Selected AbstractsIn vitro and in vivo evaluation and a case report of intense nanosecond pulsed electric field as a local therapy for human malignanciesINTERNATIONAL JOURNAL OF CANCER, Issue 3 2007Edward B. Garon Abstract When delivered to cells, very short duration, high electric field pulses (nanoelectropulses) induce primarily intracellular events. We present evidence that this emerging modality may have a role as a local cancer therapy. Five hematologic and 16 solid tumor cell lines were pulsed in vitro. Hematologic cells proved particularly sensitive to nanoelectropulses, with more than a 60% decrease in viable cells measured by MTT assay 96 hr after pulsing in 4 of 5 cell lines. In solid tumor cell lines, 10 out of 16 cell lines had more than a 10% decrease in viable cells. AsPC-1, a pancreatic cancer cell line, demonstrated the greatest in vitro sensitivity among solid tumor cell lines, with a 64% decrease in viable cells. When nanoelectropulse therapy was applied to AsPC-1 tumors in athymic nude mice, responses were seen in 4 of 6 tumors, including clinical complete responses in 3 of 6 animals. A single human subject applied nanoelectropulse therapy to his own basal cell carcinoma and had a complete pathologic response. In summary, we demonstrate that electric pulses 20 ns or less kill a wide variety of human cancer cells in vitro, induce tumor regression in vivo, and show efficacy in a single human patient. Therefore, nanoelectropulse therapy deserves further study as a potentially effective cancer therapy. © 2007 Wiley-Liss, Inc. [source] Mortality in a Swedish rubber tire manufacturing plant: Occupational risks or an "unhealthy worker" effect?AMERICAN JOURNAL OF INDUSTRIAL MEDICINE, Issue 8 2006Gun Wingren PhD Abstract Background In this cohort study a classification of 12 work categories, with specified common exposures, is used to evaluate the mortality among workers in a Swedish rubber tire plant. Methods Mortality among the cohort members is compared with expected values from national rates. Standardized mortality ratios (SMR) were calculated for the total cohort, for sub-cohorts and with the inclusion of a latency requirement. Results Male workers had increased risks for total mortality, circulatory death, respiratory death, mental disorders, epilepsy, injuries/poisoning, and for alcohol-related death. Many risks were noted among workers exposed to dust, fumes, or vapor. No trend in risk with increasing duration of employment was seen. Discussion Some established risks for rubber workers could be related to some specified work categories. The highest risks were noted among workers having a very short duration of employment indicating a possible "unhealthy worker effect" associated with life-style factors or to tasks offered to unskilled workers. [source] Persistent isolated hypocortisolism following brief treatment with trilostaneAUSTRALIAN VETERINARY JOURNAL, Issue 12 2008IK Ramsey A 12-year-old male neutered Miniature Poodle with confirmed pituitary-dependent hyperadrenocorticism was treated with trilostane. After three doses, it developed clinical and laboratory changes suggestive of isolated hypocortisolism (,atypical hypoadrenocorticism'), which persisted and progressed for more than 3 months despite immediate withdrawal of the trilostane. The clinical signs of hyperadrenocorticism resolved without further trilostane. After 3 months, prednisolone treatment was started and the clinical signs of hypocortisolism resolved. Prednisolone therapy was required for more than 1 year. Ultrasonography initially demonstrated large hypoechoic adrenal cortices, typical of dogs with hyperadrenocorticism, which then became small and heteroechoic, consistent with the development of adrenal necrosis. Persistent isolated hypocortisolism has not been reported previously as a complication of trilostane therapy. The case is also remarkable for the very short duration of trilostane therapy that elicited this complication. Clinicians should be aware that trilostane therapy may result in adrenal necrosis, even in the very earliest stages of therapy, but prompt action can prevent a life-threatening situation. [source] An investigation of the relationship between hullability and morphological features in grains of four oat varietiesANNALS OF APPLIED BIOLOGY, Issue 2 2010E. White Milling of oats involves separation of the kernel from its enclosing husk. The ease with which this happens is termed ,hullability'. The association between morphological characteristics measured using image analysis and hullability was examined in oat varieties sampled from Recommended List variety trials conducted at AFBI Crossnacreevy, Belfast, UK in 2000 and 2006. Grain features of the varieties were distinctive and showed differing relationships with weight, Barra changing only slightly, whilst Mascani showed the maximal changes with increasing grain weight. Hullability was poorer in larger grain, and better in Barra and Mascani than in Gerald and Tardis. Fitting of exponential models for the percentage of hulled grain with dehulling duration identified features associated with different aspects of dehulling behaviour. Greater width of grains of Mascani and Tardis was associated with a more rapid increase in hullability at shorter durations of dehulling. Greater roundness in Barra and Gerald in 2000 was associated with better hullability at very short durations of dehulling relative to other varieties. It is concluded that these features of the grain could be of use in selecting high hullability in breeding programmes. [source] |
