The heterogeneity of causes and courses of attention-deficit/hyperactivity disorder

Objective:, Attention-deficit / Hyperactivity Disorder (ADHD) is a frequent mental disorder with onset in childhood and persistence into adulthood in a sizeable number of people. Despite a rather simple clinical definition, ADHD has many facets because of frequent co-morbid disorders and varying impact on psychosocial functioning. Thus, there is considerable heterogeneity in various domains. Method:, A review of recent research findings in: i) selected domains of aetiology reflecting the role of genes, brain structures and functioning and the interplay of causal factors and ii) clinical heterogeneity in terms of co-morbidities, gender effects, courses and outcomes. Results:, Molecular genetic studies have identified a number of candidate genes which have a small effect on behavioural variation in ADHD. In the most recent Genome Scan Meta Analysis of seven ADHD linkage studies, genome-wide significant linkage was identified on chromosome 16. The volume of both the total brain and various regions including the prefrontal cortex, the caudate nucleus and the vermis of the cerebellum is smaller in ADHD. Functional MRI has documented a specific deficit of frontostriatal networks in ADHD. Integrative aetiological models have to take the interaction of gene and environment on various dysfunctions into account. Clinical heterogeneity results from frequent associations with various co-morbidities, the impact of the disorder on psychosocial functioning, and gender effects. Partly, these effects are evident also in the course and outcome of ADHD. Conclusion:, ADHD is a chronic mental disorder with a complex aetiology. So far, various neurobiological factors have been identified that need to be studied further to better understand their interaction with environmental factors. The clinical presentation and the long-term course of ADHD are manifold. [source]


A diffusible signal attracts olfactory sensory axons toward their target in the developing brain of the moth

Abstract The signals that olfactory receptor axons use to navigate to their target in the CNS are still not well understood. In the moth Manduca sexta, the primary olfactory pathway develops postembryonically, and the receptor axons navigate from an experimentally accessible sensory epithelium to the brain along a pathway long enough for detailed study of regions in which axon behavior changes. The current experiments ask whether diffusible factors contribute to receptor axon guidance. Explants were made from the antennal receptor epithelium and co-cultured in a collagen gel matrix with slices of various regions of the brain. Receptor axons were attracted toward the central regions of the brain, including the protocerebrum and antennal lobe. Receptor axons growing into a slice of the most proximal region of the antennal nerve, where axon sorting normally occurs, showed no directional preference. When the antennal lobe was included in the slice, the receptor axons entering the sorting region grew directly toward the antennal lobe. Taken together with the previous in vivo experiments, the current results suggest that an attractive diffusible factor can serve as one cue to direct misrouted olfactory receptor axons toward the medial regions of the brain, where local cues guide them to the antennal lobe. They also suggest that under normal circumstances, in which the receptor axons follow a pre-existing pupal nerve to the antennal lobe, the diffusible factor emanating from the lobe acts in parallel and at short range to maintain the fidelity of the path into the antennal lobe. © 2003 Wiley Periodicals, Inc. Polychlorinated biphenyls, organochlorine pesticides, tris(4-chlorophenyl)methane, and tris(4-chlorophenyl)methanol in livers of small cetaceans stranded along Florida coastal waters, USA

Abstract Concentrations of polychlorinated biphenyl congeners (PCBs) and organochlorine pesticides were determined in the livers of bottlenose dolphins, Atlantic spotted dolphins, and pygmy sperm whales found stranded along the coastal waters of Florida, USA, during 1989 to 1994. The PCBs were the most predominant contaminants followed in order by DDTs, chlordanes, tris(4-chlorophenyl)methane (TCPMe), tris(4-chlorophenyl)methanol (TCPMOH), hexachlorobenzene, and hexachlorocyclohexane isomers. Among the cetaceans analyzed, organochlorine concentrations were greatest in bottlenose dolphins followed by Atlantic spotted dolphins and pygmy sperm whales. Hexa- and heptachlorobiphenyls were the predominant PCB congeners found in the livers of dolphins. Patterns of relative concentrations of PCB congeners varied among individual bottlenose dolphins. A few individuals contained predominant concentrations of octa- (CB-199, 196/201) and nonachlorobiphenyl (CB-206, 208) congeners, which suggested exposure to the highly chlorinated PCB formulation, Aroclor® 1268, a contaminant at a coastal site in Georgia bordering northern Florida. The estimated 2,3,7,8-tetrachlorodibenzo- p -dioxin toxic equivalents (TEQs) of coplanar PCBs in bottlenose dolphins were 170 to 18,000 pg/g, lipid weight (mean: 5,400 pg/g) with mono- ortho congeners 118, 105, and 156 contributing more than 80% of the TEQs. The ratios of CB-169 to CB-126 in cetacean livers were linearly related to total PCB concentrations, which suggested a strong induction of microsomal monooxygenase enzymes in the liver. The hepatic concentrations of TCPMe and TCPMOH in bottlenose dolphins and Atlantic spotted dolphins were greater than those in the blubber of marine mammals of various regions, which suggested the presence of sources for these chemicals along the Atlantic coast of Florida. Cooperation between toll-like receptor,2 and,4 in the brain of mice challenged with cell wall components derived from gram-negative and gram-positive bacteria

Abstract In this study we investigated whether induction of toll-like receptor,2 (TLR2) amplifies the effect of a cell wall component derived from gram-positive bacteria, namely peptidoglycan (PGN). Mice received a first systemic lipopolysaccharide (LPS) injection to pre-induce TLR2 in various regions of the brain, and 6,h later, a second administration of either LPS or PGN. The data show a robust transcriptional activation of TLR2, TNF-, and monocyte chemotactic protein-1 (MCP-1) in microglial cells of mice challenged twice with LPS, whereas PGN essentially abolished this response. TLR4 plays a critical role in this process, because C3H/HeJ mice no longer responded to LPS but exhibited a normal reaction to PGN. Conversely, a robust signal for genes encoding innate immune proteinswas found in the brain of TLR2-deficient mice challenged with LPS. However, the second LPS bolus failed to trigger TNF-, and IL-12 in TLR2-deficient mice, while the same treatment caused a strong induction of these genes in the cerebral tissue of wild-type littermates. The present data provide evidence that cooperation exists between TLR4 and TLR2. While TLR4 is absolutely necessary to engage the innate immune response in the brain, TLR2 participates in the regulation of genes encoding TNF-, and IL-12 during severe endotoxemia. Such collaboration between TLR4 and TLR2 may be determinant for the transfer from the innate to the adaptive immunity within the CNS of infected animals. [source]


Identification of a cis -acting element required for dendritic targeting of activity-regulated cytoskeleton-associated protein mRNA

Abstract The mRNA encoding activity-regulated cytoskeleton-associated protein (Arc) is known to be targeted to dendritic regions that have received strong synaptic inputs. However, the cis- acting elements in Arc mRNA that mediate dendritic targeting have not been identified. To identify the dendritic targeting element (DTE) in rat Arc mRNA, we expressed reporter mRNAs containing various regions of Arc in primary hippocampal neurones and analysed their subcellular distribution by in situ hybridization. Here, we report that the 3,-untranslated region of rat Arc mRNA contains a 350-nucleotide DTE with strong dendritic targeting activity and another 370-nucleotide sequence with weaker dendritic targeting activity. The 350-nucleotide DTE does not share any obvious sequence similarity with other known DTEs previously reported. [source]


The immunocytochemical localization of connexin 36 at rod and cone gap junctions in the guinea pig retina

Abstract Connexin 36 (Cx36) is a channel-forming protein found in the membranes of apposed cells, forming the hexameric hemichannels of intercellular gap junction channels. It localizes to certain neurons in various regions of the brain including the retina. We characterized the expression pattern of neuronal Cx36 in the guinea pig retina by immunocytochemistry using specific antisera against Cx36 and green/red cone opsin or recoverin. Strong Cx36 immunoreactivity was visible in the ON sublamina of the inner plexiform layer and in the outer plexiform layer, as punctate labelling patterns. Double-labelling experiments with antibody directed against Cx36 and green/red cone opsin or recoverin showed that strong clustered Cx36 immunoreactivity localized to the axon terminals of cone or close to rod photoreceptors. By electron microscopy, Cx36 immunoreactivity was visible in the gap junctions as well as in the cytoplasmic matrices of both sides of cone photoreceptors. In the gap junctions between cone and rod photoreceptors, Cx36 immunoreactivity was only visible in the cytoplasmic matrices of cone photoreceptors. These results clearly indicate that Cx36 forms homologous gap junctions between neighbouring cone photoreceptors, and forms heterologous gap junctions between cone and rod photoreceptors in guinea pig retina. This focal location of Cx36 at the terminals of the photoreceptor suggests that rod photoreceptors can transmit rod signals to the pedicle of a neighbouring cone photoreceptor via Cx36, and that the cone in turn signals to corresponding ganglion cells via ON and OFF cone bipolar cells. Environmental and economic analysis of the fully integrated biorefinery

Abstract Cellulosic biofuel systems have the potential to significantly reduce the environmental impact of the world's transportation energy requirements. However, realizing this potential will require systems level thinking and scale integration. Until now, we have lacked modeling tools for studying the behavior of integrated cellulosic biofuel systems. In this paper, we describe a new research tool, the Biorefinery and Farm Integration Tool (BFIT) in which the production of fuel ethanol from cellulosic biomass is integrated with crop and animal (agricultural) production models. Uniting these three subsystems in a single combined model has allowed, for the first time, basic environmental and economic analysis of biomass production, possible secondary products, fertilizer production, and bioenergy production across various regions of the United States. Using BFIT, we simulate cellulosic ethanol production embedded in realistic agricultural landscapes in nine locations under a collection of farm management scenarios. This combined modeling approach permits analysis of economic profitability and highlights key areas for environmental improvement. These results show the advantages of introducing integrated biorefinery systems within agricultural landscapes. This is particularly true in the Midwest, which our results suggest is a good setting for the cellulosic ethanol industry. Specifically, results show that inclusion of cellulosic biofuel systems into existing agriculture enhances farm economics and reduces total landscape emissions. Model results also indicate a limited ethanol price effect from increased biomass transportation distance. Sensitivity analysis using BFIT revealed those variables having the strongest effects on the overall system performance, namely: biorefinery size, switchgrass yield, and biomass farm gate price. Different Helicobacter pylori Strains Colonize the Antral and Duodenal Mucosa of Duodenal Ulcer Patients

Background. We have investigated the possibility that the same patients may be colonized by Helicobacter pylori strains of different genotypes or phenotypes in the antrum as compared to in the duodenum. The strains were typed for DNA fingerprints, different lipopolysaccharides (LPS), and Lewis antigen expression on the O,side chains of LPS. Materials and Methods. Polymerase chain reaction (PCR) amplifications using primer sequences (i.e., the Enterobacterial Repetitive Intergenic Consensus [ERIC]) and randomly amplified polymorphic DNA (RAPD) elements were performed to asses chromosomal DNA diversity between H. pylori strains. The expression of different LPS types and Lewis antigens in the various H. pylori isolates were determined by whole bacterial enzyme-linked immunosorbent assays using monoclonal antibodies. Results. Duodenal ulcer patients had different H. pylori genotypes in the duodenum as compared to in the antrum as shown by ERIC-PCR (44%) and by RAPD-PCR (75%). Different DNA patterns were found among the strains that were isolated from various regions of the duodenum in 4 of 16 patients (25%) as shown by ERIC-PCR and in 8 of 16 patients (50%) as shown by RAPD-PCR. Sixty-three percent of the duodenal ulcer patients had H. pylori strains with a different Lewis antigen phenotype in the duodenum as compared to in the antrum, and 3 of 16 patients (19%) had strains with different Lewis antigens expressed by strains from different duodenal biopsies from the same patient. Conclusion. The results suggest that a mixed population of different H. pylori strains with marked variation, both genotypically and phenotypically, colonize the same patient. [source]


Increased Diversity and Deepened Uncertainty: Policy Challenges in a Zero-Inflation Economy,

The world economy today shows ,great diversity'. There are multiple engines of growth in various regions around the globe. Risks are diversified, as many novel financial products are being introduced and sold to a continuing flow of newcomers to the financial world. This increased diversity seems to deepen uncertainty surrounding monetary policy in two respects. First, coupled with increased competition, it may make prices less responsive to short-run demand changes than before, thus making monetary transmission mechanism less certain. In fact, Japanese IS and Phillips curves seem increasingly uncertain in the past 15 years. Second, we are in transition between one financial structure of little diversification and another of great diversification. In a transition period, information is scarce and rapidly becomes obsolete, posing real challenges to financial stability. I argue that the flexible gradualism, which the Bank has now adopted, is a prudent way to cope with such deepened uncertainty. [source]


Spatial distribution of cloud cover

Abstract Satellite systems and high-altitude platform systems working in Ka and V bands require the application of adaptive techniques in order to mitigate link degradations caused by atmospheric impairments such as those due to cloud cover. Among these techniques, resource sharing system techniques and site diversity need information on the spatial distribution of impairments caused by cloud cover, including the probability of simultaneous occurrence of adverse conditions in various regions. A study has been performed in Spain, within the framework of COST Action 280, to investigate the large-scale spatial distribution of cloud cover using synoptic meteorological data. Cloud cover distribution for individual sites and the spatial correlation properties for pairs of sites have been investigated. The geographical distributions of the values obtained from the statistical analysis have been represented in maps of contour lines using standard mapping procedures. Correlation parameters are expected to decrease with distance; however, the maps derived taking a given site as reference reveal a significant influence of climatic and geographic factors such as weather fronts, orography or the proximity to the sea. The statistics and maps obtained can be useful to optimize power sharing in multi-beam satellite systems, as suggested in this paper. Copyright © 2008 John Wiley & Sons, Ltd. [source]


Population management of rhinoceros in captivity

Captive-breeding programmes are important components of conservation strategies for rhinoceros. Rhinoceros in zoos can serve as (1) genetic and demographic reservoirs to reinforce wild populations as the need and opportunity occur, and (2) ambassadors to increase public awareness and support, especially financial, for conservation of wild populations. However, for these functions, rhinoceros in captivity must be managed scientifically and co-operatively to produce viable populations. Population-management programmes for Black rhinoceros Diceros bicornis, White rhinoceros Ceratotherium simum and Indian rhinoceros Rhinoceros unicornis are operating in various regions of the zoo world, especially North America [Species Survival Plans (SSP)] and Europe [European Endangered Species Programmes (EEP)]. Analyses indicate that rhinoceros populations in captivity are achieving variable levels of viability. In SSP and EEP populations Black rhinoceros and White rhinoceros are genetically but not demographically satisfactory, while Indian rhinoceros is healthy demographically but limited genetically. Improvement is needed and could be achieved through better management. [source]


Regional Alterations of Type I Collagen in Rat Tibia Induced by Skeletal Unloading,

Abstract Skeletal unloading induces loss of mineral density in weight-bearing bones that leads to inferior bone mechanical strength. This appears to be caused by a failure of bone formation; however, its mechanisms still are not well understood. The objective of this study was to characterize collagen, the predominant matrix protein in bone, in various regions of tibia of rats that were subjected to skeletal unloading by 4 weeks tail suspension. Sixteen male Sprague-Dawley rats (4 months old) were divided into tail suspension and ambulatory controls (eight rats each). After the tail suspension, tibias from each animal were collected and divided into five regions and collagen was analyzed. The collagen cross-linking and the extent of lysine (Lys) hydroxylation in unloaded bones were significantly altered in proximal epiphysis, diaphysis, and, in particular, proximal metaphysis but not in distal regions. The pool of immature/nonmineralized collagen measured by its extractability with a chaotropic solvent was significantly increased in proximal metaphysis. These results suggest that skeletal unloading induced an accumulation of post-translationally altered nonmineralized collagen and that these changes are bone region specific. These alterations might be caused by impaired osteoblastic function/differentiation resulting in a mineralization defect. [source]


Influence of salinity, competition and food supply on the growth of Gobiosoma robustum and Microgobius gulosus from Florida Bay, U. S. A.

The code Gobiosoma robustum and clown Microgobius gulosus gobies were grown in the laboratory over 27 days at two salinities (5 and 35), two food levels [low (a fixed proportion of initial mass) and high (saturation)] and both with and without the presence of the other species. Both species exhibited greatest growth at the high food level and the low (5) salinity. Neither species was affected by the presence of the other species, and there were no overall differences in growth between the two species. Thus, the observed competitive superiority of G. robustum over M. gulosus does not seem to confer an advantage relative to feeding success. Furthermore, as growth of G. robustum was greater at the lower salinity, it is clear that some factor other than salinity is restricting this species from north-eastern Florida Bay. Additional work on the importance of predation and food resources in various regions of Florida Bay is needed to further evaluate the underlying mechanisms responsible for the bay-wide distribution of these species. MRI-guided procedures in various regions of the body using a robotic assistance system in a closed-bore scanner: Preliminary clinical experience and limitations,

Abstract Purpose: To present the clinical setup and workflow of a robotic assistance system for image-guided interventions in a conventional magnetic resonance imaging (MRI) environment and to report our preliminary clinical experience with percutaneous biopsies in various body regions. Materials and Methods: The MR-compatible, servo-pneumatically driven, robotic device (Innomotion) fits into the 60-cm bore of a standard MR scanner. The needle placement (n = 25) accuracy was estimated by measuring the 3D deviation between needle tip and prescribed target point in a phantom. Percutaneous biopsies in six patients and different body regions were planned by graphically selecting entry and target points on intraoperatively acquired roadmap MR data. Results: For insertion depths between 29 and 95 mm, the average 3D needle deviation was 2.2 ± 0.7 mm (range 0.9,3.8 mm). Patients with a body mass index of up to ,30 kg/m2 fitted into the bore with the device. Clinical work steps and limitations are reported for the various applications. All biopsies were diagnostic and could be completed without any major complications. Median planning and intervention times were 25 (range 20,36) and 44 (36,68) minutes, respectively. Conclusion: Preliminary clinical results in a standard MRI environment suggest that the presented robotic device provides accurate guidance for percutaneous procedures in various body regions. Effects of Long-Term Hormone Treatment and of Tibolone on Monoamines and Monoamine Metabolites in the Brains of Ovariectomised, Cynomologous Monkeys

The effects of long-term hormone treatment on monoamines and monoamine metabolites in different regions of the primate brain were examined and compared. Ovariectomised Cynomologous monkeys received daily oral administration of either conjugated equine oestrogens (CEE), CEE + medroxyprogesterone acetate (MPA), or a low or high dose of tibolone, for a period of 2 years. Tissue punches collected from frozen sections through various regions of the forebrain, midbrain, and hindbrain were assayed for levels of dopamine, dihydroxyphenylacetic acid (DOPAC), serotonin, 5-hydroxyindole acetic acid (5-HIAA), and norepinephrine by high-performance liquid chromatography. Few differences between hormone-treated animals and ovariectomised controls were observed. No statistically significant effects of CEE relative to controls were detected in any of the seven brain regions analysed. Animals treated with CEE + MPA showed significant reductions in 5-HIAA in the dorsal raphe nucleus, a significant reduction in dopamine in the hypothalamus, and a significant reduction in serotonin (5-HT) levels in area 8AD of the frontal cortex. Similar to CEE, no significant effects of tibolone relative to controls were detected; however, animals treated with high-dose tibolone showed a decrease in 5-HT levels in the frontal cortex that approached significance and was similar to the effect of CEE + MPA. Collectively, the findings suggest that long-term oral administration of these compounds has relatively few effects on the levels of dopamine, serotonin, and their primary metabolites in the primate brain. This differs from the significant effects on serotonergic and dopaminergic systems detected following parenteral treatment with oestradiol and progesterone, and likely reflects differences between the effects of treating with CEE + MPA versus oestradiol and progesterone on brain monoaminergic systems. Description of distributed features of the nestin-containing cells in brains of adult mice: A potential source of neural precursor cells

Abstract The distribution of neural precursor cells (NPCs) in adult mice brain has so far not been described. Therefore, we investigated the distribution of NPCs by analyzing the nestin-containing cells (NCCs) in distinct brain regions of adult nestin second-intron enhancer-controlled LacZ reporter transgenic mice through LacZ staining. Results showed that NCCs existed in various regions of adult mouse brain. In cerebellum, the greatest number of NCCs existed in cortex of the simple lobule, followed by cortex of the cerebellar lobule. In olfactory bulb, NCCs were most numerous in the granular cell layer, followed by the mitral cell layer and the internal plexiform, glomerular, and external plexiform layers. In brain nuclei (nu), NCCs were most numerous in the marginal nu, followed by the brainstem and diencephalon nu. NCCs in sensory nu of brainstem were more numerous than in motor nu, and NCCs in the dorsal of sensory nu were more numerous than in the ventral part. In brain ventricle systems, NCCs were largely distributed in the center of and external to the lateral ventricle, the inferior part of the third ventricle, the dorsal and inferior parts of the fourth ventricle, and the gray matter around the cerebral aqueduct. NCCs in the left vs. right brain were not significantly different. These data collectively indicate that NCCs were extensively distributed in the cerebellum and olfactory bulb, the partial nu of the marginal system, the partial brain nu adjacent to brain ventricle systems, the subependymal zone, and the cerebral cortex around the marginal lobe and were a potential source of NPCs. © 2009 Wiley-Liss, Inc. Nurses' Use of Physical Restraints in Four Turkish Hospitals

Purpose: (a) Determine the frequency and types of physical restraints used by nurses in intensive care units, emergency departments, and neurosurgery wards; (b) understand nurses' attitudes toward physical restraint; and (c) identify complications in physically restrained patients. Design and Methods: This descriptive, cross-sectional study was carried out on 254 nurses working in intensive care units, emergency departments, and neurosurgery wards in four Turkish hospitals where physical restraints were used. The whole population was studied without any sampling, and data were collected via semistructured interviews from July 20 to September 6, 2005. Findings: Nurses used either wrist, ankle, or whole body restraints at various levels. Those nurses who worked in surgical intensive care units and emergency departments and had in-service training used more physical restraint than did others. Only a third of nurses decided on physical restraint together with physicians and three-fourths tried alternative methods. Nurses reported edema and cyanosis on wrist and arm regions, pressure ulcers on various regions, and aspiration and breathing difficulties in relation to physical restraint. Moreover, they reported 9 deaths of patients in chest restraints. Reduction in the frequency of caregiving was related to complications. Conclusions: Actions to reduce use of and complications from physical restraints should include attention to nurse staffing and education about use of restraints. [source]

Antimicrobial peptides from phagocytes and epithelia contribute to this antimicrobial barrier. Using antibodies and in situ hybridization, we explored antimicrobial peptide expression in the varied epithelia of the periodontium and in cultured gingival epithelial cells. In gingival tissue, mRNA for the ,-defensins, human beta-defensin 1 (hBD-1) and human beta-defensin 2 (hBD-2) was predominately localized in suprabasal stratified epithelium and the peptides were detected in upper epithelial layers consistent with the formation of the stratified epithelial barrier. In cultured epithelial cells, both hBD-1 and -2 peptides were detected only in differentiating, involucrin-positive epithelial cells, although hBD-2 required stimulation by proinflammatory mediators or bacterial products for expression. ,-defensins were not detected in junctional epithelium (JE) that serves as the attachment to the tooth surface. In contrast, ,-defensins and cathelicidin family member LL-37 were detected in polymorphonuclear neutrophils (PMNs) that migrate through the JE, a localization that persists during inflammation, when the JE and surrounding tissue are highly infiltrated with PMNs. Thus, the undifferentiated JE contains exogenously expressed ,-defensins and LL-37, and the stratified epithelium contains endogenously expressed ,-defensins. These findings show that defensins and other antimicrobial peptides are localized in specific sites in the gingiva, are synthesized in different cell types, and are likely to serve different roles in various regions of the periodontium. Morphological and Pathological Variability in Rice Isolates of Rhizoctonia solani and Molecular Analysis of their Genetic Variability

Abstract Nineteen isolates of Rhizoctonia solani collected from different rice varieties grown in various regions of Punjab were studied for their morphological and pathological characterization. Majority of the isolates were fast growing with raised and fluffy colonies and hyphal width of 9.6 ,m while four exhibited moderate growth rate. Colony colour in all except two isolates was light yellowish brown. While sclerotial number per 5.0 mm culture disc of the test isolates ranged between 2.1 and 11.2 mm, their size varied between 1.31 and 2.08 mm. Sclerotial colour in all except two isolates was dark brown and most of these were found scattered in the colony. There was no relationship between morphologically similar isolates and their pathogenic behaviour. Majority of the isolates produced lesion length between 45.6 and 58.2 mm on detached rice leaves (cv. PR116). Molecular characterization of genetic diversity in the test isolates was studied by using 10 inter simple sequence repeats (ISSR) and eight random amplified polymorphic DNA (RAPD) markers. The size of amplified DNA bands ranged from 0.25,3.0 to 0.5,4.0 kb with ISSR and RAPD markers, respectively. Combined data set of 155 DNA markers were analysed with UPGMA resulting five clusters with 49,89% genetic similarity. Most of the isolates showed grouping specific to the host variety. Out of these two types of DNA markers, RAPD markers were able to detect more genetic variability when compared to ISSR markers. [source]

This essay examines citizen involvement in community housing issues through Habitat for Humanity as a faith, based expansion of social capital in urban communities. This article expands Putnam's model of social capital to include criteria for evaluating the conditions under which social capital formation has a positive impact on the larger community. Using a representative sample of nine cities from various regions of the US, it examines the functioning of the Habitat affiliate in each of these cities and the attitudes and motivations of their most active volunteers. Habitat has emerged as a highly effective volunteer, non, profit producer of housing for lower income persons, yet the nature of the social capital created by this organization also reflects the contradictions raised by such an undertaking in a complex urban environment characterized by deep social divisions. Differences in hepatitis A seroprevalence among geographical regions in Turkey: a need for regional vaccination recommendations

Summary., Hepatitis A is a worldwide vaccine-preventable infection. Recommendation of vaccination depends on the endemicity of the disease. The World Health Organization recommends universal hepatitis A vaccination in intermediate areas; however, there is no need of mass vaccination in high and low endemicity regions. Therefore, most of the countries are using a vaccination policy according to the endemicity characteristic representing the whole of the country. The endemicity of this infection varies due to sanitary and hygiene conditions and socioeconomic differences among the countries and in various regions of the same country. A sample of 1173 persons between the age of 0 and 91 years from nine randomly selected medical centres from five different geographical centres of Turkey were tested for the level of anti-hepatitis A virus (anti-HAV) immunoglobulin-G antibodies using an enzyme-linked immunosorbent assay. The overall prevalence of anti-HAV antibodies was 64.4% (1142/1173). While the rate of sero-positivity was over 80% in the 5,9 age group and more than 90% after 14 years of age in south-eastern and eastern regions, it was lower than 50% at the age of 5,9 years in central and western regions and remains under 80% in those areas. We conclude that the differences observed in HAV sero-positivity among various geographical regions in Turkey support a universal HAV immunization policy for children currently living in regions of intermediate endemicity. Characterization of Bacillus subtilis strains in Thua nao, a traditional fermented soybean food in northern Thailand

Abstract Aims:, To clarify the diversity of Bacillus subtilis strains in Thua nao that produce high concentrations of products useful in food manufacturing and in health-promoting compounds. Method and Results:, Production of amylase, protease, subtilisin NAT (nattokinase), and gamma-polyglutamic acid (PGA) by the Bacillus subtilis strains in Thua nao was measured. Productivity of protease NAT by these strains tended to be higher than by Japanese commercial natto-producing strains. Molecular diversity of isolated strains was analysed via randomly amplified polymorphic DNA-PCR fingerprinting. The strains were divided into 19 types, including a type with the same pattern as a Japanese natto-producing strain. Conclusion:,B. subtilis strains that could be a resource for effective production of protease, amylase, subtilisin NAT, or PGA were evident in Thua nao produced in various regions in northern Thailand. Significance and Impact of the Study:, This study clearly demonstrated the value of Thua nao as a potential resource of food-processing enzymes and health-promoting compounds. [source]


Expression of brain natriuretic peptide in the rat heart studies during heart growth and in relation to sympathectomy

Abstract Brain natriuretic peptide (BNP) might be of importance during heart development and is described to be increasingly expressed in congestive heart failure and to affect the progress of this condition. However, details in the normal expression of BNP are still unclear in various parts of the adult and growing heart, including the conduction system. In this study, we investigated the expression of BNP in relation to that of atrial natriuretic peptide (ANP) in the growing as well as in the adult rat heart. The effects of chemical sympathectomy in adult rats were also examined. Contrary to previous BNP immunohistochemical studies, the BNP antiserum was preabsorbed with an excess of ANP before staining to abolish the crossreactivity with ANP. There was a pronounced BNP immunoreaction in the auricles, the trabeculated ventricular walls, and the peripheral parts of the conduction system at 0,1 days postnatally. The degree of immunoreaction gradually decreased with increasing age. A similar developmental pattern was seen concerning ANP expression, but the magnitude of the latter clearly exceeded that for BNP. Immunoreaction for BNP was never detected in the atrioventricular (AV) node and AV bundle at any stage. In contrast to the situation for ANP previously observed, no obvious changes in BNP immunoreaction patterns were observed in response to sympathectomy. This is the first study to thoroughly demonstrate the expression of BNP in the various regions of the rat heart during growth and in the normal and sympathectomized adult stage. Effects of FSH receptor deletion on epididymal tubules and sperm morphology, numbers, and motility

Abstract Follicle stimulating hormone (FSH) interacts with its cognate receptor (R) on Sertoli cells within the testis and plays an important role in the maintenance of spermatogenesis. Male FSH-R knockout (FORKO) mice show fewer Sertoli cells and many that are structurally abnormal and as a consequence fewer germ cells. Lower levels of serum testosterone (T) and androgen binding protein (ABP) also occur, along with reduced fertility. To assess the effects of FSH-R depletion as an outcome of testicular abnormalities, sperm from the cauda epididymidis were counted and examined ultrastructurally. As reduced fertility may also reflect changes to the epididymis, the secondary responses of the epididymis to lower T and ABP levels were also examined by comparing differences in sizes of epididymal tubules in various regions of FORKO and wild type (WT) mice. Sperm motility was evaluated in FORKO mice and compared to that of WT mice by computer assisted sperm analysis (CASA). Quantitatively, the data revealed that epithelial areas of the caput and corpus epididymidis were significantly smaller in FORKO mice compared to WT mice. Cauda epididymal sperm counts in FORKO mice were also much lower than in WT mice. This resulted in changes to 9 out of 14 sperm motility parameters, related mostly to velocity measures, which were significantly lower in the FORKO mice. The greatest change was observed relative to the percent static sperm, which was elevated by 20% in FORKO mice compared to controls. EM analyses revealed major changes to the structure of the heads and tails of cauda luminal sperm in FORKO mice. Taken together these data suggest a key role for the FSH receptor in maintaining Sertoli cells to sustain normal sperm numbers and proper shapes of their heads and tails. In addition, the shrinkage in epididymal epithelial areas observed in FORKO mice likely reflect direct and/or indirect changes in the functions of these cells and their role in promoting sperm motility, which is noticeably altered in FORKO mice. Lasting changes in NMDAR1 mRNA level in various regions of cerebral cortex in epileptogenesis of amygdaloid-kindled rat

Abstract The involvement of NMDA receptor subunit, NR1, with kindling phenomenon has been reported, but the role of NR1 in epileptogenesis is still unknown. We have examined the expression levels of NR1 mRNA in the cerebral cortices of amygdaloid-kindled rats. Northern blot analysis showed a significant increase in NR1 mRNA expression level in the ipsilateral frontal and temporal cortices at 4 weeks after the last generalized seizure. At the same time, NR1 mRNA decreased in the bilateral piriform cortices. These data suggest that NR1-mediated transmission may have an impact in the neurobiological basis of enduring epileptogenesis. Foreign debt and economic growth

This paper empirically examines the relationship between government foreign debt and the growth rate of per capita GDP based on a total sample of 77 countries, as well as sub-samples of various regions. Cross-sectional estimates of the coefficient of foreign debt based on the total sample have a negative sign, but are not always statistically significant. Available data from African countries indicate that foreign debt and the growth rate of per capita GDP were negatively related at a high level of significance. For industrialized and Latin American sub-samples, this relationship is negative but statistically insignificant. Detection of JC virus DNA fragments but not proteins in normal brain tissue

Objective Progressive multifocal leukoencephalopathy (PML) is a fatal demyelinating disease of the white matter affecting immunocompromised patients that results from the cytolytic destruction of glial cells by the human neurotropic JC virus (JCV). According to one model, during the course of immunosuppression, JCV departs from its latent state in the kidney and after entering the brain, productively infects and destroys oligodendrocytes. The goal of this study was to test the hypothesis that JCV may reside in a latent state in a specific region of the brains of immunocompetent (non-PML) individuals without any neurological conditions. Methods Gene amplification was performed together with immunohistochemistry to examine the presence of JCV DNA sequences and expression of its genome in five distinct regions of the brain from seven immunocompetent non-PML individuals. Results Although no viral proteins were expressed in any of these cases, fragments of the viral DNA were present in various regions of normal brain. Laser-capture microdissection showed the presence of JCV DNA in oligodendrocytes and astrocytes, but not in neurons. Interpretation The detection of fragments of viral DNA in non-PML brain suggests that JCV has full access to all regions of the brain in immunocompetent individuals. Thus, should the immune system become impaired, the passing and/or the resident virus may gain the opportunity to express its genome and initiate its lytic cycle in oligodendrocytes. Vegetation restoration of secondary bare saline-alkali patches in the Songnen plain, China

Abstract Questions: What factors limit vegetation restoration of secondary bare saline-alkaline patches (BSAP) in the Songnen grassland of northeast China? Is there any adaptive approach to promote revegetation in the shortest time possible and at a low cost? Location: Northeast China. Methods: Considering the climate, soil saline-alkalization and available seed sources, a new approach was adopted to restore vegetation in BSAP, which were formed by the degradation of typical Leymus chinensis grasslands owing to long-term overgrazing. The experimental treatments included no treatment (CK), fencing (F), fencing+inserting cornstalks (FS), fencing+inserting cornstalks+sowing L. chinensis (FSL) and fencing+inserting cornstalks+sowing Puccinellia chinampoensis (FSP). The assumptions behind inserting cornstalks were not only that they would create safe sites for initial revegetation but also that they would enhance seed input by trapping and containing the seeds from seed movement on the BSAP surface. Results: Seed bank shortage was an important factor limiting initial revegetation in BSAP; seed movement on the BSAP surface could provide the necessary seed source if it were contained by effective measures. Vegetation at the sites FS, FSL and FSP was restored well in terms of the above-ground biomass and coverage. Inserted cornstalks acted as safe sites that enhanced the plant survival rate in BSAP; they also enhanced the ability to contain seed movement, thus providing a seed source for initial revegetation. Along with initial revegetation, tussocks around cornstalks can provide better safe sites, which in turn can accelerate subsequent vegetation restoration in BSAP. Conclusions: The approach entails the strategic use of diverse seed sources and the construction of safe sites with agricultural byproducts (cornstalks); therefore, it is a low-cost method and can be used on a widespread scale. The results provide vigorous support in favor of vegetation restoration in BSAP and severely degraded grasslands in the region. In practice, this approach can be used in degraded ecosystems with compacted soil surfaces (including arid and salt-affected soils) to promote revegetation in various regions. [source]


Comprehensive review of the records of the biota of the Indian Seas and introduction of non-indigenous species

Abstract 1.Comparison of the pre-1960 faunal survey data for the Indian Seas with that for the post-1960 period showed that 205 non-indigenous taxa were introduced in the post-1960 period; shipping activity is considered a plausible major vector for many of these introductions. 2.Of the non-indigenous taxa, 21% were fish, followed by Polychaeta (<11%), Algae (10%), Crustacea (10%), Mollusca (10%), Ciliata (8%), Fungi (7%), Ascidians (6%) and minor invertebrates (17%). 3.An analysis of the data suggests a correspondence between the shipping routes between India and various regions. There were 75 species common to the Indian Seas and the coastal seas of China and Japan, 63 to the Indo-Malaysian region, 42 to the Mediterranean, 40 and 34 to western and eastern Atlantic respectively, and 41 to Australia and New Zealand. A further 33 species were common to the Caribbean region, 32 to the eastern Pacific, 14 and 24 to the west and east coasts of Africa respectively, 18 to the Baltic, 15 to the middle Arabian Gulf and Red Sea, and 10 to the Brazilian coast. 4.The Indo-Malaysian region can be identified as a centre of xenodiversity for biota from Southeast Asia, China, Japan, Philippines and Australian regions. 5.Of the introduced species, the bivalve Mytilopsis sallei and the serpulid Ficopomatus enigmaticus have become pests in the Indian Seas, consistent with the Williamson and Fitter ,tens rule'. Included amongst the biota with economic impact are nine fouling and six wood-destroying organisms. 6.Novel occurrences of the human pathogenic vibrios, e.g. Vibrio parahaemolyticus, non-01 Vibrio cholerae, Vibrio vulnificus and Vibrio mimicus and the harmful algal bloom species Alexandrium spp. and Gymnodinium nagasakiense in the Indian coastal waters could be attributed to ballast water introductions. 7.Introductions of alien biota could pose a threat to the highly productive tropical coastal waters, estuaries and mariculture sites and could cause economic impacts and ecological surprises. 8.In addition to strict enforcement of a national quarantine policy on ballast water discharges, long-term multidisciplinary research on ballast water invaders is crucial to enhance our understanding of the biodiversity and functioning of the ecosystem. A birth-to-death view of mRNA from the RNA recognition motif perspective,

Abstract RNA binding proteins are a large and varied group of factors that are the driving force behind post-transcriptional gene regulation. By analogy with transcription factors, RNA binding proteins bind to various regions of the mRNAs that they regulate, usually upstream or downstream from the coding region, and modulate one of the five major processes in mRNA metabolism: splicing, polyadenylation, export, translation and decay. The most abundant RNA binding protein domain is called the RNA Recognition Motif (RRM)1. It is probably safe to say that an RRM-containing protein is making some contact with an mRNA throughout its existence. The transcriptional counterpart would likely be the histones, yet the multitude of specific functions that are results of RRM based interactions belies the universality of the motif. This complex and diverse application of a single protein motif was used as the basis to develop an advanced graduate level seminar course in RNA:protein interactions. The course, utilizing a learner-centered empowerment model, was developed to dissect each step in RNA metabolism from the perspective of an RRM containing protein. This provided a framework to discuss the development of specificity for the RRM for each required process. [source]
