In this study we evaluated an assumptive anti-inflammatory effect of the nonsedative histamine H1-receptor antagonist, mizolastine, on UV-induced acute sunburn reaction. Therefore, a clinical, randomized, double-blind, four-arm, crossover study was conducted in healthy young female volunteers (skin type II) comparing the UV sensitivity under mizolastine, acetyl-salicylic acid (ASA), indomethacin or a mizolastine/ASA combination. Moreover, HaCaT keratinocytes were incubated with mizolastine under various UV treatment modalities in vitro to study its effect on the release of inflammatory cytokines, i.e. interleukin (IL)-1,, IL-6 and tumor necrosis factor , (TNF-,). All three drugs were effective in suppressing the UVB-, UVA- and combined UVA/UVB-erythema. However, the strongest effects were observed using the combined treatment with both 250 mg ASA and 10 mg mizolastine. An inhibitory effect in vitro of 10 nM mizolastine upon UV-induced cytokine release from HaCaT keratinocytes was observed for IL-1, at 24 h after 10 J/cm2 UVA1, for IL-6 at 48 h after 10 J/cm2 UVA1 and 30 mJ/cm2 UVB, and also for TNF-, at 4 h after 10 J/cm2 UVA, 10 J/cm2 UVA1 and 30 mJ/cm2 UVB, respectively. The combination of mizolastine and ASA can be strongly recommended as a protective measure against UV erythema development with a lower unwanted side effect profile than that of the hitherto treatment modality, i.e. indomethacin. [source] The importance of family management, closeness with father and family structure in early adolescent alcohol useADDICTION, Issue 10 2010Cherine Habib ABSTRACT Aims To examine the importance of family management, family structure and father,adolescent relationships on early adolescent alcohol use. Design Cross-sectional data was collected across 30 randomly selected Australian communities stratified to represent a range of socio-economic and regional variation. Setting Data were collected during school time from adolescents attending a broad range of schools. Participants The sample consisted of a combined 8256 students (aged 10,14 years). Measurements Students completed a web-based survey as part of the Healthy Neighbourhoods project. Findings Family management,which included practices such as parental monitoring and family rules about alcohol use,had the strongest and most consistent relationship with alcohol use in early adolescence. Adolescents reporting higher family management were less likely to have drunk alcohol in their life-time, less likely to drink alcohol in the preceding 30 days and less likely to have had an alcohol binge. Adolescents reporting emotionally close relationships with their fathers were less likely to have drunk alcohol in their life-time and less likely to have had an alcohol binge in the preceding fortnight. Conclusions Findings indicate that family management practices may contribute to alcohol abstinence in adolescents. Furthermore, emotionally close father,adolescent relationships may also foster abstinence; however, fathers' drinking behaviours need to be considered. [source] Comparison of different algorithms to calculate electrophoretic mobility of analytes as a function of binary solvent compositionELECTROPHORESIS, Issue 10 2003Abolghasem Jouyban Abstract Ten different mathematical models representing the electrophoretic mobility of analytes in capillary electrophoresis in mixed solvents of different composition have been compared using 32 experimental data sets. The solvents are binary mixtures of water-methanol, water-ethanol and methanol-ethanol, respectively. Mean percentage deviation (MPD), overall MPD (OMPD) and individual percentage deviation (IPD) have been considered as comparison criteria. The results showed that a reorganized solution model, namely the combined nearly ideal binary solvent/Redlich-Kister equation, is the most accurate model among other similar models concerning both correlation ability and prediction capability. [source] Early treatment after a symptomatic event is not associated with an increased risk of stroke in patients undergoing carotid stentingEUROPEAN JOURNAL OF NEUROLOGY, Issue 1 2008K. Gröschel A recently symptomatic carotid artery stenosis carries a high risk of subsequent ischaemic events and thus requires rapid treatment. We investigated the influence of the time delay between the last symptomatic event of a carotid stenosis and subsequent carotid artery stenting (CAS) with respect to the combined 30-day outcome of stroke and death. In a group of 320 patients undergoing CAS the median delay before the intervention was 19 days (interquartile range 10,36) and the combined 30-day complication rate was 8.4%. Time delay was not significantly associated with peri-procedural complications, regardless of whether this variable was dichotomized (<14 days and ,14 days), separated into interquartile ranges or analysed as a continuous variable. Our results indicate that early CAS is not associated with an increased complication rate in patients with a recently symptomatic carotid stenosis. Thus, if CAS has been selected as the treatment modality for a patient, it should be performed as soon as possible to maximize the benefit of the intervention in reducing the risk of stroke. [source] Family Predictors of Antisocial Behavior in AdolescenceFAMILY PROCESS, Issue 2 2003Maja Dekovi, Ph.D. The goal of the present study was to examine the combined and unique ability of different aspects of family functioning to predict involvement in antisocial behavior in a large nonclinical (community) sample of adolescents. Distinction was made between global (e.g., family socio-economic status), distal (dispositional characteristics of parents), contextual (family characteristics), and proximal (parent-child interaction) factors that operate within families. Results show that proximal factors were significant predictors of antisocial behavior, independent of their shared variance with other factors. Consistent with the hypothesized mediational model, the effects of distal and contextual factors appear to be mostly indirect: after their association with proximal factors was taken into account, these factors were no longer significantly related to antisocial behavior. The implications of these findings for planning of developmentally appropriate interventions for ado-lesents and their families are discussed. [source] Combining fishery prohibition with stocking of landlocked salmon, Salmo salar L.: an effort to gain bigger yield and individual sizeFISHERIES MANAGEMENT & ECOLOGY, Issue 6 2000P. Hyvärinen After stocking landlocked salmon, Salmo salar L., in Lake Änättijärvi, all fishing was prohibited for 18 months to provide time for the fish to establish and reach a larger size prior to capture. The potential benefit of the fishery prohibition was severely diminished by emigration of the stocked fish down the watercourse to other lakes. Only 19% of the recaptures of Carlin-tagged individuals came from Lake Änättijärvi. Also, the CPUE of test fishing in Lake Änättijärvi indicated a steep decline in the density of stocked fish during the fishery prohibition period. The mean weight of landlocked salmon in the combined recaptures from Lake Änättijärvi and other lakes downstream was 660 g. When fishing was not prohibited, the corresponding mean weight at recapture was 442 g. It is recommended that landlocked salmon stocking should be carried out in lakes with relatively low fishing pressure. [source] Effects of waterfowl and fish on submerged vegetation and macroinvertebratesFRESHWATER BIOLOGY, Issue 11 2002Ola Marklund SUMMARY 1. With the aim to assess the combined and separate effects of waterfowl and fish on submerged vegetation and macroinvertebrates, we performed a replicated selective exclosure study in a shallow, eutrophic lake in southern Sweden. Our results are presented together with a literature review of the effects of fish and waterfowl on macroinvertebrates and submerged vegetation. 2. Based on our experiment and on published data, we conclude that waterfowl normally will reduce submerged vegetation only at high waterfowl densities, at very low vegetation densities, or in the colonisation phase of the vegetation. 3. Further, we conclude that in shallow temperate eutrophic lakes, a naturally occurring mixed fish assemblage rarely reduces submerged vegetation. Unless the vegetation is very sparse, the risk of severe reduction of submerged vegetation as a result of waterfowl or fish grazing, should thereby be low. 4. Even relatively low densities of fish seem to reduce macroinvertebrate biomass, while a mixed waterfowl assemblage rarely has a significant effect on macroinvertebrate biomass. [source] Modelling hydrological management for the restoration of acidified floating fensHYDROLOGICAL PROCESSES, Issue 20 2005Stefan C. Dekker Abstract Wetlands show a large decline in biodiversity. To protect and restore this biodiversity, many restoration projects are carried out. Hydrology in wetlands controls the chemical and biological processes and may be the most important factor regulating wetland function and development. Hydrological models may be used to simulate these processes and to evaluate management scenarios for restoration. HYDRUS2D, a combined saturated,unsaturated groundwater flow and transport model, is presented. This simulates near-surface hydrological processes in an acidified floating fen, with the aim to evaluate the effect of hydrological restoration in terms of conditions for biodiversity. In the acidified floating fen in the nature reserve Ilperveld (The Netherlands), a trench system was dug for the purpose of creating a runoff channel for acid rainwater in wet periods and to enable circum-neutral surface water to enter the fen in dry periods. The model is calibrated against measured conductivity values for a 5 year period. From the model simulations, it was found that lateral flow in the floating raft is limited. Furthermore, the model shows that the best management option is a combination of trenches and inundation, which gave the best soil water quality in the root zone. It is concluded that hydrological models can be used for the calculation of management scenarios in restoration projects. W. van Wingerden Abstract The trend with offshore wind turbines is to increase the rotor diameter as much as possible to decrease the costs per kWh. The increasing dimensions have led to the relative increase in the loads on the wind turbine structure. Because of the increasing rotor size and the spatial load variations along the blade, it is necessary to react to turbulence in a more detailed way: each blade separately and at several separate radial distances. This combined with the strong nonlinear behavior of wind turbines motivates the need for accurate linear parameter-varying (LPV) models for which advanced control synthesis techniques exist within the robust control framework. In this paper we present a closed-loop LPV identification algorithm that uses dedicated scheduling sequences to identify the rotational dynamics of a wind turbine. We assume that the system undergoes the same time variation several times, which makes it possible to use time-invariant identification methods as the input and the output data are chosen from the same point in the variation of the system. We use time-invariant techniques to identify a number of extended observability matrices and state sequences that are inherent to subspace identification identified in a different state basis. We show that by formulating an intersection problem all states can be reconstructed in a general state basis from which the system matrices can be estimated. [source] Assessment of the 10-Year Probability of Osteoporotic Hip Fracture Combining Clinical Risk Factors and Heel Bone Ultrasound: The EPISEM Prospective Cohort of 12,958 Elderly Women,,JOURNAL OF BONE AND MINERAL RESEARCH, Issue 7 2008Didier Hans Abstract This study aimed to develop a hip screening tool that combines relevant clinical risk factors (CRFs) and quantitative ultrasound (QUS) at the heel to determine the 10-yr probability of hip fractures in elderly women. The EPISEM database, comprised of ,13,000 women ,70 yr of age, was derived from two population-based white European cohorts in France and Switzerland. All women had baseline data on CRFs and a baseline measurement of the stiffness index (SI) derived from QUS at the heel. Women were followed prospectively to identify incident fractures. Multivariate analysis was performed to determine the CRFs that contributed significantly to hip fracture risk, and these were used to generate a CRF score. Gradients of risk (GR; RR/SD change) and areas under receiver operating characteristic curves (AUC) were calculated for the CRF score, SI, and a score combining both. The 10-yr probability of hip fracture was computed for the combined model. Three hundred seven hip fractures were observed over a mean follow-up of 3.2 yr. In addition to SI, significant CRFs for hip fracture were body mass index (BMI), history of fracture, an impaired chair test, history of a recent fall, current cigarette smoking, and diabetes mellitus. The average GR for hip fracture was 2.10 per SD with the combined SI + CRF score compared with a GR of 1.77 with SI alone and of 1.52 with the CRF score alone. Thus, the use of CRFs enhanced the predictive value of SI alone. For example, in a woman 80 yr of age, the presence of two to four CRFs increased the probability of hip fracture from 16.9% to 26.6% and from 52.6% to 70.5% for SI Z-scores of +2 and ,3, respectively. The combined use of CRFs and QUS SI is a promising tool to assess hip fracture probability in elderly women, especially when access to DXA is limited. [source] Comparison of spinal anesthesia with general anesthesia on morphine requirement after abdominal hysterectomyACTA ANAESTHESIOLOGICA SCANDINAVICA, Issue 5 2009L. MASSICOTTE Purpose: The aim of this study was to compare morphine consumption with patient-controlled analgesia (PCA) between spinal anesthesia (SA) (bupivacaine, morphine and fentanyl) and general anesthesia (GA) (sufentanil) after an abdominal hysterectomy. Methods: Forty women were randomly assigned to receive SA with bupivacaine 15 mg, 0.15 mg of intrathecal morphine and 15 ,g of fentanyl or GA with sufentanil, both combined with PCA. The primary outcome was morphine consumption with the PCA device. The secondary outcomes were post-operative pain at rest and under stress on a visual analog scale, nausea, pruritus and respiratory depression on a standardized scale. Outcome measures were recorded at 6, 12, 18, 24 and 48 h post-anesthesia. The duration of post-anesthesia care unit (PACU) and hospital stay were recorded. Results: Patients in the SA group consumed at least two times less morphine at each time interval than the GA group: at 48 h, they used 19 ± 17 vs. 81 ± 31 mg (P<0.0001). Post-operative pain at rest was lower in the SA group until the 18th hour and under stress until the 48th. There was more sedation in the GA group until the 18th hour. Little difference was observed in the incidence of pruritus. Nausea was more intense at the 6th hour in the GA group. There was no difference in the respiratory rate. The duration of PACU stay was shorter for the SA group (52 ± 9 vs. 73 ± 11 min, P<0.0001) as was the duration of hospital stay (2.2 ± 0.4 vs. 3.3 ± 0.7 days, P=0.01). Conclusions: It is concluded that intrathecal morphine 0.15 mg with 15 ,g of fentanyl decreases post-operative pain and morphine consumption by PCA without increasing adverse reactions for women undergoing an abdominal hysterectomy. Murphy ABSTRACT: This study was to evaluate the effectiveness of steam or steam in combination with an antimicrobial agent to control Listeria monocytogenes on ready-to-eat (RTE) franks. The franks were surface-inoculated to contain 6 or 3 log10(colony-forming units [CFU])/cm2 of L. monocytogenes and treated with steam or steam in combination with an antimicrobial agent, immediately followed by vacuum-sealing the top films of frank packages (6 franks per package in a single layer). Three log (CFU) /cm2 of reductions were achieved at the both inoculation levels for L. monocytogenes on franks. At an inoculation level of 3 logs, no outgrowth of L. monocytogenes was obtained on the treated franks after storing at 4.4°C or 16°C for a combined 47 d. This study provided an alternative approach for controlling L. monocytogenes in packaged franks. [source] The use of desflurane or propofol in combination with remifentanil in myasthenic patients undergoing a video-assisted thoracoscopic-extended thymectomyACTA ANAESTHESIOLOGICA SCANDINAVICA, Issue 3 2009P. GRITTI Background: Although several studies of the use of desflurane in anesthesia have revealed many desirable qualities, there are no data on the use and effects especially on the neuromuscular function of desflurane on myasthenia gravis (MG) patients. The purpose of this study was to evaluate the use of either desflurane or propofol, both combined with remifentanil, in patients with MG undergoing a video-assisted thoracoscopic-extended thymectomy (VATET). Methods: Thirty-six MG patients who underwent VATET were enrolled. Nineteen patients were anesthetized with remifentanil and propofol infused with a target-controlled infusion plasma model, and 17 patients with desflurane and remifentanil. No muscle relaxant was used. The intubating conditions, hemodynamic and respiratory changes, neuromuscular transmission and post-operative complications were evaluated. Results: Neuromuscular transmission was significantly decreased in the desflurane group (6.7%, from 3% to 9% during anesthesia P=<0.05). The intubating conditions were good in all 36 patients and 35 patients were successfully extubated in the operating room. The time-to-awakening, post-operatory pH and base excess were significantly different in the two groups, with a decreasing mean arterial pressure in the group administered with desflurane. No patients required reintubation due to myasthenic or cholinergic crisis, or respiratory failure. No other significant differences between the two groups studied were observed. Conclusion: Our experience indicates that anesthesia with desflurane plus remifentanil in patients with MG could determine a reversible muscle relaxation effect, but with no clinical implication, allowing a faster recovery with no difference in extubation time and post-operative complications in the two groups. [source] Investigation of tyrosine nitration in proteins by mass spectrometryJOURNAL OF MASS SPECTROMETRY (INCORP BIOLOGICAL MASS SPECTROMETRY), Issue 6 2001Ann-Sofi Petersson Abstract In vivo nitration of tyrosine residues is a post-translational modification mediated by peroxynitrite that may be involved in a number of diseases. The aim of this study was to evaluate possibilities for site-specific detection of tyrosine nitration by mass spectrometry. Angiotensin II and bovine serum albumin (BSA) nitrated with tetranitromethane (TNM) were used as model compounds. Three strategies were investigated: (i) analysis of single peptides and protein digests by matrix-assisted laser desorption/ionization (MALDI) peptide mass mapping, (ii) peptide mass mapping by electrospray ionization (ESI) mass spectrometry and (iii) screening for nitration by selective detection of the immonium ion of nitrotyrosine by precursor ion scanning with subsequent sequencing of the modified peptides. The MALDI time-of-flight mass spectrum of nitrated angiotensin II showed an unexpected prompt fragmentation involving the nitro group, in contrast to ESI-MS, where no fragmentation of nitrated angiotensin II was observed. The ESI mass spectra showed that mono- and dinitrated angiotensin II were obtained after treatment with TNM. ESI-MS/MS revealed that the mononitrated angiotensin II was nitrated on the side-chain of tyrosine. The dinitrated angiotensin II contained two nitro groups on the tyrosine residue. Nitration of BSA was confirmed by Western blotting with an antibody against nitrotyrosine and the sites for nitration were investigated by peptide mass mapping after in-gel digestion. Direct mass mapping by ESI revealed that two peptides were nitrated. Precursor ion scanning for the immonium ion for nitrotyrosine revealed two additional partially nitrated peptides. Based on the studies with the two model compounds, we suggest that the investigation of in vivo nitration of tyrosine and identification of nitrated peptides might be performed by precursor ion scanning for the specific immonium ion at m/z 181.06 combined with ESI-MS/MS for identification of the specific nitration sites. [source] Coupled Lu,Hf and Sm,Nd geochronology constrains garnet growth in ultra-high-pressure eclogites from the Dabie orogenJOURNAL OF METAMORPHIC GEOLOGY, Issue 7 2008H. CHENG Abstract Ultra-high-pressure eclogites from the Dabie orogen that formed over a range in temperatures (,600 to > 700 °C) have been investigated with combined Lu,Hf and Sm,Nd geochronology. Three eclogites, sampled from Zhujiachong, Huangzhen and Shima, yield Lu,Hf ages of 240.0 ± 5.0, 224.4 ± 1.9 and 230.8 ± 5.0 Ma and corresponding Sm,Nd ages of 222.5 ± 5.0, 217.6 ± 6.1 and 224.2 ± 2.1 Ma respectively. Well-preserved prograde major- and trace-element zoning in garnet in the Zhujiachong eclogite suggests that the Lu,Hf age mostly reflects an early phase of garnet growth that continued over a time interval of c. 17.5 Myr. For the Huangzhen eclogite, despite preserved elemental growth zoning in garnet, textural study reveals that the Lu,Hf age is biased towards a later garnet growth episode rather than representing early growth. The narrow time interval of <6.6 Myr defined by the difference between Lu,Hf and Sm,Nd ages indicates a short final garnet growth episode and suggests a rapid cooling stage. By contrast, the rather flat element zoning in garnet in the Shima eclogite suggests that Lu,Hf and Sm,Nd ages for this sample have been reset by diffusion and are cooling ages. The new Lu,Hf ages point to an initiation of prograde metamorphism prior to c. 240 Ma for the Dabie orogen, while the exact peak metamorphic timing experienced by specific samples ranges between c. 230 to c. 220 Ma. In this report, we describe the use of a combined left trisegmentectomy and caudate lobectomy, along with resection of the inferior vena cava, to treat a large intrahepatic cholangiocarcinoma in a Jehovah's Witness. To our knowledge, this is the first report of major liver resection for a large malignant tumor in this patient population. We also discuss the perioperative strategy and surgical technique we used to minimize blood loss and avoid transfusion. This approach could be a safe alternative for use in all patients with complex liver tumor, regardless of their religious beliefs, to reduce the risks and cost associated with blood transfusion. J. Abou-Agag Background: Moderate red wine consumption has been associated with a reduced risk for coronary heart disease, and this cardioprotection may be mediated, in part, by promoting fibrinolysis. This protection may be attributed to the combined or perhaps synergistic effects of alcohol and other red wine components (i.e., polyphenolics). These studies were carried out to determine whether individual phenolics (i.e., catechin, epicatechin, quercetin, and resveratrol) affect fibrinolytic protein (tissue-type plasminogen activator [t-PA] and urokinase-type PA [u-PA]) e-pression and surface-localized fibrinolytic activity in cultured human umbilical vein endothelial cells (HUVECs). Methods: Cultured HUVECs were preincubated (1 hr, 37°C) in the absence or presence of varying concentrations of catechin, epicatechin, quercetin, and resveratrol (0.001,10 ,M) and then were washed and incubated for various times in the absence of phenolics. Secreted t-PA/u-PA antigen (24 hr, enzyme-linked immunoadsorbent assay) and mRNA [0,16 hr, reverse transcription-polymerase chain reaction(RT-PCR)] levels and fibrinolytic activity (direct activation of HUVEC-bound 125I-labeled glutamyl-plasminogen, quantitation of 125I-labeled M r 20 kDa plasmin light-chain) were measured. Transient transfections of cultured HUVECs were carried out with the pt-PA222/luc and pu-PA236/luc promoter constructs, by using lipofectamine. Results: Each of the phenolics similarly increased t-PA and u-PA antigen (2- to 3-fold) and mRNA (3- to 4-fold) levels, concomitant with an increase (2- to 3-fold) in sustained (24 hr), surface-localized fibrinolytic activity. Transcription inhibitor actinomycin D abolished the induction of t-PA and u-PA mRNA e-pression by these phenolics. Transfections with the pt-PA222/luc and pu-PA236/luc promoter constructs showed 2- to 3-fold and 2- to 4-fold increases in luciferase activity for t-PA and u-PA, respectively. Conclusions: These results demonstrate that each of these phenolics up-regulates both t-PA and u-PA gene transcription, which results in the sustained increased e-pression of surface-localized fibrinolytic activity in cultured HUVECs. Wine phenolics increase fibrinolytic activity, independent of ethanol, and it is likely that the overall cardioprotective benefits associated with moderate red wine consumption are attributable to the combined, additive, or perhaps synergistic effects of alcohol and other wine components. Lignell Abstract Aims:, Microbial concentrations in vacuumed house dust samples (n = 71) were analysed by culture and quantitative polymerase chain reaction (qPCR) methods and their association with extent of moisture damage in the house was studied. Methods and Results:, Microbial concentrations measured by qPCR correlated with concentrations obtained by culture method, but were orders of magnitude higher. qPCR also had better sensitivity. Concentrations of several microbes in house dust, determined with qPCR, were associated with the extent of moisture damage in the house. This association was strongest for Penicillium brevicompactum, one of the fungi detected in highest concentrations by qPCR. Furthermore, house dust concentrations of Wallemia sebi, Trichoderma viride, Cladosporium sphaerospermum, Eurotium amstelodami and the combined assay group for Penicillium spp., Aspergillus spp. and Paecilomyces variotii were significantly associated with the extent of the moisture damage. Conclusion:, These species or assay groups could probably be used as indicators of moisture damage in the house. Significance and Impact of the Study:, This finding indicates the benefits of the qPCR method, which is sensitive enough to reveal the differences in microbial concentrations of house dust between moisture-damaged and undamaged houses. [source] Inflammatory change of fatty liver induced by intraportal low-dose lipopolysaccharide infusion deteriorates pancreatic insulin secretion in fructose-induced insulin-resistant ratsLIVER INTERNATIONAL, Issue 8 2008Po-Shiuan Hsieh Abstract Background: This study tested whether subacute inflammatory change of fatty liver induced by portal endotoxaemia is detrimental to pancreatic insulin secretion in fructose-fed rats (FFRs) with fatty liver. Methods: Rats were randomly assigned into two groups with a regular or fructose-enriched diet for 8 weeks. Rats, after fructose feeding for 4 weeks, were further divided into three subgroups: on fructose diet alone, on fructose diet combined with intraportal saline or lipopolysaccharide (LPS) infusion (n=8 per group) for the next 4 weeks. In another set of experiments, the liver and pancreatic tissues were obtained for histological examination in these four groups. Pancreatic insulin secretion was evaluated by in vivo hyperglycaemic clamp study. Results: Fasting plasma insulin concentrations and homoeostasis model assessment-insulin resistance, an insulin resistance score, were significantly increased in FFRs but failed to change in rats with LPS treatment. The 4-week intraportal LPS infusion significantly increased circulating aspartate transaminase, alanine transaminase and C-reactive protein levels but did not alter endotoxin levels in FFRs. The increased white blood cell count was also noted in rats after intraportal LPS infusion for 2 and 4 weeks. The attenuated first-phase and second-phase insulin responses in FFRs shown in hyperglycaemic clamp were further deteriorated in those with intraportal LPS infusion. Increased histopathological scores of liver and pancreas shown in FFRs were further increased in those combined with portal endotoxaemia. Conclusion: This study demonstrates that the chronic subacute inflammatory change of fatty liver induced by mild portal endotoxaemia could deteriorate insulin secretion in a rodent model of metabolic syndrome and fatty liver. [source] The mechanical binding strengths of Helicobacter pylori BabA and SabA adhesins using an adhesion binding assay,ELISA, and its clinical relevance in JapanMICROBIOLOGY AND IMMUNOLOGY, Issue 8 2010Mitsuaki Nishioka ABSTRACT To elucidate a potential role for H. pylori BabA and SabA adhesins in the pathogenesis of gastric mucosal lesions, the MBS of BabA and SabA was examined using an in-house ABA-ELISA. Ninety isolates from Japanese patients with gastric cancer (n= 43) and non-cancerous (n= 47) lesions were subjected to an ABA-ELISA which had been developed in-house, and sequential analysis of the babA2 middle region. The BabA-MBS was significantly higher in the cancer than the non-cancer group (P= 0.019), but there was no significant difference for SabA-MBS. A weak correlation between BabA-MBS and SabA-MBS (r= 0.418) was observed, the positive correlation being higher in the cancer than the non-cancer group (r= 0.598 and 0.288, respectively). The isolates were classified into two groups: a BabA-high-binding and a BabA-low-binding group (in comparison to the average for BabA-MBS). The average SabA-MBS in the BabA-high-binding group was significantly higher than in the BabA-low-binding group (P < 0.0001). Analysis of babA2 middle region diversity (AD1,5) revealed that AD2-type was predominant in isolates irrespective of BabA-MBS. H. pylori BabA-MBS might have an effect on SabA-MBS and relate to the severity of gastric disorders, including gastric cancer. Evaluation of MBS of the combined two adhesins would be helpful for predicting damage in the H. pylori infected stomach. The new characterization of a naturally occurring allopolyploid in the genus Mimulus by Sweigart et al. in this issue of Molecular Ecology is therefore to be welcomed. Mimulus is rapidly emerging as a model system for evolutionary functional genomics. Sequences of the whole genome and 200 000 expressed sequence tags of diploid M. guttatus, a putative parent of the polyploid described in this issue, will soon be available. These will facilitate investigation of the fates of genes duplicated by whole genome duplication, and their effects on morphology, mating system and ecology in natural populations. [source] Effects of Continuous and Triggered Atrial Overdrive Pacing on Paroxysmal Atrial Fibrillation in Pacemaker PatientsPACING AND CLINICAL ELECTROPHYSIOLOGY, Issue 8 2008ANDREAS SCHUCHERT M.D. Background: The aim of the study was to compare the effects of different pacing strategies to prevent paroxysmal atrial fibrillation (AF): triggered atrial overdrive pacing versus the combination of triggered and continuous overdrive pacing. Methods: Patients with an indication for dual-chamber pacing (Selection 9000, Prevent AF; Vitatron B.V., Arnhem, the Netherlands) and a history of paroxysmal AF were randomized to triggered atrial pacing (three pacing functions, "triggered group": PAC SuppressionÔ, Post-PAC ResponseÔ, and Post-Exercise ResponseÔ) or to the combination of continuous (Pace ConditioningÔ) and triggered atrial pacing (four pacing functions, "combined group"). After 3 months, there was a crossover to the other pacemaker setting. Results: In 171 enrolled patients, the median AF burden of the combined group was with 2.1% versus 0.1% in the triggered group (P = 0.014). Fewer AF episodes were observed in the triggered (median: 7) than in the combined group (median: 116; P = 0.016). The combined group had more frequent atrial pacing (median 97%) than the triggered group with 85% (P < 0.001), but ventricular pacing was not significantly different with 95% and 96% in the combined and triggered group, respectively. After the crossover, the AF burden increased in the triggered group to 0.3% and decreased in the combined group to 0.4%. Conclusions: Triggered atrial pacing functions alone resulted in a low AF burden. The additional activation of continuous atrial overdrive pacing increased the percentage of atrial pacing, but had no beneficial effects on the prevention of paroxysmal AF. [source] Assessing levels of contaminants in breast milk: methodological issues and a framework for future researchPAEDIATRIC & PERINATAL EPIDEMIOLOGY, Issue 1 2008Mary J. Renfrew Summary To assess the scale of the possible exposure by the breast-fed infant to potentially harmful substances in breast milk, methodologically robust studies are essential. Many studies in this field, however, do not report details of crucial issues such as recruitment and milk sampling. The aims of the study reported here were to develop robust methods for the study of contaminants in breast milk, and to develop a framework for future research and population monitoring. Three cohorts of women and babies were recruited by midwives from five sites in northern England. Cohort 1 (cross-sectional, n = 322) were asked to provide two milk samples, one at one week following birth and one at a subsequent time point. Cohort 2 (longitudinal, n = 54) were asked to provide five samples at specified time points over the first 12,16 weeks after birth. Cohort 3 (convenience, n = 18), mothers of babies in the Special Care Unit, were asked to donate surplus breast milk. A novel method of analysing fat concentration in small volumes was developed and tested. A randomly selected set of samples from different donors and stages of lactation was screened for organochlorine pesticide residues, polychlorinated biphenyls, dioxins/furans, phthalates and heavy metals. A total of 453 samples were donated. Cohort 3 was the least successful route of recruitment. Cohorts 1 and 2 combined were most representative of the population. Sample collection, transport and storage procedures, and the collection of data on life style and diet, were robust and acceptable to women. Midwifery involvement in recruitment was an essential component. This study offers a framework both for the conduct of future research studies, and for the establishment of regional and national monitoring mechanisms for contaminants in breast milk. Similar work on contaminants in formula as fed to babies is needed to inform risk assessment methods. They consist of simple, narrow, elongate laminas with entire margins and intramarginal veins. The epidermal cells of adaxial cuticle shows small, rounded, blunt papillae outward that protrude inward and fuse together as rolls along and parallel to the margins, while the adaxial cuticle bears brachyparacytic stomatal apparatus that exhibit sunken guard cells and hair bases consisting of a thick-walled pore surrounded by radially arranged differentiated cells. Resin bodies occur inside the mesophyll. These characters closely resemble the lauroid taxa ,Myrtophyllum' and Pandemophyllum from the Cenomanian of the Czech Republic and Dakota (USA) respectively. The narrow angle of basilaminar secondaries and the whole suite of features in the guard cells (sunken guard cells embedded into subsidiary cells and stomatal ledges) strongly support close affinity with the Lauraceae. From the Cenomanian lauraceous reproductive organs and their related leaves already showed high disparity and diversity. In addition they displayed a broad ecological range from freshwater floodplains to brackish swamps. This combined to high diversity of reproductive organs suggest ecological radiation of Lauraceae by the Cenomanian. Geller MD Abstract Purpose A dose-escalation Phase I study of taxol (paclitaxel) administered in combination with standard dose ifosfamide was conducted in children with relapsed or refractory solid tumors. Primary objectives were to estimate the maximum tolerated dose (MTD) and to describe the dose-limiting toxicities (DLTs). Patients and Methods Paclitaxel was administered as a 6-hr continuous infusion (hr 0,6), followed by intravenous ifosfamide (2 g/m2/day,×,3 days) over 1 hr at hours 6,7, 24,25, and 48,49. Patients at dose level 1 received 250 mg/m2 paclitaxel. Subsequent dose escalation proceeded using a standard 3,×,3 Phase I design. Results Fifteen patients received a combined 46 courses of therapy. The median age was 14.5 years (range, 2,19 years), and diagnoses included sarcoma (7), neuroblastoma (3), and other (5). Three patients received paclitaxel at 250 mg/m2 (10 courses), six at 325 mg/m2 (19 courses), three at 425 mg/m2 (8 courses), and three at 550 mg/m2 (9 courses). DLTs occurred in 2/3 patients at 550 mg/m2 paclitaxel during cycle 1, including grade 3 hypotension and grade 4 anaphylaxis in 1 patient each. Common non-dose-limiting toxicities included bone marrow suppression and peripheral neuropathy. Response was evaluable in 14 patients and included mixed response (3), stable disease (5), and progressive disease (6). Conclusion Paclitaxel hypersensitivity reactions were dose limiting when the drug was administered as a 6-hr infusion. The MTD and recommended Phase II dose of paclitaxel administered as a 6-hr continuous intravenous infusion followed by standard dose intravenous ifosfamide is 425 mg/m2 paclitaxel. [source] Nitrogen and carbon source,sink relationships in trees at the Himalayan treelines compared with lower elevationsPLANT CELL & ENVIRONMENT, Issue 10 2008MAI-HE LI ABSTRACT No single hypothesis or theory has been widely accepted for explaining the functional mechanism of global alpine/arctic treeline formation. The present study tested whether the alpine treeline is determined by (1) the needle nitrogen content associated with photosynthesis (carbon gain); (2) a sufficient source,sink ratio of carbon; or (3) a sufficient C,N ratio. Nitrogen does not limit the growth and development of trees studied at the Himalayan treelines. Levels of non-structural carbohydrates (NSC) in trees were species-specific and site-dependent; therefore, the treeline cases studied did not show consistent evidence of source/carbon limitation or sink/growth limitation in treeline trees. However, results of the combined three treelines showed that the treeline trees may suffer from a winter carbon shortage. The source capacity and the sink capacity of a tree influence its tissue NSC concentrations and the carbon balance; therefore, we suggest that the persistence and development of treeline trees in a harsh alpine environment may require a minimum level of the total NSC concentration, a sufficiently high sugar:starch ratio, and a balanced carbon source,sink relationship. We report a case of maternal isodisomy 10 combined with mosaic partial trisomy 10 (p12.31-q11.1). Chromosome examinations from a CVS sample showed a karyotype 47,XY,+mar/46,XY. The additional marker chromosome which was present in 6/25 interphase nuclei was shown by fluorescence in situ hybridization (FISH) to have been derived from a pericentromeric segment of chromosome 10. DNA analysis was performed from umbilical cord blood from the fetus after termination of the pregnancy at 18 weeks. The results showed that the two structurally normal chromosomes 10 were both of maternal origin, whereas the marker chromosome derived from the father. Autopsy of the fetus revealed hypoplasia of heart, liver, kidneys and suprarenal glands, but, apart from a right bifid ureter, no structural organ abnormalities. [source] The detection, correlation, and comparison of peptide precursor and product ions from data independent LC-MS with data dependant LC-MS/MSPROTEINS: STRUCTURE, FUNCTION AND BIOINFORMATICS, Issue 6 2009Scott J. Geromanos Abstract The detection, correlation, and comparison of peptide and product ions from a data independent LC-MS acquisition strategy with data dependent LC-MS/MS is described. The data independent mode of acquisition differs from an LC-MS/MS data acquisition since no ion transmission window is applied with the first mass analyzer prior to collision induced disassociation. Alternating the energy applied to the collision cell, between low and elevated energy, on a scan-to-scan basis, provides accurate mass precursor and associated product ion spectra from every ion above the LOD of the mass spectrometer. The method therefore provides a near 100% duty cycle, with an inherent increase in signal intensity due to the fact that both precursor and product ion data are collected on all isotopes of every charge-state across the entire chromatographic peak width. The correlation of product to precursor ions, after deconvolution, is achieved by using reconstructed retention time apices and chromatographic peak shapes. Presented are the results from the comparison of a simple four protein mixture, in the presence and absence of an enzymatically digested protein extract from Escherichia coli. The samples were run in triplicate by both data dependant analysis (DDA) LC-MS/MS and data-independent, alternate scanning LC-MS. The detection and identification of precursor and product ions from the combined DDA search results of the four protein mixture were used for comparison to all other data. Each individual set of data-independent LC-MS data provides a more comprehensive set of detected ions than the combined peptide identifications from the DDA LC-MS/MS experiments. In the presence of the complex E. coli background, over 90% of the monoisotopic masses from the combined LC-MS/MS identifications were detected at the appropriate retention time. Moreover, the fragmentation pattern and number of associated elevated energy product ions in each replicate experiment was found to be very similar to the DDA identifications. In the case of the corresponding individual DDA LC-MS/MS experiment, 43% of the possible detectable peptides of interest were identified. The presented data illustrates that the time-aligned data from data-independent alternate scanning LC-MS experiments is highly comparable to the data obtained via DDA. The obtained information can therefore be effectively and correctly deconvolved to correlate product ions with parent precursor ions. The ability to generate precursor-product ion tables from this information and subsequently identify the correct parent precursor peptide will be illustrated in a companion manuscript. [source] The study of organisational autonomy: a conceptual reviewPUBLIC ADMINISTRATION & DEVELOPMENT, Issue 2 2004Koen Verhoest We argue that contemporary research on the influence of organisational autonomy on performance in public organisations uses a diverse and a too restrictive conceptualisation of autonomy. After discussing that research, the article develops six dimensions of the concept of autonomy in public organisations. Second, weaknesses of contemporary research are shown by confronting their conceptualisations with the developed taxonomy. Third, data from a survey of Flemish public organisations illuminate the need to acknowledge the six different dimensions of autonomy when studying the effect of autonomy on performance. The empirical material points at the dangers of using formal,legal status of a public organisation as an indicator of its autonomy, given substantial heterogeneity of organisations with the same formal,legal status on each dimension of autonomy. Moreover, tensions between different levels of autonomy appear in practice, indicating the need for a combined and integrated study of the effects of the different dimensions of autonomy on performance.
