DAP kinase activity is critical for C2 -ceramide-induced apoptosis in PC12 cells

Exposure of PC12 cells to C2 -ceramide results in dose-dependent apoptosis. Here, we investigate the involvement of death-associated protein (DAP) kinase, initially identified as a positive mediator of the interferon-,-induced apoptosis of HeLa cells, in the C2 -ceramide-induced apoptosis of PC12 cells. DAP kinase is endogenously expressed in these cells. On exposure of PC12 cells to 30 µm C2 -ceramide, both the total (assayed in the presence of Ca2+/calmodulin) and Ca2+/calmodulin-independent (assayed in the presence of EGTA) DAP kinase activities were transiently increased 5.0- and 12.2-fold, respectively, at 10 min, and then decreased to 1.7- and 3.4-fold at 90 min. After 10 min exposure to 30 µm C2 -ceramide, the Ca2+/calmodulin independent activity/ total activity ratio increased from 0.22 to 0.60. These effects were dependent on the C2 -ceramide concentration. C8 -ceramide, another active ceramide analog, also induced apoptosis and activated DAP kinase, while C2 -dihydroceramide, an inactive ceramide analog, failed to induce apoptosis and increase DAP kinase activity. Furthermore, transfection studies revealed that overexpression of wild-type DAP kinase enhanced the sensitivity to C2 - and C8 -ceramide, while a catalytically inactive DAP kinase mutant and a construct containing the death domain and C-terminal tail of DAP kinase, which act in a dominant-negative manner, rescued cells from C2 -, and C8 -ceramide-induced apoptosis. These findings demonstrate that DAP kinase is an important component of the apoptotic machinery involved in ceramide-induced apoptosis, and that the intrinsic DAP kinase activity is critical for ceramide-induced apoptosis. Diagnostic value of leptin in tuberculous pleural effusions

Summary It is suggested that leptin may be involved in inflammation. Although relation between leptin levels and active pulmonary tuberculosis has been studied, there is no information about relation between leptin levels and tuberculous pleural effusions (TPE). We evaluated the diagnostic value of pleural fluid and serum leptin levels in TPE and compared them with adenosine deaminase (ADA). Forty-five patients, 17 tuberculous effusion and 28 nontuberculous effusion, with exudative pleural effusions were included. Leptin and ADA levels were measured from serum and pleural fluid in all patients. There were no statistically significant differences between tuberculous and nontuberculous groups with respect to the serum ADA activity and pleural fluid/serum leptin ratio. On the contrary, pleural fluid leptin level, pleural fluid ADA activity, serum leptin level and pleural fluid/serum ADA activity ratio were statistically different between tuberculous and nontuberculous groups. When leptin levels were corrected for body mass index, serum leptin levels did not reach statistical significance. Cut-off points to predict tuberculosis were calculated as 9.85 ng/ml and 35.55 U/l for pleural fluid leptin level and pleural fluid ADA activity, respectively. Sensitivity, specificity and area under the curve ± standard error were 82.4%, 82.1%, 0.83 ± 0.07 for pleural fluid leptin levels and 100%, 100%, 1.00 ± 0.00 for pleural fluid ADA activity, respectively; the difference between these curves was significant (p = 0.01). Pleural fluid leptin levels were lower in tuberculous effusions than in other exudates. Pleural fluid leptin has a diagnostic value for TPE but not as good as that of ADA. [source]


Penicillium chrysogenum glucose oxidase , a study on its antifungal effects

Abstract Aims:, Purification and characterization of the high molecular mass Candida albicans -killing protein secreted by Penicillium chrysogenum. Methods and Results:, The protein was purified by a combination of ultrafiltration, chromatofocusing and gel filtration. Enzymological characteristics [relative molecular mass (Mr) = 155 000, subunit structure ,2 with Mr,, = 76 000, isoelectric point (pI) = 5·4] were determined using SDS-PAGE and 2D-electrophoresis. N-terminal amino acid sequencing and homology search demonstrated that the antifungal protein was the glucose oxidase (GOX) of the fungus. The enzyme was cytotoxic for a series of bacteria, yeasts and filamentous fungi. Vitamin C (1·0 mg ml,1) prevented oxidative cell injuries triggered by 0·004 U GOX in Emericella nidulans cultures but bovine liver catalase was ineffective even at a GOX : catalase activity ratio of 0·004 : 200 U. A secondary inhibition of growth in E. nidulans cultures by the oxygen-depleting GOX,catalase system was likely to replace the primary inhibition exerted by H2O2. Conclusions:,Penicillium chrysogenum GOX possesses similar enzymological features to those described earlier for other Penicillium GOXs. Its cytotoxicity was dependent on the inherent antioxidant potential of the test micro-organisms. Significance and Impact of the Study:,Penicillium chrysogenum GOX may find future applications in glucose biosensor production, the disinfection of medical implants or in the food industry as an antimicrobial and/or preservative agent. Primary Aldosteronism: Diagnosis and Treatment

Recent studies have indicated a higher prevalence of primary aldosteronism (PA) than reported historically. Aldosterone excess induces sodium and fluid retention with consequential increases in blood pressure. Patients with PA are at an increased risk of developing left ventricular hypertrophy, chronic kidney disease, and endothelial dysfunction. Measurement of the plasma aldosterone/plasma renin activity ratio is an effective screening test for PA. The majority of patients with PA do not have a discernable aldosterone-producing adenoma (APA), and the aldosterone excess is considered idiopathic in etiology and/or attributed to adrenal hyperplasia. Treatment of PA includes medical therapy with mineralocorticoid receptor antagonists and adrenalectomy for patients with a unilateral APA. A reasonable treatment strategy is to attempt medical therapy in all patients with a high plasma aldosterone/PRA ratio and reserve the extensive workup needed to identify an APA for those patients whose hypertension or hypokalemia cannot be controlled medically. EFFECT OF ARTIFICIAL FEEDING ON DIGESTIVE EFFICIENCY, GROWTH AND QUALITIES OF MUSCLE AND OOCYTE OF MATURING ATLANTIC MACKEREL (SCOMBER SCOMBRUS L.)

ABSTRACT Maturing Atlantic mackerel with and without artificial feeding, kept in sea pens (September to May), showed differences in digestive efficiency (protease activity ratio of trypsin to chymotrypsin), muscle growth (concentrations of RNA, protein, RNA/protein ratio and free amino acids [FAA]) and oocyte quality (trypsin-like specific activity, and concentrations of RNA, RNA/protein ratio and FAA). The artificially fed mackerel had higher body weights (1.7 times) but with less white muscle protein concentration (0.5 time), compared to the control group. Both groups showed higher levels of capacity for protein synthesis in the oocytes than in the white muscle, but it was about two times higher in the artificially fed fish whereas about four times higher in the control group. This indicated that, during maturation, development of oocytes and muscle for growth occurred concurrently in higher growth mackerel, while development of oocytes dominated in slower growth fish. A higher trypsin-like specific activity with higher FAA levels in the oocytes from females fed with an artificial diet, compared to the control group, suggested differences in development and quality between the gametes of the fish with different feedings. PRACTICAL APPLICATIONS The work illustrates differences in digestive efficiency and the quality of growth performance (growth and protein metabolism in muscle and oocytes) in fish with different feedings. The use of various methods for evaluating digestive efficiency and the quality of fish growth performance could provide reasonable information for some important biological differences between fish groups, especially when the number of samples are low. It is more advantageous to apply different methods simultaneously than using growth parameter alone in order to study for precise evaluation of the quality of fish growth performance. The methods are very practical for studying food utilization and growth quality of fish in different environmental conditions and with different behaviors in aquaculture as well as in natural ecosystem where food consumption rate and feeding regime cannot be under control. Cyclopentadienyl technetium (99mTc) tricarbonyl piperidine conjugates: biodistribution and imaging studies

Abstract Synthesis of organometallic complexes of 99mTc using the precursor ligands N -methylpiperidino-4[(bispentahaptocyclopentadienyl)iron] carboxylate and N -(isopropyl)-piperidino-4[(bispentahaptocyclopentadienyl)iron]carboxylate is described. The labelling method involved reaction of the ligands with 99mTcO in the presence of Mn(CO)5Br in dimethyl formamide at 150°C for 1 h in an oil bath. The purification of the complexes was carried out by preparative TLC using ethery/n -butyl methyl amine (95:5) solvent system. The purified complexes were characterized by HPLC using acetonitrile:water (80:20) solvent system in a PRP-1 column in which both the complexes were eluted as single peaks. Biodistribution studies carried out in rats showed 2.4±0.14 and 1.1±0.42% of the injected activity in the brain tissue 5 min p.i. for cytectrene I and II, respectively. The brain to blood activity ratio was >15:1 for both the complexes at 5 min p.i. Scintigraphic studies in rabbits showed significant uptake of the activity in the brain with fast clearance from blood. The complexes warrant further investigation as agents for brain imaging. Abnormality of the Left Ventricular Sympathetic Nervous Function Assessed by I-123 Metaiodobenzylguanidine Imaging in Pediatric Patients with Neurocardiogenic Syncope

The purpose of this study was to assess the left ventricular sympathetic nervous system function in the patients with neurocardiogenic syncope (NCS) using I-123 metaiodobenzylguanidine (MIBG) imaging of the heart, and to compare the plasma noradrenaline (NA) and MIBG results of tilt positive and tilt negative patients following a head-up tilt test (HUT). The study included 30 patients. Their physical and laboratory examinations did not show a pathology that may be the cause of their syncope. HUT test was positive in 13 patients and negative in 17 patients. Plasma NA concentrations were higher in the HUT positive than the HUT negative group at the beginning and at the 10th minute of the test. Specific I-123 MIBG uptake assessed as the cardiac to mediastinal activity ratio in the delayed image was significantly higher in HUT positive group. The higher levels of MIBG uptake and plasma NA observed in HUT positive patients may reflect the greater capacity of NA storage in cardiac adrenergic neuronal tissue in patients with NCS. The results of this study support the critical role of autonomic nervous system in the pathophysiology of NCS and the excessive sympathetic nervous stimulation as the trigger of paradox reflex. Effects of cellulase and xylanase enzymes on the deconstruction of solids from pretreatment of poplar by leading technologies

Abstract Comparative data is presented on glucose and xylose release for enzymatic hydrolysis of solids produced by pretreatment of poplar wood by ammonia fiber expansion (AFEX), ammonia recycled percolation (ARP), controlled pH, dilute acid, flowthrough (FT), lime, and sulfur dioxide (SO2) technologies. Sugar solubilization was measured for times of up to 72 h using cellulase supplemented with ,-glucosidase at an activity ratio of 1:2, respectively, at combined protein mass loadings of 5.8,116 mg/g of glucan in poplar wood prior to pretreatment. In addition, the enzyme cocktail was augmented with up to 11.0 g of xylanase protein per gram of cellulase protein at combined cellulase and ,-glucosidase mass loadings of 14.5 and 29.0 mg protein (about 7.5 and 15 FPU, respectively)/g of original potential glucose to evaluate cellulase,xylanase interactions. All pretreated poplar solids required high protein loadings to realize good sugar yields via enzymatic hydrolysis, and performance tended to be better for low pH pretreatments by dilute sulfuric acid and sulfur dioxide, possibly due to higher xylose removal. Glucose release increased nearly linearly with residual xylose removal by enzymes for all pretreatments, xylanase leverage on glucan removal decreased at high cellulase loadings. Washing the solids improved digestion for all pretreatments and was particularly beneficial for controlled pH pretreatment. Furthermore, incubation of pretreated solids with BSA, Tween 20, or PEG6000 prior to adding enzymes enhanced yields, but the effectiveness of these additives varied with the type of pretreatment. © 2009 American Institute of Chemical Engineers Biotechnol. Field testing of equilibrium passive samplers to determine freely dissolved native polycyclic aromatic hydrocarbon concentrations

Abstract Equilibrium passive samplers are promising tools to determine freely dissolved aqueous concentrations (CW,free) of hydrophobic organic compounds. Their use in the field, however, remains a challenge. In the present study on native polycyclic aromatic hydrocarbons (PAHs) in Oslo Harbor, Norway, two different passive sampler materials, polyoxymethylene (POM; thickness, 55 ,m [POM-55] and 500 ,m [POM-500]) and polydimethylsiloxane (PDMS; thickness, 200 ,m), were used to determine in the laboratory CW,free in sediment pore water (CPW,free), and the suitability of five passive samplers for determination of CW,free in overlying surface water was tested under field conditions. For laboratory determinations of CPW,free, both POM-55 and PDMS turned out to be suitable. In the field, the shortest equilibrium times (approximately one month) were observed for POM-55 and PDMS (thickness, 28 ,m) coatings on solid-phase microextraction fibers, with PDMS tubing as a good alternative. Low-density polyethylene (thickness, 100 ,m) and POM-500 did not reach equilibrium within 119 d in the field. Realistic values were obtained for dissolved organic carbon,water partition coefficients in the field (approximately one log unit under log KOW), which strengthened the conclusion that equilibrium was established in field-exposed passive samplers. At all four stations, chemical activity ratios between pore water and overlying water were greater than one for all PAHs, indicating that the sediment was a PAH diffusion source and that sediment remediation may be an appropriate treatment for PAH contamination in Oslo Harbor. Hybrid reuteransucrase enzymes reveal regions important for glucosidic linkage specificity and the transglucosylation/hydrolysis ratio

The reuteransucrase enzymes of Lactobacillus reuteri strain 121 (GTFA) and L. reuteri strain ATCC 55730 (GTFO) convert sucrose into ,- d -glucans (labelled reuterans) with mainly ,-(1,4) glucosidic linkages (50% and 70%, respectively), plus ,-(1,6) linkages. In the present study, we report a detailed analysis of various hybrid GTFA/O enzymes, resulting in the identification of specific regions in the N-termini of the catalytic domains of these proteins as the main determinants of glucosidic linkage specificity. These regions were divided into three equal parts (A1,3; O1,3), and used to construct six additional GTFA/O hybrids. All hybrid enzymes were able to synthesize ,-glucans from sucrose, and oligosaccharides from sucrose plus maltose or isomaltose as acceptor substrates. Interestingly, not only the A2/O2 regions, with the three catalytic residues, affect glucosidic linkage specificity, but also the upstream A1/O1 regions make a strong contribution. Some GTFO derived hybrid/mutant enzymes displayed strongly increased transglucosylation/hydrolysis activity ratios. The reduced sucrose hydrolysis allowed the much improved conversion of sucrose into oligo- and polysaccharide products. Thus, the glucosidic linkage specificity and transglucosylation/hydrolysis ratios of reuteransucrase enzymes can be manipulated in a relatively simple manner. This engineering approach has yielded clear changes in oligosaccharide product profiles, as well as a range of novel reuteran products differing in ,-(1,4) and ,-(1,6) linkage ratios. [source]


Fast set-up of doxycycline-inducible protein expression in human cell lines with a single plasmid based on Epstein,Barr virus replication and the simple tetracycline repressor

We have developed a novel plasmid vector, pEBTetD, for full establishment of doxycycline-inducible protein expression by just a single transfection. pEBTetD contains an Epstein,Barr virus origin of replication for stable and efficient episomal propagation in human cell lines, a cassette for continuous expression of the simple tetracycline repressor, and a cytomegalovirus-type 2 tetracycline operator (tetO2)-tetO2 promoter. As there is no integration of vector into the genome, clonal isolation of transfected cells is not necessary. Cells are thus ready for use 1 week after transfection; this contrasts with 3,12 weeks for other systems. Adequate regulation of protein expression was accomplished by abrogation of mRNA polyadenylation. In northern analysis of seven cDNAs coding for transport proteins, pools of transfected human embryonic kidney 293 cells showed on/off mRNA ratios in the order of 100 : 1. Cell pools were also analyzed for regulation of protein function. With two transport proteins of the plasma membrane, the on/off activity ratios were 24 : 1 and 34 : 1, respectively. With enhanced green fluorescent protein, a 23 : 1 ratio was observed based on fluorescence intensity data from flow cytometry. The unique advantage of our system rests on the unmodified tetracycline repressor, which is less likely, by relocation upon binding of doxycycline, to cause cellular disturbances than chimera of tetracycline repressor and eukaryotic transactivation domains. Thus, in a comprehensive comparison of on- and off-states, a steady cellular background is provided. Finally, in contrast to a system based on Flp recombinase, the set-up of our system is inherently reliable. [source]


Relations of clinical features, subgroups and medication to serum monoamines in schizophrenia

Abstract Background Plasma and serum indices of monoaminergic activity reflect partly the illness of schizophrenia (e.g. HVA/deficit syndrome) and sometimes the symptoms (e.g. HVA/anhedonia). But, such studies have rarely taken both metabolites and parent amines or inter-amine activity ratios into account. We hypothesized that comparing the major symptom dimensions to measures of transmitter activity (with and without control for antipsychotic drug treatment) would show differential patterns of activity useful for the design of pharmacological treatments. Methods Dopamine (DA), noradrenaline (NA), serotonin (5-HT), their three major metabolites and prolactin were measured in the serum of 108 patients with schizophrenia and 63 matched controls: DA D2-receptor blocking-activity was estimated from a regression of butyrophenone displacement in striatum in vitro on to PET reports of drug-binding in vivo. Symptoms were factored into four dimensions (disorganized/thought disorder, nonparanoid/negative, ideas-of-reference and paranoid/positive symptoms). Results (1) Patients' DA activity did not differ from controls: but their 5-HT and NA turnovers increased/decreased, respectively, and the DA/5HT-metabolite ratio was lower. Increased DA-D2-receptor occupancy was predicted by decreased DA-metabolism and its ratio to 5-HT-metabolism. (2) Patients had higher levels of NA, DA-metabolites and DA-/5-HT-metabolite ratios on atypical vs typical drugs. (3) Increased D2-occupancy was associated with lower DA metabolism in paranoid patients but was unrelated to relative increases of DA/5-HT- and NA-metabolism in nonparanoid patients. (4) Low DA-/5-HT-metabolite ratios, high prolactin and low DA-metabolism characterized thought-disordered patients. (5) High DA-/5-HT-metabolite ratios paralleled many ideas-of-reference. The metabolites were sensitive, respectively, to control for D2-occupancy and prolactin. Conclusions The role of DA in paranoid, and 5-HT in thought-disordered and ideas-of-reference dimensions point both to the mechanisms underlying the features typical of these subgroups and the type of medication appropriate. Uranium and thorium isotopes in the rivers of the Amazonian basin: hydrology and weathering processes

Abstract Two expeditions (October 1989 and May 1992) were carried out to two points of the main Amazon River channel and four tributaries. The Solimões and Madeira rivers, taking their origin in the Andes, are whitewater rivers. The Negro River is a typical acid, blackwater river. The Trombetas River flows through bauxite-rich areas, and is characterized by low concentrations of dissolved humic substances. The 238U, 234U, 232Th and 230Th activities were recorded from dissolved, suspended particulate phases and river bank sediments. The latter were analysed for their 226Ra, 228Ra and 210Pb contents, and also subjected to leaching with 0·2 M hydroxylamine,hydrochloride solution to determine the concentrations of radionuclides bound to amorphous Fe hydroxides and Mn oxides and hydroxides. The dissolved U average concentration in the Amazon system is ten times lower than the mean world river concentration. The uranium concentration observed at Óbidos in the lower Amazon (0·095 µg L,1), where the U content in the river bank sediments and suspended matter is lowest, suggests U release from the solid phase during river transport. About 485 t of U are transported annually to the Amazon delta area in dissolved form, and 1943 t bound to suspended particulate matter. Total U and Th concentrations in the river bank sediments ranged from 1·59 to 7·14 µg g,1 and from 6·74 to 32 µg g,1, respectively. The highest concentrations were observed in the Trombetas River. The proportion extracted by means of the hydroxylamine solution (HL) was relatively high for U in the Trombetas river bank sediment (31%) and for Th in the Solimões sediment (30%). According to the alpha recoil effects, the 234U/238U activity ratios of the Andean river waters and downstream Amazon water (Óbidos) were >1, but were <1 in the Negro River (at Manaus). The activity ratios of dissolved U correlate with pH and also with the U activity ratios in the river bank sediment hydroxylamine extracts. As expected, the 234U/238U activity ratios in river bank sediments were <1 in the Andean rivers and in the downstream Amazon, but they were >1 in the Trombetas and Negro rivers. Such ratios probably result from the binding of dissolved uranium to solid sediment. The 228Th/232Th ratios of river bank sediments were close to unity (except for the Negro River, where it is lower), suggesting no significant Th exchanges between the river water and the sediment. The 226Ra/232Th activity ratios were <1, and the 226Ra/228Ra activity ratios generally were significantly higher than the activity ratios of their respective parents. This perhaps is the result of easier leaching of the 226Ra parent, 230Th, from solid material (owing to the alpha recoil effect) than of the 228Ra parent. Motor control of jaw muscles in chewing and in isometric biting with graded narrowing of jaw gape

Summary, When a certain bite force is applied during unilateral chewing, the combination of jaw elevator muscle activities is different than when a comparable force is applied in unilateral isometric biting, e.g. on a force transducer. Masticatory peak force is generated in a nearly isometric phase of the chewing cycle, with a jaw gape of about 1 mm. In contrast, peak force in isometric biting on force measuring equipment usually induces jaw gapes of 6 mm or even more. Therefore, we tested the hypothesis that the jaw gape influences relative activation of elevator muscles in unilateral isometric biting. We further examined whether such influence could explain the different activity combinations of chewing and isometric biting. In thirty asymptomatic males, masseter and temporalis activities were recorded during intermittent isometric biting with jaw gapes of 6, 5, 3, 2 and 1 mm and during unilateral chewing. Activity combinations were described by working/balancing ratios and by temporalis/masseter ratios. With decreasing jaw gape the working/balancing ratio of the posterior temporalis decreased (P < 0·002) while that of the masseter increased (P < 0·001). Likewise, the temporalis/masseter ratio on the balancing side increased (P < 0·001). With decreasing jaw gape, activity ratios of isometric biting approached ratios of chewing. We conclude that: (i) relative jaw muscle activation in isometric biting depends on the jaw gape, (ii) relative muscle activation in chewing resembles relative activation of isometric biting with a small ,chewing-like' gape. This suggests that characteristic activity combinations in chewing are mainly a result of the approximately isometric contraction during the slow closing phase of the chewing cycle. [source]
