The synthesis is concise and scalable, and the compounds are conjugation-ready. In addition, these compounds permeate zebrafish embryos; this suggests that they might be useful fluorescent probes for bioimaging. [source]

Abstract Synthesis of north -5'-methylbicyclo[3.1.0]hexyl purine and pyrimidine nucleosides with an ethynyl group at C-3' position has been successfully accomplished by a facile method. Methylbicyclo[3.1.0]hexanone (±)- 5 having three contiguous chiral centers was remarkably simply constructed only by four steps containing a carbenoid insertion reaction in the presence of rhodium(II) acetate dimer and CuSO4, giving a correct relative stereochemistry of the generated three chiral centers. Upon Grignard reaction of (±)- 5 with ethynylmagnesium bromide, exclusive diastereoselectivity was observed. Condensation of glycosyl donor (±)- 9 with purine nucleobase afforded only the desired N9 -alkylated nucleoside, while condensation with pyrimidine, N3 -benzoylated uracil gave the desired N1 -alkylated nucleoside (±)- 13 with the undesired O2 -alkylated nucleoside (±)- 14. Probably, (±)- 14 would be formed due to steric hindrance caused upon approaching for N1 -alkylation. [source]

Abstract A general stereoselective synthetic route to 5-substituted and 5,8-disubstituted indolizidine alkaloids has been developed starting from commercially available L -proline. (,)-Indolizidines 209D and 209B were efficiently synthesized in 9.8% and 14.8% overall yields in seven and five-step reactions from readily available aldehyde 3 and ketone 10, respectively. The key steps of this synthesis involve a substrate-induced asymmetric addition of ethyl lithiopropiolate to aldehyde 3 or methyl ketone 10, and a two-step one-pot hydrogenation/cyclization sequence to construct the bicyclic skeleton. Toward a Total Synthesis of Macrocyclic Jatrophane Diterpenes , Concise Route to a Highly Functionalized Cyclopentane Key Intermediate

A total synthesis of the biologically potent jatrophane diterpenes pepluanin A (1) and euphosalicin A (2) is being aimed at. En route to these targets, a concise synthesis of the nonracemic cyclopentane building block 74 was developed. Key steps were a Claisen,Eschenmoser rearrangement of the enantiomerically enriched allylic alcohol 14 to amide 34 (Scheme,7), a hydroxy-lactonization of 40 to 43 (Scheme,9), followed by trans -lactonization to 72, which was subjected to a Davis hydroxylation to 69 (Scheme,17). Eventually, compound 69 was converted into the enol triflate 74. This material should prove suitable for an annulation of the macrocyclic ring characteristic of the desired jatrophanes 1 and 2. Less-successful approaches are also discussed due to their intrinsically valuable information content. [source]

Abstract An efficient synthesis of N -sulfonyl-substituted 2-imino-3,4-dihydrocoumarins and 2-iminocoumarins via a copper-catalyzed multicomponent reaction of sulfonyl azides with terminal alkynes and ,-(ortho -hydroxyphenyl)-,,,-unsaturated ketones or ortho -hydroxyphenylpropiolates has been developed. The cascade process involves trapping the keteimine by a nucleophilic addition and an intramolecular Michael addition. This methodology could well be extended to the concise synthesis of the polysubstituted piperidine scaffold. [source]
