A truncated bovine ,-1,3-galactosyltransferase, the key enzyme responsible for the biosynthesis of the terminal structure of ,-Gal, was cloned and overexpressed previously. The acceptor specificity was further studied in the present paper, and lactose and galactose derivatives were found to be good acceptors. To develop a more proficient reaction process, we report herein an example of an efficient enzymatic synthesis of ,-Gal oligosaccharides catalyzed by the combination of two recombinant Escherichia coli whole cells harboring the genes of a UDP-galactose 4-epimerase and the ,-1,3-galactosyltransferase, respectively. Using lactosyl azide (LacN3) as the acceptor for the glycosyltransferase, the combined use of the two recombinant cells efficiently produced ,-Gal epitope Gal,1,3LacN3 in 60,68% yield. [source] Parallel four-dimensional Haralick texture analysis for disk-resident image datasetsCONCURRENCY AND COMPUTATION: PRACTICE & EXPERIENCE, Issue 1 2007Brent Woods Abstract Texture analysis is one possible method of detecting features in biomedical images. During texture analysis, texture-related information is found by examining local variations in image brightness. Four-dimensional (4D) Haralick texture analysis is a method that extracts local variations along space and time dimensions and represents them as a collection of 14 statistical parameters. However, application of the 4D Haralick method on large time-dependent image datasets is hindered by data retrieval, computation, and memory requirements. This paper describes a parallel implementation using a distributed component-based framework of 4D Haralick texture analysis on PC clusters. The experimental performance results show that good performance can be achieved for this application via combined use of task- and data-parallelism. [source] ENDOSCOPIC SUBMUCOSAL DISSECTION FOR EARLY GASTRIC CANCER USING MAGNIFYING ENDOSCOPY WITH A COMBINATION OF NARROW BAND IMAGING AND ACETIC ACID INSTILLATIONDIGESTIVE ENDOSCOPY, Issue 3 2008Kyosuke Tanaka Demarcation of early gastric cancers is sometimes unclear. Enhanced-magnification endoscopy with acetic acid instillation and magnifying endoscopy with a narrow band imaging (NBI) system have been useful for recognition of demarcation of early gastric cancers. We report a patient with early gastric cancer who underwent a successful endoscopic submucosal dissection (ESD) by magnifying endoscopy with the combined use of NBI and acetic acid instillation. A 72-year-old man with early gastric cancer underwent ESD. Demarcation of the lesion was not clear, but magnifying endoscopy using the combination of NBI and acetic acid clearly revealed the demarcation. ESD was carried out after spots were marked circumferentially. We identified the positional relation between the demarcation and all markings. Resection of the lesion was on the outside of the markings. Histopathologically, the lesion was diagnosed as a well-differentiated adenocarcinoma limited to the mucosa. The margins were carcinoma free. Magnifying endoscopy combining the use of NBI with acetic acid instillation is simple and helpful for identifying the demarcation of early gastric cancer. This method may be useful in increasing the rate of complete resection by ESD for early gastric cancer. [source] Three-dimensional reconstruction of the mucosa from sequential sections of biopsy specimens of patients with ulcerative colitis: Relationship between crypt structure and vascular architectureDIGESTIVE ENDOSCOPY, Issue 2 2004Hiroo Furukawa Background:, In a previous paper, the stereographic reconstruction of the crypt structure of ulcerative colitis using the RATOCK System was described. The relationship between the blood vessels and the crypt structure is the focus of the current paper, using two kinds of tissue staining color in which the color differs. Stereographic images make the relationship between the crypt structure and blood vessel distribution understandable at a glance. Methods:, The methods used here are identical to those described in a previous paper. In the present paper, five cases of ulcerative colitis (UC) are examined. Biopsy specimens were obtained from the diseased, normal, and transitional zones (the area between the normal and diseased zones) from each patient. Three-dimensional reconstruction was created using TRI for Windows (RATOC System Tokyo, Japan) software. In the present paper, two kinds of dyeing method between H&E and monoclonal antibody staining of the tissue was used. It was proven that the distribution of gland and blood vessel is very clear in the 3-D reconstruction shown. Results:, (i) The blood vessels in the normal zones run parallel to the crypt in a regular manner and are almost identical to one another in diameter. (ii) In the transitional and diseased zones, the blood vessels show no clear direction and produce many branches without any apparent order. The blood vessels are, moreover, irregular in diameter. (iii) In short, clear parallelism is lost in both the transitional and diseased zones. Conclusion:, Stereographic reconstruction of endoscopically obtained biopsy specimens of UC-affected tissues makes it possible to understand at a glance the distribution of blood vessels and their relationship to crypts. The relationship of these was clarified by the combined use of two kinds of dyeing method with three-dimensional reconstruction. [source] Combined use of chiral ionic liquid and cyclodextrin for MEKC: Part I. Simultaneous enantioseparation of anionic profensELECTROPHORESIS, Issue 16 2009Bin Wang Abstract The enantiomers of five profen drugs were simultaneously separated by MEKC with the combined use of 2,3,6-tri- O -methyl-,-cyclodextrin and chiral cationic ionic liquid, N -undecenoxy-carbonyl- L -leucinol bromide, which formed micelles in aqueous buffers. Enantioseparations of these profen drugs were optimized by varying the chain length and concentration of the IL surfactant using a standard recipe containing 35,mM 2,3,6-tri- O -methyl-,-cyclodextrin, 5,mM sodium acetate at pH 5.0. The batch-to-batch reproducibility of N -undecenoxy-carbonyl- L -leucinol bromide was tested and found to have no significant impact in terms of enantiomeric resolution, efficiency, and migration time. Finally, this method was successfully applied for the quantitative determination of ibuprofen in pharmaceutical tablets. [source] Progression of oral snuff use among Finnish 13,16-year-old students and its relation to smoking behaviourADDICTION, Issue 4 2006Ari Haukkala ABSTRACT Aims To examine the progression of oral moist snuff use among adolescents and its relation to smoking behaviour and nicotine addiction. Design and setting A 3-year smoking prevention study in 27 schools of Helsinki, Finland, starting with the seventh grade to the ninth grade., Participants and measurements Pupils (n = 2816) completed questionnaires four times, which included information on smoking behaviour, snuff experiments, nicotine addiction (Fagerström Tolerance Questionnaire) and other activities. Findings The prevalence of snuff experimentation rose among boys from 7% in the seventh grade to 43% 3 years later in the ninth grade, and among girls from 2% to 13% for the corresponding period. Among boys, smoking predicted later snuff use in all assessments and snuff experimentation predicted later weekly smoking. The impact of snuff experimentation upon later smoking experimentation was smaller than vice versa. Among boys active in sports, smoking was less common but snuff use was more common. Combined use was common; by the end of the follow-up only 10% of weekly smokers had not tried oral snuff. Nicotine dependence scores increased linearly with snuff use among weekly smokers., Conclusions Despite the European Union sales ban on oral snuff products since 1995, in Finland snuff use is common among boys. Although combined use of snuff and cigarettes is associated with higher levels of nicotine dependence among adolescent boys, the direction of causality is not known. Unlike cigarette smoking, oral snuff use was tried among boys who spent their free time with sports-related activities. [source] Combined Ketogenic Diet and Vagus Nerve Stimulation: Rational Polytherapy?EPILEPSIA, Issue 1 2007Eric H. Kossoff Summary:,Objective: The concept of "rational polypharmacy" has been associated with anticonvulsant management for decades, but the term has not been applied to nonpharmacologic therapies. Methods: We conducted a multicenter, retrospective study of children who received concurrent diet (ketogenic or modified Atkins) and vagus nerve stimulation (VNS) treatment for medically intractable epilepsy. Results: Thirty children in total from six epilepsy centers were treated over a 6-yr period. The median age at the initiation of combination therapy was 10 yr (range, 4,24 yr). Sixteen (53%) received dietary therapy followed by VNS; no differences were noted between centers. After 3 months, 21 (70%) had seizure reduced by >50% over the previous single nonpharmacologic treatment, of whom 13 (62%) had improvement within the first month. A 5-min VNS off-time correlated with >90% seizure reduction (p = 0.02). The median duration of nonpharmacologic polytherapy was 12 months (range, 0.5,96 months); 17 (57%) remain on dual therapy at this time. No side effects were noted. Most patients who discontinued combination therapy did so because of a lack of efficacy rather than restrictiveness. Conclusions: In this small group, the combined use of diet and VNS appeared synergistic and yielded rapid benefits. It may be more effective with longer VNS off-times. Further prospective studies of this combination in refractory pediatric epilepsy are needed to help guide optimal use. [source] A case of small-cell gastric carcinoma with an adenocarcinoma component and hepatic metastases: treatment with systemic and intra-hepatic chemotherapyEUROPEAN JOURNAL OF CANCER CARE, Issue 5 2007T. CIOPPA md Primary small-cell carcinoma (SmCC) of the stomach is a rare neoplasm with a poor prognosis and unclear histogenesis: to date, only 50 cases, including ours, have been reported in the literature. In the World Health Organization gastrointestinal tumours' classification, SmCC of the stomach has been recognized as an ,independent entity affecting the stomach'. In this paper, the authors present a clinical case and the surgical treatment of an adult with a SmCC of the stomach associated with gastric adenocarcinoma. After laparotomy, a large neoplasm with locoregional extension and multiple liver metastases were found. A palliative resection, subtotal gastrectomy, was performed, followed by systemic and intra-hepatic chemotherapy: computed tomography scan demonstrated a marked response, but the patient died 15 months after the operation. A review of the literature showed that the diagnosis of gastric SmCC is based on immunohistochemical findings. Our experience confirmed the high aggressiveness of this neoplasm, which is generally diagnosed in advanced stage and is unresponsive to chemotherapy, but the combined use of systemic and intra-hepatic chemotherapy shows an acceptable result in a palliative care perspective. The titanium alkoxide catalysts are protected from the water co-product by the combined use of a tightly binding trialkanolamine ligand and molecular sieves. Nitrones can be obtained in high yields (up to 98,%) under homogeneous, anhydrous conditions and even in the absence of solvent. The reactions are fast (2,7 h) and good selectivity and complete conversion can be achieved with as little as 1,% catalyst. [source] Loss of phosphorus from soil in semi-arid northern Tanzania as a result of cropping: evidence from sequential extraction and 31P-NMR spectroscopyEUROPEAN JOURNAL OF SOIL SCIENCE, Issue 4 2000D. Solomon Summary In semi-arid northern Tanzania, the native woodland is being rapidly cleared and replaced by low input agriculture. This has resulted in pronounced environmental degradation, and in particular loss of phosphorus (P) from the soil. We have used sequential extraction and 31P-NMR to investigate the effects of land use changes, i.e. native woodland, degraded woodland, cultivation for 3 and 15 years and homestead fields where manure was applied, on the amount and structural composition of P in this soil. Clearing and continuous cultivation reduced both organic and inorganic P in the soil. The difference in the amount of organic P from the bulk soil of the fields cultivated for 3 and 15 years was not statistically significant (P <,0.05), suggesting that most of the depletion in organic P occurred during the first 3 years of cultivation. By contrast, in the homesteads, there was much organic and inorganic P in the soil. The 31P-NMR revealed that cultivation resulted in a 53% depletion of orthophosphate diester P, whereas only a 30% and 39% reduction of orthophosphate monoester P was found in the bulk soil after 3 and 15 years of cultivation, respectively. These results concur with the suggestion that diester P constitutes more easily mineralizable forms of organic P in soil than does monoester P. Our 31P-NMR also showed that 70% of the inorganic orthophosphate P was depleted from the coarse and fine sand separates as a result of cultivation. The influence of clearing and subsequent cropping on the amount and forms of P was more pronounced in the coarse and fine sand than in the silt and clay, stressing the importance of particle size and chemical properties such as organic matter and oxides in the availability of P in this soil. Our results show that the current low input agricultural practice is not sustainable, and that practices must be developed to combat the ongoing degradation of the soil. A combined use of available organic materials such as animal manure with the judicious use of inorganic fertilizers can replenish the soil's fertility. Jullien Somatic angiotensin-converting enzyme (ACE) contains two homologous domains, each bearing a functional active site. Studies on the selectivity of these ACE domains towards either substrates or inhibitors have mostly relied on the use of mutants or isolated domains of ACE. To determine directly the selectivity properties of each ACE domain, working with wild-type enzyme, we developed an approach based on the combined use of N-domain-selective and C-domain-selective ACE inhibitors and fluorogenic substrates. With this approach, marked differences in substrate selectivity were revealed between rat, mouse and human somatic ACE. In particular, the fluorogenic substrate Mca-Ala-Ser-Asp-Lys-DpaOH was shown to be a strict N-domain-selective substrate of mouse ACE, whereas with rat ACE it displayed marked C-domain selectivity. Similar differences in selectivity between these ACE species were also observed with a new fluorogenic substrate of ACE, Mca-Arg-Pro-Pro-Gly-Phe-Ser-Pro-DpaOH. In support of these results, changes in amino-acid composition in the binding site of these three ACE species were pinpointed. Together these data demonstrate that the substrate selectivity of the N-domain and C-domain depends on the ACE species. These results raise concerns about the interpretation of functional studies performed in animals using N-domain and C-domain substrate selectivity data derived only from human ACE. [source] CdS-Nanoparticle/Polymer Composite Shells Grown on Silica Nanospheres by Atom-Transfer Radical Polymerization,ADVANCED FUNCTIONAL MATERIALS, Issue 3 2005T. Cui Abstract In this paper we describe the combined use of surface-initiated atom transfer radical polymerization (ATRP) and a gas/solid reaction in the direct preparation of CdS-nanoparticle/block-copolymer composite shells on silica nanospheres. The block copolymer, consisting of poly(cadmium dimethacrylate) (PCDMA) and poly(methyl methacrylate) (PMMA), is obtained by repeatedly performing the surface-initiated ATRP procedures in N,N -dimethylformamide (DMF) solution at room temperature, using cadmium dimethacrylate (CDMA) and methyl methacrylate (MMA) as the monomers. CdS nanoparticles with an average size of about 3,nm are generated in situ by exposing the silica nanospheres coated with block-copolymer shells to H2S gas. These synthetic core,shell nanospheres were characterized using transmission electron microscopy (TEM), dynamic light scattering (DLS), thermogravimetric analysis (TGA), diffuse reflectance UV-vis spectroscopy, X-ray photoelectron spectroscopy (XPS), and powder X-ray diffraction (XRD). These composite nanospheres exhibit strong red photoluminescence in the solid state at room temperature. [source] THE "INS" AND "OUTS" OF HISTORY: REVISION AS NON-PLACEHISTORY AND THEORY, Issue 4 2007MARNIE HUGHES-WARRINGTON ABSTRACT Revision in history is conventionally characterized as a linear sequence of changes over time. Drawing together the contributions of those engaged in historiographical debates that are often associated with the term "revision," however, we find our attention directed to the spaces rather than the sequences of history. Contributions to historical debates are characterized by the marked use of spatial imagery and spatialized language. These used to suggest both the demarcation of the "space of history" and the erasure of existing historiographies from that space. Bearing these features in mind, the essay argues that traditional, temporally oriented explanations for revision in history, such as Thomas S. Kuhn's Structure of Scientific Revolutions, miss the mark, and that a more promising line of explanation arises from the combined use of Michel Foucault's idea of "heterotopias" and Marc Augé's idea of "non-places." Revision in history is to be found where writers use imagery to move readers away from rival historiographies and to control their movement in the space of history toward their desired vision. Revision is thus associated more with control than with liberation. [source] Remotely sensed data used for modelling at different hydrological scalesHYDROLOGICAL PROCESSES, Issue 8 2002Peter Droogers Abstract There is a growing awareness that water will be one of the most critical natural resources and that there is a need for better management of the limited water resources. This paper reports on a study of a water-scarce river basin in western Turkey. Hydrological analyses, emphasizing water use for irrigation, are performed at three different spatial scales (field scale, irrigation scheme scale and basin scale) using two kind of model: a parametric basin-scale model and a physically based crop-scale model. Data accessibility for this basin, especially for areal data, was low. A combined use of public domain data sets and remotely sensed data was used to solve this problem. A public domain digital elevation model was used to generate the streamflow network and the distances and slopes to streams. Land-cover data and leaf area index data were derived from public domain NOAA,AVHRR images. For one irrigation scheme in the basin, detailed areal water balances were obtained from the simulation model and a comparison was made between a normal and a water-short year. At the basin scale, observed flows were compared with simulated flows. It is concluded that remotely sensed data and other public domain data can be used with simulation models at different scales to create a powerful tool to evaluate water resources in a basin context. [source] Reproduction of temporal scaling by a rectangular pulses rainfall modelHYDROLOGICAL PROCESSES, Issue 3 2002Jonas Olsson Abstract The presence of scaling statistical properties in temporal rainfall has been well established in many empirical investigations during the latest decade. These properties have more and more come to be regarded as a fundamental feature of the rainfall process. How to best use the scaling properties for applied modelling remains to be assessed, however, particularly in the case of continuous rainfall time-series. One therefore is forced to use conventional time-series modelling, e.g. based on point process theory, which does not explicitly take scaling into account. In light of this, there is a need to investigate the degree to which point-process models are able to ,unintentionally' reproduce the empirical scaling properties. In the present study, four 25-year series of 20-min rainfall intensities observed in Arno River basin, Italy, were investigated. A Neyman,Scott rectangular pulses (NSRP) model was fitted to these series, so enabling the generation of synthetic time-series suitable for investigation. A multifractal scaling behaviour was found to characterize the raw data within a range of time-scales between approximately 20 min and 1 week. The main features of this behaviour were surprisingly well reproduced in the simulated data, although some differences were observed, particularly at small scales below the typical duration of a rain cell. This suggests the possibility of a combined use of the NSRP model and a scaling approach, in order to extend the NSRP range of applicability for simulation purposes. [source] Wrapped input selection using multilayer perceptrons for repeat-purchase modeling in direct marketingINTELLIGENT SYSTEMS IN ACCOUNTING, FINANCE & MANAGEMENT, Issue 2 2001Stijn Viaene In this paper, we try to validate existing theory on and develop additional insight into repeat-purchase behavior in a direct marketing setting by means of an illuminating case study. The case involves the detection and qualification of the most relevant RFM (Recency, Frequency and Monetary) variables, using a neural network wrapper as our input pruning method. Results indicate that elimination of redundant and/or irrelevant inputs by means of the discussed input selection method allows us to significantly reduce model complexity without degrading the predictive generalization ability. It is precisely this issue that will enable us to infer some interesting marketing conclusions concerning the relative importance of the RFM predictor categories and their operationalizations. The empirical findings highlight the importance of a combined use of RFM variables in predicting repeat-purchase behavior. However, the study also reveals the dominant role of the frequency category. Results indicate that a model including only frequency variables still yields satisfactory classification accuracy compared to the optimally reduced model. Copyright © 2001 John Wiley & Sons, Ltd. [source] Prevention of secondary stroke and transient ischaemic attack with antiplatelet therapy: the role of the primary care physician roleINTERNATIONAL JOURNAL OF CLINICAL PRACTICE, Issue 10 2007H. S. Kirshner Summary Background:, Stroke risk is heightened among patients who have had a primary stroke or transient ischaemic attack (TIA). The primary care physician is in the best position to monitor these patients for stroke recurrence. Because stroke recurrence can occur shortly after the primary event, guidelines recommend initiating antiplatelet therapy as soon as possible. Aspirin, with or without extended-release dipyridamole (ER-DP), and clopidogrel are options for such patients. Low-dose aspirin (75,150 mg/day) has the same efficacy as higher doses but with less gastrointestinal bleeding. Clopidogrel remains an option for prevention of secondary events and may benefit patients with symptomatic atherothrombosis, but its combined use with aspirin can harm patients with multiple risk factors and no history of symptomatic cerebrovascular, cardiovascular or peripheral vascular disease. Results:, Low dose aspirin is effective in secondary stroke prevention. Trials assessing aspirin plus ER-DP have shown that the combination is more effective than aspirin monotherapy in preventing stroke, with efficacy increasing among higher risk patients, notably those with prior stroke/TIA. Clopidogrel does not appear to have as much advantage over aspirin in secondary stroke prevention as aspirin plus ER-DP. Smoking cessation and cholesterol, blood glucose and blood pressure control are also important concerns in preventing recurrent stroke. In choosing pharmacological therapy, the physician must consider the individual patient's risk factors and tolerance, as well as other issues, such as use of aspirin among patients with ulcers. Conclusion:, Antiplatelet therapy is effective in secondary stroke prevention. Low dose aspirin can be used first-line, but aspirin plus ER-DP improves efficacy. Clopidogrel is another option in secondary stroke prevention, especially for aspirin-intolerant patients, but it appears to have less advantage over aspirin than aspirin plus ER-DP, and its combined use with aspirin has only marginally better efficacy and increased bleeding risk. Abonamah Abstract Shared buffering and channel grouping are powerful techniques with great benefits in terms of both performance and implementation. Shared-buffer switches are known to have better performance and better utilization than input or output queued switches. With channel grouping, a cell is routed to a group of channels instead of a specific output channel. In this way, congestion due to output contention can be minimized and the switch performance can therefore be greatly improved. Although each technique is well known by itself in the traditional study of queuing systems, their combined use in ATM networks has not been much explored previously. In this paper, we develop an analytical model for a shared-buffer ATM switch with grouped output channels. The model is then used to study the switch performance in terms of cell loss probability, cell delay and throughput. In particular, we study the impact of the channel grouping factor on the buffer requirements. [source] Design techniques of two-layer architectures for WDM optical networksINTERNATIONAL JOURNAL OF COMMUNICATION SYSTEMS, Issue 2 2001Andrea Borella Abstract A general method for designing multilayered WDM optical networks, based on the combined use of single-and multi-hop connection modes, is presented. It takes into account variable values for the number of users and wavelengths in each cluster. Closed form expressions are derived for the transmission capacity and the optimum number of channels for intra-cluster communications, either in the case of uniform or non-uniform traffic distribution. The analytical approach is particularly useful in the presence of constraints on the number of wavelengths, due to technological limits or non-linear phenomena, when sub-optimal solutions must be necessarily addressed. The proposed method is integrated with the adoption of well-known selection procedures, like simulated annealing or genetic algorithms, to reduce the computational effort. [source] Improved process monitoring using nonlinear principal component models,INTERNATIONAL JOURNAL OF INTELLIGENT SYSTEMS, Issue 5 2008David Antory This paper presents two new approaches for use in complete process monitoring. The first concerns the identification of nonlinear principal component models. This involves the application of linear principal component analysis (PCA), prior to the identification of a modified autoassociative neural network (AAN) as the required nonlinear PCA (NLPCA) model. The benefits are that (i) the number of the reduced set of linear principal components (PCs) is smaller than the number of recorded process variables, and (ii) the set of PCs is better conditioned as redundant information is removed. The result is a new set of input data for a modified neural representation, referred to as a T2T network. The T2T NLPCA model is then used for complete process monitoring, involving fault detection, identification and isolation. The second approach introduces a new variable reconstruction algorithm, developed from the T2T NLPCA model. Variable reconstruction can enhance the findings of the contribution charts still widely used in industry by reconstructing the outputs from faulty sensors to produce more accurate fault isolation. These ideas are illustrated using recorded industrial data relating to developing cracks in an industrial glass melter process. A comparison of linear and nonlinear models, together with the combined use of contribution charts and variable reconstruction, is presented. © 2008 Wiley Periodicals, Inc. Epidermal growth factor receptor (EGFR) is expressed at the cell surface by more than 90% of SCCs and its activation is responsible for cell cycle progression, proliferation, survival, angiogenesis and metastasis. Cyclooxygenase-2 (COX-2) is an enzyme up-regulated through EGFR signaling and responsible for some of the EGFR-dependent biological effects. An 88-year-old man presented with a recurrent, locoregionally meta-static SCC of the right parietal region, which was resistant to radiotherapy. With a combination therapy of an EGFR blocker (cetuximab) and a COX-2 inhibitor (celecoxib), the tumor regressed partially and the patient's Karnofsky index improved. We speculate that the combined use of cetuxi-mab and COX-2 inhibitors can be a new and effective therapy for advanced and recurrent cutaneous SCCs. The study commenced in 1994 and the main phase of data-gathering took place over a 3-week period in 1995. Having outlined the study's aim, context and theoretical background, this paper focuses on the two main methods of data-gathering used. Detailed description of the simulation method and the post-simulation interview and the rationales for their use are followed by critical discussion which identifies their particular strengths and weaknesses. Threats to validity are also considered. It is argued that the combined use of a simulated assessment and a post-simulation structured interview has great potential as a means of exploring the knowledge involved in community nursing assessment practice. However, in spite of the promising preclinical results, a first generation of antiangiogenic compounds yielded poor results in clinical trials. Conceptual errors and mistakes in the design of trials and in the definition of clinical end-points could account for these negative results. In this context of discouraging results, a second generation of antiangiogenic therapies is showing positive results in phases II and III trials at the beginning of the twenty-first century. In fact, several combined treatments with conventional chemotherapy and antiangiogenic compounds have been recently approved. The discovery and pharmacological development of future generations of angiogenesis inhibitors will benefit from further advances in the understanding of the mechanisms involved in human angiogenesis. New styles of trials are necessary, to avoid missing potential therapeutic effects. Different clinical end-points, new surrogate biomarkers and methods of imaging will be helpful in this process. Real efficacy in clinical trials may come with the combined use of antiangiogenic agents with conventional chemotherapy or radiotherapy, and combinations of several antiangiogenic compounds with different mechanisms of action. Paraskevas Abstract Aim: To examine the effect of amine fluoride/stannous fluoride (AmF/SnF2)-containing dentifrice and mouth rinse on plaque formation and gingivitis as compared with habitual oral hygiene procedures with a regular sodium fluoride (NaF) dentifrice. Material and Methods: In total, 22 general practices participated in this research project. The participants (N=281) were randomly assigned into two groups: the test group received an AmF/SnF2 dentifrice,mouth rinse combination and the control group received a NaF-containing dentifrice. The patients were requested to brush twice daily for approximately 2 min. The subjects of the test group had to rinse additionally in the evening for 30 s with 10 ml of the mouth rinse. Results: Both groups started with comparable scores of plaque, bleeding and staining. At 6 months, the plaque scores were 0.95 for the AmF/SnF2 group and 0.99 for the NaF group (decrease of 16% and 10%, respectively). Bleeding scores, although significantly different from baseline, did not show differences between the two regimes. At the end of the experimental period, the overall staining was more pronounced in the AmF/SnF2 group (41%) than the NaF group (26%). Both plaque reduction and increase in staining seemed to be correlated to the amount of mouth rinse used in the test group. Conclusion: In instruction-resistant patients recruited from dental practices, the combined use of AmF/SnF2 did not decrease gingivitis at a significant level in comparison with the regular regime of two times daily brushing with an NaF-containing dentifrice. However, the above-mentioned combination resulted in greater plaque reduction than that observed with the use of the conventional dentifrice. When used according to the manufacturer's instructions, this effect on plaque scores was more pronounced. [source] Effect of Piperine, a Major Component of Black Pepper, on the Intestinal Absorption of Fexofenadine and Its Implication on Food,Drug InteractionJOURNAL OF FOOD SCIENCE, Issue 3 2010Ming-Ji Jin ABSTRACT:, The present study aimed to investigate the effect of piperine, a major component of black pepper, on the oral exposure of fexofenadine in rats. Pharmacokinetic parameters of fexofenadine were determined in rats following an oral (10 mg/kg) or intravenous (5 mg/kg) administration of fexofenadine in the presence and absence of piperine (10 or 20 mg/kg, given orally). Compared to the control group given fexofenadine alone, the combined use of piperine increased the oral exposure (AUC) of fexofenadine by 180% to 190% while there was no significant change in,Cmax and,T1/2 of fexofenadine in rats. The bioavailability of fexofenadine was increased by approximately 2-folds via the concomitant use of piperine. Furthermore,,Tmax tends to be increased which might be attributed to the delayed gastric emptying in the presence of piperine. In contrast, piperine did not alter the intravenous pharmacokinetics of fexofenadine, implying that piperine may increase mainly the gastrointestinal absorption of fexofenadine rather than reducing hepatic extraction. In conclusion, piperine significantly enhanced the oral exposure of fexofenadine in rats likely by the inhibition of P-glycoprotein-mediated cellular efflux during the intestinal absorption, suggesting that the combined use of piperine or piperine-containing diet with fexofenadine may require close monitoring for potential drug,diet interactions. This paper reports specific methods using mass spectrometric techniques to localize protein modifications induced by lactose and galactose on ,-lactoglobulin (,-Lg) under solid-state glycation conditions. The extent of glycation was first determined by liquid chromatography/electrospray ionization mass spectrometry (LC/ESI-MS). The specific identification of lactose-modified amino acid residues was realized using both NanoESI-MS, NanoESI-MS/MS (neutral loss scanning modes) and matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOFMS) (with and without guanidination of lysine residues) on unfractionated digests. The results indicated that, after 8.25 h of incubation, the lysine residues were the main targets of lactose-induced modification. In addition to the 15 lysine residues, Leu1 (NH2 terminal) and the Arg124 were also found to be modified, thus leading to a total of 17 different modified amino acid residues (versus 15 found by LC/ESI-MS measurement). In a second set of experiments, different strategies consisting of constant neutral loss and precursor ion scanning were compared to characterize galactose-induced modifications. Owing to the high level of ,-Lg glycation, the combined use of these different strategies appeared to be necessary for determining the galactose-modified sites after 8.25 h of incubation. Thus, among the 22 galactose adducts deduced from the LC/ESI-MS measurement, apart from the N-terminal and classical lysine residues, we also observed a few arginine residues (Arg40, Arg124 and Arg148) that were modified, and also dialkylations on specific lysine residues (Lys47, Lys75). The water uptake can promote crystallographic modifications, according to the amount of water. In a previous work, the authors showed that a dihydrated form could be obtained either by crystallization in water or by exposure of the anhydrous form to a RH of 55%. In the present work, the authors report about the formation and characterization of a new tetrahydrated form, obtained by exposing the ASN to RH,,,75%. All the hydrated compounds were characterized by the combined use of several spectroscopic, thermal, and crystallographic techniques.
