Effects of soy vs. casein protein on body weight and glycemic control in female monkeys and their offspring

Abstract Nutritional interventions are important for reducing obesity and related conditions. Soy is a good source of protein and also contains isoflavones that may affect plasma lipids, body weight, and insulin action. Described here are data from a monkey breeding colony in which monkeys were initially fed a standard chow diet that is low fat with protein derived from soy. Monkeys were then randomized to a defined diet with a fat content similar to the typical American diet (TAD) containing either protein derived from soy (TAD soy) or casein,lactalbumin (TAD casein). The colony was followed for over two years to assess body weight, and carbohydrate and lipid measures in adult females (n=19) and their offspring (n=25). Serum isoflavone concentrations were higher with TAD soy than TAD casein, but not as high as when monkey chow was fed. Offspring consuming TAD soy had higher serum isoflavone concentrations than adults consuming TAD soy. Female monkeys consuming TAD soy had better glycemic control, as determined by fructosamine concentrations, but no differences in lipids or body weight compared with those consuming diets with TAD casein. Offspring born to dams consuming TAD soy had similar body weights at birth but over a two-year period weighed significantly less, had significantly lower triglyceride concentrations, and like adult females, had significantly lower fructosamine concentrations compared to TAD casein. Glucose tolerance tests in adult females were not significantly different with diet, but offspring eating TAD soy had increased glucose disappearance with overall lower glucose and insulin responses to the glucose challenge compared with TAD casein. Potential reasons for the additional benefits of TAD soy observed in offspring but not in adults may be related to higher serum isoflavone concentrations in offspring, presence of the diet differences throughout more of their lifespan (including gestation), or different tissue susceptibilities in younger animals. Ultrastructural Study of Encystation and Excystation in Acanthamoeba castellanii

Abstract. Encystation and excystation of Acanthamoeba castellanii were studied by transmission electron microscopy. The differentiation process was induced in asynchronous cultures grown axenically. Cytoplasmic vesicles containing a dense fibrous material very similar in appearance to the cyst wall were observed in trophozoites induced to encyst. When these trophozoites were incubated with calcofluor white m2r, fluorescence was observed in cytoplasmic vesicles, suggesting that the material contained in these vesicles corresponded to cyst wall precursors. Semithin cryosections of mature cysts with the same treatment showed fluorescence in the ectocyst and a less intense fluorescence in the endocyst, suggesting the presence of cellulose in both structures of the cyst wall. In mature cysts induced to excystation, small structures very similar to electron-dense granules (EDG) previously described in other amoebae were frequently observed. The EDGs were either sparsely distributed in the cytoplasm or associated with the cytoplasmic face of the plasma membrane. Many of them were located near the ostiole. In advanced phases of excystation, endocytic activity was suggested by the formation of endocytic structures and the presence of vacuoles with fibrous content similar to that of the cyst wall. Electron-dense granules in the process of dissolution were also observed in these vacuoles. Furthermore, the formation of a pseudopod suggests a displacement of the amoeba toward the ostiole. Strong decrease in lignin content without significant alteration of plant development is induced by simultaneous down-regulation of cinnamoyl CoA reductase (CCR) and cinnamyl alcohol dehydrogenase (CAD) in tobacco plants

Summary Different transgenic tobacco lines down-regulated for either one or two enzymes of the monolignol pathway were compared for their lignin content and composition, and developmental patterns. The comparison concerned CCR and CAD down-regulated lines (homozygous or heterozygous for the transgene) and the hybrids resulting from the crossing of transgenic lines individually altered for CCR or CAD activities. Surprisingly, the crosses containing only one allele of each antisense transgene, exhibit a dramatic reduction of lignin content similar to the CCR down-regulated parent but, in contrast to this transgenic line, display a normal phenotype and only slight alterations of the shape of the vessels. Qualitatively the lignin of the double transformant displays characteristics more like the wild type control than either of the other transgenics. In the transgenics with a low lignin content, the transformations induced other biochemical changes involving polysaccharides, phenolic components of the cell wall and also soluble phenolics. These results show that the ectopic expression of a specific transgene may have a different impact depending on the genetic background and suggest that the two transgenes present in the crosses may operate synergistically to reduce the lignin content. In addition, these data confirm that plants with a severe reduction in lignin content may undergo normal development at least in controlled conditions. [source]


Theory of Mind and Relational Complexity

Cognitive complexity and control theory and relational complexity theory attribute developmental changes in theory of mind (TOM) to complexity. In 3 studies, 3-, 4-, and 5-year-olds performed TOM tasks (false belief, appearance,reality), less complex connections (Level 1 perspective-taking) tasks, and transformations tasks (understanding the effects of location changes and colored filters) with content similar to TOM. There were also predictor tasks at binary-relational and ternary-relational complexity levels, with different content. Consistent with complexity theories: (a) connections and transformations were easier and mastered earlier than TOM; (b) predictor tasks accounted for more than 80% of age-related variance in TOM; and (c) ternary-relational items accounted for TOM variance, before and after controlling for age and binary-relational items. Prediction did not require hierarchically structured predictor tasks. [source]


Australasian microtektites and associated impact ejecta in the South China Sea and the Middle Pleistocene supereruption of Toba

Unmelted ejecta were found associated with the microtektites at this site and with Australasian microtektites in Core SO95,17957,2 and ODP Hole 1144A from the central and northern part of the South China Sea, respectively. A few opaque, irregular, rounded, partly melted particles containing highly fractured mineral inclusions (generally quartz and some K feldspar) and some partially melted mineral grains, in a glassy matrix were also found in the microtektite layer. The unmelted ejecta at all three sites include abundant white, opaque grains consisting of mixtures of quartz, coesite, and stishovite, and abundant rock fragments which also contain coesite and, rarely, stishovite. This is the first time that shock-metamorphosed rock fragments have been found in the Australasian microtektite layer. The rock fragments have major and trace element contents similar to the Australasian microtektites and tektites, except for higher volatile element contents. Assuming that the Australasian tektites and microtektites were formed from the same target material as the rock fragments, the parent material for the Australasian tektites and microtektites appears to have been a fine-grained sedimentary deposit. Hole 1144A has the highest abundance of microtektites (number/cm2) of any known Australasian microtektite-bearing site and may be closer to the source crater than any previously identified Australasian microtektite-bearing site. A source crater in the vicinity of 22° N and 104° E seems to explain geographic variations in abundance of both the microtektites and the unmelted ejecta the best; however, a region extending NW into southern China and SE into the Gulf of Tonkin explains the geographic variation in abundance of microtektites and unmelted ejecta almost as well. The size of the source crater is estimated to be 43 ± 9 km based on estimated thickness of the ejecta layer at each site and distance from the proposed source. A volcanic ash layer occurs just above the Australasian microtektite layer, which some authors suggest is from a supereruption of the Toba caldera complex. We estimate that deposition of the ash occurred ,800 ka ago and that it is spread over an area of at least 3.7 times 107 km2. [source]
