Selected AbstractsModels with Errors due to Misreported MeasurementsAUSTRALIAN & NEW ZEALAND JOURNAL OF STATISTICS, Issue 4 2003Brent Henderson Summary Measurement error and misclassification models feature prominently in the literature. This paper describes misreporting error, which can be considered to fall somewhere between these two broad types of model. Misreporting is concerned with situations where a continuous random variable X is measured with error and only reported as the discrete random variable Z. Data grouping or rounding are the simplest examples of this, but more generally X may be reported as a value z of Z which refers to a different interval from the one in which X lies. The paper discusses a method for handling misreported data and draws links with measurement error and misclassification models. A motivating example is considered from a prenatal Down's syndrome screening, where the gestational age at which mothers present for screening is a true continuous variable but is misreported because it is only ever observed as a discrete whole number of weeks which may in fact be in error. The implications this misreporting might have for the screening are investigated. [source] NMDA receptor-mediated metaplasticity during the induction of long-term depression by low-frequency stimulationEUROPEAN JOURNAL OF NEUROSCIENCE, Issue 11 2002Bruce Mockett Abstract Metaplasticity refers to the activity-dependent modification of the ability of synapses to undergo subsequent synaptic plasticity. Here, we have addressed the question of whether metaplasticity contributes to the induction of long-term depression (LTD) by low-frequency stimulation (LFS). The experiments were conducted using standard extracellular recording techniques in stratum radiatum of area CA1 in hippocampal slices made from adult Sprague,Dawley rats. The degree of LTD induction was found to be a nonlinear function of the number of pulses during a 1-Hz LFS. Little LTD was observed following 600 or 900 pulses, but a significant LTD occurred following 1200 pulses of LFS, whether delivered in one episode, or in two bouts of 600 pulses given 10 min apart. A similar pattern was observed for 3 Hz LFS. The data support the suggestion that pulses occurring early in the LFS train prime synapses for LTD induction, as triggered by later occurring stimuli. The priming effect lasted at least 120 min, when tested by giving two bouts of 1 Hz LFS (600 pulses each) at different intervals. Neither heterosynaptic nor homosynaptic stimulation by itself was sufficient to prime LTD. However, a combination of the stimuli, induced by increased stimulus strength during the LFS, appeared necessary for inducing the effect. An N -methyl- d -aspartate (NMDA) receptor antagonist markedly reduced total LTD induction, regardless of whether it was administered during the first or second LFS in a protocol employing two bouts of 600 pulse LFS, 30 min apart. These findings strongly support the hypothesis that NMDA receptor-dependent metaplasticity processes contribute to the induction of LTD during standard LFS protocols. [source] The life cycles of the temperate lactococcal bacteriophage ,LC3 monitored by a quantitative PCR methodFEMS MICROBIOLOGY LETTERS, Issue 1 2000Merete Lunde Abstract We present here a new and general approach for monitoring the life cycles of temperate bacteriophages which establish lysogeny by inserting their genomes site-specifically into the bacterial host chromosome. The method is based on quantitative amplification of specific DNA sites involved in various cut-and-join events during the life cycles of the phages (i.e. the cos, attP, attB, attL and attR sites) with the use of sequence-specific primers. By comparing the amounts of these specific DNA sites at different intervals, we were able to follow the development of the lytic and lysogenic life cycles of the temperate lactococcal bacteriophage ,LC3 after infection of its bacterial host Lactococcus lactis ssp. cremoris IMN-C18. [source] Cell adhesion regulates platelet-derived growth factor,induced MAP kinase and PI-3 kinase activation in stellate cellsHEPATOLOGY, Issue 3 2002Vinicio Carloni The biologic effects of growth factors are dependent on cell adhesion, and a cross talk occurs between growth factors and adhesion complexes. The aim of the present study was to evaluate the influence of cell adhesion on the major intracellular signaling pathways elicited by platelet-derived growth factor (PDGF) in hepatic stellate cells (HSC). PDGF signaling was investigated in an experimental condition characterized by lack of cell adhesion for different intervals of time. Basal and PDGF-induced focal adhesion kinase (FAK) tyrosine phosphorylation was maintained in a condition of cell suspension for 2, 4, and 6 hours, whereas it was completely lost after 12 and 24 hours. We examined MAP kinase activity at 2 and 24 hours, corresponding to the higher and lower levels of FAK phosphorylation. In these experiments, MAP kinase activity correlated with FAK phosphorylation. Stimulation with PDGF was able to cause Ras-GTP loading only in adherent cells. The ability of PDGF to induce phosphatidylinositol 3-kinase (PI 3-K) activity was abrogated in cells maintained in suspension. The Ser473 phosphorylation of Akt was only marginally affected by the lack of cell adhesion. We then evaluated the association of FAK with c-Src. This association was found to be cell adhesion dependent, and it did not appear to be dependent from phosphorylated FAK. These changes in PDGF-induced intracellular signaling were associated with a remarkable reduction of PDGF-proliferative potential in nonadherent cells, although no marked differences in the apoptotic rate were observed. In conclusion, these results suggest that cell adhesion differentially regulates major signaling pathways activated by PDGF in HSC. [source] At what costs will screening with CT colonography be competitive?INTERNATIONAL JOURNAL OF CANCER, Issue 5 2009A cost-effectiveness approach Abstract The costs of computed tomographic colonography (CTC) are not yet established for screening use. In our study, we estimated the threshold costs for which CTC screening would be a cost-effective alternative to colonoscopy for colorectal cancer (CRC) screening in the general population. We used the MISCAN-colon microsimulation model to estimate the costs and life-years gained of screening persons aged 50,80 years for 4 screening strategies: (i) optical colonoscopy; and CTC with referral to optical colonoscopy of (ii) any suspected polyp; (iii) a suspected polyp ,6 mm and (iv) a suspected polyp ,10 mm. For each of the 4 strategies, screen intervals of 5, 10, 15 and 20 years were considered. Subsequently, for each CTC strategy and interval, the threshold costs of CTC were calculated. We performed a sensitivity analysis to assess the effect of uncertain model parameters on the threshold costs. With equal costs ($662), optical colonoscopy dominated CTC screening. For CTC to gain similar life-years as colonoscopy screening every 10 years, it should be offered every 5 years with referral of polyps ,6 mm. For this strategy to be as cost-effective as colonoscopy screening, the costs must not exceed $285 or 43% of colonoscopy costs (range in sensitivity analysis: 39,47%). With 25% higher adherence than colonoscopy, CTC threshold costs could be 71% of colonoscopy costs. Our estimate of 43% is considerably lower than previous estimates in literature, because previous studies only compared CTC screening to 10-yearly colonoscopy, where we compared to different intervals of colonoscopy screening. © 2008 Wiley-Liss, Inc. [source] Bladder wall grafting in rats using salt-modified and collagen-coated polycaprolactone scaffolds: Preliminary reportINTERNATIONAL JOURNAL OF UROLOGY, Issue 10 2007Dah-Shyong Yu Aim: A rat model was used for the evaluation of collagen-coated and salt-modified polycaprolactone (PCL) scaffolds for bladder grafting after hemicystectomy. Methods: SD rats underwent partial cystectomy and cystoplasty with collagen-coated and salt-modified polycaprolactone scaffolds. The grafts of the regenerated bladder wall were harvested at different intervals and tissue regeneration was evaluated microscopically. Anatomic and functional characters were evaluated by cystography and urodynamics. Results: At harvesting, after 1 and 2 months, we found good preservation of the bladder shape and volume in all 16 rats receiving PCL cystorrhaphy. No stone formation was observed. Good epithelialization and ingrowth of smooth muscle cells were seen after 2 months grafting. Collagen-coated PCL scaffolds showed considerable encrustation, which appeared to be absorbed and disappear with time. The cystographic and urodynamic examinations revealed intact contour and a well-accommodated bladder with reservoir volume and contractility. Conclusions: In the rat model, we have successfully demonstrated the applicability of collagen coated and salt-modified PCL in reconstruction of the partial cystectomized bladder. [source] Metabolic and luteal function in winter-calving Spanish beef cows as affected by calf management and breedJOURNAL OF ANIMAL PHYSIOLOGY AND NUTRITION, Issue 3 2010J. Álvarez-Rodríguez Summary This experiment aimed at evaluating the effect of calf management and breed on the metabolic and luteal function of post-partum beef cows fed at maintenance. Fifty multiparous cows, 22 Parda de Montaña (PA) and 28 Pirenaica (PI), were assigned to either suckling once-daily for 30 min (RESTR) or ad libitum (ADLIB) from the day after calving. Blood samples were collected to analyse metabolites [non-esterified fatty acids (NEFA), ,-hydroxybutyrate, total protein and urea)], insulin-like growth factor-I (IGF-I) and progesterone (P4) at different intervals. Cows from RESTR maintained their live-weight (LW) over the first 3 months post-partum, whereas ADLIB cows lost nearly 4% LW. Both genotypes showed similar LW gains during this period (p > 0.10). Calf daily gains were lower in RESTR than in ADLIB treatment (p < 0.05), but similar across breeds (p > 0.10). Milk and lactose production were lower in RESTR cows than in ADLIB (p < 0.05). Milk and protein yield were greater in PA than in PI breed (p < 0.05). Serum NEFA, total protein and urea were higher in PI cows suckling ADLIB than in the rest (p < 0.05). Cows from PI breed had greater NEFA values than PA ones on the first week post-partum (p < 0.001). Circulating IGF-I was not affected by suckling frequency, breed nor their interaction (p > 0.10). Suckling frequency, but not breed, affected the interval from calving to first ovulation (p < 0.001), being shorter in RESTR than in ADLIB cows. In conclusion, the ad libitum suckling practice improved cow milk yield and offspring gain compared to once-daily suckling for 30 min from the day after calving, at the expense of impairing the onset of cyclicity. The effect of calf management was confounded with breed on the studied blood biochemical constituents, but any of these metabolites influenced the role of endocrine IGF-I in these genotypes. [source] Laboratory and field studies on the integrated pest management of Helicoverpa armigera (Hübner) in cotton, based on pheromone trap catch threshold levelJOURNAL OF APPLIED ENTOMOLOGY, Issue 5-6 2000G. V. P. Reddy Results revealed that egg parasitism in the laboratory by Trichogramma chilonis was 75.6%. Among the insecticides tested against T. chilonis and the predator Chrysoperla carnea, nimbecidine (neem product) and dipel resulted in zero mortality, with only a low level of mortality by dimethoate, cypermethrin, fenvalerate, alphamethrin and monocrotophos. Combinations of nimbecidine 2% + NPV at 250 larval equivalents (LE)/ha and dipel 8 l + NPV @250 LE/ha were the most effective treatments against H. armigera. The integrated pest management components (T. chilonis, C. carnea, NPV, nimbecidine, dipel and synthetic chemicals) were imposed at different intervals on the basis of pheromone trap threshold level (7 moths/trap per night) on a consolidated block of 40 ha cotton (MCU-1) fields at two locations, Shankarabanda and Korlagundi. The results demonstrated a significant superiority of the IPM strategy in terms of both cost versus benefit and environmental safety over that used in the farmer's fields where only conventional control methods were followed. Aglan Abstract In this study, the effects of environmental aging on the mechanical performance of elastomeric polyurethane (PU) were investigated using two accelerated aging techniques, namely, ultraviolet (UV) and hygrothermal (HT). Samples were prepared and subjected to UV and HT exposure for a period of 5 months and removed and mechanically tested at different time intervals. Differential scanning calorimetry (DSC) was performed. A noticeable change in the chemical structure of the PU after 1 month of UV exposure was found, however, that was not the case after 1 month of HT exposure. The stress and strain to failure, tearing energy, and storage modulus were evaluated at different intervals for both aging techniques. It was found that the UV exposure caused severe degradation of the PU in comparison with the HT. A reduction of more than 98% in the tearing energy was observed for the UV-exposed samples after 5 months when compared with only a 35% reduction in the tearing energy for the HT-exposed samples. A similar trend was observed for tear strength and storage modulus. The degradation mechanisms of the PU elastomers have been identified using SEM and correlated with the tearing energy. © 2008 Wiley Periodicals, Inc. HILDEBRANDT Protective Zones During Quatrefoil Reentry.Introduction: An S3 stimulus can exert a protective effect by terminating reentry induced by an S2 stimulus. Our goal was to examine the mechanism by which an S3 pulse terminates reentry and the role that virtual electrodes and break excitation play in this process. Methods and Results: In our simulation, the bidomain model represents the electrical properties of the tissue and the Beeler-Reuter model represents the membrane kinetics. Quatrefoil reentry is initiated by S1-S2 stimulation, and then a third stimulus S3 is applied at different intervals after S2. All stimuli are applied through the same unipolar electrode. For some S2-S3 intervals, the S2 and S3 wavefronts interact destructively, terminating reentry (protective zones). For other S2-S3 intervals, S2 and S3 wavefronts interact constructively, and reentry continues. Protective zones appear recurrently, with approximately the period of the S2 reentrant circuit. The protective zones are wider for anodal stimulation than for cathodal stimulation. Conclusion: Virtual electrodes, break excitation, and S2-S3 timing all play important roles in determining the electrical response of the tissue. [source] Porosity and surface characteristics of activated carbons produced from waste tyre rubberJOURNAL OF CHEMICAL TECHNOLOGY & BIOTECHNOLOGY, Issue 1 2002Guillermo San Miguel Abstract Waste tyre rubber has proven to be a suitable precursor for the production of high quality activated carbons. The performance of these carbons in commercial applications such as water treatment or gas purification is highly dependent on their surface characteristics. This paper presents an in-depth investigation on how production conditions may affect the yield and characteristics of activated carbons produced from tyre rubber. For this purpose, three tyre rubbers of different particle sizes were consecutively pyrolysed and then activated in a steam atmosphere at 925,°C using a laboratory-scale rotary furnace. Activation was conducted at different intervals over 80,640,min to achieve different degrees of carbon burn-off. The resulting carbons were analysed for their elemental composition, ash content and nitrogen gas adsorption characteristics. The BET and t -plot models were used to investigate various aspects of their porosity and surface area characteristics. SEM analyses were also conducted for visual examination of the carbon surface. Results show that pyrolytic chars, essentially mesoporous materials, developed a very narrow microporosity during the initial stages of the activation process (up to 15,25,wt% burn-off). Further activation resulted in the progressive enlargement of the average micropore width and a gradual development of the mesoporous structure. Total micropore volumes and BET surface areas increased continuously with the degree of activation to reach values up to 0.498,cm3g,1 and 1070,m2g,1 respectively, while external surface areas developed more rapidly at degrees of activation above 45,wt% burn-off. Results presented in this work also illustrate that carbons produced from powdered rubber developed a narrower and more extensive porosity, both in the micropore and mesopore range, than those produced from rubber of a larger particle size. © 2001 Society of Chemical Industry [source] Multishot diffusion-weighted SPLICE PROPELLER MRI of the abdomenMAGNETIC RESONANCE IN MEDICINE, Issue 5 2008Jie Deng Abstract Multishot FSE (fast spin echo)-based diffusion-weighted (DW)-PROPELLER (periodically rotated overlapping parallel lines with enhanced reconstruction) MRI offers the potential to reduce susceptibility artifacts associated with single-shot DW-EPI (echo-planar imaging) approaches. However, DW-PROPELLER in the abdomen is challenging due to the large field-of-view and respiratory motion during DW preparation. Incoherent signal phase due to motion will violate the Carr-Purcell-Meiboom-Gill (CPMG) conditions, leading to destructive interference between spin echo and stimulated echo signals and consequent signal cancellation. The SPLICE (split-echo acquisition of FSE signals) technique can mitigate non-CPMG artifacts in FSE-based sequences. For SPLICE, spin echo and stimulated echo are separated by using imbalanced readout gradients and extended acquisition window. Two signal families each with coherent phase properties are acquired at different intervals within the readout window. Separate reconstruction of these two signal families can avoid destructive phase interference. Phantom studies were performed to validate signal phase properties with different initial magnetization phases. This study evaluated the feasibility of combining SPLICE and PROPELLER for DW imaging of the abdomen. It is demonstrated that DW-SPLICE-PROPELLER can effectively mitigate non-CPMG artifacts and improve DW image quality and apparent diffusion coefficient (ADC) map homogeneity. [source] Using Cochrane reviews for oral diseasesORAL DISEASES, Issue 7 2010HV Worthington Oral Diseases (2010) 16, 592,596 Objectives:, To provide readers with information about the Cochrane Oral Health Group and how the reviews on oral diseases have contributed to guideline developments and the commissioning of trials. Materials and methods:, Examples have been selected from the reviews published on The Cochrane Library. Descriptions are given of how these reviews have been used in guideline development and commissioning of trials. Readers are updated on reviews focused on the management of oral cancer and the new venture of diagnostic test reviews. Results:, Reviews on the management of oral diseases due to cancer treatments have been included in guidelines and changed practice in the UK. Cochrane reviews on Bell's Palsy have led to a randomised controlled trial which has changed the evidence base. The Cochrane review on recall intervals between routine appointments has input into the NICE guideline and resulted in a randomised controlled trial to look at different intervals including a risk-based interval. Conclusion:, We hope this article will give readers information on the work of the Cochrane Oral Health Group and insight into the diversity of reviews in oral diseases. The reviews are successfully being used to change practice and as background for the funding of large-scale clinical trials. [source] The cost-effectiveness of cervical screening in Australia: what is the impact of screening at different intervals or over a different age range?AUSTRALIAN AND NEW ZEALAND JOURNAL OF PUBLIC HEALTH, Issue 1 2008Rob Anderson Abstract Objective: To estimate the cost-effectiveness of altering the currently recommended interval and age range for cervical screening of Australian women. Methods: The cost and effectiveness estimates of alternative screening strategies were generated using an established decision model. This model incorporated a Markov model (of the natural history of cervical cancer and pre-cancerous lesions) and decision trees which: ,mapped' the various pathways to cervical cancer screening; the follow-up of abnormal Pap test results; and the management of confirmed lesions. The model simulated a hypothetical large cohort of Australian women from age 15 to age 85 and calculated the accumulated costs and life-years under each screening strategy. Results: Our model estimated that moving from the current two-yearly screening strategy to annual screening (over the same age range) would cost $379,300 per additional life-year saved. Moving from the current strategy to three-yearly screening would yield $117,100 of savings per life-year lost (costs and effects both discounted at 5% per year), with a relatively modest (<5%) reduction in the total number of life-years saved by the program. Conclusions: Although moving to annual screening would save some additional lives, it is not a cost-effective strategy. Consideration should be given to increasing the recommended interval for cervical screening. However, the net value of any such shift to less effective (e.g. less frequent) and less costly screening strategies will require better evidence about the cost-effectiveness of strategies that encourage non-screeners or irregular screeners to have a Pap test more regularly.
