The European Union's Counter-Terrorism Strategy, its Problems
On the basis of an analysis of the European Union's common definition of the post-9/11 terrorist threat, this article provides a critical assessment of the EU's response. The EU has arrived at a reasonably specific definition of the common threat that avoids simplistic reductions and is a response that is sufficiently multidimensional to address the different aspects , internal and external, legislative and operational, repressive and preventive , of this threat. Yet the definition is undermined by differences between national threat perceptions. The preference for instruments of cooperation and coordination rather than integration, and poor implementation are having a negative impact on the effectiveness of the common response, the legitimacy of which is also weakened by limited parliamentary and judicial control. Interaction between macroinvertebrates, discharge and physical habitat in upland rivers

Abstract 1.There is a need to relate changing river flows to ecological response, particularly using methods which do not require extensive new data for water bodies without historical data. This paper describes how local-scale habitat features and changing discharge appear to influence a macroinvertebrate-based biotic index. 2.The study used 87 time-series of river biomonitoring data from upland, wadeable streams with quasi-natural flow regimes across England and Wales. Twenty-seven of the sites were matched to a nearby flow gauging station, and historical, natural flows using a generalized rainfall-runoff model were synthesized for 60 sites. All sites were matched to a River Habitat Survey (RHS) within 1,km. 3.The data were analysed using multilevel linear regression, combining sample- and site-level characteristics as predictors. Common responses were assessed across sites, using the biotic index LIFE (Lotic Invertebrate index for Flow Evaluation), an average of abundance-weighted Flow Groups which indicate the relative preference among taxa for higher velocities with gravel/cobble substrates or slow velocities with finer substrates. The aim was to understand the influence of physical habitat on macroinvertebrate response to antecedent high and low flow magnitude. 4.There was a positive relationship between LIFE score calculated from spring and autumn samples and antecedent high (Q10) and low flows (Q95). The relationship between summer Q10 and autumn LIFE score was steeper than the relationship between winter Q10 and spring LIFE score. Bed and bank resectioning reduced overall LIFE and increased the steepness of the response of LIFE to low (Q95) flow. 5.The models derived may be used to guide environmental flow allocations and to quantify the relative influence of flow and physical habitat change on macroinvertebrate responses. The interaction between resectioning and low flow has particular implications for the conservation of macroinvertebrate taxa with requirements for faster flowing water. Abiotic stress and plant responses from the whole vine to the genes

Abstract Drought, salinity and extreme temperatures significantly limit the distribution of grapes around the world. In this review, the literature of grape responses to abiotic stress with particular reference to whole plant and molecular responses observed in recent studies is discussed. A number of short-term and long-term studies on grapevine shoots and berries have been conducted using a systems biology approach. Transcripts, proteins and metabolites were profiled. Water deficit, salinity and chilling altered the steady-state abundance of a large number of transcripts. Common responses to these stresses included changes in hormone metabolism, particularly abscisic acid (ABA), photosynthesis, growth, transcription, protein synthesis, signalling and cellular defences. Some of the transcriptional changes induced by stress were confirmed by proteomic and metabolomic analyses. More than 2000 genes were identified whose transcript abundance was altered by both water deficit and ABA. Different gene sets were used to map molecular pathways regulated by ABA, water deficit, salinity and chilling in grapevine. This work supports the hypothesis that ABA is a central regulator of abiotic stress tolerance mechanisms. ABA affects signalling pathways that trigger important molecular activities involving metabolism, transcription, protein synthesis, and cellular defence and also regulates important physiological responses such as stomatal conductance, photoprotection and growth. Systems biology approaches are providing more comprehensive understanding of the complex plant responses to abiotic stress. The molecular sets generated from mapping the ABA-inducible stress responses provide numerous targets for genetic and cultural manipulation for improved plant protection and grape quality. [source]


Punishment as restoration of group and offender values following a transgression: value consensus through symbolic labelling and offender reform

Justice theory has suggested that transgressions pose a threat to the shared values that underlie broken rules or laws, suggesting that in order to address concerns over the values violated by an offence, perceived consensus regarding those values must be reaffirmed. However, little empirical research has been conducted examining how legal responses can address those value concerns. In the current research we argue that punishments, as a common response to injustice, can reaffirm perceived value consensus through two routes: (1) by symbolically labelling the offence as against group values, thus reinforcing values towards observers and (2) by attempting to reform the offender, thus reinforcing values towards the offender. Consistent with this argument, three empirical studies showed that the public and inclusive nature of punishment helps restore a perceived value consensus as such characteristics facilitate these two processes. Moreover, these characteristics had a positive effect on perceived punishment appropriateness particularly when value concerns were heightened. These findings implicate symbolic labelling and offender reform as two processes by which punishments can restore the perception of value consensus and suggest that these processes are integral to justice restoration through punishment when value consensus is a dominant concern. RESEARCH ARTICLE: Fungicidal activity of amiodarone is tightly coupled to calcium influx

Abstract The antiarrhythmic drug amiodarone has microbicidal activity against fungi, bacteria and protozoa. In Saccharomyces cerevisiae, amiodarone triggers an immediate burst of cytosolic Ca2+, followed by cell death markers. Ca2+ transients are a common response to many forms of environmental insults and toxic compounds, including osmotic and pH shock, endoplasmic reticulum stress, and high levels of mating pheromone. Downstream signaling events involving calmodulin, calcineurin and the transcription factor Crz1 are critical in mediating cell survival in response to stress. In this study we asked whether amiodarone induced Ca2+ influx was beneficial, toxic or a bystander effect unrelated to the fungicidal effect of the drug. We show that downregulation of Ca2+ channel activity in stationary phase cells correlates with increased resistance to amiodarone. In actively growing cells, extracellular Ca2+ modulated the size and shape of the Ca2+ transient and directly influenced amiodarone toxicity. Paradoxically, protection was achieved both by removal of external Ca2+ or by adding high levels of CaCl2 (10 mM) to block the drug induced Ca2+ burst. Our results support a model in which the fungicidal activity of amiodarone is mediated by Ca2+ stress, and highlight the pathway of Ca2+ mediated cell death as a promising target for antifungal drug development. A past and a future for diversification on farms?

Some evidence from large-scale, commercial farms in South East England
Abstract Diversification has been identified as a common response to the agricultural crisis of the 1980s and to the changing ethos of agricultural policy in the closing decade of the twentieth century. In particular, farmers operating large-scale farms have been prominent in adopting this approach, just as they were innovative across a range of farming practices in the expansion and modernisation of their agricultural production in earlier decades. Can we identify serial diversifiers within this sector of the farming community, who are disposed to react in an entrepreneurial fashion to the changing fortunes of agriculture? The paper draws on results from a survey of large-scale commercial farmers in South East England and, by examining the sequence in which various forms of diversification were adopted, identifies a temporal pattern as farmers responded to the fluctuating fortunes of the agricultural industry over the past thirty years. But has the potential for diversification been exhausted? The paper also considers future prospects for diversification within the large-scale, commercially oriented sector of the agricultural industry. [source]

Surgical margin determination in head and neck oncology: Current clinical practice.

Abstract Background. Our aim was to investigate the ways in which surgeons who perform head and neck ablative procedures on a regular basis define margins, how they use frozen sections to evaluate margins, and the effect of chemoradiation on determining tumor margins. Methods. A custom-designed questionnaire was mailed to members of the American Head and Neck Society asking members how they evaluate and define tumor margins. Results. Of 1500 surveys mailed, 476 completed surveys were received. The most common response for distance of a clear pathologic margin was >5 mm on microscopic evaluation. A margin containing carcinoma in situ was considered a positive margin by most, but most did not consider a margin containing dysplasia a positive margin. When initial frozen section margins are positive for tumor and further resection results in negative frozen section margins, 90% consider the patient's margin negative. Most surgeons sample the frozen section from the surgical bed rather than from the main specimen. Nearly half use wider margins when resecting tumors treated with neoadjuvant therapy. When resecting recurrent or residual tumors treated with previous chemoradiation therapy, most resect to the pretreatment margin. Conclusions. No uniform criteria to define a clear surgical margin exist among practicing head and neck surgeons. Most head and neck surgeons consider margins clear if resection completed after an initial positive frozen section margin reveals negative margins, but this view is not shared by all. Most surgeons take frozen sections from the surgical bed; however, error may occur when identifying the positive margin within the surgical bed. The definition of a clear tumor margin after chemoradiation is unclear. These questions could be addressed in a multicenter prospective trial. © 2005 Wiley Periodicals, Inc. Gliotoxin non-selectively induces apoptosis in fibrotic and normal livers

Abstract: Background: Liver fibrosis is the common response to chronic liver injury, ultimately leading to cirrhosis. Several lines of evidence indicate that inducing apoptosis of hepatic stellate cells (HSC) may lead to regression of liver fibrosis. Recently, it was shown that gliotoxin (GTX) induces apoptosis of HSC. However, the clinical use of GTX may be limited because of the lack of cell and tissue specificity, causing a high risk of potentially severe adverse effects. The aim of this study, therefore, was to study the effect of GTX on different cells of the liver. Methods: We used normal and fibrotic precision-cut rat liver slices to study the effect of GTX on the various resident liver cell types. In these slices, the complex cell,cell interactions are preserved, which closely mimics the in vivo situation. Results: GTX exhibited a potent apoptosis-inducing activity in these slices. Both immunohistochemical stainings and real-time mRNA techniques showed that this apoptosis-inducing effect was seen in HSC. However, Kupffer cells and liver endothelial cells were also affected by GTX, whereas hepatocytes were only mildly affected. Conclusions: This study indicates that the apoptosis-inducing strategy to treat liver fibrosis has high potential, but it will be necessary to develop an HSC-specific therapy to prevent adverse effects. Rangewide population genetic structure of the African malaria vector Anopheles funestus

Abstract Anopheles funestus is a primary vector of malaria in Africa south of the Sahara. We assessed its rangewide population genetic structure based on samples from 11 countries, using 10 physically mapped microsatellite loci, two per autosome arm and the X (N = 548), and 834 bp of the mitochondrial ND5 gene (N = 470). On the basis of microsatellite allele frequencies, we found three subdivisions: eastern (coastal Tanzania, Malawi, Mozambique and Madagascar), western (Burkina Faso, Mali, Nigeria and western Kenya), and central (Gabon, coastal Angola). A. funestus from the southwest of Uganda had affinities to all three subdivisions. Mitochondrial DNA (mtDNA) corroborated this structure, although mtDNA gene trees showed less resolution. The eastern subdivision had significantly lower diversity, similar to the pattern found in the codistributed malaria vector Anopheles gambiae. This suggests that both species have responded to common geographic and/or climatic constraints. The western division showed signatures of population expansion encompassing Kenya west of the Rift Valley through Burkina Faso and Mali. This pattern also bears similarity to A. gambiae, and may reflect a common response to expanding human populations following the development of agriculture. Due to the presumed recent population expansion, the correlation between genetic and geographic distance was weak. Mitochondrial DNA revealed further cryptic subdivision in A. funestus, not detected in the nuclear genome. Mozambique and Madagascar samples contained two mtDNA lineages, designated clade I and clade II, that were separated by two fixed differences and an average of 2% divergence, which implies that they have evolved independently for ,1 million years. Clade I was found in all 11 locations, whereas clade II was sampled only on Madagascar and Mozambique. We suggest that the latter clade may represent mtDNA capture by A. funestus, resulting from historical gene flow either among previously isolated and divergent populations or with a related species. Abstract Increasing water scarcity is increasing pressure on water management institutions, particularly in the area of water rights. A common response is to formalise water tenure, one of several options for securing access and resolving conflicts concerning water allocation. This article looks at four contexts where negotiation, self-governance and concepts of legal pluralism may help improve water resource management. Existing users and potential new users need to negotiate before water resources are developed. Users can participate in forums with authority to solve basin management problems through self-governance. Negotiated water transfers offer an alternative to water acquisition by expropriation. Oxidative gating of water channels (aquaporins) in Chara by hydroxyl radicals

ABSTRACT Hydroxyl radicals (*OH) as produced in the Fenton reaction (Fe2+ + H2O2 = Fe3+ + OH, + *OH) have been used to reversibly inhibit aquaporins in the plasma membrane of internodes of Chara corallina. Compared to conventional agents such as HgCl2, *OH proved to be more effective in blocking water channels and was less toxic to the cell. When internodes were treated for 30 min, cell hydraulic conductivity (Lp) decreased by 90% or even more. This effect was reversed within a few minutes after removing the radicals from the medium. In contrast to HgCl2, radical treatment reduced membrane permeability of small lipophilic organic solutes (ethanol, acetone, 1-propanol, and 2-propanol) by only 24 to 52%, indicating some continued limited movement of these solutes across aquaporins. The biggest effect of *OH treatment on solute permeability was found for isotopic water (HDO), which largely used water channels to cross the membrane. Inhibition of aquaporins reduced the diffusional water permeability (Pd) by about 70%. For the organic test solutes, which mainly use the bilayer to cross the membrane, channel closure caused anomalous (negative) osmosis; that is, cells had negative reflection coefficients (,s) and were transiently swelling in a hypertonic medium. From the ratio of bulk (Lp or osmotic permeability coefficient, Pf) to diffusional (Pd) permeability of water, the number (N) of water molecules that align in water channels was estimated to be N = Pf/Pd = 46 (on average). Radical treatment decreased N from 46 to 11, a value still larger than unity, which would be expected for a membrane lacking pores. The gating of aquaporins by *OH radicals is discussed in terms of a direct action of the radicals when passing the pores or by an indirect action via the bilayer. The rapid recovery of inhibited channels may indicate an easy access of cytoplasmic antioxidants to closed water channels. As hydrogen peroxide is a major signalling substance during different biotic and abiotic stresses, the reversible closure of water channels by *OH (as produced from H2O2 in the apoplast in the presence of transition metals such as Fe2+ or Cu+) may be downstream of the H2O2 signalling. This may provide appropriate adjustments in water relations (hydraulic conductivity), and a common response to different kinds of stresses. [source]


Evaluation of estrogenic activity of phthalate esters by gene expression profiling using a focused microarray (estrarray®),

Abstract Phthalates are used industrially as plasticizers and are known to contaminate natural environments, mostly as di-ester or mono-ester complexes. Because they are structurally similar to natural estrogens, they could act as endocrine disruptors. Here, we used a DNA microarray containing estrogen responsive genes (EstrArray®) to examine gene expression profiles in MCF-7 cells treated with 10 ,M butylbenzyl phthalate (BBP), dibutyl phthalate (DBP), diethyl phthalate (DEP), and diisopropyl phthalate (DIP) along with the natural estrogen 17,-estradiol ([E2], 10 nM). The profiles for phthalate esters and E2 were examined by correlation analysis using correlation coefficients (r -values) and cluster analysis. We found that BBP showed the highest correlation with E2 (r = 0.85), and DEP and DIP showed moderate r -values (r = 0.52 and r = 0.49, respectively). Dibutyl phthalate exhibited the lowest (but still significant) correlation with E2 (r = 0.36). Furthermore, among the pairs of chemicals, DEP-DIP and DIP-DBP showed very high correlations (r = 0.90 and r = 0.80, respectively), and the other pairs showed moderate relationships, which reflected how structurally close they are to each other. The analysis of six functional groups of genes (enzymes, signaling, proliferation, transcription, transport, and others) indicated that the genes belonging to the enzyme, transcription, and other functional groups showed common responses to phthalate esters and E2. Although the effect of BBP was similar to that of E2, the other phthalate esters showed different types of effects. These results indicate that the structure of estrogenic chemicals is strongly related to their estrogenic activity and can be evaluated by appropriate grouping of the responsive genes by focused microarray analysis. Predicting changes in community composition and ecosystem functioning from plant traits: revisiting the Holy Grail

Summary 1The concept of plant functional type proposes that species can be grouped according to common responses to the environment and/or common effects on ecosystem processes. However, the knowledge of relationships between traits associated with the response of plants to environmental factors such as resources and disturbances (response traits), and traits that determine effects of plants on ecosystem functions (effect traits), such as biogeochemical cycling or propensity to disturbance, remains rudimentary. 2We present a framework using concepts and results from community ecology, ecosystem ecology and evolutionary biology to provide this linkage. Ecosystem functioning is the end result of the operation of multiple environmental filters in a hierarchy of scales which, by selecting individuals with appropriate responses, result in assemblages with varying trait composition. Functional linkages and trade-offs among traits, each of which relates to one or several processes, determine whether or not filtering by different factors gives a match, and whether ecosystem effects can be easily deduced from the knowledge of the filters. 3To illustrate this framework we analyse a set of key environmental factors and ecosystem processes. While traits associated with response to nutrient gradients strongly overlapped with those determining net primary production, little direct overlap was found between response to fire and flammability. 4We hypothesize that these patterns reflect general trends. Responses to resource availability would be determined by traits that are also involved in biogeochemical cycling, because both these responses and effects are driven by the trade-off between acquisition and conservation. On the other hand, regeneration and demographic traits associated with response to disturbance, which are known to have little connection with adult traits involved in plant ecophysiology, would be of little relevance to ecosystem processes. 5This framework is likely to be broadly applicable, although caution must be exercised to use trait linkages and trade-offs appropriate to the scale, environmental conditions and evolutionary context. It may direct the selection of plant functional types for vegetation models at a range of scales, and help with the design of experimental studies of relationships between plant diversity and ecosystem properties. Chimpanzee responses to researchers in a disturbed forest,farm mosaic at Bulindi, western Uganda

Abstract We describe the behavior of a previously unstudied community of wild chimpanzees during opportunistic encounters with researchers in an unprotected forest,farm mosaic at Bulindi, Uganda. Data were collected during 115 encounters between May 2006 and January 2008. Individual responses were recorded during the first minute of visual contact. The most common responses were "ignore" for arboreal chimpanzees and "monitor" for terrestrial individuals. Chimpanzees rarely responded with "flight". Adult males were seen disproportionately often relative to adult females, and accounted for 90% of individual responses recorded for terrestrial animals. Entire encounters were also categorized based on the predominant response of the chimpanzee party to researcher proximity. The most frequent encounter type was "ignore" (36%), followed by "monitor" (21%), "intimidation" (18%) and "stealthy retreat" (18%). "Intimidation" encounters occurred when chimpanzees were contacted in dense forest where visibility was low, provoking intense alarm and agitation. Adult males occasionally acted together to repel researchers through aggressive mobbing and pursuit. Chimpanzee behavior during encounters reflects the familiar yet frequently agonistic relationship between apes and local people at Bulindi. The chimpanzees are not hunted but experience high levels of harassment from villagers. Human-directed aggression by chimpanzees may represent a strategy to accommodate regular disruptions to foraging effort arising from competitive encounters with people both in and outside forest. Average encounter duration and proportion of encounters categorized as "ignore" increased over time, whereas "intimidation" encounters decreased, indicating some habituation occurred during the study. Ecotourism aimed at promoting tolerance of wildlife through local revenue generation is one possible strategy for conserving great apes on public or private land. However, the data imply that habituating chimpanzees for viewing-based ecotourism in heavily human-dominated landscapes, such as Bulindi, is ill-advised since a loss of fear of humans could lead to increased negative interactions with local people. Interdependencies between agricultural commodity futures prices on the LIFFE

Interdependencies between commodity prices can arise from the impact of changing macroeconomic variables, from complementarities or substitutabilities between commodities, or from common responses by speculators. Malliaris and Urrutia (1996) found significant linkages between rollover prices of six related agricultural commodities on the Chicago Board of Trade. This article examines interdependencies between futures prices for soft commodities traded on the London International Financial Futures Exchange (LIFFE), calculated using Clark indices. Animal performance and stress: responses and tolerance limits at different levels of biological organisation

ABSTRACT Recent advances in molecular biology and the use of DNA microarrays for gene expression profiling are providing new insights into the animal stress response, particularly the effects of stress on gene regulation. However, interpretation of the complex transcriptional changes that occur during stress still poses many challenges because the relationship between changes at the transcriptional level and other levels of biological organisation is not well understood. To confront these challenges, a conceptual model linking physiological and transcriptional responses to stress would be helpful. Here, we provide the basis for one such model by synthesising data from organismal, endocrine, cellular, molecular, and genomic studies. We show using available examples from ectothermic vertebrates that reduced oxygen levels and oxidative stress are common to many stress conditions and that the responses to different types of stress, such as environmental, handling and confinement stress, often converge at the challenge of dealing with oxygen imbalance and oxidative stress. As a result, a common set of stress responses exists that is largely independent of the type of stressor applied. These common responses include the repair of DNA and protein damage, cell cycle arrest or apoptosis, changes in cellular metabolism that reflect the transition from a state of cellular growth to one of cellular repair, the release of stress hormones, changes in mitochondrial densities and properties, changes in oxygen transport capacities and changes in cardio-respiratory function. Changes at the transcriptional level recapitulate these common responses, with many stress-responsive genes functioning in cell cycle control, regulation of transcription, protein turnover, metabolism, and cellular repair. These common transcriptional responses to stress appear coordinated by only a limited number of stress-inducible and redox-sensitive transcription factors and signal transduction pathways, such as the immediate early genes c-fos and c-jun, the transcription factors NF,B and HIF - 1,, and the JNK and p38 kinase signalling pathways. As an example of environmental stress responses, we present temperature response curves at organismal, cellular and molecular levels. Acclimation and physiological adjustments that can shift the threshold temperatures for the onset of these responses are discussed and include, for example, adjustments of the oxygen delivery system, the heat shock response, cellular repair system, and transcriptome. Ultimately, however, an organism's ability to cope with environmental change is largely determined by its ability to maintain aerobic scope and to prevent loss in performance. These systemic constraints can determine an organism's long-term survival well before cellular and molecular functions are disturbed. Model identification in presence of incomplete information by generalized principal component analysis: Application to the common and differential responses of Escherichia coli to multiple pulse perturbations in continuous, high-biomass density culture

Abstract In a previous report we described a multivariate approach to discriminate between the different response mechanisms operating in Escherichia coli when a steady, continuous culture of these bacteria was perturbed by a glycerol pulse (Guebel et al., 2009, Biotechnol Bioeng 102: 910,922). Herein, we present a procedure to extend this analysis when multiple, spaced pulse perturbations (glycerol, fumarate, acetate, crotonobetaine, hypersaline plus high-glycerol basal medium and crotonobetaine plus hypersaline basal medium) are being assessed. The proposed method allows us to identify not only the common responses among different perturbation conditions, but to recognize the specific response for a given stimulus even when the dynamics of the perturbation is unknown. Components common to all conditions are determined first by Generalized Principal Components Analysis (GPCA) upon a set of covariance matrices. A metrics is then built to quantify the similitude distance. This is based on the degree of variance extraction achieved for each variable along the GPCA deflation processes by the common factors. This permits a cluster analysis, which recognizes several compact sub-sets containing only the most closely related responsive groups. The GPCA is then run again but is restricted to the groups in each sub-set. Finally, after the data have been exhaustively deflated by the common sub-set factors, the resulting residual matrices are used to determine the specific response factors by classical principal component analysis (PCA). The proposed method was validated by comparing its predictions with those obtained when the dynamics of the perturbation was determined. In addition, it showed to have a better performance than the obtained with other multivariate alternatives (e.g., orthogonal contrasts based on direct GPCA, Tucker-3 model, PARAFAC, etc.).
