Hypogonadism-related symptoms: development and evaluation of an empirically derived self-rating instrument (HRS ,Hypogonadism Related Symptom Scale')

Summary While self-report screening instruments are highly sensitive to hypogonadism in the ageing male, they have lacked specificity as evidenced by low or absent correlations with testosterone. The purpose of this paper was to develop an economical and specific screening instrument for identifying hypogonadal ageing men. Based on a comprehensive study of physical, somatoform and affective complaints, sexual behaviour and function and hormonal parameters of 263 outpatients aged 40 years and above (M = 56.2; 40,84 years) recruited from six andrological outpatient departments in Germany, we identified those items correlating significantly with testosterone. By factor analyses, five factors were identified: ,reduced activity', ,dissatisfaction with sexual function', ,negative self-concept of physical fitness', ,reduced sexual desire' and ,hot flushes'. The corresponding scales were reliable and only moderately inter-correlated. Consistent correlations were found with the level of testosterone, ageing male scales (Androgen Deficiency in the Aging Male, Aging Male Survey), specific affective, somatoform and sexual functioning scales and potential determinants of low testosterone (body mass index, physical inactivity, etc.). While further validation is needed, the new Hypogonadism Related Symptoms Scale appears to be a promising hypogonadism screening tool. [source]


Suspended sediment transport regime in a debris-flow gully on Vancouver Island, British Columbia

Abstract In debris-flow-prone channels, normal fluvial sediment transport occurs (nearly exclusively in suspended mode) between episodic debris-flow events. Observations of suspended sediment transport through a winter season in a steepland gully in logged terrain revealed two event types. When flows exceeded a threshold of 270 l s,1, events yielded significant quantities of sediment and suspended sediment concentration increased with flow. Smaller events were strongly ,supply limited'; sediment concentration decreased as flow increased. Overall, there is no consistent correlation between runoff and sediment yield. Within the season, three subseasons were identified (demarcated by periods of freezing weather) within which a pattern of fine sediment replenishment and evacuation occurred. Finally, a signature of fine sediment mobilization and exhaustion was observed within individual events. Fine sediment transport occurred in discrete pulses within storm periods, most of the yield occurring within 5 to 15% of storm runoff duration, so that it is unlikely that scheduled sampling programs would identify significant transport. Significant events are, however, generally forecastable on the basis of regional heavy rainfall warnings, providing a basis for targeted observations. Radiative snowmelt events and rain-on-snow remain difficult to forecast, since the projection of temperatures from the nearest regular weather station yields variable results. Are there correlates of male Australian Magpie Gymnorhina tibicen reproductive success in a population with high rates of extra-group paternity?

The reproductive success of a male bird is often correlated with measurable traits that predict his intrinsic quality. Females are thought to base their selection of mates on the latter's ,quality' in order to gain their ,good genes'. Male Australian Magpies Gymnorhina tibicen of the white-backed race tyrannica were trapped in two breeding seasons. Measurements were taken of morphometric and other characteristics in order to discover whether particular traits of males were associated with: (1) percentage of offspring sired in the territory, (2) number of fledglings produced in the territory per season and (3) whether females select males for their ,good genes'. There were no consistently significant correlations between any of the measured variables and male Magpie reproductive success within territories. In particular, none of the traits measured had any consistent correlation with the percentage of offspring sired in a territorial group. This was an unexpected result given that the species is strongly territorial but also engages regularly in extra-group copulations. These findings appear contrary to the predictions of the ,good genes' hypothesis. The general lack of correlation between the variables and level of genetic paternity may in fact be due to females engaging in extra-group mating primarily to avoid breeding with a close relative rather than to choose a high-quality male. In this case, males would not have to be ,high quality', but merely genetically different from the female's social mate. [source]


Pharmacological and Functional Characterization of Novel EP and DP Receptor Agonists: DP1 Receptor Mediates Penile Erection in Multiple Species

ABSTRACT Introduction., Despite the widespread use of prostaglandin E1 as an efficacious treatment for male erectile dysfunction for more than two decades, research on prostanoid function in penile physiology has been limited. Aim., To characterize the pharmacological and physiological activity of novel subtype-selective EP and DP receptor agonists. Methods., Radioligand binding and second messenger assays were used to define receptor subtype specificity of the EP and DP agonists. Functional activity was further characterized using isolated human and rabbit penile cavernosal tissue in organ baths. In vivo activity was assessed in rabbits and rats by measuring changes in cavernous pressure after intracavernosal injection of receptor agonists. Main Outcome Measures., Receptor binding and signal transduction, smooth muscle contractile activity, erectile function. Results., In organ bath preparations of human cavernosal tissue contracted with phenylephrine, EP2- and EP4-selective agonists exhibited variable potency in causing relaxation. One of the compounds caused mild contraction, and none of the compounds was as effective as PGE1 (EC50 = 0.23 µM). There was no consistent correlation between the pharmacological profile (receptor binding and second messenger assays) of the EP agonists and their effect on cavernosal tissue tone. In contrast, the DP1-selective agonist AS702224 (EC50 =29 nM) was more effective in relaxing human cavernosal tissue than either PGE1, PGD2 (EC50 = 58 nM), or the DP agonist BW245C (EC50 =59 nM). In rabbit cavernosal tissue, PGE1 and PGD2 caused only contraction, while AS702224 and BW245C caused relaxation. Intracavernosal administration of AS702224 and BW245C also caused penile tumescence in rabbits and rats. For each compound, the erectile response improved with increasing dose and was significantly higher than vehicle alone. Conclusions., These data suggest that AS702224 is a potent DP1-selective agonist that causes penile erection. The DP1 receptor mediates relaxation in human cavernosal tissue, and stimulates pro-erectile responses in rat and rabbit. Integrated genomic and expression profiling in mantle cell lymphoma: identification of gene-dosage regulated candidate genes

Summary Mantle cell lymphoma (MCL) is characterized by the t(11;14)(q13;q32) translocation and several other cytogenetic aberrations, including heterozygous loss of chromosomal arms 1p, 6q, 11q and 13q and/or gains of 3q and 8q. The common intervals of chromosomal imbalance have been narrowed down using array-comparative genomic hybridization (CGH). However, the chromosomal intervals still contain many genes potentially involved in MCL pathogeny. Combined analysis of tiling-resolution array-CGH with gene expression profiling on 11 MCL tumours enabled the identification of genomic alterations and their corresponding gene expression profiles. Only subsets of genes located within given cytogenetic anomaly-intervals showed a concomitant change in mRNA expression level. The genes that showed consistent correlation between DNA copy number and RNA expression levels are likely to be important in MCL pathology. Besides several ,anonymous genes', we also identified various fully annotated genes, whose gene products are involved in cyclic adenosine monophosphate-regulated pathways (PRKACB), DNA damage repair, maintenance of chromosome stability and prevention of rereplication (ATM, ERCC5, FBXO5), energy metabolism (such as genes that are involved in the synthesis of proteins encoded by the mitochondrial genome) and signal transduction (ARHGAP29). Deregulation of these gene products may interfere with the signalling pathways that are involved in MCL tumour development and maintenance. [source]


Position of nonmuscle myosin heavy chain IIA (NMMHC-IIA) mutations predicts the natural history of MYH9 -related disease,

Abstract MYH9 -related disease (MYH9 -RD) is a rare autosomal-dominant disorder caused by mutations in MYH9, the gene for the heavy chain of nonmuscle myosin IIA (NMMHC-IIA). All patients present from birth with macrothrombocytopenia, but in infancy or adult life, some of them develop sensorineural deafness, presenile cataracts, and/or progressive nephritis leading to end-stage renal failure. No consistent correlations have been identified between the 27 different MYH9 mutations identified so far and the variable clinical evolution of the disease. We have evaluated 108 consecutive MYH9 -RD patients belonging to 50 unrelated pedigrees. The risk of noncongenital manifestations associated with different genotypes was estimated over time by event-free survival analysis. We demonstrated that all subjects with mutations in the motor domain of NMMHC-IIA present with severe thrombocytopenia and develop nephritis and deafness before the age of 40 years, while those with mutations in the tail domain have a much lower risk of noncongenital complications and significantly higher platelet counts. We also evaluated the clinical course of patients with mutations in the four most frequently affected residues of NMMHC-IIA (responsible for 70% of MYH9 -RD cases). We concluded that mutations at residue 1933 do not induce kidney damage or cataracts and cause deafness only in the elderly, those in position 702 result in severe thrombocytopenia and produce nephritis and deafness at a juvenile age, while alterations at residue 1424 or 1841 result in intermediate clinical pictures. Cell resilience in species life spans: a link to inflammation?

Summary Species differences in life span have been attributed to cellular survival during various stressors, designated here as ,cell resilience'. In primary fibroblast cultures, cell resilience during exposure to free radicals, hypoglycemia, hyperthermia, and various toxins has shown generally consistent correlations with the species characteristic life spans of birds and mammals. However, the mechanistic links of cell resilience in fibroblast cultures to different species life spans are poorly understood. We propose that certain experimental stressors are relevant to somatic damage in vivo during inflammatory responses of innate immunity, particularly, resistance to reactive oxygen species (ROS), low glucose, and hyperthermia. According to this hypothesis, somatic cell resilience determines species differences in longevity during repeated infections and traumatic injuries in the natural environment. Infections and injury expose local fibroblasts and other cells to ROS generated by macrophages and to local temperature elevations. Systemically, acute phase immune reactions cause hypoglycemia and hyperthermia. We propose that cell resilience to somatic stressors incurred in inflammation is important in the evolution of longevity and that longer-lived species are specifically more resistant to immune-related stressors. This hypothesis further specifies Kirkwood's disposable soma theory. We suggest expanding the battery of stressors and markers used for comparative studies to additional cell types and additional parameters relevant to host defense and to their ecological specificities.
