Comparison of Different Strategies on DNA Chip Fabrication and DNA-Sensing: Optical and Electrochemical Approaches

Abstract New strategies for the construction of DNA chips and the detection of DNA hybridization will be discussed in this review. The focus will be on the use of polypyrrole as a linker between a substrate and oligonucleotide probes. The modification step is based on the electrochemical copolymerization of pyrrole and oligonucleotides bearing a pyrrole group on its 5, end. This strategy was employed for the immobilization of oligonucleotides on millimeter-sized electrodes, microelectrode arrays, as well as for the local structuring of homogeneous gold surfaces. Our approaches for the localized patterning of gold surfaces will be also discussed. Localized immobilization was achieved by using an electrospotting technique, where a micropipette served as an electrochemical cell where spot sizes with 800,,m diameters were fabricated. The use of a microcell using a Teflon covered metal needle with a cavity of 100,,m resulted in immobilized probe spots of 300,,m. Scanning electrochemical microscopy (SECM) was also used, and surface modifications of 100,,m were obtained depending on the experimental conditions. Different detection methods were employed for the reading of the hybridization event: fluorescence imaging, surface plasmon resonance imaging (SPRI), photocurrent measurements, and voltamperometric measurements using intercalators. Their advantages concerning the various immobilization strategies will also be discussed. [source]


3D Powder Printing of ,-Tricalcium Phosphate Ceramics Using Different Strategies,

Custom made macroporous ,-tricalcium phosphate (,-TCP) bone substitutes were fabricated using 3D powder printing comparing three different preparation strategies. Samples fabricated using a novel hydraulic cement setting reaction showed the best printing resolution and highest mechanical performance. This method is a significant step forward in producing ,-TCP monoliths by rapid prototyping and would decrease processing time for commercial fabrication due to their rapid hardening and ease of handling. Politics and Economic Development: Why Governments Adopt Different Strategies to Induce Economic Growth

This article assesses the forces that drive governments to engage in economic development activity and attempts to explain why communities adopt different strategies to bring about the common goal of economic well-being. I address this issue at the state policy level by assessing the relationship between economic development strategy and theories of policy adoption in a pooled times series analysis using indicators of interstate competition, fiscal stress, and state ideology collected between 1983 and 1994. I find that economic development strategy choices are largely a function of interjurisdictional competition. The implications of my findings are discussed. [source]


Numerical modelling of compensation grouting above shallow tunnels

Abstract This paper describes the development of a numerical model for compensation grouting which is a useful technique for the protection of surface structures from the potentially damaging movements arising from tunnel construction. Pipes are inserted into the ground between the tunnel and the overlaying structure from an access shaft. Buildings on the surface are instrumented and movements are carefully monitored. Once the deformations exceed a certain Trigger Level, grout is injected into the ground to prevent damage. In the finite element model described here, compensation grouting is modelled by applying an internal pressure to zero-thickness interface elements embedded in the mesh. An ,observational algorithm' is used, where the deformations of the surface are monitored and used to control the injection process. Example analyses of compensation grouting are given for three-dimensional tunnel construction underneath a greenfield site. Different strategies are used to control the injection process and their effectiveness in preventing surface movement is assessed. The numerical model is shown to replicate general behaviour expected in the field and is capable of modelling the control of ground surface movements at a greenfield site. Vertical integration and trade policy: The case of sugar

The degree of vertical integration in the U.S. sugar industry between raw-sugar processing and sugar refining cannot be explained using theories of vertical integration based only on transaction costs. We graphically decompose the economic rents accruing to each level participant in the marketing channel. Different strategies of several major sugar producing, processing, and refining entities with regard to sugar quota policy are explored. Firms that are integrated from sugar production through to sugar marketing are less impacted by freer trade than are those that concentrate solely on production. We contrast the sugarcane industry in Florida and Louisiana with sugar beet production and processing in the northern plains. The sugar industry in Florida, because of the high degree of vertical integration, is much more capable of dealing with expanded sugar imports than either sugarcane producers in Louisiana or sugar beet growers in the northern plains where integration is not as pronounced. Sharing life with a gluten-intolerant person , the perspective of close relatives

Abstract Background, Several studies indicate the importance of social support in the case of chronic disease. Aim, The aim was to explore dilemmas experienced by close relatives living with a person suffering from coeliac disease, and to describe the strategies they used to deal with these dilemmas. Methods, Twenty-three informants were interviewed. A systematic inductive qualitative method, the critical incident technique was used. Results, Disease-related worries included having bad conscience about not being affected by the disease, experiencing anxiety and witnessing the vulnerability of the affected relative in social situations. Dilemmas related to manage daily life were connected with increased domestic work, restricted freedom of action and the diseased person's preferential right of interpretation of health risks associated with the coeliac disease and deviations from the diet restrictions. Dilemmas related to disturbances in social life, concerned lack of information, knowledge and understanding. Different strategies were described to manage daily life. Conclusions, Close relatives experienced a variety of dilemmas that affected the situation of the whole family. The role of relatives in handling the coeliac disease with the diseased person in the everyday life might be underestimated, and to provide relatives with better knowledge regarding the disease might improve the situation also for patients. This paper explores the preferences that companies have as they use alternative (quasi) external sources of innovative competencies such as strategic technology alliances, mergers and acquisitions, or a mix of these. These alternatives are studied in the context of distinct industrial, technological and international settings during the first half of the 1990s. Different strategies followed by companies and the role played by routinized sets of preferences are also taken into consideration. The analysis demonstrates that these options are influenced by both different environmental conditions and firm specific circumstances, such as those related to protecting core businesses. [source]

Abstract The use of 1,3-diisopropylcarbodiimide (DIC) for the synthesis of cyclic analogues of the hypoglycaemic peptide fragment derived from the N -terminus of human growth hormone (hGH), namely hGH[6,13], is described. Different strategies were examined to achieve improved yields for the on resin side-chain to side-chain cyclization of the corresponding linear peptides containing reverse , - turn motifs. When compared with the more reactive Castro's reagent, the results confirm that DIC in the presence of HOBt can be successfully employed to minimize the formation of intermolecular oligomeric by-products associated with the preparation of cyclic hGH[6,14] peptide analogues based on an i,(i+4)Lys,Glu or Glu,Lys cyclization strategy. Balancing fertility management and economics in organic field vegetable rotations

Abstract BACKGROUND: Organic field-scale vegetables are among the most profitable enterprises in organic farming systems. They are also some of the most nutrient-demanding crops, and many organic arable systems with field-scale vegetables are stockless. Without livestock manure inputs, nutrient supply depends on fertility-building crops, which generate only costs and no income. Different strategies of fertility management were compared on a central England research farm. Fertility management treatments consisted of different lengths of fertility building with green waste compost additions. Outputs and inputs in terms of nutrients and economics were monitored for 31 rotations during 1996,2002. RESULTS: N, P and K rotational nutrient balances, as well as C inputs, showed a negative relationship with rotational gross margins. Variable and allocated fixed costs of fertility building were low, between 2 and 5% of variable costs (Ł0.5,2 ha,1 for 1 kg N ha,1 supplied to the rotation). The intensity of vegetable cropping in these rotations was moderate (25,40% vegetable crops in the rotation) and balancing of fertility management and economics was possible at this intensity without livestock manure or other permitted fertiliser additions. CONCLUSION: Completely stockless systems (in analogy may be called vegan) are possible in organic vegetable production without compromising on fertility or economics. However, for a higher vegetable-cropping intensity (up to 90%) a more sophisticated mix of short-term fertility-building and N-trapping crops will be needed and such rotations may require further external addition of green waste or livestock manure. Quality Concepts for the Improved Use of Recycled Polymeric Materials: A Review

Abstract Polymeric waste materials should be considered resources for the manufacture of new products through recycling processes, with a similar status to virgin fossil-based plastics and biopolymers from renewable resources. Several efforts can be made to achieve this qualitative quantum leap in plastics recycling, and consequently introduce recycled products, with competitive performance, to the market. Scientific knowledge about the degradation processes during the life cycle and the development of fast and reliable analytical methods for the quality assessment of recycled plastics are fundamental to guarantee their performance in new applications. Different strategies,restabilisation, rebuilding, compatibilisation, and addition of elastomers and fillers,can be used to upgrade the structure and properties of polymeric waste streams. This review discusses recent developments in the mechanical recycling of plastics, focusing on how to produce quality materials from waste streams and, thus, contribute to a sustainable management of resources and energy. Interest Group Strategies: Navigating Between Privileged Access and Strategies of Pressure

The literature often contrasts interest groups possessing insider status and outsider groups forced to seek influence through more indirect means. Drawing on data from a survey of all national Danish interest groups, this article demonstrates that most groups have an action repertoire including both direct contacts to bureaucrats and parliamentarians and indirect activities such as media campaigns and mobilizations of members. Different strategies of influence are correlated positively, hence, there is no contradiction between pursuing strategies associated with insider access to decision-making and strategies where pressure is put on decision makers through media contacts and mobilizations. An analysis of four distinct strategies , an administrative, a parliamentary, a media and a mobilization strategy , finds interesting variations in the factors that affect the pursuance of the various strategies of influence. Groups with a privileged position vis-ŕ-vis decision makers have high levels of activities targeting these decision makers, but the lack of a privileged position does not lead groups to pursue indirect strategies. Indirect strategies are most intensively pursued by cause groups and groups who find themselves in a competitive situation with regard to attracting members. [source]


Characterization of a novel bioactive poly[(lactic acid)- co -(glycolic acid)] and collagen hybrid matrix for dermal regeneration

Abstract Different strategies have been explored for the purpose of autologous or allogeneic dermal regeneration. We have developed a hybrid matrix by lyophilizing collagen within a poly[(lactic acid) -co -(glycolic acid)] (10:90, molar composition) knitted mesh, in order to assimilate the advantages of natural and synthetic materials. The porosity of the mesh was found to be almost 95 %, using Micro-Computed Tomography Analysis, while the mechanical properties were comparable to native skin. In vitro biocompatibility was analyzed by culturing rat dermal fibroblasts in the matrices over 10 days. The cells were able to attach, proliferate and remain viable within the hybrid matrices. Subsequently, in vivo biocompatibility was analyzed by implanting the matrices subcutaneously in immunocompetent rats, for 2 weeks. Histological analysis showed that the poly[(lactic acid) -co -(glycolic acid)],collagen hybrid matrices evoked minimal host tissue response in vivo. This study forms the basis of using poly[(lactic acid) -co -(glycolic acid)],collagen hybrid matrices for our future work to develop a bioactive matrix for dermal regeneration. Comparison of Combination Plasmapheresis/IVIg/Anti-CD20 Versus High-Dose IVIg in the Treatment of Antibody-Mediated Rejection

Different strategies appear to improve the success in treatment of antibody-mediated rejection (AMR), although no one best method has yet emerged. The objective of this study was to compare the efficacy of the combination of Plasmapheresis/intravenous immunoglobulin (IVIg)/anti-CD20-based regimes versus high-dose IVIg alone in the treatment of AMR. Group A (12 patients) was treated with high-dose IVIg between January 2000 and December 2003; group B (12 patients) was treated by Plasmapheresis/IVIg/anti-CD20 between January 2004 and December 2005. Graft survival at 36 months was 91.7% in group B versus 50% in group A (p = 0.02). Donor-specific human leukocyte antigens (DSA) levels detected by Luminex single antigen (Luminex SA) and ELISA, 3 months postrejection are significantly lower in group B than in group A: DSA ELISA class 2 score 6,8 (p = 0.02), DSA mean intensity of fluorescence (MFI) max (p = 0.009) and DSA mean MFI (p = 0.0004). The persistence of elevated DSA levels posttreatment is more frequent in patients with graft loss as compared to those with preserved renal function: score 6,8 on ELISA (p = 0.04); mean MFI (p = 0.00009) and MFImax (p = 0.018). We conclude that: (1) high dose IVIg alone is inferior to Plasmapheresis/IVIg/anti-CD20 as therapy for AMR and (2)DSA postrejection can be quantified using solid phase assays, showing that 3 months after AMR, DSA levels are higher in patients with graft loss. A framework for fusion methods and rendering techniques of multimodal volume data

Abstract Many different direct volume rendering methods have been developed to visualize 3D scalar fields on uniform rectilinear grids. However, little work has been done on rendering simultaneously various properties of the same 3D region measured with different registration devices or at different instants of time. The demand for this type of visualization is rapidly increasing in scientific applications such as medicine in which the visual integration of multiple modalities allows a better comprehension of the anatomy and a perception of its relationships with activity. This paper presents different strategies of direct multimodal volume rendering (DMVR). It is restricted to voxel models with a known 3D rigid alignment transformation. The paper evaluates at which steps of the rendering pipeline the data fusion must be realized in order to accomplish the desired visual integration and to provide fast re-renders when some fusion parameters are modified. In addition, it analyses how existing monomodal visualization algorithms can be extended to multiple datasets and it compares their efficiency and their computational cost. On the concept of a universal audit of quality and environmental management systems

There is a definite trend in industry today toward the integration of internal management systems (MSs), including those for managing quality, environment, health and safety, and social accountability. The standards describing the minimum requirements for such systems have been made largely compatible, but are not yet fully aligned or integrated. Apart from several national standards for integrated quality, environment and safety MSs, the world has yet to see a corresponding and internationally accepted guideline. In contrast, integrative standardization activities in the realm of MS auditing are proceeding in full force, with the introduction of the pioneering ISO 19011 guideline for quality and environmental auditing expected soon. This paper focuses on the concepts, principles and practices of a truly generic audit, applicable for the evaluation of diverse aspects of organizational performance against the criteria stated in MS standards. A universal audit model based on the systems approach and several important questions regarding the compatibility and integration of the current auditing schemes are discussed. These issues include the ability of integrated audits to foster unification of supported MSs, as well as different strategies for the development of a universal audit guideline (UAG) and integration of function-specific audits. Postoperative discomfort associated with surgical and nonsurgical endodontic retreatment

Abstract , Endodontic retreatment decision-making must include an appraisal of the costs of the different strategies proposed. In addition to direct costs, postoperative discomfort may have other consequences in terms of time off work, unscheduled visits and suffering. To establish a foundation for the appraisal of such indirect and intangible costs the present study was set up in which patients' assessments of pain and swelling after surgical and nonsurgical retreatment procedures were recorded. Ninety-two patients with 95 root-filled incisors and canine teeth exhibiting apical periodontitis were included in the study. The mode of retreatment was randomly assigned. Each day during the first post-treatment week patients assessed their degree of swelling and pain on horizontal 100-mm visual analog scales (VAS). The scales ranged from "no swelling" to "very severe swelling" and "no pain" to "intolerable pain", respectively. Consumption of self-prescribed analgesics and time off work were also recorded. Significantly more patients reported discomfort after surgical retreatment than after nonsurgical procedures. High pain scores were most frequent on the operative day while swelling reached its maximum on the first postoperative day followed by progressive decrease both in frequency and magnitude. Postoperative symptoms associated with nonsurgical retreatment were less frequent but reached high VAS values in single cases. Analgesics were significantly more often consumed after periapical surgery. Patients reported absence from work mainly due to swelling and discoloration of the skin. This was found to occur only after surgical retreatment. Conclusively, surgical retreatment resulted in more discomfort and tended to bring about greater indirect costs than nonsurgical retreatment. Transnational Advocacy Networks and Affirmative Action for Dalits in India

ABSTRACT In India, movements and parties representing the lowest ranking dalit caste groups have followed different strategies in their struggle against social, economic and cultural discrimination. In this article, a new dalit movement making use of a ,transnational advocacy network strategy' will be compared to a more ,classical'dalit political party. The main policy target for the new movement is an extension of existing affirmative action policies, while the dalit BSP party focuses more on emancipatory issues. Based on an analysis of the impacts of the BSP and of the new movement at the grassroots level, it is argued that the achievements of the new movement are tempered by the fact that in order to make use of international discourses and political pressure, the movement has had to develop a strategy and policy proposals compatible with existing mainstream neoliberal discourses. This depoliticizes the policies, and hence makes them of less importance strategically. It is argued that this is likely to be a difficulty for transnational advocacy networks in general. [source]


Theoretical influences on research on language development and intervention in individuals with mental retardation

Abstract In this article, we consider the theoretical debates and frameworks that have shaped research on language development and intervention in persons with mental retardation over the past four decades. Our starting point is the nativist theory, which has been espoused most forcefully by Chomsky. We also consider more recent alternatives to the nativist approach, including the social-interactionist and emergentist approaches, which have been developed largely within the field of child language research. We also consider the implications for language development and intervention of the genetic syndrome-based approach to behavioral research advocated by Dykens and others. We briefly review the impact and status of the debates spurred by the nativist approach in research on the course of language development in individuals with mental retardation. In addition, we characterize some of the achievements in language intervention that have been made possible by the debates spurred by nativism and the various alternatives to it. The evidence we consider provides support for all three alternatives to the nativist approach. Moreover, successful interventions appear to embody elements of several of these approaches as well as other theoretical approaches (e.g., behaviorism). We conclude that language intervention must be theoretically eclectic in its approach, with different strategies appropriate for teaching different features of language, at different points in development, and for children displaying different characteristics or learning histories. © 2004 Wiley-Liss, Inc. Design and engineering human forms of monoclonal antibodies

Abstract The antibody molecule has multiple properties that make it a key component of the immune response. These include its ability to recognize a vast array of different foreign substrates and to interact with and activate the host effector systems. Antibodies with defined specificities may serve as "magic bullets" for the diagnosis and therapy of multiple diseases. With the development of the hybridoma technology, it was possible to produce rodent (mouse or rat) monoclonal antibodies that are the product of a single clone of antibody producing cells and have only one antigen binding specificity. However, the therapeutic use of rodent monoclonals antibodies in humans is limited by their immunogenicity, short circulating half-life, and inability to efficiently trigger human effector mechanisms. However, it proved difficult to produce human monoclonal antibodies using the same methods. To address these problems genetic engineering and expression systems have instead been used to produce chimeric, humanized, and totally human antibodies as well as antibodies with novel structures and functional properties. In addition, the use of yeast and human artificial chromosome vectors for animal transgenesis has allowed the development of animal models that produce antigen specific antibodies that are totally human. As a consequence, recombinant antibody-based therapies are now used to treat a variety of clinical conditions including infectious diseases, inflammatory disorders, and cancer. Expanding field of purinergic signaling

Abstract This article attempts to paint a broad picture of the extraordinary explosive recent developments in the purinergic signaling field. After a brief historical review and update of purinoceptor subtypes, the focus is on the physiological roles of purines and pyrimidines. These are considered both in terms of short-term signaling in neurotransmission, secretion, and vasodilatation and in long-term (trophic) signaling in development, regeneration, proliferation, and cell death. Examples of trophic signaling include cartilage development in limb buds, glial cell proliferation, development of skeletal muscle, changes in receptor expression in smooth-muscle phenotypes, maturation of testicular spermatids, and bone remodeling. Plasticity of purinoceptor expression in pathological conditions is described, including the increase in the purinergic component of parasympathetic nervous control of the human bladder in interstitial cystitis and outflow obstruction and in sympathetic cotransmitter control of blood vessels in hypertensive rats, the appearance of P2X7 receptors in the glomeruli of the kidney from diabetic and transgenic hypertensive animal models, and up-regulation of P2X1 and P2Y2 receptor mRNA in hearts of rats with congestive heart failure. The role of P2X3 receptors in nociception is considered, and a new hypothesis about purinergic mechanosensory transduction in the gut is explored. A personal view of some of the areas ripe for future development concludes this article, including a discussion of different strategies that could lead to the development of purinergic therapeutic agents. Error patterns in word reading among primary school children: A cross-orthographic study

Abstract A comparative investigation of word reading efficiency indicates that different strategies may be used by English and Swedish early readers. In a first study, 328 native English speakers from UK Years 3 and 6 completed a pen-and-paper word recognition task (the Wordchains test). Results were analysed for frequency and type of errors committed. A sample of 123 chronological-age-matched Swedish children carried out the same task on a matched Swedish test. For a sub-sample of 68 English/Swedish pairs matched on word recognition score and sex, significant differences were observed in a comparison of average and low scorers from the two language groups. The English children attempted more task items and committed more errors, while the Swedish group corrected their errors more, suggesting a difference in approach to the task. A second study of a larger Swedish sample (241 participants) found the same pattern of errors as Study 1. Lateral movement of common carp (Cyprinus carpio L.) in a large lowland river and floodplain

Abstract,,, Common carp (Cyprinus carpio L.) are a major freshwater invader and knowledge of their movements is important for planning control efforts. To investigate the movement patterns of common carp, radio-tags were implanted into 46 adult fish; 37 near a large floodplain wetland, the Barmah-Millewa forest, and 9 in the Murray River approximately 175 km upstream. Tagged fish were located every second week between August 1999 and March 2001. Common carp occupied total linear ranges (TLR) between 0.4 and 238 km (mean 30 ± 61 km), with 25 fish (62.5%) occupying a TLR < 10 km. Two fish made large distance movements approximately 650 km downstream. Fish sex, the number of locations, time at large, or tagging location explained little variability (P > 0.05) in TLR. Monthly distance from release varied from 0.04 to 238 km (mean 15 ± 44 km), and was not significantly related to river discharge and water temperature, but 29 of 31 (93.5%) fish tagged at Barmah moved from the Murray River into adjacent floodplain habitats upon flooding. Five fish (12.5%) moved large distances (>127 km) upstream of the Barmah-Millewa forest. Fourteen fish (35%) showed site fidelity to within 20 m and usually occupied one or two home sites. Twenty-six fish (65%) showed site fidelity to within 100 m occupying up to five sites during the study period. Movement patterns of common carp were complex, and individuals exhibited different strategies, which is typical of invasive species. Efforts to control and potentially reduce common carp populations in regulated river-floodplain environments should target key floodplain access points and over-wintering habitats to reduce adult biomass, spawning and recruitment levels. A novel methodology for the analysis of membrane and cytosolic sub-proteomes of erythrocytes by 2-DE

Abstract With the aim of studying a wide cohort of erythrocyte samples in a clinical setting, we propose here a novel approach that allows the analysis of both human cytosolic and membrane sub-proteomes. Despite their simple structure, the high content of hemoglobin present in the red blood cells (RBCs) makes their proteome analysis enormously difficult. We investigate here different strategies for isolation of the membrane and cytosolic fractions from erythrocytes and their influence on proteome profiling by 2-DE, paying particular attention to hemoglobin removal. A simple, quick and satisfactory approach for hemoglobin depletion based on HemogloBindÔ reagent was satisfactorily applied to erythrocyte cells, allowing the analysis of the cytosolic sub-proteome by 2-DE without major interference. For membrane proteome, a novel combined strategy based on hypotonic lysis isolation and further purification on minicolumns is described here, allowing detection of high molecular weight proteins (i.e. spectrin, ankyrin) and well-resolved 2-DE patterns. An aliquot of the membrane fraction was also in solution digested and analyzed by nano-LC coupled to an LTQ-Orbitrap mass spectrometer. A total of 188 unique proteins were identified by this approach. This study sets the basis for future clinical studies where the erythrocyte cell may be implicated. [source]


Determination of trace cationic impurities in butylmethylimidazolium-based ionic liquids: From transient to comprehensive single-capillary counterflow isotachophoresis-zone electrophoresis

Abstract Determination of impurities in ionic liquids (ILs) remains a difficult task. In this work, the hyphenation of isotachophoretic,(ITP) preconcentration to zone electrophoresis,(ZE) has been explored for the trace analysis of the cationic impurities Na+, Li+, and methylimidazolium (MI+) in butylmethylimidazolium (BMI+)-based ILs. Simultaneous detection of UV-transparent and UV-absorbing impurities was ensured by a BGE composed of creatinine-acetate buffer. To induce ITP, three different strategies were evaluated: (i),Sample self-stacking ensured by the addition of ammonium acetate (NH4Ac) to 25,50-fold diluted IL solution (transient ITP). (ii),Complete ITP-ZE separation performed in a single capillary: ITP was realized in discontinuous electrolytes comprising an 80,mM NH4Ac, 40,mM acetic acid, 30,mM ,-CD, pH,5.05, leading electrolyte,(LE) and a 10,mM creatinine, 10,mM acetic acid, pH,4.9, terminating electrolyte,(TE). To create the ZE stage, the ITP stack of analytes was moved back toward the capillary inlet by pressure and simultaneously the capillary was filled with the BGE. This protocol made it possible to accommodate a 2.5-times diluted IL sample. (iii),Complete counterflow ITP-ZE with continuous electrokinetic sample supply: the ITP stage was performed in a capillary filled with a 150,mM NH4Ac, 75,mM acetic acid, 30,mM ,-CD, pH,5.0 LE, with 40-times diluted IL at the capillary inlet. BMI+ from IL acts as the terminating ion. The LODs reached in this latter case were at the 10 and 1,ppb levels for MI+ and Li+ in diluted IL matrix, respectively. [source]


Determination of dissociation constants by competitive binding in partial filling capillary electrophoresis

Abstract The determination of dissociation constands (Kd) by competitive ligand binding in partial filling capillary electrophoresis is demonstrated. Two different strategies were applied, one of which only uses a single reporter ligand and a more elaborated one which suppresses systemic disturbances by using a racemic mixture as reporter. The dissociation constants obtained by both alternatives were virtually identical and in good agreement with those previously reported. [source]

Abstract A food chain through three trophic levels on certain trees, Stomaphis aphids , a primary parasitoid, Protaphidius nawaii, a hyperparasitoid, Euneura stomaphidis, has been known in Japan. In the course of my survey on the seasonal prevalence of three Stomaphis species and their parasitoids from 1981,2007 in Kyoto, Japan, I found another hyperparasitoid, which I describe here as a new species, Dendrocerus stomaphis. From fragmentary observations, I inferred the life cycles of P. nawaii, E. stomaphidis and D. stomaphis. These three species have different strategies to escape ant aggression: P. nawaii by specific morphological and behavioral adaptation, E. stomaphidis by general anti-predator behavior like quick movement and D. stomaphis by a unique univoltine life cycle with the active phase from late autumn to early winter when the ants are less active. Efficacy of intervention strategies for bioremediation of crude oil in marine systems and effects on indigenous hydrocarbonoclastic bacteria

Summary There is little information on how different strategies for the bioremediation of marine oil spills influence the key indigenous hydrocarbon-degrading bacteria (hydrocarbonoclastic bacteria, HCB), and hence their remediation efficacy. Therefore, we have used quantitative polymerase chain reaction to analyse changes in concentrations of HCB in response to intervention strategies applied to experimental microcosms. Biostimulation with nutrients (N and P) produced no measurable increase in either biodegradation or concentration of HCB within the first 5 days, but after 15 days there was a significant increase (29%; P < 0.05) in degradation of n -alkanes, and an increase of one order of magnitude in concentration of Thalassolituus (to 107 cells ml,1). Rhamnolipid bioemulsifier additions alone had little effect on biodegradation, but, in combination with nutrient additions, provoked a significant increase: 59% (P < 0.05) more n -alkane degradation by 5 days than was achieved with nutrient additions alone. The very low Alcanivorax cell concentrations in the microcosms were hardly influenced by addition of nutrients or bioemulsifier, but strongly increased after their combined addition, reflecting the synergistic action of the two types of biostimulatory agents. Bioaugmentation with Thalassolituus positively influenced hydrocarbon degradation only during the initial 5 days and only of the n -alkane fraction. Bioaugmentation with Alcanivorax was clearly much more effective, resulting in 73% greater degradation of n -alkanes, 59% of branched alkanes, and 28% of polynuclear aromatic hydrocarbons, in the first 5 days than that obtained through nutrient addition alone (P < 0.01). Enhanced degradation due to augmentation with Alcanivorax continued throughout the 30-day period of the experiment. In addition to providing insight into the factors limiting oil biodegradation over time, and the competition and synergism between HCB, these results add weight to the use of bioaugmentation in oil pollution mitigation strategies. Perception and integration of people living with type 1 diabetes , an empirical study

Abstract Background: The chronic complications of type 1 diabetes impose a heavy physical, psychological and social burden on people living with the condition. Improved metabolic control reduces the risk of developing chronic complications and could lead to improved well-being for people with diabetes. Aims: The aim of this study was to explore the perceptions of adults with type 1 diabetes who have improved their metabolic control with respect to acceptance, knowledge, social support and their relationships with healthcare providers. Methods: The study included ten people with type 1 diabetes who had achieved and maintained a reduction of 1.5% in their HbA1C during a one-year period. A phenomenological qualitative semi-structured interview was used in the collection of information from the participants. The interviews were analysed using the method of meaning condensation; these were interpreted from a perspective of integration. Results: The study shows differences among people living with diabetes regarding their perceptions of living with the condition. The people have at least three different strategies of integrating diabetes, based on their perception. Conclusions: Integration is a life-long process and in this process, the person with diabetes has to learn to integrate diabetes into both behavioural and psychosocial aspects of life. [source]


Evaluating cluster analysis solutions: an application to the Italian NEO personality inventory

This paper is concerned with the evaluation of cluster analysis solutions. Internal criteria and replication issues are compared and applied to empirical data collected from an Italian sample of 421 young adults, using the NEO Personality Inventory. The following internal criteria were considered: C, gamma, and G(,+,) indices, and point-biserial correlation. Replication was examined (i) ,internally' using double cross-validation and bootstrap approaches and (ii) ,externally' by comparing the solution obtained on the Italian sample with the results obtained in German and Spanish samples. While replication analyses supported three- and four-cluster solutions, internal criteria (with the exception of point-biserial correlation) tended to privilege solutions with a much larger number of groups. THE EVOLUTION OF SPECIFICITY IN EVOLVING AND COEVOLVING ANTAGONISTIC INTERACTIONS BETWEEN A BACTERIA AND ITS PHAGE

The evolution of exploitative specificity can be influenced by environmental variability in space and time and the intensity of trade-offs. Coevolution, the process of reciprocal adaptation in two or more species, can produce variability in host exploitation and as such potentially drive patterns in host and parasite specificity. We employed the bacterium Pseudomonas fluorescens SBW25 and its DNA phage ,2 to investigate the role of coevolution in the evolution of phage infectivity range and its relation with phage growth rate. At the phage population level, coevolution led to the evolution of broader infectivity range, but without an associated decrease in phage growth rate relative to the ancestor, whereas phage evolution in the absence of bacterial evolution led to an increased growth rate but no increase in infectivity range. In contrast, both selection regimes led to phage adaptation (in terms of growth rates) to their respective bacterial hosts. At the level of individual phage genotypes, coevolution resulted in within-population diversification in generalist and specialist infectivity range types. This pattern was consistent with a multilocus gene-for-gene interaction, further confirmed by an observed cost of broad infectivity range for individual phage. Moreover, coevolution led to the emergence of bacterial genotype by phage genotype interactions in the reduction of bacterial growth rate by phage. Our study demonstrates that the strong reciprocal selective pressures underlying the process of coevolution lead to the emergence and coexistence of different strategies within populations and to specialization between selective environments.
