M AA was to suppress spontaneous electrical activity induced by either the ,-adrenergic agonist isoprenaline or the Na+ pump inhibitor, ouabain. This effect of AA on spontaneous activity might be associated with the observed reduction of calcium entry through L-type calcium channels, although additional effects of AA on calcium release from the sarcoplasmic reticulum might also be involved. Scott The catch of Atlantic salmon Salmo salar L., from ocean and coastal fisheries since 1960 is described and the decline starting in 1973 noted. This decline continued despite cessation of some fisheries, and evidence indicates a marine locus for the decline. Oceanic pollution has received little attention, and ozone loss has caused an increase in UV radiation reaching the surface of the North Atlantic. Direct effects of this increase include negative impacts on the food chain. Photo-induced toxicity of polycyclic aromatic hydrocarbons (PAH) is a possible effect, but concentrations of these compounds in oceanic waters are low. Chlorinated organic compounds are widely distributed in the North Atlantic, and their persistence, lipophilicity, bioaccumulation potential and toxicity represent a potential hazard to salmon. The suggested causes of decline are likely to be complementary rather than exclusive. PEARCE-HIGGINS Abstract Understanding the mechanisms by which climate change will affect animal populations is vital for adaptive management. Many studies have described changes in the timing of biological events, which can produce phenological mismatch. Direct effects on prey abundance might also be important, but have rarely been studied. We examine the likely importance of variation in prey abundance in driving the demographics of a European golden plover (Pluvialis apricaria) population at its southern range margin. Previous studies have correlated plover productivity with the abundance of their adult cranefly (Tipulidae) prey, and modelled the phenology of both plover breeding and cranefly emergence in relation to temperature. Our analyses demonstrate that abundance of adult craneflies is correlated with August temperature in the previous year. Correspondingly, changes in the golden plover population are negatively correlated with August temperature 2 years earlier. Predictions of annual productivity, based on temperature-mediated reductions in prey abundance, closely match observed trends. Modelled variation in annual productivity for a future scenario of increasing August temperatures predicts a significant risk of extinction of the golden plover population over the next 100 years, depending upon the magnitude of warming. Direct effects of climate warming upon cranefly populations may therefore cause northward range contractions of golden plovers, as predicted by climate envelope modelling. Craneflies are an important food source for many northern and upland birds, and our results are likely to have wide relevance to these other species. Research into the potential for habitat management to improve the resilience of cranefly populations to high temperature should be an urgent priority. [source] Structural response of Caribbean dry forests to hurricane winds: a case study from Guánica Forest, Puerto RicoJOURNAL OF BIOGEOGRAPHY, Issue 3 2006Skip J. Van Bloem Abstract Aim, Tropical dry forests in the Caribbean have an uniquely short, shrubby structure with a high proportion of multiple-stemmed trees compared to dry forests elsewhere in the Neotropics. Previous studies have shown that this structure can arise without the loss of main stems from cutting, grazing, or other human intervention. The Caribbean has a high frequency of hurricanes, so wind may also influence forest stature. Furthermore, these forests also tend to grow on soils with low amounts of available phosphorus, which may also influence structure. The objective of this study was to assess the role of high winds in structuring dry forest, and to determine whether soil nutrient pools influence forest response following hurricane disturbance. Location, Guánica Forest, Puerto Rico. Methods, Over 2000 stems in five plots were sampled for hurricane effects within 1 week after Hurricane Georges impacted field sites in 1998. Sprout initiation, growth, and mortality were analysed for 1407 stems for 2 years after the hurricane. Soil nutrient pools were measured at the base of 456 stems to assess association between nutrients and sprout dynamics. Results, Direct effects of the hurricane were minimal, with stem mortality at < 2% and structural damage to stems at 13%, although damage was biased toward stems of larger diameter. Sprouting response was high , over 10 times as many trees had sprouts after the hurricane as before. The number of sprouts on a stem also increased significantly. Sprouting was common on stems that only suffered defoliation or had no visible effects from the hurricane. Sprout survival after 2 years was also high (> 86%). Soil nutrient pools had little effect on forest response as a whole, but phosphorus supply did influence sprout dynamics on four of the more common tree species. Main conclusions, Hurricanes are able to influence Caribbean tropical dry forest structure by reducing average stem diameter and basal area and generating significant sprouting responses. New sprouts, with ongoing survival, will maintain the high frequency of multi-stemmed trees found in this region. Sprouting is not limited to damaged stems, indicating that trees are responding to other aspects of high winds, such as short-term gravitational displacement or sway. Soil nutrients play a secondary role in sprouting dynamics of a subset of species. The short, shrubby forest structure common to the Caribbean can arise naturally as a response to hurricane winds. Barnes Abstract There is a complex interrelationship between upper shore hermit crabs (such as Coenobita sp. and Clibanarius sp.), coastal human populations and mangrove forests in Mozambique. The abundance, activity, shell selection and behaviour of three species of hermit crab are related to the level of mangrove cover. With increased density of mangrove trees, the study species of hermit crab changed in abundance, tended to become diurnal, spent more time feeding and were clustered in larger groups when doing so, and selected longer spired shells. All five of the same variables are also linked to the proximity and activity of humans through both direct and indirect actions. Direct effects included a tendency to nocturnal activity with proximity to human activity; indirect effects included increased and more clumped food supplies, and shell middens from intertidal harvesting and deforestation. Mangroves are important to local human populations as well as to hermit crabs, for a wide variety of (similar) reasons. Mangroves provide storm shelter, fisheries and fishery nursery grounds for adjacent human settlements, but they also harbour mosquito populations and their removal provides valuable building materials and fuel. Hermit crabs may be useful (indirectly) to coastal human populations by being a source of food to certain commercial species, and by quickly consuming rotting/discarded food and faeces (thereby reducing disease and pests). They can also cause minor problems to coastal human populations because they use shells of (fisheries) target mollusc species and can be more abundant than the living molluscs, thereby slowing down effective hand collection through confusion over identification. The mixture of positive and negative attributes that the three groups impart to each other in the Quirimba Archipelago, northern Mozambique, is discussed. L'abondance, l'activité, le choix de la coquille et le comportement de trois espèces de Bernard-l'Ermite sont liés au degré de couverture de la mangrove. Lorsque la densité des arbres de la mangrove augmente, l'abondance des espèces étudiées de Bernard-l'Ermite change, ils ont tendance à devenir diurnes, passent plus de temps à se nourrir et se rassemblent à ces moments-là en plus grands groupes, et ils choisissent aussi de plus longues coquilles. Les cinq mêmes variables sont aussi liées à la proximité et à l'activité des hommes, directement et indirectement. Parmi les effets directs, on compte une tendance à une activité nocturne lorsque les activités humaines sont proches ; les effets indirects incluent des apports de nourriture et de débris de coquilles accrus et plus regroupés résultant des marées et de la déforestation. Les mangroves sont aussi importantes pour les populations locales que pour les Bernard-l'Ermite, pour toute une série de raisons (semblables). Les mangroves constituent un abri en cas de tempête, un terrain de pêche et de frai dont bénéficient les populations humaines voisines, mais elles renferment aussi beaucoup de moustiques, et leur bois fournit un bon matériau de construction et du combustible. Les Bernard-l'Ermite peuvent être (indirectement) utiles aux populations côtières car certaines espèces commerciales sont comestibles et que tous consomment rapidement les restes de nourriture en décomposition et les excréments (réduisant ainsi les risques de maladie et d'animaux nuisibles). PERSELLO-CARTIEAUX ABSTRACT Colonization of the rhizosphere by micro-organisms results in modifications in plant growth and development. This review examines the mechanisms involved in growth promotion by plant growth-promoting rhizobacteria which are divided into indirect and direct effects. Direct effects include enhanced provision of nutrients and the production of phytohormones. Indirect effects involve aspects of biological control: the production of antibiotics and iron-chelating siderophores and the induction of plant resistance mechanisms. The study of the molecular basis of growth promotion demonstrated the important role of bacterial traits (motility, adhesion and growth rate) for colonization. New research areas emerge from the discovery that molecular signalling occurs through plant perception of eubacterial flagellins. Recent perspectives in the molecular genetics of cross-talking mechanisms governing plant,rhizobacteria interactions are also discussed. [source] Direct effects of short-term psychoeducational intervention for relatives of patients with schizophrenia in JapanPSYCHIATRY AND CLINICAL NEUROSCIENCES, Issue 5 2006HAJIME YAMAGUCHI md Abstract, This study examined the direct effects of short-term psychoeducation on relatives of inpatients with schizophrenia, with the goal of introducing this type of support program into standard care. The subjects were 46 relatives of inpatients with schizophrenia who attended three or four sessions of psychoeducation. Levels of anxiety and subjective burden and distress were measured before and after sessions using self-administered rating scales. In addition, levels of expressed emotion were also measured. Results showed that both state and trait anxiety on the State-Trait Anxiety Inventory were significantly lower after psychoeducational intervention than before intervention. In addition, subjective burden and distress reported by the family significantly decreased on the subscales for family confusion resulting from a lack of knowledge of the illness and anxiety about the future, subjective burden and depression resulting from the patient's illness, and difficulties in the relatives' relationships with the patient. Comparison of high and low expressed emotion families showed that the intervention was almost equally effective for the two groups. However, its effectiveness with regard to the subjective burden and depression experienced by the families was significantly greater among high expressed emotion families. The present study confirmed that family psychoeducation during hospitalization, even for a short period, is effective for all families, whether high or low expressed emotion. Moreover, the results suggested that the intervention may have a greater effect on emotional factors in high expressed emotion families than in low expressed emotion families. [source] Developmental toxicity of indium: Embryotoxicity and teratogenicity in experimental animalsCONGENITAL ANOMALIES, Issue 4 2008Mikio Nakajima ABSTRACT Indium, a precious metal classified in group 13 (IIIB) in the periodic table, has been used increasingly in the semiconductor industry. Because indium is a rare metal, technology for indium recycling from transparent conducting films for liquid crystal displays is desired, and its safety evaluation is becoming increasingly necessary. The developmental toxicity of indium in experimental animals was summarized. The intravenous or oral administration of indium to pregnant animals causes growth inhibition and the death of embryos in hamsters, rats, and mice. The intravenous administration of indium to pregnant animals causes embryonic or fetal malformation, mainly involving digit and tail deformities, in hamsters and rats. The oral administration of indium also induces fetal malformation in rats and rabbits, but requires higher doses. No teratogenicity has been observed in mice. Caudal hypoplasia, probably due to excessive cell loss by increased apoptosis in the tailbud, in the early postimplantation stage was considered to account for indium-induced tail malformation as a possible pathogenetic mechanism. Findings from in vitro experiments indicated that the embryotoxicity of indium could have direct effects on the conceptuses. Toxicokinetic studies showed that the embryonic exposure concentration was more critical than the exposure time regarding the embryotoxicity of indium. It is considered from these findings that the risk of the developmental toxicity of indium in humans is low, unless an accidentally high level of exposure or unknown toxic interaction occurs because of possible human exposure routes and levels (i.e. oral, very low-level exposure). [source] The Effect of National Governance Codes on Firm Disclosure Practices: Evidence from Analyst Earnings ForecastsCORPORATE GOVERNANCE, Issue 6 2008John Nowland ABSTRACT Manuscript Type: Empirical Research Question: This study examines whether voluntary national governance codes have a significant effect on company disclosure practices. Two direct effects of the codes are expected: 1) an overall improvement in company disclosure practices, which is greater when the codes have a greater emphasis on disclosure; and 2) a leveling out of disclosure practices across companies (i.e., larger improvements in companies that were previously poorer disclosers) due to the codes new comply-or-explain requirements. The codes are also expected to have an indirect effect on disclosure practices through their effect on company governance practices. Research Findings/Results: The results show that the introduction of the codes in eight East Asian countries has been associated with lower analyst forecast error and a leveling out of disclosure practices across companies. The codes are also found to have an indirect effect on company disclosure practices through their effect on board independence. Practical Implications: This study shows that a regulatory approach to improving disclosure practices is not always necessary. Voluntary national governance codes are found to have both a significant direct effect and a significant indirect effect on company disclosure practices. In addition, the results indicate that analysts in Asia do react to changes in disclosure practices, so there is an incentive for small companies and family-owned companies to further improve their disclosure practices. [source] Ascorbic acid oral treatment modifies lipolytic response and behavioural activity but not glucocorticoid metabolism in cafeteria diet-fed ratsACTA PHYSIOLOGICA, Issue 4 2009D. F. Garcia-Diaz Abstract Aim:, To analyse the effects of vitamin C (VC), a potent dietary antioxidant, oral supplementation on body weight gain, behavioural activity, lipolytic response and glucocorticoid metabolism in the early stages of diet-induced overweight in rats. Methods:, Food intake, locomotive activity and faecal corticosterone were assessed during the 14 day trial period. After 2 weeks, the animals were sacrificed and the body composition, biochemical markers and lipolytic response from isolated adipocytes from retroperitoneal white adipose tissue were examined. Results:, The intake of a high-fat diet by rats induced a significant increase in body weight, adiposity and insulin resistance markers as well as a decrease in faecal corticosterone levels compared with standard diet-fed rats. Interestingly, the animals fed on the cafeteria diet showed a significant increase in the isoproterenol-induced lipolytic response in isolated adipocytes. Furthermore, this cafeteria-fed group showed a reduced locomotive behaviour than the control rats. On the other hand, oral VC supplementation in animals receiving the high-fat diet restored the cafeteria diet effect in some of the analysed variables such as final body weight and plasma insulin to control group levels. Remarkably, increases in locomotive behaviour and a significant decrease in the lipolytic response induced by isoproterenol on isolated adipocytes from animals treated with VC were observed. Conclusion:, This work demonstrates that an oral ascorbic acid supplementation has direct effects on behavioural activity and on adipocyte lipolysis in early obesity stages in rats, which could indicate a protective short-term role of this vitamin against adiposity induced by chronic high-fat diet consumption. [source] RACE, ETHNICITY, THREAT AND THE LABELING OF CONVICTED FELONS,CRIMINOLOGY, Issue 3 2005STEPHANIE BONTRAGER Florida law allows judges to withhold adjudication of guilt for persons who have either pled guilty or been found guilty of a felony. This provision may apply only to persons who will be sentenced to probation, and it allows such individuals to retain all civil rights and to truthfully assert they had not been convicted of a felony. This paper examines the effects of race and Hispanic ethnicity on the withholding of adjudication for 91,477 males sentenced to probation in Florida between 1999 and 2002. Hierarchical Generalized Linear Modeling is used to assess the direct effects of defendant attributes as well as the cross-level interactions between race, ethnicity and community level indicators of threat, such as percentage black and Hispanic and concentrated disadvantage. Our results show that Hispanics and blacks are significantly less likely to have adjudication withheld when other individual and community level factors are controlled. This effect is especially pronounced for blacks and for drug offenders. Cross-level interactions show that concentrated disadvantage has a substantial effect on the adjudication withheld outcome for both black and Hispanic defendants. Palombo Abstract Complete maternal deprivation in rats, through artificial rearing (AR), produces deficits in subsequent maternal behavior of the offspring. These deficits are partially reversed when isolated pups are provided with additional tactile stimulation designed to simulate maternal licking (e.g., Gonzalez et al. [2001] Developmental Psychobiology, 38, 11,32). These findings highlight the importance of the early maternal environment in subsequent development. However, given the possibility that prenatal environments may differ between AR and maternally reared (MR) offspring, the deficits in the behavior of AR mothers may be driven by the characteristics of their pups derived from the effects of an altered prenatal environment. Hence differences in the neonatal pups of AR mothers may produce the alterations in the AR maternal behavior. To rule out this possibility, we employed a fostering paradigm where AR and MR mothers received cross-fostered mother-reared pups. AR mothers showed the same level of deficits in maternal behavior towards MR foster pups as they do with their own pups and these deficits were partially reversed with additional tactile stimulation. Although these medications exert similar effects on blood glucose, recent clinical studies indicated that PIO has a more pronounced beneficial effect on lipid parameters than ROSI. In order to get further insight into the lipid effects of both drugs, we tested whether PIO, compared to ROSI, could exert direct effects on lipid liver metabolism in relation with plasma lipids. Methods We performed in vitro studies using mice liver slices incubated 21 h either with ROSI (1 µmol/L) or PIO (7.5 µmol/L). Results We showed that both glitazones slightly reduced HMG-CoA reductase mRNA levels at the same degree but only PIO reduced intracellular cholesterol content, suggesting an alteration of cholesterol uptake rather than an inhibition of cholesterol biosynthesis. This concept was supported by the reduction of scavenger receptor class B type I expression, hepatic lipase activity and high-density lipoprotein cholesterol uptake in PIO-treated liver explants. Conversely, hepatic lipase mRNA levels were increased 3.5-fold. ROSI, but not PIO, induced acetyl-CoA carboxylase and fatty acid synthase gene expression and increased apoB secretion suggesting a stimulation of lipogenesis. Concurrently, peroxisome proliferator-activated receptor-, mRNA levels were induced by ROSI and not significantly changed by PIO. Besides, PIO appeared to be a more potent activator of AMP-Activated Protein Kinase than ROSI. Conclusions PIO and ROSI exert specific direct effects on liver and extrapolating these data to humans could explain the significant improvements in plasma lipids observed in diabetic patients treated with PIO. No evidence of behavioural fever in response to infection by either pathogen was found, but normal thermoregulation was found to reduce virulence and spore production of B. bassiana. Normal thermoregulation did not appear to affect M. anisopliae var. acridum. 4.,These results suggest that the effects of temperature on host resistance depend on the thermal sensitivity of the pathogen and, in this case, derive from direct effects of temperature on pathogen growth rather than indirect effects mediated by host immune response. 5.,The implications for possible risks of exotic pathogens and influence of climate change are discussed. [source] Connections between species diversity and genetic diversityECOLOGY LETTERS, Issue 7 2005Mark Vellend Abstract Species diversity and genetic diversity remain the nearly exclusive domains of community ecology and population genetics, respectively, despite repeated recognition in the literature over the past 30 years of close parallels between these two levels of diversity. Species diversity within communities and genetic diversity within populations are hypothesized to co-vary in space or time because of locality characteristics that influence the two levels of diversity via parallel processes, or because of direct effects of one level of diversity on the other via several different mechanisms. Here, we draw on a wide range of studies in ecology and evolution to examine the theoretical underpinnings of these hypotheses, review relevant empirical literature, and outline an agenda for future research. The plausibility of species diversity,genetic diversity relationships is supported by a variety of theoretical and empirical studies, and several recent studies provide direct, though preliminary support. Focusing on potential connections between species diversity and genetic diversity complements other approaches to synthesis at the ecology,evolution interface, and should contribute to conceptual unification of biodiversity research at the levels of genes and species. Cruickshank ABSTRACT Aims To examine the literature regarding clinical pharmacokinetics, direct effects and adverse clinical outcomes associated with methamphetamine use. Methods Relevant literature was identified through a PubMed search. Additional literature was obtained from relevant books and monographs. Findings and conclusions The mean elimination half-life for methamphetamine is approximately 10 hours, with considerable inter-individual variability in pharmacokinetics. Direct effects at low-to-moderate methamphetamine doses (5,30 mg) include arousal, positive mood, cardiac stimulation and acute improvement in cognitive domains such as attention and psychomotor coordination. At higher doses used typically by illicit users (,50 mg), methamphetamine can produce psychosis. Its hypertensive effect can produce a number of acute and chronic cardiovascular complications. Repeated use may induce neurotoxicity, associated with prolonged psychiatric symptoms, cognitive impairment and an increased risk of developing Parkinson's disease. Abrupt cessation of repeated methamphetamine use leads to a withdrawal syndrome consisting of depressed mood, anxiety and sleep disturbance. Acute withdrawal lasts typically for 7,10 days, and residual symptoms associated with neurotoxicity may persist for several months. [source] Effects of elevated CO2 associated with maize on multiple generations of the cotton bollworm, Helicoverpa armigeraENTOMOLOGIA EXPERIMENTALIS ET APPLICATA, Issue 1 2010Jin Yin Abstract Under elevated environmental carbon dioxide (CO2), leaf chewers tend to compensate for decreased leaf nutritional quality with increased consumption; mortality and development times also increase and cause a reduction in the fitness of leaf chewers. However, the effect of elevated CO2 on multiple successive generations of these and other insects is not well understood. Furthermore, information about the direct effects of increased environmental CO2 on developmental time and consumption of herbivores is lacking. In this paper, we tested the hypothesis that cascade effects of elevated CO2 through plants, rather than the direct effects of elevated CO2, are the main factors decreasing the fitness of cotton bollworm, Helicoverpa armigera Hübner (Lepidoptera: Noctuidae). We used two series of experiments to quantify the growth, development, and consumption of H. armigera fed on an artificial diet or C4 plants (maize) grown under two CO2 levels (ambient vs. double ambient). In the first series of experiments, elevated CO2 had no effect on the population abundance or individual consumption for three successive generations of cotton bollworms fed on an artificial diet. In the second series of experiments, elevated CO2 reduced population abundance of cotton bollworm larvae for two successive generations when they were fed maize milky grains. The specific effects were longer larval duration, lower fecundity, and decreased rm of cotton bollworms. Furthermore, elevated CO2 increased individual consumption when cotton bollworm was fed maize milky grains for two successive generations and decreased the population's total consumption in the first generation but increased it in the second generation. The results from this study indicate that: (1) The effects of elevated CO2 on three successive generations of cotton bollworm fed on artificial diet were weak, or even non-existent, and (2) elevated CO2 increased the consumption when cotton bollworm were fed maize. Our study also suggests that the damage inflicted by cotton bollworm on maize (a C4 plant) will be seriously affected by the increases in atmospheric CO2, which is unlike our previous results for spring wheat (a C3 plant). Glass ABSTRACT Aims Neurocognitive deficits in chronic alcoholic men are well documented. Impairments include memory, visual,spatial processing, problem solving and executive function. The cause of impairment could include direct effects of alcohol toxicity, pre-existing cognitive deficits that predispose towards substance abuse, comorbid psychiatric disorders and abuse of substances other than alcohol. Cigarette smoking occurs at higher rates in alcoholism and has been linked to poor cognitive performance, yet the effects of smoking on cognitive function in alcoholism are often ignored. We examined whether chronic alcoholism and chronic smoking have effects on executive function. Methods Alcoholism and smoking were examined in a community-recruited sample of alcoholic and non-alcoholic men (n = 240) using standard neuropsychological and reaction-time measures of executive function. Alcoholism was measured as the average level of alcoholism diagnoses across the study duration (12 years). Smoking was measured in pack-years. Results Both alcoholism and smoking were correlated negatively with a composite executive function score. For component measures, alcoholism was correlated negatively with a broad range of measures, whereas smoking was correlated negatively with measures that emphasize response speed. In regression analyses, both smoking and alcoholism were significant predictors of executive function composite. However, when IQ is included in the regression analyses, alcoholism severity is no longer significant. Conclusions Both smoking and alcoholism were related to executive function. However, the effect of alcoholism was not independent of IQ, suggesting a generalized effect, perhaps affecting a wide range of cognitive abilities of which executive function is a component. On the other hand, the effect of smoking on measures relying on response speed were independent of IQ, suggesting a more specific processing speed deficit associated with chronic smoking. Evans-White Abstract Contaminants have direct, harmful effects across multiple ecological scales, including the individual, the community, and the ecosystem levels. Less, however, is known about how indirect effects of contaminants on consumer physiology or behavior might alter community interactions or ecosystem processes. We examined whether a potential aquatic contaminant, an ionic liquid, can indirectly alter benthic algal biomass and primary production through direct effects on herbivorous snails. Ionic liquids are nonvolatile organic salts being considered as an environmentally friendly potential replacement for volatile organic compounds in industry. In two greenhouse experiments, we factorially crossed four concentrations of 1-N-butyl-3-methylimidazolium bromide (bmimBr; experiment 1: 0 or 10 mg/L; experiment 2: 0, 1, or 100 mg/L) with the presence or absence of the snail Physa acuta in aquatic mesocosms. Experimental results were weighted by their respective control (no bmimBr or P. acuta) and combined for statistical analysis. When both bmimBr and snails were present, chlorophyll a abundance and algal biovolume were higher than would be expected if both factors acted additively. In addition, snail growth rates, relative to those of controls, declined by 41 to 101% at 10 and 100 mg/L of bmimBr. Taken together, these two results suggest that snails were less efficient grazers in the presence of bmimBr, resulting in release of algae from the grazer control. Snails stimulated periphyton primary production in the absence, but not in the presence, of bmimBr, suggesting that bmimBr also can indirectly alter ecosystem function. These findings suggest that sublethal contaminant levels can negatively impact communities and ecosystem processes via complex interactions, and they provide baseline information regarding the potential effects of an emergent industrial chemical on aquatic systems. Boone Abstract Although aquatic communities frequently are exposed to a number of pesticides, the effects of chemical mixtures are not well understood. In two separate studies, I examined how insecticide mixtures influenced the likelihood of unpredictable, nonadditive effects on American toad (Bufo americanus) and green frog (Rana clamitans) tadpoles reared in outdoor aquatic communities. I exposed tadpoles to single or multiple insecticides at approximately half the reported median lethal concentrations using insecticides that were either acetylcholinesterase inhibitors (carbaryl or malathion) or a sodium-channel disruptor (permethrin). I found that combinations of insecticides with the same mode of action were more likely to have nonadditive effects on amphibian metamorphosis compared with those having different modes of action. Additionally, in one study, a commercial formulation of permethrin led to near-complete elimination of American toads, suggesting that this formulation could have adverse effects on aquatic communities. Many community studies exploring the ecological effects of expected environmental concentrations of pesticides have suggested that indirect effects in the food web, rather than direct effects on individual physiology, have the largest effect on amphibians. The present study indicates that direct effects of pesticides may become particularly important when insecticides with the same mode of action are present in the environment. [source] Responses of zooplankton in lufenuron-stressed experimental ditches in the presence or absence of uncontaminated refuges,ENVIRONMENTAL TOXICOLOGY & CHEMISTRY, Issue 6 2008Patricia López-Mancisidor Abstract Outdoor experimental ditches were used to evaluate the influence of untreated refuges on the recovery of zooplankton communities following treatment with the fast-dissipating insecticide lufenuron. Each experimental ditch was divided into three sections of the same surface area. The treatments differed in the proportion of ditch (0, 33, 67, and 100% of the surface area) to which the insecticide was applied at the same nominal treatment (3 ,g/L). During the first week postapplication, a barrier was placed between treated and untreated ditch sections. The untreated sections were included to provide a source of organisms for recovery of affected zooplankton populations in the treated sections of the ditch after the removal of the barrier. Cyclopoida were the most affected by lufenuron treatment, followed by Daphnia gr. galeata. These and other direct effects of treatment on larvae of the phantom midge Chaoborus spp. resulted in clear indirect effects on populations of Calanoida, Ceriodaphnia, and Rotifera. Overall, faster recovery of the zooplankton community was observed in the treated sections of ditches that were sprayed for a smaller proportion of their surface area. Nevertheless, individual zooplankton populations showed considerable differences in rate of recovery. Cyclopoida showed a relatively slow rate of recovery even in the partially treated ditches. Daphnia gr. galeata recovered more rapidly in treated ditch sections in the presence of unsprayed ditch sections, illustrating the potential influence of unexposed refuges. Furthermore, the presence of refuges most likely dampened the magnitude and duration of indirect effects in the ditches treated with lufenuron. [source] Effects of lambda-cyhalothrln in two ditch microcosm systems of different trophic statusENVIRONMENTAL TOXICOLOGY & CHEMISTRY, Issue 7 2005Ivo Roessink Abstract The fate and effects of the pyrethroid insecticide lambda-cyhalothrin were compared in mesotrophic (macrophyte-dominated) and eutrophic (phytoplankton-dominated) ditch microcosms (,0.5 m3). Lambda-cyhalothrin was applied three times at one-week intervals at concentrations of 10, 25, 50, 100, and 250 ng/L. The rate of dissipation of lambda-cyhalothrin in the water column of the two types of test systems was similar. After 1 d, only 30% of the amount applied remained in the water phase. Initial, direct effects were observed primarily on arthropod taxa. The most sensitive species was the phantom midge (Chaoborus obscuripes). Threshold levels for slight and transient direct toxic effects were similar (10 ng/L) between types of test systems. At treatment levels of 25 ng/L and higher, apparent population and community responses occurred. At treatments of 100 and 250 ng/L, the rate of recovery of the macroinvertebrate community was lower in the macrophyte-dominated systems, primarily because of a prolonged decline of the amphipod Gammarus pulex. This species occurred at high densities only in the macrophyte-dominated enclosures. Indirect effects (e.g., increase of rotifers and microcrustaceans) were more pronounced in the plankton-dominated test systems, particularly at treatment levels of 25 ng/L and higher. [source] Reproductive and transgenerational effects of methylmercury or Aroclor 1268 on Fundulus heteroclitusENVIRONMENTAL TOXICOLOGY & CHEMISTRY, Issue 2 2001Mary Baker Matta Abstract This research determined the potential for methylmercury or Aroclor 1268 to disrupt reproduction and sexual differentiation in Fundulus heteroclitus. The research determined whether fish that are exposed to mercury or Aroclor 1268 survive and successfully reproduce; whether offspring of exposed fish hatch, survive, produce eggs, and fertilize them; and whether the secondgeneration offspring of exposed fish hatch and survive. Fundulus heteroclitus were exposed to mercury or Aroclor 1268 via contaminated food. Endpoints evaluated included survival, growth, fecundity, fertilization success, hatch success, larval survival, sex ratios, and the prevalence of gonadal abnormalities. In general, polychlorinated biphenyls were highly bioavailable and accumulated well through feeding. The only statistically significant effect observed as a result of treatment with Aroclor 1268 was an increase in growth in the offspring of exposed fish. Mercury was accumulated in a dose-dependent fashion via food exposures. Exposure to mercury in food increased mortality in male F. heteroclitus, which possibly occurred as a result of behavioral alterations. Increased mortality was observed at body burdens of 0.2 to 0.47 ,g/g. Offspring of F. heteroclitus fed mercury-contaminated food were less able to successfully reproduce, with reduced fertilization success observed at egg concentrations of 0.01 to 0.63 ,g/g, which corresponds with parent whole-body concentrations of 1.1 to 1.2 ,g/g. Offspring of exposed fish also had altered sex ratios, with treatment at moderate concentrations producing fewer females and treatment at the highest concentration producing more females than expected. Alterations in sex ratios were observed at concentrations of less than 0.01 ,g/g in eggs or between 0.44 and 1.1 ,g/g in parents. Offspring of mercury-exposed fish also had increased growth in moderate treatments, when egg concentrations were less than 0.02 ,g/g, or when parent whole bodies contained 0.2 to 0.47 ,g/g. In summary, exposure to mercury reduced male survival, reduced the ability of offspring to successfully reproduce, and altered sex ratios in offspring. Both direct effects on exposed fish and transgenerational effects were observed. [source] |
