Service selection and workflow mapping for Grids: an approach exploiting quality-of-service information

Abstract The advent of heterogeneous and distributed environments, such as Grid environments, made feasible the solution to computational-intensive problems in a reliable and cost-effective manner. In parallel, workflows with increased complexity that require specialized systems to deal with them are emerging, so as to carry out more composite and mission-critical applications. In that rationale, quality-of-service (QoS) issues need to be tackled in order to ensure that each application satisfies the corresponding user requirements. Therefore, considering the quality provision aspect as fundamental for enabling Grid applications to become QoS compliant, we present an approach for service selection using QoS criteria. The latter is achieved with a suite of components that allow the different mappings of application workflow processes to Grid services that not only meet the user goals and requirements but also maximize his/her benefit in terms of the offered QoS level. We also demonstrate the operation of the aforementioned suite of components and evaluate its performance and effectiveness using a Grid scenario, based on a 3D image rendering application. Establishment of a chick embryo model for analyzing liver development and a search for candidate genes

The liver plays a crucial role in metabolism. There is considerable interest in how the liver develops, as such knowledge could prove of importance in regenerative medicine. However, our understanding of liver development remains somewhat limited. We have developed a model system using the chick embryo that is cost effective and is easy to manipulate experimentally. We performed four fundamental studies: (i) construction of an atlas of the developing chick liver; (ii) identification of differentiation marker genes in the developing chick embryo; (iii) development of germ-layer specific electroporation; and (iv) establishment of organ culture from the developing chick liver. Using this system, we have been able to demonstrate the functions of candidate genes within a shorter period and in a more cost-effective manner. In parallel with the establishment of this system, we examined the expression patterns of genes known to be required for organ development in the developing chick embryo in order to identify genes also involved in liver development. To date, we have found sixteen genes that are expressed in the developing chick liver (GELD, genes expressed in liver development). This knowledge will be fundamental to the establishment of the basic technology for engineering liver tissue in the future. [source]


Rapid plant diversity assessment using a pixel nested plot design: A case study in Beaver Meadows, Rocky Mountain National Park, Colorado, USA

ABSTRACT Geospatial statistical modelling and thematic maps have recently emerged as effective tools for the management of natural areas at the landscape scale. Traditional methods for the collection of field data pertaining to questions of landscape were developed without consideration for the parameters of these applications. We introduce an alternative field sampling design based on smaller unbiased random plot and subplot locations called the pixel nested plot (PNP). We demonstrate the applicability of the PNP design of 15 m × 15 m to assess patterns of plant diversity and species richness across the landscape at Rocky Mountain National Park (RMNP), Colorado, USA in a time (cost)-efficient manner for field data collection. Our results produced comparable results to a previous study in the Beaver Meadow study (BMS) area within RMNP, where there was a demonstrated focus of plant diversity. Our study used the smaller PNP sampling design for field data collection which could be linked to geospatial information data and could be used for landscape-scale analyses and assessment applications. In 2003, we established 61 PNP in the eastern region of RMNP. We present a comparison between this approach using a sub-sample of 19 PNP from this data set and 20 of Modified Whittaker nested plots (MWNP) of 20 m × 50 m that were collected in the BMS area. The PNP captured 266 unique plant species while the MWNP captured 275 unique species. Based on a comparison of PNP and MWNP in the Beaver Meadows area, RMNP, the PNP required less time and area sampled to achieve a similar number of species sampled. Using the PNP approach for data collection can facilitate the ecological monitoring of these vulnerable areas at the landscape scale in a time- and therefore cost-effective manner. [source]


Monoclonal antibodies: a morphing landscape for therapeutics

Abstract The concept of using antibodies as therapeutics to cure human diseases was postulated nearly 100 years ago by Paul Ehrlich and subsequently enabled by the discovery of hybridoma technology by Kohler and Milstein in 1975. While the use of monoclonal antibodies (mAbs) as drugs that can specifically target a disease-associated antigen is compelling, it has taken a quarter century for these molecules to be adopted as bona fide therapeutic agents. Despite their slow pursuit in drug development during the pioneering years, it is now estimated that there are nearly 500 mAb-based therapies in development. Major factors that have influenced the acceptance of monoclonal antibodies as therapeutics include their drug safety profiles, technological advancements for facilitating mAb discovery and development, and market success. Early on, it was demonstrated that antibodies could elicit clinical benefit by antagonizing a specific antigen without the common side effects that are prevalent with small chemical entities due to their nonspecific effects on homeostatic biochemical pathways. In addition, the significant technological advances that the biotechnology industry has established for developing and producing monoclonal antibodies at commercial scale in a more efficient and cost-effective manner has broadly enabled their use as therapeutics. However, despite the beneficial pharmacologic advantages and technological advances, it has been the sheer market success that monoclonal antibody products have achieved over the past few years that has propelled their vast pursuit by the biopharmaceutical industry in light of their value-creating potential. Here we provide an overview of the monoclonal antibody industry and discuss evolving technologies and strategies that are being pursued to overcome challenges in the changing marketplace. Drug Dev. A cost-effective simulation curriculum for preclinical endodontics

A challenge in contemporary dental education is to achieve a smooth transition from preclinical teaching environments to patient-care clinics in a cost-effective manner. The preclinical endodontic courses at The University of Texas, Dental Branch at Houston provide a unique learning environment that enables the student to perform endodontic treatment on extracted teeth in a typodont, and be involved in diagnosis and treatment-planning discussions. The specially designed stone typodont used has built-in radiographic capability, and is mounted at each chair in the clinic. During each preclinical session, students are assigned clinical cubicles and proper aseptic protocol is followed. Students are required to wear gloves, masks and eyewear, and place a rubber dam during treatment. Written self-assessment evaluations based upon prescribed criteria are utilised; feedback is given by faculty composed of both full-time endodontists and graduate students who periodically rotate and are calibrated on a regular basis. In the lecture phase, clinical case scenarios are presented to reinforce concepts of diagnosis and emergency care and to help integrate endodontics with other disciplines; a Socratic-like teaching style is established by the faculty facilitator to create an environment for developing critical-thinking and problem-solving skills. The overall feedback from graduating students has been very positive. Advantages of this format are an easier transition to patient management, a more keen interest in specialsation and a perceived increase in levels of confidence. [source]


An Innovative Strategy for Conducting Clinical Research: The Academic Associate Program

Performance of high-quality clinical research usually requires procurement of substantial funding to support and sustain the research infrastructure. The authors have developed a program to support a research infrastructure that minimizes financial start-up costs and maximizes productivity of clinical-educator faculty with limited protected time. The program integrates clinical research with undergraduate education. The authors report the experience with the academic associate program at two academic institutions. They describe the program, provide measures of program success, and discuss potential methods for other institutions to initiate similar programs. Similar programs can be developed in a variety of outpatient and inpatient settings allowing the performance of high-quality clinical research in a time-efficient and cost-effective manner. Bioaccumulation Assessment Using Predictive Approaches,

Abstract Mandated efforts to assess chemicals for their potential to bioaccumulate within the environment are increasingly moving into the realm of data inadequacy. Consequently, there is an increasing reliance on predictive tools to complete regulatory requirements in a timely and cost-effective manner. The kinetic processes of absorption, distribution, metabolism, and elimination (ADME) determine the extent to which chemicals accumulate in fish and other biota. Current mathematical models of bioaccumulation implicitly or explicitly consider these ADME processes, but there is a lack of data needed to specify critical model input parameters. This is particularly true for compounds that are metabolized, exhibit restricted diffusion across biological membranes, or do not partition simply to tissue lipid. Here we discuss the potential of in vitro test systems to provide needed data for bioaccumulation modeling efforts. Recent studies demonstrate the utility of these systems and provide a "proof of concept" for the prediction models. Computational methods that predict ADME processes from an evaluation of chemical structure are also described. Most regulatory agencies perform bioaccumulation assessments using a weight-of-evidence approach. A strategy is presented for incorporating predictive methods into this approach. To implement this strategy it is important to understand the "domain of applicability" of both in vitro and structure-based approaches, and the context in which they are applied. [source]


A novel fault management approach for DWDM optical networks

Connection availability is considered as a critical metric when providing differentiated services in Wavelength-Division Multiplexing mesh networks. Indeed, one of the major concerns of optical network operators is related to improving the availability of services provided to their highest-class clients. Achieving this objective is possible through managing faults using the different classical protection schemes, namely the so-called dedicated and shared protection schemes. However, the majority of the work concerning protection schemes has considered the primary connections as equally important when contending for the use of the backup resources. As a main contribution in this paper, we therefore propose an improvement of the existing protection schemes through the introduction of relative priorities among the different primary connections contending for the access to the protection path. To evaluate numerically the benefits of the service differentiation feature introduced in our proposal, we first develop a mathematical model, based on which we derive explicit expressions for the average connection availabilities that result from both the classical protection schemes and the proposed priority-aware one. Through this model, we show how the availability of the highest-class clients is improved when deploying the proposed priority-aware protection scheme. Finally, with the same objective in mind, we develop a simulation study, where a given set of connection demands with predefined availability requirements is provisioned using different protection strategies. Through this study, we show that the priority-aware protection strategy satisfies service-availability requirements in a cost-effective manner compared with the classical protection schemes.Copyright © 2006 John Wiley & Sons, Ltd. Sensitivity and specificity of current diagnostic tests for gill-associated virus in Penaeus monodon

Abstract This study reports the sensitivity, specificity, positive predictive value (PPV), negative predictive value (NPV) and accuracy between a reverse transcriptase-nested polymerase chain reaction (RT-nPCR) and an enzyme-linked immunosorbent assay (ELISA) for the detection of gill-associated virus (GAV) from a sample of 120 Penaeus monodon. Subsequently, the same comparisons were applied to the ELISA and haemagglutination (HA) assays for detection of GAV from a second 120 prawns. The optical density (OD) or dilution cut-off point had a direct influence on the tested parameters. The cut-off OD of 0.5,0.6 with the ELISA produced a sensitivity of 98% compared with RT-nPCR. However, these OD produced the lowest accuracy (85.8% and 86.7%, respectively). The OD cut off of 0.75 resulted in the highest accuracy (91.7%) and NPV (81.3%) while it had the second highest sensitivity (97%) and PPV (93.3%). However, the OD cut off of 0.9 had the highest specificity (80%). With regards to HA, the titre cut off at 8 resulted in the highest sensitivity, specificity and NPV (94%, 100% and 100%, respectively) compared with the ELISA, while the HA titre of 16 gave the highest accuracy (73%) and the second highest specificity (75%). A HA titre of 64 gave the highest PPV (81%). Using the RT-nPCR as the gold standard, the ELISA had an accuracy of 91.7% when using a cut off >0.75 as a positive result. When compared with the ELISA, the HA had an accuracy of 73% when using an HA titre cut off greater than 16 as a positive result. These results indicate that alternative tests for GAV (ELISA and HA) can be used to explore multiple questions about the disease status of P. monodon stocks in a cost-effective manner. [source]


Compartmentalisation: flood consequence reduction by splitting up large polder areas

Abstract River and coastal floodplains are often protected by vast systems of connected embankments. In the Netherlands, about 55% of the country is protected in this way, by some 3600 km of primary defences. The protection level is probably the highest in the world, and annual mean flood risks are low. Nevertheless, the consequences of a major flood event might be unacceptable. This is a reason to consider whether and how the consequences of floods could be reduced in a cost-effective manner. Splitting up large polder areas into smaller portions, the so-called compartmentalisation, would reduce the area subject to flooding, and thus the economic damage, the number of people exposed and the fatality risk. We carried out a policy analysis for the national authorities in order to establish whether, where and under which conditions a further compartmentalisation of the country would be desirable. This paper gives some results, discusses our experiences in four case studies and finally focuses on the fundamental questions of assessment and the trade-off between better flood protection and the benefit/cost ratio of reducing consequences. Cambium cell stimulation from surgical release of the periosteum

An autograft of periosteal tissue containing cambium cells has potential to become chondrogenic or osteogenic depending on the regeneration repair strategies. The potential number of harvestable cambium cells diminishes with age. Other factors may be associated with a reduction in the number or variable yields of cambium cells including harvest technique, harvest site location, and the time interval from harvest to implantation. Attempts to increase the number of cambium cells have included improvements in harvesting and handling technique, and expansion of the cells in tissue culture. An ,in situ" stimulation and proliferation technique would offer the potential for increasing the number of cambium cells in a cost-effective manner for transplantation without the need for expansion in tissue culture. The hypothesis tested was that surgical release of the periosteum and its deep inner underlying cambium layer by sharply incising through the superficial periosteal fibrous layer down to and scoring the cortical bone surface would increase the number of cambium cells that could be harvested at a later time period. Two techniques for periosteal release were used to stimulate a proliferation of the underlying cambium layer and increase the cambium cells for harvest in skeletally mature goats: (1) sharply scoring all four-sides of the tissue test site perimeter, and (2) sharply scoring only two sides of the tissue test site. The two-sided and four-sided release scoring of the periosteum induced stimulatory responses in the number of cambium cells. In addition, a marked increase in mRNA expression for BMP-2 (p < 0.001) was observed within 24 h and remained elevated over baseline values for up to 96 h after this stimulation to the cambium layer. © 2002 Orthopaedic Research Society. PV thermal systems: PV panels supplying renewable electricity and heat

Abstract With PV Thermal panels sunlight is converted into electricity and heat simultaneously. Per unit area the total efficiency of a PVT panel is higher than the sum of the efficiencies of separate PV panels and solar thermal collectors. During the last 20 years research into PVT techniques and concepts has been widespread, but rather scattered. This reflects the number of possible PVT concepts and the accompanying research and development problems, for which it is the general goal to optimise both electrical and thermal efficiency of a device simultaneously. The aspects that can be optimised are, amongst others, the spectral characteristics of the PV cell, its solar absorption and the internal heat transfer between cells and heat-collecting system. Another important level of optimisation is for the PVT device geometry and the integration into a system. The electricity and heat demand and the temperature level of the heat determine the choice for a certain system set-up. With an optimal design, PVT systems can supply buildings with 100% renewable electricity and heat in a more cost-effective manner than separate PV and solar thermal systems and thus contribute to the long-term international targets on implementation of renewable energy in the built environment. Promoting Infant Health Through Home Visiting By a Nurse-Managed Community Worker Team

This article describes the Resources, Education and Care in the Home program (REACH-Futures), an infant mortality reduction initiative in the inner city of Chicago built on the World Health Organization (WHO) primary health care model and over a decade of experience administering programs to reduce infant mortality through home visits. The program uses a nurse-managed team, which includes community residents selected, trained, and integrated as health advocates. Service participants were predominately African American families. All participants were low-income and resided in inner-city neighborhoods with high unemployment, high teen birth rates, violent crime, and deteriorated neighborhoods. Outcomes for the first 666 participants are compared to a previous home-visiting program that used only nurses. Participant retention rates were equivalent overall and significantly higher in the first months of the REACH-Futures program. There were two infant deaths during the course of the study, a lower death rate than the previous program or the city. Infant health problems and developmental levels were equivalent to the prior program and significantly more infants were fully immunized at 12 months. The authors conclude that the use of community workers as a part of the home-visiting team is as effective as the nurse-only team in meeting the needs of families at high risk of poor infant outcomes. This approach is of national interest because of its potential to achieve the desired outcomes in a cost-effective manner. [source]


Characterizing a Brownfields recreational reuse scenario using the Triad Approach,Assunpink Creek Greenways project

Since the early 1990s the U.S. government has been developing and implementing public policies that advance the redevelopment of brownfields, and the recent passage of the Small Business Liability Relief and Brownfields Revitalization Act (SBLRBRA) will significantly advance efforts to integrate environmental contamination mitigation and redevelopment. Experience has demonstrated that successful redevelopment requires the collection, analysis, and interpretation of environmental data in a timely and cost-effective manner in order to allow developers and lenders to efficiently use cleanup resources, develop response strategies that integrate cleanup with redevelopment, and support meaningful outreach to involved stakeholders. Recent advances in the science and technology of site characterization hold the promise of improved site characterization outcomes while saving time and money. One such advancement, the Triad Approach, combines systematic up-front planning with the use of a dynamic field investigation process and the generation of real time data to allow in-field decision making on sample location selection. This article describes an application of the Triad Approach to redevelopment of an urban greenway in Trenton, New Jersey. The Triad Approach, initiated through a partnership between the City of Trenton, New Jersey Department of Environmental Protection, New Jersey Institute of Technology, and the U.S. Environmental Protection Agency, demonstrated that this approach could accelerate the characterization of the 60-acre, 11-parcel project area. Environmental issues that were solved using the Triad Approach included the delineation of the extent of historic fill, determination of no further action for several areas of concern, detailed investigation of specific impacted areas and the acquisition of sufficient data to allow the city to make important decisions regarding remediation costs and property acquisition. © 2003 Wiley Periodicals, Inc. Development and optimization of a process for automated recovery of single cells identified by microengraving

Abstract Microfabricated devices are useful tools for manipulating and interrogating large numbers of single cells in a rapid and cost-effective manner, but connecting these systems to the existing platforms used in routine high-throughput screening of libraries of cells remains challenging. Methods to sort individual cells of interest from custom microscale devices to standardized culture dishes in an efficient and automated manner without affecting the viability of the cells are critical. Combining a commercially available instrument for colony picking (CellCelector, AVISO GmbH) and a customized software module, we have established an optimized process for the automated retrieval of individual antibody-producing cells, secreting desirable antibodies, from dense arrays of subnanoliter containers. The selection of cells for retrieval is guided by data obtained from a high-throughput, single-cell screening method called microengraving. Using this system, 100 clones from a mixed population of two cell lines secreting different antibodies (12CA5 and HYB099-01) were sorted with 100% accuracy (50 clones of each) in ,2 h, and the cells retained viability. © 2010 American Institute of Chemical Engineers Biotechnol.
