Avian Risk Assessment: Effects of Perching Height and Detectability

We studied two components of predator risk assessment in birds. While many species are limited to seeking safety under cover or under ground, some birds can fly away from their predators and escape to trees. If birds in fact ,feel' safer (e.g. perceive less risk) in trees, we would expect them to tolerate closer approach by a potential terrestrial predator. Another component of safety is at which point the animal detects an approaching threat, which we expected to increase with eye size, assuming eye size is a surrogate for visual acuity. We used the distance birds moved away from an approaching human [flight initiation distance (FID)] as a metric to determine whether birds associated a lower risk of predation by being in trees, and we used the distance at which birds first displayed alert behaviors from an approaching human (alert distance) to determine if birds with larger eyes had higher detection distances. Although some species were affected by tree height, we found no clear pattern that birds assessed themselves to be at a lower risk of predation when they were ,3 m above the ground compared with being <3 m above ground. In the 10 species for which height had any significant effect on FID, birds ,3 m off the ground had greater FIDs in six species, but the remaining three species had the opposite response. While we found a significant positive relationship between eye size and alert distance in 23 species, the relationship was not present in a phylogenetic analysis using independent contrasts, which suggests that the apparent relationship was influenced strongly by the association between the studied species. Together, these results suggest that birds do not obviously associate being in a tree with safety, and that variations in visual acuity, per se, cannot be used as a general indicator of differences in alert distances, as previously suggested in the literature. Decreased Detectability of Grapevine Leafroll-associated virus 3 in Sakasly Grapevines Cultivated Under the Sahara Conditions

Abstract The detectability of grapevine-leafroll-associated virus 3 (GLRaV-3) was investigated by enzyme-linked immunosorbent assay (ELISA) and reverse-transcriptase polymerase chain reaction (RT,PCR) in the domestic grapevine cultivar Sakasly, grown over two successive years in the Sahara, at Rjim-Mâatoug in Tunisia. Self-rooted cuttings, infected with GLRaV-3 were cultivated and the presence of the virus was checked over 2 years and compared with controls. During the first year, 80% of the originally infected vines were negative for GLRaV-3 using ELISA. After the second year, 93% and 95% of these plants were negative for GLRaV-3 using ELISA and RT,PCR, respectively. Furthermore, rooted cuttings derived from GLRaV-3-negative plants and grown under controlled conditions in a greenhouse (at 16,20°C) were ELISA-negative in most cases (84%). In addition, biological indexing on Vitis vinifera cv. Gamay Rouge de la Loire showed no leafroll symptoms on this indicator in 92% of the grafted vines. These results suggest a naturally occurring heat therapy in the Tunisian Sahara, which could be of practical importance for the production of GLRaV-3-free grapevine cuttings, especially as scale insect and mealybug vectors have not been observed in this area. Biased Reduced Sampling: Detectability of an Attribute and Estimation of Prevalence

Abstract In surveilling a population, detection of systems with an attribute of interest and estimation of the prevalence of the attribute in the population are two main goals. Due to cost constraints, only a subset of all components of sampled systems may be fully tested. Biasing the sampling to increase the probability of choosing a component with an attribute of interest ameliorates the impact of reduced sampling. In this paper, we consider the impact of biased reduced sampling on detection and propose an approach for estimating the prevalence of the attribute in the population which properly accounts for the biasing. Estimates of population size of white-chinned petrels and grey petrels at Kerguelen Islands and sensitivity to fisheries

Abstract White-chinned petrels Procellaria aequinoctialis and grey petrels Procellaria cinerea are among the most frequently killed seabird species by accidental bycatch, and both species have received strong conservation concern. Data on population size are required to evaluate the impact of bycatch and to establish management plans. We estimated the population size of both species at Kerguelen, Southern Indian Ocean, from 2004 to 2006 by explicitly taking into account detection probability of burrows using distance sampling and burrow occupancy. A total of 31 line-transects were distributed across the eastern part of Kerguelen, representing a total length of 566 km. Detectability was low (from 0.19 to 0.54 for white-chinned petrels, 0.58 for grey petrels). Burrow densities varied from 1.37±0.67 to 25.77±5.23 burrows ha,1 for white-chinned petrels and was 2.78±0.79 burrows ha,1 for grey petrels. For white-chinned petrels, these densities were extrapolated to the entire surface area of vegetation and there were 234 000 (186 000,297 000) active burrows on Kerguelen. For grey petrels, the number of active burrows for the eastern part of Kerguelen was 3400 (1900,5600). Based on these estimates, the potential biological removal method suggests that the additional mortality on birds caused by the fisheries operating around Kerguelen can be considered a serious threat for the species at least at the regional scale of the Southern Indian Ocean, especially for grey petrels. Direct on-line analysis of neutral analytes by dual sweeping via complexation and organic solvent field enhancement in nonionic MEKC

Abstract Conventionally, neutral compounds cannot be separated by nonionic micelle capillary electrophoresis. In this report, the development of a novel on-line preconcentration technique combining dual sweeping based on complexation and organic solvent field enhancement is applied to the sensitive and selective analysis of three neutral glucosides: ginsenoside Rf, ginsenoside Rg1, and ginsenoside Re. Nonionic micelle detectability by CE is demonstrated through effective focusing of large sample volumes (up to 38% capillary length) using a dual sweeping mode. This results in a 50- to 130-fold improvement in the LODs relative to conventional injection method. Neutral compounds sweeping is examined in terms of analyte mobility dependence on borate complexation, solvent viscosity difference, and Brij-35 interaction. Enhanced focusing performance by this hyphenated method was demonstrated by a greater than fourfold reduction in glucoside bandwidth, as compared with common sweeping (devoid of organic solvent-mediated sweeping method in the sample matrices). Moreover, separation efficiencies greater than a million theoretical plates can be achieved by sweeping large sample volumes into narrow zones. The designated method was also tested for its ability to determine the presence of glucosides in the crude extracts obtained from plant sample. [source]


Application of polymeric surfactants in micellar electrokinetic chromatography-electrospray ionization mass spectrometry of benzodiazepines and benzoxazocine chiral drugs

Abstract Chiral micellar EKC (CMEKC) coupled to ESI-MS using polymeric surfactants as pseudostationary phases is investigated for simultaneous enantioseparation of two benzodiazepines, (±)-oxazepam ((±)-OXA) and (±)-lorazepam ((±)-LOR), and one benzoxazocine, (±)-nefopam ((±)-NEF). First, enantioselectivity and electrospray sensitivity of six chiral polymeric surfactants for all three chiral compounds are compared. Second, using poly(sodium N -undecenoyl- L -leucinate) as pseudostationary phase, the organic modifiers (methanol (MeOH), isopropanol, and ACN) are added into the running buffer to further improve chiral resolution (RS). Next, a CMEKC-ESI-MS method for the simultaneous enantioseparation of two benzodiazepines is further developed by using a dipeptide polymeric surfactant, poly(sodium N -undecenoxy carbonyl- L,L -leucyl-valinate) (poly- L,L -SUCLV). The CMEKC conditions including nebulizer pressure, capillary length, ammonium acetate concentration, pH, poly- L,L -SUCLV concentration, and capillary temperature were optimized to achieve maximum chiral RS and highest sensitivity of MS detection. The spray chamber parameters (drying gas temperature and drying gas flow rate) as well as sheath liquid conditions (MeOH content, pH, flow rate, and ionic strength) were found to significantly influence MS S/N of both (±)-OXA and (±)-LOR. Finally, a comparative study between simultaneous UV and MS detection showed high plate numbers, better chiral RS, and enhanced detectability with CMEKC-MS. However, speed of analysis was faster using CMEKC-UV. [source]


Development and evaluation of an enzyme-linked immunosorbent assay to detect Pieris rapae remains in guts of arthropod predators

Abstract An enzyme-linked immunosorbent assay (ELISA) was developed to detect remains of Pieris rapae L. (Lepidoptera: Pieridae) immature stages in the guts of field collected arthropod predators. The assay can be used to help ascertain the relative importance of arthropod predator species in suppressing P. rapae in cabbage, Brassica oleracea var. capitata L. The ELISA is sensitive to all immature stages of P. rapae, although first and fifth instars can be detected more readily than eggs or pupae and third instars showed intermediate detectability. Assays on whole body homogenates of predators readily detected predation on P. rapae first instars by all seven of the predator species tested, although response generally declined with increasing predator size. Together the results show that the P. rapae ELISA possesses a sufficiently high level of sensitivity and specificity to be a useful tool in helping to elucidate the roles of arthropod predator species in reducing populations of P. rapae in cabbage. Plesiomorphic Escape Decisions in Cryptic Horned Lizards (Phrynosoma) Having Highly Derived Antipredatory Defenses

Escape theory predicts that the probability of fleeing and flight initiation distance (predator,prey distance when escape begins) increase as predation risk increases and decrease as escape cost increases. These factors may apply even to highly cryptic species that sometimes must flee. Horned lizards (Phrynosoma) rely on crypsis because of coloration, flattened body form, and lateral fringe scales that reduce detectability. At close range they sometimes squirt blood-containing noxious substances and defend themselves with cranial spines. These antipredatory traits are highly derived, but little is known about the escape behavior of horned lizards. Of particular interest is whether their escape decisions bear the same relationships to predation risk and opportunity costs of escaping as in typical prey lacking such derived defenses. We investigated the effects of repeated attack and direction of predator turning on P. cornutum and of opportunity cost of fleeing during a social encounter in P. modestum. Flight initiation distance was greater for the second of two successive approaches and probability of fleeing decreased as distance between the turning predator and prey increased, but was greater when the predator turned toward than away from a lizard. Flight initiation distance was shorter during social encounters than when lizards were solitary. For all variables studied, risk assessment by horned lizards conforms to the predictions of escape theory and is similar to that in other prey despite their specialized defenses. Our findings show that these specialized, derived defenses coexist with a taxonomically widespread, plesiomorphic method of making escape decisions. They suggest that escape theory based on costs and benefits, as intended, applies very generally, even to highly cryptic prey that have specialized defense mechanisms. [source]


THE EVOLUTION OF WING COLOR: MALE MATE CHOICE OPPOSES ADAPTIVE WING COLOR DIVERGENCE IN COLIAS BUTTERFLIES

Abstract Correlated evolution of mate signals and mate preference may be constrained if selection pressures acting on mate preference differ from those acting on mate signals. In particular, opposing selection pressures may act on mate preference and signals when traits have sexual as well as nonsexual functions. In the butterfly Colias philodice eriphyle, divergent selection on wing color across an elevational gradient in response to the thermal environment has led to increasing wing melanization at higher elevations. Wing color is also a long-range signal used by males in mate searching. We conducted experiments to test whether sexual selection on wing melanization via male mate choice acts in the same direction as natural selection on mate signals due to the thermal environment. We performed controlled mate choice experiments in the field over an elevational range of 1500 meters using decoy butterflies with different melanization levels. Also, we obtained a more direct estimate of the relation between wing color and sexual selection by measuring mating success in wild-caught females. Both our experiments showed that wing melanization is an important determinant of female mating success in C. p. eriphyle. However, a lack of elevational variation in male mate preference prevents coevolution of mate signals and mate preference, as males at all elevations prefer less-melanized females. We suggest that this apparently maladaptive mate choice may be maintained by differences in detectability between the morphs or by preservation of species recognition. [source]


Respiratory oscillations in yeast: mitochondrial reactive oxygen species, apoptosis and time; a hypothesis

Abstract Oscillatory metabolic activities occur more widely than is generally realised; detectability requires observation over extended times of single yeast cells or synchrony of individuals to provide a coherent population. Where oscillations in intracellular metabolite concentrations are observed, the phenomenon has been ascribed to sloppy control, energetic optimisation, signalling, temporal compartmentation of incompatible reactions, or timekeeping functions. Here we emphasise the consequences of respiratory oscillations as a source of mitochondrially generated reactive O2 metabolites. Temporal co-ordination of intracellular activities necessitates a time base. This is provided by an ultradian clock, and one result of its long-term operation is cyclic energisation of mitochondria, and thereby the generation of deleterious free radical species. Our hypothesis is that unrepaired cellular constituents and components (especially mitochondria) eventually lead to cellular senescence and apoptosis when a finite number of respiratory cycles has occurred. Seasonal and inter-stream variations in the population dynamics, growth and secondary production of an algivorous fish (Pseudogastromyzon myersi: Balitoridae) in monsoonal Hong Kong

Summary 1.,Balitorid loaches are widespread and highly diverse in Asian streams, yet their life history and ecology have received little attention. We investigated seasonal (wet versus dry season) and spatial variation in populations of algivorous Pseudogastromyzon myersi in Hong Kong, and estimated the magnitude of secondary production by this fish in pools in four streams (two shaded and two unshaded) over a 15-month period. 2.,Mean population densities of P. myersi ranged from 6.0 to 23.2 individuals m,2, constituting more than half (and typically >70%) of benthic fishes censused. Abundance was c. 25% greater in the wet season, when recruitment occurred. Significant density differences among streams were not related to shading conditions and were evident despite small-scale variations in P. myersi abundance among pools. Mean biomass varied among streams from 0.85 to 3.87 g ash-free dry weight (AFDW) m,2. Spatial and seasonal patterns in biomass and density were similar, apart from some minor disparities attributable to differences in mean body size among populations. 3.,All four P. myersi populations bred once a year in June and July, and life spans varied from 24 to 26 months. Populations consisted of three cohorts immediately after recruitment but, for most of the study period, only two cohorts were evident. Cohort-specific growth rates did not differ significantly among streams but, in all streams, younger cohorts had higher cohort-specific growth rates. 4.,Secondary production of P. myersi estimated by the size-frequency (SF) method was 2.7,11.5 g AFDW m,2 year,1 and almost twice that calculated by the increment-summation (IS) method (1.2,6.6 g AFDW m,2 year,1). Annual P/B ratios were 1.17,2.16 year,1 (IS) and 2.73,3.22 year,1 (SF). Highest production was recorded in an unshaded stream and the lowest in a shaded stream, but site rankings by production did not otherwise match shading conditions. Wet-season production was six times greater than dry-season production, and daily production fell to almost zero during January and February. Cool temperatures (<17 °C) may have limited fish activity and influenced detectability during some dry-season censuses. Estimates of abundance and annual production by P. myersi are therefore conservative. 5.,Comparisons with the literature indicate that the abundance and production of P. myersi in Hong Kong was high relative to other benthic fishes in tropical Asia, or their temperate counterparts in small streams. Manipulative experiments are needed to determine the influence of P. myersi, and algivorous balitorids in general, on periphyton dynamics and energy flow in Asian streams. [source]


Carotenoid and melanin-based ornaments signal similar aspects of male quality in two populations of the common yellowthroat

Summary 1.,Female preferences for particular male ornaments may shift between populations as a consequence of ecological differences that change the reliability and detectability of the ornament, but few studies have examined how ornaments function in different populations. 2.,We examined the signalling function of male plumage ornaments in a warbler, the common yellowthroat (Geothlypis trichas), breeding in New York (NY) and Wisconsin (WI), USA. Males have two prominent ornaments: a black facial mask pigmented with melanin and a yellow bib pigmented by carotenoids. Previous studies in WI indicate that the size of the mask, and not the bib, is primarily related to female choice and male reproductive success. In NY, however, the pattern is reversed and attributes of the bib (size and colour), and not the mask, are the target of sexual selection. 3.,We found that brightness of the yellow bib was the best signal of humoral immunity (immunoglobulin G) in NY and mask size was the best signal in WI, after controlling for breeding experience and capture date. Thus, similar aspects of male quality appeared to be signalled by different ornaments in different populations. 4.,There was no difference between populations in the level of plasma carotenoids or the prevalence of malarial parasites, which may affect the costs and benefits of choosing males with particular ornaments in each location. 5.,Even though females in different populations prefer different ornaments produced by different types of pigments, these ornaments appear to be signalling similar aspects of male quality. Our results caution against inferring the function of particular ornaments based simply on their type of pigment. [source]


A transportable mb(Lg) scale for central Europe and implications for low-magnitude Ms,mb discrimination

SUMMARY We have extended the mb(Lg) method of Nuttli using root-mean-square (rms) amplitudes corrected for noise and a ,,1 dependence for geometrical spreading. Lg waves recorded on the German Regional Seismic Network (GRSN) for earthquakes in south-central Europe were used to develop an mb(Lg) formula requiring a new calibration constant Crms to keep rms mb(Lg) on the same baseline as Nuttli's traditional formula based on 3rd-peak amplitudes. GRSN stations had to be calibrated for site terms and for Lg attenuation. Lateral variations in LgQ appear to be significant across the study area, and a regional Q model consisting of constant- Q partitions north, south and in the central Alps was developed using measurements based on interstation and two-event, single-station methods. When plotted against surface wave estimates of Mw, rms mb(Lg) measurements in central Europe are found to be consistent with Mw,mb(Lg) relationships for north America and southern Asia, thus supporting the transportability of our mb(Lg) formula. Frequency,wavenumber processing of Gräfenberg Array data enabled us to extract Rayleigh waves for small events, and regional Ms were measured using the Marshall and Basham formula. Our Ms,mb(Lg) relationship extends to Ms 2.5 and agrees well with observations in other regions including the western United States. The discrimination potential of Ms,mb(Lg) observations was examined under realistic monitoring conditions, where path corrections were inferred from earthquake data and applied uniformly to natural sources and explosions. Under these conditions, mb(Pn, P) are greater than mb(Lg) for large NTS explosions; however, Ms,mb scaling slopes are steeper for P waves than they are for Lg, and Ms,mb observations for NTS explosions converge near mb 4. Thus, allowing for measurement errors and additional uncertainty in mb(Pn) due to regional bias, there is little difference in the discrimination potential for Pn and Lg waves at small magnitudes. As such, a regional Ms,mb discriminant based on Lg might be preferred owing to the better detectability of Lg waves for small earthquakes. These results need to be confirmed for explosions at other test sites. Compared to teleseismic experience, regional Ms,mb observations extend the discrimination capability to lower magnitudes by at least one Ms unit. [source]


The seismic response to overpressure: a modelling study based on laboratory, well and seismic data

We investigate the seismic detectability of an overpressured reservoir in the North Sea by computing synthetic seismograms for different pore-pressure conditions. The modelling procedure requires the construction of a geological model from seismic, well and laboratory data. Seismic inversion and AVO techniques are used to obtain the P-wave velocity with higher reliability than conventional velocity analysis. From laboratory experiments, we obtain the wave velocities of the reservoir units versus confining and pore pressures. Laboratory experiments yield an estimate of the relationship between wave velocities and effective pressure under in situ conditions. These measurements provide the basis for calibrating the pressure model. Overpressures are caused by different mechanisms. We do not consider processes such as gas generation and diagenesis, which imply changes in phase composition, but focus on the effects of pure pore-pressure variations. The results indicate that changes in pore pressure can be detected with seismic methods under circumstances such as those of moderately deep North Sea reservoirs. [source]

Formation of nonculturable Escherichia coli in drinking water

Abstract Aims:, To examine whether incubation of Escherichia coli in nondisinfected drinking water result in development of cells that are not detectable using standard procedures but maintain a potential for metabolic activity and cell division. Methods and Results:, Survival and detectability of four different E. coli strains were studied using drinking water microcosms and samples from contaminated drinking water wells. Recovery of E. coli was compared using different cultivation-dependent methods, fluorescence in situ hybridization (FISH) using specific oligonucleotide probes, direct viable counts (DVC), and by enumeration of gfp -tagged E. coli (green fluorescent protein, GFP). Two levels of stress responses were observed after incubation of E. coli in nondisinfected drinking water: (i) the presence of cells that were not detected using standard cultivation methods but could be cultivated after gentle resuscitation on nonselective nutrient-rich media, and (ii) the presence of cells that responded to nutrient addition but could only be detected by cultivation-independent methods (DVC, FISH and GFP). Collectively, the experiments demonstrated that incubation for 20,60 days in nondisinfected drinking water resulted in detection of only 0·7,5% of the initial E. coli population using standard cultivation methods, whereas 1,20% could be resuscitated to a culturable state, and 17,49% could be clearly detected using cultivation-independent methods. Conclusions:, Resuscitation of stressed E. coli on nonselective nutrient-rich media increased cell counts in drinking water using both traditional (CFU), and cultivation-independent methods (DVC, FISH and GFP). The cultivation-independent methods resulted in detection of 10,20 times more E. coli than the traditional methods. The results indicate that a subpopulation of substrate-responsive but apparent nonculturable E. coli may develop in drinking water during long-term starvation survival. Significance and Impact of the Study:, The existence of substrate-responsive but nonculturable cells should be considered when evaluating the survival potential of E. coli in nondisinfected drinking water. On the importance of estimating detection probabilities from at-sea surveys of flying seabirds

The primary and accepted method used to estimate seabird densities at sea from ships is the strip transect method, designed to correct for the effect of random directional bird movement relative to that of the ship. However, this method relies on the critical assumption that all of the birds within the survey strip are detected. We used the distance sampling method from line-transects to estimate detection probability of a number of species of flying seabirds, and to test whether distance from the ship and bird body size affected detectability. Detection probability decreased from 0.987 (SE=0.029) to 0.269 (SE=0.035) with increasing strip half-width from 100 to 1400,m. Detection probability also varied between size-groups of species with strip half-width. For all size-groups, this probability was close to 1 for strip half-width of 100,m, but was 0.869 (SE=0.115), 0.725 (SE=0.096) and 0.693 (SE=0.091) for strip half-width of 300,m, a typical strip width used in seabird surveys, for respectively large, medium and small size flying seabirds. For larger strip half-width, detection probability was higher for large sized species, intermediate for medium sized species and lower for smaller sized species. For strip half-width larger than 100,m we suggest that more attention should be paid to testing the assumption of perfect detectability, because abundance estimates may be underestimated when this assumption is violated. Finally, the effect of the speed of travel of flying seabird on the detection probability was estimated in a simulation study, which suggests that detection probability was underestimated with increasing flying speed. Detection, survival rates and dynamics of a cryptic plant, Asclepias meadii: applications of mark-recapture models to long-term monitoring studies

Summary 1Analysis of population trajectories is central to assessing risk in populations of conservation concern. In animal studies, researchers realize that probabilities of detection of individuals are often less than one. Plants can also escape detection due to dormancy, herbivory, or observer error, but such information is rarely incorporated into population studies. 2We monitored a population of Asclepias meadii, a rare long-lived prairie perennial. Despite standardized methods, numbers of observed plants fluctuated greatly from 1992 to 2006. Individual plants often had periods of 1,5 years between initial and final sighting when no stems were found. To determine the actual population trajectories, we estimated rates of survival and population growth using mark-recapture models. We also estimated initial and resighting probabilities of detection. In 2007, we repeated surveys to identify reasons for low detection probabilities. 3We estimated 95% annual survival and a population growth rate of 1.023. Probabilities of initial detection were low (typically from 0.120 to 0.311 depending on prairie burn treatment), whereas average probability of detection for marked plants was 0.728. 4Comparisons of survival estimates from 15- and 8-year data sets revealed that survival estimates decline in the final years of a multi-year period, probably due to heterogeneity in encounter histories. 5By conducting three different surveys in 2007, we found that both herbivory over a multiple-week period and observer error contributed substantially to gaps in detection. 6Synthesis. Probabilities of detection that are less than one complicate interpretation of population dynamics, whether of mobile animals or inconspicuous plants. Our work illustrates three general points that could apply to many plant population studies: (i) mark-recapture models may provide insights on vital rates and population trajectories despite the extreme variability in count data that can arise because of low detectability, (ii) probabilities of initial detection can be quantified and can be considerably less than probabilities of resighting, and (iii) repeated surveys can help researchers determine the degree to which dormancy, herbivory, or observer error contribute to low probabilities of detection. Consideration of these points can improve the design and analysis of monitoring programs. [source]

Abstract We have recently published support to the hypothesis that visual systems of parents could affect nestling detectability and, consequently, influences the evolution of nestling colour designs in altricial birds. We provided comparative evidence of an adjustment of nestling colour designs to the visual system of parents that we have found in a comparative study on 22 altricial bird species. In this issue, however, Renoult et al. (J. Evol. Biol., 2009) question some of the assumptions and statistical approaches in our study. Their argumentation relied on two major points: (1) an incorrect assignment of vision system to four out of 22 sampled species in our study; and (2) the use of an incorrect approach for phylogenetic correction of the predicted associations. Here, we discuss in detail re-assignation of vision systems in that study and propose alternative interpretation for current knowledge on spectrophotometric data of avian pigments. We reanalysed the data by using phylogenetic generalized least squares analyses that account for the alluded limitations of phylogenetically independent contrasts and, in accordance with the hypothesis, confirmed a significant influence of parental visual system on gape coloration. Our results proved to be robust to the assumptions on visual system evolution for Laniidae and nocturnal owls that Renoult et al. (J. Evol. Biol., 2009) study suggested may have flawed our early findings. Thus, the hypothesis that selection has resulted in increased detectability of nestling by adjusting gape coloration to parental visual systems is currently supported by our comparative data. Environmental stress and the costs of whole-organism phenotypic plasticity in tadpoles

Abstract Costs of phenotypic plasticity are important for the evolution of plasticity because they prevent organisms from shaping themselves at will to match heterogeneous environments. These costs occur when plastic genotypes have relatively low fitness regardless of the trait value expressed. We report two experiments in which we measured selection on predator-induced plasticity in the behaviour and external morphology of frog tadpoles (Rana temporaria). We assessed costs under stressful and benign conditions, measured fitness as larval growth rate or competitive ability and focused analysis on aggregate measures of whole-organism plasticity. There was little convincing evidence for a cost of phenotypic plasticity in our experiments, and costs of canalization were nearly as frequent as costs of plasticity. Neither the magnitude of the cost nor the variation around the estimate (detectability) was sensitive to environmental stress. [source]


Colour polymorphism in birds: causes and functions

Abstract We studied polymorphism in all species of birds that are presently known to show intraspecific variation in plumage colour. At least three main mechanisms have been put forward to explain the maintenance of polymorphism: apostatic, disruptive and sexual selection. All of them make partly different predictions. Our aims were to investigate evolutionary causes and adaptive functions of colour polymorphism by taking into account a number of ecological and morphological features of polymorphic species. Overall, we found 334 species showing colour polymorphism, which is 3.5% of all bird species. The occurrence of colour polymorphism was very high in Strigiformes, Ciconiiformes, Cuculiformes and Galliformes. Phylogenetically corrected analysis using independent contrasts revealed that colour polymorphism was maximally expressed in species showing a daily activity rhythm extended to day/night, living in both open and closed habitats. All these findings support the hypothesis that colour polymorphism probably evolved under selective pressures linked to bird detectability as affected by variable light conditions during activity period. Thus, we conclude that selective agents may be prey, predators and competitors, and that colour polymorphism in birds may be maintained by disruptive selection. [source]


Habitat use, abundance, and persistence of Neotropical migrant birds in a habitat matrix in northeast Belize

ABSTRACT To ensure adequate protection of nonbreeding habitats used by Neotropical migratory landbirds, we must first address questions about habitat use and quality. On the Yucatan peninsula, migrants use many habitats, several of which remain unstudied, and methodological differences preclude interhabitat comparisons based on studies to date. We used distance sampling along line transects in six habitats in northeast Belize to examine use of previously unstudied habitats (e.g., salt marsh) by Neotropical migrants and to permit comparison across habitats. We calculated unadjusted and adjusted (for detectability) density estimates for individual migrant species and for all species combined to generate hypotheses about habitat quality based on the assumption that density and quality are positively correlated. Adjusted density estimates for all migrants were highest in black mangrove habitat (1799 ± 110 ind/km2), intermediate in three forest types and milpa (range 598,802 ind/km2), and lowest in salt marsh (207 ± 32.3 ind/km2). By combining density estimates with habitat availability in our study region, we estimated that evergreen forest and black mangrove supported 70% and 9% of the region's migrant population, respectively. At the species level, five of the 10 most common species had habitat preferences (>50% detections in one habitat). Given the diversity of habitat preferences among species and apparent seasonal movements, our results indicate that Neotropical migrants in northeast Belize are dependent on a matrix of interconnected habitats. RESUMEN Para asegurar la protección adecuada del hábitat no- reproductivo utilizado por aves migratorias Neotropicales terrestres, debemos responder preguntas sobre el uso y la calidad del hábitat. En la península de Yucatán, lasaves migratorias utilizan muchos hábitats, varios de los cuales aún continúan sin ser estudiados. Además las diferencias metodológicas evitan hacer comparaciones inter-hábitat basadas en los estudios que se han hecho hasta ahora. Utilizamos unmuestreo a lo largo de transectos de distancia variable, en seis hábitats, algunos previamente no estudiados (ej. ciénagas salobres), en el noreste de Belize para examinar y comparar el uso por parte de las migratorias Neotropicales, Calculamos estimados de densidad, ajustados y no ajustados (para detectabilidad), para especies particulares de migratorias y para todas las especies combinadas para generar una hipótesis sobre la calidad del hábitat basándonos en la presunción que la densidad estaría positivamente correlacionada con la calidad del hábitat. Los estimados de densidad ajustados para todos los migratorios fueron más altos en mangle negro (1799 ± 100 ind/km2), intermedios en tres tipos de bosque y milpa rango 598,802 ind/km2) y menores en ciénagas salobres (207 ± 32.3 ind/km2). Combinando los estimados de densidad con la disponibilidad de hábitats en nuestra región de estudio, estimamos que el bosque siempreverde y el mangle negro sostienen el 70% y 9% de los migratorios en la región, respectivamente. A nivel de especies, cinco de las 10 especies más comunes tienen preferencias de hábitat (>50% de las detecciones en un hábitat). Dada la diversidad de preferencias de hábitat entre especies y el aparente movimiento estacional, nuestros resultados indican que las aves migratorias Neotropicales en el noreste de Belize dependen de una matriz de hábitats interconectados. Effects of nest-site characteristics and parental activity on cowbird parasitism and nest predation in Brown-and-yellow Marshbirds

ABSTRACT Nest-site selection and nest defense are strategies for reducing the costs of brood parasitism and nest predation, two selective forces that can influence avian nesting success and fitness. During 2001,2002, we analyzed the effect of nest-site characteristics, nesting pattern, and parental activity on nest predation and brood parasitism by cowbirds (Molothrus spp.) in a population of Brown-and-yellow Marshbirds (Pseudoleistes virescens) in the Buenos Aires province, Argentina. We examined the possible effects of nest detectability, nest accessibility, and nest defense on rates of parasitism and nest predation. We also compared rates of parasitism and nest predation and nest survival time of marshbird nests during the egg stage (active nests) with those of the same nests artificially baited with passerine eggs after young fledged or nests failed (experimental nests). Most nests (45 of 48, or 94%) found during the building or laying stages were parasitized, and 79% suffered at least one egg-predation event. Cowbirds were responsible for most egg predation, with 82 of 107 (77%) egg-predation events corresponding to eggs punctured by cowbirds. Nests built in thistles had higher rates of parasitism and egg predation than nests in other plant, probably because cowbirds were most active in the area where thistles were almost the only available nesting substrate. Parasitism rates also tended to increase as the distance to conspecific nests increased, possibly due to cooperative mobbing and parental defense by marshbirds. The proportion of nests discovered by cowbirds was higher for active (95%) than for experimental (29%) nests, suggesting that cowbirds used host parental activity to locate nests. Despite active nest defense, parental activity did not affect either predation rates or nest-survival time. Thus, although nest defense by Brown-and-yellow Marshbirds appears to be based on cooperative group defense, such behavior did not reduce the impact of brood parasites and predators. RESUMEN La selección del sitio de nidificación y defensa del nido son estrategias que reducen los costos del parasitismo de cría y predación de nidos, dos fuerzas selectivas que pueden afectar el éxito reproductivo y la eficacia biológica de las aves. Durante 2001,2002, analizamos el efecto de las características del sitio de nidificación, patrón de nidificación, y actividad parental en la predación de nidos y parasitismo de cría por tordos (Molothrus spp.) en una población de Pseudoleistes virescens en la provincia de Buenos Aires, Argentina. Examinamos los posibles efectos de la detectabilidad, accesibilidad y defensa del nido en las tasas de parasitismo y depredación de nidos. Asimismo, se compararon las tasas de parasitismo, depredación, y tiempo de supervivencia de los nidos entre los estadios de huevo (nidos activos) con los de los mismos nidos cebados artificialmente con huevos de passerinos (nidos experimentales). La mayoría de los nidos (45 de 48, o 94%) hallados durante los estadios de construcción y puesta fueron parasitados, donde el 79% de los nidos sufrieron al menos un evento de predación de huevos. Los tordos fueron responsables de la mayoría de los eventos de predación de huevos, donde 82 de 107 (77%) eventos de predación correspondieron a huevos picados por tordos. Los nidos construidos en cardos tuvieron tasas de predación y parasitismo más altas que las correspondientes a nidos construidos en otros sustratos, probablemente debido a que los tordos exhiben una mayor actividad en el área donde los cardos son el único sustrato de nidificación disponible. La tasa de parasitismo mostró una tendencia a crecer con la distancia al nido más próximo, probablemente debido a la defensa parental y los despliegues de defensa comunales de Pseudoleistes virescens. La proporción de nidos descubiertos por tordos fue mayor para nidos activos (95%) que para nidos experimentales (29%), lo cual sugiere que los tordos utilizaron la actividad parental para detectar los nidos. A pesar de la activa defensa del nido, descubrimos que la actividad parental no afectó ni la tasa de predación ni el tiempo de supervivencia de los nidos. De esta forma, a pesar de que la defensa del nido en Pseudoleistes virescens aparenta estar basada en la defensa cooperativa grupal, tal comportamiento no redujo el impacto de los parásitos de cría o predadores de nidos. Behavioural responses of the seven-spot ladybird Coccinella septempunctata to plant headspace chemicals collected from four crop Brassicas and Arabidopsis thaliana, infested with Myzus persicae

Abstract 1,Insects using olfactory stimuli to forage for prey/hosts are proposed to encounter a ,reliability,detectability problem', where the usability of a stimulus depends on its reliability as an indicator of herbivore presence and its detectability. 2,We investigated this theory using the responses of female seven-spot ladybirds Coccinella septempunctata (Coleoptera: Coccinellidae) to plant headspace chemicals collected from the peach-potato aphid Myzus persicae and four commercially available Brassica cultivars; Brassica rapa L. cultivar ,turnip purple top', Brassica juncea L. cultivar ,red giant mustard', Brassica napus L. cultivar ,Apex', Brassica napus L. cultivar ,Courage' and Arabidopsis thaliana. For each cultivar/species, responses to plants that were undamaged, previously infested by M. persicae and infested with M. persicae, were investigated using dual-choice Petri dish bioassays and circular arenas. 3,There was no evidence that ladybirds responded to headspace chemicals from aphids alone. Ladybirds significantly preferred headspace chemicals from B. napus cv. Apex that were undamaged compared with those from plants infested with aphids. For the other four species/cultivars, there was a consistent trend of the predators being recorded more often in the half of the Petri dish containing plant headspace chemicals from previously damaged and infested plants compared with those from undamaged ones. Furthermore, the mean distance ladybirds walked to reach aphid-infested A. thaliana was significantly shorter than to reach undamaged plants. These results suggest that aphid-induced plant chemicals could act as an arrestment or possibly an attractant stimulus to C. septempunctata. However, it is also possible that C. septempunctata could have been responding to aphid products, such as honeydew, transferred to the previously damaged and infested plants. 4,The results provide evidence to support the ,reliability,detectability' theory and suggest that the effectiveness of C. septempunctata as a natural enemy of aphids may be strongly affected by which species and cultivar of Brassica are being grown. Effect of instrument tuning on the detectabilityof biopolymers in electrospray ionization mass spectrometry

Abstract Electrospray ionization mass spectrometry of multiply charged biopolymer ions of different molecular size revealed a strong influence of tuning parameters on their detectability in quadrupole ion trap and triple quadrupole mass spectrometers. Hence, after optimizing the ion optical parameters with the signal of the 4, charge state of (dT)24 (low charge state tuning), a tenfold increase in the signal-to-noise ratio for a mixture of oligodeoxythymidylic acids (n = 12,18) was obtained compared with the results achieved with tune parameters optimized with a synthetic 80-mer oligodeoxynucleotide. By contrast, a detection limit in the upper femtomole region could only be reached for a 104-mer oligodeoxynucleotide utilizing the 24, charge state of the 80-mer (high charge state tuning). The same effect was observed for proteins investigated in the positive ion mode using low and high charge states of cytochrome c and carbonic anhydrase, respectively, for instrument tuning. By comparing the settings for low and high charge state tuning, it became obvious that the most significant difference was observed in the potential applied to the heated metal capillary used to transfer ions from the atmospheric pressure to the vacuum region of the ion source. Taking advantage of the optimized tuning procedure, the molecular mass of a 61 base pair product of polymerase chain reaction was accurately determined by electrospray ionization mass spectrometry on-line interfaced to ion-pair reversed-phase high-performance liquid chromatography. Monitoring of herpes simplex virus DNA types 1 and 2 viral load in cerebrospinal fluid by real-time PCR in patients with herpes simplex encephalitis

Abstract A quantitative polymerase chain reaction (PCR) assay was evaluated retrospectively on 92 cerebrospinal fluid (CSF) samples from 29 patients with herpes simplex virus (HSV) encephalitis with the aim to study if the concentration of HSV genomes can be used as a prognostic marker and for monitoring of antiviral therapy. The results were compared to those obtained previously by nested PCR, and the numbers of HSV genomes/ml were evaluated in correlation to patient outcome and treatment. The aims were to compare the sensitivity of a conventional nested PCR to a quantitative PCR, to investigate the range of HSV genome concentration in initial samples and to evaluate possible relationships between the HSV DNA concentrations in CSF, neopterin levels, and outcome of disease. The 29 initial samples contained between 2,×,102 and 42,×,106 HSV genomes/ml. There was no apparent correlation between the amount of HSV DNA in the initial samples and income status, initial neopterin levels, or prognosis. The number of HSV genomes/ml declined after treatment in all patients, but HSV DNA was still detectable after day 20 in 3 out of 16 patients. A long duration of genome detectability was found to correlate with poor outcome. There was no difference in sensitivity between the nested PCR and the quantitative PCR. While the quantitative PCR is more rational than a nested PCR, the quantitation of HSV genomes does not seem very useful as a prognostic marker in HSV encephalitis. J. Med. The urinary excretion of ,-hydroxybutyric acid in man

,-Hydroxybutyric acid (GHB) has been widely associated with drug-facilitated sexual assault (DFSA). However, its excretion profile in man has not been well characterized. To assess the detectability of GHB for forensic cases and to correlate urinary levels with dose, we have examined the excretion profiles of 1- and 2-g doses of GHB (sodium salt) in a healthy male volunteer. The urinary levels were measured by a novel, simple and highly reproducible method. The drug was found to be excreted in small amounts in the free form (0.86 and 1.16% for 1- and 2-g doses, respectively) rapidly in urine (, 10 h). The urinary levels were found to be in the low mg L,1 range (up to 29.1 mg L,1). The work presented demonstrates that it is of the utmost importance to collect the samples as soon as possible following the alleged assault. [source]


Fast miniaturised sample preparation for the screening and comprehensive two-dimensional gas chromatographic determination of polychlorinated biphenyls in sludge

Abstract Polychlorinated biphenyls (PCBs) in sludge are usually extracted by a technique such as Soxhlet with subsequent fractionation prior to long GC runs using GC,ECD or GC,HRMS. In this study, the extraction of selected chlorinated biphenyls (CBs) from a spiked sludge sample by three rapid techniques, i.e. ultrasonic (USE), pressurised-liquid (PLE), and microwave-assisted (MAE) extraction using a domestic microwave, was studied, with subsequent direct GC,ECD, GC,MS, or GC×GC,,ECD analysis of the extracts. The main goal was to select an appropriate, and miniaturised, extraction method after only a brief optimisation and demonstrate the power of GC×GC analysis of dirty extracts. For PLE similar CB recoveries were found when extracting with either n -hexane or n -hexane/acetone (1/1). For USE and MAE, n- hexane/acetone (1/1) was the preferred extraction solvent. USE gave the best recoveries (80,95%; except 130% for CB 105). The only clean-up needed prior to GC,MS or GC×GC,,ECD analysis was the removal of sulphur-containing compounds. GC,ECD was not suitable for these dirty extracts. The lowest LODs for the CBs (20 fg or 0.1 ng/g sludge) were found when combining USE and GC×GC,,ECD, because of the powerful extraction, high separation power and excellent detectability provided by this technique. Challenges of gas chromatography,high-resolution time-of-flight mass spectrometry for simultaneous analysis of polybrominated diphenyl ethers and other halogenated persistent organic pollutants in environmental samples

Abstract The potential of a gas chromatographic method employing high-resolution time-of-flight (TOF) mass spectrometry was evaluated for detection of polybrominated diphenyl ethers (PBDEs) in the environmental matrices represented by fish and river sediment. Two ionisation techniques, viz. electron ionisation (EI) and negative ion chemical ionisation (NICI), the latter with methane as a reagent gas, were used in this study. While the instrumental lowest calibration levels (LCLs) obtained in EI were in the range from 1 to 5 pg, their values ranged between 10 to 250 fg in NICI mode. This enhancement in detectability of target analytes enabled identification/quantification of even minor PBDE congeners, and consequently, improved characterisation of particular sample contamination patterns. In addition, this method allowed estimation of the PCB levels in examined samples. CB 153 was used as a contamination marker in this study. [source]


Reduced-bore monolithic silica column modified with C8 -TEOS for reversed-phase electrochromatography

Abstract Monolithic silica columns of 2.7 mm ID were prepared and derivatized with C8 -TEOS and TEOS by on-column sol-gel reaction. These C8 large diameter monolithic silica columns gave 21 000 theoretical plates for aromatic hydrocarbons in 60% acetonitrile and 40% Tris-HCl buffer. The surface areas as well as the separation reproducibility were improved on coating by the sol-gel approach. Joule heating was greatly reduced by using monolithic columns to which fine quartz sand had been added during column preparation. Since this is a preliminary investigation on a monolithic column with such a large inner diameter, the separation efficiency was not so high as that presently achieved in normal capillary electrochromatography (CEC). However, use of the columns improved sample loadability and concentration detectability of electrochromatography, and semi-preparative separations could be performed. [source]
