Selected AbstractsA DIAGNOSTIC READING OF SCIENTIFICALLY BASED RESEARCH FOR EDUCATIONEDUCATIONAL THEORY, Issue 3 2005Thomas A. Schwandt This essay offers a diagnosis of what may be at stake in the current preoccupation with defining science-based educational research. The diagnosis unfolds in several readings: The first is a charitable and considerate appraisal that draws attention to the fact that advocating experimental methods as important to a science of educational research is not an inherently evil thing to do. Subsequent readings are grimmer, suggesting more deleterious consequences of the science-based research movement for the entire enterprise of educational practice and research. The central thesis of the essay is that making arguments about method and science the focal point in the current quarrel may be largely beside the point. Instead, educational researchers should join the political and public (not just the academic) conversation about the place of educational science in society and about how science is both implicated in and confronts the politics of what counts as knowledge. [source] Transcriptional readthrough of Hox genes Ubx and Antp and their divergent post-transcriptional control during crustacean evolutionEVOLUTION AND DEVELOPMENT, Issue 5 2006Yasuhiro Shiga SUMMARY Hox genes are in principle tandemly arranged in an order colinear with their order of expression along the anterior,posterior axis. Combinations of Hox proteins encode information that specifies the unique characteristics of axial regions in the metazoan body plan. The independent regulation of Hox genes achieved by differential promoter activity is essential for the expression of Hox proteins in distinct territories and thereby creating a full repertoire of Hox codes. Here we report the abundant expression of transcriptional readthrough products of two adjacent Hox genes, Ubx, and Antp, in five crustacean species of Branchiopoda and Malacostraca. Bicistronic mRNA places Antp under the control of the Ubx promoter, which is active in the posterior segments of two branchiopodans Daphnia and Artemia, and would normally reduce the complexity of Hox codes if translated. This does not occur, however, as the translational capability of the bicistronic mRNA is limited. In Daphnia, bicistronic Ubx/Antp mRNA produced no significant level of either UBX or ANTP. In Artemia, on the other hand, the bicistronic mRNA produced only UBX, and replaced the role of monocistronic Ubx mRNA. In this way, multiple post-transcriptional control mechanisms in two extant branchiopodans can be seen as preventing the potentially deleterious consequences of Hox gene fusion. [source] Physiological response to stress in fledgling Lesser Kestrels Falco naumanni: the role of physical condition, sex and individual genetic diversityIBIS, Issue 3 2009JOAQUÍN ORTEGO Exposure to chronic stress early on during development has important deleterious consequences later in life, reducing important components of individual fitness such as survival and future reproduction. In this study, we evaluate the factors associated with physiological response to stress in fledgling Lesser Kestrels Falco naumanni, paying particular attention to the potential role of individual genetic diversity. For this purpose, we used heterophil/lymphocyte ratios (H/L ratio) as a haematological stress indicator and typed the analysed individuals at 11 highly polymorphic microsatellite loci, which allowed us to estimate their genetic diversity. We found that the H/L ratio decreases with fledgling physical condition, suggesting that this parameter is a good indicator of nutritionally based physiological stress. Physiological response to stress was higher in males than in females and this effect was independent of physical condition, suggesting that the observed pattern is due to inherent sexual differences in the factors influencing H/L ratios. Finally, the H/L ratio was positively associated with the genetic diversity of offspring. Previous experimental studies have found that individuals with higher genetic diversity show increased levels of circulating glucocorticoids, which in turn are directly responsible for increasing H/L ratios. On this basis, we suggest that a positive effect of genetic diversity on corticosterone levels may explain the observed association between H/L ratios and individual heterozygosity. Overall, this study highlights the utility of leucocyte profiles to study stress in wild bird populations and poses an interesting question about the effects of individual genetic diversity on haematological response to stress. [source] Heptachlor and o-p,DDT effects on protein kinase activities associated with human placenta particulate fractionsJOURNAL OF BIOCHEMICAL AND MOLECULAR TOXICOLOGY, Issue 3 2009Gladis Magnarelli Abstract Organochlorine pesticides have been detected in placenta. The ability of heptachlor (HC) and 1,1,1-tricholoro-2-(2-chlorophenyl)-2-4-chlorophenyl)ethane (o-p,DDT) to interfere with protein phosphorylation was evaluated. In vitro incubations of cell-free placental villi homogenates with a concentration range 1,100 µM were performed. In particulate fractions, total serine/threonine kinase activity was increased by 10 µM HC and o-p, DDT (59% and 82%, respectively). Maximum eightfold increase was observed with 10 µM o-p, DDT on protein kinase A activity. By contrast, protein kinase C activity was reduced by 10 µM HC and o-p, DDT (40% and 52%, respectively). Endogenous substrate phosphorylation studies demonstrated that slight but significant increase in 24-kDa band labeling was produced in nuclear samples with 1, 10, and 100 µM HC and 100 µM o-p, DDT. Exposition to 100 µM HC increased 85-kDa band labeling. In mitochondrial fractions, 10 µM HC and o-p, DDT increased 24- and 65-kDa bands' labeling. These data indicate that both pesticides affect protein kinase activities in particulate fraction. Nuclear compartmentalization of these compounds, insertion in membranes, and chemical stress production may be associated to the observed effects, thus suggesting deleterious consequences in signaling pathways. © 2009 Wiley Periodicals, Inc. STEINBERG M.D. Introduction: This study was designed to assess whether right ventricular pacing in the implantable cardioverter defibrillator (ICD) arm of the Multicenter Automatic Defibrillator Implantation Trial (MADIT) II was associated with an unfavorable outcome. Methods and Results: Data on the number of ventricular paced beats were available in 567 (76%) of 742 MADIT II patients with ICDs. The number of ventricular paced beats over the total number of beats showed a bimodal distribution with patients being predominantly paced or nonpaced. Therefore, patients were dichotomized at 0,50% and 51,100% of cumulative pacing with median pacing rate 0.2% and 95.6%, respectively. Endpoints included new or worsening heart failure, appropriate ICD therapy for VT/VF, and the combined endpoint of heart failure or death. Clinical features associated with frequent ventricular pacing included age ,65 years, advanced NYHA heart failure class, LVEF < 0.25, first degree AV and bundle branch block, and amiodarone use. During follow-up, 119 patients (21%) had new or worsened heart failure, 130 (23%) had new or worsened heart failure or death, and 142 (25%) had appropriate therapy for VT/VF. In comparison to patients with infrequent pacing, those with frequent pacing had significantly higher risk of new or worsened heart failure (hazard ratio = 1.93; P = 0.002) and VT/VF requiring ICD therapy (HR = 1.50; P = 0.02). Conclusions: Patients in MADIT II who were predominantly paced had a higher rate of new or worsened heart failure and were more likely to receive therapy for VT/VF. These results suggest the deleterious consequences of RV pacing, particularly in the setting of severe LV dysfunction. Tumor necrosis factor (TNF) is a key mediator of cutaneous inflammation and the keratinocyte an important protagonist of skin immunity. Calcitriol, the hormonally active vitamin D metabolite, and its analogs attenuate epidermal inflammation and inhibit the hyperproliferation of keratinocytes associated with the inflammatory disorder, psoriasis. Since activation of extracellular signal-regulated kinase (ERK) promotes keratinocyte proliferation and mediates epidermal inflammation, we studied the effect of calcitriol on ERK activation in HaCaT keratinocytes exposed to the ubiquitous inflammatory cytokine TNF. By using the EGF receptor (EGFR) tyrosine kinase inhibitor, AG1487 and the Src family inhibitor, PP-1, we established that TNF activated ERK in an EGFR and Src dependent and an EGFR and Src independent modes. EGFR dependent activation resulted in the upregulation of the transcription factor, c-Fos, while the EGFR independent activation mode was of a shorter duration, did not affect c-Fos expression but induced IL-8 mRNA expression. Pretreatment with calcitriol, enhanced TNF-induced EGFR-Src dependent ERK activation and tyrosine phosphorylation of the EGFR, but abolished the EGFR-Src independent ERK activation. These effects were mirrored by enhancement of c-Fos and inhibition of IL-8 induction by TNF. Treatment with calcitriol increased the rate of the de-phosphorylation of activated ERK, accounting for the inhibition of EGFR-Src independent ERK activation by TNF. It is possible that effects on the ERK cascade contribute to the effects of calcitriol and its synthetic analogs on cutaneous inflammation and keratinocyte proliferation. J. Cell. ORTEGA Summary, Cerebral palsy (CP) is one of the most frequent conditions encountered in the daily practice of dentists who treat special-needs patients and it seems that parafunctional oral habits are often present in such individuals. The aim of this study was to investigate the frequency of occurrence of parafunctional habits in individuals with CP. Sixty-five patients with CP were evaluated through a questionnaire and clinical observation, regarding the following habits: pacifier-sucking, finger-sucking, biting objects, tongue interposition, and bruxism. The results showed that nine (13·8%) patients presented with pacifier-sucking, four (6·1%) showed finger-sucking, 12 (18·4%) had the habit of biting objects, 27 (41·5%) presented with tongue interposition, and 24 (36·9%) had eccentric bruxism. The significance of the presence of oral parafunctional habits in individuals with CP, revealed in this study, justifies the need to establish protocols for adequate prevention and clinical intervention in order to minimize the deleterious consequences that may result from such habits. [source] Motivation for Alcohol Becomes Resistant to Quinine Adulteration After 3 to 4 Months of Intermittent Alcohol Self-AdministrationALCOHOLISM, Issue 9 2010Frederic W. Hopf Background:, Continued consumption of alcohol despite deleterious consequences is a hallmark of alcoholism and represents a critical challenge to therapeutic intervention. Previous rat studies showed that enduring alcohol self-administration despite pairing alcohol with normally aversive stimuli was only observed after very long-term intake (>8 months). Aversion-resistant alcohol intake has been previously interpreted to indicate pathological or compulsive motivation to consume alcohol. However, given the time required to model compulsive alcohol seeking in previous studies, there is considerable interest in developing more efficient and quantitative rodent models of aversion-resistant alcohol self-administration. Methods:, Outbred Wistar rats underwent 3 to 4 months or approximately 1.5 months of intermittent, home-cage, two-bottle access (IAA) to 20% alcohol (v/v) or water. Then, after brief operant training, the effect of the bitter-tasting quinine (0.1 g/l) on the motivation to seek alcohol was quantified via progressive ratio (PR). Motivation for quinine-adulterated 2% sucrose under PR was assayed in a separate cohort of 3 to 4 months IAA rats. The effects of quinine on home-cage alcohol consumption in IAA rats and rats with continuous access to alcohol were also examined. Finally, a dose,response for quinine taste preference in IAA and continuous-access animals was determined. Results:, Motivation for alcohol after 3 to 4 months IAA, measured using an operant PR procedure, was not altered by adulteration of alcohol with 0.1 g/l quinine. In contrast, after 3 to 4 months of IAA, motivation for sucrose under PR was significantly reduced by adulteration of sucrose with 0.1 g/l quinine. In addition, motivation for alcohol after only approximately 1.5 months IAA was significantly reduced by adulteration of alcohol with 0.1 g/l quinine. Furthermore, home-cage alcohol intake by IAA rats was insensitive to quinine at concentrations (0.01, 0.03 g/l) that significantly reduced alcohol drinking in animals with continuous access to alcohol. Finally, no changes in quinine taste preference after 3 to 4 months IAA or continuous access to alcohol were observed. Conclusions:, We have developed a novel and technically simple hybrid operant/IAA model in which quinine-resistant motivation for alcohol is evident after an experimentally tractable period of time (3 to 4 months vs. 8 months). Quinine dramatically reduced sucrose and water intake by IAA rats, indicating that continued responding for alcohol in IAA rats despite adulteration with the normally aversive quinine might reflect maladaptive or compulsive motivation for alcohol. This model could facilitate identification of novel therapeutic interventions for pathological alcohol seeking in humans. [source] Meta-analysis of transmitter effects on avian behaviour and ecologyMETHODS IN ECOLOGY AND EVOLUTION, Issue 2 2010Douglas G. Barron Summary 1. Researchers often attach transmitters and other devices to free-living birds without a clear understanding of potential deleterious consequences to their study organisms, and thus to their data. Studies investigating this topic have generally been limited to a single species or type of device. 2. To achieve a broader understanding we used a meta-analysis of 84 studies to ask: (1) Do devices have an overall effect on birds? (2) Which aspects of avian behaviour and ecology are affected? (3) What attributes of birds influence transmitter effects? (4) What attributes of devices influence their effects? (5) Are effects partially a consequence of capture and restraint? 3. We found a significant negative effect of devices on birds, both overall and for 8 of the 12 specific aspects analysed. The most substantial effects were that birds with devices had markedly increased energy expenditure and were much less likely to nest. 4. Effects were independent of attributes of the birds (sex, age, primary method of locomotion and body mass). We also found no evidence that proportionally heavier devices had greater effects, although researchers generally avoided using heavy devices. Breast-mounted and harness attachments increased device-induced behaviours such as preening, however, and the risk of device-induced mortality differed between attachment methods. 5. Other than foraging behaviours, no effects were a consequence of capture or restraint. 6.Synthesis and applications. We provide the first comprehensive evidence that transmitters and other devices negatively affect birds and may bias resulting data. Researchers should balance the benefits of using these techniques against potential costs to the birds and reliability of the data obtained. [source] Ultrastructural aspects of tomato leaves infected by Tomato torrado virus (ToTV) and co-infected by other virusesPLANT PATHOLOGY, Issue 2 2010A. Alfaro-Fernández Optical and electron microscopy studies were carried out to investigate the cytopathology induced in tomato leaves infected by Tomato torrado virus (ToTV), a new picorna-like virus associated with the ,Torrado' disease. Infected leaves, showing typical Torrado disease symptoms were surveyed in commercial greenhouses in the main tomato production areas of Spain. The effect of the co-infection of ToTV with other viruses which commonly infect tomato crops was also studied. Ultra-thin sections of ToTV-infected tomato leaves did not show a strong cellular alteration. However, crystalline arrays of isometric virus-like particles (VLPs) of 20,30 nm in the inclusion bodies were observed in phloem parenchyma cells of the infected tissues. Tissues co-infected by ToTV and either Tomato chlorosis virus (ToCV) or Pepino mosaic virus (PepMV) presented more severe cellular alterations. The most deleterious consequences for tomato cells were found in triple infections of ToTV, PepMV and Tomato spotted wilt virus (TSWV), where characteristic cell wall overgrowth was distinguishable, together with a large amount of necrotic cells. Changes in the NO/NOS (Nitric Oxide/Nitric Oxide Synthase) system after hypoxia situations remain controversial in this organ, so that the aim of this work is to perform a complete study of this system in the hypoxic lung after different reoxygenation times ranging from 0 h to 5 days posthypoxia. This is a novel follow-up study carried out in Wistar rats submitted for 30 min to acute hypobaric hypoxia. We measured endothelial and inducible NOS (eNOS, iNOS) mRNA and protein expression, location, and in situ NOS activity as well as nitrated protein expression and location. In addition, NO levels were indirectly quantified (NOx) as well as the apoptosis level. Results showed an increase in eNOS mRNA, protein, activity as well as eNOS positive immunostaining at 0 h posthypoxia, coinciding with raised NOx levels. Contrary, iNOS, nitrated protein expression and apoptosis level augmented during the final reoxygenation times. The lung NO/NOS system provokes two responses to the hypoxia/reoxygenation processes: (i) eNOS is responsible of the immediate response, producing NO, which causes vasodilation and bronchodilation, and (ii) iNOS is related to the second late response, which seems to be involved in some of the deleterious consequences that hypoxia induces in the lung. Chidgey The intrathymic differentiation events leading to the development and export of mature T cells tolerant to self yet capable of responding to foreign peptide antigen in the context of self-MHC are clearly both dynamic and complex. The changing phenotype of the developing thymocyte as it migrates through and interacts with the heterogeneous thymic microenvironment and the intracellular signalling events associated with such interactions are being extensively studied, yet many aspects remain poorly defined, such as the precise relationship between stromal cells and thymic selection. Positive and negative selection are crucial events in the development of T cells, leading to a diverse yet non-autoreactive immune system. A breakdown in either of these processes could lead to either a reduced T cell repertoire or the escape into the periphery of autoreactive T cells , both clearly having deleterious consequences for the health of the individual. This review aims to summarise the current status of research in thymic positive selection with emphasis on the role of different cell types and peptides. [source] Effects of Poverty and Maternal Depression on Early Child DevelopmentCHILD DEVELOPMENT, Issue 6 2001Stephen M. Petterson Researchers have renewed an interest in the harmful consequences of poverty on child development. This study builds on this work by focusing on one mechanism that links material hardship to child outcomes, namely the mediating effect of maternal depression. Using data from the National Maternal and Infant Health Survey, we found that maternal depression and poverty jeopardized the development of very young boys and girls, and to a certain extent, affluence buffered the deleterious consequences of depression. Results also showed that chronic maternal depression had severe implications for both boys and girls, whereas persistent poverty had a strong effect for the development of girls. The measures of poverty and maternal depression used in this study generally had a greater impact on measures of cognitive development than motor development. [source] |
