Changes in toxicity and bioavailability of lead in contaminated soils to the earthworm Eisenia fetida (savigny 1826) after bone meal amendments to the soil

Abstract The effect of bone meal (Ca5(PO4)3OH) amendments on lead (Pb) bioavailability to Eisenia fetida (Savigny 1826) was investigated. A standard uncontaminated soil was amended with Pb(NO3)2 solution to give Pb concentrations of 7,000 ,g/g of soil. After one week, bone meal was added to one half of the soil in the ratio 1:20 bone meal:soil. Immediately after addition of the bone meal, survival times of E. fetida were 23 and 41 h in the bone meal-free and bone meal-amended soil, respectively. Twentyeight days after addition of the bone meal, survival times of Eisenia fetida were 67 h in the bone meal-free soil and more than 168 h in the bone meal-amended soil. In a second experiment, a standard Organisation for Economic Co-operation and Development reproduction toxicity test was carried out, but in addition to Pb(NO3)2 solution, bone meal was added to the soil in the ratio 1:20 bone meal:soil. The bone meal-free soil was left for five weeks before addition of E. fetida. In the bone meal-amended soil, bon emeal was added to the soil one week after addition of the Pb. The soil was left for a further four weeks before addition of Eisenia fetida. Calculated toxicities were significantly lower for the bone meal-amended soil than those calculated for the bone meal-free soil. Twenty-eight-day median lethal concentrations (LC50s; concentration that is statistically likely to kill 50% of the exposed test organism within a given time period ± 95% confidence intervals) of Pb were 4,379 ± 356 ,g/g of soil for bone meal-free soil and 5,203 ± 401 ,g/g of soil for bone meal-amended soil. Twenty-eight-day median effect concentrations (EC50s; concentration causing a reduction by 50% of a stated parameter) of Pb for weight change were 1,408 ± 198 ,g/g of soil for bone meal-free soil and 3,334 ± 731 ,g/g of soil for bone meal-amended soil and EC50s for cocoon production were 971 ± 633 ,g/g of soil for bone meal-free soil and 1,814 ± 613 ,g/g of soil for bone meal-amended soil. Significant mortalities occurred at Pb concentrations of 2,000 ,g/g of soil in the bone meal-free soil and 5,000 ,g/g of soil in the bone meal-amended soil. Earthworm Pb body load was lower in the bone meal-treated soil than in the bone meal-free soil up to a Pb concentration of 5,000 ,g/g of soil. Earthworm Pb body load was approximately 100 ,g/g of worm in surviving earthworms in both experiments when significant mortality occurred. Water and diethylenetriamine pentaacetic acid-extractable soil Pb showed good correlations with earthworm Pb body load. These extractions could be used as estimates for Pb bioavailability. [source]


Combination of support vector machines (SVM) and near-infrared (NIR) imaging spectroscopy for the detection of meat and bone meal (MBM) in compound feeds

Abstract This study concerns the development of a new system to detect meat and bone meal (MBM) in compound feeds, which will be used to enforce legislation concerning feedstuffs enacted after the European mad cow crisis. Focal plane array near-infrared (NIR) imaging spectroscopy, which collects thousands of spatially resolved spectra in a massively parallel fashion, has been suggested as a more efficient alternative to the current methods, which are tedious and require significant expert human analysis. Chemometric classification strategies have been applied to automate the method and reduce the need for constant expert analysis of the data. In this work the performance of a new method for multivariate classification, support vector machines (SVM), was compared with that of two classical chemometric methods, partial least squares (PLS) and artificial neural networks (ANN), in classifying feed particles as either MBM or vegetal using the spectra from NIR images. While all three methods were able to effectively model the data, SVM was found to perform substantially better than PLS and ANN, exhibiting a much lower rate of false positive detection. Sensitive Monoclonal Antibody-based Sandwich ELISA for the Detection of Porcine Skeletal Muscle in Meat and Feed Products

ABSTRACT: A monoclonal antibody-based sandwich enzyme-linked immunosorbent assay (ELISA) was developed for the sensitive detection of porcine skeletal muscle in raw and heat-processed meat and feed products. Heat treatment of meat samples up to 132 °C for 2 h did not affect the assay performance. The assay uses a pair of monoclonal antibodies (MAbs 8F10 and 5H9) specific to skeletal muscle troponin I (TnI). MAb 8F10, reacting to mammalian TnI, is the capture antibody and the biotin-conjugated MAb 5H9, specific to porcine TnI, the detection antibody. The sandwich ELISA is able to detect 0.05% (w/w) of laboratory-adulterated pork in chicken, 0.1% (w/w) pork in beef mixtures, 0.05% (w/w) pork meal in soy-based feed, and 1% commercial meat and bone meal (MBM), containing an unknown amount of pork, in soy-based feed. This new assay provides a rapid and reliable means to detect the contamination of meat and feed products with trace amounts of porcine muscle tissue to ensure product quality and safety. [source]


Long-term results of subtotal colectomy for acquired hypertrophic megacolon in eight dogs

Objectives: To evaluate the long-term results of subtotal colectomy for acquired hypertrophic megacolon in the dog. Methods: Eight dogs with acquired hypertrophic megacolon underwent subtotal colectomy with preservation of the ileocolic junction. Long-term follow-up was obtained by clinical records and telephone interviews with the owners. Results: Eight large-breed dogs (age range: 6 to 12 years; mean age: 10·75 years) were enrolled. The use of bone meal, low levels of exercise, chronic constipation with dyschesia and tenesmus refractory to medical management were factors predisposing dogs to acquired hypertrophic megacolon. The diagnosis was confirmed in all animals by abdominal palpation, plain radiography and postoperative histopathological findings. There were no intraoperative complications. One dog died as a result of septic peritonitis. The clinical conditions (that is, resolution of obstipation and stool consistency) of the remaining seven dogs were improved at discharge; all animals returned to normal defecation in five to 10 weeks (mean: 7·3 weeks) and were alive 11 to 48 months (mean: 40·5 months) after surgery. Clinical Significance: Predominantly bony diet and/or low levels of physical activity may predispose dogs to acquired hypertrophic megacolon. Our results emphasise the long-term effectiveness of subtotal colectomy with preservation of the ileocolic junction in this condition. [source]


Evaluation of Alternative Protein Sources to Replace Fish Meal in Practical Diets for Juvenile Tilapia, Oreochromis spp

Two feeding experiments were conducted to evaluate if methionine is limiting in practical grow-out diets for tilapia, Oreochromis spp. Four diets containing 32% protein and 5% lipid were designed to compare the use of diets high in dehulled solvent-extracted soybean meal (DSESM) and expeller pressed soybean meal (EPSM) compared with a diet containing 6% fish meal (FM). Tilapia (4.78 ± 0.07 g, mean ± SD) were randomly stocked into twelve 600-L flow-through tanks at 20 fish per tank. After 6 wk, there were no notable trends or statistically significant differences (P > 0.05) in final mean weight, survival rate, and feed conversion ratio (FCR) among the treatments. Because results of this study indicated that DSESM could totally replace FM in practical diets for juvenile tilapia, a second batch of diets were formulated using other protein sources. Typical levels of cottonseed meal (CSM), DSESM, and meat and bone meal (MBM) were used to evaluate whether methionine could be limiting. Two basal diet formulations were tested either without or with methionine supplement (0.06/100 g diet). The first diet contained 15% CSM, 27% DSESM, and 10% MBM and the second diet contained 15% CSM and 37% DSESM. These diets contained 28% protein and 5% lipid. Tilapia (3.90 ± 0.05 g) were randomly stocked into twelve 60-L glass aquaria of a recirculation system at 18 fish per aquarium for 5 wk and then moved to the 600-L flow-through tanks for five more weeks. After 10 wk, there were no statistically significant differences (P > 0.05) in final mean weight, survival rate, and FCR among the four treatments. Results of the present study indicated that DSESM and EPSM could totally replace FM's inclusion rate in commercial diets for juvenile tilapia. Furthermore, methionine did not appear to be limiting in practical diets using typical levels of CSM, DSESM, and MBM as primary protein sources. [source]


The potential of land animal protein ingredients to replace fish meal in diets for cuneate drum, Nibea miichthioides, is affected by dietary protein level

Abstract A net pen experiment was carried out to examine the effect of dietary protein level on the potential of land animal protein ingredients as fish meal substitutes in practical diets for cuneate drum Nibea miichthioides. Two isocaloric basal (control) diets were formulated to contain 400 g kg,1 herring meal but two different digestible protein (DP) levels (400 versus 350 g kg,1). At each DP level, dietary fish meal level was reduced from 400 to 280, 200, 80 and 0 g kg,1 by incorporating a blend that comprised of 600 g kg,1 poultry by-products meal (PBM), 200 g kg,1 meat and bone meal (MBM), 100 g kg,1 feather meal (FEM) and 100 g kg,1 blood meal (BLM). Cuneate drum fingerling (initial weight 42 g fish,1) were fed the test diets for 8 weeks. Fish fed the test diets exhibited similar feed intake. Final body weight, feed conversion ratio and nitrogen retention efficiency was not significantly different between fish fed the basal diets containing 350 and 400 g kg,1 DP. Weight gain decreased linearly with the reduction of dietary fish meal level at the 350 g kg,1 DP level, but did not decrease with the reduction of dietary fish meal level at the 400 g kg,1 DP level. Results of the present study suggest that fish meal in cuneate drum diets can be completely replaced with the blend of PBM, MBM, FEM and BLM at the 400 g kg,1 DP level, based on a mechanism that excessive dietary protein compensate lower contents of bio-available essential amino acid in the land animal protein ingredients relative to fish meal. Nutrients apparent digestibility coefficients of selected protein sources for juvenile Siberian sturgeon (Acipenser baerii Brandt), compared by two chromic oxide analyses methods

Abstract Apparent digestibility coefficients (ADCs) of dry matter (ADCd), crude protein (ADCp), energy (ADCe) and amino acids in selected feedstuffs were determined for juvenile Siberian sturgeon (8.38 ± 0.20 g). The tested feedstuffs were fishmeal (FM), meat and bone meal (MBM), poultry by-product meal, hydrolysed feather meal, fermented feather meal solvent-extracted cottonseed meal and soybean meal. ADCs were determined using a reference diet and test diets at 7 : 3 ratios with 5 g kg,1 chromic oxide (Cr2O3) as an inert marker. Fish were reared in a recirculating system and fed to apparent satiation five times daily. Cr2O3 in diets and faeces samples were determined by inductively coupled plasma atomic emission spectrometry (ICP-AES) and acid-digestion colorimetry (AC) methods, respectively. The results showed that ICP-AES method was more accurate for Cr2O3 determination than AC method, and the results determined by ICP-AES method were used in this study. ADCd and ADCp of seven tested ingredients were lowest for MBM (59.1 and 84.5%) and highest for FM (79.9 and 94.5%); ADCe of tested ingredients were from 71.8% for SECM to 93.2% for FM. ADCs of amino acid in test ingredients followed similar trend to the ADCp. The ADCs of individual amino acids varied from 61.6% (histidine in MBM) to 98.8% (valine in FM). [source]


Apparent digestibilities of common feedstuffs for bluegill Lepomis macrochirus and largemouth bass Micropterus salmoides using individual test ingredients

Abstract Apparent digestibility of dry matter and energy, and availability of amino acids from blood meal (BM), fish meal (FM), meat and bone meal (MBM), poultry by-product meal (PBM), soybean meal (SBM), corn, wheat and yellow grease (YG) were determined for bluegill Lepomis macrochirus (mean weight, 57 g), and likewise, but not for BM or wheat, for largemouth bass Micropterus salmoides (mean weight, 30 g). To avoid nutrient interaction from a reference diet, diets containing 98.5% (985 g kg,1) of test ingredients were used, except for semi-solid, YG which was mixed with corn to permit pelletization. Faeces were collected by a siphoning method. Apparent dry matter digestibility values ranged from 50% (corn) to 87% (BM) for bluegill and from 53% (MBM) to 76% (PBM) for largemouth bass. Apparent energy digestibility values ranged from 53% (corn) to 92% (BM) for bluegill and from 63% (MBM) to 93% (YG) for largemouth bass. Apparent digestibility of most amino acids exceeded 90% for evaluated protein sources, except for MBM which showed slightly lower values (80,90%) for both fishes. Isoleucine digestibility from BM was relatively low (82%) for bluegill. High digestibility values for SBM, PBM and BM, indicate good potential for replacing FM in diets for both fishes. [source]


Amino acid availability and protein digestibility of several protein sources for Nile tilapia, Oreochromis niloticus

Abstract Apparent amino acid availability coefficients and protein digestibility of four animal products [fish meal (FM), meat and bone meal (MBM), poultry by-product and feather meal] and four plant protein-rich products [soybean meal (SBM), cottonseed meal-28, cottonseed meal-38 and corn gluten meal (CGM)] were determined for Nile tilapia, Oreochromis niloticus. Ingredients were incorporated to a practical reference diet at a 7 : 3 ratio (70% of reference diet and 30% of test ingredient). Chromic oxide was used as external digestibility marker. Among animal products poultry by-product meal (PBM; 89.7%) and FM (88.6%) presented the highest apparent protein digestibility (APD) while MBM (78.4%) and feather meal (78.5%) presented the lowest APD. Among plant protein-rich products CGM (91.4%) and SBM (92.4%) presented the highest APD values while cottonseed meal-28 presented the lowest APD (78.6%). Average apparent amino acid availability of feed ingredients was similar to protein digestibility with 92.3%, 89.6%, 73.4%, 80.7%, 88.9%, 84.4%, 91.2% and 79.7% values for SBM, CGM, cottonseed meal-28 and 38, FM, MBM, PBM and feather meal respectively. These results indicate that O. niloticus is able to utilize efficiently different feedstuffs. [source]


Effect of fishmeal substitution with various animal and/or plant protein sources in the diet of the abalone Haliotis discus hannai Ino

Abstract The effect of dietary substitution of animal and/or plant protein sources for fishmeal on the growth and body composition of juvenile abalone was determined. Nine experimental diets in triplicate were prepared: Con, SM, PM, CM, SPM, MB, SM+CM, SM+SPM and CM+SPM diets. A 350 g kg,1 fishmeal was included in the Con diet, and the whole fishmeal was substituted with a 580 g kg,1 soybean meal (SM), 335 g kg,1 poultry meal (PM), 370 g kg,1 corn gluten meal (CM), 325 g kg,1 silkworm pupae meal dehydrated (SPM), 590 g kg,1 meat and bone meal (MB), the combined 290 g kg,1 soybean meal and 180 g kg,1 corn gluten meal (SM+CM), 290 g kg,1 soybean meal and 160 g kg,1 silkworm pupae meal dehydrated (SM+SPM) and 180 g kg,1 corn gluten meal and 170 g kg,1 silkworm pupae meal dehydrated (CM+SPM) diets respectively. All experimental diets were iso-nitronic and iso-lipidic. The survival of abalone fed with the SM and SM+SPM diets was higher than that of abalone fed with the CM, SPM, MB and CM+SPM diets. Weight gain of abalone fed with the SM+SPM diet was higher than that of abalone fed with all the other experimental diets, except for that of abalone fed with the SM+CM diet. The combined soybean meal and corn gluten meal (SM+CM) or silkworm pupae meal (SM+SPM) could be replaced with the whole fishmeal in the diet for abalone and improved its performance. [source]


Apparent digestibility of selected feed ingredients for white shrimp Litopenaeus vannamei, Boone

Abstract Apparent digestibility coefficients of dry matter (DM), crude protein, crude lipid, gross energy, phosphorus and amino acids in Peruvian fish meal (FM), fermented soybean meal, extruded soybean meal, soybean meal, peanut meal, wheat gluten meal, corn gluten meal, shrimp byproduct meal, meat and bone meal (MBM), poultry meat meal and plasma protein meal (PPM) were determined for white shrimp (Litopenaeus vannamei). A reference diet (RF) and test diets (consisting of 70% RF diet and 30% of the feedstuff) were used with 0.5% chromic oxide as an external indicator. A total of 1440 shrimp (initial mean body weight 1.05 ± 0.01 g) were randomly stocked into thirty-six 500-L fibreglass tanks with 40 shrimp per tank and three tanks per diet. Faeces were collected from triplicate groups of shrimp by a faecal collection vessel attached to the shrimp-rearing tank. The shrimp were fed to apparent satiation four times a day and the feeding experiment lasted for 6 weeks. Statistics indicate that apparent DM digestibilities for white shrimp (L. vannamei) were the highest for FM, ranged 52.83,71.23% for other animal products and 69.98,77.10% for plant products. The protein and lipid from plant and animal sources were well digested by white shrimp. Apparent protein and lipid digestibility were in the range 87.89,93.18% and 91.57,95.28%, respectively, in plant products, and 75.00,92.34% and 83.72,92.79%, respectively, for animal products. The white shrimp demonstrated a high capacity to utilize phosphorus in the ingredients. The apparent phosphorus digestibility ranges of animal feedstuffs and plant feedstuffs were 58.90,71.61% and 75.77,82.30% respectively. Amino acid availability reflected protein digestibility, except that in MBM, for which the availability of some amino acid was lower, possibly due to protein damage during processing. Digestibility information could promote the use of ingredient substitution in least-cost formulated diets for white shrimp. [source]


Evaluation of rendered animal protein ingredients for replacement of fish meal in practical diets for gibel carp, Carassius auratus gibelio (Bloch)

Abstract A 12-week feeding trial was carried out in fibreglass tanks to examine partial replacement of fish meal (FM) with poultry by-product meal (PBM), meat and bone meal (MBM) and blood meal (BM) in practical diets for gibel carp Carassius auratus gibelio (Bloch). Triplicate groups of fish (initial body weight 15.3 g fish,1) were fed eight isonitrogenous (crude protein: 37.5%) and isolipidic diets (crude lipid: 7%). The control diet is the commercial diet of gibel carp, which used 18% of FM as the sole animal protein source. In the other seven diets (Diet 2,Diet 8), 17,83% of FM protein was substituted by a blend of PBM and BM or a blend of PBM, MBM and BM. The final body weight and thermal-unit growth coefficient of fish fed the feeds in Diet 8 in which 83% of FM protein was replaced by the blend of 3% BM, 10% PBM and 5% MBM is significantly lower than Diet 1 (the control). The feed efficiency ratio in Diet 8 group is significantly lower than Diet 1, 2, 4 and 7 groups. The results of the present study indicated that a combination of PBM, BM and MBM can replace most of the FM protein and the FM level can be reduced to about 6% with satisfactory growth and feed utilization in practical diets for gibel carp. Low-cost diet for monoculture of giant freshwater prawn (Macrobrachium rosenbergii de Man) in Bangladesh

Abstract An experiment was conducted for 3 months in 12 experimental ponds, each of 30 m2, with a view to develop a low-cost diet for monoculture of Macrobrachium rosenbergii in ponds. Three experimental diets (30% protein) were formulated using fish meal, meat and bone meal, mustard oilcake, sesame meal and rice bran in different combinations partially replacing fish meal by meat and bone meal and sesame meal and assigned to treatments T1, T2 and T3 respectively. A commercial golda feed (Starter-III) was assigned to T4 (reference diet). Each treatment had three replicates. Juveniles of M. rosenbergii (2.90±0.21 g) were stocked at the rate of 40 000 ha,1. Prawns were fed three times daily at the rate of 10% and 5% of their body weight at the beginning and for the last 2 months respectively. The ponds were provided with aeration during the night using air pumps. The ranges of water quality parameters recorded in different ponds were: temperature 28.9,32.5°C, dissolved oxygen 5.1,8.1 mg L,1 and pH 6.4,7.7. The results showed that the weight gain of prawns fed diet 1 was significantly higher (P<0.05) than those fed diets 2 and 3, but was not significantly different from those fed diet 4 (reference diet). The feed conversion ratio (FCR) values of diets ranged between 2.21 and 2.96 with diets 1 and 4 showing significantly lower (P<0.05) FCR values. The survivals (%) ranged between 68% and 78% with prawns fed diets 1 and 4 showing significantly higher survival. The production of prawn ranged between 921 and 1428 kg ha,1 and diet 1 resulted in a significantly high (P<0.05) production. A simple economic analysis showed that diet 1 generated the maximum net profit of Tk 159 178 ha,1. The results of the study showed that a diet containing 20% fish meal, 10% meat and bone meal, 15% mustard oilcake, 15% sesame meal, 35% rice bran, 4% molasses and 1% vitamin,mineral premixes may be recommended to the farmers for monoculture of M. rosenbergii in ponds.
