Development of Cu and Zn Isotope MC-ICP-MS Measurements: Application to Suspended Particulate Matter and Sediments from the Scheldt Estuary

Calibration of reference materials (such as the IRMM 3702 Zn) against the JMC Zn and NIST Cu reference materials were performed in wet and/or dry plasma modes (Aridus I and DSN-100) on a Nu Plasma MC-ICP-MS. Different mass bias correction methods were compared. More than 100 analyses of certified reference materials suggested that the sample-calibrator bracketing correction and the empirical external normalisation methods provide the most reliable corrections, with long term external precisions of 0.06 and 0.07, (2SD), respectively. Investigation of the effect of variable analyte to spike concentration ratios on Zn and Cu isotopic determinations indicated that the accuracy of Cu measurements in dry plasma is very sensitive to the relative Cu and Zn concentrations, with deviations of ,65Cu from ,0.4, (Cu/Zn = 4) to +0.4, (Cu/Zn = 0.2). A quantitative assessment (with instrumental mass bias corrections) of spectral and non-spectral interferences (Ti, Cr, Co, Fe, Ca, Mg, Na) was performed. Titanium and Cr were the most severe interfering constituents, contributing to inaccuracies of ,5.1, and +0.60, on ,68/64Zn, respectively (for 500 ,g l,1 Cu and Zn standard solutions spiked with 1000 ,g l,1 of Ti or Cr). Preliminary isotopic results were obtained on contrasting sediment matrices from the Scheldt estuary. Significant isotopic fractionation of zinc (from 0.21, to 1.13, for ,66Zn) and copper (from ,0.38, to 0.23, for ,65Cu), suggest a control by physical mixing of continental and marine water masses, characterized by distinct Cu and Zn isotopic signatures. These results provide a stepping-stone to further evaluate the use of Cu and Zn isotopes as biogeochemical tracers in estuarine environments. L'étude présentée ici porte sur l'évaluation critique d'un certain nombre de paramètres contrôlant la précision et la justesse des mesures des isotopes de Cu et Zn, dans le cadre d'une application à du matériel particulaire estuarien. Une calibration de matériaux de référence (tels que le Zn IRMM 3702) par rapport aux matériaux de référence JMC Zn et NIST Cu a été effectuée avec des plasmas humides et secs (avec Aridus I et DSN-100) sur un MC-ICP-MS Nu. Différentes méthodes de correction de biais de masse ont été comparées. Plus de 100 analyses de matériaux de référence certifiés ont montré que la correction par l'intercalation d'un calibrateur entre chaque échantillon et la calibration externe empirique fournissaient les corrections les plus fiables, avec des précisions externes sur le long terme de 0.06 et 0.07, (2SD) respectivement. Les effets de la variation des rapports de concentrations entre analyte et spike sur les mesures des rapports isotopiques de Cu et Zn ont montré que la justesse des mesures pour Cu en plasma sec est très tributaire des concentrations relatives de Cu et Zn, avec des déviations de ,65Cu allant de ,0.4, (Cu/Zn = 4) à+0.4, (Cu/Zn = 0.2). Ti et Cr se sont révélés être les constituants interférents les plus importants pouvant entraîner des erreurs de ,5.1, et +0.60, sur ,68/64Zn respectivement (pour des solutions standards à 500 ,g l,1 de Cu et Zn dopées avec 1000 ,g l,1 de Ti ou Cr). Des données isotopiques préliminaires ont été obtenues sur des matrices sédimentaires très différentes provenant de l'estuaire de Scheldt. Les fractionnements significatifs du zinc (de 0.21,à 1.13, pour ,66Zn) et du cuivre (de ,0.38,à 0.23, pour ,65Cu) suggèrent un contrôle par un processus physique de mélange entre des masses d'eaux continentales et marines ayant des signatures isotopiques de Cu et Zn distinctes. An analysis and comparison of the time accuracy of fractional-step methods for the Navier,Stokes equations on staggered grids

Abstract Fractional-step methods solve the unsteady Navier,Stokes equations in a segregated manner, and can be implemented with only a single solution of the momentum/pressure equations being obtained at each time step, or with the momentum/pressure system being iterated until a convergence criterion is attained. The time accuracy of such methods can be determined by the accuracy of the momentum/pressure coupling, irrespective of the accuracy to which the momentum equations are solved. It is shown that the time accuracy of the basic projection method is first-order as a result of the momentum/pressure coupling, but that by modifying the coupling directly, or by modifying the intermediate velocity boundary conditions, it is possible to recover second-order behaviour. It is also shown that pressure correction methods, implemented in non-iterative or iterative form and without special boundary conditions, are second-order in time, and that a form of the non-iterative pressure correction method is the most efficient for the problems considered. Wavelet analysis applied to removing non-constant, varying spectroscopic background in multivariate calibration

Abstract Multiresolution, the ability to separate signals according to frequency, is one of the main advantages offered by the wavelet transform. However, the coarsening of resolution associated with this method may be problematic in some applications. The ,wavelet prism' (WP) method proposed here can split the signal into different frequency components, which retain the original resolution of the signal. In conjunction with a maximum information gain criterion developed here, this new method can be used to judge and remove the low-frequency non-constant background variation reasonably and automatically. In this paper the theory and background concerning this wavelet baseline correction method are introduced. The method is successfully applied to simulated and real near-infrared (NIR) spectral data to deal with non-constant background for multivariate calibration. Its performance compares favorably with the current signal correction methods for background removal. The new method appears to be an efficient method for removal of non-constant, varying spectroscopic background, leading to a simpler and more parsimonious multivariate linear model. Tropical forest tree mortality, recruitment and turnover rates: calculation, interpretation and comparison when census intervals vary

Summary 1Mathematical proofs show that rate estimates, for example of mortality and recruitment, will decrease with increasing census interval when obtained from censuses of non-homogeneous populations. This census interval effect could be confounding or perhaps even driving conclusions from comparative studies involving such rate estimates. 2We quantify this artefact for tropical forest trees, develop correction methods and re-assess some previously published conclusions about forest dynamics. 3Mortality rates of > 50 species at each of seven sites in Africa, Latin America, Asia and Australia were used as subpopulations to simulate stand-level mortality rates in a heterogeneous population when census intervals varied: all sites showed decreasing stand mortality rates with increasing census interval length. 4Stand-level mortality rates from 14 multicensus long-term forest plots from Africa, Latin America, Asia and Australia also showed that, on average, mortality rates decreased with increasing census interval length. 5Mortality, recruitment or turnover rates with differing census interval lengths can be compared using the mean rate of decline from the 14 long-term plots to standardize estimates to a common census length using ,corr = , × t0.08, where , is the rate and t is time between censuses in years. This simple general correction should reduce the bias associated with census interval variation, where it is unavoidable. 6Re-analysis of published results shows that the pan-tropical increase in stem turnover rates over the late 20th century cannot be attributed to combining data with differing census intervals. In addition, after correction, Old World tropical forests do not have significantly lower turnover rates than New World sites, as previously reported. Our pan-tropical best estimate adjusted stem turnover rate is 1.81 ± 0.16% a,1 (mean ± 95% CI, n = 65). 7As differing census intervals affect comparisons of mortality, recruitment and turnover rates, and can lead to erroneous conclusions, standardized field methods, the calculation of local correction factors at sites where adequate data are available, or the use of our general standardizing formula to take account of sample intervals, are to be recommended. The effects of preservation on fish tissue stable isotope signatures

The effects of formalin and ethanol preservation on the ,13C and ,15N isotope signatures of Arctic charr Salvelinus alpinus muscle tissue were examined. The lipid content of the tissue samples studied ranged from 3·6 to 6·1% and was not correlated with the magnitude of observed isotopic shifts in preserved samples. Ethanol and formalin significantly depleted and enriched, respectively, the ,13C isotope signatures of preserved tissues when compared to control samples. Ethanol did not significantly enrich ,15N signatures in comparison to controls, whereas formalin did. A meta-analysis of multiple species effects further demonstrated significant preservation effects in fish tissue. Statistical analysis of data obtained by correcting preserved tissue isotope signatures with literature, bootstrapped or meta-analysis derived correction factors demonstrated significant differences between corrected and control sample isotope signatures or failure to produce a unity slope when the data sets were regressed against one another. Species-specific, bootstrapped linear correction models resulted in no such errors. Results suggest that species-specific correction methods should be used for fishes because of the known wide variation in fish tissue lipid content and composition. Accordingly, the use of pilot studies will be required to develop correction factors that properly adjust for preservation effects when interpreting temporal patterns in historic analyses of food webs. Assessing accuracy of a continuous screening test in the presence of verification bias

Summary., In studies to assess the accuracy of a screening test, often definitive disease assessment is too invasive or expensive to be ascertained on all the study subjects. Although it may be more ethical or cost effective to ascertain the true disease status with a higher rate in study subjects where the screening test or additional information is suggestive of disease, estimates of accuracy can be biased in a study with such a design. This bias is known as verification bias. Verification bias correction methods that accommodate screening tests with binary or ordinal responses have been developed; however, no verification bias correction methods exist for tests with continuous results. We propose and compare imputation and reweighting bias-corrected estimators of true and false positive rates, receiver operating characteristic curves and area under the receiver operating characteristic curve for continuous tests. Distribution theory and simulation studies are used to compare the proposed estimators with respect to bias, relative efficiency and robustness to model misspecification. The bias correction estimators proposed are applied to data from a study of screening tests for neonatal hearing loss. [source]


Independent estimation of T*2 for water and fat for improved accuracy of fat quantification

Abstract Noninvasive biomarkers of intracellular accumulation of fat within the liver (hepatic steatosis) are urgently needed for detection and quantitative grading of nonalcoholic fatty liver disease, the most common cause of chronic liver disease in the United States. Accurate quantification of fat with MRI is challenging due the presence of several confounding factors, including T*2 decay. The specific purpose of this work is to quantify the impact of T*2 decay and develop a multiexponential T*2 correction method for improved accuracy of fat quantification, relaxing assumptions made by previous T*2 correction methods. A modified Gauss-Newton algorithm is used to estimate the T*2 for water and fat independently. Improved quantification of fat is demonstrated, with independent estimation of T*2 for water and fat using phantom experiments. Correction of concomitant magnetic field-induced image artifacts in nonaxial echo-planar imaging,

Abstract Echo-planar images acquired in nonaxial planes are often distorted. Such image distortion has limited the applications of the echo-planar imaging (EPI) technique. In this article, it is demonstrated that a considerable amount of the distortion is caused by the higher-order magnetic field concomitant with the linear magnetic field gradient, or the concomitant magnetic field. The image distortion caused by the concomitant magnetic field is more prominent when a higher gradient amplitude is used for readout. It is also shown that the concomitant magnetic field can cause ghosting and blurring. A theoretical analysis is performed for the concomitant field effect in nonaxial EPI images. A point-by-point (or line-by-line) phase correction algorithm is developed to correct the image distortion, ghosting, and blurring. A postreconstruction processing algorithm is also developed to correct image distortion with much higher computational efficiency. Evaluation of a Single-monochromator Diode Array Spectroradiometer for Sunbed UV-radiation Measurements,

ABSTRACT The suitability of a new technology single-monochromator diode array spectroradiometer for UV-radiation safety measurements, in particular for sunbed measurements, was evaluated. The linearity, cosine response, temperature response, wavelength scale, stray-light and slit function of the spectroradiometer were determined and their effects on the measurement accuracy evaluated. The main error sources were stray-light and nonideal cosine response, for which correction methods are presented. Without correction, the stray-light may reduce the accuracy of the measurement excessively, particularly in the UV-B range. The expanded uncertainty of the corrected UV measurements is estimated to be 14%, which is confirmed with the comparative measurements carried out with a well-characterized double-monochromator spectroradiometer. The measurement accuracy is sufficient for sunbed measurements, provided that all corrections described above have been done and the user of the instrument has a good understanding of the instrument's operating principles and potential error sources. If these requirements are met, the tested spectroradiometer improves and facilitates market surveillance field measurements of sunbeds. [source]


Magnitude of Error Introduced by Application of Heart Rate Correction Formulas to the Canine QT Interval

Background: Accurate detection of drug-induced QT interval changes is often confounded by concurrent heart rate changes. Application of heart rate correction formulas has been the traditional approach to account for heart rate,induced QT interval changes, and thereby identify the direct effect of the test article on cardiac repolarization. Despite numerous recent studies identifying the imprecision of these formulas they continue to be applied. Methods: Using a chronic atrioventricular dissociated His-paced canine model, heart rate correction methods were evaluated for their ability to generate a corrected QT interval independent of original heart rate. Additionally, His bundle pacing at a heart rate of 60 beats/min allowed calculation of the magnitude of error introduced by application of heart rate correction formulas. Results: Of the fixed parameter heart rate correction formulas, only Van de Water was able to predict corrected QT values independent of the original heart rate. The magnitude of error discovered by application of heart rate correction formulas varied, but in many cases was very large. Bazett's formula was associated with a mean overcorrection of 67.9 ms; Fridericia's 28.7 ms. Van de Water was the best fixed parameter formula with a mean error of 10.8 ms. As expected, group and individual corrections derived from linear regression of the HR-QT data offered improvement over the traditional formulas. Both were able to predict QTc values independent of the heart rate. However, errors of the magnitude of 10 and 6 ms, respectively, were still introduced. Conclusion: Van de Water and linear regression correction methods were superior to others in this study, but all methods generated QTc errors equal to or much greater than the magnitude of interest for drug safety evaluation. Youden Index and Optimal Cut-Point Estimated from Observations Affected by a Lower Limit of Detection

Abstract The receiver operating characteristic (ROC) curve is used to evaluate a biomarker's ability for classifying disease status. The Youden Index (J), the maximum potential effectiveness of a biomarker, is a common summary measure of the ROC curve. In biomarker development, levels may be unquantifiable below a limit of detection (LOD) and missing from the overall dataset. Disregarding these observations may negatively bias the ROC curve and thus J. Several correction methods have been suggested for mean estimation and testing; however, little has been written about the ROC curve or its summary measures. We adapt non-parametric (empirical) and semi-parametric (ROC-GLM [generalized linear model]) methods and propose parametric methods (maximum likelihood (ML)) to estimate J and the optimal cut-point (c *) for a biomarker affected by a LOD. We develop unbiased estimators of J and c * via ML for normally and gamma distributed biomarkers. Alpha level confidence intervals are proposed using delta and bootstrap methods for the ML, semi-parametric, and non-parametric approaches respectively. Simulation studies are conducted over a range of distributional scenarios and sample sizes evaluating estimators' bias, root-mean square error, and coverage probability; the average bias was less than one percent for ML and GLM methods across scenarios and decreases with increased sample size. An example using polychlorinated biphenyl levels to classify women with and without endometriosis illustrates the potential benefits of these methods. We address the limitations and usefulness of each method in order to give researchers guidance in constructing appropriate estimates of biomarkers' true discriminating capabilities. Lamotrigine does not prolong QTc in a thorough QT/QTc study in healthy subjects

WHAT IS ALREADY KNOWN ABOUT THIS SUBJECT , Drugs that inhibit the human cardiac delayed rectifier potassium current may lead to prolongation of the cardiac QT interval and are associated with a fatal, polymorphic, ventricular tachycardia known as torsades de pointes. , Lamotrigine is indicated in the treatment of epilepsy and the prevention of mood episodes in patients with bipolar disorder. , Lamotrigine inhibits the human cardiac delayed rectifier potassium current in vitro, and it has been hypothesized that QT prolongation may contribute to the risk of sudden unexpected death in epilepsy patients. WHAT THIS STUDY ADDS , This is the first reported thorough QT/QTc study with lamotrigine conducted to International Conference on Harmonization guidelines. , The mean QTc interval was not prolonged by lamotrigine in healthy subjects, as assessed by the standard heart rate correction methods (Fridericia's and Bazett's). , The in vitro inhibition of the delayed rectifier potassium current does not translate into an effect on QT in man. AIM To characterize the effects of lamotrigine on QT interval in healthy subjects. METHODS Healthy subjects received a single oral dose of moxifloxacin (400 mg) or placebo in crossover design, followed by a dose-escalating regimen of lamotrigine (n = 76) over a 77-day period, or matched placebo (n = 76). Blood samples were taken for determination of moxifloxacin and lamotrigine concentrations and digital 12-lead ECGs were recorded. The relationships between individual QT values and respective individual moxifloxacin or lamotrigine concentrations were explored using population pharmacokinetic,pharmacodynamic (PK,PD) modelling. RESULTS Moxifloxacin was associated with a maximum mean increase from baseline in QTcF of 14.81 ms [90% confidence interval (CI) 13.50, 16.11] 2.5 h after dosing. Steady-state exposure to lamotrigine (50, 150 or 200 mg b.d.) was not associated with an increase in QTc interval. Small reductions in QTcF (maximum mean difference from placebo ,7.48 ms, 90% CI ,10.49, ,4.46) and small increases in heart rate (maximum mean difference from placebo 5.94 bpm, 90% CI 3.81, 8.06) were observed with lamotrigine 200 mg b.d. vs. placebo. No effect of lamotrigine on QRS duration or blood pressure was observed. No outliers with QTcF > 450 ms, or with an increase from baseline of >60 ms were observed in the lamotrigine group. PK,PD modelling indicated statistically significant decreases in individually corrected QT intervals for lamotrigine and statistically significant increases in individually corrected QT intervals for moxifloxacin over the concentration ranges studied. CONCLUSIONS Therapeutic doses of lamotrigine (50,200 mg b.d.) were not associated with QT prolongation in healthy subjects. [source]
