Predicting world health organization toxic equivalency factor dioxin and dioxin-like polychlorinated biphenyl levels in farmed atlantic salmon (Salmo salar) based on known levels in feed

Abstract Assimilation and elimination rate constant of dietary polychlorinated dibenzo- p -dioxins and dibenzofurans (PCDD/Fs) and dioxin-like polychlorinated biphenyls (DLPCBs) with a World Health Organization toxic equivalency factor (WHO-TEF) were estimated in market-size Atlantic salmon (Salmo salar) using fish that were previously fed vegetable oil,based (low in PCDD/Fs and DLPCBs) or fish oil,based (high in PCDD/Fs and PCBs) diets. At the start of the kinetic trial, half the fish that were fed fish oils were fed vegetable oil feeds and inverted (cross-over design) for five months. The assimilation efficiencies of the PCDD/F congeners were more variable (3,89%) and, generally, were lower than those of the DLPCBs (70,80%). Among the PCDD/F congeners, the assimilation efficiency of the most toxic tetra- and pentachlorinated PCDD/Fs was greater than that of higher-chlorinated PCDD/Fs. Elimination rates for DLPCBs were higher than those for PCDD/Fs. Lower-chlorinated PCDDs had a lower elimination rate than the higher-chlorinated PCDDs, but no differences were observed among PCDF congeners or DLPCB congeners. Kinetic parameters were used to predict the level of WHO-TEF dioxins and DLPCBs in Atlantic salmon reared in a large-scale facility under commercial conditions. Predictions were based on preanalyzed levels of these organochlorines in feeds with three different replacement levels (0, 30, and 60%) of vegetable oil. A simple one-compartmental, first-order kinetic model was used to predict the level of sum WHO toxic equivalents for PCDD/Fs and DL-PCBs. The predicted values varied by 0 to 11% from the measured values in the commercially reared salmon. [source]


The influence of pre-fermentative practices on the dominance of inoculated yeast starter under industrial conditions

Abstract The influence of pre-fermentative practices on the growth dynamics of a ,natural' starter culture with specific phenotype (H2S,) concurrently with wild yeast populations was evaluated under winery conditions. Different clarification procedures and added SO2 strongly influenced species and cell numbers isolable at the pre-fermentation stage. Independent treatments of must with sulphite addition or vacuum-filtering clarification caused a 30-fold reduction in viable cells. Clarification procedures, enhanced by the selective effect of SO2 addition, induced the appearance of Saccharomyces cerevisiae ,wild' yeasts. Correct application of the inoculum generally guarantees the dominance of fermentation by starter cultures. However, inoculated fermentations using unclarified white and red musts exhibited a consistent presence and persistence of non- Saccharomyces and/or Saccharomyces ,wild' yeasts during fermentation. The extent and composition of the initial wild microflora at the start of fermentation may affect the presence and persistence of wild Saccharomyces and non- Saccharomyces yeasts during guided fermentations under commercial conditions. The above findings confirm the results of previous works carried out at laboratory- or pilot-scale level. Furthermore, they suggest a clear correlation between the modality of pre-fermentative practices and the presence and persistence of ,wild' yeasts during fermentation. © 2002 Society of Chemical Industry [source]


Volatile metabolite profiling to detect and discriminate stem-end rot and anthracnose diseases of mango fruits

The volatile metabolites from the headspace gas of containerised mango (Mangifera indica) cv. Tommy Atkins fruits, surface wounded and inoculated with the two fungal anamorphic pathogens Colletotrichum gloeosporioides and Lasiodiplodia theobromae, or non-inoculated (controls), were profiled using a portable gas chromatograph/mass spectrometer to discriminate diseases of mango. Thirty-four compounds were detected relatively consistently among replicates. Several of these were disease/inoculation-discriminatory and were classified into three groups: (i) compounds unique to only one treatment; (ii) compounds common to two or more treatments, but not to all; and (iii) compounds common to all treatments, but varying in their abundance. Two compounds, 1-pentanol and ethyl boronate, were detected in L. theobromae- inoculated mangoes alone, while thujol was observed only in C. gloeosporioides- inoculated mangoes. Discriminant analysis models based on the abundance of significant mass ions and consistent compounds correctly classified diseases/inoculations in up to 100% of cases. The disease-discriminatory compounds and discriminant analysis models developed here have the potential to be used in the early detection of postharvest diseases of mango fruits after validation under commercial conditions. Identification of genetic markers associated with fatness and leg weakness traits in the pig

Summary Pigs have undergone long-term selection in commercial conditions for improved rate and efficiency of lean gain. Interestingly, it has been observed in both experimental and field conditions that leg weakness has increased over time, concurrent with the selection for improved rate of lean gain, while fatter animals tend to have better leg action, and foot and leg (FL) structure. The exact molecular mechanisms or individual genes responsible for this apparent genetic correlation between fatness and leg weakness and other physical adaptability traits have been less well reported. Based on our recent studies involving candidate genes and leg weakness traits, the present investigation has identified 30 SNPs from 26 genes that were found to be associated with 10th rib backfat in a sow population consisting of 2066 animals. The specific alleles associated with increased backfat tended to be associated with better overall leg action, as shown for the genes including MTHFR, WNT2, APOE, BMP8, GNRHR and OXTR, while inconsistent associations with the single FL structure trait and backfat were observed for other genes. This study suggests that in some cases there may be a common genetic mechanism or linked genes regulating fatness and leg weakness. Such relationships are clearly complex, and the utilization of genetic markers associated with both traits should be treated cautiously. [source]


A comparison of two faecal collection methods for protein and amino acid digestibility coefficients of menhaden fish meal and two grades of poultry by-product meals for market-size sunshine bass (Morone chrysops × M. saxatilis)

Abstract Apparent digestibility and availability coefficients for protein and amino acids in menhaden fish meal (MEN), pet-food grade (PBM-pet,) and feed-grade poultry by-product meal (PBM-feed) were determined for market-size (500 g) sunshine bass in two consecutive trials using passive netting (1.6 mm mesh) followed by manual stripping of faeces. A reference diet resembling a commercial feed was formulated to meet or exceed all known nutritional requirements of hybrid striped bass. Test diets were formulated to contain a 70 : 30 mixture of reference diet to test ingredient with chromic oxide (1%) serving as the inert marker. Diets were extruded under commercial conditions and the reference diet was fed for two weeks in order to acclimate fish to experimental conditions. Each diet was randomly assigned to triplicate tanks containing 30 fish each. Fish were fed their respective diet twice daily to apparent satiation for two weeks, with faecal collections being conducted on the 7th and 14th day, in each trial. ADC values determined in the net method were highly variable and generally lower than ADCs obtained by stripping. Consistently lower and highly variable chromium concentrations were found in the net method faecal samples and suggest that marker loss relative to nutrient content, or dilution of marker with non-faecal matter, on the net collectors influenced results in that trial. In contrast, the standard errors of ADCs determined in the strip method were less than 5 percentage points in most cases. Protein digestibility ranged from a low of 51% (PBM-feed) to a high of 87% (PBM-pet) in the net method, and from a low of 80% (PBM-feed) to a high of 99% (MEN) in the strip method. With the exception of Lys, no differences in amino acid availabilities among diets were found in the net method. In the strip method, protein digestibility and amino acid availabilities in MEN were generally greater than those found in PBM-pet or PBM-feed, whereas ADCs were not significantly different between the two poultry by-products. Based on the conditions of the present study, net collection of faecal matter can not be recommended for determining the digestibility of nutrients in feed ingredients for market-size sunshine bass. Digestibility coefficients obtained by the strip method for feed and petfood grades poultry by-product were higher than those previously reported and will be facilitate more efficient and economical diet formulations for larger sunshine bass. Commercial assessment of growth and mortality of fifth-generation Sydney rock oysters Saccostrea glomerata (Gould, 1850) selectively bred for faster growth

Abstract The Sydney rock oyster Saccostrea glomerata (Gould, 1850) industry acquired hatchery-produced spat selected for faster growth for the first time in January 2004. Selectively bred and non-selected (Control) spat produced concurrently were used to compare performance when grown under commercial conditions. Spat were distributed to farmers in seven estuaries in New South Wales. Individual farmers cultivated these oysters using their own techniques and growth and mortality were recorded quarterly. At each site, the two oyster types were cultivated using the same culture method, location and density. Growth was compared when oysters were 27 months of age. At this time, selectively bred oysters were significantly larger and heavier than Control oysters. The same result was obtained when oysters were compared at the point in time when selectively bred oysters had reached 50 g at each site. No significant difference was found for cumulative mortality between the selectively bred oysters and Control oysters across all sites. However, the seven sites had significantly different levels of cumulative mortality. Overall, the performance of selectively bred oysters was superior to the Control oysters and selectively bred oysters reached the 50 g bench mark within 29.3 months when averaged across all sites.
