Carrique-Mas Abstract Aims:, To evaluate the performance of three Salmonella plating media (Rambach, Xylose Lysine Deoxycholate agar and modified Brilliant Green Agar plus Novobiocin) as part of the ISO 6579: 2002 (Annex D) on poultry environmental samples. Methods and Results:, The samples analysed were those for the European Union Salmonella baseline surveys of laying (N = 3087), broiler (N = 1550), turkey fattening (N = 1540) and turkey breeding (N = 580) flocks for Great Britain. Results were considered separately for Rambach (including and excluding pale orange colonies) and for growth on selective media [Modified semi-solid Rappaport Vassiliadis (MSRV)] after 24 and 48 h of incubation. Overall, Rambach was the most sensitive medium, provided that pale orange colonies were checked. In all cases, an increase in the sensitivity of detection was obtained by plating growth on MSRV after 48 h of incubation. In broilers and laying flocks, the specificity significantly improved when Rambach only was used. Conclusion:, The use of Rambach results in considerable savings compared with the two-plate method prescribed by ISO 6579:2002 (Annex D) without compromising sensitivity. Significance and Impact of the Study:,Salmonella isolation protocols should be reviewed in terms of their efficiency and cost. [source] Effects of Monochromatic Light on Proliferation Response of Splencyte in BroilersANATOMIA, HISTOLOGIA, EMBRYOLOGIA, Issue 5 2008D. Xie Summary To investigate the effects of various monochromatic lights on splenocyte proliferation responses, a total of 260 Arbor Acre male broilers on P1 (post-hatching day 1) were exposed to blue light (BL), green light (GL), red light (RL) and white light treatments by light emitting diode system for 7 weeks, respectively. All light sources were equalized on the intensity of 15 lx and light period of 23 h daily. Morphological change of spleen and response of splenocyte proliferation were assessed by using histochemistry staining and colorimetric test in cultures of purified splenic cells. The results were as follows: (1) At P21, GL increased significantly the spleen weight by 163.6% and spleen index by 118.8% compared with RL (P < 0.05). Until P49, BL enhanced significantly the spleen weights by 42.2% compared with RL (P < 0.05), but no significant difference was found in the spleen index among four light-treated groups (P > 0.05). (2) Compared with RL, GL increased significantly the diameter of splenic nodule and area of periarterial lymphatic sheath at P21 by 87.2 and 58.1%, respectively (P < 0.05); BL increased significantly the diameter of splenic nodule and area of periarterial lymphatic sheath at P49 by 64.4 and 50.5%, respectively (P < 0.05). (3) At P21, GL enhanced spleen lymphocytes proliferation in response to concanavalin A compared with RL by 50.0% (P < 0.05). Until P49, the mitogenic response in BL was significantly higher (29.4%) than that of RL (P < 0.05). (4) The interleukin-2 (IL-2) bioactivity was significantly increased to 34.3% in GL than in RL at P21 (P < 0.05). Until P49, the IL-2 bioactivity in BL was significantly higher (62.2%) than that of RL (P < 0.05). (5) There was no significant difference in the nitric oxide (NO) concentration of splenocyte among RL, GL and BL groups at P21 (P > 0.05), but the concentration in RL group at P49 was significantly increased, 59.0 and 63.7% compared to that of GL and BL groups, respectively (P < 0.05). These results suggested that the monochromatic light affected splenocyte proliferation mainly because of alterations in IL-2 bioactivity and NO production in splenocyte of broiler. In early stage of broiler growth, the action of GL was obvious, while the response of BL was stronger in later stage. [source] Quantitative trait loci associated with fatness in a broiler,layer crossANIMAL GENETICS, Issue 5 2009R. L. R. Campos Summary An F2 population established by crossing a broiler male line and a layer line was used to map quantitative trait loci (QTL) affecting abdominal fat weight, abdominal fat percentage and serum cholesterol and triglyceride concentrations. Two genetic models, the line-cross and the half-sib, were applied in the QTL analysis, both using the regression interval method. Three significant QTL and four suggestive QTL were mapped in the line-cross analysis and four significant and four suggestive QTL were mapped in the half-sib analysis. A total of five QTL were mapped for abdominal fat weight, six for abdominal fat percentage and four for triglyceride concentration in both analyses. New QTL associated with serum triglyceride concentration were mapped on GGA5, GGA23 and GG27. QTL mapped between markers LEI0029 and ADL0371 on GGA3 for abdominal fat percentage and abdominal fat weight and a suggestive QTL on GGA12 for abdominal fat percentage showed significant parent-of-origin effects. Some QTL mapped here match QTL regions mapped in previous studies using different populations, suggesting good candidate regions for fine-mapping and candidate gene searches. [source] Quantitative trait loci for performance traits in a broiler × layer crossANIMAL GENETICS, Issue 2 2009M. Ambo Summary An F2 resource population, derived from a broiler × layer cross, was used to map quantitative trait loci (QTL) for body weights at days 1, 35 and 41, weight gain, feed intake, feed efficiency from 35 to 41 days and intestinal length. Up to 577 F2 chickens were genotyped with 103 genetic markers covering 21 linkage groups. A preliminary QTL mapping report using this same population focused exclusively on GGA1. Regression methods were applied to line-cross and half-sib models for QTL interval mapping. Under the line-cross model, eight QTL were detected for body weight at 35 days (GGA2, 3 and 4), body weight at 41 days (GGA2, 3, 4 and 10) and intestine length (GGA4). Under the half-sib model, using sire as common parent, five QTL were detected for body weight at day 1 (GGA3 and 18), body weight at 35 days (GGA2 and 3) and body weight at 41 days (GGA3). When dam was used as common parent, seven QTL were mapped for body weight at day 1 (GGA2), body weight at day 35 (GGA2, 3 and 4) and body weight at day 41 (GGA2, 3 and 4). Growth differences in chicken lines appear to be controlled by a chronological change in a limited number of chromosomal regions. [source] Identification of quantitative trait loci associated with egg quality, egg production, and body weight in an F2 resource population of chickens,ANIMAL GENETICS, Issue 2 2006M. A. Schreiweis Summary Egg production and egg quality are complex sex-limited traits that may benefit from the implementation of marker-assisted selection. The primary objective of the current study was to identify quantitative trait loci (QTL) associated with egg traits, egg production, and body weight in a chicken resource population. Layer (White Leghorn hens) and broiler (Cobb-Cobb roosters) lines were crossed to generate an F2 population of 508 hens over seven hatches. Phenotypes for 29 traits (weekly body weight from hatch to 6 weeks, egg traits including egg, albumen, yolk, and shell weight, shell thickness, shell puncture score, percentage of shell, and egg shell colour at 35 and 55 weeks of age, as well as egg production between 16 and 55 weeks of age) were measured in hens of the resource population. Genotypes of 120 microsatellite markers on 28 autosomal groups were determined, and interval mapping was conducted to identify putative QTL. Eleven QTL tests representing two regions on chromosomes 2 and 4 surpassed the 5% genome-wise significance threshold. These QTL influenced egg colour, egg and albumen weight, percent shell, body weight, and egg production. The chromosome 4 QTL region is consistent with multiple QTL studies that define chromosome 4 as a critical region significantly associated with a variety of traits across multiple resource populations. An additional 64 QTL tests surpassed the 5% chromosome-wise significance threshold. [source] Genetic diversity of Forest and Savannah chicken populations of Ghana as estimated by microsatellite markersANIMAL SCIENCE JOURNAL, Issue 3 2010Richard OSEI-AMPONSAH ABSTRACT The characterization of indigenous animal genetic resources is a requisite step in providing needed information for the conservation of useful genotypes against future needs. Thus, in this study, 22 microsatellite markers were used to genotype 114 local chickens from the Forest (n = 59) and Savannah (n = 55) eco-zones of Ghana and the results compared to those of the ancestral red junglefowl (n = 15) and two European commercial chicken populations , a broiler (n = 25) and white leghorn (n = 25). A total of 171 alleles were observed, with an average of 7.8 alleles per locus. The local Ghanaian chickens showed higher diversity in terms of the observed number of alleles per locus (6.6) and observed heterozygosity (0.568) compared with the combined control populations (6.0 and 0.458, respectively). However, Wright's F -statistics revealed negligible genetic differentiation (FST) in local Ghanaian chicken populations. In addition, 65% of the Savannah chickens were inferred to be more likely from the Forest, suggesting a south-north dispersal of chickens from their probable original location in the Forest zone to the Savannah areas. It is concluded that the Forest and Savannah chickens of Ghana are a single, randomly mating unselected population, characterized by high genetic diversity and constitute a valuable resource for conservation and improvement. [source] Feeding of different levels of metabolite combinations produced by Lactobacillus plantarum on growth performance, fecal microflora, volatile fatty acids and villi height in broilersANIMAL SCIENCE JOURNAL, Issue 2 2010Teck C. LOH ABSTRACT The effects of feeding different dosages of metabolite combination of L. plantarum RS5, RI11, RG14 and RG11 strains (Com3456) on the performance of broiler chickens was studied. A total of 504 male Ross broilers were grouped into 7 treatments and offered different diets: (i) standard corn-soybean based diet (negative control); (ii) standard corn-soybean based diet +100 ppm neomycin and oxytetracycline (positive control); (iii) standard corn-soybean based diet + 0.1% metabolite combination of L. plantarum RS5, RI11, RG14 and RG11 strains (Com3456); (iv) standard corn-soybean based diet + 0.2% of Com3456; (v) standard corn-soybean based diet + 0.3% of Com3456 (vi) standard corn-soybean based diet + 0.4% of Com3456 and (vii) standard corn-soybean based diet + 0.5% of Com3456. Supplementation of Com3456 with different dosages improved growth performance, reduced Enterobacteriaceae and increased lactic acid bacteria count, and increased villi height of small intestine and fecal volatile fatty acid concentration. Treatment with 0.4% and 0.2% Com3456 had the best results, especially in terms of growth performance, feed conversion ratio and villi height among other dosages. However, the dosage of 0.2% was recommended due to its lower concentration yielding a similar effect as 0.4% supplementation. These results indicate that 0.2% is an optimum level to be included in the diets of broiler in order to replace antibiotic growth promoters. [source] A study on feeding hazelnut kernel oil meal as a protein source for broiler chickensANIMAL SCIENCE JOURNAL, Issue 3 2009Guray ERENER ABSTRACT An experiment was conducted to evaluate the effect of substituting different levels of hazelnut kernel oil meal (HKM) for soybean meal (SBM) in diets for broiler. A total of 450 one-day-old female Ross 308 broiler chicks were allocated randomly to three treatment groups of 150 birds each in a randomized design. Each treatment group consisted of five replicates each of 30 chicks. All diets (in mash form) were formulated to meet nutrient concentrations recommended for broilers. The experiment lasted for six weeks. In the experiment, an SBM control (SBM) diet was compared to two HKM diets, replacing 50 (50HKM) and 100% (HKM) of SBM protein, respectively. Body weight gain, feed intake and feed conversion ratio of broilers were adversely affected (P < 0.05) by the HKM diets at 42 days of age. Broilers fed 50HKM and HKM had growth performances similar (P > 0.05). The carcass yield and abdominal fat pads of birds fed diets with SBM were higher (P < 0.05) than those of chicks fed the 50HKM and HKM diets. The edible inner organ weight of chicks fed diets with HKM was the heaviest (P < 0.05). It is concluded that SBM cannot be replaced even up to 50% with HKM in commercial broiler diet. [source] ORIGINAL ARTICLE: Effects of dietary supplementation of synbiotics and phytobiotics on performance, caecal coliform population and some oxidant/antioxidant parameters of broilersJOURNAL OF ANIMAL PHYSIOLOGY AND NUTRITION, Issue 5 2010Z. Erdo Summary The current study was conducted to evaluate the effects of dietary supplementation of synbiotics and phytobiotics on performance, small intestine weight, pH and caecal coliform counts of broilers. The influences of synbiotics and phytobiotics on oxidant/antioxidant status in the blood of broilers were also assessed. A total of 200 broiler chicks were randomly allotted to four dietary treatments, either fed a basal diet or the same diet supplemented with 1 g/kg synbiotic, 1 g/kg phytobiotic or 1 g/kg synbiotic plus 1 g/kg phytobiotic. The diet supplemented with both synbiotic and phytobiotic had no effect on body weight, body weight gain, feed intake and feed efficiency of broilers at the end of the study (p > 0.05). Neither small intestine weight nor pH was affected by any of the treatments. Supplementation of both synbiotic and phytobiotic to diet decreased the caecal coliform count (p < 0.01). Addition of synbiotics and phytobiotics in combination significantly increased plasma malondialdehyde (MDA) levels (p , 0.05), whereasphytobiotic addition alone showed only a slight increase. Similarly, elevated nitric oxide (NO) level was recorded in the synbiotic- and phytobiotic-fed group and in the phytobiotic-fed group (p , 0.001). Superoxide dismutase (SOD) activities did not differ between the groups. In conclusion, dietary supplementation of synbiotic and phytobiotic improved the gut health by decreasing the caecal total coliform count, but growth performance was not affected by the supplementations. Further investigations are needed to determine the effects of phytobiotics on oxidative/antioxidative metabolism as regards their compositional analysis. [source] ORIGINAL ARTICLE: Intestinal villus histological alterations in broilers fed dietary dried fermented gingerJOURNAL OF ANIMAL PHYSIOLOGY AND NUTRITION, Issue 5 2010T. Incharoen Summary To evaluate the effect of dietary dried fermented ginger (DFG) on intestinal villous histological alteration and growth performance, 64 Marshall Chunky male broilers were divided into four groups, each with four replicates of four chickens. Birds were fed the basal commercial mash diet supplemented with DFG at 0 (control), 5, 10 and 20 g/kg for 42 days. With increasing dietary DFG levels, feed intake tended to decrease and significantly decreased in the 20 g/kg DFG group (p < 0.05). Weight gain was higher in all the DFG groups, with the highest in the 10 g/kg DFG group (p < 0.05), resulting in an improved feed efficiency in all the DFG groups. Intestinal villus height, villus area, cell area and cell mitosis in all the intestinal segments were higher in all the DFG groups than in the control group. Protuberated cells and cell clusters were found in all the DFG groups, suggesting that the intestinal villi and cells might be hypertrophied. The present results indicate that dietary DFG can be used as a natural feed additive to induce broiler growth performance as a result of stimulation of morphological maturation and in consequence intestinal function. [source] The impact of nutrient density in terms of energy and/or protein on live performance, metabolism and carcass composition of female and male broiler chickens of two commercial broiler strainsJOURNAL OF ANIMAL PHYSIOLOGY AND NUTRITION, Issue 4 2010E. Delezie Summary The objective of this study was to investigate the effects of diet composition on performance, slaughter yield and plasma metabolites, as different modern broiler strains show different responses to feed intake. Broilers of two commercial strains and of both sexes received one of three diets being different in energy and/or protein level [control diet, low energy/low protein diet (LM/LP) and low protein diet (LP)]. Low energy/low protein diet chickens were characterized by significantly lower body weights and feed intake compared with their LP and control counterparts. Broilers of the Cobb strain or broilers that were fed the control diet were most efficient in converting energy to body weight. No significant differences in plasma metabolites were detected due to diet composition or genotype. The diet with the lower energy and crude protein levels reached the lowest slaughter yield but the highest drumstick and wing percentages. The lowest mortality percentages were observed for broilers fed the LM/LP diet, and Cobb birds appeared to be more sensitive for metabolic disorders resulting in death. It is obvious from this study that different genotypes respond differently to changes in diet composition and therefore have adjusted nutritional requirements. [source] Effects of different dietary phytase activities on the concentration of antioxidants in the liver of growing broilersJOURNAL OF ANIMAL PHYSIOLOGY AND NUTRITION, Issue 4 2010F. Karadas Summary One-hundred and fifty male chickens were used to evaluate the effects of different activities (0, 250, 500, 12 500 FTU/kg) of phytase on their performance and antioxidant concentration in the liver. The chicks were housed in 30 cages and were allocated to six replicates of five dietary treatments. All diets were formulated to be adequate in energy and protein (12.90 MJ/kg metabolizable energy, 214 g/kg crude protein), however, the negative control (NC) was lower in available P compared with the positive control (PC) (2.5 vs. 4.5 g/kg diet). The other three diets were the NC supplemented with phytase at 250, 500 and 12 500 FTU/kg (NC + 250, NC + 500 and NC + 12 500 FTU respectively). The concentration of antioxidants in the liver of the birds was determined using HPLC at 21 days of age. Low P diets (NC) reduced weight gain, however, supplementation with phytase improved weight gain to the extent that it was better than the PC at the 12 500 FTU treatment (p < 0.05). Feed conversion ratio was also improved by the high level of phytase supplement more than other treatments (p < 0.05). Feed consumption was not affected either by dietary phosphorus concentration or by different phytase supplementation. The antioxidant data showed that the unsupplemented diet with low phosphorus (NC) decreased the concentration of coenzyme Q10 and retinol-linoleate in the liver compared with that of birds on the adequate phosphorus treatment (PC). Phytase supplementation, especially at the higher doses (500 and 12 500 FTU) increased the level of coenzyme Q10 to the same level as the PC treatment. In addition, the highest dose (12 500 FTU) of phytase increased retinol concentration in the liver of chickens compared with those on the NC treatment. The highest inclusion level of phytase increased the ,-tocopherol level in the liver compared with the lower levels of phytase (NC + 250 and NC + 500 FTU). [source] Effect of an organic acid blend and phytase added to a rapeseed cake-containing diet on performance, intestinal morphology, caecal microflora activity and thyroid status of broiler chickensJOURNAL OF ANIMAL PHYSIOLOGY AND NUTRITION, Issue 1 2010S. Smulikowska Summary The experiment was carried out on 96 female broilers, allocated to eight groups of 12 birds kept in individual cages. Two basal wheat- and soyabean meal-based diets containing 150 g/kg of rapeseed expeller cake were formulated, differing in the level of P: 7.1 g/kg in diet H or 5.9 g/kg in diet L. Rapeseed cake supplied 3.15 ,mol alkenyl glucosinolates per gram of diet. The eight treatments were: basal diets only, basal diets + phytase (1000 U/kg), basal diets + organic acid blend (OA, 6 g/kg), or basal diets + both additives. Diets were fed from day 8 to 28 of life. The results showed that the lower dietary P content and OA supplementation did not significantly affect feed intake or BWG, while both increased (p < 0.001) after phytase supplementation. Tibia ash content as well as tibia ultimate strength were lower (p < 0.001) in birds fed diets L compared with diets H, and increased (p < 0.01) with phytase supplementation of diet L, while OA had no influence on either parameter. Dietary P levels and OA supplementation had no influence on the pH of gut digesta, but the pH of jejunal digesta increased following phytase supplementation (p < 0.01). Morphological measurements of the small intestinal mucosa of chicks indicated that OA added to diet L depressed villi height (p < 0.001) and crypt depth (p < 0.001); both parameters increased after phytase supplementation (p < 0.01). The lower total SCFA as well as acetic, propionic and butyric acid concentrations in caecal digesta indicated lower activity of caecal microflora in birds fed diets L compared with H. OA supplementation had no influence, while phytase supplementation increased the concentration of acetic acid in caecal digesta. Supplementation of diets with either phytase or OA increased thyroid weight by 16% (p < 0.01) and 11% (p < 0.05) respectively. The increase in thyroid weight because of phytase supplementation was greater at the lower dietary P level, and the greatest when both phytase and OA were added to the diet. [source] Intestinal function and gut microflora of broiler chickens as influenced by cereal grains and microbial enzyme supplementationJOURNAL OF ANIMAL PHYSIOLOGY AND NUTRITION, Issue 5 2009M. D. Shakouri Summary A study was conducted to investigate the effect of the key cereal grains and a microbial enzyme supplement on broiler chicken performance, gut microflora and intestinal function. Ingestion of the barley-based diet was associated with low 28-day body weight, decreased feed intake and high FCR. The supplemental enzyme increased feed intake and weight gain of the chickens on a wheat-based diet. The pH of the gizzard and caecal contents varied with the grain type. Enzyme supplementation reduced ileal viscosity, particularly in birds that received the diet based on wheat. The birds on the barley-based diet had lower ileal digestibility of dry matter, protein and energy than those given maize and sorghum-based diets. The ileal digestibility of starch was increased by enzyme supplementation. Enzyme supplementation increased the number of total anaerobic bacteria in the gizzard of birds fed on sorghum and increased lactobacilli in the gizzard of those fed both sorghum and wheat. The birds fed the sorghum-based diet had the lowest counts of caecal total anaerobic bacteria and lactobacilli. Jejunal villus height and villus:crypt ratio of birds fed the barley-based diet were the lowest when compared with those fed the other diets. Enzyme application induced an increase in villus height and villus:crypt ratio of birds on wheat, crypt depth on barley and a reduction in crypt depth of chickens on the sorghum-based diets. The highest activity of maltase and the lowest activity of sucrase were observed in tissue from birds fed on maize and sorghum-based diets respectively. The differences in the performance of broilers on cereal grains could be explained by changes in intestinal morphology, enzyme activities and gut microflora as well as nutrient digestibility. The improved performance by supplemental enzyme in wheat-fed chickens was associated with beneficial changes in intestinal morphology and digesta viscosity. [source] Evaluation of supplementary stevia (Stevia rebaudiana, bertoni) leaves and stevioside in broiler diets: effects on feed intake, nutrient metabolism, blood parameters and growth performanceJOURNAL OF ANIMAL PHYSIOLOGY AND NUTRITION, Issue 6 2008J. O. Atteh Summary A perennial schrub, stevia, and its extracts are used as a natural sweetener and have been shown to possess antimicrobial properties. Stevia contains high levels of sweetening glycosides including stevioside which is thought to possess antimicrobial and antifungal properties. Little is known about the nutritional value of the schrub in livestock. This study determined the potential use of the shrub as a prebiotic animal feed supplement in light of the recent ban on the use of antibiotics in animal feed and the role of its constituent stevioside in the effects of the shrub. Male Cobb broiler chicks were fed a basal broiler diet without antibiotic but with performance enhancing enzyme mix (positive control), a basal diet without antibiotic and enzymes (negative control), or diets in which 2% of the negative control diet was replaced with either dried ground stevia leaves or 130 ppm pure stevioside during 2 week starter and 2 week grower periods. Body weight gains, feed conversion, abdominal fat deposition, plasma hormone and metabolites and caecal short chain fatty acids (SCFA) were measured in the broilers at 2 and 4 weeks of age. There was no significant effect of the treatments on feed intake during the starter period but birds fed diet supplemented with stevia leaves and stevioside consumed more feed (p < 0.05) than those fed the positive control diet during the grower period. Weight gain by birds fed the positive control and stevioside diets was higher (p < 0.05) than those fed other diets only during the starter period. Feed/gain ratio of birds fed the positive control and stevioside diets was superior (p < 0.05) to others. There was no effect of the treatments on nutrient retention and water content of the excreta. Dietary stevia leave and stevioside decreased total concentration of SCFA and changed their profile in the ceca. There was no effect of the treatments on pancreas weight. Dietary stevia reduced blood levels of glucose, triglycerides and triiodothyronine (T3) but had no effect on non-esterified fatty acids. In contrast, stevioside only decreased T3. Both the stevia leaves and stevioside diets significantly increased abdominal fat content. It is concluded that dietary enzyme growth promoters are beneficial to the broilers only during the starter stage and that inclusion of stevia leaves or stevioside has no beneficial effect on the performance of broilers. [source] Body composition and heat expenditure in broiler chickens fed diets with or without trans fatty acidsJOURNAL OF ANIMAL PHYSIOLOGY AND NUTRITION, Issue 1 2008M. Javadi Summary The effect of a diet containing trans fatty acids (TFA) on the fatty acid composition and fat accumulation was investigated in broiler chickens. Female broilers were fed a control or a TFA-containing diet. The difference between the diets was that a part of cis 18:1 in the control diet was replaced by the TFA. Body composition, energy balance and the fatty acid composition were examined. Over the time-period studied (15 days), the body fat content and the protein content did not differ significantly between the TFA-fed group and the control. In energy balance studies, total energy intake, energy loss in excreta, energy expenditure and energy storage did not differ between the treatments. Compared to the control diet, the TFA diet resulted in significantly higher amounts of 14:0 and 18:1n-7 and lower amounts of 18:1n-9 and 20:4n-6 in the body. In conclusion, the data suggest that feeding TFA for 15 days to female broilers had no effect on energy retention, energy expenditure and energy loss in excreta or in body composition in terms of fat and protein. Only the fatty acid composition in the body was affected by the treatment with TFA. In addition, 50% of ingested TFA was incorporated into the body fat. Methods and Results:, The samples analysed were those for the European Union Salmonella baseline surveys of laying (N = 3087), broiler (N = 1550), turkey fattening (N = 1540) and turkey breeding (N = 580) flocks for Great Britain. Results were considered separately for Rambach (including and excluding pale orange colonies) and for growth on selective media [Modified semi-solid Rappaport Vassiliadis (MSRV)] after 24 and 48 h of incubation. Overall, Rambach was the most sensitive medium, provided that pale orange colonies were checked. In all cases, an increase in the sensitivity of detection was obtained by plating growth on MSRV after 48 h of incubation. In broilers and laying flocks, the specificity significantly improved when Rambach only was used. Conclusion:, The use of Rambach results in considerable savings compared with the two-plate method prescribed by ISO 6579:2002 (Annex D) without compromising sensitivity. Significance and Impact of the Study:,Salmonella isolation protocols should be reviewed in terms of their efficiency and cost. [source] Genotyping of thermotolerant Campylobacter from poultry slaughterhouse by amplified fragment length polymorphismJOURNAL OF APPLIED MICROBIOLOGY, Issue 2 2007G. Johnsen Abstract Aim:, To examine the occurrence, diversity and transmission of Campylobacter in a poultry slaughterhouse. Methods and Results:, During a 4-week period, a slaughterhouse was sampled alternately during slaughtering and the following mornings post-disinfection. Samples were taken from poultry at six stages in the slaughter process and from 25 environmental sites. For positive broiler flocks slaughtered on one occasion, 92% and 48% of the environmental sites were positive during slaughter and post-disinfection, respectively. For positive laying hen flocks slaughtered on three occasions, 8,56% and 12,20% of the environmental sites were positive during slaughter and post-disinfection, respectively. Genetic fingerprinting by amplified fragment length polymorphism (AFLP) of the 109 isolates obtained resulted in 28 different AFLP clones. Five AFLP clones were present for more than 1 week. Conclusions:, Slaughtering of Campylobacter -positive broilers resulted in extensive contamination of the slaughterhouse, including the air. A high proportion of the laying hen flocks were Campylobacter positive, but these caused less environmental contamination than the broilers. This, together with the freezing of all layer carcasses, results in a lower public health risk from laying hens, when compared with broilers. Adkin Abstract Aims:, Produce an evidence-based ranking of the major contributing factors and sources of Campylobacter occurrence in broilers produced in England, Scotland and Wales , Great Britain (GB). Method and Results:, Relevant data were extracted from 159 research papers and findings were grouped into 14 sources of on-farm contamination and 37 contributing factors. A relevancy score was developed to take into account various measures from each study of applicability to GB broilers and strength of findings. Results indicate that major sources of Campylobacter include a depopulation event, another house on-farm, on-farm staff, and other animals on farm. The depopulation schedule (staggered slaughter) and multiple houses on-farm were identified as contributing factors associated with increasing the risk, and those decreasing the risk were use of a hygiene barrier, parent company and certain seasons of rearing. Conclusions:, Although the review was more resource intensive compared to narrative studies, the system allows an increased level of transparency and the ability to investigate patterns and trends. Wainų Abstract Aims: To validate a phenotypic Campylobacter species identification method employed to identify campylobacters in broilers by comparison with campylobacterial species identification using various species-specific PCR analyses. Methods and Results: From a collection of 2733 phenotypically identified campylobacterial cultures, 108 Campylobacter jejuni cultures and 351 campylobacterial cultures other than Camp. jejuni were subjected to various species-specific PCR assays. On the basis of the genotypic tests, it was demonstrated that Camp. jejuni and Camp. coli constituted approx. 99% of all cultures, while other species identified were Helicobacter pullorum, Camp. lari and Camp. upsaliensis. However, 29% of the 309 Camp. coli cultures identified by phenotypic tests were hippurate-variable or negative Camp. jejuni cultures, whereas some Camp. lari cultures and unspeciated campylobacter cultures belonged to H. pullorum. It was also notable that 2,6% of the cultures were, in fact, mixed cultures. Conclusions: The phenotypic identification scheme employed failed to appropriately differentiate Campylobacter species and particularly to identify the closely related species, H. pullorum. Significance and Impact of the Study: Future phenotypic test schemes should be designed to allow a more accurate differentiation of Campylobacter and related species. Preferably, the phenotypic tests should be supplemented with a genotypic strategy to disclose the true campylobacterial species diversity in broilers. Bamiro In this article, the influence of vertical integration on cost behavior in poultry farming in southwestern Nigeria is examined. The study was based on primary data obtained in cross-section survey of 211 randomly selected poultry farms in the study area. An average farmer in the sample was 44 years old; 85% were males and 72% had tertiary education. An average poultry farm in the sample had 4,342 birds, about half of which were laying birds. A set of cost and revenue share equations estimated by Zellner's seemingly unrelated regression (SUR) method revealed that vertical integration (measured in proxy by value-added sales ratio) is feed and veterinary service using, labor saving, and output augmenting. However, the scale effect of vertical integration was found to be higher in layers production than what obtains in broilers and cock/cockerel productions. Nganje Turkey is an important food commodity whose total value of U.S. production amounted to $2.72 billion in 2003. Empirical evidence suggests that among broilers, eggs, turkeys, and chickens, Salmonella contamination of ground turkey is highest at 49.9% prior to hazard analysis and critical control point (HACCP) implementation and 26.6% after HACCP implementation. Salmonella and other microbial outbreaks have greatly contributed to the large number of food recalls in the meat and poultry industry; therefore, processed turkey constitutes a prime commodity for HACCP intervention analysis. Value-at-risk provides a framework for assisting firm management to assess food safety risks in monetary terms, and to evaluate the economic incentives of control measures like HACCP. Results show that food safety losses as measured by downside risk significantly declined following HACCP implementation. Medium- and large-scale turkey processors are more likely to derive more benefit from implementing an augmented HACCP plan than a generic HACCP plan. OSCAR ABSTRACT One strain of 11 serotypes or 11 strains of Salmonella, which were isolated from the ceca of broilers, were surveyed for their growth kinetics on sterile ground chicken breast burgers incubated at 25C to determine the variation of lag time and specific growth rate. Growth curves, four per strain, were fit to a two-phase linear model to determine lag time (h) and specific growth rate (log10/h). Repeatability of growth kinetics measurements for individual strains had a mean coefficient of variation of 11.7% for lag time (range: 5.8 to 17.3%) and a mean coefficient of variation of 6.7% for specific growth rate (range: 2.7 to 13.3%). Lag time among strains ranged from 2.2 to 3.1 h with a mean of 2.8 h for all strains, whereas specific growth rate among strains ranged from 0.3 to 0.38 log10 per h with a mean of 0.35 log10per h for all strains. One-way analysis of variance indicated that lag time (P =0.029) and specific growth rate (P =0.025) differed slightly among strains. S. Haardt had a shorter (P < 0.05) lag time than S. Agona and S. Brandenburg, whereas the specific growth rate of S. Enteritidis was less than (P < 0.05) the specific growth rates of S. Typhimurium and S. Brandenburg. All other strains had similar lag times and specific growth rates. The coefficient of variation among strains was 9.4% for lag time and 5.7% for specific growth rate. These results indicate that there were only minor differences in the lag times and specific growth rates among the strains of Salmonella surveyed. Thus, the growth kinetic values obtained with one strain of Salmonella may be useful for predicting the growth of other strains of Salmonella for which data do not currently exist. [source] Changes in Tenderness, Color, and Water Holding Capacity of Broiler Breast Meat during Postdeboning AgingJOURNAL OF FOOD SCIENCE, Issue 8 2009Y.S. Lee ABSTRACT:, The impact of postharvest aging on the tenderness, color, water holding capacity, and appearance of broiler breast fillets after deboning was investigated. A total of 360 broilers were processed and deboned at either 1.5-, 3-, or 6-h postmortem (PM) and aged at 4 ± 1 °C for up to 6 d. Tenderness was predicted by the Meullenet,Owens razor shear. Drip loss, cook loss, color, and muscle-shape profiles were also evaluated during the 6-d aging duration. Deboned fillets were in the tenderness range corresponding to "neither tough nor tender" for the first 2 d of aging and changed into "slightly tender" after 3 d of aging according to the instrument-tenderness perception equivalent scale. Tenderization due to postdeboning aging seemed to be more pronounced for fillets deboned in a prerigor state (that is, 1.5- and 3-h PM). Over the aging period, tenderness improved by 6.9 and 7.4 percentage points for the 1.5- and 3-h PM treatments, respectively, while those fillets deboned in a postrigor state (6-h PM) exhibited no significant difference in tenderness. Drip and cook loss of fillets consistently increased over the aging period. The color of fillets tended to become less red and more yellow during aging although there was no significant difference in,L*. Overall, the tenderizing effects of deboned broiler breast fillets during the storage of 6 d were minimal but seemed to be affected by fillet height and length as determined through analysis of covariance (ANCOVA) and K-means clustering analysis. Thicker and tougher fillets were more susceptible to the tenderizing effects during postdeboning aging. [source] THE MEULLENET-OWENS RAZOR SHEAR (MORS) FOR PREDICTING POULTRY MEAT TENDERNESS: ITS APPLICATIONS AND OPTIMIZATIONJOURNAL OF TEXTURE STUDIES, Issue 6 2008Y.S. LEE ABSTRACT The Meullenet-Owens Razor Shear (MORS), recently developed for the assessment of poultry meat tenderness, is a reliable instrumental method. Three different studies were conducted to (1) investigate the adaptation of MORS to an Instron InSpec 2200 tester (InSpec); (2) optimize the number of replications necessary per fillet to obtain a reliable instrumental tenderness mean; and (3) test the efficacy of a blunt version of MORS (BMORS). In study 1, the tenderness of 157 cooked broiler breast fillets was predicted by the MORS performed with both a texture analyzer (MORS standard) and InSpec. The correlation coefficient of 0.95 was reported for the MORS energy obtained from the both tests, indicating that the MORS performed with an InSpec is equivalent to that performed on the more expensive texture analyzer. In study 2, eight shears were taken on each cooked fillet (101 fillets) to determine a recommended number of shears per fillet for the MORS. The composite hypothesis test was conducted considering the average of 8 shears as Y (representative estimated tenderness of a fillet) and the average of 2, 3, 4, 5, 6 or 7 as X (potentials for recommended number of shears). The results showed that the optimal number of replications of the MORS for a reliable estimate of tenderness to be four shears or greater per fillet. A blunt version of MORS (BMORS) was introduced in study 3. A total of 288 broilers (576 fillets) were deboned at eight different postmortem deboning times. Tenderness of cooked fillets was assessed by both the MORS and BMORS on the same individual fillets. Both methods were equivalent in performance for predicting broiler breast meat tenderness, giving a correlation coefficient of 0.99 with all instrumental parameters obtained from both methods. Tenderness intensity perceived by consumers was slightly more highly correlated to BMORS energy (r = ,0.90) than MORS energy (r = ,0.87). The BMORS was recommended to use especially for tough meat because of its better discrimination ability among tough meat. Overall, both the MORS and BMORS were proven to be reliable predictors for broiler breast meat tenderness. PRACTICAL APPLICATIONS The incidence of tough meat has been a major issue the poultry industry faces. Therefore, the need to ensure consumer acceptance and the increased recognition of the importance of tenderness has led to the development of instrumental methods for monitoring meat tenderness. To date, a great deal of efforts has been devoted to the development of such instrumental methods. One promising method is the Meullenet-Owens Razor Shear (MORS). The method has gained in popularity for predicting poultry meat tenderness because of its high reliability as well as simplicity compared with that of other industry standards (Warner-Bratzler shear or Allo-Kramer shear). The MORS is not only as reliable as the industry standards, but also more rapid because of the elimination of the sample cutting steps. The application of the MORS will be of benefit to the poultry industry as it could significantly save labor or time to implement for routine quality control. [source] Effects of dietary humate supplementation to broilers on performance, slaughter, carcass and meat colourJOURNAL OF THE SCIENCE OF FOOD AND AGRICULTURE, Issue 7 2008Dr Nurinisa Esenbu Abstract BACKGROUND: This experiment was designed to examine the effect of dietary humate supplementation primarily on pH and colour parameters of carcasses, breast fillets and drumsticks and, secondarily on performance and carcass characteristics in broilers. RESULTS: A total of 240 male broiler chicks (Ross-308) were randomly allocated to four dietary treatments varying in supplemental humate level (0, 0.1, 0.2 and 0.3% for H0, H1, H2 and H3). Dietary humate supplementation did not affect performance traits and slaughter, hot carcass weights and yields. Carcass-related variables (pH, L*, a*, b*, H* and C*) were responsive to the dietary treatments. The L*, a*, b*, and C* values for drumstick muscles were higher than those for breast muscles. Except for the L* value, meat colour parameters changed due to packaging. The a* value was higher and b* value was lower for vacuum packaged breast and drumsticks than for those aerobic packaged. Storage period affected colour parameters; while L*, b*, H* and C* values were higher for drumstick skin than for drumstick meat; the a* value was greater in drumstick meat than in skin. CONCLUSION: pH and colour parameters of carcasses, breast fillets and drumsticks of broilers were improved by dietary humate supplementation. However, responses of broiler performance and slaughter and carcass characteristics were minimal. The effect of dietary COS supplementation on immune function in broilers was assessed by feeding graded levels (0, 50, 100, 150 mg kg,1) of COS. Two thousand four hundred male commercial Avian® broilers (1 day old) were assigned randomly to five dietary treatment groups (60 birds/pen with eight pens per treatment). Diet A was a typical maize- and soybean meal-based diet supplemented with 6 mg kg,1 antibiotics of flavomycin (positive control). Diet B was the basal diet without any supplement. Diets C, D and E were formulated by addition of 50, 100, 150 mg kg,1 of COS to the basal diet, respectively. On the morning of days 21 and 42, a total of 64 (eight birds/pen with eight pens per treatment) of the growth experimental birds were killed by cervical dislocation, respectively for determination of serum concentrations of IgG, IgA and IgM, and weights of spleen, bursa and thymus. Compared with broilers fed the control and the positive control diets, dietary supplementation with COS increased (P < 0.001) serum concentrations of IgG, IgA, and IgM, with the greatest response for the 100 mg kg,1COS supplementation. Chitosan oligosaccharides supplementation also enhanced the immune organ development. Results suggest COS can improve immune response in birds. Through positive modulation of the immune response, COS has great potential as an effective, antibiotic-like growth promoter for poultry. For wheat and barley, the influence of phytase, individually or in combination with glycanases, was also evaluated. Microbial phytase improved (p < 0.05) apparent ileal phosphorus digestibility in all cereals. Phytase supplementation improved (p < 0.05) the apparent metabolisable energy of maize and barley by 2.6 and 7.8%, respectively. Numerical improvements in apparent metabolisable energy were observed in sorghum (1.9%) and wheat (2.1%), but were not statistically significant (p > 0.05). Further improvements (p < 0.05) in the apparent metabolisable energy of wheat and barley were observed when the phytase was combined with glycanases. However, the observed improvements in apparent metabolisable energy were not always associated with enhanced ileal digestibility of protein and starch. Flaxseed was included in the diet fed to day-old broilers for the first 3 weeks as whole seed, ground seed, autoclaved whole seed, ground autoclaved whole seed or whole seed pelleted with the other ingredients, at levels of 0, 100, 120 and 140 g kg,1. Chicks fed the pelleted flaxseed-containing diets had heavier body weights, consumed more feed and had better feed/gain ratios than those fed the other flaxseed-containing diets during the 3 weeks period (P < 0.01). The flaxseed level in the diet also had very significant (P < 0.01) effects on body weight and feed/gain ratio at the end of weeks 1 and 3, with the diets containing 100 g kg,1 flaxseed resulting in better performance than the other flaxseed-containing diets. Among the flaxseed containing diets, the pelleted diets led to higher apparent ether extract digestibilities, with values of 778 and 770 g kg,1 for the diets containing 100 and 140 g kg,1 flaxseed respectively. This better utilisation of ether extract by young broilers may explain the significantly (P < 0.05) higher AMEn (apparent metabolisable energy) value of 2924 kcal kg,1 for the pelleted 140 g kg,1 flaxseed-containing diet. This was 15.4,17.5% higher than for the diets with the same level of flaxseed but provided as raw or autoclaved whole seed. The pelleting of flaxseed allowed an inclusion rate of 100 g kg,1 without any reduction in the performance and nutrient utilisation of broilers. CHAUVIN Several measurement units are available to quantify antimicrobial usage in veterinary medicine, to obtain diverse measures such as the weight of active substance used, the live weight (LW) treated, the fraction of animals exposed, the number of treatments recorded or the cost represented. These measures can be applied to study practices variability between farms, to characterize patterns of usage of the different antimicrobial classes or to follow evolution of antimicrobial usage with time. An investigation was carried out to specifically explore the influence of measurement units on the conclusions obtained from such studies. Antimicrobial exposure was explored in a sample of turkey and chicken broiler flocks, using six different units [kg of active compound, treatments, days of administration, kg of LW treated, animal daily dose to treat 1 kg of LW (ADDkg) and euros] to compare flocks usage variability and patterns of use of the different antimicrobial classes. Time-trends evolutions of macrolides usage in turkey broilers, characterized by percentage of flocks exposed and LW treated, were also compared. In all analyses, the measure gave different results without equivalences, highlighting the necessity for care in choosing the measurement unit and caution in interpreting the figures obtained.
