Selected AbstractsSimulation of groundwater dynamics in the North China Plain by coupled hydrology and agricultural modelsHYDROLOGICAL PROCESSES, Issue 16 2006Tadanobu Nakayama Abstract We simulated the effects of irrigation on groundwater flow dynamics in the North China Plain by coupling the NIES Integrated Catchment-based Ecohydrology (NICE) model with DSSAT-wheat and DSSAT-maize, two agricultural models. This combined model (NICE-AGR) was applied to the Hai River catchment and the lower reach of the Yellow River (530 km wide by 840 km long) at a resolution of 5 km. It reproduced excellently the soil moisture, evapotranspiration and crop production of summer maize and winter wheat, correctly estimating crop water use. So, the spatial distribution of crop water use was reasonably estimated at daily steps in the simulation area. In particular, NICE-AGR reproduced groundwater levels better than the use of statistical water use data. This indicates that NICE-AGR does not need detailed statistical data on water use, making it very powerful for evaluating and estimating the water dynamics of catchments with little statistical data on seasonal water use. Furthermore, the simulation reproduced the spatial distribution of groundwater level in 1987 and 1988 in the Hebei Plain, showing a major reduction of groundwater level due mainly to overpumping for irrigation. [source] Welfare-improving adverse selection in credit markets,INTERNATIONAL ECONOMIC REVIEW, Issue 4 2002James Vercammen A model of simultaneous adverse selection and moral hazard in a competitive credit market is developed and used to show that aggregate borrower welfare may be higher in the combined case than in the moral-hazard-only case. Adverse selection can be welfare improving because in the pooling equilibrium of the combined model, high-quality borrowers cross subsidize low-quality borrowers. The cross subsidization reduces the overall moral hazard effort effects, and the resulting gain in welfare may more than offset the welfare loss stemming from distorted investment choices. The analysis focuses on pooling equilibria because model structure precludes separating equilibria. [source] Assessment of the 10-Year Probability of Osteoporotic Hip Fracture Combining Clinical Risk Factors and Heel Bone Ultrasound: The EPISEM Prospective Cohort of 12,958 Elderly Women,,JOURNAL OF BONE AND MINERAL RESEARCH, Issue 7 2008Didier Hans Abstract This study aimed to develop a hip screening tool that combines relevant clinical risk factors (CRFs) and quantitative ultrasound (QUS) at the heel to determine the 10-yr probability of hip fractures in elderly women. The EPISEM database, comprised of ,13,000 women ,70 yr of age, was derived from two population-based white European cohorts in France and Switzerland. All women had baseline data on CRFs and a baseline measurement of the stiffness index (SI) derived from QUS at the heel. Women were followed prospectively to identify incident fractures. Multivariate analysis was performed to determine the CRFs that contributed significantly to hip fracture risk, and these were used to generate a CRF score. Gradients of risk (GR; RR/SD change) and areas under receiver operating characteristic curves (AUC) were calculated for the CRF score, SI, and a score combining both. The 10-yr probability of hip fracture was computed for the combined model. Three hundred seven hip fractures were observed over a mean follow-up of 3.2 yr. In addition to SI, significant CRFs for hip fracture were body mass index (BMI), history of fracture, an impaired chair test, history of a recent fall, current cigarette smoking, and diabetes mellitus. The average GR for hip fracture was 2.10 per SD with the combined SI + CRF score compared with a GR of 1.77 with SI alone and of 1.52 with the CRF score alone. Thus, the use of CRFs enhanced the predictive value of SI alone. For example, in a woman 80 yr of age, the presence of two to four CRFs increased the probability of hip fracture from 16.9% to 26.6% and from 52.6% to 70.5% for SI Z-scores of +2 and ,3, respectively. The combined use of CRFs and QUS SI is a promising tool to assess hip fracture probability in elderly women, especially when access to DXA is limited. Sandy Pang The administration of metabolites arising from new drug entities is often employed in drug discovery to investigate their associated toxicity. It is expected that administration of metabolites can predict the exposure of metabolites originating from the administration of precursor drug. Whether exact and meaningful information can be obtained from this has been a topic of debate. This communication summarizes observations and theoretical relationships based on physiological modelling for the liver, kidney and intestine, three major eliminating organs/tissues. Theoretical solutions based on physiological modelling of organs were solved, and the results suggest that deviations are expected. Here, examples of metabolite kinetics observed mostly in perfused organs that did not match predictions are provided. For the liver, discrepancies in fate between formed and preformed metabolites may be explained by the heterogeneity of enzymes, the presence of membrane barriers and whether transporters are involved. For the kidney, differences have been attributed to glomerular filtration of the preformed but not the formed metabolite. For the intestine, the complexity of segregated flows to the enterocyte and serosal layers and differences in metabolism due to the route of administration are addressed. Administration of the metabolite may or may not directly reflect the toxicity associated with drug use. However, kinetic data on the preformed metabolite will be extremely useful to develop a sound model for modelling and simulations; in-vitro evidence on metabolite handling at the target organ is also paramount. Subsequent modelling and simulation of metabolite data arising from a combined model based on both drug and preformed metabolite data are needed to improve predictions on the behaviours of formed metabolites. [source] Heritability of diurnal type: a nationwide study of 8753 adult twin pairsJOURNAL OF SLEEP RESEARCH, Issue 2 2007MARKKU KOSKENVUO Summary Twin studies suggest a genetic component in diurnal types. In 1981, a questionnaire sent to the Older Finnish Twin Cohort yielded responses from 2836 adult monozygotic (MZ) and 5917 like-sexed dizygotic (DZ) twin pairs with four category self-report on diurnal type. We used structural equation modelling to estimate genetic and environmental components of variance in morningness and eveningness. The model fitting was best when the morningness and the eveningness were analysed together. The ADE-model (including additive genetic, dominant genetic and non-shared environmental effects) fitted best to the data. ADE-models for men and women separately did not differ in a statistically significant manner from the combined model, and similarly ADE-models for young and old age groups separately did not differ either. The estimate for overall genetic effect (broad sense heritability) was 49.7% (95% confidence interval 46.4,52.8), with the remainder accounted for by environmental factors not shared by siblings. The variance component estimates for the underlying liability to diurnal type were 11.7% (95% CI 0,23.7) for additive genetic factors, 38.0% (24.7,51.3) for genetic factors due to dominance. Genetic effects thus account for about one-half of the interindividual variability in diurnal type in adults. [source] Use of Topological Indices of Organic Sulfur Compounds in Quantitative Structure-Retention Relationship StudyMOLECULAR INFORMATICS, Issue 9 2005F. Safa Abstract Structure-gas chromatographic retention index models were developed for some organic sulfur compounds at four different temperatures (60, 80, 100 and 120,°C) using only topological descriptors. At first, regression models were generated for each temperature separately with high values of multiple correlation coefficient and Fisher-ratio statistics. The results of cross validation test using leave-one-out (Q2,0.956) and leave-two-out (Q2,0.953) methods showed good predictive ability of the models developed. Then, a single combined quantitative structure-retention relationship model, added temperature as a parameter, was also developed for all the temperatures, showing good statistical parameters (R=0.991 and F=728.474). The stability and validity of the combined model were verified by both internal (Q2>0.970) and external validation (Q=0.993) techniques. The results of the study indicated the efficiency of the classical topological descriptors in simultaneous prediction of retention index values of sulfur compounds at different temperatures. The topological descriptors well covered the molecular properties known to be relevant for gas chromatographic retention data, such as molecular size and degree of branching. Gray ABSTRACT Observations show that 1612-MHz masers of OH/IR stars can fade on a time-scale of a decade. This fading is probably associated with the switch from rapid mass loss, which is ultimately linked with an internal He-shell flash, to the much slower mass loss supported by more quiescent conditions. We study the observed maser decay with a composite computational model, comprising a time-dependent chemical model of the envelope, and a radiation transfer model which provides the maser pumping. Our combined model is able to reproduce the rapid decay of maser intensity, following a sudden drop in the stellar mass-loss rate. The explanation for the rapid fall in maser emission is not a fall in the OH number density, or the kinetic temperature in the inverted layers, but the loss of a radiative pump route which carries population from levels 1 to 4 via levels 16, 15 and 11. The loss of these pump routes is a result, in turn, of a greatly reduced energy density of 53-,m radiation. KOSUGI ABSTRACT A combined model to simulate CO2 and H2O gas exchange at the leaf scale was parameterized using data obtained from in situ leaf-scale observations of diurnal and seasonal changes in the CO2 and H2O gas exchange of four temperate deciduous broad-leaved trees using a porometric method. The model consists of a Ball et al. type stomatal conductance submodel [Ball, Woodrow & Berry, pp. 221,224 in Progress in Photosynthesis Research (ed. I. Biggins), Martinus-Nijhoff Publishers, Dordrecht, The Netherlands, 1987] and a Farquhar et al. type biochemical submodel of photosynthesis (Farquhar, von Caemmerer & Berry, Planta 149, 78,90, 1980). In these submodels, several parameters were optimized for each tree species as representative of the quantitative characteristics related to gas exchange. The results show that the seasonal physiological changes of Vcmax25 in the biochemical model of photosynthesis should be used to estimate the long-term CO2 gas exchange. For Rd25 in the biochemical model of photosynthesis and m in the Ball et al. type stomatal conductance model, the difference should be counted during the leaf expansion period. [source] A Holistic Simulation Approach from a Measured Load to Element Stress Using Combined Multi-body Simulation and Finite Element ModellingPROCEEDINGS IN APPLIED MATHEMATICS & MECHANICS, Issue 1 2009Matthias Harter The design of vehicle bodies requires the knowledge of the vehicle's structural response to external loads and disturbances. In rigid multi-body simulation the dynamic behaviour of complex systems is calculated with rigid bodies and neglect of body elasticity. On the other hand, in finite element models large degree of freedom numbers are used to represent the elastic properties of a single body. Both simulation methods can be combined, if the finite element model size is reduced to a degree of freedom number feasible to multi-body simulation. The application to practical purposes requires the use and interconnection of several different software tools. In this contribution a holistic method is presented, which starts with the measurement or synthesis of loads and excitations, continues with the integration of a reduced finite element model into a multi-body system, the dynamic response calculation of this combined model, and concludes with the result expansion to the full finite element model for calculating strain and stress values at any point of the finite element mesh. Clapp We model competing risks of mortgage termination where the borrower faces a repeated choice to continue to pay, refinance the loan, move or default. Most previous empirical work on mortgage prepayment has ignored the distinction between prepayments triggered by refinancing and moving, combining them into a single prepayment rate. We show that financial considerations are the primary drivers of the refinance choice while homeowner characteristics have more influence on the move decision. We demonstrate that these differences are statistically significant and that combining these two distinct choices into a single measure of prepayment shifts coefficients toward zero and produces inaccurate predictions of aggregate termination rates. For example, a combined model underestimates the effect of the market price of the loan on refinancing; it misses entirely the opposite effects of borrower income on moving and refinancing. Our results suggest that existing prepayment models are inconsistent predictors of mobility-driven prepayment and underestimate the effect of market conditions and borrower characteristics on refinancing and housing decisions. Our findings have great significance to mortgage investors because mobility-driven prepayments are likely to be a more significant source of prepayments in thenext decade. WHAT THIS STUDY ADDS , The data on aripiprazole and dehydroaripiprazole in psychiatric patients were modelled jointly using a population approach. , The apparent clearance of aripiprazole in cytochrome P450 (CYP) 2D6 intermediate metabolizers (IM) was approximately 60% of that in CYP2D6 extensive metabolizers (EM) having two functional alleles, but the exposure to dehydroaripiprazole in CYP2D6 IM was similar to that in EM. AIMS The aims of this study were to develop a combined population pharmacokinetic model for both aripiprazole and its active metabolite, dehydroaripiprazole, in psychiatric patients and to identify to what extent the genetic polymorphisms of cytochrome P450 (CYP) enzymes contribute to the variability in pharmacokinetics (PK). METHODS A population pharmacokinetic analysis was performed using NONMEM software based on 141 plasma concentrations at steady state from 80 patients receiving multiple oral doses of aripiprazole (10,30 mg day,1). RESULTS A one-compartment model with first-order kinetics for aripiprazole and dehydroaripiprazole each was developed to describe simultaneously the concentration data. The absorption rate constant was fixed to 1.06 h,1. The typical value of apparent distribution volume of aripiprazole was estimated to be 192 l. Covariate analysis showed that CYP2D6 genetic polymorphisms significantly influenced the apparent clearance of aripiprazole (CL/F), reducing the interindividual variability on CL/F from 37.8% CV (coefficient of variation) to 30.5%. The CL/F in the CYP2D6 IMs was approximately 60% of that in CYP2D6 EMs having two functional alleles. Based on the CYP2D6 genotype, the metabolic ratios were calculated at 0.20,0.34. However, the plasma concentration : dose ratios of dehydroaripiprazole were not different across the CYP2D6 genotype. CONCLUSIONS This population pharmacokinetic model provided an adequate fit to the data for both aripiprazole and dehydroaripiprazole in psychiatric patients. The usefulness of CYP genotyping as an aid to select the starting dose should be further investigated. [source] |
