The effect of adding cadmium and lead alone or in combination to the diet of pigs on their growth, carcase composition and reproduction

Abstract Limits for cadmium and lead concentrations in food animal products have been established independently, whereas these two toxic metals often co-exist in polluted regions. Weaned pigs (60) were allocated to ten treatments: control; low (0.5 mg kg,1), medium (1 mg kg,1) or high cadmium (2.5 mg kg,1) in feed; low (5 mg kg,1) medium (10 mg kg,1) or high (25 mg kg,1) lead in feed; and low, medium and high cadmium plus lead in feed. Growth rates and concentrations of cadmium and lead in body tissues (kidney, liver, spleen, lungs, heart, testicle, ribs, hair and teeth) were measured after 137 days. There was a similar reduction in weight gain for pigs in the cadmium and lead treatments, compared with the control, and a greater reduction for the pigs in the cadmium plus lead treatments. The reduction increased with the level of metal included. There was an increase in cadmium concentration of all tissues and blood with increasing feed cadmium concentration, which was usually less when lead was also included in the feed. There was also an increase in tissue lead concentration with increasing dietary lead, and this was in most cases increased when cadmium was also included in the feed. The most sensitive tissues for cadmium and lead exposure were the kidney, liver, hair and teeth, and regression equations were developed for the accumulation rates in these tissues. Tissue and blood cadmium concentrations increased gradually with increasing dietary lead, whereas tissue lead concentration was not sensitive to dietary cadmium, except in the ribs and heart. In a second experiment, 10 sows were allocated to a control diet or the same diet but with a supplement of cadmium and lead. The birth weight of piglets was decreased by the supplement and their mortality increased. Lead accumulated most in the ovary and oviduct of the sows, and there were increases in the lead and, to a lesser extent, cadmium concentrations of tissues of the piglets from these sows. Supplementation of diets for lactating sows with zinc amino acid complex and gastric nutriment-intubation of suckling pigs with zinc methionine on mineral status, intestinal morphology and bacterial translocation in lipopolysaccharide-challenged weaned pigs

Summary Sixty-four pigs from 16 sows were used to evaluate addition of zinc amino acid complex (ZnAA) to lactating sows and gastric nutriment-intubation of zinc methionine (ZnMet) to suckling pigs on mineral status, intestinal morphology and bacterial translocation after weaning. Sows were fed a barley-based diet supplying 120 ppm zinc (Zn; control) or the control diet supplemented with 240 ppm Zn from ZnAA. At birth, day-10 and day-21 (weaning) of age, pigs from each litter were nutriment-intubated with 5 ml of an electrolyte solution without or with 40 mg Zn from ZnMet. At weaning, 24 h prior to the collection of small and large intestinal lymph nodes and sections of the duodenum, jejunum and ileum, the pigs received an intramuscular injection of saline without or with 150 ,g/kg body weight of Escherichia coli O26:B6 lipopolysaccharide (LPS). With the exception of a tendency (p = 0.09) for lower serum concentration of copper in pigs at weaning from ZnAA-supplemented sows, there were no differences (p > 0.1) than for pigs from control-fed sows for mineral status or intestinal morphology. Nutriment-intubation of ZnMet increased serum (p = 0.001) and liver (p = 0.003) Zn concentrations, number of goblet cells per 250 ,m length of jejunal villous epithelium (p = 0.001) and tended (p = 0.06) to enhance jejunum mucosa thickness. Interactive effects (p < 0.05) for higher jejunal villi height and villi:crypt ratio and increased ileal goblet cell counts were apparent for pigs from ZnAA-supplemented sows that also received nutriment-intubation of ZnMet. Challenge with LPS increased (p = 0.05) ileal villous width. Nutriment-intubation of ZnMet decreased (p = 0.05) anaerobic bacteria colony forming unit counts in the large intestinal mesenteric lymph nodes. In conclusion, nutriment-intubation of ZnMet increased serum and liver tissue concentrations of Zn and resulted in limited improvement to intestinal morphology of weaned pigs. [source]


In vitro assessment of antimicrobial activity of carvacrol, thymol and cinnamaldehyde towards Salmonella serotype Typhimurium DT104: effects of pig diets and emulsification in hydrocolloids

Abstract Aims:, To determine the effect of pig diets in vitro on the antimicrobial activity of carvacrol, thymol and cinnamaldehyde, and to identify an emulsifier/stabilizer that can stabilize the essential oil (EO) components in aqueous solution and retain their antimicrobial activity in the presence of the diets. Methods and Results:, Emulsification of essential oil components with hydrocolloid solution was achieved by blending with a Polytron. Antimicrobial activity was measured through in vitro assays to determine the inhibition of bacterial growth by measuring the optical density at 600 nm or plating on nutrition agar after incubation of the mixtures of an EO component with the culture of Salmonella serotype Typhimurium DT104 in the presence or absence of pig diets. The results generated through the in vitro assays indicated that pig diets were able to abolish the antimicrobial activity of EOs. Xanthan, fenugreek and yellow mustard gums were the best in forming stable emulsions of five different EO components among ten different plant polysaccharides and surfactants examined. Emulsification of all the EO components in the fenugreek gum solution did not alter their antimicrobial activity. However, the antimicrobial activity of geraniol was significantly reduced when emulsified with other polysaccharides and surfactants. Both fenugreek and xanthan gum solutions were unable to protect the antimicrobial activity of carvacrol and thymol when mixed with the diets. Although cinnamaldehyde required no emulsification, but a high concentration (equivalent to at least three times of minimum bactericidal concentration for cinnamon oil) to inhibit Salmonella growth significantly in the presence of the diets, emulsification in fenugreek gum appeared to be essential for cinnamaldehyde solution to retain its antimicrobial activity during storage. Conclusions:, The diets for newly weaned pigs were a significant factor limiting the antimicrobial activity of EOs and their components. Cinnamaldehyde required a high concentration to retain its antimicrobial activity in the diets, in addition to its requirement for emulsification to stabilize its activity during the storage. Significance and Impact of the Study:, The assay with the diets used in this study for measuring the antimicrobial activity can be used in vitro for rapid and effective screening of potential antimicrobials for swine production. This study has identified polysaccharides that are able to stabilize EO component solutions. It has also identified cinnamaldehyde for further in vivo studies that may have potential in future application in controlling Salmonella and possibly other enteric pathogens in swine production. Estimates of basal ileal endogenous losses of amino acids by regression analysis and determination of standardised ileal amino acid digestibilities from casein in newly weaned pigs,

Abstract BACKGROUND: This study was carried out to estimate basal ileal endogenous crude protein (CP) and amino acid (AA) losses (IAALB) in newly weaned pigs by regression analysis from apparent ileal digestible and the corresponding total dietary contents of CP and AA in order to obtain standardised ileal digestibilities (SID) of CP and AA originating from casein. A total of fourteen 3-week-old barrows were fitted with simple T cannulas at the distal ileum and were fed six diets with two pigs per diet in 4 weekly repeated measurements. Corn starch-based diets containing six graded levels of CP from casein with 90, 155, 220, 285, 350 and 415 g kg,1 CP (as-fed basis) were formulated. RESULTS: Linear relationships (P < 0.001) between apparent ileal digestible and total dietary contents of CP and AA (g kg,1 dry matter intake (DMI)) exist for CP and all AA in the range of 90,220 g kg,1 CP content in the diet. The IAALB extrapolated from the range of 90,220 g kg,1 CP content are different from zero (P < 0.001 to P = 0.019) except for tryptophan (P = 0.220). Based on regression analysis in the range of 90,220 g kg,1 CP, estimates of IAALB, expressed as g kg,1 DMI, and SID of CP and AA in casein (in parentheses, %) were as follows: CP 9.3 (98.0); arginine 0.4 (98.9); histidine 0.2 (98.5); isoleucine 0.2 (98.1); leucine 0.5 (99.2); lysine 0.4 (99.0); methionine 0.1 (99.1); phenylalanine 0.4 (99.9); threonine 0.6 (97.8); tryptophan 0.1 (96.2); valine 0.4 (98.5). CONCLUSION: The IAALB and SID of CP and AA originating from casein in newly weaned pigs are in close agreement with values obtained in grower finisher pigs.
