Campos Abstract In spite of the worldwide occurrence of domestic cats and dogs, and their close relationship with humans, the number of published papers on free-ranging cats Felis catus and dogs Canis familiaris, is small. The diet of both species was estimated in a suburban and rural environment in July 2002 and January 2003. Visual observations and scat collection of both species were accomplished along a 10 km transect line in the Campus ,Luiz de Queiroz', University of São Paulo, Piracicaba, south-eastern Brazil. The diet of both species was determined by analysis of sterilized, washed, dried and sorted scats. Estimated abundances of free-ranging cats and dogs in the sampled area were 81 (±4.32) and 42 (±2.96), respectively. Cats and dogs were more abundant in the suburban than in the rural environment (t=3.78, P<0.001, N=55; t=8.38, P<0.001, N=55, respectively) and cats were more abundant than dogs in the suburban environment (t=6.76, P<0.001, N=55), even though there was no significant difference between the abundance of both species in the rural environment (t=0.82, P=0.46, N=55). Invertebrates were the most commonly consumed item by both species, followed by mammals (cats: 63.24 and 20.51%; dogs: 57.05 and 25.15%, respectively). Niche breadth was 0.4892 for cats and 0.4463 for dogs. Niche overlap was almost complete (0.97108). The consumption of mammals was estimated to be between 16.76 and 25.42 kg individual,1year,1 for dogs and between 2.01 and 2.9 kg individual,1year,1 for cats. These data might be useful to establish a management program to minimize the predation pressure of free-ranging cats and dogs on wildlife. [source] Wolves in the Great Lakes region: a phylogeographic puzzleMOLECULAR ECOLOGY, Issue 20 2010ETTORE RANDI Empirical studies demonstrate that natural hybridization in animals is more common than thought so far (Mallet 2005), particularly among species that originated recently through cycles of population contraction,expansion arising from climate changes over the last glacial period, the Pleistocene. In addition, the post-glacial global growth of human populations has fostered anthropogenic hybridization events, mediated by habitat changes, the persecution of large predators and the introduction of alien species (Allendorf et al. 2001). The Canis lineage shows cases of both natural and anthropogenic hybridization, exacerbating the controversy about the number of species that should be formally validated in the taxonomic lists, the evolutionary role of genetic introgression and the ways to manage hybrids with invading wild or domesticated populations. The study by Wheeldon et al. (2010), published in this issue of Molecular Ecology, adds a new piece to the intricate puzzle of evolution and taxonomy of Canis in North America. They show that sympatric wolves (C. lupus) and coyotes (C. latrans) are not (extensively) hybridizing in the western North American Great Lakes region (GLR). Widespread hybridization between coyotes and a genetically distinct, but closely related, wolf-like population (the eastern wolf) occurred in the northeastern regions of North America. In Wheeldon et al.'s (2010) opinion, these data should prove definitely that two different species of wolf (the western gray wolf C. lupus and the eastern wolf C. lycaon) and their hybrids are distributed across the GLR. [source] Reconstruction and Morphometric Analysis of the Nasal Airway of the Dog (Canis familiaris) and Implications Regarding Olfactory AirflowTHE ANATOMICAL RECORD : ADVANCES IN INTEGRATIVE ANATOMY AND EVOLUTIONARY BIOLOGY, Issue 11 2007Brent A. Craven Abstract The canine nasal airway is an impressively complex anatomical structure, having many functional roles. The complicated branching and intricate scrollwork of the nasal conchae provide large surface area for heat, moisture, and odorant transfer. Of the previous anatomical studies of the canine nasal airway, none have included a detailed rendering of the maxilloturbinate and ethmoidal regions of the nose. Here, we present a high-resolution view of the nasal airway of a large dog, using magnetic resonance imaging scans. Representative airway sections are shown, and a three-dimensional surface model of the airway is reconstructed from the image data. The resulting anatomic structure and detailed morphometric data of the airway provide insight into the functional nature of canine olfaction. A complex airway network is revealed, wherein the branched maxilloturbinate and ethmoturbinate scrolls appear structurally distinct. This is quantitatively confirmed by considering the fractal dimension of each airway, which shows that the maxilloturbinate airways are more highly contorted than the ethmoidal airways. Furthermore, surface areas of the maxilloturbinate and ethmoidal airways are shown to be much different, despite having analogous physiological functions. Functionally, the dorsal meatus of the canine nasal airway is shown to be a bypass for odorant-bearing inspired air around the complicated maxilloturbinate during sniffing for olfaction. Finally, nondimensional analysis is used to show that the airflow within both the maxilloturbinate and ethmoturbinate regions must be laminar. This work has direct relevance to biomimetic sniffer design, chemical trace detector development, intranasal drug delivery, and inhalation toxicology. [source] Coding repeats and evolutionary "agility"BIOESSAYS, Issue 6 2005Sandrine Caburet The rapid generation of new shapes observed in the living world is the result of genetic variation, especially in "morphological" developmental genes. Many of these genes contain coding tandem repeats. Fondon and Garner have shown that expansions and contractions of these repeats are associated with the great diversity of morphologies observed in the domestic dog, Canis familiaris.1 In particular, they found that the repeat variations in two genes were significantly associated with changes in limb and skull morphology. [source] Mammalian Diet and Broad Hunting Strategy of the Dingo (Canis familiaris dingo) in the Wet Tropical Rain Forests of Northeastern Australia,BIOTROPICA, Issue 2 2001Karl Vernes ABSTRACT The diet of dingoes (Canis familiaris dingo) in the Australian Wet Tropics was examined by analyzing 383 dingo scats collected throughout the region for the presence of mammal prey remains. The scats yielded 29 native and 4 introduced mammal prey species from 14 families. The most important species in terms of percentage occurrence in the scats were Melomys cervinipes (22.2%), hoodon macrourus (17.0%), Perameles nasuta (12.5%), and Thylogale stigmatica (12.5%). The most important families were Muridae (37.1%), Peramelidae (29.5%), and Macropodidae (25.8%). Examination of small-scale habitat preferences revealed species that preferentially use the forest edge ranked significantly higher in the diet than those that do not, and species that are terrestrial ranked higher in the diet than those that are arboreal. Relative abundance was also a significant factor in the ranked dietary occurrence of each species, with abundant species ranked significantly higher than those that are less abundant. These results suggest that dingoes in the Australian Wet Tropics are opportunistic predators of a wide variety of mammal species, with abundant terrestrial and forest edge-dwelling taxa the most susceptible to predation. [source] Electrophoretic analysis of sequence variability in three mitochondrial DNA regions for ascaridoid parasites of human and animal health significanceELECTROPHORESIS, Issue 13 2008Ming-Wei Li Abstract Sequence variability in three mitochondrial DNA (mtDNA) regions, namely cytochrome c oxidase subunit 1 (cox1), NADH dehydrogenase subunits 1 and 4 (nad1 and nad4), among and within Toxocara canis, T. cati, T. malaysiensis, T. vitulorum and Toxascaris leonina from different geographical origins was examined by a mutation-scanning approach. A portion of the cox1 gene (pcox1), a portion of the nad1 and nad4 genes (pnad1 and pnad4) were amplified separately from individual ascaridoid nematodes by polymerase chain reaction and the amplicons analyzed by single-strand conformation polymorphism (SSCP). Representative samples displaying sequence variation in SSCP profiles were subjected to sequencing in order to define genetic markers for their specific identification and differentiation. While the intra-specific sequence variations within each of the five ascaridoid species were 0.2,3.7% for pcox1, 0,2.8% for pnad1 and 0,2.3% for pnad4, the inter-specific sequence differences were significantly higher, being 7.9,12.9% for pcox1, 10.7,21.1% for pnad1 and 12.9,21.7% for pnad4, respectively. Phylogenetic analyses based on the combined sequences of pcox1, pnad1 and pnad4 revealed that the recently described species T. malaysiensis was more closely related to T. cati than to T. canis. These findings provided mtDNA evidence for the validity of T. malaysiensis and also demonstrated clearly the usefulness and attributes of the mutation-scanning sequencing approach for studying the population genetic structures of these and other nematodes of socio-economic importance. [source] Utilization of essential oil as natural antifungal against nail-infective fungiFLAVOUR AND FRAGRANCE JOURNAL, Issue 2 2002Mamta Patra Abstract During antifungal screening of some essential oils, Foeniculum vulgare exhibited the strongest activity, completely inhibiting the mycelial growth of the nail-infective fungi, Trichophyton rubrum, T. mentagrophytes and Scytalidium dimidiatum. The essential oil was found to be fungicidal at 0.2, 0.4 and 0.5 µl/ml concentrations. The oil was efficiently active against heavy doses of inoculum at minimum fungicidal concentrations. The fungicidal activity of the oil was found to be thermostable up to 80 °C, with no descramble decrease in activity after 48 months of storage. The oil also showed a broad fungitoxic spectrum, inhibiting the mycelial growth of other nail-infective fungi, viz. Aspergillus flavus, A. fumigatus, A. niger,A. ustus, Candida albicans, Epidermophyton floccosum, Microporum audouinii, M. canis, M. gypseum, M. nanum, Rhizopus nigricans, Trichophyton tonsurans and T. violaceum. Moreover, it did not exhibit any adverse effects on mammalian skin and nails up to 5% concentration. [source] Detection of Helicobacter ganmani -Like 16S rDNA in Pediatric Liver TissueHELICOBACTER, Issue 5 2004Vasundhara Tolia ABSTRACT Background., To determine the presence of Helicobacter species in the liver biopsy specimens from children with various chronic liver diseases as data in adult literature suggests a possible role of these bacteria in their pathogenesis. Materials and methods., Paraffin sections of 61 liver biopsies of pediatric patients with miscellaneous diseases and autopsy liver tissue from 10 control subjects with no evidence of preexisting liver disease were examined for the presence of Helicobacter species by a genus-specific seminested polymerase chain reaction (PCR) assay. PCR,products of positive samples were further characterized by denaturing gradient gel electrophoresis (DGGE) and DNA-sequence analysis. Based on those results, a seminested PCR assay for H. ganmani was developed and applied to the samples. Results., On analysis, 40/61 patient samples were positive in the genus-specific Helicobacter PCR and 4/10 from the control group. The nucleotide sequences of 16S rDNA fragments were 99,100% similar to mainly Helicobacter sp. ,liver' and H. ganmani. PCR-products similar to H. canis and H. bilis were also found. The 16S rDNAs of control specimens showed similarity to Helicobacter sp. ,liver'. In the H. ganmani -specific PCR analysis 19 patients, but none of the controls, were positive. Conclusions., Amplified Helicobacter 16S rDNAs were related to Helicobacter sp. ,liver' or H. ganmani in liver biopsy specimens of pediatric patients. The possible significance of Helicobacter species in pediatric liver diseases needs to be evaluated further in prospective studies. [source] Kerion and dermatophytic granuloma.INTERNATIONAL JOURNAL OF DERMATOLOGY, Issue 3 2006Mycological, histopathological findings in 19 children with inflammatory tinea capitis of the scalp Background, Inflammatory tinea capitis or kerion is the result of a hypersensitivity reaction to a dermatophytic infection. Majocchi's granuloma, in contrast, usually begins as a suppurative folliculitis and culminates in a granulomatous reaction. Objectives, To present clinical, mycological and histopathological findings for 19 cases of kerion of the scalp in children. Methods, Nineteen children were investigated (14 boys and five girls) with a mean age of 6.5 years. A potassium hydroxide (KOH) exam and culture in Sabouraud dextrose agar were performed, followed by a biopsy with hematoxylin and eosin, periodic acid-schiff (PAS) and Gomori-Grocott stains. The same investigations were carried out in four control cases of noninflammatory tinea capitis. Results, Clinical history varied from 2 to 16 weeks (mean 6.6 weeks). Diagnosis was confirmed by a positive KOH exam: all cases except one had a positive culture. The following dermatophytes were isolated: Microsporum canis (32%), Trichophyton mentagrophytes (27%), Trichophyton tonsurans (21%), Trichophyton rubrum (10%) and Microsporum gypseum (5%). The histopathological findings were: suppurative folliculitis (SF) 11%, SF plus suppurative dermatitis 37%, suppurative and granulomatous dermatitis (SGD) 26% and SGD plus fibrosing dermatitis 26%. Fungi were observed in 63% of the histopathological sections. Perifollicular infiltrates (PF) around the parasitized hair follicles were identified in the four noninflammatory control cases due to M. canis. Conclusions, Kerion Celsi is an inflammatory or suppurative type of tinea capitis caused by zoophylic dermatophytes (M. canis and T. mentagrophytes), but also by antrophophylic (T. tonsurans and T. rubrum) and geophylic (M. gypseum) dermatophytes. Histopathological findings showed a spectrum from mild suppurative folliculitis to dense granulomatous infiltrates without a clear relationship with the clinical features. [source] Survey of dermatophytes on clinically normal cats in the southeast of EnglandJOURNAL OF SMALL ANIMAL PRACTICE, Issue 9 2005A. Patel Objectives: To report the incidence of dermatophytes on the hair coat of asymptomatic cats in the southeast of England. Asymptomatic cats are often blamed for transmission of dermatophytes between animals and humans. This study may help to clarify whether cats are responsible for the increase in fungal infections among the human population. Methods: A total of 169 clinically healthy cats without any dermatological signs were sampled using the Mackenzie brush technique and cultured for dermatophytes. Thirty cats were from a closed colony and 139 were feral or from domestic households in the southeast of England. Results: The incidence of Microsporum canis and Trichophyton mentagrophytes in household and feral cats was 2.16 per cent for each dermatophyte. This survey shows little difference in the isolation rates of M canis between the southeast and southwest of England, which was reported on in 1994. Clinical Significance: Given the low number of dermatophytes isolated, asymptomatic cats are unlikely to be responsible for the increasing incidence of human infection. Asymptomatic carriers with T mentagrophytes in the hair coat have not been previously reported and may need to be considered when treating humans with trichophytosis. Wills DVM Abstract Objective: To present a case of clinical hypocalcemia in a critically ill septic dog. Case summary: A 12-year old, female spayed English sheepdog presented in septic shock 5 days following hemilaminectomy surgery. Streptococcus canis was cultured from the incision site. Seven days after surgery, muscle tremors were noted and a subsequent low serum ionized calcium level was measured and treated. Intensive monitoring, fluid therapy, and antibiotic treatment were continued because of the sepsis and hypocalcemia, but the dog was euthanized 2 weeks after surgery. New or unique information provided: Low serum ionized calcium levels are a common finding in critically ill human patients, especially in cases of sepsis, pancreatitis, and rhabdomyolysis. In veterinary patients, sepsis or streptococcal infections are not commonly thought of as a contributing factor for hypocalcemia. Potential mechanisms of low serum ionized calcium levels in critically ill patients include intracellular accumulation of calcium ions, altered sensitivity and function of the parathyroid gland, alterations in Vitamin D levels or activity, renal loss of calcium, and severe hypomagnesemia. Pro-inflammatory cytokines and calcitonin have also been proposed to contribute to low ionized calcium in the critically ill. Many veterinarians rely on total calcium levels instead of serum ionized calcium levels to assess critical patients and may be missing the development of hypocalcemia. Serum ionized calcium levels are recommended over total calcium levels to evaluate critically ill veterinary patients. [source] Babesiosis Caused by a Large Babesia Species in 7 Immunocompromised DogsJOURNAL OF VETERINARY INTERNAL MEDICINE, Issue 1 2010L.E. Sikorski Background: A large unnamed Babesia species was detected in a dog with lymphoma. It was unknown if this was an underrecognized pathogen. Objective: Report the historical and clinicopathologic findings in 7 dogs with babesiosis caused by a large unnamed Babesia species characterize the 18S ribosomal ribonucleic acid (rRNA) genes. Animals: Seven immunocompromised dogs from which the Babesia was isolated. Methods: Retrospective case review. Cases were identified by a diagnostic laboratory, the attending clinicians were contacted and the medical records were reviewed. The Babesia sp. 18S rRNA genes were amplified and sequenced. Results: Six of 7 dogs had been splenectomized; the remaining dog was receiving oncolytic drugs. Lethargy, anorexia, fever, and pigmenturia were reported in 6/7, 6/7, 4/7, and 3/7 dogs. Laboratory findings included mild anemia (7/7) and severe thrombocytopenia (6/7). Polymerase chain reaction (PCR) assays used to detect Babesia sensu stricto species were all positive, but specific PCR assays for Babesia canis and Babesia gibsoni were negative in all dogs. The 18S rRNA gene sequences were determined to be identical to a large unnamed Babesia sp. previously isolated. Cross-reactive antibodies against other Babesia spp. were not always detectable. Five dogs were treated with imidocarb dipropionate and 1 dog with atovaquone/azithromycin; some favorable responses were noted. The remaining dog was untreated and remained a clinically stable carrier. Conclusions and Clinical Importance: Dogs with pigmenturia, anemia, and thrombocytopenia should be tested for Babesia sp. by PCR. Serology is not sufficient for diagnosis of this Babesia sp. Asplenia, chemotherapy, or both might represent risk factors for persistent infection, illness, or both. [source] Serum Cardiac Troponin I Concentration in Dogs with EhrlichiosisJOURNAL OF VETERINARY INTERNAL MEDICINE, Issue 5 2008P.P.V.P. Diniz Background: Ehrlichiosis is a multisystemic disease with the potential to cause cardiomyocyte injury in naturally infected dogs. Hypothesis: Myocardial injury occurs in dogs infected with Ehrlichia canis. Animals: One-hundred and ninety-four dogs from Brazil with clinical and laboratory abnormalities indicative of ehrlichiosis. Sixteen healthy dogs served as controls. Methods: Electrocardiogram, echocardiogram, noninvasive blood pressure measurement, and serum cardiac troponin I (cTnI) concentrations were evaluated. Serologic assays and PCR determined the exposure and infection status for E. canis, Anaplasma spp., Babesia canis vogeli, Bartonella spp., Borrelia burgdorferi, Dirofilaria immitis, Ehrlichia chaffeensis, Ehrlichia ewingii, Leishmania chagasi, and spotted-fever group Rickettsia. Dogs were assigned to groups according to PCR status: E. canis infected, infected with other vector-borne organisms, sick dogs lacking PCR evidence for infection, and healthy controls. Results: E. canis -infected dogs had higher serum cTnI concentrations than controls (median: 0.04 ng/dL; range 0.04,9.12 ng/dL; control median: 0.04 ng/dL; range: 0.04,0.10 ng/dL; P= .012), and acute E. canis infection was associated with myocardial injury (odds ratio [OR]: 2.67, confidence interval [CI] 95%: 1.12,6.40, P= .027). Severity of anemia was correlated with increased risk of cardiomyocyte damage (r= 0.84, P < .001). Dogs with clinical signs of systemic inflammatory response syndrome (SIRS) were at higher risk for myocardial injury than were other sick dogs (OR: 2.55, CI 95%: 1.31,4.95, P= .005). Conclusions and Clinical Importance: Acute infection with E. canis is a risk factor for myocardial injury in naturally infected Brazilian dogs. Severity of anemia and SIRS might contribute to the pathophysiology of myocardial damage. [source] Detection and identification of Bartonella sp. in fleas from carnivorous mammals in Andalusia, SpainMEDICAL AND VETERINARY ENTOMOLOGY, Issue 4 2009F. J. MÁRQUEZ Abstract. A total of 559 fleas representing four species (Pulex irritans, Ctenocephalides felis, Ctenocephalides canis and Spilopsyllus cuniculi) collected on carnivores (five Iberian lynx Lynx pardinus, six European wildcat Felis silvestris, 10 common genet Genetta genetta, three Eurasian badger Meles meles, 22 red fox Vulpes vulpes, 87 dogs and 23 cats) in Andalusia, southern Spain, were distributed in 156 pools of monospecific flea from each carnivore, and tested for Bartonella infection in an assay based on polymerase chain reaction (PCR) amplification of the 16 S,23 S rRNA intergenic spacer region. Twenty-one samples (13.5%) were positive and the sequence data showed the presence of four different Bartonella species. Bartonella henselae was detected in nine pools of Ctenocephalides felis from cats and dogs and in three pools of Ctenocephalides canis from cats; Bartonella clarridgeiae in Ctenocephalides felis from a cat, and Bartonella alsatica in Spilopsyllus cuniculi from a wildcat. DNA of Bartonella sp., closely related to Bartonella rochalimae, was found in seven pools of Pulex irritans from foxes. This is the first detection of B. alsatica and Bartonella sp. in the Iberian Peninsula. All of these Bartonella species have been implicated as agents of human diseases. The present survey confirms that carnivores are major reservoirs for Bartonella spp. [source] Dermatophytoses in outpatients attending the Dermatology Center of Avicenna Hospital in Qazvin, IranMYCOSES, Issue 2 2008Mohammad Reza Aghamirian Summary Dermatophytosis is still being considered as one of the major public health problems in many parts of the world. To identify the prevalence and etiological agents of dermatophytoses, a study was carried out between 2004 and 2006. Out of 1023 subjects suspected to have cutaneous mycoses, 348 (34%) patients were affected with dermatophytoses. The causative agents were identified macroscopically and microscopically after the clinical samples were subjected to potassium hydroxide examination and culture isolation. Epidermophyton floccosum was the most frequently isolated species representing 32.8% of isolates, followed by T. rubrum (18.1%), T. verrucosum (17.2%), T. mentagrophytes var. interdigitale (15.8%), T. mentagrophytes var. mentagrophytes (6.6%) and T. violaceum, T. schoenleinii and Microsporum canis (0.9%). The most common type of infection was tinea cruris (31.9%) affecting in particular male patients. Some other most prevalent tinea types were tinea corporis (20.7%), tinea pedis (19%), and tinea unguium (11.2%). To the best of our knowledge, this is the first report on dermatophytoses in Qazvin and the first study that shows tinea cruris as a major type of superficial fungal infection in Iran. The frequency of tinea was higher in males and tinea cruris showed a remarkably increasing tendency and was an important public health issue in Qazvin. Iorio Summary Two hundred hair/skin samples were collected from 2002 to 2004 from two groups of cats (privately owned and stray cats from a shelter) and 165 samples were obtained during the same period from persons in whom dermatophyte infection was highly suspected. The epidemiological data were statistically evaluated. Thirteen of the 100 privately owned cats (13%) and 100% of the stray cats were positive; of the 165 human samples examined 109 (66%) were positive for dermatophytes. Microsporum canis was the most common dermatophyte isolated in both cat groups while Trichophyton mentagrophytes was the most common in humans. Interestingly, a geophylic dermatophyte species (Microsporum gypseum) was found to be present and associated with clinical signs. Living in the countryside proved to be a risk factor for dermatophytoses in privately owned cats while in humans the main risk factor for M. canis was contact with animals followed by young age. None of the variables considered was associated with positivity for T. mentagrophytes while positivity for other fungi was correlated with life in the countryside. LIAO SUMMARY Infection by Toxocara canis in humans may cause cerebral toxocariasis (CT). Appreciable numbers of T. canis larvae cross the blood,brain barrier (BBB) to invade the brain thus causing CT. In the present studies, we evaluated the BBB permeability and BBB injury as assessed by the cerebral Evans blue (EB) concentration as well as by pathological changes and glial fibrillary acidic protein (GFAP) expression in T. canis -infected mice monitored from 3 days (dpi) to 8 weeks post-infection (wpi). The vasodilation neuropeptides, the expressions of substance P (SP) and its preferred binding neurokinin-1 receptor (NK-1R) as well as claudin-5 of tight-junction proteins associated with BBB impairment were also assessed by Western blotting and reverse-transcriptase polymerase chain reaction. Results revealed that BBB permeability increased as evidenced by a significantly elevated EB concentration in brains of infected mice. BBB injury appeared due to enhanced GFAP protein and mRNA expressions from 4 to 8 wpi. Leukocytes might have been unrelated to BBB impairment because there was no inflammatory cell infiltration despite T. canis larvae having invaded the brain; whereas markedly elevated SP protein and NK-1R mRNA expressions concomitant with enhanced claudin-5 expression seemed to be associated with persistent BBB impairment in this experimental CT model. PILARCZYK SUMMARY The effect of oral and intraperitoneal supply of sodium selenite on the immune response to, and the course of T. canis larvae infection in mice were determined. The number of worms in the host tissue was reduced but the migratory route of larvae was not affected. Selenite (Se) supplementation influences Se retention in the liver, enhanced IL-5 and eosinophil responses and evoked IL-6 production in mice infected with T. canis. The enhanced protection in mice given Se intraperitoneally was associated with high levels of parasite-specific IgE, and enhanced concentration of Th1-related cytokines such IL-12p70, TNF-, and IFN-,. In mice given Se orally, the predominant cytokines produced were IL-10, MCP-1 and IL-6 and these mice had lower protection. In conclusion, Se supplementation increases production of specific cytokines in mice infected with T. canis and increases protection against infection. Espinoza SUMMARY The direct effect of nitric oxide (NO) on the viability of Toxocara canis larvae was studied. We observed that the nitric oxide donors, SIN-1 and SNOG, exert no cytotoxic effect on the in vitro viability of T. canis larvae. In addition, we developed a model in rats to elucidate the role of NO during T. canis infection. We evaluated different indicators in four experimental groups: morphological parameters, the total number cells and cell types recovered, nitrite and protein concentration, lactate dehydrogenase and alkaline phosphatase enzymatic activity in the bronchoalveolar lavage fluid, lung index and detection of anti- T. canis specific antibodies. We observed significant differences between non-infected and infected groups. The infected animals treated with the inducible nitric oxide synthase (iNOS) inhibitor aminoguanidine were less damaged than infected, non-treated animals. Our results suggest that the in vivo inhibition of the synthesis of NO triggered by iNOS diminishes the deleterious effects of the parasite upon the host, especially the vascular alterations in the lungs. We could show that in vivo production of NO induced by infection with T. canis results in direct host damage. Thus, this induction may constitute an evasion/adaptation mechanism of the parasite. Romano MD Tinea capitis is the most frequent manifestation of dermatophyte infection in children, but because it is rare in the first months of life it is often misdiagnosed. Here we report 15 cases of tinea capitis observed in Italy in infants less than 1 year of age. There were 10 boys and 5 girls (mean age 6 months). Diagnosis was confirmed by mycologic examination. Microsporum canis was isolated in nine cases and Trichophyton mentagrophytes in three. These 12 infants were Italian and animals were the source of infection. Trichophyton erinacei was isolated in one Italian infant, and the source was soil. In the other two cases, Trichophyton tonsurans and Trichophyton violaceum were isolated; these infants were from Central America and India, respectively, and had contracted the infection from humans. All achieved clinical and mycologic recovery after systemic and topical antimycotic therapy. The relationship between toxocariasis and asthma is complex, with some studies demonstrating that children with asthma were more likely to be Toxocara seropositive as compared to non-asthmatic children, and other studies indicating no such significant relationship. The aim of the present study was to investigate Toxocara seropositivity and its association with asthma in a selected group of Sri Lankan children. Methods:, Two groups of children were studied: group 1 included 100 children with confirmed bronchial asthma who were on regular inhaler steroid treatment for asthma; group 2 included 96 children who did not have physician-diagnosed asthma or upper respiratory tract infections, attending the same hospital. Diagnosis of Toxocara seropositivity was based on IgG Toxocara Microwell Serum Elisa Kits. Enzyme-linked immunosorbent assay was regarded as positive for a reading of 0.3 optical density units. Stool samples were examined for helminth ova. Results:,Toxocara seropositivity in children with asthma was 29% and this was significantly more than Toxocara seropositivity among non-asthmatic children (P < 0.001). Toxocara seropositivity was identified as a significant risk factor of asthma in a univariate model. Eosinophilia was seen in a significantly higher proportion of non-asthmatic and asthmatic children who were Toxocara seropositive. Toxocara seropositivity, however, was not identified as a significant risk factor in a multivariate model. Conclusions:, The analysis confirmed previously identified risk factors for asthma but there was no association between the helminth parasitic infection, toxocariasis and bronchial asthma in children. Cama ABSTRACT: Cryptosporidium parasites from a cross-sectional study conducted in two national hospitals in Lima, Peru were genetically characterized to deteimine the diversity of Cryptosporidium spp. in HIV-positive people. A total of 2,672 patients participated in this study and provided 13,937 specimens. Cryptosporidium oocysts were detected by microscopy in 354 (13.3%) of the patients. Analysis of 951 Cryptosporidium - positive specimens from 300 patients using a small subunit rRNA-based PCR-RFLP tool identified 6 genotypes; Cryptosporidium hominis was the species most frequently detected (67.5%), followed by C. meleagridis (12.6%) and C. parvum (11.3%). Cryptosporidium canis (4.0%), C. felis (3.3%), and Cryptosporidium pig genotype (0.5%) were also found. These findings indicate that C. hominis is the predominant species in Peruvian HIV-positive persons, and that zoonotic Cryptosporidium spp. account for about 30% of cryptosporidiosis in these patients. Chohan Abstract A series of triazole-derived Schiff bases (L1,L5) and their oxovanadium(IV) complexes have been synthesized. The chemical structures of Schiff bases were characterized by their analytical (CHN analysis) and spectral (IR, 1H and 13C NMR and mass spectrometry) data, and oxovanadium(IV) complexes were elucidated by their physical (magnetic susceptibility and conductivity), analytical (CHN analysis), conductance measurements and electronic spectral data. The molar conductivity data indicate the oxovanadium(IV) complexes to be non-electrolyte. The Schiff bases act as bidentate and coordinate with the oxovanadium(IV)-forming stoichiometry of a complex as [M (L-H)2] where M = VO and L = L1,L5 in a square-pyramidal geometry. The agar well diffusion method was used for in vitro antibacterial screening against E. coli, S. flexenari, P. aeruginosa, S. typhi, S. aureus and B. subtilis and for antifungal activity against T. longifucus, C. albican, A. flavus, M. canis, F. solani and C. glaberata. The biological activity data show the oxovanadium(IV) complexes to be more antibacterial and antifungal than the parent Schiff bases against one or more bacterial and fungal strains. The compounds obtained have been further used as ligands for their ligand and antimicrobial properties with cobalt(II), copper(II), nickel(II) and zinc(II) metal ions. The compounds synthesized have been characterized by physical, spectral and analytical methods and have been screened for antibacterial activity against Escherichia coli, Bacillus subtillis, Staphylococcus aureus, Pseudomonas aeruginosa and Salmonella typhi, and for antifungal activity against Trichophyton longifusus, Candida albicans, Aspergillus flavus, Microsporum canis, Fusarium solani and Candida glaberata using the agar well-diffusion method. All the compounds synthesized have shown good affinity as antibacterial and antifungal agents, which increased in most of the cases on complexation with the metal ions. The aryl part was: C6H5 for 1a, 1b; 4-CH3C6H4 for 2a, 2b; 4-CH3OC6H4 for 3a, 3b; 4-ClC6H4 for 4a, 4b; 4-FC6H4 for 5a, 5b; 4-BrC6H4 for 6a, 6b; 2,4-(Cl)2C6H3 for 7a, 7b; 4-NO2C6H4 for 8a, 8b; and C4H3S(2-yl) i. e., 2-thienyl for 9a, 9b. Piperidinol compounds 2b, 3b, 4b, 5b, 7b, 8b, and 9b are reported here for the first time. The synthesized compounds were tested against seven types of plant pathogenic fungi and three types of human pathogenic fungi using the agar dilution assay. Itraconazole was tested against Candida parapsilosis as the reference compound, while Nystatin was tested as the reference compound against the other fungi. Compounds 1a, 1b, 2a, 4a, 4b, 5a, 5b, 6a, 7a, 8a, 9a, and 9b can be selected as model compounds to develop new antifungal agents against the human pathogen Microsporum canis. Compounds 8a and 8b, which had a similar antifungal activity compared with the reference compound Nystatin against the plant pathogen Aspergillus flavus, can serve as model compounds to develop new antifungal agents to solve agricultural problems. [source] Encroachment of Echinococcus granulosus into urban areas in eastern Queensland, AustraliaAUSTRALIAN VETERINARY JOURNAL, Issue 8 2008DJ Jenkins Objective To investigate the prevalence of Echinococcus granulosus in wild dogs (dingos and dingo,domestic dog hybrids) living in and around human habitation on Fraser Island and in townships of the Maroochy Shire, on Queensland's Sunshine Coast, Australia. Design Wild dogs were humanely killed on Fraser Island and in the Maroochy Shire because they were deemed a potential danger to the public. Their intestines were collected and the contents examined for intestinal parasites. Procedure Intestines were removed as soon after death as possible, packed in plastic bags and kept frozen until examination. The intestinal contents were washed, sieved and examined microscopically for the presence of helminths, which were identified and counted. Results Intestines from 108 wild dogs, 7 foxes and 18 Fraser Island dingoes were examined. Echinococcus granulosus was only present in the wild dogs from Maroochy Shire (46.3%) with worm burdens of between 30 and 104,000. Other helminths included Spirometra erinacei, Dipylidium caninum, Taenia spp., Ancylostoma caninum and Toxocara canis. Two specimens of a trematode (Haplorchinae sp.) usually found infecting fish and seabirds were recovered from a Fraser Island dingo. Conclusion Dingoes on Fraser Island are not infected with E. granulosus and do not pose a hydatid disease public health risk to residents or visitors. However, wild dogs examined from the Maroochy Shire do present a potential hydatid disease public health risk. [source] |
