Analysis of nubbin expression patterns in insects

Summary Previous studies have shown that the gene nubbin (nub) exhibits large differences in expression patterns between major groups of arthropods. This led us to hypothesize that nub may have evolved roles that are unique to particular arthropod lineages. However, in insects, nub has been studied only in Drosophila. To further explore its role in insects in general, we analyzed nub expression patterns in three hemimetabolous insect groups: zygentomans (Thermobia domestica, firebrat), dyctiopterans (Periplaneta americana, cockroach), and hemipterans (Oncopeltus fasciatus, milkweed bug). We discovered three major findings. First, observed nub patterns in the ventral central nervous system ectoderm represent a synapomorphy (shared derived feature) that is not present in other arthropods. Furthermore, each of the analyzed insects exhibits a species-specific nub expression in the central nervous system. Second, recruitment of nub for a role in leg segmentation occurred early during insect evolution. Subsequently, in some insect lineages (cockroaches and flies), this original role was expanded to include joints between all the leg segments. Third, the nub expression in the head region shows a coordinated change in association with particular mouthpart morphology. This suggests that nub has also gained an important role in the morphological diversification of insect mouthparts. Overall, the obtained data reveal an extraordinary dynamic and diverse pattern of nub evolution that has not been observed previously for other developmental genes. [source]


Uptake and diverse effects of polycyclic aromatic hydrocarbons on the metabolic activity of Elliptio complanata measured by calorespirometry

Abstract Polycyclic aromatic hydrocarbons (PAHs) are important contaminants of world water resources, with effects on aquatic life. Metabolic responses to short-term acute toxicities of naphthalene, anthracene, and chrysene were investigated in the freshwater bivalve mollusk Elliptio complanata using differential scanning calorespirometry coupled with uptake and scanning electron microscopy. Comparing the uptakes of naphthalene, anthracene, and chrysene with that of inulin, which is known to occupy only extracellular space, showed that all compounds studied were taken up. The PAHs studied had diverse effects on the metabolic activity of E. complanata. Naphthalene and, to a lesser degree, chrysene caused stimulation of heat rates, possibly due to uncoupling of oxidative metabolism. Differential scanning calorespirometry coupled with studies of rates of oxygen consumption by the gill tissue exposed to the PAHs showed similar diverse patterns of respiratory rate stimulation and inhibition. Analysis of results of scanning electron microscopy suggested that irreversible damage to the gill tissue occurred in the presence of anthracene but not in the presence on naphthalene or chrysene. The batch calorespirometric method coupled with uptake and spectroscopy proved to be a useful technique to assess the toxicity of PAHs on the control of energy flux in gills of a freshwater bivalve mollusk. Variation in Reproductive Behaviour within a Sex:Neural Systems and Endocrine Activation

Abstract Intrasexual variation in reproductive behaviour, morphology and physiology is taxonomically widespread in vertebrates, and is as biologically and ecologically significant as the differences between the sexes. In this review, we examine the diverse patterns of intrasexual variation in reproductive behaviours within vertebrates. By illustrating the genetic, cellular, hormonal and/or neural mechanisms underlying behavioural variation in a number of species, another level of complexity is added to studies of brain organization and function. Such information increases our understanding of the unique and conserved mechanisms underlying sex and individual differences in behaviour in vertebrates as a whole. Here, we show that intrasexual variation in behaviour may be discrete or continuous in nature. Moreover, this variation may be due to polymorphism at a single genetic locus or many loci, or may even be the result of phenotypic plasticity. Phenotypic plasticity simply refers to cases where a single genotype (or individual) can produce (or display) different phenotypes. Defined in this way, plasticity subsumes many different types of behavioural variation. For example, some behavioural phenotypes are established by environmental factors during early ontogeny, others are the result of developmental transitions from one phenotype early in life to another later in life, and still other strategies are facultative with different behaviours displayed in different social contexts. On Nursing Theories and Evidence

Purpose: To expand the understanding of what constitutes evidence for theory-guided, evidence-based nursing practice from a narrow focus on empirics to a more comprehensive focus on diverse patterns of knowing. Organizing construct: Carper's four fundamental patterns of knowing in nursing,empirical, ethical, personal, and aesthetic,are required for nursing practice. A different mode of inquiry is required to develop knowledge about and evidence for each pattern. Conclusions: Theory, inquiry, and evidence are inextricably linked. Each pattern of knowing can be considered a type of theory, and the modes of inquiry appropriate to the generation and testing of each type of theory provide diverse sources of data for evidence-based nursing practice. Different kinds of nursing theories provide different lenses for critiquing and interpreting the different kinds of evidence essential for theory-guided, evidence-based holistic nursing practice. Genetic composition of Brazilian population samples based on a set of twenty-eight ancestry informative SNPs

Ancestry informative SNPs can be useful to estimate individual and population biogeographical ancestry. Brazilian population is characterized by a genetic background of three parental populations (European, African, and Brazilian Native Amerindians) with a wide degree and diverse patterns of admixture. In this work we analyzed the information content of 28 ancestry-informative SNPs into multiplexed panels using three parental population sources (African, Amerindian, and European) to infer the genetic admixture in an urban sample of the five Brazilian geopolitical regions. The SNPs assigned apart the parental populations from each other and thus can be applied for ancestry estimation in a three hybrid admixed population. Data was used to infer genetic ancestry in Brazilians with an admixture model. Pairwise estimates of Fst among the five Brazilian geopolitical regions suggested little genetic differentiation only between the South and the remaining regions. Estimates of ancestry results are consistent with the heterogeneous genetic profile of Brazilian population, with a major contribution of European ancestry (0.771) followed by African (0.143) and Amerindian contributions (0.085). The described multiplexed SNP panels can be useful tool for bioanthropological studies but it can be mainly valuable to control for spurious results in genetic association studies in admixed populations. Neo-liberalization and Incorporation in Advanced Newly Industrialized Countries: A View from South Korea

Historically closely associated with interventionist growth strategies, the demise of corporatism was widely predicted as neo-liberalism became the dominant economic paradigm from the late 1970s. The experience of social dialogue and social pacts in continental Europe during the 1980s and 90s, however, suggests that corporatism may have found a new economic purpose in the era of neo-liberalism. Similarly, in the developing world, the intellectual aversion to government accommodation of ,special interest groups' that accompanied the early phase of economic liberalization has given way to a more sophisticated appreciation of the economic advantages of social compromise. Using the case of South Korea, a country that recently established a social pact in support of its economic liberalization drive, this paper seeks to extend our knowledge about the compatibility between neo-liberalism and incorporation. The results contribute to the wider debate about the possibilities of diverse patterns of government,business,labour relations under ,actually existing' neo-liberalism. As one of the most advanced and successful newly industrialized countries, South Korea shares structural,institutional affinities with both advanced and developing societies. Hence, it represents an intermediate case of relevance to both the mature European and ,third wave' democracies. [source]


Customising an antibody leukocyte capture microarray for systemic lupus erythematosus: Beyond biomarker discovery

Abstract Systemic lupus erythematosus (SLE) is a complex autoimmune disease that has heterogeneous clinical manifestation with diverse patterns of organ involvement, autoantibody profiles and varying degrees of severity of disease. Research and clinical experience indicate that different subtypes of SLE patients will likely benefit from more tailored treatment regimes, but we currently lack a fast and objective test with high enough sensitivity to enable us to perform such sub-grouping for clinical use. In this article, we review how proteomic technologies could be used as such an objective test. In particular, we extensively review many leukocyte surface markers that are known to have an association with the pathogenesis of SLE, and we discuss how these markers can be used in the further development of a novel SLE-specific antibody leukocyte capture microarray. In addition, we review some bioinformatics challenges and current methods for using the data generated by these cell-capture microarrays in clinical use. In a broader context, we hope our experience in developing a disease specific cell-capture microarray for clinical application can be a guide to other proteomic practitioners who intend to extend their technologies to develop clinical diagnostic and prognostic tests for complex diseases. The soybean Dof-type transcription factor genes, GmDof4 and GmDof11, enhance lipid content in the seeds of transgenic Arabidopsis plants

Summary Soybean is one of the most important leguminous seed crops among the oil crops. Although the pathways for lipid biosynthesis have been identified, the factors that regulate the biosynthetic pathways at the transcriptional level are largely unknown. Here, we report our findings on the involvement of soybean Dof-type transcription factor genes in the regulation of the lipid content in soybean seeds. We identified 28 Dof-type transcription factor genes in soybean plants, and these genes displayed diverse patterns of expression in various organs. Seven flower/pod-specific genes and one constitutively expressed gene were further investigated. The proteins encoded by these seven genes were localized in the nucleus, and exhibited different abilities for transcriptional activation and DNA binding. Two genes, GmDof4 and GmDof11, were found to increase the content of total fatty acids and lipids in GmDof4 and GmDof11 transgenic Arabidopsis seeds. We also found that the 1000-seed weight was increased in the GmDof4 and GmDof11 transgenic plants. Using microarray and DNA binding analysis, we found that the two Dof-like proteins, GmDof4 and GmDof11, activated the acetyl CoA carboxylase gene and long-chain-acyl CoA synthetase gene, respectively, by direct binding to the cis -DNA elements in their promoter regions. In addition, both proteins downregulated the storage protein gene, CRA1, through direct binding. These results suggest that the two GmDof genes may augment the lipid content of soybean seeds by upregulating genes that are associated with the biosynthesis of fatty acids. [source]
