Properties of ZrB2 -Reinforced Ternary Composites,

Different amounts of ZrB2 particles are added to a matrix constituted by AlN+SiC for the production of novel electro-conductive composites by hot pressing. The presence of ZrB2 particles makes the composites electro-conductive and improves many relevant mechanical properties, such as toughness, hardness and strength. The new materials can reach a fracture toughness of 4 MPa.m1/2 and strength of 504 MPa at 1400°C. [source]


Influence of magnesium aluminium silicate on rheological, release and permeation characteristics of diclofenac sodium aqueous gels in-vitro

The effect of magnesium aluminium silicate (MAS) on rheological, release and permeation characteristics of diclofenac sodium (DS) aqueous gels was investigated. DS aqueous gels were prepared using various gelling agents, such as 15% w/w poloxamer 407 (PM407), 1% w/w hydroxypropylmethylcellulose (HPMC), and 1% w/w high and low viscosity grades of sodium alginate (HV-SA and LV-SA, respectively). Different amounts of MAS (0.5, 1.0 and 1.5% w/w) were incorporated into the DS gels. Incorporation of MAS into the DS gels prepared using SA or PM407 caused a statistical increase in viscosity (P<0.05) and a shift from Newtonian flow to pseudoplastic flow with thixotropic property. The DS release rates of these composite gels were significantly decreased (P<0.05) when compared with the control gels. This was due to an interaction between MAS and PM407 or SA, and adsorption of DS onto MAS particles. Moreover, a longer lag time and no change in DS permeation flux were found when MAS was added to the gels. The findings suggest that the rheological characteristics of gels prepared using PM407 or SA could be improved by incorporating MAS. However, the use of MAS could retard the DS release and extend the lag time of DS permeation. [source]


Effectiveness of an ultrafiltration device for use with the enzyme-hydrolysed protein method for determining endogenous ileal nitrogen and amino acid excretion in the pig

Abstract The aim of the work was to perform an in vitro study to determine the effectiveness of Centriprep-10 concentrator devices for use with the enzyme-hydrolysed protein method for the determination of endogenous ileal nitrogen and amino acid flows. Different amounts of enzyme-hydrolysed casein (EHC) were added to tubes containing digesta collected from pigs that had received a protein-free diet for 5,8 days. The samples were centrifuged and then ultrafiltered using Centriprep-10 concentrators. The precipitate from the centrifugation step was added to the retentate from the ultrafiltration, and this material was analysed for nitrogen and amino acids. The ultrafiltrates were also analysed for nitrogen. The amount of nitrogen that was deemed to have originated from the EHC and remained in the precipitate plus retentate fraction of digesta after processing, expressed as a percentage of the total amount of nitrogen added to the tubes as EHC, ranged from 1.0 to 5.0%. The overall mean amounts of amino acid in the precipitate plus retentate fractions originating from the EHC, expressed as a percentage of the amino acids added to the tubes as EHC, ranged from 2.4 to 5.8%. The results demonstrate that with Centriprep-10 concentrators there is a less than complete separation of nitrogen and amino acids originating from EHC from endogenous material in digesta, but for most amino acids this is unlikely to be due to binding of the amino acids to digesta. The incomplete separation of EHC from the endogenous fraction of digesta by Centriprep-10 concentrators may lead to a small overestimation (approximately 2%) of endogenous ileal nitrogen and amino acid flows. © 2001 Society of Chemical Industry [source]


Different amounts of isoflavones in various commercially available soy extracts in the light of gene expression-targeted isoflavone therapy

Abstract Isoflavones are plant-derived, biologically active compounds that are commonly used as natural drugs or diet supplements in the treatment of menopausal symptoms and as antioxidants. Recently, it was proposed that genistein (4,,5,7-trihydroxyisoflavone) may be used in the treatment of patients suffering from Sanfilippo disease (mucopolysaccharidosis type III), a severe genetic disorder for which no therapy is available. A pilot clinical study with this novel therapy, called ,gene expression-targeted isoflavone therapy' (GET IT), indicated that a standardized, genistin-rich soy isoflavone extract is effective in the treatment of such patients. Since various isoflavone-containing products are commercially available, the content of the main isoflavones were measured in such products. Extremely different amounts of isoflavones were determined in various products, from 0.13 to 39 mg per tablet. Only some of these products were found to be effective in inhibition of the synthesis of glycosaminoglycans (compounds whose degradation is severely impaired in mucopolysaccharidoses, including Sanfilippo disease) in cultured fibroblasts. Since in GET IT the dose of genistein is calculated per patient's body weight, the amount of this isoflavone in a tablet is crucial for this therapy. Adsorptive removal of textile dyes from aqueous solutions by dead fungal biomass

Dead fungal biomass prepared from Phanerochaete chrysosporium and Funalia trogii was tested for their efficiency in removal of textile dyes. The effects of contact time, initial dye concentration, amount of dead biomass and agitation rate on dye removal have been determined. Removal of all dyes required a very short time (60 min). Experimental results show that, P. chrysosporium was more effective than F. trogii . An increase in the amount of dead biomass positively affected of the dye removal. The removal efficiency of different amount of biomass was in order 1 g > 0.5 g > 0.2 g > 0.1 g. The highest removal was obtained at 150,200 rpm. Slightly lower removing activities were found at lower agitation rates. This study showed that it was possible to remove textile dyes by dead biomass of P. chrysosporium . Good Maintenance of Exercise-Induced Bone Gain with Decreased Training of Female Tennis and Squash Players: A Prospective 5-Year Follow-Up Study of Young and Old Starters and Controls

Abstract This prospective 5-year follow-up study of 64 adult female racquet sports players and 27 controls assessed the changes in the playing-to-nonplaying arm bone mineral content (BMC) differences to answer three questions: (1) Are training-induced bone gains lost with decreased training? (2) Is the bone response to decreased training different if the playing career has been started before or at puberty rather than after it? (3) Are the possible bone changes related to the changes in training? The players were divided into two groups according to the starting age of their tennis or squash playing. The mean starting age was 10.5 years (SD, 2.2) among the players who had started training before or at menarche (young starters; n = 36) while 26.4 years (SD, 8.0) among those players who had begun training a minimum of 1 year after menarche (old starters; n = 28). At baseline of the 5-year follow-up, the mean age of the young starters was 21.6 years (SD, 7.6) and that of old starters was 39.4 years (SD, 10.5). During the follow-up, the young starters had reduced the average training frequency from 4.7 times a week (2.7) to 1.4 times a week (1.3) and the old starters from 4.0 times a week (1.4) to 2.0 times a week (1.4), respectively. The 5-year follow-up revealed that despite reduced training the exercise-induced bone gain was well maintained in both groups of players regardless of their clearly different starting age of activity and different amount of exercise-induced bone gain. The gain was still 1.3,2.2 times greater in favor of the young starters (at the follow-up, the dominant-to-nondominant arm BMC difference was 22% [8.4] in the humeral shaft of the young starters versus 10% [3.8] in the old starters, and 3.5% [2.4] in controls). In the players, changes in training were only weakly related to changes in the side-to-side BMC difference (rs = 0.05,0.34, all NS), and this was true even among the players who had stopped training completely a minimum 1 year before the follow-up. In conclusion, if controlled interventions will confirm our findings that an exercise-induced bone gain can be well maintained with decreased activity and that the maintenance of the bone gain is independent of the starting age of activity, exercise can be recommended for preventing osteoporosis and related fractures. Wear of human enamel and nano-filled composite resin denture teeth under different loading forces

Summary, To evaluate and correlate the two-body wear of human enamel and nano-filled composite resin teeth with the loading forces used in a dual-axis chewing simulator. Three groups of human enamel and three of nano-filled composite resin teeth were tested in a chewing simulator. Zirconia ceramic balls were used as antagonists. The teeth were tested with three different loading forces (20, 49 and 78 N). Wear was analysed by measuring the volume and vertical substance loss using a laser scanner after 300000 chewing cycles. Data were statistically analysed using two-way anova followed by the Scheffé test (P , 0·05). Spearman correlation test was used to determine whether there was a relationship between the loading force and the degree to which the human enamel and composite resin had worn. An increase in the loading force significantly increased the wear of composite resin and of human enamel. The effect of the loading force on the wear was statistically significant at the 0·001 level. Human enamel showed a lower volume and vertical substance loss than composite resin under loading forces of 20 and 49 N and lower vertical loss under loading force of 78 N. The correlation between the volume loss and loading force was statistically significant (r = 0·616, P < 0·001). Nano-filled composite resin and human enamel exhibited different amount of wear under different loading forces. In general, human enamel showed less vertical substance loss than nano-filled composite resin. [source]


Differential activity patterns in the masseter muscle under simulated clenching and grinding forces

summary, The aim of this study was to investigate (i) whether the masseter muscle shows differential activation under experimental conditions which simulate force generation during clenching and grinding activities; and (ii) whether there are (a) preferentially active muscle regions or (b) force directions which show enhanced muscle activation. To answer these questions, the electromyographic (EMG) activity of the right masseter muscle was recorded with five intramuscular electrodes placed in two deep muscle areas and in three surface regions. Intraoral force transfer and force measurement were achieved by a central bearing pin device equipped with three strain gauges (SG). The activity distribution in the muscle was recorded in four different mandibular positions (central, left, right, anterior). In each position, maximum voluntary contraction (MVC) was exerted in vertical, posterior, anterior, medial and lateral directions. The investigated muscle regions showed different amount of EMG activity. The relative intensity of the activation, with respect to other regions, changed depending on the task. In other words, the muscle regions demonstrated heterogeneous changes of the EMG pattern for the various motor tasks. The resultant force vectors demonstrated similar amounts in all horizontal bite directions. Protrusive force directions revealed the highest relative activation of the masseter muscle. The posterior deep muscle region seemed to be the most active compartment during the different motor tasks. The results indicate a heterogeneous activation of the masseter muscle under test conditions simulating force generation during clenching and grinding. Protrusively directed bite forces were accompanied by the highest activation in the muscle, with the posterior deep region as the most active area. [source]


Wet granulation as innovative and fast method to prepare controlled release granules based on an ion-exchange resin

Abstract The goal of this work was to evaluate the suitability of wet granulation as an innovative and fast method for the preparation of granules containing a drug,resin complex (resinate), having cholestyramine as resin and potassium diclofenac (KD) as drug. Resinate and granules were prepared directly by steam granulation in high shear mixer (method A), using two different amount of resin (granules 1 and 2). For comparison granules 1 were also prepared by conventional batch method followed by steam granulation (method B). All granules showed quite irregular shape, main size fractions between 75 and 500 µm, good flowability and uniform KD distribution. Granules 1A exhibited controlled release profiles at pH 7.4, while granules 2A showed a burst effect due to KD free crystals. FT-IR studies confirmed the complete complexation between resin and KD during the granulation process with method A for granules 1. Finally, the dissolution test of granules 1A in different media revealed a controlled drug release in 12 h, providing the utility of this system for enteric drug delivery. Nanoclay-reinforced syntactic foams: Flexure and thermal behavior

Syntactic foams containing 60 vol% of hollow glass microballoons in epoxy matrix are modified with untreated nanoclays using combined mechanical and ultrasonication methods. Effects of nanoclays on flexure and thermal behavior of syntactic foams are investigated by adding different amount of nanoclays in the range of 1,3% by weight. Microscopic examinations and physical property characterization are performed to determine the interactions among constituent materials and the void formation during fabrication. It is found that the syntactic foams with 2 wt% nanoclays show the highest improvement in flexural properties (,42% strength and ,18% modulus) and dynamic mechanical properties (,30% storage modulus and ,28% loss modulus) properties. Thermal decomposition temperature is found to be unaffected by the addition of nanoclays, whereas a continuous reduction in the coefficient of thermal expansion (CTE) is observed. An examination of failure surface indicates that the failure is initiated on the tension side of the flexure sample due to fracturing of microballoons. POLYM. Rapid determination of short-chain fatty acids in colonic contents and faeces of humans and rats by acidified water-extraction and direct-injection gas chromatography

Abstract Short-chain fatty acids (SCFAs) have attracted much attention recently because of their positive physiological effects. In this work, a rapid and reliable gas chromatographic method for determination of eight SCFAs, in colonic and faecal samples from rats and humans has been developed and validated. The methodology involves extraction of the SCFAs in water before a direct injection procedure on a FFAP capillary column. A stock standard solution containing acetic acid, propionic acid, n -butyric acid, i -butyric acid, n -valeric acid, i -valeric acid, n -caproic acid and n -heptanoic acid was prepared and used. A high line-arity (r2 > 0.9990), low quantification limit (2.38,30.14 µm) and high recovery for most acids were obtained. Acidification of faecal samples was found to be crucial for quantitative determination of the SCFAs, and adjustment of pH to 2,3 was regarded as necessary. Glass wool inserted in the glass liner of the injection port proved effective in preventing the contamination of the column by non-volatiles, and 12% formic acid reduced the ghost peak that appeared gradually after several injections. After validation, the methodology was applied on two faecal samples from rats fed diets containing different amount of dietary fibre and one faecal sample from human fed a normal diet to test the accuracy of the developed method. Solvothermal production of CdS nanorods using polyvinylpyrrolidone as a template

Abstract CdS nanorods were solvothermally produced using Cd(NO3)2 and S powder in ethylenediamine containing different amounts of polyvinylpyrrolidone (PVP). The phase with hexagonal structure was detected using X-ray diffraction (XRD) and selected area electron diffraction (SAED). Their SAED patterns were in accordance with those of the simulations. Scanning and transmission electron microscopies (SEM and TEM) revealed the presence of CdS nanorods with their lengths influenced by different amounts of PVP. The nanorods were also characterized using high resolution TEM (HRTEM). They grew in the [001] direction normal to the (002) parallel crystallographic planes composing the nanorods. Raman spectra showed the 1LO (first harmonic) and 2LO (second harmonic) modes at the same wavenumbers although the products were produced under different conditions. Critical and sensitive periods for reversing the effects of mechanosensory deprivation on behavior, nervous system, and development in Caenorhabditis elegans

Abstract In these studies the nematode Caenorhabditis elegans was used as a model to investigate ways to reverse the effects of mechanosensory deprivation on behavior and development. Rose et al. (J Neurosci 2005; 25:7159,7168) showed that worms reared in isolation responded significantly less to a mechanical tap stimulus, were significantly smaller, and expressed significantly lower levels of a postsynaptic glutamate receptor subunit on the command interneurons of the tap response circuit and a presynaptic vesicle marker in the tap sensory neurons compared with worms raised in groups. Here, brief mechanical stimulation at any time throughout development reversed the effects of isolation on the response to tap and on postsynaptic glutamate receptor expression on the command interneurons, suggesting there is no critical period for these measures. In contrast to the high level of plasticity in glutamate receptor subunit expression on the interneurons, low levels of stimulation only rescued vesicle expression in the tap sensory neurons early in development and progressively higher levels of stimulation were required as the worm developed, suggesting a sensitive period immediately after hatching, followed by a period of decreasing plasticity. Stimulation during the first three stages of larval development, but not later, rescued the effects of isolation on worm length, suggesting there is a critical period for this measure that ends in the third larval stage. The costs and benefits of fast living

Abstract Growth rates play a fundamental role in many areas of biology (Q. Rev. Biol., 67, 1992, 283; Life History Invariants. Some Explorations of Symmetry in Evolutionary Biology, 1993; Philos. Trans. R. Soc. Lond. B Biol. Sci., 351, 1996, 1341; Plant Strategies, Vegetation Processes, and Ecosystem Properties, 2002; Trends Ecol. Evol., 18, 2003, 471; Q. Rev. Biol., 78, 2003, 23; J. Ecol., 95, 2007, 926.) but the cost and benefits of different growth rates are notoriously difficult to quantify (Q. Rev. Biol., 72, 1997, 149; Funct. Ecol., 17, 2003, 328). This is because (1) growth rate typically declines with size and yet the most widely used growth measure , relative growth rate or RGR (conventionally measured as the log of the ratio of successive sizes divided by the time interval) , is not size-corrected and so confounds growth and size, (2) organisms have access to different amounts of resource and (3) it is essential to allow for the long-term benefits of larger size. Here we experimentally demonstrate delayed costs and benefits of rapid growth in seven plant species using a novel method to calculate size-corrected RGR. In control treatments, fast-growing plants benefited from increased reproduction the following year; however, fast-growing plants subjected to an experimental stress treatment (defoliation) showed strongly reduced survival and reproduction the following year. Importantly, when growth was estimated using the classical RGR measure, no costs or benefits were found. These results support the idea that life-history trade-offs have a dominant role in life-history and ecological theory and that the widespread failure to detect them is partly due to methodological shortcomings. Voltammetric Monitoring and Speciation of Copper Ions in Italian "Grappa" with Platinum Microelectrodes

Abstract A linear sweep voltammetric (LSV) investigation and the anodic stripping voltammetric (ASV) detection of copper ions in ethanol-water mixtures and grappa samples are reported. The measurements are carried out by using platinum microdisk electrodes. Ethanol-water mixtures with ethanol content in the range 40,100,vol%, commercially available and raw grappa samples having ethanol content in the above range are examined. From LSV measurements of copper (II) ions added to the samples, the formation of intermediate copper (I) soluble species, which are stabilized mainly by the naturally occurring organic compounds present in the real samples, is observed. The analysis of LSV and ASV current responses against added Cu2+ ions provides linear trends over the concentration range 5×10,5,5×10,3,M and 5×10,7,5×10,5,M, respectively. The sensitivity depends on the ethanol content in the mixture and, as expected, it is the higher the lower the viscosity of the medium. In particular, it varies from 1.54 to 3.53,nA mM,1 and from 0.114 to 0.263,nA ,M,1 for LSV and ASV measurements, respectively, upon changing the ethanol content from 40 to 100,vol%. In the same range of ethanol content, detection limits obtained by ASV vary from 0.27 to 0.15,,M, respectively. Labile or total copper contents in the grappa samples are determined by ASV measurements performed in the untreated matrices or in the samples acidified with 0.1,M HClO4, respectively. Finally, acidification of the samples with different amounts of HClO4 allows also some speciation investigations to be performed. [source]


Crash of a population of the marine heterotrophic flagellate Cafeteria roenbergensis by viral infection

Summary Viruses are known as important mortality agents of marine microorganisms. Most studies focus on bacterial and algal viruses, and few reports exist on viruses infecting marine heterotrophic protists. Here we show results from several incubations initiated with a microbial assemblage from the central Indian Ocean and amended with different amounts of organic matter. Heterotrophic flagellates developed up to 30 000 cells ml,1 in the most enriched incubation. A 18S rDNA clone library and fluorescent in situ hybridization counts with newly designed probes indicated that the peak was formed by Cafeteria roenbergensis and Caecitellus paraparvulus (90% and 10% of the cells respectively). Both taxa were below detection in the original sample, indicating a strong positive selective bias during the enrichment. During the peak, C. roenbergensis cells were observed with virus-like particles in the cytoplasm, and 4 days later this taxa could not be detected. Transmission electron microscopy confirmed the viral nature of these particles, which were large (280 nm), had double-stranded DNA, and were produced with a burst size of ,70. This virus was specific of C. roenbergensis as neither C. paraparvulus that was never seen infected, nor other flagellate taxa that developed in later stages of the incubation, appeared attacked. This is one of the few reports on a heterotrophic flagellate virus and the implications of this finding in the Indian Ocean are discussed. [source]


A method for the determination of polyunsaturated fatty acid methyl esters in biodiesel: Results of an interlaboratory study

Abstract A gas chromatographic method for the determination of fatty acid methyl esters with four or more double bonds in biodiesel was developed and tested. The method is based on gas chromatographic separation on a wax capillary column using methyl tricosanoate as internal standard. The performance of the method was proved with the participation of 11,European laboratories by a Round Robin test on six different biodiesel samples containing different amounts of polyunsaturated fatty acid methyl esters. The results showed that the precision is sufficient around the EN,14214 limit of 1,% (m/m). At lower concentrations the variation is too high. The scope of the application can be given between as 0.6 and 1.5%. [source]


Neural activity related to the processing of increasing monetary reward in smokers and nonsmokers

Abstract This study investigated the processing of increasing monetary reward in nonsmoking and smoking subjects. The choice of the subject populations has been motivated by the observation of differences between nonsmokers and smokers in response to rewarding stimuli in a previous study. Subjects performed a pattern recognition task with delayed response, while rCBF was measured with [\mathrm{H}^{15}_{2}O] PET. Correct responses to the task were reinforced with three different amounts of monetary reward. The subjects received the sum of the rewards at the end of the experiment. The results show that a cortico-subcortical loop, including the dorsolateral prefrontal cortex, the orbitofrontal cortex, the cingulate gyrus and the thalamus is involved in processing increasing monetary reward. Furthermore, the striatal response differentiates nonsmokers from smokers. Thus, we found significant correlations between rCBF increases in striatum and increasing monetary reward and between striatal rCBF increases and mood in nonsmokers, but not in smokers. Moreover, no significant mood changes among the different monetary rewards could be observed in smokers. We infer that the response of the striatum to reward is related to changes in subjective feelings. The differences between smokers and nonsmokers confirm our previous conclusions that the association between blood flow, performance, mood and amount of reward is more direct in nonsmokers. Hydroxyapatite/SiO2 Composites via Freeze Casting for Bone Tissue Engineering,

Freeze casting is a fabrication method that allows producing near-net-shaped ceramics with variable porosity. Hydroxyapatite (HA) was modified by the addition of different amounts of SiO2 nanoparticles during freeze cast preparation. The addition of SiO2 introduced a partial phase transformation of HA to , -tricalcium phosphate and improved the form stability due to less shrinkage after sintering. The impact of surface roughness of pure HA ceramics and the influence of SiO2 introduction during freeze casting on adhesion, proliferation, and differentiation of human osteoblast-like cells (MG-63) was investigated. While both cell attachment and proliferation of smooth pressed HA was significantly enhanced compared to rough freeze cast HA, the addition of SiO2 improved the cell numbers of the latter. Microfibril-associated glycoprotein-1 binding to tropoelastin

Multiple binding sites, the role of divalent cations
Microfibrils and elastin are major constituents of elastic fibers, the assembly of which is dictated by multimolecular interactions. Microfibril-associated glycoprotein-1 (MAGP-1) is a microfibrillar component that interacts with the soluble elastin precursor, tropoelastin. We describe here the adaptation of a solid-phase binding assay that defines the effect of divalent cations on the interactions between MAGP-1 and tropoelastin. Using this assay, a strong calcium-dependent interaction was demonstrated, with a dissociation constant of 2.8 ± 0.3 nm, which fits a single-site binding model. Manganese and magnesium bestowed a weaker association, and copper did not facilitate the protein interactions. Three constructs spanning tropoelastin were used to quantify their relative contributions to calcium-dependent MAGP-1 binding. Binding to a construct spanning a region from the N-terminus to domain 18 followed a single-site binding model with a dissociation constant of 12.0 ± 2.2 nm, which contrasted with the complex binding behavior observed for fragments spanning domains 17,27 and domain 27 to the C-terminus. To further elucidate binding sites around the kallikrein cleavage site of domains 25/26, MAGP-1 was presented with constructs containing C-terminal deletions within the region. Construct M1659, which spans a region from the N-terminus of tropoelastin to domain 26, inclusive, bound MAGP-1 with a dissociation constant of 9.7 ± 2.0 nm, which decreased to 4.9 ± 1.0 nm following the removal of domain 26 (M155n), thus displaying only half the total capacity to bind MAGP-1. These results demonstrate that MAGP-1 is capable of cumulative binding to distinct regions on tropoelastin, with different apparent dissociation constants and different amounts of bound protein. Lep d 2 polymorphisms in wild and cultured Lepidoglyphus destructor mites

We have previously cloned, expressed and characterized two variants of the major allergen Lep d 2 from cultured Lepidoglyphus destructor mites. These variants, Lep d 2.0101 and Lep d 2.0201, differ at 13 amino acid positions. In this study we investigated Lep d 2 sequence diversity between wild and cultured mites. PCR, Southern blot and DNA sequence analysis revealed the presence of two different Lep d 2 genes, one with and one without an intron. In addition, two new variants of Lep d 2, Lep d 2.0102 and Lep d 2.0202, were found at different frequencies in wild and cultured mites. When we expressed the Lep d 2 variants and compared their IgE binding properties by ELISA inhibition, we found that Lep d 2.0102 was a more potent inhibitor than Lep d 2.0101, and to a lesser extent Lep d 2.0202 was more potent than Lep d 2.0201. Long-term cultures of peripheral blood mononuclear cells were used to assess the ability of the expressed Lep d 2 variants to induce cytokine release. Although cells from different individuals released different amounts of interferon-, and interleukin-5, no consistent cytokine release pattern could be linked to any specific Lep d 2 variant. In conclusion, we show that both cultured and wild Lepidoglyphus destructor mites contain the same pattern of polymorphism. Furthermore, this Lep d 2 sequence diversity seems not to have any significant impact on the allergens IgE binding or its ability to induce T cell cytokine release. [source]


Reversible transition between active and dormant microbial states in soil

Abstract The rate of respiration obtained in the substrate-induced respiration (SIR) method can be divided into the respiration rate of growing (r) and non-growing (K) microorganisms. The fraction of r is generally small (5,20%) in soils with no recent addition of substrates, but can be 100% in soils with high substrate availability. This suggests that substrate availability determines the proportion of biomass between these groups, and implies that transitions between them can take place reversibly. These hypotheses were tested by adding three different amounts of glucose which induced first-order, zero-order, and growth-associated respiration kinetics to three soils at four pre-incubation times (4, 12, 27, and 46 days) before the SIR measurement. An abiotic flush of CO2 in the SIR measurement was detected and corrected for before data analysis. Accumulated CO2 -C over 4 days after glucose addition, corrected for the respiration in unamended controls, corresponded to 41,50% mineralization of the glucose-C, and the relative amount mineralized by each soil was independent of the glucose amount added. The high glucose concentration gave an increased SIR, which reverted to the initial value within 27,46 days. In a specific sample, the maximum respiration rate induced during the pre-incubation, and the amount of organisms transformed from the K to the r state, as quantified in respiration rate units in the SIR measurement, were identical to each other, and these parameters were also highly correlated to the initial glucose concentration. The K,r transition was very fast, probably concurrent with the instantaneous increase in the respiration rate obtained by the glucose amendment. Thereafter, a slow first-order back-transition from the r to the K state ensued, with half-lives of 12, 23, and 70 days for the three soils. The results suggest the existence of community-level controls by which growth within or of the whole biomass is inhibited until it has been completely transformed into the r state. The data also suggest that the microbial specific activity is not related to the availability of exogenous substrate in a continuous fashion, rather it responds as a sharp transition between dormant and fully active. Furthermore, the inherent physiological state of the microbial biomass is strongly related to its history. It is proposed that the normal dynamics of the soil microbial biomass is an oscillation between active and dormant physiological states, while significant growth occurs only at substantial substrate amendment. Effects of size and fragmentation of marine reserves and fisher infringement on the catch and biomass of coral trout, Plectropomus leopardus, on the Great Barrier Reef, Australia

Abstract, A spatially structured simulation model of the population dynamics and line fishing exploitation of common coral trout, Plectropomus leopardus Lacepčde, was used to evaluate the effects of infringement and different amounts and arrangements of marine reserves on the Reef Line Fishery of the Great Barrier Reef, Australia. With no marine reserves and under a constant future effort level equal to that for 1996, the size of the population was reduced and the biomass stabilised at about 40% of pre-exploitation levels. Marine reserves were ineffective at conserving biomass when limited infringement was allowed throughout an entire reserve. When infringement was absent altogether or limited to the edges of reserves, larger marine reserves lead to lower total catches and higher overall biomass. When infringement was limited to reserve edges, a single large closure was more effective at conserving biomass than more fragmented arrangements. Simulations suggested that marine reserves might lead to better conservation of a fishery-targeted species if infringement is negligible or limited to reserve margins. Even where infringement occurred only at the edges of reserves, a network of small reserves may be less effective at conserving a targeted species than a smaller number of larger reserves. [source]

Summary 1Large female insects usually have high potential fecundity. Therefore selection should favour an increase in body size given that these females get opportunities to realize their potential advantage by maturing and laying more eggs. However, ectotherm physiology is strongly temperature-dependent, and activities are carried out sufficiently only within certain temperature ranges. Thus it remains unclear if the fecundity advantage of a large size is fully realized in natural environments, where thermal conditions are limiting. 2Insect fecundity might be limited by temperature at two levels; first eggs need to mature, and then the female needs time for strategic ovipositing of the egg. Since a female cannot foresee the number of oviposition opportunities that she will encounter on a given day, the optimal rate of egg maturation will be governed by trade-offs associated with egg- and time-limited oviposition. As females of different sizes will have different amounts of body reserves, size-dependent allocation trade-offs between the mother's condition and her egg production might be expected. 3In the temperate butterfly Pararge aegeria, the time and temperature dependence of oviposition and egg maturation, and the interrelatedness of these two processes were investigated in a series of laboratory experiments, allowing a decoupling of the time budgets for the respective processes. 4The results show that realized fecundity of this species can be limited by both the temperature dependence of egg maturation and oviposition under certain thermal regimes. Furthermore, rates of oviposition and egg maturation seemed to have regulatory effects upon each other. Early reproductive output was correlated with short life span, indicating a cost of reproduction. Finally, large females matured more eggs than small females when deprived of oviposition opportunities. Thus, the optimal allocation of resources to egg production seems dependent on female size. 5This study highlights the complexity of processes underlying rates of egg maturation and oviposition in ectotherms under natural conditions. We further discuss the importance of temperature variation for egg- vs. time-limited fecundity and the consequences for the evolution of female body size in insects. [source]


Effects of ultraviolet radiation on litter decomposition depend on precipitation and litter chemistry in a shortgrass steppe ecosystem

Abstract We examined the effect of altered levels of ultraviolet (UV) radiation (280,400 nm) and different amounts of precipitation on the decomposition rates of litter of contrasting carbon to nitrogen ratio (C : N) in a 3-year field experiment in a shortgrass steppe (SGS) ecosystem. UV radiation was either blocked or passed under clear plastic tents where precipitation was applied to simulate a very dry or very wet year. These treatments minimized or maximized the abiotic component (UV) or the biotic component (biological activity of decomposer organisms) of decomposition to assess potential interactions between the two. Initial litter chemistry varied in response to having been grown under ambient or elevated atmospheric CO2 concentrations. While precipitation and litter chemistry were the most important drivers in decomposition in this system, UV radiation increased decomposition rates under dry conditions in litter with higher C : N ratios. Exposure to UV radiation slightly increased the amount of holocellulose that was lost from the litter. UV exposure did not affect the decomposition of the lignin fraction. Increased decomposition with UV radiation was accompanied by a decrease in N immobilization over the summer months. These results suggest that the effects of UV radiation on decomposition rates may be primarily abiotic, caused by direct photochemical degradation of the litter. Our results demonstrate that the role of UV radiation in litter decomposition in semiarid systems depends on the aridity of the system and the chemistry of the litter. [source]


Responses of shoot growth and survival to water stress gradient in diploid and tetraploid populations of Lolium multiflorum and L. perenne

Abstract Drought stress is one of the critical environmental factors in determining growth and survival of herbage grasses. In this study, by using a hydroponic culture system including different amounts of polyethylene glycol (PEG), responses of plant shoots to water stress in four different intensities (0 Mpa, ,0.6 Mpa, ,1.2 Mpa and ,1.8 Mpa) were examined in diploid and tetraploid cultivars of Italian ryegrass (Lolium multiflorum) and perennial ryegrass (L. perenne). Since freezing injury is caused by cell dehydration, freezing tolerance was also examined for six subzero temperatures (,11, ,12, ,14, ,16, ,18 and ,20°C) in both species. L. multiflorum had a larger shoot biomass at all stress intensities and a lower survival rate under severe water stress and freezing stress conditions than L. perenne. Thus, there was a trade-off (negative correlation) between potential growth under a stress-free condition and survival under a severe stress condition in diploid and tetraploid cultivars of both species. This trade-off was mediated by tissue water content. High water content led to a high growth rate through increasing specific leaf area, while low water content resulted in a high tissue osmotic potential that could confer high cell dehydration tolerance. Multiple Functionalization of Mesoporous Silica in One-Pot: Direct Synthesis of Aluminum-Containing Plugged SBA-15 from Aqueous Nitrate Solutions,

Abstract Aluminum-containing plugged mesoporous silica has been successfully prepared in an aqueous solution that contains triblock copolymer templates, nitrates, and silica sources but without using mineral acid. The acidity of the solution can be finely tuned from pH 1.4 to 2.8 according to the amount of the introduced aluminum species which ranged from an Al/Si molar ratio of 0.25/1 to 4.0/1. The aluminum nitrate additive in the starting mixture, along with the weak acidity produced by the nitrates, contributes to the formation of plugged hexagonal structures and the introduction of different amounts of aluminum species into the mesostructure. Characterization by X-ray diffraction, transmission electron microscopy, and N2 sorption measurements show that the Al-containing plugged silicas possess well-ordered hexagonal mesostructures with high surface areas (700,860 m2,g,1), large pore volume (0.77,1.05 cm3,g,1) and, more importantly, combined micropores and/or small mesopores in the cylindrical channels. Inductively coupled plasma,atomic emission spectrometry results show that 0.7,3.0 wt,% aluminum can be introduced into the final samples. 27Al MAS NMR results display that about 43,60% aluminum species are incorporated into the skeleton of the Al-containing silicas and the amount of the framework aluminum increases as the initial added nitrates rises. Scanning electron microscopy images reveal that the directly synthesized Al-containing plugged silica has a similar morphology to that of traditional SBA-15. Furthermore, the Al-containing plugged samples have excellent performances in the adsorption and the catalytic decomposition of isopropyl alcohol and nitrosamine. Finally, the direct synthesis method is used to produce plugged mesoporous silicas that contain other metals such as chromium and copper, and the resultant samples also show good catalytic activities. A welfare analysis of social security in a dynastic framework*

In this article, we study the welfare effects of unfunded social security in a general equilibrium model populated with overlapping generations of altruistic individuals that differ in lifetime expectancy and earnings ability. Contrary to previous research, our results indicate that steady-state welfare increases with social security for most households, although by very different amounts. This result is mainly due to two factors. First, the presence of two-sided altruism significantly mitigates the crowding out effect of unfunded social security. Second, ability shocks and uncertain lifetimes generate significant heterogeneity among households to yield different induced preferences for social security. [source]


Influence of residual milk-clotting enzyme and proteolysis on melting properties of soft cheese

In this work, we assessed the influence of coagulant residual activity and primary proteolysis on Cremoso Argentino cheese melting properties. For that purpose, we made Cremoso soft cheeses using different amounts of coagulant, and also obtained samples in which milk-clotting enzyme was inactivated. Primary proteolysis correlated with residual activity of coagulant in early stages of cheese ripening; however, it was similar in all cheeses after 30 days. The hydrolysis of caseins did not significantly affect the melting ability of the cheeses, expressed as the area increase after heating samples under standardized conditions. Samples with similar proximate composition showed some changes in meltability; those seemed related to pH evolution during ripening. [source]


Detecting vegetable oil adulteration in hazelnut paste (Corylus avellana L.)

Summary The possibilities of detecting hazelnut paste adulterated with refined and non-refined vegetable oils have been studied. Research was focussed mainly on peanut, high oleic-acid sunflower, corn and soybean oils which have a similar composition to hazelnut oil. The analytical procedures to detect fatty acid (FA), triacylglycerol (TAG) and tocopherol profiles as indicators of adulteration were determined. The better indicators experimentally determined were seven FA (palmitic, stearic, linoleic, linolenic, arachidonic, behenic and lignoceric acids) and different TAG with three unsaturated FA (the code letters used for FA are: P = C16:0; S = C18:0; O = C18:1; L = C18:2;; Ln = C18:3) (LLLn, LLL and OOO), two unsaturated FA (POL, PLL and SOO), and one unsaturated FA (PPL). As expected, when refined vegetable oils were added to hazelnut paste, the increment of stigmasta-3,5-diene allowed detection at levels of 2% oil added. Limits of detection were measured using standard and adulterated hazelnut with different amounts of non-refined vegetable oils added (5%, 10%, 20% and 30%). The distribution of tocopherols and tocotrienols is highly useful, except in the case of added sunflower oil. The differences between the experimental and theoretical values of the TAG with equivalent carbon number (,ECN) of 42 does not improve the detection limit of hazelnut paste adulterated with peanut or sunflower oils. Similarly, tocopherols usually added to refined vegetable oils as an antioxidant were also determined. [source]
