DNAX accessory molecule-1 (CD226) promotes human hepatocellular carcinoma cell lysis by V,9V,2 T cells

Abstract Human V,9V,2 T lymphocytes can be activated by nonpeptidic antigens such as the mevalonate pathway-derived isopentenyl pyrophosphate or synthetic phosphoantigen such as bromohydrin pyrophosphate. They display a strong cytotoxic activity against several tumor types, including hepatocellular carcinoma (HCC). Little is known about the mechanisms underlying V,9V,2 T-cell recognition of tumor cells, but there is strong evidence that activating NK receptors play a role in ,, T-cell cytotoxicity. In this study, we showed that the two NK receptors DNAX accessory molecule-1 (DNAM-1) and CD96 were expressed by V,9V,2 T cells. The ligands Nectin-like-5 specific of both DNAM-1 and CD96, and also Nectin-2, an additional ligand of DNAM-1, were present on all HCC cell lines analyzed. Furthermore, we demonstrated by mAb-mediated masking experiments that cytotoxicity against HCC cells as well as IFN-, production in ,, T cells were dependent on DNAM-1. Our experiments indicated that Nectin-like-5 but not Nectin-2 was involved in DNAM-1-dependent ,, T-cell functions. We did not reveal a role for CD96 in the killing of HCC cells. Finally, we showed by combined mAb-mediated blockade that DNAM-1 and NKG2D could cooperate in the cell lysis of HCC. [source]


Large Second-Order NLO Activity in Poly(4-vinylpyridine) Grafted with PdII and CuII Chromophoric Complexes with Tridentate Bent Ligands Containing Heterocycles

Abstract Two N -salicylidene- N,-acylhydrazines tridentate ligands containing heterocycles were prepared and utilized for the synthesis of mononuclear acentric complexes of CuII and PdII and pyridine serves as an additional ligand (MLxPy, M = metal, Lx = ligand). Characterization also includes single-crystal X-ray diffraction analysis. These complexesshow high second-order nonlinear optical activity (,, = 620,×,10,48esu for both CuLIIPy and PdLIIPy and ,, = 1600,×,10,48 and 1400,×,10,48 esu for CuLIPy and PdLIPy, respectively, at incident wavelength of 1907 nm). The properties of polymers (PMLx) obtained by grafting poly(4-vinylpyridine) with fragments of the complexes are also reported. The polymers exhibit good thermal stability, high glass-transition temperatures and the absence of crystallinity. The second-order nonlinear optical (NLO) properties of thin, transparent poled films, prepared by spin coating and thermal corona poling, were investigated for some of the polymers. The value of the d33 parameter was measured at 1064 and 1500 nm (i.e., with and without resonance contribute). In the latter case, the d33 values are 21 and 12 pm/V for PCuLII and PPdLII, respectively. A high time stability of the NLO properties of these materials was found. (© Wiley-VCH Verlag GmbH & Co. Tuning the Magnetic Properties of a Dinuclear Copper Complex: From Ferromagnetic to Antiferromagnetic Coupling

Abstract A novel structural motif in oxalate bridged copper(II) complexes has been prepared namely a metallamacrocyclic compound of the dinucleating ligand 1,3-bis[bis(2-pyridylmethyl)amino]benzene (= 1,3-tpbd). It could be demonstrated that the magnetic properties of copper(II) complexes of the 1,3-tpbd ligand can be adjusted from weakly ferromagnetic (J = +9.3 cm,1) to strongly antiferromagnetic [J = ,366(9) cm,1] by variation of the additional ligands (e.g. perchlorate, azide, oxalate etc.). Highly Practical "Ligand-Free-Like" Copper-Catalyzed N -Arylation of Azoles in Lower Nitrile Solvents

Synthesis of Schiff Base-Ruthenium Complexes and Their Applications in Catalytic Processes

Abstract The synthesis of various Schiff base mononuclear and binuclear ruthenium complexes, whose additional ligands around the metal core have been selected from an array of motifs, is described. These types of ruthenium complexes, conveniently prepared from commonly available ruthenium sources, are rather stable, display good tolerance towards diverse organic functionalities and also to air and moisture. Remarkably, they exhibit a high activity and chemoselectivity in a variety of catalytic processes such as ring-closing metathesis (RCM), Kharasch addition, alkyne dimerization, enol ester synthesis, ring-opening metathesis polymerization (ROMP) and atom-transfer radical polymerization (ATRP). This review covers both homogeneous and heterogeneous hybrid Schiff base-ruthenium complexes. [source]
