A novel role for polyamines in adult neurogenesis in rodent brain

Abstract Although neurogenesis in the adult is known to be regulated by various internal cues such as hormones, growth factors and cell-adherence molecules, downstream elements underlying their action at the cellular level still remain unclear. We previously showed in an insect model that polyamines (putrescine, spermidine and spermine) play specific roles in adult brain neurogenesis. Here, we demonstrate their involvement in the regulation of secondary neurogenesis in the rodent brain. Using neurosphere assays, we show that putrescine addition stimulates neural progenitor proliferation. Furthermore, in vivo depletion of putrescine by specific and irreversible inhibition of ornithine decarboxylase, the first key enzyme of the polyamine synthesis pathway, induces a consistent decrease in neural progenitor cell proliferation in the two neurogenic areas, the dentate gyrus and the subventricular zone. The present study reveals common mechanisms underlying birth of new neurons in vertebrate and invertebrate species. [source]


Soil organic carbon contents in long-term experimental grassland plots in the UK (Palace Leas and Park Grass) have not changed consistently in recent decades

Abstract A recent report of widespread declines in soil organic C (SOC) in the UK over the 10,25 years until the early 2000s has focussed attention on the importance of resampling previously characterized sites to assess long-term trends in SOC contents and the importance of soils as a potentially volatile and globally significant reservoir of terrestrial C. We have used two sets of long-term experimental plots which have been under constant and known management for over a century and for which historical data exist that allow comparison over recent decades to determine what, if any, changes in SOC content have occurred. The plots used are the Palace Leas (PL) Meadow Hay Plots in north-east England (UK) established in 1897, and from the Park Grass (PG) Continuous Hay experiment established in 1856 at Rothamsted in south-east England. Collectively, these plots represent the only grassland sites in the UK under long-term management where changes in SOC over several decades can be assessed, and are probably unique in the world. The plots have received different manure and fertilizer treatment and have been under known management for at least 100 years. In 1982, total SOC contents were determined for the 0,27 cm layer of six of the PL plots using measurements of SOC concentrations, bulk density and soil depth. In 2006, the same six PL plots were resampled and SOC contents determined again. Four of the plots showed no net change in SOC content, but two plots showed net loss of SOC of 15% and 17% (amounting to decreases of 18 and 15 t C ha,1) since 1982. However, these differences in total SOC content were in a similar range to the variations in bulk density (6,31%) with changing soil water content. In 1959, the soil masses and SOC concentrations to 23 cm depth were measured on six PG plots with fertilizer and manure treatments corresponding closely with those measured on PL. In 2002, the SOC concentrations on the same plots were measured again. On three of the PG plots, SOC concentrations had declined by 2,10%, but in the other three it had increased by 4,8% between 1959 and 2002. If it is assumed that the soil bulk density had not changed over this period, the losses of SOC from the top soils ranged range from 10 to 3 t C ha,1, while the gains ranged from 4 to 7 t C ha,1. When the differences with time in SOC contents for the six PL and the six PG plots were examined using paired t -tests, that is, regarding the plots as two sets of six replicate permanent grasslands, there were no significant differences between 1982 and 2006 for the PL plots or between 1959 and 2002 for the PG plots. Thus, these independent observations on similar plots at PL and PG indicate there has been no consistent decrease in SOC stocks in surface soils under old, permanent grassland in England in recent decades, even though meteorological records for both sites indicate significant warming of the soil and air between 1980 and 2000. Because the potential influences of changes in management or land use have been definitively excluded, and measured rather than derived bulk densities have been used to convert from SOC concentrations to SOC amounts, our observations question whether for permanent grassland in England, losses in SOC in recent decades reported elsewhere can be attributed to widespread environmental change. Favorable outcome of ex-vivo purging of monocytes after the reintroduction of treatment after interruption in patients infected with multidrug resistant HIV-1,

Abstract In multidrug resistant patients treatment interruptions allow the selection of archived wild-type drug-susceptible viruses that compete for the less fit drug-resistant strains. However, the selection of viruses with increased replicative capacity is often followed by a loss of CD4+ T cells. In addition, drug resistant variants later re-emerge limiting the overall clinical benefit of treatment interruption. Blood monocytes are a key component of the HIV reservoir and can be partially removed by a system for purging of myeloid cells (MYP). This study tested the safety and efficacy of MYP on multidrug resistant patients who underwent treatment interruption. Twelve patients were randomized to receive or not six cycles of MYP during treatment interruption. An optimized antiretroviral regimen was reintroduced after the reappearance of a drug susceptible genotype. Following therapy reintroduction, a long lasting increase in CD4+ T cell counts was observed only in the treatment interruption,+,MYP patients but not in the control patients. Five/six treatment interruption,+,MYP patients never experienced virological rebound during a median follow up period of 98 weeks. In contrast, 4/6 patients who did not receive MYP never reached complete viral suppression and had a virological rebound after a median of 16.5 weeks after treatment reintroduction. The difference between the two groups in the time to virological rebound was statistically significant (P,=,0.021). A consistent decrease of HIV DNA load in CD14+ purified cells was observed only in treatment interruption,+,MYP patients. Human population genetic diversity as a function of SNP type from HapMap data

Data from the international HapMap project were mined to determine if the degree of genetic differentiation (Fst) is dependent on single nucleotide polymorphism (SNP) category. The Fst statistic was evaluated across all SNPs for each of 30 genes and for each of five chromosomes. A consistent decrease in diversity between Europeans and Africans was seen for nonsynonymous coding region SNPs compared to the three other SNP categories: synonymous SNPs, UTR, and intronic SNPs. This suggests an effect of balancing selection in reducing interpopulation genetic diversity at sites that would be expected to influence phenotype and therefore be subject to selection. Maleated amorphous ethylene propylene compatibilized polyethylene nanocomposites: Room temperature nonlinear creep response

Nonlinear creep of polyethylene and its nanocomposites remains an area of significant interest. Maleated polyethylene is often used as a compatibilizer to ensure enhanced dispersion. This article investigates blown films of linear low-density polyethylene and its nanocomposites with montmorillonite-layered silicate (MLS). An amorphous ethylene propylene copolymer grafted maleic anhydride (amEP) was added to enhance the interaction between the PE and the MLS. Tensile results indicate that the addition of amEP and MLS separately and together produces a synergistic effect on the mechanical properties of the neat PE. Nonlinear creep was analyzed by examining creep and recovery of the films with a Burger model and the Kohlrausch-Williams-Watts relation. A consistent decrease in unrecoverable plastic strain was obtained in the nanocomposite samples. A decreased retardation time associated with MLS presence was determined. POLYM. ENG. Trends in childhood leukemia mortality in Brazil and correlation with social inequalities

Abstract BACKGROUND. Mortality from childhood leukemia has declined substantially in developed countries but less markedly in the developing world. This study was designed to describe mortality trends in childhood leukemia and the impact of social inequalities on these trends in Brazil from 1980 to 2002. METHODS. Cancer mortality data by cause and estimates of resident population stratified by age and sex were obtained from the Brazilian Mortality Information System (SIM) for the years 1980 to 2002. Age-standardized (ages 0-19 years) mortality rates were calculated by the direct method using the 1960 world standard population. Trends were modeled using linear regression with 3-year moving average rates as the dependent variable and with the midpoint of the calendar year interval (1991) as the independent variable. The Index of Social Exclusion was used to classify the 27 Brazilian states. Pearson correlation was used to describe the correlation between social exclusion and variations in mortality in each state. RESULTS. Age-standardized mortality rates for boys decreased from 2.05 per 100,000 habitants in 1984 to 1.44 100,000 habitants in 1995, whereas the observed corresponding decline among girls was from 1.60 per 100,000 habitants in 1986 to 1.14 per 100,000 habitants in 1995. Statistically significant declining trends in mortality rates were observed for boys (adjusted correlation coefficient [r2] = 0.68; P < .001) and girls (adjusted r2 = 0.62; P < .001). Significant negative correlations between social inequality and changes in mortality were noted for boys (r = ,0.66; P = .001) and for girls (r = ,0.78; P < .001). CONCLUSIONS. A consistent decrease in mortality rates from childhood leukemia was noted in Brazil. Higher decreases in mortality were observed in more developed states, possibly reflecting better health care.
