Kinds of Contamination Terms modified by Contamination Selected AbstractsDETERMINATION OF AFLATOXIN CONTAMINATION IN OLIVES BY IMMUNOAFFINITY COLUMN USING HIGH-PERFORMANCE LIQUID CHROMATOGRAPHYJOURNAL OF FOOD QUALITY, Issue 2 2006CAVIT BIRCAN ABSTRACT Eighty-two whole black olive samples gathered from six different olive oil processing facilities were surveyed to determine levels of aflatoxins using immunoaffinity column extraction and reversed-phase high-performance liquid chromatography. Two different analytical procedures adopted for the analysis of aflatoxins were investigated for their suitability by spiking the blank olive samples with five different known levels of aflatoxins to determine which one had higher recovery rates. Although some of the olive samples had been exposed to adverse conditions, such as rain and high temperatures, none were found to contain aflatoxins at the determined detection limit. Although the samples were kept in high relative humidity (75%) and high temperature (30C) for 3 months and were tested at 1-month intervals, no aflatoxins were detected. In addition, the olives were inoculated on a potato dextrose agar medium and incubated for 7 days at 25C to characterize the microflora. Because there is no evidence of aflatoxins in fresh whole olives, the next step of processing the contaminated olives into olive oils and testing them for the aflatoxins was not pursued. [source] CONTAMINATION OF GRAINS BY MYCOTOXIN-PRODUCING MOLDS AND MYCOTOXINS AND CONTROL BY GAMMA IRRADIATIONJOURNAL OF FOOD SAFETY, Issue 3 2006NAGY H. AZIZ ABSTRACT Ninety random grain samples were collected and analyzed for mycotoxins, and the effect of gamma irradiation on the production of mycotoxins in grains was studied. Aspergillus, Penicillium, Mucor, Rhizopus, Fusarium, Alternaria, Scopulariopsis and Cladosporium were the most common fungal genera isolated from grains. Aspergillus flavus, Aspergillus niger, Aspergillus candidus, Aspergillus ochraceus, Penicillium citrinum, Penicillium expansum, Penicillium citreonigrum, Penicillium purpurogenum, Penicillium griseofulvum and Penicillium verrucosumwere the most common Aspergillus and Penicillium species in grains. Out of 120 Aspergillus and Penicillium isolates, 80 were mycotoxin producers. Analysis of grains revealed the occurrence of aflatoxin B1 ochratoxin A, cycolopiazonic acid and citrinin. Of the 90 samples, 67 were positive for one or more mycotoxin. Irradiation of grains at dose of 2.0 and 4.0 kGy decreased significantly the total fungal counts compared with unirradiated controls. After 100 days of storage at room temperature, the unirradiated grains were contaminated with high concentrations of mycotoxins as compared with irradiated 4.0-kGy samples. Mycotoxin production in grains decreased with increasing irradiation doses and was not detected at 6.0 kGy over 100 days of storage. [source] FECAL COLIFORM CONTAMINATION OF BEEF CARCASSES DURING THE SLAUGHTERING PROCESSJOURNAL OF FOOD SAFETY, Issue 4 2001SUZAN YALÇIN ABSTRACT The purpose of this study was to determine the fecal coliform counts of beef carcasses during different stages in the slaughter process. A total of nine carcasses were selected at random in the abattoir. The samples were taken by excision from three different sites; rump, brisket and shoulder. The samples were collected from the same carcasses at four different stages of processing; after dressing, after evisceration, after washing and after chilling for 24 h in a chilling room. The processing steps did not increase the fecal coliform counts on the rump samples. There were no significant differences in the samples of rump and shoulder among different processing steps. The contamination level of the brisket after washing was significantly higher than other processing steps. Brisket and shoulder parts are critical points for microbiological sampling as these sites showed higher microbial counts after chilling steps. The data obtained have relevance for the planning of washing methods for the production of clean and safe carcasses. [source] DIRECT PLATING: A METHOD FOR DETECTING FUNGAL CONTAMINATION IN PAPERBOARD CARTONSJOURNAL OF FOOD SAFETY, Issue 3 2001JAN A. NARCISO ABSTRACT Contamination of refrigerated juice products in gable-top cartons can occur by filamentous fungi that are present in the paperboard. A method was developed to assay the mycoflora of paperboard carton material used in beverage packaging. This method involved direct plating on an agar surface of 1 cm2 carton pieces rather than disintegration of carton material in a blender prior to plating. When compared to the standard disintegration method traditionally used for monitoring contamination of paperboard, the new method is less cumbersome, more efficient, and reduces opportunities for contamination. The number of colonies produced by the direct plating method was greater than or equal to the modified standard disintegration method. Direct plating also resulted in a larger number of different genera isolated. [source] STATEWIDE EMPIRICAL MODELING OF BACTERIAL CONTAMINATION OF SURFACE WATERS,JOURNAL OF THE AMERICAN WATER RESOURCES ASSOCIATION, Issue 3 2006James D. Wickham ABSTRACT: Bacterial contamination of surface waters is attributed to both urban and agricultural land use practices and is one of the most frequently cited reasons for failure to meet standards established under the Clean Water Act (CWA) (P.L. 92,500). Statewide modeling can be used to determine if bacterial contamination occurs predominantly in urban or agricultural settings. Such information is useful for directing future monitoring and allocating resources for protection and restoration activities. Logistic regression was used to model the likelihood of bacterial contamination using watershed factors for the state of Maryland. Watershed factors included land cover, soils, topography, hydrography, locations of septic systems, and animal feeding operations. Results indicated that bacterial contamination occurred predominantly in urban settings. Likelihood of bacterial contamination was highest for small watersheds with well drained and erodible soils and a high proportion of urban land adjacent to streams. The number of septic systems and animal feeding operations and the amount of agricultural land were not significant explanatory factors. The urban infrastructure tends to "connect" more of the watershed to the stream network through the creation of roads, storm sewers, and wastewater treatment plants. This may partly explain the relationship between urbanization and bacterial contamination found in this study. [source] Contamination by nickel, copper and zinc during the handling of euro coinsCONTACT DERMATITIS, Issue 4 2003Paul-Guy Fournier The introduction of the euro has revived interest in the risk of nickel allergy due to the handling of coins. In the present work, the transfer of metallic contamination during the manipulation of coins is examined by means of leaching experiments and manipulation tests. It is shown that pre-existing metallic species present on the surface of the coins are the major source of contamination during manipulation, and that friction inherent to everyday usage contributes predominantly to their transfer to the hands. The comparison of coins as to their relative risks of metal contamination should therefore rely on tests that simulate the friction inherent in everyday human handling. Carrying out such tests with the newly issued 1, and 2, pieces, we find, contrary to long-term leaching measurements, that the euros release less nickel than previously circulated pure-nickel coins, but that this decrease is less pronounced than might have been hoped for on the basis of their surface composition. When the coins are rubbed to a shiny polish before manipulation, contamination of the fingers is reduced by more than a factor of 10. A comparison of coins used in France indicates that the introduction of the common currency has led to a fourfold reduction in contamination by nickel, while causing a 45% increase in contamination by copper. [source] A New Indirect Electroanalytical Method to Monitor the Contamination of Natural Waters with 4-Nitrophenol Using Multiwall Carbon NanotubesELECTROANALYSIS, Issue 9 2009Cruz Moraes, Fernando Abstract The electrochemical detection of the hazardous pollutant 4-nitrophenol (4-NP) at low potentials, in order to avoid matrix interferences, is an important research challenge. This study describes the development, electrochemical characterization and utilization of a multiwall carbon nanotube (MWCNT) film electrode for the quantitative determination of 4-NP in natural water. Electrochemical impedance spectroscopy measurements showed that the modified surface exhibits a decrease of ca. 13 times in the charge transfer resistance when compared with a bare glassy carbon (GC) surface. Voltammetric experiments showed the possibility to oxidize a hydroxylamine layer (produced by the electrochemical reduction of 4-NP on the GC/MWNCT surface) in a potential region which is approximately 700,mV less positive than that needed to oxidize 4-NP, thus minimizing the interference of matrix components. The limit of detection for 4-NP obtained using square-wave voltammetry (0.12,,mol L,1) was lower than the value advised by EPA. A natural water sample from a dam located in São Carlos (Brazil) was spiked with 4-NP and analyzed by the standard addition method using the GC/MWCNT electrode, without any further purification step. The recovery procedure yielded a value of 96.5% for such sample, thus confirming the suitability of the developed method to determine 4-NP in natural water samples. The electrochemical determination was compared with that obtained by HPLC with UV-vis detection. [source] In vivo genotoxic effects of industrial waste leachates in mice following oral exposureENVIRONMENTAL AND MOLECULAR MUTAGENESIS, Issue 5 2006Saurabh Chandra Abstract Contamination of ground water by industrial waste poses potential health hazards for man and his environment. The improper disposal of toxic wastes could allow genotoxic chemicals to percolate into ground waters, and these contaminated ground waters may produce toxicity, including mutation and eventually cancer, in exposed individuals. In the present study, we evaluated the in vivo genotoxic potential of leachates made from three different kinds of industrial waste (tannery waste, metal-based waste, and waste containing dyes and pigments) that are disposed of in areas adjoining human habitation. Three different doses of test leachates were administered by oral gavage for 15 consecutive days to Swiss albino mice; their bone marrow cells were examined for chromosome aberrations (CAs), micronucleated polychromatic erythrocytes (MNPCEs), and DNA damage using the alkaline Comet assay. Exposure to the leachates resulted in significant (P < 0.05 or P < 0.001) dose-dependent increases in chromosome and DNA damage. Fragmented chromosomes and chromatid breaks were the major CAs observed. Chemical analysis of the leachates indicated that chromium and nickel were elevated above the limits established by health organizations. The highest levels of genotoxicity were produced by the metal-based leachate and the tannery-waste leachate, while the dye-waste leachate produced weaker genotoxic responses. The cytogenetic abnormalities and DNA damage produced by the leachates indicate that humans consuming water contaminated with these materials are at increased risk of developing adverse health consequences. (Crustacea, Stomatopoda)ENVIRONMENTAL TOXICOLOGY, Issue 1 2008Antonio Dell'Anno Abstract Contamination of marine environments by hexachlorobenzene (HCB) represents a serious concern for potential consequences on ecosystem and human health. Despite this, information on cytotoxic effects on marine organisms is still largely lacking. In this study, we investigated cytotoxic effects induced by HCB on gonads and muscular tissue of Squilla mantis by analysing Na+/K+ -ATPase activity and plasma membrane fluidity. This crustacean species was selected as a model for its habitat, trophic level, feeding behavior, and commercial exploitation for human consumption. Time course experiments revealed that low concentrations of HCB (i.e. 50 nM) determine an exponentially decrease of Na+/K+ -ATPase activity and a significant modification of cellular membrane fluidity. Significant negative relationships between Na+/K+ -ATPase activity and membrane fluidity were observed, suggesting that changes in the structure and packing of cellular membranes induced by HCB may be the primary factor affecting the activity of essential bilayer-associated enzymes. Overall these findings suggest that even small concentrations of HCB may determine important changes on cell metabolism with potential cascade effects on recruitment of this commercial species. © 2008 Wiley Periodicals, Inc. Environ Toxicol, 2008. [source] Toxicity of oral exposure to 2,4,6-trinitrotoluene in the western fence lizard (Sceloporus occidentalis),ENVIRONMENTAL TOXICOLOGY & CHEMISTRY, Issue 5 2008Craig A. McFarland Abstract Contamination of the soil with the explosive 2,4,6-trinitrotoluene (TNT) has been found at military sites, many of which are habitats used by reptiles. To provide data useful in assessing ecological risk for reptilian species, acute, subacute, and subchronic oral toxicity studies were conducted with the western fence lizard (Sceloporus occidentalis). Oral median lethal dose (LD50) values for TNT in corn oil were 1,038 and 1,579 mg/kg of body weight for male and female lizards, respectively. Overt signs of toxicity included chromaturia, abdominal enlargement, and tremors. A 14-d subacute study followed in which male lizards were orally dosed with TNT (corn oil) at 0, 33, 66, 132, 263, 525, and 1,050 mg/kg of body weight each day. Clinical signs of toxicity, while similar to the LD50 study, were more subtle and noted in lizards receiving TNT amounts of at least 66 mg/kg/d. Chromaturia was an early consistent sign, often preceding the onset of adverse effects. Male lizards in the 60-d subchronic study were dosed at 0, 3, 15, 25, 35, and 45 mg/kg/d with nearly complete survival (>90%) for lizards in all treatments. Changes in food consumption and body weight were observed at 35 and 45 mg/kg/d. Alterations in hematological end points; blood chemistries (albumin, total protein, alkaline phosphatase, calcium); kidney, spleen, and liver weights; and adverse histopathology were observed in lizards exposed at 25 to 45 mg/kg/d. Testosterone concentration, sperm count, and motility were variable between treatments. Although not significant, incidences of hypospermia and testicular atrophy were observed in some individuals. Together, these data suggest a lowest-observed-adverse effect level of 25 mg/kg/d and a no-observed-adverse effect level of 15 mg/kg/d in S. occidentalis. [source] Polychlorinated naphthalenes and other dioxin-like compounds in Elbe River sedimentsENVIRONMENTAL TOXICOLOGY & CHEMISTRY, Issue 3 2008Werner Brack Abstract Contamination of Elbe River (Germany) sediments with dioxin-like toxicants was investigated following the 500-year flood (flood that statistically occurs once in 500 years) of 2002. It was hypothesized that large amounts of particulate matter from river beds and associated dioxin-like toxicants were mobilized and transported during this flood event. The investigation focused on polychlorinated naphthalenes (PCNs) that have not been determined previously in the Elbe River. The in vitro H4IIE- luc assay was used as an overall measure for toxicants capable of binding to the aryl hydrocarbon receptor (AhR). The assay was combined with congener-specific instrumental analyses and fractionation to quantify PCN contributions to total AhR-mediated activity relative to polychlorinated dibenzo- p -dioxins and dibenzofurans (PCDD/Fs) and polychlorinated biphenyls (PCBs). Penta- to octachloron-aphthalene concentrations of 30 ng/kg dry weight up to 13 ,g/kg dry weight were found in Elbe River sediments downstream of Bitterfeld. Concentrations of penta- to octachloronaphthalenes, however, were only approximately 3 ,g/kg dry weight at a site in the vicinity of Bitterfeld, where a level of approximately 3 mg/kg dry weight was reported before the flood. Also, the congener pattern of PCNs at this site changed after the flood, and PCN patterns reported previously for Bitterfeld and assigned to chlor-alkali electrolysis with graphite electrodes could now be observed at the sites from downstream of Bitterfeld and Magdeburg. Whereas PCDD/Fs dominated the dioxin-like activity in the middle and lower Elbe River, PCNs contributed as much as 10% of the total AhR-mediated activity. The contribution of PCBs was less significant (maximum, 0.2%). Thus, in Elbe River sediments, PCNs should be considered as relevant contaminants and be included in future monitoring and risk assessment programs. [source] Influence of contamination by organochlorine pesticides and polychlorinated biphenyls on the breeding of the Spanish Imperial Eagle (Aquila adalberti)ENVIRONMENTAL TOXICOLOGY & CHEMISTRY, Issue 2 2008Mauro Hernández Abstract We evaluated temporal and regional trends of organochlorine (OC) pesticide (including polychlorinated biphenyl [PCB]) levels in eggs of the Spanish Imperial Eagle (Aquila adalberti) collected in Spain between 1972 and 2003. Levels of p,p,-dichlorodiphenyldichloroethylene (DDE) and PCBs varied significantly (p = 0.022) among regions (central, western, and Doñana), being higher in Doñana than in the central and western populations (DDE: 1.64 ± 5.56, 0.816 ± 1.70, and 1.1 ± 2.66 ,g/g, respectively; PCBs: 1.189 ± 5.0, 0.517 ± 1.55, and 0.578 ± 1.75 ,g/g, respectively). Levels of DDE decreased with time, but a significant interaction was observed between region and time. In Doñana, egg volume and breadth as well as Ratcliffe Index were significantly lower after DDT use (p = 0.0018) than during the pre-DDT period (p = 0.0018); eggs were significantly smaller overall than in the other two regions (p = 0.04) and were smaller when DDE levels increased, even when controlling for regional differences (p = 0.04). Productivity in Doñana was significantly lower than in the other regions (p < 0.001). Clutch size in Doñana varied according to DDE concentrations (p = 0.01), with the highest DDE concentrations found in clutches consisting of one egg. When considering eggs with DDE levels greater than 3.5 ,g/g, a significant effect of DDE on fertility was found (p = 0.03). Clutches with DDE levels greater than 4.0 ,g/g had a higher probability of hatching failure (p = 0.07) and produced fewer fledglings (p = 0.03). If we consider 3.5 ,g/g as the lowest-observable-adverse-effect level, the proportion of sampled clutches that exceeded that level in Doñana (29%) was significantly higher than in other regions (p < 0.001). These eggs showed a mean percentage of thinning of 16.72%. Contamination by OCs, mainly DDE, could explain, at least in part, the low productivity of the Spanish Imperial Eagles in Doñana. [source] Sponges as biomonitors of heavy metals in spatial and temporal surveys in northwestern Mediterranean: Multispecies comparison,ENVIRONMENTAL TOXICOLOGY & CHEMISTRY, Issue 11 2007Emma Cebrian Abstract Contamination by heavy metals has increased drastically in the coastal Mediterranean during the last 20 years. A comparative study on metal bioaccumulation by four widespread sponge species (Crambe crambe, Chondrosia reniformis, Phorbas tenacior, and Dysidea avara) has been performed to select the most suitable species for metal monitoring. Copper bioaccumulation fits an accumulation strategy while Pb concentration seems to be regulated in most sponges. Crambe crambe was the only studied species that bioaccumulated Pb and Cu as a function of the available metal, proving its suitability for monitoring purposes. Then, we examined its effectiveness as a bioindicator at large spatial and temporal scales, comparing metal accumulation in this species and in sediments. Crambe crambe provided accurate information on the background levels of metals in the area at both spatial and temporal scales, and furthermore it reflected seasonal fluctuations of the bioavailable metals, which would be impossible to assess by means of a sediment survey. [source] Acute and chronic toxicity of five selective serotonin reuptake inhibitors in Ceriodaphnia dubiaENVIRONMENTAL TOXICOLOGY & CHEMISTRY, Issue 9 2004Theodore B. Henry Abstract Contamination of surface waters by pharmaceutical chemicals has raised concern among environmental scientists because of the potential for negative effects on aquatic organisms. Of particular importance are pharmaceutical compounds that affect the nervous or endocrine systems because effects on aquatic organisms are possible at low environmental concentrations. Selective serotonin reuptake inhibitors (SSRIs) are drugs used to treat clinical depression in humans, and have been detected in low concentrations in surface waters. In this investigation, the acute and chronic toxicity of five SSRIs (fluoxetine, Prozac®; fluvoxamine, Luvox®; paroxetine, Paxil®; citalopram, Celexa®; and sertraline, Zoloft®) were evaluated in the daphnid Ceriodaphnia dubia. For each SSRI, the 48-h median lethal concentration (LC50) was determined in three static tests with neonate C. dubia, and chronic (8-d) tests were conducted to determine no-observable-effect concentrations (NOEC) and lowest-observable-effect concentrations (LOEC) for reproduction endpoints. The 48-h LC50 for the SSRIs ranged from 0.12 to 3.90 mg/L and the order of toxicity of the compounds was (lowest to highest): Citalopram, fluvoxamine, paroxetine, fluoxetine, sertraline. Mortality data for the 8-d chronic tests were similar to the 48-h acute data. The SSRIs negatively affected C. dubia reproduction by reducing the number of neonates per female, and for some SSRIs, by reducing the number of broods per female. For sertraline, the most toxic SSRI, the LOEC for the number of neonates per female was 0.045 mg/L and the NOEC was 0.009 mg/L. Results indicate that SSRIs can impact survival and reproduction of C. dubia; however, only at concentrations that are considerably higher than those expected in the environment. [source] Contamination of fish in streams of the Mid-Atlantic Region: An approach to regional indicator selection and wildlife assessmentENVIRONMENTAL TOXICOLOGY & CHEMISTRY, Issue 3 2003James M. Lazorchak Abstract The extent of contamination of fish in the Mid-Atlantic Region was evaluated as part of the U.S. Environmental Protection Agency's Monitoring and Assessment Program's regional assessment in 1993 through 1994. Fish assemblages from wadeable streams were dominated by small, short-lived fishes (e.g., minnows, darters, and sculpins) that were more widely distributed and abundant than large fishes typically chosen for tissue contaminant studies (e.g., trout, black bass, sunfish, common carp). Chemical concentrations in whole-fish homogenates exceeded detection limits for mercury, DDT, and polychlorinated biphenyls (PCBs) in 75 to 100% of the stream length assessed using small fishes and 84 to 100% of the stream length assessed using large fishes. Wildlife values (WVs) representing a threshold for toxic effect were developed to allow examination of the spatial extent of potential risk to piscivorous wildlife. For mercury, DDT, dieldrin, and chlordane, estimates of the regional extent of streams where fish contaminant concentrations exceeded the WVs were greater when based on small fishes than on large fishes. However, within the distribution of stream lengths assessed using small and large fishes, the percentage of stream kilometers exceeding the WVs were quite similar. Our data demonstrate that the greater abundance and distribution of small, short-lived fishes provide greater estimates of regional extent of contamination for first- through third-order streams and can be used for regional assessments of potential exposure and effects in wildlife. [source] Sensors for the Detection and Quantification of Bacterial Contamination in Water for Human Use,ADVANCED ENGINEERING MATERIALS, Issue 5 2010Raquel Barbosa Queirós The deterioration of water quality by Cyanobacteria cause outbreaks and epidemics associated with harmful diseases in Humans and animals because of the toxins that they release. Microcystin-LR is one of the hepatotoxins most widely studied and the World Health Organization, recommend a maximum value of 1,µg,L,1 in drinking water. Highly specific recognition molecules, such as molecular imprinted polymers are developed to quantify microcystins in waters for human use and shown to be of great potential in the analysis of these kinds of samples. The obtained results were auspicious, the detection limit found, 1.5,µg,L,1, being of the same order of magnitude as the guideline limit recommended by the WHO. This technology is very promising because the sensors are stable and specific, and the technology is inexpensive and allows for rapid on-site monitoring. Contamination of food with viruses may also promote disease outbreaks. A number of studies have investigated the survival characteristics of several enteric viruses in various environments and foodstuffs, to help explain the transmissibility of these pathogens. This review deals with published work on enteric virus survival on fomites, and in waters, soil, and foods; the results of these studies have illustrated the robust survival of viruses in these environments. Much information is lacking, however, especially for foodstuffs and soils, and no detailed information is available concerning the survival of noroviruses, the most significant foodborne type. [source] Physiological and oenological traits of different Dekkera/Brettanomyces bruxellensis strains under wine-model conditionsFEMS YEAST RESEARCH, Issue 7 2008Ileana Vigentini Abstract Contamination of wine by Dekkera/Brettanomyces bruxellensis is mostly due to the production of off-flavours identified as vinyl- and especially ethyl-phenols, but these yeasts can also produce several other spoiling metabolites, such as acetic acid and biogenic amines. Little information is available about the correlation between growth, viability and off-flavour and biogenic amine production. In the present work, five strains of Dekkera bruxellensis isolated from wine were analysed over 3 months in wine-like environment for growth, cell survival, carbon source utilization and production of volatile phenols and biogenic amines. Our data indicate that the wine spoilage potential of D. bruxellensis is strain dependent, being strictly associated with the ability to grow under oenological conditions. 4-Ethyl-phenol and 4-ethyl-guaiacol production ranged between 0 and 2.7 and 2 mg L,1, respectively, depending on the growth conditions. Putrescine, cadaverine and spermidine were the biogenic amines found. [source] Monitoring Subsurface Contamination Using Tree BranchesGROUND WATER MONITORING & REMEDIATION, Issue 1 2007Gayathri Gopalakrishnan This paper proposes a method of assessing the distribution of chlorinated solvents in soil and ground water using tree branches. Sampling branches is a potentially more cost-effective and easier method than sampling tree cores, with less risk of damage to the tree. This approach was tested at Argonne National Laboratory, where phytoremediation is being used to remove tetrachloroethene (PCE), trichloroethene (TCE), and carbon tetrachloride (CCl4) from soil and ground water. The phytoremediation system consists of shallow-rooted willows planted in an area with contaminated soil and deep-rooted poplars planted in an area with clean soil and contaminated ground water. Branch samples were collected from 126 willows and 120 poplars. Contaminant concentrations from 31 soil borings and six monitoring wells were compared to those from branches of adjacent trees. Regression equations with correlation coefficients of at least 0.89 were obtained, which were found to be chemical specific. Kriged profiles of TCE concentration based on soil and willow branch data were developed and showed good agreement. Profiles based on ground water data could not be developed due to lack of sufficient monitoring wells for a meaningful statistical analysis. An analytical model was used to simulate TCE concentrations in tree branches from soil concentrations; the diffusion coefficient for TCE in the tree was used as the fitting parameter and the best-fit value was two orders of magnitude greater than literature values. This work indicates that tree branch sampling is a useful approach to assess contaminant distribution and potentially to determine where to locate monitoring wells or perform detailed soil analysis. Further research is necessary prior to using this method as a quantitative monitoring tool for soil and ground water. Kram Contamination from the use of chlorinated solvents, often classified as dense nonaqueous phase liquids (DNAPLs) when in an undissolved state, pose environmental threats to ground water resources worldwide. DNAPL site characterization method performance comparisons are presented in a companion paper (Kram et al. 2001). This study compares the costs for implementing various characterization approaches using synthetic unit model scenarios (UMSs), each with particular physical characteristics. Unit costs and assumptions related to labor, equipment, and consumables are applied to determine costs associated with each approach for various UMSs. In general, the direct-push sensor systems provide cost-effective characterization information in soils that are penetrable with relatively shallow (less than 10 to 15 m) water tables. For sites with impenetrable lithology using direct-push techniques, the Ribbon NAPL Sampler Flexible Liner Underground Technologies Everting (FLUTe) membrane appears to be the most cost-effective approach. For all scenarios studied, partitioning interwell tracer tests (PITTs) are the most expensive approach due to the extensive pre-and post-PITT requirements. However, the PITT is capable of providing useful additional information, such as approximate DNAPL saturation, which is not generally available from any of the other approaches included in this comparison. [source] A Decision-Making Framework for Sediment ContaminationINTEGRATED ENVIRONMENTAL ASSESSMENT AND MANAGEMENT, Issue 3 2005Peter M. Chapman Abstract A decision-making framework for determining whether or not contaminated sediments are polluted is described. This framework is intended to be sufficiently prescriptive to standardize the decision-making process but without using "cook book" assessments. It emphasizes 4 guidance "rules": (1) sediment chemistry data are only to be used alone for remediation decisions when the costs of further investigation outweigh the costs of remediation and there is agreement among all stakeholders to act; (2) remediation decisions are based primarily on biology; (3) lines of evidence (LOE), such as laboratory toxicity tests and models that contradict the results of properly conducted field surveys, are assumed incorrect; and (4) if the impacts of a remedial alternative will cause more environmental harm than good, then it should not be implemented. Sediments with contaminant concentrations below sediment quality guidelines (SQGs) that predict toxicity to less than 5% of sediment-dwelling infauna and that contain no quantifiable concentrations of substances capable of biomagnifying are excluded from further consideration, as are sediments that do not meet these criteria but have contaminant concentrations equal to or below reference concentrations. Biomagnification potential is initially addressed by conservative (worst case) modeling based on benthos and sediments and, subsequently, by additional food chain data and more realistic assumptions. Toxicity (acute and chronic) and alterations to resident communities are addressed by, respectively, laboratory studies and field observations. The integrative decision point for sediments is a weight of evidence (WOE) matrix combining up to 4 main LOE: chemistry, toxicity, community alteration, and biomagnification potential. Of 16 possible WOE scenarios, 6 result in definite decisions, and 10 require additional assessment. Typically, this framework will be applied to surficial sediments. The possibility that deeper sediments may be uncovered as a result of natural or other processes must also be investigated and may require similar assessment. CONCLUSIONS.,An environmental nursing diagnosis schema, with its emphasis on contamination, infection, and violence, provides nurses with a holistic framework for making judgments about environmental influences related to individual, family, community, and global health. The diagnoses of Contamination and Risk for Contamination provide necessary language to describe human responses and risk states that may arise following exposure to environmental contaminants. PRACTICE IMPLICATIONS.,Development of environmental diagnostic labels and delineation of the linkages to nursing outcomes and interventions will allow nurses to take active roles in identifying environmental components that affect health and planning care that responds to environmental health needs. [source] Transgenic weed beets: possible, probable, avoidable?JOURNAL OF APPLIED ECOLOGY, Issue 4 2002Benoît Desplanque Summary 1Weed beets pose a serious problem for sugar beet Beta vulgaris crops. Traditionally, the only efficient method of weed control has been manual removal, but the introduction of transgenic herbicide-tolerant sugar beets may provide an alternative solution because non-tolerant weed beets can be destroyed by herbicide. We evaluated the possibility that new, transgenic, weed beets may arise by gene flow between wild and crop plants. 2In a study area in northern France, weed beets were present in variable densities in sugar beet fields of up to 80 weed beet plants m,2. Weed beets arise from a long-lived seed bank, with seeds germinating from depths of 5 cm or less. In addition, diploid F1 crop,wild hybrids and triploid variety bolters (individuals with a low vernalization requirement) were present in low densities in virtually all sugar beet fields. We found gene flow to be possible between all forms, illustrated by both overlapping flowering periods in the field and successful controlled cross-pollinations. 3The F1 crop,wild hybrids result from pollination in the seed-production region by wild plants possessing the dominant bolting allele B for flowering without experiencing a period of cold. In the case of a transgene for herbicide tolerance incorporated into male-sterile seed-bearer plants, such hybrids will contain both the herbicide-tolerance and the bolting allele. Contamination of the fields by transgenic weed beets will be the result unless bolters are removed manually. The same will apply in the case of a cytoplasmically inherited transgene. 4Incorporation of the transgene into the pollinator plants will prevent the immediate formation of transgenic weed beets. However, in sugar beet fields, variety bolters may successfully cross-pollinate with weed beets in neighbouring fields. The use of diploid pollinator plants instead of tetraploids will considerably enhance gene flow towards wild beets, and is not, therefore, an attractive option. 5In conclusion, the appearance of transgenic weed beets is possible but can best be retarded if the transgene for herbicide tolerance is incorporated into the tetraploid pollinator breeding line. Dacarro Abstract Aims:, In order to assess possible occupational risk for workers in a grain mill, we evaluated aerial microbiological contamination in different areas of the mill and at different points of the production line. We also measured the concentration of aerodispersed dust particles. Methods and Results:, An assessment of microbiological contamination levels based on a Global Index of Microbial Contamination per cubic metre (GIMC per m3), an Index of Mesophilic Bacterial Contamination, and an Amplification Index is proposed. The indices were obtained from total and fungal counts. The cleaning sector is the most contaminated area of the mill: the mean GIMC per m3 was 17 213·6. In this area, the average microbial contamination was 11·41 times higher than that in the external environment. The highest concentrations of aerodispersed dust (inhalable 2·763 mg m,3; respirable 1·400 mg m,3) were found in the cleaning area. Conclusions:, The proposed microbiological indices and the concentrations of aerodispersed dust particles show that the most hazardous section of the mill is the cleaning area. The large variation in the data does not depend on seasonal factors, but rather on not easily identifiable conditions of the internal environment which facilitate diffusion and/or proliferation of the micro-organisms. Østensvik Abstract Aims:, To investigate the presence and numbers of Bacillus spp. spores in surface waters and examine isolates belonging to the B. cereus and B. subtilis groups for cytotoxicity, and to discuss the presence of cytotoxic Bacillus spp. in surface water as hazard identification in a risk assessment approach in the food industry. Methods and Results:, Samples from eight different rivers with variable degree of faecal pollution, and two drinking water sources, were heat shocked and examined for the presence of Bacillus spp. spores using membrane filtration followed by cultivation on bovine blood agar plates. Bacillus spp. was present in all samples. The numbers varied from 15 to 1400 CFU 100 ml,1. Pure cultures of 86 Bacillus spp. isolates representing all sampling sites were characterized using colony morphology, atmospheric requirements, spore and sporangium morphology, and API 50 CHB and API 20E. Bacillus spp. representing the B. cereus and B. subtilis groups were isolated from all samples. Twenty-one isolates belonging to the B. cereus and B. subtilis groups, representing eight samples, were screened for cytotoxicity. Nine strains of B. cereus and five strains belonging to the B. subtilis group were cytotoxic. Conclusions:, The presence of cytotoxic Bacillus spp. in surface water represents a possible source for food contamination. Filtration and chlorination of surface water, the most common drinking water treatment in Norway, do not remove Bacillus spores efficiently. This was confirmed by isolation of spores from tap water samples. Significance and Impact of the Study:, Contamination of food with water containing low numbers of Bacillus spores implies a risk for bacterial growth in foods. A method was developed to assay the mycoflora of paperboard carton material used in beverage packaging. This method involved direct plating on an agar surface of 1 cm2 carton pieces rather than disintegration of carton material in a blender prior to plating. When compared to the standard disintegration method traditionally used for monitoring contamination of paperboard, the new method is less cumbersome, more efficient, and reduces opportunities for contamination. The number of colonies produced by the direct plating method was greater than or equal to the modified standard disintegration method. Direct plating also resulted in a larger number of different genera isolated. Over Abstract:, Contamination of poultry with pathogenic bacteria contributes to human foodborne disease, causes damage to industry brand names, and has a significant economic impact on the food industry in the form of both damage to industry brand names and losses associated with recalls. Irradiation is a safe and effective means of decontaminating poultry products, but the maximum dose strengths allowed negatively impact poultry sensory quality characteristics. The 1st objective of this study was to investigate the potential interactive inhibitory effects of natural antimicrobials as components of a vacuum-marination in addition to various dose levels of irradiation. Tartaric acid (TA) at 2 levels and grape seed (GS) and green tea (GT) extracts were combined, vacuum-infused into chicken breast fillets, and irradiated at 1, 2, and 3 kGy by electron beam irradiation. The 2nd objective was to use a consumer test group to evaluate TA and plant extract infusion into chicken breast fillets with and without irradiation at 2 kGy on overall impression, flavor, texture, appearance, and tenderness. The results showed that samples vacuum-infused with TA at 37.5 and 75.0 mM and irradiated at 1 kGy significantly reduced Listeria monocytogenes (L.m.) levels by 2 and 3 log CFU/g compared to the control after 12 d of refrigerated storage. Vacuum-infusion of TA at 37.5 and 75.0 mM at 2 and 3 kGy irradiation, reduced L.m. to near nondetectable levels. The addition of TA and GS and GT to chicken breast fillets with and without irradiation did not significantly impact consumer preference, tenderness, appearance, or flavor. The addition of tartaric acid and natural plant extracts to chicken marinades could contribute to the prevention of L.m. contamination. [source] Detection of Fecal Contamination on Cantaloupes Using Hyperspectral Fluorescence ImageryJOURNAL OF FOOD SCIENCE, Issue 8 2005Angela M. Vargas ABSTRACT To determine whether detection of fecal contamination on cantaloupes is possible using fluorescence imaging, hyperspectral images of cantaloupes artificially contaminated with a range of diluted bovine feces were acquired from 425 to 774 nm in responses to ultraviolet-A (320 to 400 nm) excitation. Evaluation of images at emission peak wavelengths indicated that 675 nm exhibited the greatest contrast between feces contaminated and untreated surface areas. Two-band ratios compared with the single-band images enhanced the contrast between the feces contaminated spots and untreated cantaloupe surfaces. The 595/655-nm, 655/520-nm, and 555/655-nm ratio images provided relatively high detection rates ranging from 79% to 96% across all feces dilutions. However, both single band and ratio methods showed a number of false positives caused by such features as scarred tissues on cantaloupes. Principal component analysis (PCA) was performed using the entire hyperspectral images data; 2nd and 5th principal component (PC) image exhibited differential responses between feces spots and false positives. The combined use of the 2 PC images demonstrated the detection of feces spots (for example, minimum level of 16-,g/mL dry fecal matter) with minimal false positives. Based on the PC weighing coefficients, the dominant wavelengths were 465, 487, 531, 607, 643, and 688 nm. This research demonstrated the potential of multispectral-based fluorescence imaging for online applications for detection of fecal contamination on cantaloupes. [source] Identification of Biological Samples in a Case of Contamination of a Cytological Slide Preparation,JOURNAL OF FORENSIC SCIENCES, Issue 3 2008Anke Junge Ph.D. Abstract:, Here we report a case where a discrete section of the cytological slide preparation of a female individual was obviously contaminated with pleura liquid of a female tumor patient. Analysis of the cancerous pleura liquid and the healthy cells of the slide preparation showed different DNA profiles, indicating that the material originated from two different female individuals. The DNA profile of the cell mixture revealed a heterogenous pattern whereby the alleles could be assigned to the healthy and the tumor patient. Loss of heterozygosity (LOH) was observed in four of eight short tandem repeat systems for the pleura liquid and the cell mixture. Despite the low amount of DNA on the slide preparation and the occurrence of LOH, it was possible to clarify the case and to support the assumption that a drop of cancerous pleura liquid contaminated the cytological slide.
