Variable extent of sex-biased dispersal in a strongly polygynous mammal

Abstract For mammals with a polygynous mating system, dispersal is expected to be male-biased. However, with the increase in empirical studies, discrepancies are arising between the expected and observed direction/extent of the bias in dispersal. In this study, we assessed sex-biased dispersal in red deer (Cervus elaphus) on 13 estates from the Scottish Highlands. A total of 568 adult individuals were genotyped at 21 microsatellite markers and sequenced for 821 bp of the mitochondrial control region. Estimates of population structure with mitochondrial sequences were eight times larger than that obtained with microsatellite data (Fst,-mtDNA = 0.831; Fst,-micros = 0.096) indicating overall male-biased dispersal in the study area. Comparisons of microsatellite data between the sexes indicated a predominance of male-biased dispersal in the study area but values of FST and relatedness were only slighter larger for females. Individual-based spatial autocorrelation analysis generated a similar pattern of relatedness across geographical distances for both sexes, with differences only significant at two distance intervals (25,30 and 70,112 km). Patterns of relatedness differed between estates, male biased-dispersal was detected in eight estates but no sex-biased dispersal was found in the remaining five. Neither population density nor landscape cover was found to be associated with the patterns of relatedness found across the estates. Differences in management strategies that could influence age structure, sex ratio and dispersal behaviour are proposed as potential factors influencing the relatedness patterns observed. This study provides new insights on dispersal of a strongly polygynous mammal at geographical scales relevant for management and conservation. [source]


High frequency of chimerism in transplanted livers

Recent studies have shown that primitive stem cells can mobilize and differentiate into hepatocytes. We investigated the time and extent in which cells of recipient origins could differentiate into hepatocytes and other cells in human liver allografts. Microsatellite analysis, which can assess quantitatively the proportions of recipient and donor DNA, was performed in posttransplantation liver biopsy specimens from 17 patients at various times. Combined fluorescence in situ hybridization (FISH) for Y chromosome and immunofluorescence for different cell types was also performed in 10 of these cases with sex mismatch. Organ chimerism in the transplanted livers was found to be of variable extent, and the recipients' DNA in the posttransplantation liver biopsy specimens (excluding portal tracts) amounted up to 50%. The recipient DNA in the posttransplantation liver biopsy specimens increased after liver transplantation by as early as 1 week, peaked at around 30 to 40 weeks, and could be shown 63 weeks after transplantation. Most (64%,75%) of the recipientderived cells showed macrophage/Kupffer cell differentiation. Only up to 1.6% of the recipient-derived cells in the liver grafts showed hepatocytic differentiation in the liver grafts and made up 0.62% of all hepatocytes of both donor and recipient origins. These livers had mild or minimal injury histologically. In conclusion, our results show that most of the recipient-derived cells in the liver allografts were macrophages/Kupffer cells and only a small proportion of hepatocytes was recipient derived. However, with regard to recipient-derived hepatocytes, our data cannot distinguish between transdifferentiation and cell fusion. (Hepatology 2003;38:989,998). [source]


CRETACEOUS CARBONATES IN THE ADIYAMAN REGION, SE TURKEY: AN ASSESSMENT OF BURIAL HISTORY AND SOURCE-ROCK POTENTIAL

The burial history and source-rock potential of Cretaceous carbonates in the Adiyaman region of SE Turkey have been investigated. The carbonates belong to the Aptian-Campanian Mardin Group and the overlying Karabogaz Formation. The stratigraphy of these carbonates at four well locations was recorded. At each well, the carbonate succession was found to be incomplete, and important unconformities were present indicating periods of non-deposition and/or erosion. These unconformities are of variable extent. When combined with the effects of rapid subsidence and sedimentation which took place in the SW of the Adiyaman region during end-Cretaceous foredeep development, they have resulted in variations in the carbonates' present-day burial depths, thereby influencing the regional pattern of source-rock maturation and the timing of oil generation. Burial history curves indicate that the carbonates' maturity increases from SW to NE, towards the Late Cretaceous thrust belt. Predicted levels of maturity for the Mardin Group are consistent with measured geochemical data from three of the wells in the study area (the exception being well Karadag-1). Three potential source-rock intervals of Cretaceous age have been identified. Two of these units , the Derdere and Karababa Formations of the Mardin Group , are composed of shallow-water carbonates which were deposited on the northern margin of the Arabian Platform. The third source-rock unit, the overlying Karabogaz Formation, is composed of pelagic carbonates which were deposited during a regional transgression. These potential source-rock intervals contain marine organic matter dominated by Type II kerogen. Total organic carbon contents range from 0.5 to 2.9 %. Time-temperature analyses indicate that the Mardin Group carbonates are immature to marginally mature at well locations in the SW of the study area, and are mature at western and NE well locations. The onset of oil generation in these Cretaceous source rocks took place between the middle Eocene (48 million yrs ago) and the Oligocene (28 million yrs ago). [source]


Loss of tocopherols and formation of degradation compounds at frying temperatures in oils differing in degree of unsaturation and natural antioxidant content

Abstract Samples of oils of different degrees of unsaturation, namely palm olein, olive oil, high-linoleic sunflower oil, high-oleic sunflower oil, rapeseed oil and soybean oil, were heated at 180,°C for 2, 4, 6, 8 and 10,h in the presence or absence of their natural antioxidants. Also, tocopherol-stripped oils were supplemented with ,-tocopherol (500,mg,kg,1), ,-tocopherol (500,mg,kg,1) or a mixture of ,-, ,-, ,- and ,-tocopherols (250,mg,kg,1 each) and heated under the same conditions. Losses of tocopherols and formation of polymeric triacylglycerols were followed. Total polar compounds were also evaluated after 10,h of heating. Results demonstrated that tocopherols were lost very rapidly, in the expected order, with ,-tocopherol being the least stable. Polymeric and polar compound formation during heating was inhibited to a variable extent, being more dependent on the natural content and type of tocopherols than on the degree of unsaturation of the oil. For example, polymeric and polar compound contents in soybean oil were significantly lower than those found in high-linoleic sunflower oil. However, the expected influence of the degree of unsaturation was evident when oils were unprotected or possessed identical initial antioxidant contents. Finally, levels of degradation compounds after 10,h of heating were not dependent on the remaining content of antioxidants. © 2002 Society of Chemical Industry [source]


Review: Dupuytren's disease in Asia and the migration theory of Dupuytren's disease

Abstract Background:, The presentation of a Chinese patient with unilateral Dupuytren's disease (DD) prompted a literature search and a review of the epidemiology of DD in the Asian population as it has never been cumulatively reported. The purpose of this paper is to review all the reported cases of DD in the literature and aetiological links to DD elsewhere. Methods:, The literature was searched with a wide variety of terms, and subsequent references were analysed and further references investigated for other reported cases of DD in the Asian population. Results:, This review found 595 cases and has shown that DD is present to a variable extent in China (96 cases), Thailand (19 cases), Vietnam (one case), India (15 cases) and Japan (474 cases). A total of 54% had bilateral disease. Risk factors (diabetes, trauma, epilepsy, alcoholism, manual labour) were reportedly present in 65% of the patients, and a positive family history was reported in 9%. The average patient age was 67 years. Conclusion:, This review shows that there is a low but significant incidence of DD across Asia, which supports the hypothesis of a widespread genetic susceptibility to the disease. Therefore, the prevalence of DD in this community is not likely due to sporadic genetic mutation as previously presumed but rather individual genetic susceptibility and that risk factors play a major role in the expression of DD in this population. Immunohistochemical expression of vascular endothelial growth factor, matrix metalloproteinase 2, and matrix metalloproteinase 9 in cutaneous melanocytic lesions

Abstract BACKGROUND Vascular endothelial growth factor (VEGF), an endothelial cell mitogen, plays a hierarchical role in regulating physiologic and pathologic angiogenesis. Moreover, the transformation from noninvasive to invasive carcinomas is accompanied by focal disruption and discontinuity of the basement membrane. Several groups of proteases have been implicated in tumor cell invasion, including the 72-kDa gelatinase A/Type IV collagenase (matrix metalloproteinase 2 [MMP-2]) and the 92-kDa gelatinase B/Type IV collagenase (MMP-9). METHODS The authors assessed the immunohistochemical expression of VEGF and metalloproteinases MMP-2 and MMP-9 in paraffin embedded biopsy specimens of malignant melanomas (18 invasive melanomas and 10 in situ melanomas); dysplastic nevi with architectural disorder and cytologic atypia of melanocytes; Spitz nevi; and compound or predominantly intradermal, ordinary, benign melanocytic nevi. RESULTS Strong cytoplasmic staining for VEGF was observed in melanoma cells in as many as 77% of primary invasive melanomas, whereas only 25% of the in situ melanomas exhibited a detectable immunoreactivity for VEGF. It is interesting to note that no immunoreactivity was shown by any nevi; Spitz nevi, in particular, showed negative immunoreactivity to VEGF. Invasive melanomas and in situ melanomas displayed coexpression of MMP-2 and MMP-9, although to a variable extent. In particular, high MMP-2 staining was observed in 14 of 18 invasive melanomas; moreover, strong MMP-2 expression also was observed in 60% of in situ melanomas, whereas the residual 40% of those melanomas showed a moderate level of positivity. CONCLUSIONS On the basis of the current data showing that malignant melanocytic tumors displayed strong VEGF expression, whereas benign melanocytic proliferations showed no immunoreactivity for VEGF, VEGF also may be used as a discriminating factor to distinguish malignant melanoma from lesions of uncertain histology. FSH and ovarian response: spontaneous recovery of pituitary,ovarian activity during the pill-free period vs. exogenous recombinant FSH during high-dose combined oral contraceptives

Summary ojbective Compare spontaneous recovery of pituitary,ovarian activity during the pill-free period following the correct use of low-dose oral contraceptives and subsequent ovarian function during the administration of exogenous recombinant FSH (recFSH) after switching to continued Lyndiol® (2·5 mg lynestrenol + 0·05 mg ethinyl-oestradiol) medication. design Prospective, randomized, group-comparative, single-centre study. Following the monitoring of the pill-free period (week 1) and subsequent treatment with Lyndiol® (for a total of 5 weeks), all subjects were randomly allocated to one of four groups receiving daily FSH injections for 1 week [75, 150, 225 IU recFSH or 150 IU purified urinary FSH (uFSH)] during the fourth week of Lyndiol® use. patients Thirty-six healthy volunteers aged 18,39 years, prestudy oral contraceptive use for at least 3 months, cycle length between 24 and 35 days. measurements Serum FSH, LH and oestradiol (E2) concentrations as well as transvaginal ultrasound assessment of the number and diameter of follicles > 2 mm were used to monitor pituitary ovarian function. results At the start of the pill-free period following the prestudy contraceptive medication, 67% of the women presented with LH and FSH levels < 1 IU/l and only one follicle > 10 mm was observed. Initial levels of LH and FSH correlated (P < 0·05) with the extent of pituitary,ovarian activity during the pill-free period. At the end of the pill-free period a follicle > 10 mm had emerged in one subject only. During the first 3 days of Lyndiol® use, seven women (19%) eventually showed at least one follicle > 10 mm. During combined exogenous FSH and Lyndiol® administration, LH levels remained completely suppressed (, 0·5 IU/l) in all women studied. FSH levels and number and size of follicles increased with increasing doses of exogenous FSH in a dose-dependent manner. E2 levels remained low in all groups (< 150 pmol/l). During the week following FSH administration, FSH levels and E2 levels decreased gradually while the number of follicles > 10 mm still increased. conclusions We have confirmed that dominant follicles > 10 mm are present at the end of the pill-free period and during the first days after resumption of pill intake. Once follicles > 10 mm arose at the end of the pill-free period, continued use of Lyndiol® did not reduce follicle diameters. One week of Lyndiol® reduces pituitary,ovarian activity to levels observed after 3 weeks of low-dose pills. FSH administration during Lyndiol® resulted in dose-dependent follicle growth despite extremely low LH levels. E2 secretion (56 ± 51 pmol/l) occurred to a limited and variable extent along with extremely low serum LH concentrations. Recovery of pituitary,ovarian activity at the end of the pill-free period is comparable to FSH levels and follicle dynamics following 7 days of 75,150 IU/l recFSH. [source]


Human granulocytic ehrlichiosis in Europe

Ehrlichiosis comprises a group of emerging tick-borne infectious diseases caused by obligate intracellular Gram-negative bacteria that infect leukocytes. Infections caused by members of the genus Ehrlichia have been described in animals and humans, but to date there are no convincing reports of the presence of other types of human ehrlichiosis different from human granulocytic ehrlichiosis (HGE) in Europe. The European vector is the same as that of Lyme borreliosis, the hard tick Ixodes ricinus, and HGE has a similar epidemiology to that of Borrelia burgdorferi infection. Across Europe, I. ricinus is infected to a variable extent (0.4,66.7%) with the causative agent Ehrlichia (Anaplasma) phagocytophila genogroup, and since its first description in Slovenia in 1997, details of 15 patients have been published. Diagnosis requires careful consideration of all circumstances and symptoms (history of tick bite and the presence of a flu-like syndrome with variable degrees of anemia, thrombocytopenia, and leukopenia, and elevated liver enzymes). Some differences can be seen between US and European HGE patients. European HGE cases have a less severe course, and the presence of morulae is uncommon. In Europe, verification of HGE has been based on PCR and immunofluorescence antibody tests, because no isolation from humans has been reported. [source]


Mono and double polar [4 + 2+] Diels,Alder cycloaddition of acylium ions with O -heterodienes

Abstract Gas-phase reactions of acylium ions with ,,,-unsaturated carbonyl compounds were investigated using pentaquadrupole multiple-stage mass spectrometry. With acrolein and metacrolein, CH3,C+O, CH2CH,C+O, C6H5,C+O, and (CH3)2N,C+O react to variable extents by mono and double polar [4 + 2+] Diels,Alder cycloaddition. With ethyl vinyl ketone, CH3,C+O reacts exclusively by proton transfer and C6H5,C+O forms only the mono cycloadduct whereas CH2CH,C+O and (CH3)2N,C+O reacts to great extents by mono and double cycloaddition. The positively charged acylium ions are activated O -heterodienophiles, and mono cycloaddition occurs readily across their C+O bonds to form resonance-stabilized 1,3-dioxinylium ions which, upon collisional activation, dissociate predominantly by retro-addition. The mono cycloadducts are also dienophiles activated by resonance-stabilized and chemically inert 1,3-dioxonium ion groups, hence they undergo a second cycloaddition across their polarized CC ring double bonds. 18O labeling and characteristic dissociations displayed by the double cycloadducts indicate the site and regioselectivity of double cycloaddition, which are corroborated by Becke3LYP/6,311++G(d,p) calculations. Most double cycloadducts dissociate by the loss of a RCO2COR1 molecule and by a pathway that reforms the acylium ion directly. The double cycloadduct of the thioacylium ion (CH3)2N,C+S with acrolein dissociates to (CH3)2N,C+O in a sulfur-by-oxygen replacement process intermediated by the cyclic monoadduct.
