3D resistivity inversion using 2D measurements of the electric field

Field and ,noisy' synthetic measurements of electric-field components have been inverted into 3D resistivities by smoothness-constrained inversion. Values of electrical field can incorporate changes in polarity of the measured potential differences seen when 2D electrode arrays are used with heterogeneous ,geology', without utilizing negative apparent resistivities or singular geometrical factors. Using both the X - and Y -components of the electric field as measurements resulted in faster convergence of the smoothness-constrained inversion compared with using one component alone. Geological structure and resistivity were reconstructed as well as, or better than, comparable published examples based on traditional measurement types. A 2D electrode grid (20 × 10), incorporating 12 current-source electrodes, was used for both the practical and numerical experiments; this resulted in 366 measurements being made for each current-electrode configuration. Consequently, when using this array for practical field surveys, 366 measurements could be acquired simultaneously, making the upper limit on the speed of acquisition an order of magnitude faster than a comparable conventional pole,dipole survey. Other practical advantages accrue from the closely spaced potential dipoles being insensitive to common-mode noise (e.g. telluric) and only 7% of the electrodes (i.e. those used as current sources) being susceptible to recently reported electrode charge-up effects. Effects of 2-hydroxypropyl-,-cyclodextrin and cholesterol on porcine sperm viability and capacitation status following cold shock or incubation

Abstract Porcine sperm are extremely sensitive to the damaging effects of cold shock. It has been shown that cholesterol-binding molecules, such as 2-hydroxypropyl-,-cyclodextrin (HBCD), improve post-cooling porcine sperm viability when added to an egg yolk-based extender, but also enhance sperm capacitation in other species. The objective of this study was to determine the effects of HBCD and cholesterol 3-sulfate (ChS) on porcine sperm viability and capacitation following cold shock or incubation under conditions that support capacitation using a defined medium. We report here that porcine sperm incubated in medium containing both HBCD and ChS have significantly improved viability following cold shock (10 min at 10°C) when compared to sperm incubated without HBCD or ChS, or with either component alone. Treatment with HBCD plus ChS also completely inhibited the increase in protein tyrosine phosphorylation induced by the cold shock treatment or by incubation for 3 hr under conditions that support capacitation. Two assays of sperm capacitation, the rate of calcium ionophore-induced acrosome reactions and chlortetracycline (CTC) staining, were not significantly altered by HBCD and ChS following cold shock. However, 3-hr incubation with HBCD plus ChS or with 1 mM ChS alone decreased the percentage of sperm undergoing the induced acrosome reaction without significantly affecting viability when compared to the control. These results indicate that the manipulation of sperm plasma membrane cholesterol content affects porcine sperm viability and capacitation status and could therefore be useful to protect sperm from cold shock during cryopreservation by improving viability without promoting premature capacitation. Effects of PEGylation on the preservation of cationic lipid/DNA complexes during freeze-thawing and lyophilization

Abstract The incorporation of components with covalently attached polyethylene glycol (PEG) into nonviral vectors has been shown to prevent aggregation in serum and extend the circulating half-life of lipid/DNA complexes (lipoplexes) in vivo. The tendency of synthetic vectors to aggregate during processing and storage also represents a significant obstacle in the development of lipoplexes as marketable pharmaceutical products. The extreme instability of lipoplexes formulated as aqueous suspensions has generated interest in preserving nonviral vectors as frozen or lyophilized formulations. Previous work has demonstrated that stabilizing excipients are capable of protecting lipoplexes during freezing and lyophilization, but there is little known about the ability of PEGylation to protect vectors during these stresses. This study incorporates up to 10% by weight dioleoyl phosphatidylethanolamine conjugated to PEG-2000 and PEG-5000 into lipoplexes and assesses the maintenance of particle size and transfection after agitation, freeze-thawing, and lyophilization. Our results indicate that the incorporation of PEGylated components alone (up to 10% by weight) is insufficient to preserve particle size during these stresses. Complementary (secondary) metabolites in a soft coral: sex-specific variability, inter-clonal variability, and competition

Abstract Sex-specific interactions involving competition for space between the dioecious alcyonacean soft coral Sarcophyton glaucum and the scleractinian coral Acropora robusta were assessed experimentally on Bald Rock, central region of the Great Barrier Reef. To examine this, plus inter-clonal responses, one male colony of S. glaucum, known to produce sarcophytoxide as its predominant complementary (secondary) metabolite, was sectioned, producing 10 clones. The same was done for a female colony. These two sets of clones were then relocated to grids and placed in contact with Acropora clones. Relocated and non-relocated controls were also monitored. High levels of tissue necrosis were observed in the hard coral under contact conditions with both the male and female clones after 20 days. The development of a protective polysaccharide layer in the alcyonacean was also observed. Differences observed in the concentrations of complementary metabolites within the two different S. glaucum colonies were related to sex. Both under competition and non-competition conditions, females exhibited significantly higher concentrations of sarcophytoxide than males, and this increased with time. Fatty ester concentration was also higher in females than males, varying significantly through time, and falling dramatically just after spawning. Fatty ester concentrations decreased linearly through time in the male clones. When involved in competition for space, females possessed higher concentrations of fatty esters than males, both at the site of contact and in non-contact sites, again, decreasing after spawning. No significant changes in sarcophytoxide levels were noted in the parental colonies, but such changes were observed in fatty esters, with the female producing higher concentrations until after spawning. The use of these two variates in the form of a ratio (sarcophytoxide concentration:fatty ester concentration) yielded a variable Rho (,) which was a more sensitive indicator of biochemical change than either of its components alone. These two sets of compounds appeared to have a negative association through time and varied highly significantly between sexes. The diterpene sarcophytoxide may be considered an allelopathic or stress metabolite, while the lipids act as energy storage metabolites. [source]


Synergistic Effect of Porcine Follicular Fluid and Dibutyryl Cyclic Adenosine Monophosphate on Development of Parthenogenetically Activated Oocytes from Pre-Pubertal Gilts

Contents This study investigated the effect of porcine follicular fluid (PFF) and dibutyryl cyclic adenosine monophosphate (dbcAMP) during in vitro maturation (IVM) of porcine oocytes on meiotic maturation, fertilization and embryo development, and compared the effect of supplementing the embryo culture media with PFF or foetal bovine serum (FBS) on embryo development. Oocytes from pre-pubertal gilts were IVM for 44 h, and parthenogenetically activated or in vitro -fertilized. Embryos were cultured in porcine zygote medium (PZM3) for 7 days. Cleavage and blastocyst rates were evaluated at 48 h and 7 days of culture. The supplementation of the IVM medium with 25% PFF and 1 mm dbcAMP for the first 22 h resulted in more (p < 0.05) embryos developing to the blastocyst stage as compared with the inclusion of dbcAMP alone. The dbcAMP + PFF combination increased (p < 0.05) the average number of nuclei per blastocyst as compared with either of these components alone or in its absence. A synergistic effect of dbcAMP + PFF during IVM was also reflected in the capacity of oocytes to regulate sperm penetration and prevent polyspermy, as twice as many oocytes from the control group were penetrated by more than one sperm as compared with those matured in the presence of both dbcAMP and PFF. The supplementation of PZM3 with 10% FBS from days 5 to 7 of culture significantly improved the total cell quantity in embryos derived either from control or dbcAMP + PFF matured oocytes. There was no effect on the total cell quantity when FBS was replaced by the same concentration of PFF. These studies showed that dbcAMP, PFF and FBS can improve both the quantity (57.3% vs 41.5%) and quality (74.8 vs 33.3 nuclei) of porcine blastocysts derived from oocytes recovered of pre-pubertal gilts. Acoustic matrix microseeding: improving protein crystal growth with minimal chemical bias

A crystal seeding technique is introduced that uses acoustic waves to deliver nanolitre volumes of seed suspension into protein drops. The reduction in delivery volume enables enhanced crystal growth in matrix-seeding experiments without concern for bias from chemical components in the seed-carrying buffer suspension. Using this technique, it was found that while buffer components alone without seed can marginally promote crystal growth in some cases, crystal seeding is far more effective in boosting the number of sparse-matrix conditions that yield protein crystals. Comparison of ethanol and methanol crossover through different MEA components and structures by cyclic voltammetry

Abstract The crossover rate of ethanol or methanol through membrane electrode assembly (MEA) and MEA components was studied quantitatively at 25 and 60 °C by cyclic voltammetry method. The results obtained in this work show that cyclic voltammetry is a powerful technique to assess the crossover phenomenon through MEA components and structures. In all cases, the ethanol crossover rates are lower than those of methanol. The ethanol and methanol crossover rates depend upon time and temperature. For an initial concentration of 1 M of ethanol or methanol, the crossover rate increases to a maximum after the first hour of the cell operation and then decreases gradually to a certain concentration after the third hour. At 60 °C, the maximum concentration of ethanol crossover rate is lower than that obtained at 25 °C. The crossover rate of ethanol or methanol through MEA is lower than through the components alone (pressed or nonpressed membranes or gas diffusion electrode).
