Numeric Simulation of Ion-Site Association Effects in Ion-Selective Electrode Response

Abstract Effects in ion-selective electrode response and selectivity, caused by ion-site association in membranes are studied by means of numeric simulations based on a generalized model, which does not rely on a certain degree of the dissociation of electrolytes in membranes. The variability of the experimental values of the potentiometric selectivity coefficients is considered in view of the association in membranes. The reasons why the "dissociation approach" often fits experimental data are also discussed. A novel version of segmented sandwich membrane method is proposed for direct potentiometric measurements of ion-site association constants in membranes, and preliminary experimental results are presented. Host-related life history traits in interspecific hybrids of cactophilic Drosophila

Abstract In the genus Drosophila (Diptera: Drosophilidae), interspecific hybridization is a rare phenomenon. However, recent evidence suggests a certain degree of introgression between the cactophilic siblings Drosophila buzzatii Patterson & Wheeler and Drosophila koepferae Fontdevila & Wasserman. In this article, we analyzed larval viability and developmental time of hybrids between males of D. buzzatii and females of D. koepferae, raised in media prepared with fermenting tissues of natural host plants that these species utilize in nature as breeding sites. In all cases, developmental time and larval viability in hybrids was not significantly different from parental lines and, depending on the cross, hybrids developed faster than both parental species or than the slowest species. When data of wing length were included in a discriminant function analysis, we observed that both species can be clearly differentiated, while hybrids fell in two categories, one intermediate between parental species and the other consisting of extreme phenotypes. Thus, our results point out that hybrid fitness, as measured by developmental time and viability, is not lower than in the parental species. [source]


Clonal dynamics of tumor-infiltrating lymphocytes

Abstract The presence of tumor-infiltrating lymphocytes (TIL) provides important evidence of anti-tumor immunity in vivo. However, TIL are usually not sufficient for inhibiting tumor growth. We explored the spatial and temporal aspects of clonal accumulation of TIL using RT-PCR/single-strand conformation polymorphism analysis. In CMS5 fibrosarcomas in BALB/c mice, accumulated T,cell clones were specific in that dominant TIL were identical between distant tumors. Moreover, dominant TIL in the first tumor appeared consistently in the second tumor inoculated after formation of the first tumor. These results suggest that TIL show a certain level of specific tumor surveillance. When we characterized CD4+ and CD8+ TIL separately, CD8+ TIL were highly concentrated and persistently localized at the tumor site, while most CD4+ TIL clones were less concentrated and less persistent. A functional analysis showed that TIL had a certain degree of anti-tumor activity when CD4+ and CD8+ TIL were co-transferred. Co-transfer of CD4+ and CD8+ TIL exhibited equivalent anti-tumor activity, irrespective of tumor stage. However, the numbers of TIL did not increase after the early phase of tumor progression. These data suggest that TIL are specific to the tumor and potentially retain anti-tumor activity, although their accumulation in mice is impaired. [source]


Metal Ion Complementarity: Effect of Ring-Size Variation on the Conformation and Stability of Lead(II) and Cadmium(II) Complexes with Pendant-Armed Crowns

Abstract The binding tendencies of the pendant-armed crown ethers L1,L3 [L1 = N,N, -bis(benzimidazol-2-ylmethyl)-1,7-diaza-12-crown-4, L2 = N,N, -bis(benzimidazol-2-ylmethyl)-1,10-diaza-15-crown-5) and L3 = N,N, -bis(benzimidazol-2-ylmethyl)-4,13-diaza-18-crown-6] towards PbII and CdII have been investigated. The X-ray crystal structure of [Cd(L3)](ClO4)2·EtOH shows that, in the solid state, the CdII ion is eight-coordinate and fits quite well into the crown hole, favouring an anti arrangement of the organic receptor. NMR measurements recorded in acetonitrile solution indicate that increasing the crown size induces a conformational change in the series of CdII complexes. The conformation goes from a syn arrangement for L1 to an anti arrangement for L3, passing through a syn [lrarr2] anti equilibrium in the complex derived from L2. On the contrary, no conformational change was observed for the corresponding PbII complexes, which have a syn conformation in all cases. These results have been confirmed by means of density functional theory (DFT) calculations performed by using the B3LYP model. The binding constants obtained from UV/Vis titration experiments in DMSO solution demonstrate that a decrease in the crown size provokes a 102 -fold enhancement of the stability for this series of CdII complexes, whereas for PbII a gradual decrease of the binding constants is observed. Spontaneous recurrent network activity in organotypic rat hippocampal slices

Abstract Organotypic hippocampal slices were prepared from postnatal day 4 rats and maintained in culture for >6 weeks. Cultured slices exhibited from 12 days in vitro spontaneous events which closely resembled giant depolarizing potentials (GDPs) recorded in neonatal hippocampal slices. GDP-like events occurred over the entire hippocampus with a delay of 30,60 ms between two adjacent regions as demonstrated by pair recordings from CA3,CA3, CA3,CA1 and interneurone,CA3 pyramidal cells. As in acute slices, spontaneous recurrent events were generated by the interplay of GABA and glutamate acting on AMPA receptors as they were reversibly blocked by bicuculline and 6,7-dinitroquinoxaline-2,3-dione but not by dl -2-amino-5-phosphonopentaoic acid. The equilibrium potentials for GABA measured in whole cell and gramicidin-perforated patch from interconnected interneurones,CA3 pyramidal cells were ,70 and ,56 mV, respectively. The resting membrane potential estimated from the reversal of N -methyl- d -aspartate-induced single-channel currents in cell-attach experiments was ,75 mV. In spite of its depolarizing action, in the majority of cases GABA was still inhibitory as it blocked the firing of principal cells. The increased level of glutamatergic connectivity certainly contributed to network synchronization and to the development of interictal discharges after prolonged exposure to bicuculline. In spite of its inhibitory action, in a minority of cells GABA was still depolarizing and excitatory as it was able to bring principal cells to fire, suggesting that a certain degree of immaturity is still present in cultured slices. This was in line with the transient bicuculline-induced block of GDPs and with the isoguvacine-induced increase of GDP frequency. [source]


Hedging Affecting Firm Value via Financing and Investment: Evidence from Property Insurance Use

I provide evidence about the value effects of alternative risk management by examining corporate purchase of property insurance, a commonly used pure hedge of asset-loss risks. Using an insurance data set from China, I find that there is an inverted U-shape effect of the extent of property insurance use on firm value measured by several versions of Tobin's Q. Therefore, the use of property insurance, to a certain degree, has a positive effect on firm value; however, over insurance appears detrimental to firm value. Given that the inflection points occur at relatively high levels of the observed insurance spending, insurance use appears beneficial to the majority of my sample firms. The estimated average hedging premium is about 1.5%. I demonstrate that an avenue for insurance to create value in China is that it helps firms secure valuable new debt financing and enhance investment. [source]


Effect of the Molecular Size of Analytes on Polydiacetylene Chromism

Abstract The pH chromism of polydiacetylenes (PDAs) is examined with respect to the molecular size and acidity of acid analytes, along with the alkyl spacer length of primary-amine-functionalized diacetylene (DA) lipids. pH turns out to be an important parameter to charge amine headgroups of PDA but a change in pH does not necessarily result in a PDA color change. The molecular size of acid analytes is identified as another factor that can produce a configurational change in PDA amine headgroups, followed by perturbation of the ene,yne conjugated backbone. In addition, the length of a flexible alkyl spacer between the amine headgroup and the amide group of the diacetylene lipids is found to strongly affect the degree of PDA chromatic transition. The longer alkyl spacer shows a smaller chromatic transition from blue to red phase. The alkyl spacer seems to provide a certain degree of freedom to the amine headgroup, thus decreasing the transfer of headgroup steric effects to the PDA backbone. These correlations found for PDA chromism are applied to the development of a system that colorimetrically detects diethyl phosphate (DEP), a degraded nerve agent simulant. PDA liposomes show a selective chromatic transition upon binding with DEP compared to other acid analytes. [source]


On scientists' discomfort in fisheries advisory science: the example of simulation-based fisheries management-strategy evaluations

Abstract Scientists feel discomfort when they are asked to create certainty, where none exists, for use as an alibi in policy-making. Recently, the scientific literature has drawn attention to some pitfalls of simulation-based fisheries management-strategy evaluation (MSE). For example, while estimates concerning central tendencies of distributions of simulation outcomes are usually fairly robust because they are conditioned on ample data, estimates concerning the tails of distributions (such as the probability of falling below a critical biomass) are usually conditional on few data and thus often rely on assumptions that have no strong knowledge base. The clients of scientific advice, such as the European Commission, are embracing the mechanization of the evaluation of proposed Harvest Control Rules against the precautionary principle and management objectives. Where the fisheries management institutions aim for simple answers from the scientists, giving ,green/red light' to a proposed management strategy, the scientists are forced into a split position between satisfying the demands of their advisory role and living up to the standards of scientific rigour. We argue against the mechanization of scientific advice that aims to incorporate all relevant processes into one big model algorithm that, after construction, can be run without circumspection. We rather encourage that fisheries advice should be a dynamic process of expert judgement, incorporating separate parallel concurrent, lines of scientific evidence, from quantitative and qualitative modelling exercises and factual knowledge of the biology and the fishery dynamics. This process can be formalized to a certain degree and can easily accommodate stakeholder viewpoints. [source]


Evaluation of the clinical efficacy of a mouthwash and oral gel containing the antimicrobial proteins lactoperoxidase, lysozyme and lactoferrin in elderly patients with dry mouth , a pilot study

Objectives:, To evaluate the clinical efficacy of a mouthwash and oral gel containing the antimicrobial proteins lactoperoxidase, lactoferrin and lysozyme, in a sample of elderly individuals with dry mouth. Material and methods:, Twenty elderly institutionalised subjects with dry mouth and with a certain degree of independence for daily life activities were included in this pilot study. A randomised, double blind and cross-over design was used. The study variables comprised subjective dry mouth sensation, the severity of discomfort assessed by means of a visual analogical scale (VAS), the Oral Health Impact Profile (OHIP), the presence of signs and symptoms of dry mouth, sialometry and Candida albicans culture. All the variables were recorded before and after each of the two periods of the study. Results:, The 20 selected subjects we made up of 16 women and four men, with a mean age of 81.3 years. Improvement was observed on analysing the data between the first and second intervention period in terms of the OHIP values, the presence of dry mouth, and the need to drink fluids to swallow. However, the improvement in certain variables before and after treatment did not take a positive course in all cases, and some subjects even improved with placebo. Conclusions:, The evaluated mouthwash and oral gel improved some subjective and clinical aspects in elderly individuals with dry mouth, though a placebo effect cannot be entirely discarded. Numerical Modeling of Unsaturated Flow in Wastewater Soil Absorption Systems

It is common practice in the United States to use wastewater soil absorption systems (WSAS) to treat domestic wastewater. WSAS are expected to provide efficient, long-term removal of wastewater contaminants prior to ground water recharge. Soil clogging at the infiltrative surface of WSAS occurs due to the accumulation of suspended solids, organic matter, and chemical precipitates during continued wastewater infiltration. This clogging zone (CZ) creates an impedance to flow, restricting the hydraulic conductivity and rate of infiltration. A certain degree of clogging may improve the treatment of wastewater by enhancing purification processes, in part because unsaturated flow is induced and residence times are significantly increased. However, if clogging becomes excessive, the wastewater pond height at the infiltrative surface can rise to a level where system failure occurs. The numerical model HYDRUS-2D is used to simulate unsaturated flow within WSAS to better understand the effect of CZs on unsaturated flow behavior and hydraulic retention times in sandy and silty soil. The simulations indicate that sand-based WSAS with mature CZs are characterized by a more widely distributed flow regime and longer hydraulic retention times. The impact of clogging on water flow within the silt is not as substantial. For sand, increasing the hydraulic resistance of the CZ by a factor of three to four requires an increase in the pond height by as much as a factor of five to achieve the same wastewater loading. Because the degree of CZ resistance directly influences the pond height within a system, understanding the influence of the CZ on flow regimes in WSAS is critical in optimizing system design to achieve the desired pollutant-treatment efficiency and to prolong system life. Southern hemisphere cyclones and anticyclones: recent trends and links with decadal variability in the Pacific Ocean

Abstract The aim of this paper is to study the association between the extratropical Southern Hemisphere and the decadal variability in the Pacific Ocean (PO). We discuss a pattern of coherent large-scale anomalies and trends in cyclone and anticyclone behaviour in light of the climate variability in the PO over the ERA40 reanalysis period (1957,2002). The two representative PO indices are the Pacific Decadal and Interdecadal Oscillations (PDO and IPO), and here the PDO is chosen owing to it being less associated with the southern oscillation index (SOI). Composites of the indicators of the density and intensity of cyclones/anticyclones given by an automatic tracking scheme were calculated for the years when the PDOI was more than one standard deviation above or below its mean. Although the ERA40 is not free from noise and assimilation changes, the results show a large-scale feature, which seems to be robust and agrees with earlier studies using different data sets. The sea-level pressure shows a strong annular structure related to the PDO, which is not seen for the SOI, with lower pressure around Antarctica during the positive phase and vice versa. More intense (and fewer) cyclones and anticyclones were observed during the positive PDO. This is less consistent for the SOI, particularly during the summer when a different PDO/SOI pattern arises at high latitudes. The trends project a pattern coincident with the positive PDO phase and seem to be linked with the main climate shift in the late seventies. Trends observed over the Tasman Sea are consistent with declining winter rainfall over southeastern Australia. Most patterns are statistically significant and seem robust, but random changes in ENSO may play a part, to a certain degree, in modulating the results, and a physical mechanism of causality has not been demonstrated. Although global warming and related changes in the Southern Annular Mode (SAM) may also help explain the observed behaviour, the large-scale response presented here provides a new insight and would be of considerable interest for further modelling studies. Comparison of different earth radiation budget experiment data sets over tropical oceans

Abstract We compare radiation budgets derived from different Earth Radiation Budget Experiment (ERBE) archives over the tropical oceans (30S and 30N) from 1985 to 1989. Two ERBE data sets are used. One is taken from the Earth Radiation Budget Satellite (ERBS), and the other from the combined ERBS, National Oceanic and Atmospheric Administration (NOAA) 9, and NOAA 10 satellites. The domain-mean all-sky outgoing longwave radiation (OLR) derived from the combined data set shows a notable change in early 1987 when NOAA 10 replaced NOAA 9. This change is also found in longwave (LW) cloud radiative forcing (CRF), all-sky shortwave (SW) radiation, and SWCRF. The ERBS, however, does not show such changes. We also examine the sensitivity of cloud,radiation interaction to the sea surface temperature (SST) of the tropical oceans. In general, each component of radiative feedbacks derived from the two ERBE data have the same sign, although they show a certain degree of discrepancy in the magnitude. The discrepancy is more notable for averaged quantities over the entire tropical oceans, particularly over the subtropics where convective activities are relatively weak. The combined data show a larger sensitivity of LWCRF and SWCRF to SST than those of the ERBS, consistent with the above results. The response of clouds to an increase in SST has a net cooling effect when using the combined data but has a net heating effect when using the ERBS data (,0.80 W m,2 K,1 versus 0.48 W m,2 K,1). Most of the discrepancies of the net CRF between the two ERBE data sets can be accounted for by the difference in the sensitivity of all-sky OLR (4.52 W m,2 K,1 versus 1.73 W m,2 K,1). Hepatic progenitor cells, stem cells, and AFP expression in models of liver injury

Abstract Adult hepatocytes and liver-cell progenitors play a role in restoring liver tissue after injury. For the study of progenitor cells in liver repair, experimental models included (a) surgical removal of liver tissue by partial hepatectomy; (b) acute injury by carbontetrachloride; (c) acute injury by d -galactosamine (GalN) and N -nitrosomorpholine (NNM); and (d) chemical hepatocarcinogenesis by feeding NNM in low and high doses. Serological and immunohistological detection of alpha-fetoprotein gene expression served to follow pathways of cellular differentiation. Stem cells were not required in models of surgical removal of parenchyma and in carbon tetrachloride intoxication of adult hepatocytes. In contrast, regeneration of liver occurred through biliary epithelial cells in injuries induced by GalN and NNM. These biliary epithelial cells, collectively called oval cells, are most probably derived from the canals of Hering. Proliferating bile duct cells reached a level of differentiation with reactivation of foetal genes and significant alpha-1-fetoprotein (AFP) synthesis signalling a certain degree of retrodifferentiation with potential stemness. Due to the same embryonic origin of bile ducts and hepatocytes, biliary epithelium and its proliferating progeny (oval cells) have a defined role in liver regeneration as a transit and amplification compartment. In their early proliferation stage, oval cells were heavily engaged in DNA synthesis ([3H]thymidine labelling). Pulse-chase experiments during experimental hepatocarcinogenesis exhibited their development into hepatocytes with high risk for transformation and leading to foci of altered hepatocytes. Hepatocellular carcinomas may arise either from proliferating/differentiating oval cells or from adult hepatocytes; both cell types have stem-like properties. AFP-positive and AFP-negative carcinomas occurred in the same liver. They may represent random clonal origin. The heterogeneity of phenotypic marker (AFP) correlated with a process of retrodifferentiation. [source]

Summary Protein gelation is important to obtain desirable sensory and textural structures in foods. Gelation phenomenon requires a driving force to unfold the native protein structure, followed by an aggregation retaining a certain degree of order in the matrix formed by association between protein strands. Protein gelation has been traditionally achieved by heating, but some physical and chemical processes form protein gels in an analogous way to heat-induction. A physical means, besides heat, is high pressure. Chemical means are acidification, enzymatic cross-linking, and use of salts and urea, causing modifications in protein,protein and protein,medium interactions. The characteristics of each gel are different and dependent upon factors like protein concentration, degree of denaturation caused by pH, temperature, ionic strength and/or pressure. [source]


Migrants as Minorities: Integration and Inclusion in the Enlarged European Union,

The developing migration and asylum law and policy of the European Union aim to construct a common normative framework to address the admission and residence of diverse categories of third-country nationals in EU territory. The principles of minority protection, however, are absent from EU law, with the exception of some references in the new Constitutional Treaty and the incorporated Charter of Fundamental Rights, although they have been employed, to a certain degree, in a prescriptive and pragmatic way in the context of the accession of new Member States. However, increased EU attention to the concept of integration in recent Council policy pronouncements and newly adopted legal measures, aimed almost exclusively at lawfully resident third-country nationals, provides a space where migration policy and minority protection principles may engage more directly. This article undertakes a preliminary assessment of the points of convergence and divergence in these two sets of principles, and argues that greater convergence would result in a more coherent EU policy on integration. Domestic Politics, European Norms, the Sanctions Against Austria
In February 2000, 14 EU Member States collectively took the unprecedented step of imposing bilateral sanctions on their Austrian EU partner. How can this be explained? Was it, as the 14 governments argued, because the inclusion in the Austrian government of Jörg Haider's extreme right FPö opposes many of the ideas making up the common identity of the EU? Or, were the sanctions motivated, as the Austrian government argued, by narrow-minded party political interests that lurked beneath the rhetoric of shared European norms and values? Our analysis suggests that, without the particular concerns about domestic politics of certain politicians, it is unlikely that the sanctions against Austria would have been adopted in this form. On the other hand, without the recent establishment of concerns about human rights and democratic principles as an EU norm, it is unlikely that these particular sanctions would have been adopted collectively by all member governments. Thus, while norms might have been used instrumentally, such instrumental use only works, in the sense of inducing compliant behaviour, if the norms have acquired a certain degree of taken-for-grantedness within the relevant group of actors or institution. Background., Demands, thresholds and fragmentation of services hinder the accessibility of health care, such that some severe mentally ill people do not receive the treatment they need or avoid healthcare services. Methods of establishing contact and gaining trust in mental health care include practical assistance, realistic expectations, establishing long-term goals, empathy and a client-centred and flexible approach. Method., A public mental healthcare practice in The Netherlands with outstanding performance was studied from 2002,2007 using participant observation, interviews with experienced care providers and interviews with clients with a long history of avoiding care facilities, conflicts and troubled relationships with care providers. Findings., A number of personal qualities are vital for establishing contact and gaining trust with these clients: altruism, a degree of compassion, loyalty, involvement, tenacity, a critical attitude to the mainstream, flexibility, optimism, diplomacy, patience, creativity, and a certain degree of immunity to stress. Conclusion., Care providers who establish contact and win trust employ ,non-judgemental appreciation'. They start from the acceptance of what is and try to connect with the client and their world. These professionals use their initial actions to identify and praise qualities and achievements of clients. This style of work is supported by a set of deep-rooted personal qualities we can summarize as ,empathy'. [source]


Structural Diversification of Macrolactones by Substrate-Flexible Cytochrome P450 Monooxygenases

Abstract The substrate flexibilities of several cytochrome P450 monooxygenases involved in macrolide biosynthesis were investigated to test their potential for the generation of novel macrolides. PikC hydroxylase in the pikromycin producer Streptomyces venezuelae accepted oleandomycin as an alternative substrate and introduced a hydroxy group at the C-4 position, which is different from the intrinsic C-12 hydroxylation position in the natural substrate. This is the first report of C-4 hydroxylation activity of cytochrome P450 monooxygenase involved in the biosynthesis of 14-membered macrolides. EryF hydroxylase from the erythromycin biosynthetic pathway of Saccharopolyspora erythraea and OleP oxidase from the oleandomycin biosynthetic pathway of Streptomyces antibioticus also showed a certain degree of plasticity towards alternative substrates. In particular, EryF and OleP were found to oxidize a 12-membered macrolactone as an alternative substrate. These results demonstrate the potential usefulness of these enzymes to diversify macrolactones by post-PKS oxidations. [source]

Summary The distinctive morphological features of the ovaries the ,cachara', Pseudoplatystoma fasciatum were characterized macroscopically, and by histology, when reared in cages, from March 2005 to February 2006. Forty eight females (mean total weight = 2.7 kg, mean standard length = 65.1 cm) were allocated to four cages of 2.7 m3 (20 fish/cage) which were installed in four 600 m2 ponds, located at the IP, Pariquera-Açu, São Paulo, Brazil. The monthly, samples were fixed in 4%-buffered formalin before preparation for histological examination, ovaries were removed and weighted. The gonadosomatic index (GSI) was calculated as = 100 × weight ovaries/total fish weight. The ovaries are the cystovarian type and macroscopically, were established three stages of ovarian maturation: Resting, developing Maturation (initial, intermediate, final) and Regression (initial, intermediate, final). Based on morphological criteria of those ovaries, the oocyte development has been divided into distinct stages: (i) oocyte growth (vitellogenesis); (ii) oocyte maturation, along which it goes through different phases of development, before (iii) ovulation and, (iv) spawning. When the P. fasciatum were kept in confinement and not induced to breed occurs fail to attain final oocyte maturation, start the process of degeneration. Consequently, the weight started to decline and 45% of the ovaries showed atresia of vitellogenic follicles. This was considered indicative of a recent cessation of the reproductive activity. Such failure could have been caused by stress of the monthly sampling involving a certain degree of disturbance, and perhaps also by the existence of stressors while in captivity. The synchronous ovary contained oocytes in an unique stage of development and had potential to perform total spawning up to one time a year, with the period reproductive beginning in the end of November to the beginning of February, coinciding with the highest water temperatures in the experimental cages (29.0,31.5°C) and the increase of mean values of GSI. During the regression phase, residual oocytes could be observed together with decrease of the mean values of GSI and, the temperatures. [source]


Distribution of acrylic acid grafted chains introduced into polyethylene film by simultaneous radiation grafting with water and ethanol as solvents

Abstract The graft copolymerization of acrylic acid onto low-density polyethylene films by simultaneous ,-ray irradiation was carried out. The effect of water and ethanol as grafting solvents on the distribution of grafted poly (acrylic acid) in the low-density polyethylene films was studied with optical microscopy observations of dyed and sliced samples and attenuated total reflection/Fourier infrared spectroscopy analysis. When no vigorous homopolymerization occurred, both polyethylene and poly(acrylic acid) existed in the grafted layer, and the thickness of the grafted layer and the poly(acrylic acid) concentration in the grafted layer increased with an increasing degree of grafting, regardless of the grafting conditions, the former increasing faster than the latter. In comparison with water as the solvent, in the absence of the inhibitor, homopolymerization could be suppressed to a certain degree in the ethanol solvent system, whereas in the presence of the inhibitor, obvious homopolymerization occurred at a lower monomer concentration, and both the degree of grafting and the thickness of the grafted layer were lower. Such differences could be explained by the chain transfer and the relatively low solubility of poly(acrylic acid) in ethanol. Jurors' Perception of Witnesses with Intellectual Disabilities and the Influence of Expert Evidence

Purpose, The aim of this study was to assess mock-jurors' perceptions of the evidence of witnesses with intellectual disabilities either with or without expert evidence and in comparison with witnesses from the general population. Method, Sixty participants read transcripts of a mock trial focusing on the testimony of an eyewitness. Participants were assigned to one of three groups. The first was told that the witness was a person from the general population. The second was told that the witness had mild learning disabilities. The third group was told that the witness had mild learning disabilities and was given expert evidence concerning his abilities. Results, While mock-jurors perceive witnesses with learning disabilities to be fundamentally honest, they are reluctant to rely on the evidence provided by witnesses with learning disabilities. Expert evidence can go some way to ameliorating the negative perceptions of the reliability of witnesses with learning disabilities. Conclusions, Expert evidence can provide jurors with a certain degree of insight and understanding of an individual witness with intellectual disabilities that potentially increases the likelihood of achieving justice. Noninvasive Control of Adequate Cerebral Oxygenation During Low-Flow Antegrade Selective Cerebral Perfusion on Adults and Infants in the Aortic Arch Surgery

Background: Aortic arch repair techniques using low-flow antegrade selective cerebral perfusion have been standardized to a certain degree. However, some of the often-stated beneficial effects have never been proven. Especially, the existence of an adequate continuous flow in both cerebral hemispheres during the surgical procedure still remains unclear as the monitoring of an effective perfusion remains a nonstandardized technique. Methods: Seventeen patients underwent surgical reconstruction of the aortic arch due to aortic aneurysm surgery (adult group n = 8 patients) or of the hypoplastic aortic arch due to hypoplastic left heart syndrome (HLHS) or aortic coarctation (infant group n = 9 patients) under general anesthesia and mild hypothermia (adult group 28 °C; infant group 25 °C). Mean weights were 92.75 ± 14.00 kg and 4.29 ± 1.32 kg, and mean ages were 58.25 ± 10.19 years and 55.67 ± 51.11 days in the adult group and the infant group, respectively. The cerebral O2 saturation measurement was performed by continuous plotting of the somatic reflectance oximetry of the frontal regional tissue on both cerebral hemispheres (rSO2, INVOS®; Somanetics Corporation, Troy, MI, USA). Results: During low-flow antegrade perfusion via innominate artery, continuous plots with similar values of O2 saturation (rSO2) in both cerebral hemispheres were observed, whereas a decrease in the rSO2 values below the desaturation threshold correlated with a displacement or an incorrect positioning of the arterial cannula in the right subclavian artery. Conclusions: Continuous monitorization of the cerebral O2 saturation during aortic arch surgery in adults and infants is a feasible technique to control an adequate cannula positioning and to optimize clinical outcomes avoiding neurological complications related to cerebral malperfusion. Restoration of DNA-binding and growth-suppressive activity of mutant forms of p53 via a PCAF-mediated acetylation pathway,

Tumor-derived mutant forms of p53 compromise its DNA binding, transcriptional, and growth regulatory activity in a manner that is dependent upon the cell-type and the type of mutation. Given the high frequency of p53 mutations in human tumors, reactivation of the p53 pathway has been widely proposed as beneficial for cancer therapy. In support of this possibility p53 mutants possess a certain degree of conformational flexibility that allows for re-induction of function by a number of structurally different artificial compounds or by short peptides. This raises the question of whether physiological pathways for p53 mutant reactivation also exist and can be exploited therapeutically. The activity of wild-type p53 is modulated by various acetyl-transferases and deacetylases, but whether acetylation influences signaling by p53 mutant is still unknown. Here, we show that the PCAF acetyl-transferase is down-regulated in tumors harboring p53 mutants, where its re-expression leads to p53 acetylation and to cell death. Furthermore, acetylation restores the DNA-binding ability of p53 mutants in vitro and expression of PCAF, or treatment with deacetylase inhibitors, promotes their binding to p53-regulated promoters and transcriptional activity in vivo. These data suggest that PCAF-mediated acetylation rescues activity of at least a set of p53 mutations. Therefore, we propose that dis-regulation of PCAF activity is a pre-requisite for p53 mutant loss of function and for the oncogenic potential acquired by neoplastic cells expressing these proteins. Development of a membrane-assisted hybrid bioreactor for ammonia and COD removal in wastewaters

Abstract A new membrane-assisted hybrid bioreactor was developed to remove ammonia and organic matter. This system was composed of a hybrid circulating bed reactor (CBR) coupled in series to an ultrafiltration membrane module for biomass separation. The growth of biomass both in suspension and biofilms was promoted in the hybrid reactor. The system was operated for 103 days, during which a constant ammonia loading rate (ALR) was fed to the system. The COD/N-NH4+ ratio was manipulated between 0 and 4, in order to study the effects of different organic matter concentrations on the nitrification capacity of the system. Experimental results have shown that it was feasible to operate with a membrane hybrid system attaining 99% chemical oxygen demand (COD) removal and ammonia conversion. The ALR was 0.92 kg N-NH4+ m,3 d,1 and the organic loading rate (OLR) achieved up to 3.6 kg COD m,3 d,1. Also, the concentration of ammonia in the effluent was low, 1 mg N-NH4+ dm,3. Specific activity determinations have shown that there was a certain degree of segregation of nitrifiers and heterotrophs between the two biomass phases in the system. Growth of the slow-growing nitrifiers took place preferentially in the biofilm and the fast-growing heterotrophs grew in suspension. This fact allowed the nitrifying activity in the biofilm be maintained around 0.8 g N g,1 protein d,1, regardless of the addition of organic matter in the influent. Multi-way models for sensory profiling data

Abstract One of the problems in analyzing sensory profiling data is to handle the systematic individual differences in the assessments from different panelists. It is unavoidable that different persons have, at least to a certain degree, different perceptions of the samples as well as a different understanding of the attributes or of the scales used for quantifying the assessments. Hence, any model attempting to describe sensory profiling data needs to deal with individual differences; either implicitly or explicitly. In this paper, a unifying family of models is proposed based on (i) the assumption that latent variables are appropriate for sensory data, and (ii) that individual differences occur. Based on how individual differences occur, various mathematical models can be constructed, all aiming at modeling simultaneously the sample-specific variation and the panelist-specific variation. The model family includes Principal Component Analysis (PCA) and PARAllel FACtor analysis (PARAFAC). The paper can be viewed as extending the latent variable approach commonly based on PCA to multi-way models that specifically take certain panelist-variations into account. The proposed model family is focused on analyzing data from quantitative descriptive analysis with fixed vocabulary, but it also provides a foundation upon which comparisons, extensions and further developments can be made. An example is given which shows that even for well-working data, models handling individual differences can shed important light on differences between the quality of the data from individual panelists. Notes on quantitative structure-properties relationships (QSPR) (1): A discussion on a QSPR dimensionality paradox (QSPR DP) and its quantum resolution

Abstract Classical quantitative structure-properties relationship (QSPR) statistical techniques unavoidably present an inherent paradoxical computational context. They rely on the definition of a Gram matrix in descriptor spaces, which is used afterwards to reduce the original dimension via several possible kinds of algebraic manipulations. From there, effective models for the computation of unknown properties of known molecular structures are obtained. However, the reduced descriptor dimension causes linear dependence within the set of discrete vector molecular representations, leading to positive semi-definite Gram matrices in molecular spaces. To resolve this QSPR dimensionality paradox (QSPR DP) here is proposed to adopt as starting point the quantum QSPR (QQSPR) computational framework perspective, where density functions act as infinite dimensional descriptors. The fundamental QQSPR equation, deduced from employing quantum expectation value numerical evaluation, can be approximately solved in order to obtain models exempt of the QSPR DP. The substitution of the quantum similarity matrix by an empirical Gram matrix in molecular spaces, build up with the original non manipulated discrete molecular descriptor vectors, permits to obtain classical QSPR models with the same characteristics as in QQSPR, that is: possessing a certain degree of causality and explicitly independent of the descriptor dimension. © 2008 Wiley Periodicals, Inc. Prediction of integral membrane protein type by collocated hydrophobic amino acid pairs

Abstract A computational model, IMP-TYPE, is proposed for the classification of five types of integral membrane proteins from protein sequence. The proposed model aims not only at providing accurate predictions but most importantly it incorporates interesting and transparent biological patterns. When contrasted with the best-performing existing models, IMP-TYPE reduces the error rates of these methods by 19 and 34% for two out-of-sample tests performed on benchmark datasets. Our empirical evaluations also show that the proposed method provides even bigger improvements, i.e., 29 and 45% error rate reductions, when predictions are performed for sequences that share low (40%) identity with sequences from the training dataset. We also show that IMP-TYPE can be used in a standalone mode, i.e., it duplicates significant majority of correct predictions provided by other leading methods, while providing additional correct predictions which are incorrectly classified by the other methods. Our method computes predictions using a Support Vector Machine classifier that takes feature-based encoded sequence as its input. The input feature set includes hydrophobic AA pairs, which were selected by utilizing a consensus of three feature selection algorithms. The hydrophobic residues that build up the AA pairs used by our method are shown to be associated with the formation of transmembrane helices in a few recent studies concerning integral membrane proteins. Our study also indicates that Met and Phe display a certain degree of hydrophobicity, which may be more crucial than their polarity or aromaticity when they occur in the transmembrane segments. This conclusion is supported by a recent study on potential of mean force for membrane protein folding and a study of scales for membrane propensity of amino acids. © 2008 Wiley Periodicals, Inc. Song perception among incipient species as a mechanism for reproductive isolation

Abstract Many functions in behavioural processes of small passerines are regulated via vocal messages. Song plays an important role in the development of reproductive barriers and thus playback experiments can often be used for investigating the potential for reproductive isolation through behavioural mechanisms. Moltoni's warbler Sylvia (cantillans) moltonii is characterized by diagnostic vocalizations and a peculiar pattern of distribution, being parapatric and partly sympatric with the nominate Sylvia c. cantillans. With this work, we test whether these two closely related taxa react equally to their own song and to the song of the other taxon, shedding light on whether they perceive each other's songs as coming from the same species. We carried out 184 playback experiments within the mainland range of the two forms. We judged the response of the bird on a scale of scores. Each taxon responded more strongly to playback when faced with the song of its own taxon. This held true when applied only to males or females. Additionally, birds tested for both songs showed a stronger response to the song of their own taxon. The distributional context (sympatry vs. allopatry) did not affect bird response. Results indicate that a certain degree of reproductive isolation between the two taxa (because of diverged mate recognition systems) already exists; consistently with genetic data and with the peculiar pattern of distribution; this suggests that the two taxa have reached species status. Magnesium and biological activity of oxytocin analogues modified on aromatic ring of amino acid in position 2

Abstract For the purpose of evaluating substitution effects in the ortho, meta or para positions of the aromatic ring of tyrosine or phenylalanine in position 2 of oxytocin on uterotonic activity in vitro in the presence and absence of magnesium ions, six new analogues of oxytocin ([,,- and ,,- m -methylphenylalanine2]oxytocin, [,,- and ,,- m -methoxyphenylalanine2]oxytocin and [,,- and ,,- o -methyltyrosine2]oxytocin) were synthesized and several previously described analogues resynthesized. For the phenylalanine series, it is found that, in the absence of magnesium ions, substitution of the ortho and meta positions leads to loss of intrinsic activity (the analogues are antagonists) in contrast to the para position. In the tyrosine series, only methyl substitution in the meta position has this effect (substitution of ortho position only attenuates the agonistic biological activity). Addition of Mg ions restores to a certain degree the agonistic activity in the case of the o -methylphenylalanine analogue and enhances the agonistic activity of o -methyltyrosine oxytocin. All other analogues keep the original qualities as in the absence of Mg. Molecular modelling calculations of the structure of the above analogues was carried out to help explain these findings of the molecular level. The Interaction of Plasmodiophora brassicae and Arabidopsis thaliana: Parameters for Disease Quantification and Screening of Mutant Lines

Abstract The soil-borne obligate pathogen Plasmodiophora brassicae causes clubroot disease in species of Brassicaceae, including Arabidopsis thaliana. The host,pathogen interaction was studied with respect to the age of the plant at the time point of inoculation and to different infection pressures in order to establish a standardization of infection parameters and evaluation of disease extent for A. thaliana lines. Spore number per root weight, root and shoot weight of inoculated and non-inoculated plants as well as infection rate and disease index (DI) were analysed and correlated. The disease extent of different lines was comparable as measured by the relation of root weight of inoculated and non-inoculated plants (Ri/Rni index) and the DI. Most of the 71 screened A. thaliana lines turned out to be susceptible. However, the mutant lines tu8, tu3, det1-1, and rhd3-1 showed a certain degree of tolerance under specific culture conditions. The reactions of rhd3-1 indicate that hypertrophy is a prerequisite for maturation of the pathogen. The reactions of the tu3 and tu8 mutants indicate a role of indole glucosinolates and indole-3-acetonitrile/IAA in development of clubroot disease. [source]
