The effect of topically applied salicylic compounds on serotonin-induced scratching behaviour in hairless rats

Abstract: There is a strong need for antipruritic substances for treating itch in clinical dermatology. In one recent human study, topically applied acetylsalicylic acid has been described to rapidly decrease histamine-induced itch. We have established a model for periferally elicited pruritus by injecting serotonin into the rostral back area (neck) in rats. Using this model, we aimed to investigate the antipruritic potential of four different salicylic compounds, which all possess different skin penetration characteristics. Eighteen rats were studied for 6 weeks. Prior to serotonin injections (2 mg/ml, 50 µl), 10 µl of test substances was applied to a circular area 18 mm in diameter. The four substances were salicylic acid, butyl salicylate, diethylamine salicylate and salicylamide, all solubilized in dimethyl isosorbide to a concentration of 5% w/w. Diethylamine salicylate and salicylamide were previously shown to be slowly absorbed through rat skin in contrast to salicylic acid and butyl salicylate. After serotonin injections, scratching was monitored by video recording for 1.5 h. Compared with the vehicle, a lower number of scratch sequences were seen when diethylamine salicylate (P < 0.001) and salicylamide (P = 0.005) had been applied. The numbers of scratch sequences were lower with diethylamine salicylate and salicylamide than with the vehicle throughout the 1.5-h study period. We conclude that topical application of diethylamine salicylate and salicylamide could suppress serotonin-induced scratching in rats. The antipruritic effect seems to be related to the slow drug release of the two substances. The results may be clinically relevant as serotonin induces itch in humans. [source]


Disposition of rainwater under creosotebush

Abstract In desert shrubland ecosystems water and nutrients are concentrated beneath shrub canopies in ,resource islands'. Rain falling on to these islands reaches the ground as either stemflow or throughfall and then either infiltrates into the soil or runs off as overland flow. This study investigates the partitioning of rainwater between stemflow and throughfall in the first instance and between infiltration and runoff in the second. Two series of 40 rainfall simulation experiments were performed on 16 creosotebush shrubs in the Jornada Basin, New Mexico. The first series of experiments was designed to measure the surface runoff and was performed with each shrub in its growth position. The second series was designed to measure stemflow reaching the shrub base and was conducted with the shrub suspended above the ground. The experimental data show that once equilibrium is achieved, 16% of the rainfall intercepted by the canopy or 6·7% of the rain falling inside the shrub area (i.e. the area inside the shrub's circumscribing ellipse) is funnelled to the shrub base as stemflow. This redistribution of the rainfall by stemflow is a function of the ratio of canopy area (i.e. the area covered by the shrub canopy) to collar area (i.e. a circular area centred on the shrub base), with stemflow rate being positively correlated and throughfall rate being negatively correlated with this ratio. The surface runoff rate expressed as a proportion of the rate at which rainwater arrives at a point (i.e. stemflow rate plus throughfall rate) is the runoff coefficient. A multiple regression reveals that 75% of the variance in the runoff coefficient can be explained by three independent variables: the rainfall rate, the ratio of the canopy area to the collar area, and the presence or absence of subcanopy vegetation. Although the last variable is a dummy variable, it accounts for 66·4% of the variance in the runoff coefficient. This suggests that the density and extent of the subcanopy vegetation is the single most important control of the partitioning of rainwater between runoff and infiltration beneath creosotebush. Although these findings pertain to creosotebush, similar findings might be expected for other desert shrubs that generate significant stemflow and have subcanopy vegetation. New Shade Guide for Evaluation of Tooth Whitening,Colorimetric Study

ABSTRACT Statement of the Problem:, Available shade guides lack colorimetric uniformity, which compromises the validity of visual evaluation of tooth whitening efficacy. Purpose:, The objective of this study was to perform a colorimetric analysis of a new shade guide designed primarily for the visual evaluation of tooth whitening efficacy and to compare this shade guide with two commercial shade guides. Materials and Methods:, Color ranges and color distribution of three shade guides (prototype of the new Vita Bleachedguide 3D-Master [BG, Vita Zahnfabrik, Bad Säckingen, Germany], value scale of Vitapan Classical [VC, Vita], and color-ordered Trubyte Bioform porcelain shade guide [TB, Dentsply International, York, PA, USA]) were analyzed (N=3). A circular area (d=1.7 mm) on the middle of the labial surface of the tab, excluding the cervical portion, was measured with a spectroradiometer (D65, 2). Whiteness and yellowness indices were computed. Data were analyzed by analysis of variance and Fisher's PLSD test at a 0.05 level of significance. Results:, The range of color difference (,E*) from the lightest to the darkest tab was 33.8 (BG), 17.1 (VC), and 23.2 (TB). Mean values of ,E* among pairs of adjacent tabs were 3.0 (BG), 4.2 (VC), and 3.3 (TB). BG exhibited the highest R2 values between color coordinate pairs and between whiteness and yellowness indices with the respective color coordinates. Conclusions:, BG exhibited the widest color range and had the most consistent color distribution as compared with the two commercial products. Extension of the lightness range of BG toward higher L* values (bleach shades) was confirmed. CLINICAL SIGNIFICANCE A dental shade guide that is colorimetrically uniform might increase the reliability of visual comparisons of tooth whitening efficacy, whereas the inclusion of realistic bleaching shades in the shade guide will complement contemporary esthetic dentistry. Disturbance and reef topography maintain high local diversity in Ecklonia radiata kelp forests

Disturbance of competitive-dominant plant and algae canopies often lead to increased diversity of the assemblage. Kelp forests, particularly those of temperate Western Australia, are habitats with high alpha diversity. This study investigated the roles of broad-scale canopy loss and local scale reef topography on structuring the kelp-dominated macroalgal forests in Western Australia. Eighteen 314,m2 circular areas were cleared of their Ecklonia radiata canopy and eighteen controls were established across three locations. The patterns of macroalgal recolonisation in replicate clearances were observed over a 34,month period. Macroalgal species richness initially increased after canopy removal with a turf of filamentous and foliose macroalgae dominating cleared areas for up to seven months. A dense Sargassum canopy dominated cleared areas from 11 to 22,months. By 34,months, partial recovery of the kelp canopy into cleared areas had occurred. Some cleared areas did not follow this trajectory but remained dominated by turfing, foliose and filamentous algae. As kelp canopies developed, the initial high species diversity declined but still remained elevated relative to undisturbed controls, even after 34 months. More complex reef topography was associated with greater variability in the algal assemblage between replicate quadrats suggesting colonising algae had a greater choice of microhabitats available to them on topographically complex reefs. Shading by canopies of either Sargassum spp. and E. radiata are proposed to highly influence the abundance of algae through competitive exclusion that is relaxed by disturbance of the canopy. Disturbance of the canopy in E. radiata kelp forests created a mosaic of different patch types (turf, Sargassum -dominated, kelp-dominated). These patch types were both transient and stable over the 34 months of this study, and are a potential contemporary process that maintains high species diversity in temperate kelp-dominated reefs.
