Badyaev Divergent selection on traits involved in both local adaptation and the production of mating signals can strongly facilitate population differentiation. Because of its links to foraging morphologies and cultural inheritance song of birds can contribute particularly strongly to maintenance of local adaptations. In two adjacent habitats,native Sonoran desert and urban areas,house finches (Carpodacus mexicanus) forage on seeds that are highly distinct in size and shell hardness and require different bite forces and bill morphologies. Here, we first document strong and habitat-specific natural selection on bill traits linked to bite force and find adaptive modifications of bite force and bill morphology and associated divergence in courtship song between the two habitats. Second, we investigate the developmental basis of this divergence and find that early ontogenetic tissue transformation in bill, but not skeletal traits, is accelerated in the urban population and that the mandibular primordia of the large-beaked urban finches express bone morphogenetic proteins (BMP) earlier and at higher level than those of the desert finches. Further, we show that despite being geographically adjacent, urban and desert populations are nevertheless genetically distinct corroborating findings of early developmental divergence between them. Taken together, these results suggest that divergent selection on function and development of traits involved in production of mating signals, in combination with localized learning of such signals, can be very effective at maintaining local adaptations, even at small spatial scales and in highly mobile animals. Poon Abstract: Examining "embedded" economic and social relations has become a popular theme among economic geographers who are interested in explaining the durability of place in supporting economic activities. This article explores the relationship between embeddedness and technology-oriented functions among three types of subsidiaries (regional headquarters, regional offices, and local offices) and for four cities: Hong Kong, Shanghai, Singapore, and Sydney. Using survey data from firms, we show that quiescent or branch plant-like subsidiaries, rather than developmental firms, dominate the region. But among developmental subsidiaries, returns on embeddedness are not always obvious. Embeddedness and developmental subsidiaries are most significantly correlated with manufacturing regional headquarters. However, a small group of subsidiaries (local and regional offices) also perform developmental functions, despite their relative newness and lack of embed-dedness in the region. [source] Developmental and Therapeutic Pharmacology of Antiepileptic DrugsEPILEPSIA, Issue 2000Hisao Miura Summary: We investigated the clinical effects and plasma levels of zonisamide (ZNS) in children with cryptogenic localization-related epilepsies. ZNS is absorbed slowly from the gastrointestinal tract, and its biological half-life is long as compared with that of other common antiepileptic drugs. The peak-to-trough plasma level ratios during a day were as small as 1.28 ± 0.15 in children taking a daily dose of 8 mg/kg of ZNS once a day as a single drug. The plasma level (,g/ml) to dose (mg/kg/day) ratios estimated by the trough and peak plasma levels both increased with advancing age, but the peak-to-trough plasma level ratios were maintained almost uniformly throughout the pediatric age period. A wide range of the plasma levels was associated with complete freedom from seizures. The range of the plasma levels in patients who did not respond to ZNS was higher than that in the controlled group. However, the clinical effects of ZNS were in agreement with the range of generally accepted therapeutic plasma levels of ZNS, 15,40 ,g/ml. Any patient who receives polytherapy is at risk to develop 1 or more drug interactions. Concurrent administration of carbamazepine (CBZ) decreases plasma concentrations of ZNS. However, ZNS does not alter plasma concentrations of CBZ or its primary metabolite, carbamazepine-10,11-epoxide (CBZ-E). It is evident that the concurrent administration of lamotrigine (LTG) affects plasma concentrations of CBZ-E, while plasma CBZ levels remain unaltered. However, the effect of LTG on plasma concentrations of CBZ-E is small, and none of the study patients showed toxic plasma concentrations of CBZ-E or associated clinical toxicity. Drug-protein binding interactions are another source of side effects. A simultaneous administration of valproic acid increases the total plasma CBZ-E levels relative to the CBZ dose associated with the raised free fractions of CBZ and CBZ-E. The high free plasma concentrations of CBZ-E above 1.5 ,g/ml may be responsible for the side effects. [source] Developmental and environmental regulation of antifreeze proteins in the mealworm beetle Tenebrio molitorFEBS JOURNAL, Issue 21 2000Laurie A. Graham The yellow mealworm beetle, Tenebrio molitor, contains a family of small Cys-rich and Thr-rich thermal hysteresis proteins that depress the hemolymph freezing point below the melting point by as much as 5.5 °C (,T = thermal hysteresis). Thermal hysteresis protein expression was evaluated throughout development and after exposure to altered environmental conditions. Under favorable growth conditions, small larvae (11,13 mg) had only low levels of thermal hysteresis proteins or thermal hysteresis protein message, but these levels increased 10-fold and 18-fold, respectively, by the final larval instar (> 190 mg), resulting in thermal hysteresis >,3 °C. Exposure of small larvae (11,13 mg) to 4 weeks of cold (4 °C) caused an ,,20-fold increase in thermal hysteresis protein concentration, well in excess of the less than threefold developmental increase seen after 4 weeks at 22 °C. Exposure of large larvae (100,120 mg) to cold caused 12-fold and sixfold increases in thermal hysteresis protein message and protein levels, respectively, approximately double the maximum levels they would have attained in the final larval instar at 22 °C. Thus, thermal hysteresis increased to similar levels (> 4 °C) in the cold, irrespective of the size of the larvae (the overwintering stage). At pupation, thermal hysteresis protein message levels decreased >,20-fold and remained low thereafter, but thermal hysteresis activity decreased much more slowly. Exposure to cold did not reverse this decline. Desiccation or starvation of larvae had comparable effects to cold exposure, but surprisingly, short daylength photoperiod or total darkness had no effect on either thermal hysteresis or message levels. As all environmental conditions that caused increased thermal hysteresis also inhibited growth, we postulate that developmental arrest is a primary factor in the regulation of T. molitor thermal hysteresis proteins. [source] Developmental, metabolic and immunological costs of flea infestation in the common voleFUNCTIONAL ECOLOGY, Issue 6 2008Godefroy Devevey Summary 1Parasites use resources from their hosts, which can indirectly affect a number of host functions because of trade-offs in resource allocation. In order to get a comprehensive view of the costs imposed by blood sucking parasites to their hosts, it is important to monitor multiple components of the development and physiology of parasitized hosts over long time periods. 2The effect of infestation by fleas on body mass, body length growth, haematocrit, resistance to oxidative stress, resting metabolic rate and humoral immune response were experimentally evaluated. During a 3-month period, male common voles, Microtus arvalis, were either parasitized by rat fleas (Nosopsyllus fasciatus), which are naturally occurring generalist ectoparasites of voles, or reared without fleas. Then voles were challenged twice by injecting Keyhole Limpet Haemocyanin (KLH) to assess whether the presence of fleas affects the ability of voles to produce antibodies against a novel antigen. During the immune challenge we measured the evolution of body mass, haematocrit, resistance to oxidative stress and antibody production. 3Flea infestation negatively influenced the growth of voles. Moreover, parasitized voles had reduced haematocrit, higher resting metabolic rate and lower production of antibodies against the KLH. Resistance to oxidative stress was not influenced by the presence of fleas. 4During the immune challenge with KLH, body mass decreased in both groups, while the resistance to oxidative stress remained stable. In contrast, the haematocrit decreased only in parasitized voles. 5Our experiment shows that infestation by a haematophageous parasite negatively affects multiple traits like growth, energy consumption and immune response. Fleas may severely reduce the survival probability and reproductive success of their host in natural conditions. [source] Developmental and activity-dependent genomic occupancy profiles of CREB in monkey area V1GENES, BRAIN AND BEHAVIOR, Issue 2 2009J. Lalonde The mammalian neocortex displays significant plastic rearrangement in response to altered sensory input, especially during early postnatal development. It is believed that cyclic AMP-response element-binding (CREB) plays an important role in orchestrating the molecular events that guide neuroplastic change, although the details of its genomic targets during normal postnatal development or in response to sensory deprivation remain unknown. Here, we performed CREB chromatin immunoprecipitation (ChIP) from monkey area V1 tissue and hybridized enriched DNA fragments to promoter microarrays (ChIP chip analysis). Our goal was to determine and categorize the CREB regulon in monkey area V1 at two distinct developmental stages (peak of critical period vs. adulthood) and after 5 days of monocular enucleation (ME) at both ages. Classification of enriched candidates showed that the majority of isolated promoter loci (n = 795) were common to all four conditions. A particularly interesting group of candidates (n = 192) was specific to samples derived from enucleated infant area V1. Gene ontology analysis of CREB targets during early postnatal development showed a subgroup of genes implicated in cytoskeleton-based structural modification. Analysis of messenger RNA expression (quantitative real-time,polymerase chain reaction) of candidate genes showed striking differences in expression profiles between infant and adult area V1 after ME. Our study represents the first extensive genomic analysis of CREB DNA occupancy in monkey neocortex and provides new insight into the multifaceted transcriptional role of CREB in guiding neuroplastic change. [source] Observing Purchase-Related Parent,Child Communication in Retail Environments: A Developmental and Socialization PerspectiveHUMAN COMMUNICATION RESEARCH, Issue 1 2008Moniek Buijzen In a quantitative observation study, we unobtrusively examined purchase-related communication between 0- to 12-year-old children and their parents (N= 269 dyads) during supermarket and toy store visits. The aims of the study were to determine (a) the development of purchase-related parent,child communication (i.e., children's purchase influence attempts, their coercive behavior, parent-initiated communication) and (b) the relative influence of different socialization variables (e.g., television viewing, family communication patterns) on these communication variables. Our inverted-U hypothesis for the effect of developmental level on purchase influence attempts received support: Children's purchase influence attempts increased until early elementary school and started to decline in late elementary school. Our inverted-U hypothesis for the effect of developmental level on coercive behavior was also supported: Children's coercive behavior was highest among preschoolers. With increasing age, children were more likely to be involved in the purchase decision-making process, and parent,child communication more often resulted in a product purchase. Finally, children's television viewing was the most important (positive) predictor of their purchase influence attempts. Ziele der Studie waren: a) die Entwicklung von kaufbezogener Kommunikation zwischen Elternteil und Kind und b) den relativen Einfluss verschiedener Sozialisationsvariablen (z.B. Fernsehnutzung, Familienkommunikationsmuster) auf diese Variablen zu untersuchen. Unsere umgekehrte U-Hypothese bezüglich des Einflusses des Entwicklungsstadiums auf den Grad der Einflussnahmeversuche auf den Kauf wurde gestützt: Die Einflussnahmeversuche nahmen bis zur frühen Grundschulzeit zu und gingen in der späten Grundschulzeit zurück. It is also used in cosmetic products such as hair dyes and products for coating finger nails. Beginning in 2001, HQ is no longer authorized for use in cosmetic skin lightening formulations in European Union countries, although products containing arbutin, an analogue of HQ, and botanicals, including plants that naturally contain HQ and arbutin, continue to remain available in European countries. The potential toxicity of HQ is dependent on the route of exposure, and toxicity in rodents is highly sex-, species-, and strain-specific. Subchronic and chronic toxicity in experimental animals is primarily limited to nephrotoxicity in male F-344 rats. Dermal toxicity studies, even those conducted in the sensitive male F-344 rat, are essentially devoid of systemic toxicity. Developmental and reproductive toxicity studies with HQ in rats and rabbits have not demonstrated significant effects. Cancer bioassay data for HQ demonstrate limited effects and are not sufficient to classify HQ for human carcinogenicity. Epidemiology and occupational studies of workers with extensive exposure to HQ have not reported any evidence of adverse systemic health effects or carcinogenicity. A risk-benefit approach is recommended for assessing the available data for HQ, arbutin, and other materials in use as, or proposed for use as, skin lighteners to provide optimal therapeutic benefits to patients with pigmentary changes of the skin. Design:, Quasi-experimental, repeated measures design. Setting:, Developmental and a control nursery in a 78-bed, level II/III neonatal intensive-care unit. Participants:, A convenience sample of 114 premature infants and their parents. Interventions:, Control group infants received the routine neonatal intensive-care unit standard of care. Experimental infants received routine care plus the addition of individualized, developmentally supportive family-centered interventions. Main Outcome measures:, Between groups, there were no statistically significant differences in demographic factors, days to medical or developmental milestones, length of stay, or direct cost/case. Repeated measures analysis of variance determined that at every point of data collection, the average number of baseline, activity, and postactivity stress cues were lower in the developmentally supportive group. Infants in the developmental group had 8% less sedatives/narcotics and 15% less vasopressors costs than the control group. There were no differences in complication rates, parental perceptions of the neonatal intensive-care unit experience, or parental satisfaction between groups. Conclusions:, Preterm infants who received developmentally supportive family-centered care demonstrated fewer behavioral stress cues and comparable short-term outcomes and resource utilization than infants who received routine care. Knowledge of the clinical, neurophysiological, and pathogenetic aspects of these disorders is poor. Based on a comprehensive review of the literature and our practical experience, this article summarizes current knowledge in this area. Changes in mean values and covariances of nutrients during leaf development may be relevant for plant fitness through herbivory if variation in leaf nutrients influences plant defoliation. , , We analysed the between-population and developmental covariation of leaf nutrients by using common principal components (CPC) analysis. We also studied the relationships between leaf nutrient covariates and natural levels of plant defoliation. , , Plants at our two study sites shared a CPC structure of covariation between concentrations of the leaf nutrients. Collectively, these two eating disorders are characterized by severe disturbances in eating behaviors and acute distress over body shape and weight. In AN, fear of weight gain coupled with a distorted body image leads to refusal to maintain a minimally accepted body weight (American Psychiatric Association [APA], 1994). Although women with BN also overvalue body size and shape, this disorder is characterized by episodes of binge eating followed by efforts to undo the binge episode (oral purging is the most widely known effort) (APA, 1994). Ted Brown PhD, OT(C) Abstract Paediatric occupational therapists were surveyed regarding their practices in Canada and Australia. Two hundred and eighty-nine Canadian occupational therapists and 330 Australian occupational therapists participated representing response rates of 28.9% and 55% respectively. The majority of respondents were female (98%), between 30 and 49 years of age (69%), had a bachelor's degree, worked on average 10.5 years in paediatrics and spent well over 50% of their work time in direct client care. The largest client diagnostic groups in both countries were those with developmental delays, learning disabilities and neurological disorders. Diagnostic groups were used as an organizing framework to portray theory, assessment and intervention use. Overall, the theoretical models cited most frequently in both countries were: Sensory Integration, Sensory Processing/Sensory Diet, Client-Centred Practice, and Occupational Performance Model. Australian therapists employed the Occupational Performance Model (Australia) for all groups, while it was rarely utilized in Canada. Common assessment tools in both Australia and Canada were the Peabody Developmental Motor Scales, Developmental Test of Visual Motor Integration, and the Bruininks-Oseretsky Test of Motor Proficiency. Intervention methods focused on: parental/care-giver education; activities of daily living/self-care skills training; client education; environmental modification; assistive devices; sensory integration techniques; sensory stimulation and sensory diet treatment methods; and neurodevelopmental techniques. [source] Perceptual, motor and cognitive performance components of Bedouin children in IsraelOCCUPATIONAL THERAPY INTERNATIONAL, Issue 4 2000Shula Parush PhD Abstract The purpose of this study was to evaluate the perceptual, motor and cognitive performance abilities of Bedouin children as compared with mainstream Israeli children of the same age. The study population comprised two age groups (6,8 year olds and 10,12 year olds) living in two different types of settlements (rural and town). Participants included 20 children in each group. The Test of Visual-Perceptual Skills (TVPS), the Developmental Test of Visual-Motor Integration (VMI), selected subtests from the Bruininks-Oseretsky Test of Motor Proficiency and the Loewenstein Occupational Therapy Cognitive Assessment (LOTCA) were used. Older participants in all three groups performed better than the younger subjects. There were no significant differences between the two Bedouin groups in either age category. The mainstream Israeli subjects scored significantly higher than the two Bedouin groups on most variables tested. The lack of differences between the two Bedouin groups may indicate that the transition from nomadic communities to urban communities is mainly external and has not caused a significant change in perception, perceptual-motor and cognitive performance abilities. The results of this study may be used as a base for developing preventive intervention programmes to meet the specific needs of Bedouin children. A limitation of this study was the classification of the Bedouin population according to their dwelling place and not according to their sociodemographic background, such as parents' economic status, parents' education and so on. Recommendations for future research include expanding the number of participants, including children from additional rural schools of various distant tribes and incorporating more detailed cognitive assessments. [source] Nutritional, developmental, and genetic influences on relative sitting height at high altitude,AMERICAN JOURNAL OF HUMAN BIOLOGY, Issue 5 2009Sara Stinson The study explores how nutritional status, developmental exposure to high-altitude hypoxia, and genetic ancestry influence relative sitting height in two groups of high-altitude Bolivian children aged 8 through 13 years of age: 253 rural Aymara children of very low socioeconomic status and 273 children of upper socioeconomic status from the capital city of La Paz. The rural Aymara children on average have longer trunks relative to stature, but there is also overlap in body proportions between the two groups of children. The 20% of each sample in the region of overlap was examined to investigate influences on relative sitting height. Nutritional effects on relative sitting height are suggested by the finding that Aymara children with relatively long legs are taller, heavier, and fatter than other Aymara children. Developmental and genetic influences on relative sitting height are suggested by the finding that high relative sitting heights in elite urban children are associated with a greater percentage of time lived at high altitude and with parents born in Bolivia. Separating developmental and ancestry effects is difficult because the two are closely interconnected in the urban children. The results of this study suggest that influences on growth in relative trunk and leg length are similar to those that affect other aspects of growth in Andean populations. They also highlight the fact that because relative sitting height gradually decreases prior to adolescence and then increases, the interpretation of variation in body proportions in children is not always straightforward. [source] Mixed-longitudinal growth of Karimojong girls and boys in Moroto District, UgandaAMERICAN JOURNAL OF HUMAN BIOLOGY, Issue 1 2009Sandra Gray In this article we examine results of a mixed-longitudinal study of child and adolescent growth among agropastoral Karimojong children in Moroto District, northeast Uganda. During a 5-month period from August to December, 2004, longitudinal data were collected for a mixed sample of 104 Karimojong children, aged from birth to 18 years. During a previous study in 1998,1999,we had measured 26 of these children who then ranged in Age between 3 months and 7 years. Most of the children were small and thin relative to accepted growth standards, and prevalence of stunting and wasting in childhood was high. In the period from the end of childhood through adolescence, however, Karimojong girls showed marked variability in annual growth, with some attaining a large adult size relative to what we predicted based on their poor childhood growth. Developmental, evolutionary, and environmental determinants are considered. We conclude that growth of these children reflects exposure to environmental insults that vary unpredictably within relatively short intervals. Variability in the magnitude and timing of these insults among children from different birth-cohorts is probably sufficient to account for so-called "shifting" of growth percentiles in childhood and adolescence in this mixed sample. Determining these patterns from the valuable samples typically used in anthropology and palaeoanthropology necessarily implies nondestructive procedures. [source] Developmental, gender, and practical considerations in scoring curriculum-based measurement writing probesPSYCHOLOGY IN THE SCHOOLS, Issue 4 2003Christine Kerres Malecki The present study focused on CBM written language procedures by conducting an investigation of the developmental, gender, and practical considerations surrounding three categories of CBM written language scoring indices: production-dependent, production-independent, and accurate-production. Students in first- through eighth-grade generated a three-minute writing sample in the fall and spring of the school year using standard CBM procedures. The writing samples were scored using all three types of scoring indices to assess the trends in scoring indices for students of varying ages and gender and of the time required to score writing samples using various scoring indices. With only one exception, older students outperformed younger students on all of the scoring indices. Although at the middle school level students' levels of writing fluency and writing accuracy were not closely associated, at the younger grade levels the CBM indices were significantly related. With regard to gender differences, girls outperformed boys on measures of writing fluency at all grade levels. Smith Abstract Developmental and structural affinities between modern human and Neanderthal dental remains continue to be a subject of debate as well as their utility for informing assessments of life history and taxonomy. Excavation of the Middle Paleolithic cave site Lakonis in southern Greece has yielded a lower third molar (LKH 1). Here, we detail the crown development and enamel thickness of the distal cusps of the LKH 1 specimen, which has been classified as a Neanderthal based on the presence of an anterior fovea and mid-trigonid crest. Crown formation was determined using standard histological techniques, and enamel thickness was measured from a virtual plane of section. Developmental differences include thinner cuspal enamel and a lower periodicity than modern humans. Crown formation in the LKH 1 hypoconid is estimated to be 2.6,2.7 years, which is shorter than modern human times. The LKH 1 hypoconid also shows a more rapid overall crown extension rate than modern humans. Relative enamel thickness was approximately half that of a modern human sample mean; enamel on the distal cusps of modern human third molars is extremely thick in absolute and relative terms. These findings are consistent with recent studies that demonstrate differences in crown development, tissue proportions, and enamel thickness between Neanderthals and modern humans. Although overlap in some developmental variables may be found, the results of this and other studies suggest that Neanderthal molars formed in shorter periods of time than modern humans, due in part to thinner enamel and faster crown extension rates. Maynard Abstract Developmental guidance cues act to direct growth cones to their correct targets in the nervous system. Recent experiments also demonstrate that developmental cues are expressed in the adult mammalian nervous system, although their function in the brain is not yet clear. The semaphorin gene family has been implicated in the growth of dendrites and axons in a number of different species. While the expression of semaphorin and its influence on tibial pioneer neurons in the developing limb bud have been well characterized in the grasshopper, the expression of semaphorin 2a (sema2a) has not been explored in the adult insect. In this study we used polymerase chain reaction (PCR) with degenerate and gene-specific primers to clone part of the secreted form of sema2a from Gryllus bimaculatus. Using in situ hybridization and immunohistochemistry, we confirmed that sema2a mRNA and protein expression patterns in the embryonic cricket were similar to that seen in the grasshopper. We also showed that tibial neuron development in crickets was comparable to that described in grasshopper. An examination of both developing and adult cricket brains showed that sema2a mRNA and protein were expressed in the Kenyon cells in mushroom bodies, an area involved in learning and memory. Sema2a expression was most obvious near the apex of the mushroom body in a region surrounding the neurogenic tip, which produces neurons throughout the life of the cricket. [source] Developmental and light effects on the accumulation of FtsH protease in Arabidopsis chloroplasts , implications for thylakoid formation and photosystem II maintenanceTHE PLANT JOURNAL, Issue 5 2005Adi Zaltsman Summary The chloroplast ATP-dependent metalloprotease FtsH is involved in the degradation of unassembled proteins, the repair of photosystem II (PSII) from photoinhibition, and, apparently, the formation of thylakoids. In Arabidopsis, it is encoded by a family of 12 genes. However, the products of only four of them, FtsH1, 2, 5 and 8, have been found in chloroplasts to date. Mutations in two of these, FtsH2 and 5, demonstrate a visible phenotype of variegated leaves, with the phenotype of the FtsH2 mutant being more pronounced. Moreover, the degree of variegation appears to be dependent on developmental stage and environmental factors, suggesting an intricate relationship between the different gene products. To explore this, developmental and light effects on the accumulation of FtsH protease were studied in wild-type (WT) and FtsH2-mutant plants. Whereas cotyledons of the mutant were indistinguishable from those of the WT, the first true leaves were almost completely white. Subsequent leaves contained increasing proportions of green sectors. Analysis of the mRNA of the four FtsH genes, in cotyledons, first and second leaves of WT and mutant plants, revealed that: (i) transcript level increases during development, and (ii) transcript level in the mutant is higher than in the WT. FtsH protein level in the mutant was ca. 50% of that found in the WT, whereas the levels of other thylakoid proteins were the same. In individual leaves, the level of FtsH protein increased during development as well. Exposure of seedlings to different light intensities did not affect the degree of variegation, suggesting that it is due to a defect in chloroplast development rather than photobleaching. Examination of FtsH protein during exposure to high light revealed a decrease in its level, concomitant with a decrease in PSII potential, suggesting that the kinetics of photoinhibition reflects not only photodamage to PSII and induction of protective mechanisms, but also a decrease in repair capacity due to a reduction in the level of FtsH protease. Chiara Manzini PhD Objective We sought to explore the genetic and molecular causes of Troyer syndrome, one of several complicated hereditary spastic paraplegias (HSPs). Troyer syndrome had been thought to be restricted to the Amish; however, we identified 2 Omani families with HSP, short stature, dysarthria and developmental delay,core features of Troyer syndrome,and a novel mutation in the SPG20 gene, which is also mutated in the Amish. In addition, we analyzed SPG20 expression throughout development to infer how disruption of this gene might generate the constellation of developmental and degenerative Troyer syndrome phenotypes. Methods Clinical characterization of 2 non-Amish families with Troyer syndrome was followed by linkage and sequencing analysis. Quantitative polymerase chain reaction and in situ hybridization analysis of SPG20 expression were carried out in embryonic and adult human and mouse tissue. Results Two Omani families carrying a novel SPG20 mutation displayed clinical features remarkably similar to the Amish patients with Troyer syndrome. SPG20 mRNA is expressed broadly but at low relative levels in the adult brain; however, it is robustly and specifically expressed in the limbs, face, and brain during early morphogenesis. Interpretation Null mutations in SPG20 cause Troyer syndrome, a specific clinical entity with developmental and degenerative features. Maximal expression of SPG20 in the limb buds and forebrain during embryogenesis may explain the developmental origin of the skeletal and cognitive defects observed in this disorder. Alexopoulos Objective Chondrocytes, the sole cell type in articular cartilage, maintain the extracellular matrix (ECM) through a homeostatic balance of anabolic and catabolic activities that are influenced by genetic factors, soluble mediators, and biophysical factors such as mechanical stress. Chondrocytes are encapsulated by a narrow tissue region termed the "pericellular matrix" (PCM), which in normal cartilage is defined by the exclusive presence of type VI collagen. Because the PCM completely surrounds each cell, it has been hypothesized that it serves as a filter or transducer for biochemical and/or biomechanical signals from the cartilage ECM. The present study was undertaken to investigate whether lack of type VI collagen may affect the development and biomechanical function of the PCM and alter the mechanical environment of chondrocytes during joint loading. Methods Col6a1,/, mice, which lack type VI collagen in their organs, were generated for use in these studies. At ages 1, 3, 6, and 11 months, bone mineral density (BMD) was measured, and osteoarthritic (OA) and developmental changes in the femoral head were evaluated histomorphometrically. Mechanical properties of articular cartilage from the hip joints of 1-month-old Col6a1,/,, Col6a1+/,, and Col6a1+/+ mice were assessed using an electromechanical test system, and mechanical properties of the PCM were measured using the micropipette aspiration technique. Results In Col6a1,/, and Col6a1+/, mice the PCM was structurally intact, but exhibited significantly reduced mechanical properties as compared with wild-type controls. With age, Col6a1,/, mice showed accelerated development of OA joint degeneration, as well as other musculoskeletal abnormalities such as delayed secondary ossification and reduced BMD. Conclusion These findings suggest that type VI collagen has an important role in regulating the physiology of the synovial joint and provide indirect evidence that alterations in the mechanical environment of chondrocytes, due to either loss of PCM properties or Col6a1,/, -derived joint laxity, can lead to progression of OA. [source] Temperature-dependent development and distribution in the soil profile of pupae of greyback canegrub Dermolepida albohirtum (Waterhouse) (Coleoptera: Scarabaeidae) in Queensland sugarcaneAUSTRALIAN JOURNAL OF ENTOMOLOGY, Issue 1 2007David P Logan Abstract, The temperature-dependent development rate of pupae of greyback canegrub, Dermolepida albohirtum (Waterhouse) (Coleoptera: Scarabaeidae), a major pest of sugarcane in central and northern Queensland, was determined under six constant temperature regimes: 18, 20, 23, 25, 27 and 30°C, and for four geographically separated populations. Development rate increased significantly with increasing temperature. Parameters of the linear regression equation did not differ among populations and common coefficients were calculated. Developmental zero, at and below which no development occurs, for pupae was 12.0°C and the thermal constant was 476 day-degrees (D°). Minimum and maximum periods for pupal development were 26 days at 30°C and 75 days at 18°C, respectively. The phenology of pupae was determined in soil-filled cubicles in a shade house and in the field at Ayr (19°35,S, 147°25,E), north Queensland, using D. albohirtum field-collected as late-stage third-instar larvae and kept in containers. Pupation of D. albohirtum began in late August or early September and eclosion was complete by mid- to late October. The phenology data were used to validate the development model. Eclosion was predicted by summing hourly fractions of day-degrees until 476 D° was reached and was close to actual eclosion. As temperature, and hence pupal development rate, varies with soil depth, the distribution of the third instars in pupal cells in the soil profile was determined in recently harvested fields of sugarcane in the Burdekin sugarcane district centred on Ayr. Numbers of late third instars in pupal cells peaked at 300,400 mm, with pupae found from 30 to 700 mm. Pupal development was simulated using hourly soil temperatures measured at depths of 200 and 400 mm at Ayr and at Sarina (21°22,S, 149°13,E). The pupal stage was predicted to take up to 2,10 days longer at 200 mm deep than at 400 mm depending on pupation site and date. When pupation was simulated in late August, as is likely in the field, pupal development at 400 mm deep took 48,56 days at Ayr and 58,62 days at Sarina. [source] Embryonic development of verongid demosponges supports the independent acquisition of spongin skeletons as an alternative to the siliceous skeleton of spongesBIOLOGICAL JOURNAL OF THE LINNEAN SOCIETY, Issue 2 2009MANUEL MALDONADO Approximately 85% of extant sponges (phylum Porifera) belong to the class Demospongiae, which contains 14 taxonomic orders. In the orders Verongida, Dictyoceratida, and Dendroceratida, jointly referred to as ,keratose demosponges', the skeleton does not contain siliceous spicules but only spongin fibres. This shared trait has encouraged placement of these orders together within Demospongiae, although their relationships remain uncertain. The present study documents for the first time embryo development in the order Verongida (Aplysina aerophoba), providing some clues for phylogenetic inference. Spawned eggs were enveloped by a follicle of maternal cells. Embryos and larvae were chimeric organisms, the blastocoel of which was filled with symbionts and maternal cells migrated from the follicle. The ultrastructure of epithelial larval cells revealed: (1) a basal apparatus characterized by a peculiar, angling accessory centriole; (2) a pear-shaped nucleus with a protruding beak connected to the rootlets of the basal body; and (3) a distinctive Golgi apparatus encircling the nuclear apex. Developmental and ultrastructural findings support the concept, in congruence with recent molecular studies, that Verongida are more closely related to Halisarcida (askeletal sponges) and Chondrosida (askeletal sponges + sponges with spongin + spiculate sponges) than to the remaining ,keratose' orders, making a monophyletic ,supra-ordinal unit' equivalent to a subclass (Myxospongia, new subclass). Hence, spongin skeletons have evolved at least twice in Demospongiae. Whiteley Abstract Objectives To ascertain the frequency of parental reporting of selected variables related to development, behaviour and physiology in subgroups diagnosed with pervasive developmental disorders (PDDs) and identify any significant intragroup differences. Design Retrospective cross-sectional analysis of records of patients (n = 512) held on a computerized database with a chronological age between 3 and 11 years resident in the UK/Republic of Ireland and with a formal diagnosis of autism, Asperger syndrome (AS) or autism spectrum disorder (ASD). Methods Non-parametric analysis (P > 0.01) of the frequency of specific variables for PDD subgroups reported by parents/primary caregiver. Variables included timing of symptom onset, presence of skills acquired prior to symptom onset, indications of regression and regression events, current language, history of viral infections, history of ear problems, achievement of continence, current skin complaints, current bowel habits and adverse events at parturition. Results Preliminary results showed general agreement with the principle diagnostic differences between the PDD subgroups with patients diagnosed with AS showing an increased frequency of skills acquired before symptom onset (two- to three-word phrase speech, toileting skills) and a decreased frequency of regression in acquired skills when compared with other PDD subgroups. Developmental milestones such as the achievement of bowel and bladder continence were also more frequently reported for the AS group. Infantile feeding problems defined as vomiting, reflux, colic and failure to feed were more frequently reported for the AS group as was a reported history of the bacterial skin infection impetigo. Results are discussed with reference to relationships between behavioural and somatic factors in PDD. [source] |
