Activities of glucose phosphorylation, glucose-6-phosphatase and lipogenic enzymes in the liver of perch, Perca fluviatilis, after different dietary treatment

Abstract Glucose phosphorylation was increased and the activity of glucose-6-phosphatase was decreased in the liver of perch Perca fluviatilis after feeding previously fasted fish with a high protein/low carbohydrate diet as well as with a diet containing 23% carbohydrate. Activity of the low affinity hexokinase IV (or D), also called glucokinase (GK), was not observed in the liver of perch on the natural diet, fasted perch or perch after feeding with the high protein/low-carbohydrate diet (< 0.2% CHO). How ever, hepatic GK-activity appeared after feeding with the carbohydrate containing diet. By contrast, the activity of hepatic high affinity hexokinase (HK), which was very low in fasted fish, was strongly increased after feeding with the low-carbohydrate as well as the carbohydrate-containing diet. Apparently, HK rather than GK is the hexokinase isoenzyme that is consistently regulated inversely to glucose-6-phosphatase. Activities of the lipogenic enzymes glucose-6-phosphate dehydrogenase, ATP-citrate lyase and malic enzyme were increased by feeding, particularly with the high protein/low carbohydrate diet. Very high levels of hepatic glycogen were observed after both diets. The results are in accordance with the hypothesis that the hepatic high affinity isoenzyme (HK) has a particular anabolic role. In vitro studies on the effects of Saccharomyces boulardii and Bacillus cereus var. toyoi on nutrient transport in pig jejunum

The probiotics Saccharomyces boulardii and Bacillus cereus var. toyoi are nonpathogenic microbes which have been shown to affect certain functions of the mucosal barrier in pig jejunum such as electrogenic ion transport capacity and paracellular permeability. The present studies were performed to investigate potential effects of the probiotics on jejunal nutrient transport such as sodium-dependent glucose transport or proton-dependent dipeptide transport. For this purpose the in vitro Ussing-chamber technique was applied in order to examine net electrogenic ion flux rates (short circuit currents, Isc) across isolated intact jejunal epithelia in the absence and presence of either 10 mmol/l glucose (mucosal side) or two-fold application of 5 mmol/l glycyl- l -sarcosine or glycyl- l -glutamine to the mucosal bathing solution. Brush border membrane vesicles (BBMV) were prepared in order to characterize kinetic parameters (Vmax, Km) of Na-dependent glucose transport. Intestinal tissues were obtained from growing pigs in a weight range between 23 and 33 kg. All animals were fed twice daily and received 0.8,0.9 kg/day of a standard diet. After a 9- to 10-day adaptation period the diets for treated animals were either supplemented for 8 days with 1.7×107 colony-forming units (CFU)/g feed of S. boulardii or for 3 weeks with 106 CFU/g feed B. cereus var. toyoi. Under basal conditions Isc values were not affected by different treatment protocols (controls: 0.74 ± 0.04 µeq/cm2 per h, n=9; S. boulardii: 0.74 ± 0.12 µeq/cm2 per h, n=7; B. cereus 0.68 ± 0.09 µeq/cm2 per h, n=5). Irrespective of dietary treatment, the addition of glucose resulted in significant increases of Isc indicating substantial onset of electrogenic net Na/glucose cotransport. Maximal Isc values occurred within 30 min and reached 2.79 ± 0.41 µeq/cm2 per h in control epithelia. This was significantly lower than found in S. boulardii (4.47 ± 0.43 µeq/cm2 per h, p < 0.05) and B. cereus var. toyoi tissues (4.45 ± 0.31 µeq/cm2 per h, p < 0.05). Gt values were 22.4 ± 1.3 mS/cm2 in control animals and were significantly lower as shown in S. boulardii (p < 0.01) and B. cereus var. toyoi (p < 0.01)-treated animals (28.4 ± 1.3 and 29.9 ± 0.8 mS/cm2, respectively). Vmax values of Na-dependent glucose uptake into BBMV differed significantly between controls (0.64 ± 0.08 nmol/mg protein per 10 s; n=5), S. boulardii (0.89 ± 0.06 nmol/mg protein per 10 s; n=5, p < 0.05) and B. cereus var. toyoi preparations (1.08 ± 0.05 nmol/mg protein per 10 s; n=3, p < 0.01). Km values were not significantly affected (control: 0.31 ± 0.04 mmol/l, S. boulardii: 0.29 ± 0.05 mmol/l, B. cereus var. toyoi: 0.21 ± 0.01 mmol/l). Irrespective of dietary treatment, application of the dipeptide model substances glycyl- l -sarcosine or glycyl- l -glutamine resulted in significant increases of Isc indicating marked stimulation of electrogenic net H+/dipeptide cotransport. Highest Isc responses occurred in B. cereus var. toyoi preparations and lowest were found in control tissues. However, these differences were not significant. Gt values were not affected by different dietary treatments. The results clearly demonstrate that oral administration of either S. boulardii or B. cereus var. toyoi stimulates Na-dependent glucose absorption in pig jejunum. [source]


Growth performance and body composition of sub-yearling Persian sturgeon, (Acipenser persicus, Borodin, 1897), fed different dietary protein and lipid levels

Summary In order to evaluate the protein and energy requirement of Persian sturgeon (Acipenser persicus) sub-yearlings, eight experimental diets containing two protein levels (40% and 45%) and four lipid levels (10%, 15%, 20% and 25%) were tested. Sturgeons (W0 = 136.8 g) were fed the experimental diets to satiation four times daily for 150 days, resulting in a final mean weight of 375.8 g. Growth was significantly affected by lipid content of the diets. At 40% protein level, weight gain and specific growth rate (% per day) were significantly improved (P < 0.05) by increasing the dietary lipid (energy) content. Protein efficiency ratio (PER) was significantly affected by different dietary treatments for each dietary protein level tested, reaching a mean value of 3.58 in fish fed high lipid diets and a PER of 2.77 in low lipid diets. Results obtained in the present study suggest that the optimum dietary protein content for Persian sturgeon is 40%, with an estimated optimum protein-to-energy ratio of 18,20 mg kJ,1. [source]


Practical diet evaluation with hatchlings of soft-shell turtle, Trionyx sinensis Wiegmann, in the tropics

Summary An 8-week feeding trial was undertaken to evaluate the growth and survival of soft-shell turtle, Trionyx sinensis, hatchlings fed with trash fish (TF), fish meal (FM), poultry by-product meal (PM) and a 1:1 mixture of FM and PM (MIX). No significant difference (P>0.05) was observed for survival; however, the weight gain and carapace length among different dietary treatments were significantly different (P<0.05). The weight gain and carapace length for hatchlings fed with MIX and PM were highest and lowest (P<0.05), respectively. It is concluded that the inexpensive and readily available poultry by-product can be substituted in part for FM in feed formulation for soft-shell turtle culture. [source]


Effects of dietary protein level on growth, feed utilization and digestive enzyme activity of the Chinese mitten crab, Eriocheir sinensis

Abstract A feeding trial was conducted using isoenergetic practical diets to evaluate the effects of the dietary protein level on growth performance, feed utilization and digestive enzyme activity of the Chinese mitten crab, Eriocheir sinensis. Four experimental diets were formulated containing 250, 300, 350 and 400 g kg,1 protein and 16 kJ g,1 gross energy. Each diet was randomly assigned to triplicate groups of juvenile crab with mean initial body weight 3.56 ± 0.16 g and mean shell width 15.31 ± 0.06 mm. Juvenile crab were reared in indoor flow-through system consisting of 12 plastic tanks (1.0 m × 0.6 m × 0.5 m) and fed diets twice daily at 6,8% of body weight for 12 weeks. Performance was judged on the basis of growth (specific growth rate of weight, SGRG; specific growth rate of shell width, SGRSW), feed conversion ratio (FCR) and protein efficiency ratio (PER). A decreased FCR was observed with increasing dietary protein levels. Both SGRG and SGRSW significantly increased with increasing dietary protein levels up to 350 g kg,1, whereas there were no significant differences for protein levels from 350,400 g kg,1. Application of broken line regression analysis to SGRG provided an estimate of 347.8 g kg,1 dietary protein for maximal growth. The highest PER was observed in crab fed the diet containing 350 g kg,1 protein (P < 0.05). The percent survival was not affected (P > 0.05) by the different dietary treatments. No significant differences were observed in the apparent digestibility coefficients of crude lipid and dry matter among dietary treatments (P > 0.05). However, the apparent digestibility coefficients of crude protein and energy in crab fed different protein levels significantly increased with increasing dietary protein level (P < 0.05). Both amylase and protease activities in the intestine of E. sinensis were studied. The amylase activity decreased significantly (P < 0.05) with increased dietary protein level and protease activity increased. Regression analysis showed a negative effect of inclusion of dietary protein level on amylase activity (P < 0.05). However, protease activities were found to be positively correlated (P < 0.05) with dietary protein level. The protein content of the crab significantly increased with dietary protein levels up to 350 g kg,1 (P < 0.05), but no significant differences (P > 0.05) were founded with protein levels higher than 350 g kg,1. Quantitative l -lysine requirement of juvenile black sea bream (Sparus macrocephalus)

Abstract An 8-week feeding experiment was conducted to determine the quantitative l -lysine requirement of juvenile black sea bream Sparus macrocephalus (initial mean weight: 9.13 ± 0.09 g, SD) in eighteen 300-L indoors flow-through circular fibreglass tanks provided with sand-filtered aerated seawater. The experimental diets contained six levels of l -lysine ranging from 20.8 to 40.5 g kg,1 dry diet at about 4 g kg,1 increments. All the experiment diets were formulated to be isoenergetic and isonitrogenous. Each diet was assigned to triplicate groups of 20 fish in a completely randomized design. Weight gain and specific growth rate (SGR) increased with increasing levels of dietary lysine up to 32.5 g kg,1 (P < 0.05) and both showed a declining tendency thereafter. Feed efficiency ratio and protein efficiency ratio was poorer for fish fed the lower lysine level diets (P < 0.05) and showed no significant differences among other treatments (P > 0.05). All groups showed high survival (above 90%) and no significant differences were observed. The whole body crude protein and crude lipid contents were significantly affected (P < 0.05) by dietary lysine level, while moisture and ash showed no significant differences. The composition of muscle and liver also presented similar change tendency. Total essential amino acid and lysine contents in muscle both obtained the highest value when fish fed 32.5 g kg,1 lysine diet (P < 0.05). Serum protein, cholesterol and free lysine concentration were affected by different dietary treatments (P < 0.05), triacylglyceride and glucose contents were more variable and could not be related to dietary lysine levels. Dietary lysine level significantly affected condition factor and intraperitoneal fat ratio of juvenile black sea bream (P < 0.05) except for hepatosomatic index. There were no significant differences in white blood cell count and red blood cell count (P > 0.05), however, haemoglobin level was significantly influenced by different diets (P < 0.05). Analysis of dose (lysine level)-response (SGR) with second order polynomial regression suggested the dietary lysine requirement of juvenile black sea bream to be 33.2 g kg,1 dry diet or 86.4 g lysine kg,1 protein. Preliminary investigations on the effects of dietary lipid on the spawning performance and egg quality of black sea bass Centropristis striata L

Abstract Adult black sea bass Centropristis striata broodstock (N=162) were fed three different dietary treatments: two commercially prepared diets with 45% protein and two different lipid levels (12% and 20%) (diets 1 and 2), and a diet of frozen Atlantic silversides Menidia menidia (SS, diet 3). Broodstock were held under controlled photothermal conditions and induced to spawn with an LHRHa pellet (72 ,g kg,1 bw). Dietary lipid had pronounced effects on spawning performance and egg quality. Diet 3 (SS) produced a significantly (P<0.05) higher fertilization success (22.4%) than diets 1 (0.6%) and 2 (4.8%). The hatching success of fertilized eggs was similar in all diets (range=40,58.6%), but only two spawns from diet 1 (12% lipid) yielded viable yolk-sac larvae (YSL). Diet 3 (SS) also produced significantly more YSL per female (21.8 × 103) than the diet 1 (0.3 × 103). Eggs from diet 3 (SS) contained a significantly greater proportion of n-3 series fatty acids, with docosahexaenoic acid (DHA) as the largest fraction. Eggs from commercially prepared dietary treatments contained significantly more n-6 fatty acids. The poor spawning performance of fish fed diet 1 (12% lipid) may be related to higher levels of linoleic acid and lower levels of DHA in the diet. [source]


Effect of crude oil extracts from trout offal as a replacement for fish oil in the diets of the Australian native fish Murray cod Maccullochella peelii peelii

Abstract The efficacy of trout oil (TO), extracted from trout offal from the aquaculture industry, was evaluated in juvenile Murray cod Maccullochella peelii peelii (25.4±0.81 g) diets in an experiment conducted over 60 days at 23.7±0.8 °C. Five isonitrogenous (48% protein), isolipidic (16%) and isoenergetic (21.8 kJ g,1) diets, in which the fish oil fraction was replaced in increments of 25% (0,100%), were used. The best growth and feed efficiency was observed in fish fed diets containing 50,75% TO. The relationship of specific growth rate (SGR), food conversion ratio (FCR) and protein efficiency ratio (PER) to the amount of TO in the diets was described in each case by second-order polynomial equations (P<0.05), which were: SGR=,0.44TO2+0.52TO+1.23 (r2=0.90, P<0.05); FCR=0.53TO2,0.64TO+1.21 (r2=0.95, P<0.05); and PER=,0.73TO2+0.90TO+1.54 (r2=0.90, P<0.05). Significant differences in carcass and muscle proximate compositions were noted among the different dietary treatments. Less lipid was found in muscle than in carcass. The fatty acids found in highest amounts in Murray cod, irrespective of the dietary treatment, were palmitic acid (16:0), oleic acid (18:1n-9), linoleic acid (18:2n-6) and eicosapentaenoic acid (20:5n-3). The fatty acid composition of the muscle reflected that of the diets. Both the n-6 fatty acid content and the n-3 to n-6 ratio were significantly (P<0.05) related to growth parameters, the relationships being as follows. Percentage of n-6 in diet (X) to SGR and FCR: SGR=,0.12X2+3.96X,32.51 (r2=0.96) and FCR=0.13X2,4.47X+39.39 (r2=0.98); and n-3:n-6 ratio (Z) to SGR, FCR, PER: SGR=,2.02Z2+5.01Z,1.74 (r2=0.88), FCR=2.31Z2,5.70Z+4.54 (r2=0.93) and PER=,3.12Z2,7.56Z+2.80 (r2=0.88) respectively. It is evident from this study that TO could be used effectively in Murray cod diets, and that an n-3:n-6 ratio of 1.2 results in the best growth performance in Murray cod. [source]
