Geminal Ionic Liquids: A Combined Approach to Investigate Their Three-Dimensional Organisation

Abstract Different investigations, such as 1D and 2D NMR spectroscopy, resonance light scattering spectroscopy and molecular dynamics simulations, have been jointly used to achieve a deeper understanding of the degree of structural order in two geminal ionic liquids. In particular, 3,3,-di- n -butyl-1,1,-(1,3-phenylenedimethylene)diimidazolium and 3,3,-di- n -butyl-1,1,-(1,4-phenylenedimethylene)diimidazolium bis[bis(trifluoromethanesulfonyl)imide] have been studied. These geminal ionic liquids were chosen because of the presence of both a rigid phenylenedimethylene link between two imidazolium rings, which should give a high degree of order to the solvent system, and the different shapes of the two cations of the isomers, which could induce different properties and packing in the liquid state. Data collected here show that the two geminal ionic liquids are characterised by a different degree of structural order that induces, for example, a different sensitivity of the two solvent systems to temperature changes or to the presence of a co-solvent such as methanol. Thermodynamic Analysis of Energy Transfer in Acidogenic Cultures

Abstract A global thermodynamic analysis, normally used for pure cultures, has been performed for steady-state data sets from acidogenic mixed cultures. This analysis is a combination of two different thermodynamic approaches, based on tabulated standard Gibbs energy of formation, global stoichiometry and medium compositions. It takes into account the energy transfer efficiency, ,, together with the Gibbs free energy dissipation, ,Go, analysis of the different data. The objective is to describe these systems thermodynamically without any heat measurement. The results show that , is influenced by environmental conditions, where increasing hydraulic retention time increases its value all cases. The pH effect on , is related to metabolic shifts and osmoregulation. Within the environmental conditions analyzed, , ranges from 0.23 for a hydraulic retention time of 20,h and pH,4, to 0.42 for a hydraulic retention time of 8,h and a pH ranging from 7,8.5. The estimated values of ,Go are comparable to standard Gibbs energy of dissipation reported in the literature. For the data sets analyzed, ,Go ranges from ,1210,kJ/molx, corresponding to a stirring velocity of 300,rpm, pH,6 and a hydraulic retention time of 6,h, to ,20744,kJ/molx for pH,4 and a hydraulic retention time of 20,h. For average conclusions, the combined approach based on standard Gibbs energy of formation and global stoichiometry, used in this thermodynamic analysis, allows for the estimation of Gibbs energy dissipation values from the extracellular medium compositions in acidogenic mixed cultures. Such estimated values are comparable to the standard Gibbs energy dissipation values reported in the literature. It is demonstrated that , is affected by the environmental conditions, i.e., stirring velocity, hydraulic retention time and pH. However, a relationship that relates this parameter to environmental conditions was not found and will be the focus of further research. [source]


What Females Tell Males About Their Reproductive Status: Are Morphological and Behavioural Cues Reliable Signals of Ovulation in Bonobos (Pan paniscus)?

In many Old World primate species, female attractivity increases during the tumescent phase of the sexual swelling for a period that lasts considerably longer than oestrus-related attractivity in other mammals. We examined the reliability of the swelling as an indicator of ovulation in captive bonobos, a species with a long and variable phase of maximum tumescence. Using a combined approach of (1) observations of sexual behaviour, (2) visual scoring of the sexual swelling and (3) analysis of faecal progestin to assess the timing of ovulation during 23 ovulatory cycles of eight adult females, we found that in 30% of these cycles the presumed day of ovulation did not fall within the period of maximum tumescence. When ovulation did occur during maximum swelling, it was more closely related to the end rather than the onset of the maximum swelling period. However, the pattern of sexual swelling was not a reliable indicator of ovulation. In addition, sexual behaviour of both sexes increased in frequency with the degree of the swelling but not around the time of ovulation. We conclude that swellings in bonobos provide honest information on the probability of ovulation, but not its exact timing, and that therefore the `obvious ovulation'-hypothesis cannot explain the function of sexual swellings in bonobos. Immunolocalization of BK channels in hippocampal pyramidal neurons

Abstract Neurons are highly specialized cells in which the integration and processing of electrical signals critically depends on the precise localization of ion channels. For large-conductance Ca2+ - activated K+ (BK) channels, targeting to presynaptic membranes in hippocampal pyramidal cells was reported; however, functional evidence also suggests a somatodendritic localization. Therefore we re-examined the subcellular distribution of BK channels in mouse hippocampus using a panel of independent antibodies in a combined approach of conventional immunocytochemistry on cultured neurons, pre- and postembedding electron microscopy and immunoprecipitation. In cultured murine hippocampal neurons, the colocalization of BK channels with both pre- and postsynaptic marker proteins was observed. Electron microscopy confirmed targeting of BK channels to axonal as well as dendritic membranes of glutamatergic synapses in hippocampus. A postsynaptic localization of BK channels was also supported by the finding that the channel coimmunoprecipitated with PSD95, a protein solely expressed in the postsynaptic compartment. These results thus demonstrate that BK channels reside in both post- and presynaptic compartments of hippocampal pyramidal neurons. [source]


Simulation of Welding and Distortion in Ship Building,

Simulation tools will continue to gain importance for both scientific investigations and industrial applications. This further applies to welding technology. The present work focuses on the simulation of distortions due to the welding of stiffeners on heavy plates in shipbuilding. An equivalent heat source (EHS) was computed by the software SimWeld to describe the energy input caused by welding. The EHS was combined with an FEM simulation of the global structure in the commercial program SYSWELD. Therefore an interface between the SimWeld platform and SYSWELD was implemented. The simulation results were compared with a welded demonstrator. The predicted displacements correlate closely with the experimental data. Using the combined approach the quality of the prediction was significantly improved against the typical method of heat source parameter identification, which is based on experimental results. The results allow for the optimization of welding sequences and for the minimization of buckling in shipbuilding. [source]


Molecular determinants of ligand specificity in family 11 carbohydrate binding modules , an NMR, X-ray crystallography and computational chemistry approach

The direct conversion of plant cell wall polysaccharides into soluble sugars is one of the most important reactions on earth, and is performed by certain microorganisms such as Clostridium thermocellum (Ct). These organisms produce extracellular multi-subunit complexes (i.e. cellulosomes) comprising a consortium of enzymes, which contain noncatalytic carbohydrate-binding modules (CBM) that increase the activity of the catalytic module. In the present study, we describe a combined approach by X-ray crystallography, NMR and computational chemistry that aimed to gain further insight into the binding mode of different carbohydrates (cellobiose, cellotetraose and cellohexaose) to the binding pocket of the family 11 CBM. The crystal structure of C. thermocellum CBM11 has been resolved to 1.98 Å in the apo form. Since the structure with a bound substrate could not be obtained, computational studies with cellobiose, cellotetraose and cellohexaose were carried out to determine the molecular recognition of glucose polymers by CtCBM11. These studies revealed a specificity area at the CtCBM11 binding cleft, which is lined with several aspartate residues. In addition, a cluster of aromatic residues was found to be important for guiding and packing of the polysaccharide. The binding cleft of CtCBM11 interacts more strongly with the central glucose units of cellotetraose and cellohexaose, mainly through interactions with the sugar units at positions 2 and 6. This model of binding is supported by saturation transfer difference NMR experiments and linebroadening NMR studies. [source]


Customized Electronic Coupling in Self-Assembled Donor,Acceptor Nanostructures

Abstract Charge transfer processes between donor,acceptor complexes and metallic electrodes are at the heart of novel organic optoelectronic devices such as solar cells. Here, a combined approach of surface-sensitive microscopy, synchrotron radiation spectroscopy, and state-of-the-art ab initio calculations is used to demonstrate the delicate balance that exists between intermolecular and molecule,substrate interactions, hybridization, and charge transfer in model donor,acceptor assemblies at metal-organic interfaces. It is shown that charge transfer and chemical properties of interfaces based on single component layers cannot be naively extrapolated to binary donor,acceptor assemblies. In particular, studying the self-assembly of supramolecular nanostructures on Cu(111), composed of fluorinated copper-phthalocyanines (F16CuPc) and diindenoperylene (DIP), it is found that, in reference to the associated single component layers, the donor (DIP) decouples electronically from the metal surface, while the acceptor (F16CuPc) suffers strong hybridization with the substrate. Imbalances of chromosome arm 1p in pediatric and adult germ cell tumors are caused by true allelic loss: A combined comparative genomic hybridization and microsatellite analysis

Previous studies on childhood germ cell tumors (GCTs) report highly variable frequencies of losses at chromosome arm 1p. Since deletions at 1p portend a poor prognosis in other embryonal tumors, this study aims to clarify the question of the frequency of true allelic loss at 1p and whether it constitutes a prognostic parameter. We analyzed 13 GCTs from different gonadal and extragonadal sites of children (4 teratomas, 9 malignant GCTs) and 18 GCTs of adolescents and adults (3 teratomas; 15 malignant GCTs) using automated microsatellite analysis with 23 polymorphic markers and chromosomal "high resolution" comparative genomic hybridization (HR-CGH). With this combined approach, we detected loss of heterozygosity (LOH) at 1p in 8/9 childhood malignant GCTs with concordant data from HR-CGH and microsatellite analyses. In contrast, LOH at 1p was not detected in childhood teratomas (0/4) and constituted a rare event in GCTs of adolescence and adulthood (3/18). The commonly deleted region was located at distal 1p36-pter, with a proximal boundary between the markers D1S450 and D1S2870. These data unequivocally demonstrate that deletion at 1p is common in childhood GCTs and results in allelic loss. This observation argues for the presence of a classical tumor suppressor at distal 1p. Considering the high frequency of LOH at 1p and the overall favorable prognosis of childhood GCTs, a prognostic impact of LOH at 1p in childhood GCTs appears unlikely. However, since two postpubertal tumors with LOH at 1p progressed, a prognostic relevance in this age group seems possible, warranting a prospective evaluation. © 2006 Wiley-Liss, Inc. [source]


Patterns and Determinants of Historical Woodland Clearing in Central-Western New South Wales, Australia

Abstract We consider the history of woodland clearing in central western New South Wales, Australia, which has led to the present highly cleared and fragmented landscape. A combined approach is used examining available historical land-use data and using regression analysis to relate the pattern of cleared and wooded areas in the recent landscape to environmental variables, taking into account the contagious nature of clearing. We also ask whether it would be possible to apply a simple simulation modelling approach to reconstruct a credible historical sequence of clearing in the study area. The historical data indicate that annual clearing rates have varied substantially in the study area and selective tree removal (ringbarking and thinning) has been common. These findings make it unlikely that a simple simulation approach would replicate the spatial and temporal sequence of woodland loss. Our regression results show that clearing patterns can be related to environmental variables, particularly annual rainfall and estimated pre-European vegetation type, but that patterns are dominated by contagion. Combined homology modelling and evolutionary significance evaluation of missense mutations in blood clotting factor VIII to highlight aspects of structure and function

Summary., Most small lesions in the factor VIII (FVIII) gene that cause haemophilia A (HA) are single nucleotide substitutions resulting in amino acid replacing (missense) mutations and leading to various phenotypes, ranging from mild to severe. We took a combined approach of homology modelling and quantitative evaluation of evolutionary significance of amino acid replacing alterations using the Grantham Matrix Score (GMS) to assess their structural effects and significance of pathological expression. Comparative homology models of all amino acid substitutions summarized in the FVIII mutations database plus these identified and reported lately by us or by our collaborators were evaluated. Altogether 640 amino acid replacing mutations were scored for potential distant or local conformation changes, influence on the molecular stability and predicted contact residues, using available FVIII domain models. The average propensity to substitute amino acid residues by mutation was found comparable to the overall probability of de novo mutations. Missense changes reported with various HA phenotypes were all confirmed significant using GMS. The fraction of these, comprising residues apparently involved in intermolecular interactions, exceeds the average proportion of such residues for FVIII. Predicted contact residues changed through mutation were visualized on the surface of FVIII domains and their possible functional implications were verified from the literature and are discussed considering available structural information. Our predictive modelling adds on the current view of domain interface molecular contacts. This structural insight could aid in part to the design of engineered FVIII constructs for therapy, to possibly enhance their stability and prolong circulating lifetime. [source]


Mutation analysis in nephronophthisis using a combined approach of homozygosity mapping, CEL I endonuclease cleavage, and direct sequencing,

Abstract Nephronophthisis (NPHP), an autosomal recessive kidney disease, is the most frequent genetic cause of chronic renal failure in the first three decades of life. Mutations in eight genes (NPHP1,8) have been identified. We here describe a combined approach for mutation screening of NPHP1, NPHP2, NPHP3, NPHP4, and NPHP5 in a worldwide cohort of 470 unrelated patients with NPHP. First, homozygous NPHP1 deletions were detected in 97 patients (21%) by multiplex PCR. Second, 25 patients with infantile NPHP were screened for mutations in inversin (NPHP2/INVS). We detected a novel compound heterozygous frameshift mutation (p.[Q485fs]+[R687fs]), and a homozygous nonsense mutation (p.R899X). Third, 37 patients presenting with NPHP and retinitis pigmentosa (Senior-Løken syndrome [SLS]) were screened for NPHP5/IQCB1 mutations by direct sequencing. We discovered five different (three novel) homozygous premature termination codon (PTC) mutations (p.F142fsX; p.R461X; p.R489X; p.W444X; and c.488,1G>A). The remaining 366 patients were further investigated for mutations in NPHP1, NPHP3, and NPHP4. We applied a "homozygosity only" strategy and typed three highly polymorphic microsatellite markers at the respective loci. A total of 32, eight, and 14 patients showed homozygosity, and were screened by heteroduplex crude celery extract (CEL I) endonuclease digests. The sensitivity of CEL I was established as 92%, as it detected 73 out of 79 different known mutations simply on agarose gels. A total of 10 novel PTC mutations were found in NPHP1 (p.P186fs, p.R347X, p.V492fs, p.Y509X, and c.1884+1G>A), in NPHP3 (c.3812+2T>C and p.R1259X), and in NPHP4 (p.R59X, p.T1004fs, and p.V1091fs). Karakorum,Hindukush,western Himalaya: assessing high-altitude water resources

Abstract The high mountains of Central and South Asia provide irrigation water for their adjacent lowlands. The Indus Irrigation Scheme depends on approximately 50% of its runoff originating from snowmelt and glacier melt from the eastern Hindukush, Karakorum and western Himalaya. The Atlas of Pakistan indicates that these mountains gain a total annual rainfall of between 200 and 500 mm, amounts that are generally derived from valley-based stations and not representative for elevated zones. High-altitude snowfall seems to be neglected and is obviously still rather unknown. Estimates derived from accumulation pits runoff above 4000 m range from 1000 mm to more than 3000 mm, depending on the site and time of investigation, as well as on the method applied. To assess the vertical spatio-temporal distribution of total annual precipitation, a combined approach is presented. This approach links in situ measurements of snow depth and water equivalent (10-year time series derived from automatic weather stations at elevations between 1500 and 4700 m a.s.l.), the spatial distribution and period of snow coverage (remotely sensed data and digital elevation models), and the runoff characteristics of streams originating from snow or snow/ice-covered watersheds (modified snowmelt runoff model, including intermediate snowfall and glacier runoff). Based on conservative assumptions, the vertically changing seasonal ratio between liquid and solid precipitation is calculated. Using a combined snow cover and ablation model, total annual amounts of precipitation are derived for different altitudinal zones. Amounts of modelled and measured runoff complement the investigation. Horizontal gradients along the Indus,Gilgit,Hunza transect indicate the varying dominance of seasonal precipitation regimes (monsoonal, Mediterranean and continental disturbances) south of Nanga Parbat, between Nanga Parbat and Batura Wall (=West Karakorum rainfall regime: 1500,1800 mm year,1 at 5000 m) and areas north of Batura (=Central Asian rainfall regime: ,600 mm year,1 at 5000 m). A numerical,analytical combined method for vibration of a beam excited by a moving flexible body

Abstract The vibration of a beam excited by a moving simple oscillator has been extensively studied. However, the vibration of a beam excited by an elastic body with conformal contact has attracted much less attention. This is the subject of the present paper. The established model is more complicated but has a much wider range of applications than the moving-oscillator model. Because the moving body is flexible, the moving loads at the contact interface are not known a priori and must be determined together with the dynamics of the whole system. In this paper, the equation of motion of the beam and the moving body are established separately using a numerical,analytical combined approach. It is found from the numerical results of the simulated example that the vibrations of the moving body and the beam excited by the moving body are significantly influenced by the travelling speed. At very low or very high speeds the dynamic effect is small and the beam deforms to take the shape of its static deflection. Vibrations tend to be greater in the intermediate speed range and the total moving force at the interface of the beam and the moving body can be compressive and tensile. A fast implementation of the FETI-DP method: FETI-DP-RBS-LNA and applications on large scale problems with localized non-linearities

Abstract As parallel and distributed computing gradually becomes the computing standard for large scale problems, the domain decomposition method (DD) has received growing attention since it provides a natural basis for splitting a large problem into many small problems, which can be submitted to individual computing nodes and processed in a parallel fashion. This approach not only provides a method to solve large scale problems that are not solvable on a single computer by using direct sparse solvers but also gives a flexible solution to deal with large scale problems with localized non-linearities. When some parts of the structure are modified, only the corresponding subdomains and the interface equation that connects all the subdomains need to be recomputed. In this paper, the dual,primal finite element tearing and interconnecting method (FETI-DP) is carefully investigated, and a reduced back-substitution (RBS) algorithm is proposed to accelerate the time-consuming preconditioned conjugate gradient (PCG) iterations involved in the interface problems. Linear,non-linear analysis (LNA) is also adopted for large scale problems with localized non-linearities based on subdomain linear,non-linear identification criteria. This combined approach is named as the FETI-DP-RBS-LNA algorithm and demonstrated on the mechanical analyses of a welding problem. Serial CPU costs of this algorithm are measured at each solution stage and compared with that from the IBM Watson direct sparse solver and the FETI-DP method. The results demonstrate the effectiveness of the proposed computational approach for simulating welding problems, which is representative of a large class of three-dimensional large scale problems with localized non-linearities. Use of vibrational spectroscopy to study protein and DNA structure, hydration, and binding of biomolecules: A combined theoretical and experimental approach

Abstract We report on our work with vibrational absorption, vibrational circular dichroism, Raman scattering, Raman optical activity, and surface-enhanced Raman spectroscopy to study protein and DNA structure, hydration, and the binding of ligands, drugs, pesticides, or herbicides via a combined theoretical and experimental approach. The systems we have studied systematically are the amino acids (L -alanine, L -tryptophan, and L -histidine), peptides (N -4271 acetyl L -alanine N,-methyl amide, N -acetyl L -tryptophan N,-methyl amide, N -acetyl L -histidine N,-methyl amide, L -alanyl L -alanine, tri- L -serine, N -acetyl L -alanine L -proline L -tyrosine N,-methyl amide, Leu-enkephalin, cyclo-(gly- L -pro)3, N -acetyl (L -alanine)nN,-methyl amide), 3-methyl indole, and a variety of small molecules (dichlobenil and 2,6-dochlorobenzamide) of relevance to the protein systems under study. We have used molecular mechanics, the SCC-DFTB, SCC-DFTB+disp, RHF, MP2, and DFT methodologies for the modeling studies with the goal of interpreting the experimentally measured vibrational spectra for these molecules to the greatest extent possible and to use this combined approach to understand the structure, function, and electronic properties of these molecules in their various environments. The application of these spectroscopies to biophysical and environmental assays is expanding, and therefore a thorough understanding of the phenomenon from a rigorous theoretical basis is required. In addition, we give some exciting and new preliminary results which allow us to extend our methods to even larger and more complex systems. Automatic mining of the literature to generate new hypotheses for the possible link between periodontitis and atherosclerosis: lipopolysaccharide as a case study

Abstract Aim: The aim of the current report was to generate and explore new hypotheses into how, in a pathophysiological sense, atherosclerosis and periodontitis could be linked. Material and Methods: Two different biomedical informatics techniques were used: an association-based technique that generated a ranked list of genes associated with the diseases, and a natural language processing tool that extracted the relationships between the retrieved genes and lipopolysaccharide (LPS). Results: This combined approach of association-based and natural language processing-based literature mining identified a hit list of 16 candidate genes, with PON1 as the primary candidate. Conclusions: Further study of the literature prompted the hypothesis that PON1 might connect periodontitis with atherosclerosis in both an LPS-dependent and a non-LPS-dependent manner. Furthermore, the resulting genes not only confirmed already known associations between the two diseases, but also provided genes or gene products that have only been investigated separately in the two disease states, and genes or gene products previously reported to be involved in atherosclerosis. These findings remain to be investigated through clinical studies. This example of multidisciplinary research illustrates how collaborative efforts of investigators from different fields of expertise can result in the discovery of new hypotheses. [source]


Sperm competition and maternal effects differentially influence testis and sperm size in Callosobruchus maculatus

Abstract The evolutionary factors affecting testis size are well documented, with sperm competition being of major importance. However, the factors affecting sperm length are not well understood; there are no clear theoretical predictions and the empirical evidence is inconsistent. Recently, maternal effects have been implicated in sperm length variation, a finding that may offer insights into its evolution. We investigated potential proximate and microevolutionary factors influencing testis and sperm size in the bruchid beetle Callosobruchus maculatus using a combined approach of an artificial evolution experiment over 90 generations and an environmental effects study. We found that while polyandry seems to select for larger testes, it had no detectable effect on sperm length. Furthermore, population density, a proximate indicator of sperm competition risk, was not significantly associated with sperm length or testis size variation. However, there were strong maternal effects influencing sperm length. [source]

Abstract Background The descriptive phenomenological categorical psychiatric diagnostic systems that are currently being used in the field of intellectual disability do not adequately provide for the special needs of persons with intellectual disability. Many relevant diagnostic questions are left unanswered or are only partially accounted for. This is particularly true for persons with low developmental levels. Method A solution to these stumbling blocks is sought in enhancing the contemporary categorical diagnostic systems by also applying methods derived from the developmental perspective. Result By taking the levels of emotional and personality development, in addition to other developmental aspects into account, the clinical picture becomes more comprehensible and explainable. Conclusion The integrative diagnosis that results from this combined approach provides an insight into the processes that have led to the disorder and enriches one's understanding of the presentation form of the disorder. This diagnosis is process- rather than symptom-oriented and is particularly useful with persons who have a low level of psychosocial development. [source]


Scintigraphic study to investigate the effect of food on a HPMC modified release formulation of UK-294,315

Abstract The objective of the study was to use the combined approach of gamma scintigraphy and pharmacokinetics, in order to understand the mechanisms explaining the pharmacokinetic differences observed for a modified release (MR) formulation, when administered either in the fed or fasted state. Ten healthy subjects were recruited into a randomized three period single dose study, each subject receiving UK-294,315 40 mg IR (fasted), 100 mg MR (fasted) or 100 mg MR (after a high fat meal). Cmax values were markedly higher for the MR tablet in the fed state versus fasted and mean residence time was about 3 h longer for fasted versus fed; there was little difference in apparent oral clearance. In the fasted state, average gastric emptying of the intact tablet occurred at 1.2 h postdose, with gastric emptying of intact tablet observed in all subjects. In the fed state, rapid disintegration of the MR tablet was observed by scintigraphy, with 7/9 subjects showing complete disintegration in the stomach. Complete disintegration occurred 10.1 h postdose in the fasted state versus 5.9 h after a high fat meal. Fluorescence background suppression in Raman spectroscopy using combined Kerr gated and shifted excitation Raman difference techniques

An exceptionally high level of fluorescence rejection from resonance Raman spectra was achieved using a combination of two techniques, namely Kerr gated temporal rejection with shifted excitation Raman difference spectroscopy. The method was able to recover the resonance Raman spectrum from the intense fluorescence background with a signal-to-noise ratio at least 10 times higher than that achievable with either of the two approaches used individually. To demonstrate the effectiveness of the technique we obtained the resonance Raman spectrum of the laser dye rhodamine 6G (1 × 10,3 mol dm,3) in methanol by excitation at 532 nm and measuring under the maximum of fluorescence emission at 560,590 nm. The method reached the photon shot noise limit of the residual fluorescence providing a detection limit for Raman spectra 106 times lower than the original fluorescence intensity in an accumulation time of 800 s. A unique feature of the experiment was the way in which the optical parametric amplifier light source was configured to alternate automatically between the two excitation wavelengths using an optogalvanic mirror arrangement. The ultra-high sensitivity of the combined approach holds great promise for selective probing of complex biological systems using resonance Raman spectroscopy. Consensus statement on the loco-regional treatment of appendiceal mucinous neoplasms with peritoneal dissemination (pseudomyxoma peritonei),

Abstract Pseudomyxoma peritonei (PMP) is a rare condition mostly originating from low malignant potential mucinous tumours of the appendix. Although this disease process is minimally invasive and rarely causes haematogenous or lymphatic metastases, expectation of long-term survival are limited with no prospect of cure. Recently, the combined approach of cytoreductive surgery (CRS) and perioperative loco-regional chemotherapy (PLC) has been proposed as the standard of treatment for the disease. The present paper summarizes the available literature data and the main features of the comprehensive loco-regional treatment of PMP. The controversial issues concerning the indications and technical methodology in PMP management were discussed through a web-based voting system by internationally known experts. Results were presented for further evaluation during a dedicated session of "The Fifth International Workshop on Peritoneal Surface Malignancy (Milan, Italy, December 4,6, 2006)". The experts agreed that multiple prospective trials support a benefit of the procedure in terms of improved survival, as compared with historical controls. Sentinel lymph node as a new marker for therapeutic planning in breast cancer patients

Abstract Background and Objectives Literature review suggests that the sentinel lymph node (sN) represents a reliable predictor of axillary lymph node status in breast cancer patients; however, some important issues, such as the optimisation of the technique for the intraoperative identification of the sN, the role of intraoperative frozen section examination of the sN, and the clinical implications of sN metastasis as regards the surgical management of the axilla, still require further confirmation. The authors aimed (1) to assess the feasibility of sN identification with a combined approach (vital blue dye lymphatic mapping and radioguided surgery, RGS) and the specific contribution of either techniques to the detection of the sN, (2) to determine the accuracy and usefulness of intraoperative frozen section examination of the sN in order to perform a one-stage surgical procedure, and (3) to define how the sN might modulate the therapeutic planning in different stages of disease. Materials and Methods From October 1997 to June 2001, 334 patients with early-stage (T1,2 N0 M0) invasive mammary carcinoma underwent sN biopsy; the average age of patients was 61.5 years (range, 39,75 years). In a subset of 153 patients, both vital blue dye (Patent Blue-V) lymphatic mapping and RGS were used to identify the sN, and the relative contribution of each of the two techniques was assessed. Results In the whole group, the sN was identified in 326 of 334 patients (97.6%), and 105 of 326 patients (37.3%) had positive axillary lymph nodes (pN+). In 9 of 105 pN+ patients, the definitive histologic examination of the sN did not show metastases but these were detected in non-sN, thus giving an 8.6% false-negative rate, a negative predictive value of 94.5% (156/165), and an accuracy of 96.5% (252/261). As regards the specific contribution of the two different techniques used in the identification of the sN, the detection rate was 73.8% (113/153) with Patent Blue-V alone, 94.1% (144/153) with RGS alone, and 98.7% (151/153) with Patent Blue-V combined with RGS (P,<,0.001). Noteworthy, whenever the sN was identified, the prediction of axillary lymph node status was remarkably similar (93,95% sensitivity; 100% specificity; 95,97% negative predictive value, and 97,98% accuracy) whichever of the three procedures was adopted (Patent Blue-V alone, RGS alone, or combined Patent Blue-V and RGS). Intraoperative frozen section examination was performed in 261 patients, who had at least one sN identified, out of 267 patients who underwent complete axillary dissection; 170 patients had histologically negative sN (i.o. sN,) and 91 patients histologically positive sN (i.o. sN+). All 91 i.o. sN+ were confirmed by definitive histology, whereas in 14 of 170 i.o. sN, patients (8.2%) metastases were detected at definitive histology. As regards the correlation between the size of sN metastasis, the primary tumour size, and the status of non-sN in the axilla, micrometastases were detected at final histology in 23 patients and macrometastases in 82 patients. When only micrometastases were detected, the sN was the exclusive site of nodal metastasis in 20 of 23 patients (86.9%) while in 3 patients with tumour size larger than 10 mm micrometastases were detected also in non-sN. Macrometastases were never detected in pT1a breast cancer patients; the sN was the exclusive site of these metastases in 30 patients (36.6%), while in 52 patients (63.4%) there were metastases both in sN and non-sN. Conclusions Sentinel lymphadenectomy can better be accomplished when both procedures (lymphatic mapping with vital blue dye and RGS) are used, because of the significantly higher sN detection rate, although the prediction of axillary lymph node status remains remarkably similar whichever method is used. The intraoperative frozen section examination proved to be rather accurate in predicting the actual pathologic status of the sN, with a negative predictive value of 91.8%; in 35% of patients it allowed sN biopsy and axillary dissection to be performed in a one-stage surgical procedure. Finally, specific clinical and histopathologic features of the primary tumour and sN might be used to tailor the loco-regional and systemic treatment in different clinical settings, such as in ductal carcinoma in-situ (DCIS), early-stage invasive breast cancer, and patients with large breast cancer undergoing neo-adjuvant CT for breast-saving surgery as well as elderly patients with operable breast cancer. Temperature-Gradient Effects in Thermal Barrier Coatings: An Investigation Through Modeling, High Heat Flux Test, and Embedded Sensor

The harsh thermal environment in gas turbines, including elevated temperatures and high heat fluxes, induces significant thermal gradients in ceramic thermal barrier coatings (TBCs), which are used to protect metallic components. However, the thermal conductivity of plasma-sprayed TBC increases with exposure at high temperatures mainly due to sintering phenomena and possible phase transformation, resulting in coating performance degradation and potential thermal runaway issues. An analytical thermal model and experimentally obtained coating thermal conductivity data are used to determine the coating through-thickness temperature profile and effective thermal conductivity under gradient conditions at high temperatures. High heat flux tests are then performed on TBCs to evaluate coating thermal behavior under temperature gradients close to service conditions. Coating internal temperature during the tests was also measured by thermally sprayed embedded thermocouples within the top coat. This combined approach provides a sintering map with a new model and allows for the assessment of temperature-gradient effects on the thermal performance of plasma-sprayed TBCs. [source]


Combined use of nuclear magnetic resonance and infrared spectroscopy for studying recognition processes between amphenicolic antibiotics and albumin

Abstract Biological reactions are mostly concerned with selective interactions between small ligands and macromolecular receptors. The same ligands may activate responses of different intensities and/or effects in the presence of different receptors. Many approaches based on spectroscopic and non-spectroscopic methods have been used to study interactions between small ligands and macromolecular receptors, including methods based on NMR and IR spectroscopic analysis of the solution behaviour of the ligand in the presence of receptors. In this work, we investigated the interaction between ovine serum albumin with two amphenicolic antibiotics [chloramphenicol (CAP) and thiamphenicol (TAP)], using a combined approach based on NMR and IR methodologies, furnishing complementary information about the recognition process occurring within the two systems. The two ligands, despite their similar structures, showed different affinities towards albumin. NMR methodology is based on the comparison of selective () and non-selective () spin,lattice relaxation rates of the ligands in the presence and absence of macromolecular receptors and and temperature dependence analysis. From these studies, the ligand,receptor binding strength was evaluated on the basis of the ,affinity index.' The derivation of the affinity index from chemical equilibrium kinetics for both the CAP,albumin and TAP,albumin systems allowed a comparison of the abilities of the two amphenicolic antibiotics to interact with the protein. IR methodology is based on the comparison of the ligand,protein ,complex' spectra with those of the non-interacting systems. On the basis of the differences revealed, a more thorough IR analysis was performed in order to understand the structural changes which occurred on both ligand and protein molecules within the interacting system. Carbon source accounting for fish using combined DNA and stable isotope analyses in a regulated lowland river weir pool

Abstract Determining the source and flow of carbon, energy and nutrients through food webs is essential for understanding ecological connectivity and thus determining the impact of management practices on biodiversity. We combined DNA sequencing, microarrays and stable isotope analyses to test whether this approach would allow us to resolve the carbon flows through food webs in a weir pool on the lower Murray River, a highly impacted, complex and regulated ecosystem in southern Australia. We demonstrate that small fish in the Murray River consume a wide range of food items, but that a significant component of carbon and nitrogen entering the food web during dry periods in summer, but not spring, is derived from nonconventional sources other than in-channel primary producers. This study also showed that isotopic analyses alone cannot distinguish food sources and that a combined approach is better able to elucidate food-consumer dynamics. Our results highlight that a major river ecosystem, stressed by reduced environmental flows, can rapidly undergo significant and previously undetected changes that impact on the ecology of the system as a whole. Biological and Cultural Anthropology of a Changing Tropical Forest: A Fruitful Collaboration across Subfields

In this article, we integrate approaches from biological and cultural anthropology to describe changing relationships between humans and animals in the Dzanga-Ndoki Park and Dzanga-Sangha Dense Forest Reserve (RDS), Central African Republic (CAR). Recent decades have seen a rapid proliferation of human activities, with striking tensions between logging and conservation economies. Our data suggest that certain animals and humans initially adapted successfully to these forest uses, and that local residents have crafted culturally rich new ways of living in the forest. However, our longitudinal data indicate animal declines and expanding frontiers of increasingly intensive human use. These trends are altering previous territorial arrangements and coming to undermine today's remarkably rich spectrum of human,animal encounters there. Our combined approach offers an alternative to increasingly distinct method and theory between anthropology's subfields. We sketch a research agenda for integrated anthropological attention to environmental change, especially to transformations in human,animal interactions and entanglements. [source]

Abstract In this article we apply the subdomain-Galerkin/least squares method, which is first proposed by Chang and Gunzburger for first-order elliptic systems without reaction terms in the plane, to solve second-order non-selfadjoint elliptic problems in two- and three-dimensional bounded domains with triangular or tetrahedral regular triangulations. This method can be viewed as a combination of a direct cell vertex finite volume discretization step and an algebraic least-squares minimization step in which the pressure is approximated by piecewise linear elements and the flux by the lowest order Raviart-Thomas space. This combined approach has the advantages of both finite volume and least-squares methods. Among other things, the combined method is not subject to the Ladyzhenskaya-Babus,ka-Brezzi condition, and the resulting linear system is symmetric and positive definite. High throughput testing platform for organic Solar Cells

Abstract In this paper we present a high throughput testing setup for organic solar cells that is necessary for an efficient analysis of their behaviour. The setup comprises process parameter logging, automated measurement data acquisition and subsequent data management and analysis. Utilising this setup the reproducibility of solar cells and the effect of production parameter variations has been tested with a set of 360 solar cells based on the poly-3-hexylthiophene:1-(3-methoxycarbonyl)-propyl-1-1-phenyl-(6,6)C61 bulk heterojunction. Variations in power conversion efficiency between 1 and 3% were observed on varying production parameters hardly mentioned in literature. The conditions during the vacuum deposition of the aluminium cathode turned out to have a significant effect. The key solar cell parameter affecting the performance was the fill factor (FF). As such the work exemplifies the necessity for a combined approach to analyse the complex behaviour of organic solar cells. The developed high throughput testing setup provides a basis for an efficient testing of production parameter variations and materials and additionally opens the door for statistical analysis. Functional site profiling and electrostatic analysis of cysteines modifiable to cysteine sulfenic acid

Abstract Cysteine sulfenic acid (Cys-SOH), a reversible modification, is a catalytic intermediate at enzyme active sites, a sensor for oxidative stress, a regulator of some transcription factors, and a redox-signaling intermediate. This post-translational modification is not random: specific features near the cysteine control its reactivity. To identify features responsible for the propensity of cysteines to be modified to sulfenic acid, a list of 47 proteins (containing 49 known Cys-SOH sites) was compiled. Modifiable cysteines are found in proteins from most structural classes and many functional classes, but have no propensity for any one type of protein secondary structure. To identify features affecting cysteine reactivity, these sites were analyzed using both functional site profiling and electrostatic analysis. Overall, the solvent exposure of modifiable cysteines is not different from the average cysteine. The combined sequence, structure, and electrostatic approaches reveal mechanistic determinants not obvious from overall sequence comparison, including: (1) pKas of some modifiable cysteines are affected by backbone features only; (2) charged residues are underrepresented in the structure near modifiable sites; (3) threonine and other polar residues can exert a large influence on the cysteine pKa; and (4) hydrogen bonding patterns are suggested to be important. This compilation of Cys-SOH modification sites and their features provides a quantitative assessment of previous observations and a basis for further analysis and prediction of these sites. Agreement with known experimental data indicates the utility of this combined approach for identifying mechanistic determinants at protein functional sites. [source]


Determination of phenolic compounds in rose hip (Rosa canina) using liquid chromatography coupled to electrospray ionisation tandem mass spectrometry and diode-array detection

Liquid chromatography coupled with negative and positive electrospray ionisation (ESI) tandem mass spectrometry (MS/MS) and diode-array detection (DAD) was used for determination of phenols in rose hip (Rosa canina) extract. ESI mass spectra of the chromatographically separated phenols gave the molecular weight of the compounds through prominent [M,,,H], ions for most of the compounds and M+ ions for the anthocyanins. Collision induced dissociation (CID) of the [M,,,H], (or M+) precursor ions yielded product ions which determined the molecular weight of the aglycones. In-source fragmentation followed by CID of the resulting deprotonated aglycone ([A,,,H],) provided product ions for the identification of the unconjugated phenols. The identification was based on comparison with product ion spectra of commercial standards. UV-diode-array spectra were used for identity confirmation. This combined approach allowed the identification in rose hip extract of an anthocyanin, i.e. cyanidin-3- O -glucoside, several glycosides of quercetin and glycosides of taxifolin and eriodictyol. Phloridzin was identified, and several conjugates of methyl gallate were also found, one of which was tentatively identified as methyl gallate-rutinoside.
